We provide the data of the olfactory bulbs (OB) development in the human fetuses on the stages from 8 week to birth. Immunochistochemical markers of presynaptic terminals (anti-SNAP-25, -synapsin-I, -synaptophysin) were used to evaluate the maturation of the OB. Differentiation of the OB layers begins from periphery, which implicitly evidences that growth of the olfactory nerves fibers induses not only anatomical differentiation of the OB, but also differentiation of its functional layers. The sites of the developing glomerulus are revealed using the immunochistochemical prosedure on the stage before distinct glomerulus can be identified with common histological procedure. OB conductive system demonstrates immunoreactivity with the antibodies to the presynaptic proteins on the all stages from 10-11 weeks of fetus development. Four stages of the OB development are described. All functional layers of the OB are mature at the 22-weeks stage. Further differentiation of the OB neuroblasts, including lamina formation of the internal granular leyer, glomerular layer development, OB growth continue after 20-22 weeks stage until 38-40 weeks of the fetus develoment. Patterns of the immunoreactivity with antibodies to SNAP-25, synapsin-I and synaptophysin are completely appropriate to those of adult's OB on the 38-40 weeks of the prenatal development. Complete maturity of the human OB is achived at 38-40 weeks of the prenatal development.

The effect of bacterial lipopolysaccharide endotoxin (LPS), immune system activator, on differentiation and migration of gonadotropin-releasing, hormone producing neurons in rat embryogenesis has been studied. Intraperitoneal introduction of LPS (18 jg/kg) to pregnant rats on the 12th day of pregnancy led to 50% decrease in total number of GRH-neurons in the forebrain of 17-day-old embryos and 17% decrease in 19-day-old embryos. At the same time, the number of GRH-neurons in the nasal area of the head of 17- and 19-day-old embryos increased by 40 and 50%, respectively, whereas it increased by 20% in olfactory bulbs of 17-day-old embryos and did not changed in olfactory bulbs of 19-day-old embryos. Neither the total number of neurons nor their distribution patterns were affected by the introduction of LPS into pregnant rats on the 15th day of pregnancy. Singular localization of GRH-neurons in embryo forebrain was observed after LPS administration, whereas the neurons were located by groups of 3-4 cells in rostral areas. Therefore, at the early stages of pregnancy, LPS was shown to suppress initial stages of differentiation and migration of GRH producing neurons. The effects observed in our study may be mediated by LPS-induced, proinflammatory cytokines.

The distribution of c-Fos-immunopositive neurons was examined in the mitral/tufted and granular cell layers in the medium part of the main olfactory bulbs of 18-day-old rats after they had been trained for propionic acid vapour-guided search for dam in the Y-maze. On the next day these pups exhibited a strong preference for the propionic acid odor as compared to the control pups trained for this task without the odor cue and odor-familiarized pups exposed to propionic acid as a novel neutral stimulus. Exposure to propionic acid produced a moderate activation of c-Fos expression, mainly in the granular layer of the dorsomedial part of the bulb. Training in the Y-maze devoid of odor cues resulted in diffuse increase in the number of c-Fos-positive neurons both in the mitral and granular cell layers in all parts of the olfactory bulb. Maze training with the odor cue produced activation of c-Fos expression (which significantly exceeded the non-odor Y-maze group) in the dorsomedial olfactory bulb. These data suggest that associative olfactory conditioning results in activation of c-Fos expression that combines the effect of diffuse motivational excitation and specific olfactory input to the neurons which process odor cues.

This review is concerned with neurogenesis in the mature mammalian brain with emphasis on cell population renewal in the olfactory bulb (OB). The structural and functional features of the OB are considered along with data on neurotropic viruses and toxic dust penetration into the CNS through the OB. We hypothesize a protective role of neurogenesis in the mature OB. This suggests that normal renewal of cell populations in the OB is an important barrier mechanism protecting the brain from invasion of small amounts of harmful neurotropic agents (ex. viruses and particles of toxic dust), which can cause various neurodegenerative diseases.

Olfactory placodes, that give rise to the olfactory and respiratory epithelia during ontogenesis, are a source of many neurons migrating into forebrain in the direction of growth of the olfactory nerves. The neurons expressing gonadotropin releasing hormone (GnRH) are among the best studied in the population in question. This hormone is responsible for the central regulation of reproduction in adult animals. It was already shown that, in addition to the GnRH-immunoreactive neurons, a small amount of neurons expressing tyrosine hydroxylase (TH), the first enzyme of catecholamine synthesis, migrates into the forebrain. Such a transient population of TH-immunoreactive neurons was shown by means of single and double immmunohistochemical labeling. The TH neurons were first found on branches of the olfactory, terminal, and vomeronasal nerves, along the trajectory of migration of GnRH-immunoreactive neurons on day 15 of embryogenesis, which preceded the appearance of GnRH-immunoreactive neurons. On days 17-21 of embryogenesis, both populations of neurons were found in almost the same areas and on day 21 single neurons contained both GnRH and TH. There were no neurons expressing decarboxylase of aromatic acids (DAA), the second enzyme of catecholamine synthesis, among TH-immunoreactive neurons, thus suggesting noncatecholaminergic nature of these neurons. However, single nonenzymatic DAA-immunoreactive neurons were found in the area of anterior olfactory nuclei in the forebrain, which suggests their involvement in local cooperative synthesis of catecholamines in the area where GnRH-immunoreactive neurons penetrate in the forebrain. Thus, the neurons expressing TH, TH and GnRH, and DAA were found in rats during prenatal period in the nasal part of the head along the nerves projecting into the forebrain and in the rostral part of forebrain. The origin and functional significance of these neurons are discussed.

A mathematical model of the movement of olfactory cilia in different conditions was constructed. The realization of the model includes the development of a mechanical mathematical rheological model of the behavior of a continuous deformable medium, the development of the method of solution adapted to this problem, and obtaining a numerical solution, which takes into account different starting data. The mathematical modeling of the dynamic behavior of a deformable medium was performed using a system of equations for the dynamics of the deformable medium and the solution of the corresponding nonstationary system of equations in partial derivatives.

A computer model of the olfactory bulb was constructed. The paper describes: 1) the general architecture of a model neuron network that reflects the neurophysiological experimental and theoretical data on the structural and functional organization of the peripheral part of the olfactory system, the olfactory bulb with inputs from olfactory receptor neurons; 2) the organization of each of three levels of the model: receptors, olfactory glomeruli, and basic neurons; and 3) a scenario of the computer model work. In some aspects, in particular, in the principle of information presentation, the treatment of the role of basic neurons (mitral and tufted cells), and their interrelations in modules, the model favorably differs from the available olfactory bulb models. The model is basic and provides further refinement of the architecture, an increase in the number of modules, and the modeling of the learning process.

The results of experimentation with the computer model of the olfactory bulb are presented. The architecture and scenario of the work of the model were described previously. The dynamic character of the identification process and the mechanism of memorizing short-term of smell stimuli are described. During the identification, a self-adjustment of the olfactory bulb to incoming signals occurs. The self-modification of mitral and tufted cell synapses enhances responses of the cells; upon subsequent presentation of the stimulus, the olfactory bulb responds with a higher activity. The modeling confirmed the validity of the assumption that the functions of mitral and tufted cells are to identify the components of a complex smell and the image of the smell as the whole.

Formation of structures in the cochlear complex, olfactory bulb and cerebellum has been studied during postnatal development, with special reference to the development of granular cells in the structures mentioned. The granular cells have been demonstrated to be formed rather synchronously and according to a common plan By the time of birth, every structure has a layer of the granular cells formed which is cambial for most of the granular cells in the given structure. It has bene found out that in the cochlear complex, by the time of birth, there is an external layer of the granular cells which is analogous to the external cerebellar granular layer and is connected with it by a cellular cord. Morphological changes in the structure of the granular cells have been described in the process of their differentiation.

Fast prepotentials in the granular interneurons of carp olfactory bulb were recorded by means of intracellular microelectrodes. Data are presented indicating that fast prepotentials are generated in the electrogenic membrane of granular cell dendrites. It is supposed that dendritic spikes play an important role in the inhibitory synaptic influences of granular cells upon secondary olfactory neurons.

The dynamics of DNA synthesis, its ratio to the proliferative activity and changes in the amount of genetic material in mitral cells of the white rat olfactory bulb during prenatal ontogenesis, as well as in newborn rats are considered. The lengthening of G2-phase, decrease of mitotic activity and increase of DNA content in the nuclei of forming mitral cells were shown to occur in the period from 18 till 20 days of embryogenesis.

The lateral olfactory tract nucleus of the cat brain has been studied by means of Nissl's and Golgi's methods. This structure is a cortical formation according to its neuronal organization; four layers can be detected in it (zonal, polymorphic, external and internal, and pyramidal). Among long-axonal neurons, besides cells of the subcortical type (dense-branching, treelike), there are neuronal types specific for the cortex--pyramidal, spindle-like, as well as short-axonal neurons of two types--smooth dendritic and neurons having spinelets. Afferent transmitters are mainly presented by olfactory fibres (it is a rather thick fibre forming at its end not a very dense and long branching brush consisting of terminal fibres with beads along their course).

The cat cerebral olfactory tubercle (OT) includes into its composition certain cellular ensembles distinguished by means of morphological criteria. The ensembles consist of three cellular components: clusters of granular neurons (islands of Calleja), pyramid-like neurons of the layer II, situating along the periphery of the islands, and groupings made by polygonal and spindle-like neurons of the layer III. Morphometrical analysis of every of the three cellular complex components has been carried out. About 7-10 islands of Calleja are situated on the OT territory, which makes nearly 75% of the whole surface of the OT. Neuronal composition of the cellular ensembles has been studied by Golgi method. Varieties of long and short axonal neurons, included into the ensemble composition, have been characterized. Presence of projections of the macrocellular neurons of the layer III (a part of the third component of the ensemble) has been revealed in the posterior part of the lateral hypothalamus and in the field of Forel H1. Possible role of the cellular ensembles is discussed for ensuring various functions of the OT.

Using the intracellular recording technique the electrical activity of the carp olfactory bulb granular cells and secondary neurons was studied. Early and late EPSP and IPSP were the synaptic responses of granular cells both to the olfactory nerve and olfactory tract electrical stimulation. The comparison of responses of the granular cells and secondary neurons led to the conclusion that the granular cells are interneurons producing post-synaptic inhibition of the secondary neurons. It was supposed that the granular neurons are excited by dendro-dendritic and reccurent-collateral pathways and that the inhibitory synapses are located on the dendrites of secondary neurons which are in contact with the olfactory nerve terminals.

Following horseradish peroxidase iontophoretic application into the main olfactory bulb (MOB) retrograde neuronal labeling was examined in the telencephalon in the frog. Labeled neurons, the sources of the MOB afferents are found in the mitral cell layer of the contralateral MOB, pallial and some subpallial areas. Very heavy labeling is observed in the pars ventralis of the lateral pallium, and to a lesser extent in the medial pallium, pars dorsalis of the lateral pallium and in the dorsal pallium. In subpallium labeled neurons are found in the eminentia postolfactoria, the rostral part of the medial septal nucleus, and in the nucleus of the ventro-medial telencephalic wall, which is probably homologous to the nucleus of the diagonal band (Broca) of mammals. No labelled neurons were found in the caudal portion of the MOB granular layer, usually referred to as the anterior olfactory nucleus. The arrangement of the MOB centrifugal innervation in amphibians is discussed in comparison with that in mammals.
