Non-small cell lung cancer (NSCLC) is prevalent worldwide and has a high mortality rate. Even if mesenchymal stem cells (MSCs) are suggested as cancer treatment, the studies of their effects on NSCLC cells contradict each other, mainly due to utilization of two-dimensional (2D) culture system. Three-dimensional (3D) culture systems resemble tissue organization in vivo. Here we comprehensively explore the inhibitory effects of MSCs on NSCLC cells in a 3D culture system. We confirmed that the inhibitory effects of 3D-cultured MSCs (3D-MSCs) on the proliferation and migration of NSCLC cells are greater than that of the 2D-cultured MSCs. 3D-MSCs overexpress IL-24, which serve as the key factor enhancing antitumor effects of MSCs. In these cells, IL-24 affects p38 MAPK and CXCR4/AKT pathways. Overall, this study provides the support for use of MSCs in tumor.

P28/PSMD10, a regulatory complex of the human 26S proteasome, plays a critical role in tumor genesis. This study was designed to clarify the clinical significance of p28GANK in gastric cancer. In order to demonstrate the importance of p28GANK expression for the prognosis of gastric cancer, p28GANK expression in 124 paired cases of gastric cancer and noncancerous regions and immortal gastric epithelial cell GES-1 and 5 human gastric cancer cell lines was analyzed by RT-PCR in real-time. MTT was used to observe the effect of P28GANK on cell growth. P28GANK expression was higher in gastric cancer tissues than in corresponding normal tissues (p = 0.0033), and patients comprising the group with p28GANK high expression had a poorer overall survival rate than those from the low expression group (p = 0.0037). Further, the results of multivariate analysis demonstrated that the high p28GANK expression was an independent prognostic factor of gastric cancer process. P28GANK expression was also up-regulated in five gastric cancer cell lines. As it has been shown by in vitro proliferation assay, p28GANK expression correlated with tumor growth. On the base results of present study one can suggests the p28GANK being useful as a predictive marker for patient prognosis and a novel therapeutic target for gastric cancer.

Natural antisense transcripts (NATs) are widespread in prokaryotes and eukaryotes. They have very important functions in regulating expression of their target genes. However, until now, NATs haven't been systematic identified in high throughput for a lack of effective experimental method. Here, we have developed a strategy based on RNase I protection assay, which permit us to identify NATs in large scale which presented inside Pseudomonas aeruginosa. After being isolated from total RNA of P. aeruginosa strain ATCC 6872 (PAO1), NATs that encoded chromosomally were cloned in a simple and efficient way and a NATs gene library was constructed successfully. 78 random ly selected positive clones were sequenced and analyzed by bioinformatic tools. There are several candidate molecules which are located in PAO1 genome precisely. This study suggests that there may be exist some gene regulatory mechanism acted by NATs in PAO1 and this worth further analysis.
