Possibility to use blood dried on filter paper for serological testing on antibodies to tick-borne encephalitis (TBE). Sensitivity and specificity of specific IgM detection in dry stains of blood by lanthanide fluorescence immunoassay was 94.9% (86.9-100%) and 97.5% (94-100%) respectively, compared with results obtained in tests of sera. Agreement in positive and negative results of tests for IgM against TBE in 562 serum samples and dry blood stains was 95.3%. During analysis of both types of biomaterial high degree of correlation was observed between intensity of fluorescence when testing for both IgM (r=0.86700; p=0.05; n=562), and IgG (r=0.83883; p=0.05; n=337) toTBE virus. Use of this mildly invasive technique of blood draw is reasonable during conduction of large-scale population studies for seroepidemiologic monitoring, investigation of disease outbreaks, control of effectiveness of vaccination against TBE, assessing of level of specific immunity in population of endemic regions, control of treatment, and serologic diagnostics of acute TBE in hospitalized patients, in which blood draw is difficult to perform.

A paper indicator system has been developed in the USSR for the accelerated identification of vibrios and proposed for the identification of enterobacteria. This system consists of paper discs or strips impregnated with definite substrates (carbohydrates, polyatomic alcohols, amino acids, etc.) in combination with an indicator and stabilized with the polymer coating. Two kits for the identification of enterobacteria to the level of the genus (kit A for 9 tests) and to the level of the species (kit B for 25 tests) are presented.

Filter paper discs have been used in the enzyme immunoassay (EIA) as solid phase instead of polystyrene plates. The use of paper discs has made it possible to achieve a multiple increase in the sensitivity of sandwich EIA, thus permitting the detection of Yersinia pestis capsular antigen at a concentration of 0.4 ng/ml. Paper discs can be used not only for the sorption of antigen and antibodies, but also for the affinity purification of preparations.

A total of 5082 cultures belonging to the family Vibrionaceae and 100 strains of enterobacteria were identified by means of the paper indicator systems manufactured by the Gorky Research Institute of Epidemiology and Microbiology and with the use of the classical method. The results thus obtained indicate that these systems proved to be sufficiently sensitive and specific, which allows one to recommend them for wide introduction into laboratory practice.

The method for the determination of the toxigenicity of C. diphtheriae by means of discs impregnated with diphtheria antitoxin is proposed. The discs can be prepared long before use and stored for a year. The determination of toxigenicity with these discs is not inferior in its sensitivity to the routine procedure of the immunoprecipitation test. The disc method decreases the consumption of unavailable diphtheria antitoxin by 10 times, that of culture media by 2-2.5 times: besides, it is less labor consuming.

The possibility of using paper indicator discs with glucose, saccharose, lactose and urea, manufactured at the Gorky Research Institute of Epidemiology and Microbiology, has been studied and the methods for the preparation of indicator discs with fructose, maltose, starch and sodium phenolphthalein-phosphate for the biochemical identification of C. diphtheriae, N. meningitidis and staphylococci have been proposed. The study of 395 C. diphtheriae strains, 98 N. meningitidis strains and 328 staphylococcal strains has shown that paper indicator systems are highly sensitive and specific, which makes it possible to recommend their introduction in laboratory practice for the rapid identification of the above-mentioned organisms.

The authors prepared under experimental-industrial conditions paper indicator systems for the express identification of microbes, including carbohydrate discs (by the method of Nikitin et al.), and newly worked out types for the determination of the activity of cytochromoxidase, urease, indol formation, and indicator amino acid decarboxylases (lysin, ornithin, arginine). The use of paper indicator discs proved to be expedient for rationalization of laboratory diagnosis of bacterial intestinal infections.

For the first time the metabolic processes, taking place during "cold" cultivation of Y. pseudotuberculosis and involving organic and amino acids, were shown to be more economical, thus permitting the bacteria to maintain its viability for a rather long time at low temperature without the need to consume these substances from outside. At higher temperature the bacteria switches its metabolic paths to the consumption of nutrient substances in greater amounts from outside, increasing its constructive metabolism.

The polysaccharide component contained in the slime of the causative agent of melioidosis was obtained. This component was found to be a biopolymer, mainly of the carbohydrate nature, consisting of galactose, glucose, mannose, rhamnose and two unidentified carbohydrates. The slime polysaccharide component contained two thermostable and acid-resistant antigens. The action of alkali led to the loss of one of these antigens. Rabbit antisera to the preparations of the slime polysaccharide component with titers of 1:64 to 1:256, determined in the immunodiffusion test, were obtained. In the precipitation test the slime polysaccharide component reacted with antisera from sick experimental animals, thus confirming the suggestion of its secretion in the process of the development of melioidosis infection.

Physico-chemical parameters and features of macromolecular orgnization of FI-5 phage were studied. This virus was shown to contain a molecule of double-strander DNA with the standard set of nitrous bases (37.8 mol% GC). The molecule of this DNA in situ is characterized by partial disorder of the second structure. Phage virions contain about 47% of DNA and 53% of protein. The genome of the phage is represented by a DNA molecule with molecular weight 65X10(6) daltons and is capable of coding for a least 15 different proteins.

In experiments on Fischer rats (F-344), both with common microflora and germ-free, the influence of the systemic destination of different antibiotics (tetracycline + ampicillin, gentamicin + kefzol, gentamicin, fradizine) on the intestinal microflora, the content of beta-ospartylglycine in feces and the colonization resistance of the intestinal tract to Staphylococcus aureus B-243 and Pseudomonas aeruginosa No. 93 has been studied. The early appearance of beta-aspartyl-glycine in the supernatant of fecal samples has been shown to be the first sign of dysbacteriosis and to indicate the decrease of the colonization resistance of the intestine to opportunistic bacteria.

Possible causes of disturbances of the amino acid pool of the blood serum and the tissue, as well as the presence of an association between these shifts and the changes in the ultrastructure of the striated columnar epithelium of the small intestine were studied. Investigations were conducted in 138 patients suffereing from salmonellosis and in 120 albino rats experimentally-infected with S. typhimurium culture. Free amino acids of the blood serum and the tissue were determined by paper chromatography and on the automatic amino acid analyzer; ultrastructure of the small intestinal mucosa was studied by electron microscopy. The height of salmonella infection was attended by dysproportional changes in the amino acid pool of the tissue and the blood caused by disturbance of absorption and intracellular transformation of amino acids. Ultrastructural changes in the microvilli of the absorbing cells of the small intestine underlied absorption disturbances: at the height of salmonella infection some of the microvilli of the striated columnar small intestine proved to show poor contours; reduction of mitochondrial cristae was observed in almost all the mitochondria.

It was shown that Y. pseudotuberculosis strains causing the Far-Eastern scarlatina-like fever in the Primorsk region belonged to subtype IB-ipopolysaccharides of the standard strain of subtype IB and of the local strain were closely affiliated by the analytic data and monosaccharide composition, but differed from the lipopolysaccharide of strains belonging to subtype IA. Living vaccines should be used to obtain the sera against the subtypes IA and IB of the causative agent.

The virologists' attention to bats (Сhiroptera) changed in the late 20th century as the concept of emerging infections grew in popularity. Since the beginning of the COVID-19 pandemic, the number of publications on bat viruses has increased profoundly.History of the problem; biodiversity of Chiroptera and related viruses; medical and veterinary significance of some viral genera and subgenera (Lyssavirus, Henipavirus, Marburgvirus, Ebolavirus, Sarbecovirus, Merbecovirus), as well as problems of bat protection, are addressed in a concise form. Literature search was carried out in electronic databases, mainly for the period of 2000-2021. Publications in Russian that are poorly represented in English-language reviews are also included. The purpose of the review is to substantiate the importance of an interdisciplinary approach in the context of increased interest in the study of viral infections in bats. This review was written for researchers who have not previously dealt with this problem.Since the beginning of this century, the number of known virus species associated with bats has increased by an order of magnitude (>200). The families Rhabdoviridae, Coronaviridae, Paramyxoviridae are in the first ranks according to the number of findings, and the highest diversity of viruses has been established for the families Vespertilionidae, Pteropodidae, Molossidae. Interdisciplinary cooperation positively influences the efficiency, biological safety and practical significance of the ongoing research. The best results were achieved by multidisciplinary teams with good cross-training in several specialties. Many papers emphasize the need to balance health and conservation interests.The analysis of scientific publications indicates a change in research approaches in this area: from collecting individual facts within the framework of narrow specialties to a comprehensive assessment of new knowledge from ecological, evolutionary and socio-economic positions. Results of the research emphasize the need to maintain complex approaches addressing public health needs and environmental protection. The importance of bat-borne viral infections determines the necessity for correction and interdepartmental coordination of scientific research and surveillance of wildlife zoonoses in the Russian Federation.

Due to the new coronavirus infection pandemic, the global scientific community has been forced to change the direction of the most research, focusing on vaccine development as well as the search for new antiviral drugs to treat COVID-19. The choice of experimental models, timeframe and approaches for evaluating drugs and vaccines under development is crucial for the development of effective measures to prevent and control this disease.The purpose of this review was to summarize the relevant data concerning the susceptibility of laboratory animals to SARS-CoV-2. This paper describes the most virus-susceptible animal species that can be used to reproduce coronavirus infection, stressing the main advantages and disadvantages of each of them.According to the latest data, small rodents (Rodentia) and non-human primates (Strepsirrhini) are commonly used in the scientific community to model coronavirus infection. The viral load in the upper and lower parts of the respiratory system, clinical symptoms of infection (weight loss, body temperature and general health status), pathomorphological picture in target organs and the production of antibodies after infection are considered to the main markers of pathology. Despite the vast amount of data, none of the described models of SARS-CoV-2 infection may be considered a gold standard, since they do not reproduce all spectrum of morphological and pathogenetic mechanisms of infection, and do not fully reflect the clinical picture observed in patients in human population.Based on the analyzed literature data, we suppose that Syrian hamster (Mesocricetus auratus) and mice (Muridae) expressing the angiotensin converting enzyme receptor 2 (ACE2) are the most suitable animal species for their use in experiments with SARS-CoV-2 infection. The development of neutralizing antibodies makes it possible to evaluate the efficacy of vaccines, while the course and severity of symptoms infection makes the use of mice and hamsters especially popular for screening pharmacological substances with antiviral mechanism of action, when their administration can prevent or slow the disease progression.

Introduction: Currently, new directions in cancer therapy are actively developing, one of which is oncolytic immunotherapy. This approach would be to use of viruses as cancer specific cytolytic agents capable of stimulating both the tumor-specific and non-specific immune response. The objective paper was obtain a recombinant vaccinia virus containing genes encoding immunostimulating molecules and study oncolytic and immunostimulating properties of recombinant virus., Material and Methods: MTT test, ELISA, methods of transient dominant selection., Results: The recombinant vaccinia virus (L-IVP_oncoB) were obtained with deletion of the gene encoding thymidine kinase and had an integrated gene encoding GM-CSF. Also the virus have deletion of the gene encoding viral growth factor and integrated genes encoding synthetic tumor-specific polyepitopic immunogens. It was shown that the modifications made to the viral genome did not affect the growth characteristics of the virus when cultured on CV-1 and 4647 cell cultures, and the cytopathogenic efficacy of the virus was determined in relation to cancer cultures of cells of various genesis. In in vivo experiment, it was revealed that the polyepitopic construct in the genome L-IVP_oncoB is able to initiate a change in the profile of cytokines., Discussion: The obtained data characterized L-IVP_oncoB as a promising cytopathogenic and immunostimulating agent and showed the need for further study of its properties as means of oncolytic immunotherapy., Conclusion: The basic experiments on the evaluation of the biological properties of the obtained L-IVP_oncoB, which are necessary for the characterization of the oncolytic virus, have been carried out., Competing Interests: The authors declare no conflict of interest.

Lujo hemorrhagic fever (LHF) is a viral disease accompanied with fever, headache, vomiting, diarrhea, arthralgia, myalgia and numerous signs of hemorrhagic syndrome. LHF causes a clinical syndrome remarkably similar to Lassa hemorrhagic fever. The first case of LHF occurred in Johannesburg, South Africa, in 2008. There was a secondary transmission from the index patient to four healthcare workers. Four of the five patients died. The etiologic agent of LHF is Lujo virus (LUJV) belonging to Arenavirus genus of the Arenaviridae Family. Virus Lujo is the second pathogenic arenavirus, after Lassa virus, to be recognized in Africa during the last 40 years. Data about epidemiology, clinical characteristics and diagnostics of LHF, properties of Lujo virus (according to phylogenetic analysis), and recommended precautions for preventing secondary transmission are considered in this paper., Competing Interests: The authors declare no conflict of interest.

Pathogenicity factors of Corynebacterium non diphtheriae - pili, microcapsule, cell wall, pathogenicity enzymes, toxins, that determine the ability of microorganisms to consequentially interact with epithelium of entry gates of the organism, replicate in vivo, overcome cell and hu- moral mechanisms of protection, are examined in the review. Particular attention in the paper is given to species of non-diphtheria corynebacteria, that are pathogenic for human and able to produce toxins - Corynebacterium ulcerans and Corynebacterium pseudotuberculosis. Mechanisms of expression regulation of PLD-exotoxins, its interaction with immune system cells are described.

Five influenza pandemics emerged in the 20th and 21st centuries. Data regarding possibility of infection of swine with human influenza viruses-and persistent circulation of these strains among swine with subsequent infection of humans with these viruses were obtained in the recent years. A possibility of prediction of influenza pandemics by constant observation and study of influenza viruses circulating among swine is discussed in the paper.

Papillomaviruses are a large and diverse group of viruses. It includes approximately 200 fully described types that have been detected in humans. Human papilloma viruses (HPV) are etiologic agents during various, benign and malignant lesions of mucous membrane and skin epithelium. Very importantly, persistent HPV infection of certain types is a leading cause of carcinoma of uterine cervix, penis, vulva; vagina, anal canal and fauces (including tongue base and tonsils). HPV infection prophylaxis is the best means to control HPV-conditioned diseases, and vaccination, as had been demonstrated, --the most effective method of its prophylaxis. In this paper principle characteristics and clinical manifestations of papillomavirus infection, as well as effectiveness of vaccination against HPV are examined.

Systemized data on epidemiology, pathogenesis, clinical manifestation, diagnostics and therapy of VZV-vasculopathy--a disease, occurring due to damage of arteries of the central nervous system by Varicella Zoster virus, are presented in the review. A special attention in the paper is given to the effect of vaccine prophylaxis of chicken pox and herpes zoster on the frequency of development and course of VZV-vasculopathy.

This paper considers the experience of genotyping and sequencing technologies in laboratories of specialized anti-epidemic team (SAET) during the XXII Olympic Winter Games and XI Paralympic Winter Games of 2014 in Sochi. The work carried out during the pre-Olympic period on performance of readiness by SAET for these studies is analyzed. The results of genotyping strains of pathogens during the Olympic Games are presented. A conclusion about the effectiveness of the use of molecular genetic techniques in terms of SAET is made.
