O problema da pesquisa surge da experi?ncia pastoral como presb?tero na diocese de Inhambane, em Mo?ambique. O tema engloba de modo transversal as ?reas da Religi?o Tradicional Africana (RTA) e da teologia cat?lica na dimens?o lit?rgico-pastoral do Sacramento da Ordem; concretamente no grau presbiteral. O estudo apresenta a rela??o entre o ?sacerdote tradicional? dos vatshwa1 e o presb?tero cat?lico no concernente ?s fun??es exercidas na ora??o pelos defuntos, pelas duas figuras em destaque no presente trabalho. Aborda-se, brevemente, a historiografia eclesiol?gica de Mo?ambique, desde a primeira presen?a dos mission?rios at? a fase atual. Em seguida, faz-se a descri??o das fun??es do ?sacerdote tradicional? no rito das ex?quias no contexto dos vatshwa. Ali?s, ? fundamental que se tenha presente que este sacerdote faz parte do cl?; n?o ? um desconhecido. A sua ascens?o a esta categoria n?o ? por voca??o, mas por direito ou por indica??o dos antepassados. Descreve-se, outrossim, o minist?rio presbiteral na sua extens?o e depois se focaliza o mesmo como homem chamado a rezar pelos defuntos. Assim, a pesquisa mostra semelhan?as e diferen?as entre o ?sacerdote tradicional? e o presb?tero cat?lico. Nesta pesquisa concluiu-se que os problemas pastorais relativos ? ora??o pelos defuntos devem-se, em grande parte, ?s falhas da primeira evangeliza??o no que diz respeito ? compreens?o da cultura africana por parte dos mission?rios cat?licos. The issue of this research arises from the pastoral experience as a priest in the diocese of Inhambane, Mozambique. The theme encompasses, in a transversal way, the areas of African Traditional Religions (ATR) and Catholic theology in the liturgical-pastoral dimension of the Sacrament of Order; specifically in the presbyteral degree. The study presents the relationship between the ?traditional priest? of the vatshwa and the Catholic priest regarding the functions performed in the prayer for the dead, by the two characters highlighted in the present work. The ecclesiological historiography of Mozambique is briefly discussed, from the first initial presence of the missionaries to the current phase. Next, there is a description of the functions of the ?traditional priest? in the rite of the funeral in the context of the vatshwa. In fact, it is essential to keep in mind that this priest is part of the clan; it is not an unknown person. His ascension to this category is not by vocation, but by right or by indication of ancestors. The priestly ministry is also described in its scope and then focused on a man called to pray for the dead. Thus, the research shows similarities and differences between the ?traditional priest? and the Catholic priest. In this research it was concluded that the pastoral problems related to prayer for the dead are largely due to the failures of the first evangelization regarding the understanding of African culture on the part of catholic missionaries. Ricinus communis, a plant known in Brazil as castor bean, has gained attention from industry, media and governments, due to the properties of its derivatives, especially ricin and ricinoleic oil. The latter corresponds to 90% of the total oil that is extracted from the seed of the fruit and presents an important saving for the chemical industry. Already ricin is a potent toxin capable of efficiently inactivating eukaryotic ribosomes. It consists of two subunits, RTA and RTB. Functionally, RTB is responsible for the recognition and internalization of ricin in the target cells and RTA for the catalysis of the depurination of adenine 4324 located in the Sarcin-Ricin Loop region of rRNA 28S. Due to its cytotoxicity, ease of extraction and purification, ricin is classified as a category B biological threat agent according to the Center for Disease Control and Prevention (CDC), worrying world authorities for its potential use as a biological weapon. Thus, the development of an antitoxin would be an important pharmaceutical and biotechnological achievement. However, the great difficulty in designing inhibitors capable of acting efficiently as RTA blockers is attributed to the incomplete understanding of its reaction mechanism. In order to obtain further clarification about the mechanism of action of RTA, a theoretical study was carried out in this dissertation using the Molecular Dynamics (DM) to monitor the interaction of the RTA catalytic subunit with a natural substrate analog. Quantum Chemistry calculations were also performed using Reactivity Descriptors (DR) based on the Functional Theory of Conceptual Density (CDFT). In short, the DM trajectory revealed the main hydrogen bonds present in the active site of the RTA between the Val81, Arg180 and Gly121 residues with the target adenine of the mimic ligand. In addition, it evidenced the hydrogen bond that activates the target adenine throughout the simulation. The attractive interactions of adenine stabilization by residues Tyr80, Tyr123, Val81, Gly121 and Glu208 and repulsive interactions by residues Glu177 and Trp211, were identified by the study of the interaction energy. Residue 332 was also recognized as catalytic water in the mechanism. Finally, the reactivity descriptors pointed to an RTA mechanism of action where there is a susceptibility to electrophilic attack on the target adenine through the Arg180 residue, a stabilization of the adenine in a π stack between the Tyr80 and Tyr123 residues and a Glu177 propensity to activate catalytic water by donating electron density and receiving a proton. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES A Ricinus communis, planta conhecida no Brasil como mamoneira, tem ganhado atenção da indústria, mídia e governos, em razão das propriedades de seus derivados, em especial a ricina e o óleo ricinoleico. Esse último, corresponde 90% do óleo total que é extraído da semente do fruto e apresenta uma importante economia para a indústria química. Já a ricina, é uma potente toxina capaz de inativar de forma eficiente ribossomos eucarióticos. É constituída de duas subunidades, a RTA e a RTB. Funcionalmente a RTB é responsável pelo reconhecimento e internalização da ricina nas células-alvo e a RTA pela catálise da depurinação da adenina 4324 localizada na região Sarcin-Ricin Loop do rRNA 28S. Devido a sua citotoxicidade, facilidade de extração e purificação, a ricina é classificada como agente de ameaça biológica de categoria B segundo o Centro de Controle e Prevenção de Doença (CDC), preocupando autoridades mundiais pelo seu potencial uso como arma biológica. Dessa forma, o desenvolvimento de uma antitoxina seria uma importante conquista farmacêutica e biotecnológica. Porém, a grande dificuldade em projetar inibidores capazes de agir eficientemente como bloqueadores da RTA se atribui a compreensão incompleta do seu mecanismo de reação. Com a finalidade de obter um maior esclarecimento acerca do mecanismo de ação da RTA, foi realizado neste trabalho de dissertação um estudo teórico utilizando a Dinâmica Molecular (DM) para o monitoramento da interação da subunidade catalítica RTA com um análogo do substrato natural. Também foram realizados cálculos de Química Quântica por meio de Descritores de Reatividades (DR) baseados na Teoria Funcional da Densidade Conceitual (CDFT). Em suma, a trajetória de DM revelou as principais ligações de hidrogênio presentes no sítio ativo da RTA entre os resíduos Val81, Arg180 e Gly121 com a adenina alvo do ligante mímico. Além disso, evidenciou a ligação de hidrogênio que ativa a adenina alvo ao longo de toda simulação. As interações atrativas de estabilização da adenina pelos resíduos Tyr80, Tyr123, Val81, Gly121 e Glu208 e interações repulsivas pelos resíduos Glu177 e Trp211, foram identificadas pelo estudo da energia de interação. Foi reconhecido ainda o resíduo 332 como água catalítica do mecanismo. Por fim, os descritores de reatividade apontaram para um mecanismo de ação da RTA onde há uma susceptibilidade ao ataque eletrofílico na adenina alvo por meio do resíduo Arg180, uma estabilização da adenina em um empilhamento π entre os resíduos Tyr80 e Tyr123 e uma propensão do Glu177 em ativar a água catalítica doando densidade de elétrons e recebendo um próton.

O presente estudo pretende salientar o trânsito das grandes formulações da concepção da temporalidade na tradição hindu: do tempo (kāla) à sua compreensão como instantâneo (kṣaṇa). Num ponto preliminar, salientam-se as condições linguísticas do pensar indo-europeu e a "gramática" sanscrítica a propósito do tempo. Sublinham-se de seguida os enquadramentos rituais e míticos, tanto ligados com a ciclicidade (saṃsāra), como com o imutável e eterno (ānantya). Depois percorrem-se, de forma sintética, os vários "sistemas" filosóficos salientando respectivamente: a concepção metafísica (no Vedānta e Mīmānsā), a recorrência e irrealidade já num plano gnosiológico (no Sāṃkhya e no Yoga), e a formulação psicológica (tanto no Vaiśeṣika, como no Nyāya, além disso presente no Jainismo). Enfim, na última parte, faz-se menção aos principais traços característicos da doutrina budista da instantaneidade (muito característica sobretudo das escolas “subitistas”). Valoriza-se conclusivamente esta instância de um "agora" também como lição da espiritualidade hindu em geral. The present study intends to point out the conveyance of the great formulations in Hindu tradition of temporality's conception: from time (kāla) to its understanding as momentary (kṣaṇa). In a preliminary point, we enhance linguistic conditions of Indo-European thought and sanskritic "grammar" with regard to time. Then, we underline ritual and mythical framings, linked to cyclicity (saṃsāra), as well as to the unchangeable and eternal (ānantya). Afterwards, in a synthetic way, we go through different philosophical "systems", pointing out, respectively: the metaphysical conception (in Vedānta and Mīmānsā), the recurrence and unreality already in a gnoseological levei (in Sāṃkhya and in Yoga), and the psychological formulation (as well as in Vaiśeṣika, as in Nyāya, and besides present in Jainism). Finally, in the last part, we make mention to the principal aspects characteristics of Buddhist doctrine of momentariness (very characteristic of `subitists' schools). Conclusively, we value this instancy of the "now", also as a lesson of Hindu spirituality in general.

Mestrado em Engenharia Física Os óxidos de metais, como é o caso dos óxidos de cálcio e cobalto, são materiais de elevado interesse tecnológico atendendo às suas propriedades termoelétricas, o que poderá favorecer a sua utilização como conversor de energia térmica em elétrica em aplicações futuras. Parte dos esforços para aperfeiçoar as suas propriedades termoelétricas têm passado pela modificação da composição do material ou, em alternativa, a alteração da microestrutura do material policristalino. Com o objetivo de otimizar as propriedades termoelétricas do óxido de cálcio cobalto (Ca3Co4O9) foram aplicadas neste projeto duas técnicas de síntese distintas: método de Pechini modificado e síntese por reação de estado sólido com ativação mecânica por moagem de elevada energia. Além disso, foram ainda aplicadas diferentes técnicas de sinterização, com variadas taxas de aquecimento, tendo por objetivo analisar a sua influência na evolução da microestrutura das amostras em estudo e respetivas consequências ao nível das propriedades térmicas e elétricas. A aplicação do método de Pechini resultou na redução da temperatura de síntese, quando comparado ao método de moagem de elevada energia. A temperatura escolhida para sintetizar o pó dos precursores foi a de 800 ºC, em ambos os métodos de síntese. Para sinterizar as pastilhas precursoras, foram utilizados como equipamentos um forno de microondas convencional adaptado para o controle da temperatura de aquecimento, o forno convencional e o forno de recozimento rápido (RTA, Rapid Thermal Anealing). As pastilhas sinterizadas por microondas foram aquelas em que se obteve maior densificação, alcançando mais de 80 % do valor ideal, e melhores propriedades termoelétricas, tal como a condutividade térmica reduzida de 1.3 W.m-1.K-1 à temperatura ambiente. Com a exceção da sinterização por forno de RTA, todas as técnicas de sinterização usadas permitiram, quando aplicadas as condições de preparação adequadas, a obtenção de pastilhas com uma única fase. As propriedades estruturais dos pós e pastilhas foram analisadas por difração de raios-X, microscopia ótica, análise térmica diferencial e método de Arquimedes, de modo a averiguar as melhores condições de síntese/sinterização de Ca3Co4O9. A condutividade elétrica e o coeficiente de Seebeck foram registadas num mesmo aparato de sonda de quatro pontos. A condutividade térmica foi medida através do equipamento Hot Disk TPS2500 à temperatura ambiente. Metal oxides like the calcium and cobalt oxide have attracted the interest of a lot of researchers due to their thermoelectric properties,. There are two preferable ways to improve the cobaltite performance: with the modification of its composition or microstructure. Within this project distinct methods were applied to the synthesis and sintering of the compost. At first, the powder necessary to make the pellets was prepared by the modified Pechini method and alternatively by the high energy ball-milling method. In the first case, the temperature of synthesis of the compound Ca3Co4O9 was reduced but in both cases, it was deduced that the best temperature of synthesis was at 800 ºC. Through the first method it was possible to archive lower calcining temperatures when compared to the second method. For the sintering of these pellets a customized kitchen microwave oven with heating control was used along with a fast heating oven (RTA, Rapid Thermal Annealing) and a conventional oven. The pellets prepared with the microwave oven had the highest density, surpassing 80% of the ideal density. They also presented good thermoelectric properties, such as thermal conductivity as low as 1.3 W.m-1.K-1 at room temperature. Through these sintering techniques, the archive of single phase pellets was possible when using the right conditions, with the exception of the sintering by RTA furnace. The structural characterization of these materials was performed using techniques such as scanning electron microscopy, differential thermal analysis and X-Ray diffraction. From these analysis it was possible to determine the best preparing conditions. The electrical conductivity and Seebeck coefficient were measured with a four probe equipment from room temperature till 700ºC, the thermal conductivity was studied with a Hot Disk TPS2500 equipment at lower temperatures.
