No. of bitstreams: 1 dissertacao_final_felipe_andres.pdf: 613014 bytes, checksum: 1040ec3d54249c0b8fc0ef85581966bd (MD5) Previous issue date: 2018-02-27 Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES The phytonematoids of the genus Meloidogyne present a great agricultural importance, because they decrease from 30 to 50% of the productivity of a wide range of hosts.The objective of this search was to evaluate the effect of the essential oils of oregano (Origanum vulgare), eucalyptus (Eucalyptus globolus), aniseed (Pimpinella anisum) and depleted mushroom substrate, Agaricus bisporus, in the vitro and greenhouse control, in the population of Meloidogyne javanica in tomato plants (Solanum lycopersicum) cv. Saint Clara.To verify the in vitro effect, 1 mL of suspension containing 100 eggs of M. javanica to hatching and 50 J2 to mobility were deposited in glass tubes with 1 mL of the treatments (oregano essential oil, anise and eucalyptus at a concentration of 0.3%), abamectina (Vertimec®, 2,5 mL L-1), evidence (water), Paecilomyces lilacinus (Nemat®, 3g L-1), Bacteria Pseudomonas frederiksbergensis (PSF) and Stenotrophomonas rizophila (STR) (1x108 UFC) and filtrate of the depleted substrate (FSE) 1:5 (p:v). Both experiments were performed in a completely randomized design (DIC) with nine treatments and five replicates. To determine the presence of the volatile substances in the depleted substrate of Agaricus bisporus, two-part plates were used, on one side of the plate 2 ml of the suspension containing 100 eggs was deposited on the other side of the plate, wetted depleted substrate was deposited at the field capacity, and the plate was subsequently closed in the dark at 25 °C for 15 days. The experiment was done in DIC with four treatments, evidence (water) and depleted substrate: solo in relationships 1:1, 1:2, 1:3 (v:v) and five replications. The essential oils were submitted to gas chromatographic analysis and the depleted substrate of A. bisporus was subjected to chemical analysis of micro and macronutrients and isolation of microorganisms. From this substrate were isolated bacterias that were identified by DNA analysis. In greenhouse 20-day tomato seedlings were inoculated with 5 mL of suspension containing 4000 nematode eggs. After 24 hours of inoculation the treatments were applied, evidence (water), oregano oil, anise and eucalyptus in the concentration of 0,3%, P. lilacinus, FSE and abamectina, keeping the vessels moistened at field capacity for 75 days . The anise essential oil and chemical control (abamectina) suppressed the population by more than 90% while the control and other oils reduced by 50%. The experiment was carried out in a randomized complete block design (DBC) with seven treatments and five replications. A second experiment was performed by mixing the depleted substrate of Agaricus bisporus with soil in three proportions 1:1, 1:2 and 1:3 (v:v). After mixing, 5 ml of suspension containing 4000 eggs per vessel was put and covered with black plastic for 15 days. At the end of the 15 days the pots were uncovered and transplanted the tomato seedlings cv. Santa Clara with 20 days of age, keeping the vessels moistened to field capacity for 75 days. The experiment was performed in DBC with four treatments and five replicates. All depleted substrate mixtures decreased more than 90% of the M. javanica population, differing statistically from the control by the Scoktt Knott test at 95% reliability. Os fitonematoides do gênero Meloidogyne apresentam grande importância agrícola, pois podem diminuir de 30 a 50% da produtividade em ampla gama de hospedeiros. O objetivo deste trabalho foi avaliar o efeito dos óleos essenciais de orégano (Origanum vulgare), eucalipto (Eucalyptus globolus), anis (Pimpinella anisum) e o substrato exaurido de cogumelo, Agaricus bisporus, no controle in vitro e em casa de vegetação, da população de Meloidogyne javanica em tomateiro (Solanum lycopersicum) cv. Santa Clara. Para verificação do efeito in vitro, depositou-se 1 mL de suspensão contendo 100 ovos de M. javanica para eclosão e 50 J2 para mobilidade, em tubos de vidro com 1 mL dos tratamentos (óleo essencial de orégano, anis e eucalipto na concentração de 0,3%), abamectina (Vertimec ®, 2,5 mL L-1), testemunha (água), Paecilomyces lilacinus (Nemat®, 3 g L-1), bactérias Pseudomonas frederiksbergensis (PSF) e Stenotrophomonas rizophila (STR) (1x108 UFC) e filtrado do substrato exaurido (FSE) 1:5 (p:v). Ambos os experimentos foram realizados em delineamento inteiramente casualizado (DIC) com nove tratamentos e cinco repetições. Para determinar a presença de substâncias voláteis no substrato exaurido de Agaricus bisporus, utilizaram-se placas bipartidas, e, de um lado da placa depositou-se 2 mL da suspensão contendo 100 ovos, e do outro lado substrato exaurido umedecido à capacidade de campo fechando-se a placa deixando-as no escuro a 25 °C por 15 dias. O experimento foi feito em DIC com quatro tratamentos, testemunha (água) e substrato exaurido:solo nas relações 1:1, 1:2, 1:3 (v:v) e cinco repetições. Os óleos essenciais foram submetidos à análise cromatográfica de gases e o substrato exaurido de A. bisporus foi submetido à análise química de micro e macronutrientes e isolamento de microrganismos. Deste substrato foram isoladas bactérias que foram identificadas pela análise de DNA. Em casa de vegetação inocularam-se as mudas de tomate de 20 dias com 5 mL de suspensão contendo 4000 ovos do nematoide. Após 24 horas da inoculação aplicaram-se os tratamentos, testemunha (água), óleo de orégano, anis e eucalipto na concentração de 0,3%, P. lilacinus, FSE e abamectina, mantendo os vasos umedecidos a capacidade de campo durante 75 dias. O óleo essencial de anis e o controle químico (abamectina) suprimiram em mais de 90% a população do nematoide, enquanto a testemunha e os demais óleos reduziram cerca de 50%. Os compostos majoritários identificados no óleo essencial de orégano foram thymol e cavracol, para o óleo essencial de anis foi o estragol, e o composto majoritário para o óleo essencial de eucalipto foi o eucaliptol. O experimento em casa de vegetação com óleos essenciais foi realizado em delineamento em blocos casualizados (DBC,) com sete tratamentos e cinco repetições. Realizou-se um segundo experimento, misturando-se substrato exaurido de Agaricus bisporus com solo em três proporções 1:1, 1:2 e 1:3 (v:v). Após realizar a mistura, adicionou-se 5 mL de suspensão contendo 4000 ovos por vaso e deixou-os tampados com plástico preto por 15 dias em sala com ambiente controlado a 24° C. Finalizando os 15 dias foram destampados os vasos e transplantadas as mudas de tomateiro cv. Santa Cara com 20 dias de idade, mantendo os vasos umedecidos a capacidade de campo durante 75 dias. O experimento foi realizado em DBC com quatro tratamentos e cinco repetições. Todas as misturas com substrato exaurido diminuíram em mais de 90% a população de M. javanica e influenciaram positivamente no desenvolvimento das plantas de tomateiro, diferenciando-se estatisticamente da testemunha pelo teste de Scoktt Knott a 95% de probabilidade.

Cereus jamacaru, xerofilous plant used as eatable and ornamental farraginous, show higher distribution among plants in Brazilian Northeast. Considering hard studies with sanity and seeds treatment with alternative products in native species, the objective of this work was to determine pathogens incidence and control of those using essentials oils with fungistatic potential, besides the observation of the effect of those products about the germination of the seeds. Seeds of C. jamacaru (mandacaru) were collected in Areia, in the Brejo Paraibano. Pathogens incidence were evaluated by “Blotter test”, accomplished without previous disinfection of the seeds, to the ambient temperature, for seven days (control), which was compared with the seeds submitted to different concentrations of anise oil (Pimpinella anisum) and of citronela (Cymbopogon winterianus), composing the following treatments: 1% (T1), 1,5% (T2), 2,0% (T3) and 2,5% (T4). It was used experimental design random entirely, with five treatments and ten repetitions. In the control treatment the fungus incidence: Aspergillus sp. (0,5%), Penicillium sp. (0,5%), Cladosporium sp. (13,5%), Curvularia sp. (3%), Nigrospora sp. (1%) and Rhizophus sp. (1,5%). The results showed that the growth of Cladosporium sp., Curvularia sp. and Nigrospora sp. were controlled with P. anisum oil in all the used concentrations, however, the essential oil of C. winterianus just controlled Cladosporium sp. and Nigrospora sp. Inside of the tested conditions, larger efficiency of the P. anisum oil was verified when compared with the one of C. winterianus in the control of the fungus mentioned. The essential oils products increased the germination of the seeds, when compared to the control., Cereus jamacaru, xerófila nativa utilizada como forrageira comestível e ornamental, apresenta ampla distribuição na vegetação nordestina. Tendo em vista o reduzido número de estudos da sanidade e tratamento de sementes de espécies nativas com produtos alternativos, o objetivo deste trabalho foi determinar a incidência de patógenos e detectar o controle dos mesmos, à base de óleos essenciais, além da observação do efeito desses produtos sobre a germinação das sementes. As sementes foram coletadas no município de Areia, Brejo Paraibano. A incidência de patógenos foi avaliada pelo “Blotter test”, realizado sem desinfecção prévia das sementes, à temperatura ambiente, durante sete dias (controle), a qual foi comparada com as sementes submetidas a diferentes concentrações de óleo de erva-doce (Pimpinella anisum) e de citronela (Cymbopogon winterianus), compondo os seguintes tratamentos: 1% (T1), 1,5% (T2), 2,0% (T3) e 2,5% (T4). Empregou-se delineamento experimental inteiramente casualizado, com cinco tratamentos e dez repetições. No tratamento controle, incidiram os fungos Aspergillus sp. (0,5%), Penicillium sp. (0,5%), Cladosporium sp. (13,5%), Curvularia sp. (3%), Nigrospora sp. (1%) e Rhizophus sp. (1,5%). Os resultados mostraram que o crescimento de Cladosporium sp., Curvularia sp. e Nigrospora sp. foram controlados com óleo de P. anisum em todas as concentrações utilizadas, porém o óleo essencial de C. winterianus controlou apenas Cladosporium sp. e Nigrospora sp. Dentro das condições testadas, constatou-se maior eficiência do óleo essencial de P. anisum quando comparado com o de C. winterianus no controle dos fungos citados. Os óleos essenciais empregados aumentaram a germinação das sementes, quando comparadas à testemunha.

The objective of that work was to evaluate the effects of light, of temperature and of hydric stress in the germination and vitality of seeds of anise, fennel and dill. The experiment was led in two stages. In stage I, the seeds were placed to germinate to constant temperatures of 20, 25, 30 ºC and alternate temperatures of 20-30 ºC in the presence and absence of light. The assessed parameters were: germination percentage, first count, index of germination speed, length and dry mass of seedlings. In stage II, the seeds were placed in soaked substratum in polyethylene glicol (PEG 6000) in the osmotic potentials equivalent to zero; -0,05; -0,10; -0,15; - 0,20; -0,25; -0,30 MPa. Germination percentage, first count and index of germination speed were evaluated. In stage I, the procedure employed was reached by chance, with the data submitted to variance analysis and to the Tukey test to a 5% of probability, and in stage II, analysis of polinomial regression was used. According to the results, it is possible to conclude that germination of seeds anise, fennel and dill happen in presence and absence light, however the manifestation of the vigor is favored by light. The germination of dill seeds is larger in light presence. The seeds anise, fennel and dill, without dormancy, germinate better in the constant temperatures of 20 and 25 °C and temperature of 30 °C is not adapted for germination test in these species. The dill seeds are more tolerant to hydric stress than the anise and fennel seeds. The decrease of the osmotic potentials reduces the germination and vigor of seeds anise, fennel and dill, being the most affected vigor than germination. The germination is reduced drastically, starting from -0,1 MPa for fennel and -0,2 MPa for dill, when there is decrease of osmotic potentials with polyethylene glicol 6000. O objetivo desse trabalho foi avaliar os efeitos da luz, da temperatura e do estresse hídrico na germinação e no vigor de sementes de anis, funcho e endro. O experimento foi conduzido em duas etapas. Na etapa I, as sementes foram colocadas para germinar nas temperaturas constantes de 20, 25, 30 ºC e alternada de 20-30 ºC na presença e ausência de luz. Os parâmetros avaliados foram: percentagem de germinação, primeira contagem, índice de velocidade de germinação, comprimento e massa seca das plântulas. Na etapa II, as sementes foram colocadas em substrato embebido em solução de polietileno glicol (PEG 6000) nos potenciais osmóticos correspondentes a zero; -0,05; -0,10; -0,15; -0,20; -0,25; -0,30 MPa. Avaliou-se a percentagem de germinação, primeira contagem e índice de velocidade de germinação. O delineamento utilizado foi o inteiramente casualizado com os dados submetidos à análise de variância e teste de Tukey a 5% de probabilidade, na etapa I e análise de regressão polinomial, na etapa II. Conforme os resultados pode-se concluir que a germinação das sementes de anis, funcho e endro ocorre tanto na presença quanto na ausência de luz, porém a manifestação do vigor é favorecida pela luz. A germinação das sementes de endro é maior na presença de luz. As sementes de anis, funcho e endro, sem dormência, germinam melhor nas temperaturas constantes de 20 e 25 °C e a temperatura de 30 °C não é adequada para o teste de germinação nestas espécies. As sementes de endro são mais tolerantes ao estresse hídrico que as sementes de anis e funcho. A diminuição dos potenciais osmóticos reduz a germinação e o vigor das sementes de anis, funcho e endro, sendo o vigor mais afetado que a germinação. A germinação é reduzida drasticamente, a partir de -0,1 MPa para funcho e -0,2 MPa para endro, quando há diminuição dos potenciais osmóticos com polietileno glicol 6000.

Hyadaphis foeniculi (Passerini, 1860) (Hemiptera: Aphididae) tem sido observado causando danos à cultura de erva-doce, Pimpinella anisum L., na região do agreste meridional de Pernambuco. Este é o primeiro registro da espécie no Estado. Hyadaphis foeniculi (Passerini, 1860) (Hemiptera: Aphididae) was observed causing damage on anise crop, Pimpinella anisum L., in the Meridional "Agreste" region of Pernambuco State, Brazil. This is the first report of this species in the region.
