Cattle tick fever and bovine trypanosomosis are diseases that negatively impact cattle production in Brazil, causing considerably productive losses and animals death. In this study, the objective was to investigate the presence of the four hemoparasites that mostly affect dairy herds (A. marginale, B. bovis, B. bigemina and T. vivax) using the qPCR technique in discarded dairy cows in the northwestern region of Rio Grande do Sul. For this purpose, brain and spleen tissue biopsies were collected at a local slaughterhouse from 64 dairy cattle from this region. DNA extraction from the samples was performed using the Brazol® reagent, their quantity and purity were obtained through spectrophotometer analysis with NanoDropTM. In the brain samples, DNA detection was performed for T. vivax and B. bovis, and in the spleen, DNA of the four agents was verified. From a total of 64 brain samples analyzed, 14.06 % were positive for Babesia bovis. For spleen samples, Babesia bovis was detected in 17.19 % of the samples, while 29.69 % tested positive for B. bigemina and 37.50 % for Anaplasma marginale. Regarding Trypanosoma vivax, none of the analyzed samples resulted in a positive diagnosis in both tissues. Thus, the findings highlighted that B. bovis, B. bigemina and A. marginale are present in the dairy cattle herd of the northwestern regions of Rio Grande do Sul, with a higher percentage of A. marginale; on the other hand, T. vivax was not diagnosed in the present study.

ABSTRACT: Protocols to artificially induce cows and heifers into lactation are effective and commercially available in Brazil. However, these protocols demand long hormonal treatments, which is debatable since little research has been done in the subject. To further understand artificial induction to lactation (AIL) protocols, we conducted two experiments. In experiment 1, our objective was to better characterize steroidal profile during an AIL protocol and assess the estradiol (E2) and progesterone (P4) serum profiles of Jersey cows (n=6) subjected to a conventional protocol. In experiment 2, we aimed to compare milk production and serum E2 concentrations of Holstein heifers induced into lactation by the standard AIL protocol (15 injections of E2 and 8 P4 injections) and by a short protocol in which the number of E2 injections and, consequently, the overall dosage of E2, were reduced (8 injections of E2 and 8 P4 injections). We hypothesized that a short AIL protocol (8 days) would be as efficient as the long standard protocol of 15 days. Our hypothesis was confirmed, since we demonstrated that a shorter protocol was able to induce lactation in Holstein heifers without hindering milk production.

Abstract The aim of this study was to investigate the association between chronic Anaplasma marginale and Babesia spp. infection and hematological parameters of pregnant and non-pregnant taurine heifers. Blood samples from 94 females were collected on the first day (D-10) of timed artificial insemination (TAI) protocol and on pregnancy diagnosis (D+34). Hematological parameters were determined and compared between pregnant (PG) and non-pregnant (NPG) heifers, and within group at different sampling days. Real-time PCR (qPCR) was used to determine A. marginale and Babesia bovis infection, and for absolute quantification of Babesia spp. between PG and NPG groups. Correlation analysis was performed between the number of gDNA copies (CN) of Babesia spp. and hematological parameters. On D-10, mean hemoglobin concentration was higher for NPG, and hematocrit and total plasma protein were higher on D+34 for both groups. There was no difference in Babesia spp. CN between groups. In the first qPCR, all heifers were positive for A. marginale and B. bovis. Significant correlations were found between hemoglobin and erythrocyte and between hemoglobin and hematocrit (r = 0.8082 and r = 0.3009, respectively). Low levels of A. marginale and Babesia spp. did not affect hematological parameters of chronically infected pregnant and non-pregnant taurine heifers.

The aim of this study was to compare the J-Synch and conventional protocols associated with estrus detection in beef heifers and to compare pregnancy rate between non-lactating cows displaying estrus or not during the J-Synch protocol. In Experiment 1, heifers were subjected to timed artificial insemination (AI) in a conventional protocol with ECP (n=147) or J-Synch protocol plus eCG (n=149). The AI occurred 12 hours after estrus expression; or 48 (Conventional protocol) and 72 hours (J-Synch protocol) after device removal for animals not displaying estrus. The J-Synch group received 10 µg of GnRH at AI. In Experiment 2, the JSynch was performed (n=116 cows), but without eCG injection, and estrus was monitored. Pregnancy rate was not different between protocols in Experiment 1 (Conventional: 50.68%; J-Synch: 60.4%). Heifers that displayed estrus had higher pregnancy rate only in the conventional protocol. In Experiment 2, pregnancy rate was not different between cows that displayed estrus or not. Therefore, performing AI earlier according to estrus expression increases pregnancy rate in conventional protocol, however it does not increase pregnancy rate in the J-Synch protocol.

ABSTRACT: Previous studies have evaluated the effects of different reproductive procedures on discomfort markers in sheep and cattle. Such studies may help stimulate the adoption of techniques that are more beneficial for animal welfare. However, markers that are commonly used to evaluate discomfort are highly influenced by external factors. To overcome this, several systemic markers can be evaluated to more precisely identify stress, pain, and inflammation. Such markers include cortisol, acute phase proteins, bradykinin, and substance P. We aimed to review the potential markers of stress, pain, and inflammation, and discuss how and when they are regulated after different stimuli related to reproductive procedures in cattle and sheep. Furthermore, we aimed to review how reproductive procedures with different degrees of invasiveness cause stress and provide information that may help develop strategies to limit animal discomfort.

O presente estudo teve como objetivo avaliar a capacidade crioprotetora das lipoproteínas de baixa densidade (LDL) presentes no plasma de gema de ovo, adicionado ao trihidroxiaminometano (TRIS) para congelar sêmen ovino. Trinta e seis ejaculados foram coletados para formar 12 "pool". Cada alíquota de sêmen foi diluída em TRIS-gema de ovo (TRISG) ou TRIS- plasma de gema de ovo (TRISP) antes de congelar o sêmen. Para a obtenção do plasma da gema de ovo, foi utilizado o método de ultracentrifugação. Após o descongelamento, não houve diferença entre os dois extensores em relação aos parâmetros seminais (motilidade, viabilidade, membrana acrossômica e plasma). No entanto, no Teste de Termo Resistência Lenta (TTRL - 4h/38°C), o sêmen congelado com TRISP resultou no aumento do número de espermatozoides com acrossoma intacto (P

ABSTRACT: Gestation length in swine has a considerable amplitude and both early and delayed parturition are common. This variation increases the occurrence of unassisted farrowing and could lead to a wide-ranging age at weaning for piglets born from one batch. Supervision of sow parturition is crucial to reduce mortality of neonate piglets. To facilitate assistance, induction of farrowing using prostaglandin F2α (PGF) has been widely used in batch farrowing systems, whereby synchronization would concentrate the time of farrowing, allowing for better organization of employees. However, a viable alternative method that can be implemented to manage farrowing is to sustain high progestagen levels in the final days of gestation and, consequently, prevent early parturition. Efficient techniques to delay farrowing such as using oral progestagen supplementation have been previously described, but are only recently being considered for commercial use. The present manuscript reviews publications regarding delaying parturition and discusses the use of intravaginal devices (IVDs) containing progestagen. There is limited data addressing the effect of progestagen treatment during gestation on productive and reproductive performance. Therefore, future studies should focus on improving synchronization protocols following progestagen supplementation and evaluating piglet viability and sow fertility, before widely using progestagen supplementation to manipulate parturition.

ABSTRACT: Pregnancy diagnosis is an important tool for farm management. Ultrasonography is the main technique used for pregnancy diagnosis in ewes. As an alternative, radioimmunoassay (RIA) allows accurate and early detection of pregnancy-associated glycoproteins (PAGs) in sheep blood. However, radioactive-based techniques, as RIA, have been increasingly inadvisable due to environmental risk. Homology between ovine and bovine PAGs is high, and ELISA kits used for PAGs detection in cattle are safer than RIA. Thus, this study aimed to evaluate the feasibility of PAGs detection for pregnancy diagnosis in sheep serum samples using an ELISA kit produced for cattle. The sensitivity and specificity of the ELISA kit were 93.5% and 98.9%, respectively, whereas positive and negative predictive values were 99.0% and 93.1%, respectively, in comparison to ultrasonography diagnostic (control). PAGs reached consistently detectable concentrations in ovine serum around 33 days after mating. Accuracy of the ELISA test was 96.1% from 33 days of pregnancy until lambing. After parturition, PAGs were still detectable seven days post-lambing. However, from 21 days post-parturition, PAGs from the previous pregnancy were no longer detected in serum samples. In conclusion, the bovine ELISA kit can accurately detect pregnancy in sheep 33 days following mating, while PAGs levels from the previous gestation are no longer detected from 21 days post-partum. The evaluated ELISA test is a reliable tool for pregnancy diagnosis in sheep at random stages and as a complementary exam at early gestation.
