No presente trabalho desafiamos pintinhos Gallus gallus com altas doses de endotoxina para estudar os mecanismos fisiológicos e bioquímicos da termorregulação de aves frente à inflamação sistêmica. Primeiramente, descrevemos a queda de temperatura corporal durante a inflamação sistêmica em aves como uma resposta regulada (anapirexia). Essa resposta ocorreu por uma inibição de termogênese (redução da taxa metabólica) não relacionada a impedimento da função mitocondrial ou aporte de oxigênio, além de ser apoiada por mecanismos de perda de calor, como ofego e vasodilatação. Em sequência, mostramos que a resposta anapirética à endotoxina parece ser independente da mediação por ciclo-oxigenases (COX), assim o perfil de prostaglandinas (PG) observado neste momento parece não ter função térmica. Também descrevemos que a febre em aves depende da sinalização específica pela COX-2, enzima que quando inibida bloqueia febre via inibição de mecanismos de perda de calor como vasoconstrição e comportamento de agrupamento. Ainda, a febre parece ser majoritariamente dependente da sinalização por PGE2, mas também tem participação das PGD2 e PGF2. No presente trabalho apontamos ainda que a competição com uma demanda energética aumentada por frio, e frio combinado à jejum favorece anapirexia e elimina a resposta de febre nos pintinhos. Finalmente mostramos que a exposição de pintinhos ao poluente dioxina durante o desenvolvimento embrionário resultou em aumento de taxa metabólica (demanda energética). Assim, quando foi induzida a inflamação, os pintinhos apresentaram uma resposta hipometabólica persistente acompanhada da eliminação das respostas hematológicas e osmóticas normais à endotoxina. Concluímos que pintinhos podem responder a um desafio imune tanto com anapirexia como com febre e que a resposta anapirética é favorecida em caso de demandas metabólicas aumentadas. Ainda, a anapirexia não é mediada pela via das COX-PG enquanto a febre é mediada especificamente pela COX-2 e principalmente por PGE2. Although thermal responses to pathogen infections are decisive for birds’ survival, the mechanisms of fever are largely unknown and no study has addressed the decrease in body temperature during systemic inflammation for birds. Here we challenged Gallus gallus chicks with high doses of endotoxin to study the physiological and biochemical mechanisms of birds’ thermoregulation during systemic inflammation. Firstly, we described the decrease in body temperature during systemic inflammation in birds as a regulated response (anapyrexia). This response was supported by a thermogenesis inhibition (reduced metabolic rate) unrelated to impairment of mitochondrial function or oxygen supply, and also by heat loss mechanisms, such as tachypnea and vasodilation. In sequence, we presented that anapirexia in response to endotoxin was independent of cyclooxygenases mediation (COX), thus the prostaglandin (PG) profile observed during this response did not reflect a thermal function. Moreover, the fever response depended specifically on COX-2 signaling in our chicks, and when it was blocked fever was eliminated by inhibition of heat loss mechanisms such as vasoconstriction and huddling behavior. Yet, fever seems to be mostly dependent on PGE2 signaling, but PGD2 and PGF2 had also a minor increase in expression during fever. Our data also pointed out that competing energetic demands caused by cold, and cold combined with fasting favors anapyrexia and eliminates the fever response in chicks. Finally, we show that the exposure of chicks to the pollutant dioxin during embryonic development resulted in increased in metabolic rate. Thus, when inflammation was induced, these chicks showed a persistent hypometabolic response accompanied by the elimination of normal hematological and osmotic responses to endotoxin. We conclude that chicks can respond to an immune challenge with both anapyrexia and fever, and the anapyretic response is favored in case of increased metabolic demands. Furthermore, anapyrexia is not mediated by the COX-PG pathway, whereas fever is mediated specifically by COX-2 and mainly by PGE2. NOS and COX-2 enzymes are key enzymes in the inflammatory response when stimulated primarily by pro-inflammatory cytokines. Compounds that act to modulate the activity of these enzymes are promising in the fight against inflammation, for example the N-acylhydrazonic derivatives that prove to be effective in classic models of inflammation in vivo and in vitro. This work aimed to evaluate in vitro, in vitro and in vivo, the possible mechanisms of anti-inflammatory action of the derivatives 2-cyanoacetohydrazide of the JR series (04, 09 and 19). Initially, the in silico study of the three compounds and in vitro (from JR04, JR09 and JR19) against cyclooxygenase isoforms (COXs) was performed. Next, gastric toxicity in mice and cytotoxicity in human erythrocytes were evaluated with compounds JR09 and 19. To investigate the participation of the nitric oxide / guanylate cyclase pathway of compound JR19, the carrageenan-induced peritonitis assay was performed in pre-treatment animals with L-NAME (non-specific nitric oxide synthase blocker) or methylene blue (non-specific guanylate cyclase blocker). In the same model the cytokine dosage was performed. The in silico results showed that JR04, JR09 and JR9 interacted specifically with the catalytic site of the COXs, and the compound JR04 had better affinity for COX-1. The in vitro test for the molecules JR04 and JR09 and JR19 corroborated with the in silico assay, where JR04 was shown to be more selective for COX-1 with a selectivity index (IS) of 2.22 for COX-2, whereas the molecules JR09 and 19 showed an IS of 1.67 and IS of 1.51, respectively with a percentage of inhibition of 100% of COX-2 in concentration of 25mM. In the gastric toxicity test the two compounds (100 mg / kg oral and subcutaneous) did not induce ulcerative lesions, even though they were able to interact with COX-1. In the erythrocyte cytotoxicity assay the compounds JR09 and JR19 showed low haemolytic activity, with an IC 50 of 51.42 and 85.02 mg / mL, respectively. In carrageenan-induced peritonitis, the compound JR19 was able to significantly reduce leukocyte counts of the peritoneal exudate by 48.3% compared to the control group, but this effect was reversed in animals pretreated with L-NAME or methylene blue, showing that this effect is dependent on the NO / GCs pathway. The levels of proinflammatory cytokines TNF-α, IFN-γ, IL-6 and IL-17 were also significantly reduced with administration of compound JR19 and reversed in animals pretreated with L-NAME, corroborating the probable participation of anti-inflammatory effect of this compound. The concentrations of IL-2 and IL-4 cytokines were not altered. Thus, the compounds JR04, JR09 and JR19 probably act in interaction with the COXs, compounds JR09 and 19 acting selectively on COX-2, without gastric toxic effects. In addition, compound JR19 has its anti-inflammatory action likely linked to maintenance of NO levels and reducing proinflammatory cytokines. O processo inflamatório é uma reação do organismo a uma infecção ou lesão dos tecidos. As enzimas NOS e COX-2 são enzimas chaves na resposta inflamatória quando estimuladas principalmente por citocinas pró-inflamatórias. Compostos que atuem modulando a atividade dessas enzimas são promissores no combate a inflamação, a exemplo dos derivados N-acilhidrazônicos que mostram ser eficazes em modelos clássicos de inflamação in vivo e in vitro. Este trabalho teve como objetivo avaliar in silico, in vitro e in vivo, os possíveis mecanismos de ação anti-inflamatória das moléculas derivadas 2-cianoacetohidrazida da série JR (04, 09 e 19). Inicialmente foi realizado o estudo in silico dos três compostos e in vitro (de JR04, JR09 e JR19) frente às isoformas de cicloxigenase (COXs). Em seguida foi avaliada a toxicidade gástrica em camundongo e a citotoxicidade em eritrócitos humanos com os compostos JR09 e 19. Para investigar a participação da via óxido nítrico/guanilato ciclase do composto JR19 foi realizado o ensaio de peritonite induzida por carragenina em animais pré-tratandos com L-NAME (bloqueador inespecífico da oxido nítrico sintase) ou azul de metileno (bloqueador inespecífico da guanilato ciclase). Neste mesmo modelo foi realizada a dosagem de citocinas. Os resultados in silico mostraram que JR04, JR09 e JR9 interagem de forma específica com o sítio catalítico das COXs, e o composto JR04 teve melhor afinidade pela COX-1. O teste in vitro para as moléculas JR04 e JR09 e JR19 corroborou com o ensaio in silico, onde o JR04 mostrou-se mais seletivo para COX-1 com um índice de seletividade (IS) de 2,22 para COX-2, enquanto que as moléculas JR09 e 19 apresentaram um IS de 1,67 e IS de 1,51, respectivamente com uma porcentagem de inibição de 100% de COX-2 na concentração de 25mM. No ensaio de toxicidade gástrica os dois compostos (100 mg/kg via oral e subcutânea) não induziram lesões ulcerativas, mesmo sendo capazes de interagir com a COX-1. No ensaio de citotoxicidade em eritrócitos os compostos JR09 e JR19 apresentaram baixa atividade hemolítica, apresentando ainda um IC50 de 51,42 e 85,02 mg/mL, respectivamente. Na peritonite induzida por carragenina o composto JR19 foi capaz de reduzir significativamente a contagem de leucócitos do exsudato peritoneal em 48,3% em relação ao grupo controle, porem este efeito foi revertido nos animais pré-tratados com L-NAME ou azul de metileno, mostrando ser este efeito dependente da via NO/GCs. Os níveis das citocinas pró- inflamatórias TNF-α, IFN-γ, IL-6 e IL-17 também foram significativamente reduzidas com administração do composto JR19 e foi revertido nos animais pré-tratados com L-NAME, corroborando com a provável participação da via do NO no efeito anti-inflamatório deste composto. As concentrações das citocinas IL-2 e IL-4 não foram alteradas. Dessa forma, os compostos JR04, JR09 e JR19 provavelmente atuam por interação com as COXs, com os compostos JR09 e 19 atuando de forma seletiva sobre a COX-2, sem efeitos tóxicos gástricos. Além disso, o composto JR19 tem sua ação anti-inflamatória provavelmente ligada a manutenção dos níveis de NO e reduzindo citocinas pró-inflamatórias.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Toxoplasma gondii é um protozoário parasito intracelular obrigatório que infecta uma ampla variedade de vertebrados de sangue quente, incluindo seres humanos. Muitos estudos demonstram que a ciclooxygenase-2 (COX-2) é um potente modulador da resposta imune em vários tipos de infecção. No entanto, o papel da ciclooxygenase ainda não é claro durante uma infecção causada por T. gondii. Neste sentido, o objetivo deste estudo foi verificar o papel funcional da COX-2 em roedores Calomys callosus infectados por T. gondii, um excelente modelo experimental in vivo tradicionalmente utilizado para estudar a toxoplasmose. Para este fim, fêmeas de C. callosus foram infectadas com cinquenta cistos de T. gondii (cepa ME49), tratadas com inibidores da ciclooxygenase Meloxicam (inibidor de COX-1 e COX-2) e Celecoxibe (inibidor seletivo de COX-2), e avaliadas a cada 48 horas para a verificação da alteração de peso corporal e morbidade durante 40 dias consecutivos. Como controle, os animais foram infectados e não tratados com inibidores da ciclooxygenase. Depois de 40 dias de infecção, os cérebros foram coletados e processados para imuno-histoquímica ou PCR em tempo real para detecção de T. gondii, enquanto o soro dos animais foi avaliado para detecção de citocinas. Além disso, macrófagos peritoneais de C. callosusnão infectados ou infectados com a cepa RH de T. gondii foram tratados com Meloxicam ou Celecoxibe a fim de avaliar a proliferação intracelular do parasito por ensaio de beta-galactosidase, produção de citocinas por ELISA e produção de nitrito por meio do método de Griess, após 24 h de infecção. Os resultados mostraram que a morbidade dos animais foi alterada após a infecção por T. gondii, independentementedo tratamento. Em relação à alteração de peso corporal, animais tratados com Celecoxibe apresentaram uma perda de peso menor quando comparados com seu respectivo controle. A imuno-histoquímica e PCR em tempo real realizados no tecido cerebral mostraram uma redução significativa de cistos teciduais em animais tratados com ambos os inibidores, quando comparados com animais infectados e não tratados. Além disso, observou-se que ambos os tratamentos com os inibidores foram capazes de controlar a proliferação intracelular de T. gondii em macrófagos peritoneais. No soro de C.callosus, os dados do ELISA mostraram que animais tratados com Meloxicam apresentaram uma tendência para maior produção de MIF quando comparados ao controle. Já no sobrenadante de macrófagos peritoneais, houve um aumento na produção de TNF-α e uma redução de IL-6, e o ensaio de Griess demonstrou que ambos os tratamentos induziram uma alta produção denitrito. Todos estes resultados indicam que as ciclooxygenases são capazes de aumentar a susceptibilidade a infecção por T. gondii, uma vez que a inibição destes mediadores induz o controle significativo da infecção. Em conclusão, os nossos resultados mostram que a COX-1 e COX-2 são importantes para aumentar o processo infeccioso por T. gondii no cérebro e macrófagos peritoneais de C.callosus. Toxoplasma gondii is an obligate intracellular protozoan parasite that infects a wide range of warm-blooded vertebrates, including humans. Many studies show that the ciclooxygenase-2 (COX-2) is a potent modulator of immune response in multiple types of infection. However, the role of cyclooxygenase during an infection by T. gondii is still not clear. Therefore, the aim of this study was to investigate the functional role of COX-2 in Calomys callosus rodents infected with T. gondii, an excellent in vivo experimental model traditionally used for studying toxoplasmosis. For this purpose, C. callosus females were infected with fifty cysts of T. gondii (ME49 strain), treated with ciclooxygenase inhibitors as Meloxicam, COX-1 and COX-2 inhibitor, and Celecoxib, COX-2 selective inhibitor, and evaluated every 48 hours to check the body weight change and morbidity for 40 consecutive days. As a control, the animals were infected and not treated with ciclooxygenase inhibitors. After 40 days of infection was performed collection and processing of the brains for immunohistochemistry or real-time PCR for the detection of T. gondii, while evaluating the serum of animals for the detection of cytokines by ELISA. Furthermore, the peritoneal macrophages of C. callosus, uninfected or infected with T. gondii RH strain, were treated with Meloxicam or Celecoxib in order to evaluate the parasite intracellular proliferation by beta-galactosidase assay, and the supernatant was collected for cytokine detection by ELISA and production of nitrite by Griess method, after 24h of infection. The results showed that the morbidity of the animals changed after infection by T. gondii, regardless of both treatments. Regarding the change in body weight, animals treated with Celecoxib had a smaller loss of weight when compared to their respective control. Immunohistochemistry and real-time PCR performed on brain tissue showed a significant reduction of tissue cysts in animals treated with both inhibitors when compared to infected and untreated animals. Besides, it was observed that both treatments with the inhibitors were able to control the T. gondii intracellular proliferation in peritoneal macrophages. On the serum of C. callosus, the ELISA data shows that animals treated with Meloxicam showed a tendency to increase the MIF production when compared to the PBS control. In the supernatant of peritoneal macrophages, there was an increase in TNF-α production and IL-6 reduction, and Griess assay showed that both treatments induced a high production of nitrite. All these results indicate that cyclooxygenases are capable of favoring infection by T. gondii, since inhibition of these mediators induced significant control of infection. In conclusion, our results show that COX-1 and COX-2 are important to facilitate the T. gondii infection in the brain and peritoneal macrophages of C. callosus. Dissertação (Mestrado)
