No. of bitstreams: 1 Luis Fernando Zamuner.pdf: 1709325 bytes, checksum: 0c0988cd726b220df3411fb7e529b5f0 (MD5) Previous issue date: 2020-05-01 This study aims to understand and compare the effects of the low level laser therapy (LLLT) and magnetotherapy (MAG) on C2C12 cells of new passage (representing cells with 6 passages) and old (representing cells with 76 passages). For this study, C2C12 cells were cultured, with cell growth monitoring performed every 24 h. LLLT was used at a wavelength of 660 nm. In this study, it was employ new equipment, a magnet device, which induces an oscillating magnetic field, with 12 Hz rate. The effects of LLLT and MAG on cell viability and integrity were analyzed, in addition to the migration of cells by mechanical injury. In addition, the release of pro and anti-inflammatory cytokines (IL-1β, IL-6, TNF-α and IL-10) was analyzed after the incubation of cells with LPS, by enzyme immunoassay. The expression of MyF-5, Pax7, MyoD, myogenin, caspase-3, p38 MAPK and NF-B proteins was analyzed using the western blot method. The intracellular calcium concentration and the concentration of the membrane potential in the cells were measured. The results showed an increase in cell viability and detachment in the two treatment methods used. In addition, it was observed an increase in cell migration after the using LLLT and MAG, when compared to natural migration. The expression of proteins in all myogenic factors evaluated denotes a significant increase by both LLLT and MAG. The group treated with LLLT was effective in both cell passages, in relation to the other groups, when the intracellular calcium concentration was assessed. The concentration of the membrane potential had a significant response post photobiomodulation and MAG. The LLLT, as well as MAG, reduced the release of the cytokines TNF-α, Il-1β and IL-6 in C2C12 cells, in the studied time and in the two cell passages evaluated. There was also a significant increase in the release of the cytokine IL-10. These data suggested that both treatments improved the process of migration and regeneration, facilitating the myogenic response, using the therapy with energy-based devices, as LLLT and MAG. Este estudo tem o objetivo de compreender e comparar os efeitos do Laser de Baixa Intensidade (LBI) e da Magnetoterapia (MAG) em células C2C12 de passagem nova (representando células com 6 passagens) e antiga (representando células com 76 passagens). Para o estudo, foi realizado o cultivo de células C2C12, com o monitoramento do crescimento celular realizado a cada 24 h. Foi utilizado o LBI no comprimento de onda 660 nm. Neste estudo empregamos um novo instrumento, o aparelho magneto, que induz um campo magnético oscilante, com frequência de 12 Hz. Foram analisados os efeitos do LBI e MAG na viabilidade e integridade celular, além da migração das células por lesão mecânica. Ainda, foi analisado a liberação de citocinas pró e anti-inflamatórias (IL-1β, IL-6, TNF-α e IL-10), após a incubação das células com LPS, por ensaio imunoenzimático. Foi analisado a expressão de proteínas MyF-5, Pax7, MyoD, miogenina, caspase-3, p38 MAPK e NF-B através do método de western blot. Foi mensurada a concentração de cálcio intracelular e a concentração do potencial de membrana nas células. Os resultados demonstraram aumento da migração, viabilidade celular e integridade pelos dois métodos de tratamento utilizados. A expressão de todos os fatores miogênicos avaliados teve elevação significativa tanto com o LBI quanto com a MAG. O grupo tratado com LBI foi eficaz nas duas situações de passagem, em relação aos outros grupos, quando avaliada a concentração de cálcio intracelular. A concentração do potencial de membrana teve resposta significativa com o LBI e MAG. O LBI, assim como a MAG, reduziram a liberação das citocinas TNF-α, IL-1β e IL-6 nas células C2C12, no tempo estudado e nas duas passagens de células avaliadas. Também ocorreu incremento da liberação da citocina IL-10. Esses dados sugerem que ambos tratamentos melhoram o processo de migração e regeneração, facilitando a resposta miogênica, frente a terapia com aparelhos baseados em energia (LBI e MAG).
