Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Adventure games, as a genre defined in the context of electronic games, have had throughout their history evolutions and transformations that allowed more, and more, complex stories to be told through video games. The objective of this work starts from a macro analysis, bringing the history of adventure games as a genre, and then, in a second moment, analyzes the style and trajectory of one of the main companies recognized as a reference in the production of narrative games, Telltale Games, starting from a panorama that begins in the history of its predecessor, LucasArts, and ends in the analysis of the particular characteristics of the games produced by the company. From this context, the objective is to carry out a specific analysis of one of the company's titles: Game of Thrones - A Telltale Games Series. It is with the construction of such an analysis that it will be possible to understand how adventure games have effectively evolved as a genre and how narrative and gameplay come together around building an experience based on the player's choices and the development of the story told by the game in question. Os adventure games, como um gênero definido no contexto dos jogos eletrônicos, tiveram ao longo de sua história evoluções e transformações que permitiram que, cada vez mais, histórias mais complexas fossem contadas através dos videogames. O objetivo deste trabalho é partir de uma análise macro, trazendo a história dos adventure games como gênero, para então, em um segundo momento, analisar o estilo e trajetória de uma das principais empresas reconhecidas como referência na produção de jogos majoritariamente narrativos, a Telltale Games, partindo de um panorama que se inicia na história de sua predecessora, a LucasArts, e se encerra na análise das características particulares dos jogos produzidos pela empresa. A partir desse contexto, o objetivo é realizar uma análise específica de um dos títulos da empresa: Game of Thrones – A Telltale Games Series. É com a construção de tal análise que será possível compreender como os adventure games evoluíram efetivamente como gênero e como narrativa e jogabilidade se unem em torno de construir uma experiência baseada nas escolhas do jogador e no desenvolvimento da história contada pelo jogo em questão. This article has the goal of presenting the development of a mobile application as a complementary way of vaccine registry, besides motivating people to abide by a vaccination calendar, thus avoiding unnecessary spending with revaccination and diminishing both the treatment time and dose waste. It was developed in a web software named APP Inventor. Foi analisada a influência do gerenciamento de terminologia (BOWKER, 2015) no processo, buscando observar se a identificação e a tradução prévia dos termos colaboram com a qualidade e a produtividade. A pesquisa foi realizada a partir da tradução de seis textos feita por quatro tradutores profissionais voluntários em três cenários diferentes (Cenário 1: utilizando somente processador de texto, Cenário 2: utilizando somente Lilt e Cenário 3: utilizando Lilt aliado ao gerenciamento de terminologia). A análise qualitativa das traduções nos três referidos cenários baseou-se em uma planilha de controle de qualidade à luz da concepção de qualidade para objetivos específicos (TAUS, 2017). Também se examinou a existência de diferenças de produtividade e qualidade a partir da análise contrastiva entre os cenários propostos, dos resultados da planilha e do tempo gasto na realização das traduções. Os resultados das análises indicaram que a abordagem Lilt, aliada à pós-edição humana e o gerenciamento de terminologia pode aumentar significativamente a produtividade e a qualidade das traduções especializadas, um indício da importância do uso de tecnologia como auxílio ao tradutor. Outros benefícios encontrados incluem a aplicabilidade em outras áreas de especialidade, como engenharia, computação, medicina etc., mais facilidade na manutenção de registros terminológicos e também maior controle da terminologia no caso das agências de tradução, que podem garantir um uso consistente da terminologia independentemente da equipe. Espera-se que esta pesquisa possa abrir caminhos para o aprimoramento dos processos de tradução envolvendo mecanismos de tradução automática adaptativa, ainda pouco explorados. Based on a study on the historical journey of the translation automation, we present an analysis of translations carried out by professional translators in order to observe challenges and benefits of this approach. The influence of terminology management in the process was also analyzed (BOWKER, 2015), seeking to observe whether the prior identification and translation of terms collaborate with quality and productivity. The research examined the translation of six texts completed by four voluntary professional translators in three different scenarios (Scenario 1: using only text processor, Scenario 2: using Lilt and Scenario 3: using Lilt combined with terminology management). The qualitative analysis of translations in the three scenarios was based on a quality control sheet in the light of the quality for specific purposes concept (TAUS, 2017). Finally, there was also a difference of productivity and quality from the contrasting analysis between the proposed scenarios, the spreadsheet results and the time spent to complete the translations. This research can open paths for improving translation processes involving adaptive MT mechanisms that have been not been fully exploited so far. The results of the analyses suggested that Lilt and human post-editing combined with terminology management approach can significantly increase productivity and quality of specialized translations, an indication of the importance of the use of technology as aid to translators. Furthermore, other benefits and challenges have also been observed in adopting this approach. Among the benefits are the applicability in other areas of expertise, such as engineering, computing, and medicine, easier terminology maintenance and greater terminology control for translation agencies, thus enhancing consistent use of terminology independently of the translation team.

Londrina é um dos maiores e mais urbanizados municípios do Estado do Paraná e da região Sul do Brasil. No município, afim de evitar a infestação do mosquito vetor e a circulação de vírus, o uso de inseticidas piretroides é bastante comum. Esse composto químico têm um custo acessível e é utilizado principalmente no ambiente doméstico, porém seu uso descontrolado pode ocasionar a seleção de alelos de resistência nas populações (resistência knockdown). O monitoramento de mutações de resistência assim como a variabilidade genética do mosquito é importante para conhecer a estrutura das populações e direcionar campanhas de controle do inseto. Sendo assim, o presente estudo avaliou onze localidades em diferentes regiões da cidade e três localidades dentro da Universidade durante os meses de novembro e dezembro de 2017. Em relação a variabilidade genética (gene mitocondrial ND4), dezessete haplótipos foram encontrados nas cinco regiões do município, enquanto na Universidade dez haplótipos foram observados. Em ambas análises, o número haplotípico pode ser considerado elevado. O fluxo gênico foi baixo e as populações avaliadas se demonstraram estruturadas: a variação foi maior entre populações que dentro delas em ambas localidades (77,9% em Londrina e 67,7 na UEL). Em relação às mutações de resistência (Val1016Ile e Phe1534Cys kdr), as populações avaliadas apresentaram uma elevada frequência dos alelos mutantes: 50% para o alelo Ilekdr na cidade de Londrina e 57,3% na UEL, enquanto para o alelo Cyskdr, a frequência foi de 52% na cidade de Londrina e 62,2% nos três centros avaliados da UEL. Na Universidade, 3.321 ovos de Ae. aegypti foram coletados para monitoramento, com positividade de 54% nas armadilhas e proporções semelhantes de infestação nas três localidades. Os resultados demonstraram a diferenciação de cada ponto de coleta, sugerindo que ações de controle devem ser direcionadas considerando a particularidade de cada região e de cada localidade. A partir da forte presença dos alelos de resistência, é possível que a aplicação de inseticidas piretroides não seja a forma de controle ideal no município, tornando necessário o uso de outras alternativas para evitar a proliferação do mosquito e surtos de dengue e outras arboviroses. Londrina is one of the biggest and most urban cities in Paraná and in southern Brazil. In Londrina, in order to avoid the infestation of the vector mosquito and the spread of the virus, the usage of pyrethroid insecticides is common. This low-cost chemical compound is used mainly in the household environment. The uncontrolled pyrethroid use may cause the selection of resistance alleles in populations (knockdown resistance). Monitoring resistance mutations, as well as the mosquito?s genetic variability, is important to learn the structure of the populations and direct insect control campaigns. Therefore, this study evaluated eleven locations in different regions of the city and three locations within the University during the months of November and December of 2017. Genetic variability-wise (ND4 mitochondrial gene), seventeen haplotypes have been found in the five regions of the city, whereas in the University, ten haplotypes have been observed. In both analyses, the haplotype number may be considered elevated. The gene flow was low, and the evaluated populations have shown to be structured: the variance was higher between populations than within themselves in both locations (77.9% in Londrina and 67.7% in UEL). In relation to resistance mutations (Val1016Ile and Phe1534Cys kdr), the evaluated populations represented a high frequency of mutant alleles: 50% for the Ilekdr allele in Londrina, and 57.3% in UEL, whereas for the Cyskdr allele, the frequency was 52% in Londrina, and 62.2% for the three evaluated centers in UEL. In the University, 3,321 Ae. aegypti eggs were collected for monitoring, with a positivity of 54% in the traps and similar rates of infestation on the three locations. The results demonstrated differentiation in each collect spot, which suggests control actions tailored to each region considering their own particularity. From the strong presence of resistance alleles, pyrethroid insecticides might not be the ideal control mechanism for Londrina, making it necessary the use of alternatives to avoid mosquito proliferation and dengue and other arbovirosis outbreaks.

FtsZ é homóloga de tubulina, presente em quase todas as bactérias, que se autoassocia em filamentos que formam uma estrutura chamada anel Z dentro das células. O anel Z quando formado recruta de um macrocomplexo proteico chamado divisomo, que é responsável pela síntese do septo de divisão, formando duas células filhas. Diversos moduladores se ligam diretamente a FtsZ regulam sua polimerização, controlando o momento e o local onde o anel Z é formado. MciZ é um peptídeo de 40 aminoácidos expresso durante a esporulação de Bacillus subtilis e inibe a formação do do anel Z na célula mãe. O objetivo do presente trabalho foi estudar a interação entre as proteínas FtsZ e MciZ e investigar os mecanismos envolvidos na inibição da polimerização de FtsZ por MciZ. Através de uma triagem genética, usando uma biblioteca de mutantes de ftsZ, identificamos treze mutações em ftsZ que conferiram resistência à superexpressão de MciZ in vivo. Sete delas eram capazes de crescer na presença e na ausência da superexpressão de MciZ e as outras seis se mostraram dependentes da superexpressão de MciZ. A partir da coexpressão e copurificação do complexo FtsZ:MciZ, observamos que todas as proteínas mutantes ainda continuavam interagindo com MciZ in vitro. O Kd estimado para a interação entre as proteínas foi de 150±50nM, e mostrou-se que MciZ não se liga nem ao CTP (C-Terminal Peptide) de FtsZ, nem compete com GTP para a ligação no mesmo sítio. Usando construções truncadas de MciZ, determinou-se que o N-terminal da proteína (resíduos 1 ao 27) é suficiente para inibição. A partir das estruturas tridimensionais de MciZ (RMN) e do complexo FtsZ:MciZ (cristalografia de raios x), determinou-se que MciZ é um peptídeo desenovelado, que assume uma estrutura terciária ao interagir através da sua α-hélice H2 e folha-β B2 com a α-hélice H10 e a folha-β S9 de FtsZ. MciZ mostrou-se capaz de reduzir o tamanho dos protofilamentos de FtsZ de forma subestequiométrica, gerando fragmentos menores de filamentos. Proporções de MciZ:FtsZ de 1:10 foram suficientes para extinguir completamente o anel Z, confirmando a inibição subestequiométrica também in vivo. A conservação da inibição da fusão FtsZ-MciZ e a cinética de despolimerização de FtsZ induzida por MciZ provaram que MciZ não é um simples sequestrador. Marcações fluorescentes de MciZ sugeriram que o peptídeo é capaz de interagir com o anel Z in vivo, e também decorar feixes de FtsZ in vitro, formando focos localizados frequentemente na ponta dos filamentos. Cossedimentações com polímeros de FtsZ mostraram a presença de MciZ ou da fusão FtsZ-MciZ. Apesar de MciZ induzir o aumento da atividade GTPáscia específica de FtsZ, a ausência de hidrólise de GTP não eliminou o efeito subestequiométrico de MciZ. Nossos resultados em conjunto mostram que MciZ é um capeador dos filamentos de FtsZ, bloqueando a elongação pela ponta menos e bloqueando o anelamento entre protofilamentos FtsZ is a tubulin-like protein present in most bacteria, that self-assembles into filaments forming a structure known as Z-ring in the cells. Following formation, the Z- ring recruits a protein macrocomplex, the divisome, which is responsible by the division septum synthesis, resulting in two daughter cells. Many modulators interact directly to FtsZ, regulating its polymerization and controlling the time and place of the Z-ring formation. MciZ is a 40-amino-acid peptide that is expressed during sporulation in Bacillus subtilis and inhibits the formation of the Z-ring in the mother-cell. The aim of this work was to study the interaction between FtsZ and MciZ proteins, and to investigate the mechanisms involved in FtsZ inhibition by MciZ. Applying a genetic screening, using an ftsZ mutant library, we identified 13 mutations on ftsZ that conferred resistance to MciZ overexpression in vivo. Seven of them were able to grow either in the presence or absence of MciZ overexpression, and the other six showed to be dependent on it. With the co-expression and co-purification of the FtsZ:MciZ complex, we observed all mutant proteins still interact with MciZ in vitro. Estimated Kd for the interaction was 150±50nM, and it was demonstrated that MciZ does not bind to FtsZ CTP (C-Terminal Peptide), nor does it compete with GTP for the same binding site. Using truncated versions of MciZ, it was determined that its N-terminal (residues 1 to 27) is sufficient for the inhibition. Based on the tridimensional structure of MciZ (NMR) and of the FtsZ:MciZ complex (x- ray crystallography), it was determined that MciZ is an unstructured peptide that assumes a tertiary structure by interacting with FtsZ α-helix H10 and β-sheet S9 through its α-helix H2 and β-sheet B2. MciZ was able to reduce the size of FtsZ protofilaments in a substoichiometric manner, generating smaller fragmented filaments. 1:10 ratios of MciZ:FtsZ were sufficient to completely extinguish the Z-ring, thus confirming the substoichiometric inhibition in vivo as well. The inhibition of FtsZ polymerization by the FtsZ-MciZ fusion and the FtsZ depolymerization kinetics induced by MciZ proved that MciZ is not a simple sequesterer. Fluorescent dyeing of MciZ suggests the peptide is able to interact with the Z-ring in vivo, as well as decorate FtsZ bundles in vivo, forming localized spots frequently at the filaments\' ends. Co- sedimentations with FtsZ polymers showed the presence of MciZ or of the FtsZ-MciZ fusion. Despite MciZ-induced increase in specific GTPase activity of FtsZ, the lack of GTP hydrolysis did not eliminate the substoichiometric effect of MciZ. Combined, our results show that MciZ is an FtsZ filament capper, blocking elongation at the minus end and blocking the annealing between protofilaments

The Sado River Drainage Survey project (2004-2008) was designed to fill a significant gap in our knowledge of the prehistory of Portugal. Southern Alentejo constitutes nearly one third of the total land mass of continental Portugal, but has received comparatively little attention from Palaeolithic archaeologists. Practically nothing was known about the prehistory of the Sado River basin, which includes the southern Alentejo plain, before now. The results of the Sado River Drainage Survey (SRDS) indicate that the Sado River basin was likely occupied at low population densities during the Middle Palaeolithic. There is some evidence for a Lower Palaeolithic presence but little or no evidence of an Upper Palaeolithic occupation. The emerging pattern suggests either an occupational hiatus or a major shift in settlement pattern towards the end of the Middle Palaeolithic. Possible explanations for this pattern, including aridification driven by climate change, are explored here.

A citocinese bacteriana é controlada por diversas proteínas que se agrupam em um complexo chamado divisomo. O cerne do divisomo é constituído por FtsZ, uma proteína homóloga à tubulina eucariótica, que se auto-associa formando uma estrutura chamada anel Z. O anel Z serve como arcabouço e recruta diversas outras proteínas componentes do divisomo para o sítio onde o septo será sintetizado na célula. A formação do anel Z é modulada por proteínas que se ligam diretamente a FtsZ e regulam a sua auto-associação, tanto induzindo como inibindo a sua polimerização. Apesar de muitos destes moduladores de FtsZ já serem conhecidos, muito pouco se sabe sobre o mecanismo pelo qual eles controlam a estruturação do anel Z in vivo. O objetivo do presente trabalho foi estudar a interação entre FtsZ e um modulador de divisão, a proteína ZapA, da bactéria gram-positiva Bacillus subtilis. Para isso construímos uma biblioteca de mutantes de ftsZ por \"Error Prone PCR\", com aproximadamente 1 substituição por cópia de ftsZ e contendo um total de 1x105 clones. A partir dessa biblioteca, utilizamos duas triagens genéticas para identificar mutantes incapazes de interagir com ZapA. Na primeira estratégia, selecionamos 12 mutantes de FtsZ resistentes à superexpressão de uma forma tóxica de ZapA, que bloqueia a divisão, causando filamentação e morte das células. Surpreendentemente, apesar destes mutantes serem insensíveis ao efeito de ZapA, ensaios citológicos mostraram que nenhum deles perdeu a interação com ZapA. Como as mutações foram mapeadas nas vizinhanças do sítio catalítico e de polimerização de FtsZ, e como a maioria delas confere resistência cruzada aos efeitos de outros moduladores de FtsZ, suspeitamos que elas afetassem a estabilidade do polímero de FtsZ e, consequentemente, o comportamento do anel Z. Essas suspeitas foram confirmadas em ensaios de FRAP e cálculos de proporção de FtsZ no anel Z, indicando que os mutantes formam um anel Z mais estável que o normal. Como não obtivemos mutantes que perderam a interação com ZapA na primeira triagem, aplicamos a biblioteca em uma segunda estratégia de triagem genética, procurando um mutante de FtsZ que voltasse a interagir com um mutante de ZapA que não se liga mais a FtsZ (ZapAN62A). Esta estratégia de ganho de função identificou um candidato, FtsZE91V , que, tanto por critérios genéticos como citológicos, voltou a interagir com ZapAN62A. Apesar do mutante FtsZE91V mostrar-se capaz de restaurar a interação com ZapAN62A, ele não afetou a interação com ZapA selvagem, segundo nossos ensaios de microscopia de fluorescência e viabilidade. O mutante FtsZE91V, mapeia na hélice H3 de FtsZ. Esta hélice está exposta na superfície de FtsZ (compõe um dos lados da molécula de FtsZ) de uma maneira compatível com a idéia de que ela seria importante para interações laterais entre polímeros de FtsZ. Nossos resultados apontam, portanto, que a hélice H3 deve ser o sítio de interação para ZapA em FtsZ. The bacterial cytokinesis is ruled by a number of proteins that constitute the divisome complex. FtsZ, a homologue of eukaryotic tubulin, is the main component of the divisome and self-associates in a structure named Z ring. The Z ring works as a scaffold and recruits the other components of divisome, establishing itself where the septum will be synthesized in the cell. Some of these proteins interact directly with FtsZ and control self-association, promoting polymerization or preventing it. Although there have been discovered many of FtsZ modulators, little is known about the mechanisms that control the formation of the Z ring in vivo. The aim of this work was study de interaction between FtsZ e one of its division modulators, ZapA protein, on Bacillus subtilis grampositive bacteria. We created a mutagenized ftsZ plasmid library by error prone PCR, which contained 1,0x105 transformants and exhibited a mutation rate of one substitution per ftsZ copy. The library was transformed into a modified Bacillus subtilis strain and we performed two genetic screenings to select cells with FtsZ mutants incapable of interacting with ZapA. In first strategy, we selected 12 resistant ftsZ mutants for a toxic ZapA overexpression, that blocked division and caused filamentation and cell death. Surprisingly, although these mutants were insensitive to ZapA effect, cytological assays showed that none of them lost interaction with ZapA. As the substitutions were mapped around the catalytic and interaction site of FtsZ structure and showed resistance to other modulators, we suspected that the mutations were affecting the polymer stability of FtsZ and, consequently, the behavior of Z ring. This hypothesis was confirmed by FRAP experiments and by calculations of FtsZ proportions in Z ring, pointing out that the mutants form more stable Z rings. As we didnt\' find mutants that lost their ZapA´s interaction, we applied our library in a second genetic screen, looking for mutants that return to interact with a ZapA mutant (ZapAN62A) that doesn´t bind to FtsZ anymore. This gain of function strategy identified one candidate, FtsZE91V, which returns to interact with ZapAN62A in our genetic and cytological assays. Although the mutant FtsZE91V showed itself capable to interact with ZapAN62A, that didn´t affect the interaction with wild type ZapA by our fluorescent microscopy and viability assays. The substitution E91V was mapped on H3 helix of FtsZ structure. This helix is exposed on FtsZ surfaces (on FtsZ´s lateral side), being compatible with the idea that lateral interaction is important in FtsZ polymers. So, we concluded that helix H3 is the binding site of ZapA in FtsZ.

A bactéria Gram-negativa Pseudomonas aeruginosa é um patógeno oportunista frequentemente associado a vítimas de queimaduras graves ou indivíduos com fibrose cística, sendo os isolados resistentes a carbepenêmicos dessa espécie considerados pela OMS como uma das maiores ameaças ao controle de infecções. O estabelecimento da infecção por esse patógeno é dependente de uma série de fatores de virulência, entre eles o pilus tipo IV (T4P), que possui papel importante na adesão a superfícies e motilidade do tipo twitching, essenciais para a colonização do hospedeiro. Uma das moléculas importantes na diferenciação entre as formas séssil e planctônica de P. aeruginosa é o segundo mensageiro bis-(3,5)-di-guanosina monofosfato cíclico (c-di-GMP), cuja síntese é feita enzimaticamente por diguanilato ciclases (DGCs). DgcP é uma DGC localizada nos polos da célula, que tem sua atividade de síntese de c-di-GMP aumentada na presença da proteína FimV, essencial para a montagem do T4P em P. aeruginosa. Neste trabalho, ensaios de microscopia de fluorescência, organização e expressão gênica foram realizados com o objetivo de aumentar a compreensão sobre o papel de DgcP em relação a sua expressão e aos fatores que regulam o T4P de P. aeruginosa. A proteína DgcP em fusão com mNeonGreen no C-terminal, expressa a partir do locus cromossômico, se localiza de maneira predominantemente bipolar tanto na linhagem selvagem quanto nos mutantes ΔpilA, ΔpilR e ΔchpA, evidenciando que seu padrão de localização não depende dos sistemas de regulação Pil-Chp e PilS-PilR. Ensaios de RT-PCRmostraram que dgcP se encontra em operon com PA14_72430 e dsbA1, indicando um papel celular conjunto entre esses genes, até o momento, desconhecido. Por fim, ensaios de qRT-PCR revelaram que os níveis de mRNA de dgcP são invariáveis nas linhagens WT, ΔpilA, ΔpilR, ΔchpA e ΔfimV, cultivadas em meio líquido ou meio sólido. Os resultados aqui mostrados, combinados com trabalhos prévios do nosso e de outros grupos, sugerem que DgcP é uma diguanilato ciclase responsável por geração constante de c-di-GMP nos polos da célula, possivelmente, atuando na sinalização local dependente do dinucleotídeo cíclico, cuja localização e atividade não são dependentes dos sistemas de regulação que atuam sobre o T4P. The Gram-negative bacterium Pseudomonas aeruginosa is an opportunistic pathogen often associated with severe burn victims or individuals with cystic fibrosis, which carbapenem-resistant isolates were classified by th World Health Organization classified one of the greatest threats to infection control. The establishment of infection by this pathogen is dependent on a series of virulence factors, including the type IV pilus (T4P), which plays an important role in adhesion to surfaces and twitching motility, essential features for host colonization. Bis-(3\',5\')-cyclic dimeric guanosine monophosphate (c-di-GMP) is a second messenger that involved in processes of biofilm formation, motility, and virulence. The diguanylate cyclase DgcP synthetizes cdi-GMP and it is located at the cell poles, and its activity depends on the scaffold protein FimV, essential for T4P assembly in P. aeruginosa. By increasing c-di-GMP levels, DgcP decreases flagellum-dependent motility and increases biofilm formation. In this work, fluorescence microscopy, gene organization and expression assays were performed to understand the whether DgcP localization and expression are under the control of T4P regulatory proteins. Fluorescence microscopy analysis showed that DgcP localizes predominantly at both cell poles in ΔpilA, ΔpilR, and ΔchpA mutants, showing that its localization pattern does not depend on the Pil-Chp and PilS-PilR systems. Furthermore, RT-PCR assays showed that dgcP is found in an operon with PA14_72430 and dsbA1, indicating an unknown putative related cellular role for these genes. Finally, qRT-PCR assays indicated that DgcP expression is invariant in ΔpilA, ΔpilR, ΔchpA, and ΔfimV mutants, either in liquid or solid medium. The results shownhere, combined with previous work by ours and other groups, suggest that DgcP is a diguanylate cyclase responsible for constant generation of c-di-GMP at the cell poles, possibly acting in local signaling dependent on the cyclic dinucleotide, but that is not under the control of the known T4P regulatory systems.

Estima-se que 80% da população sexualmente ativa será acometida por pelo menos um subtipo de papilomavírus humano (HPV) em algum momento da vida, o que representa uma alta prevalência e incidência dessa infecção sexualmente transmissível (IST), principalmente em países subdesenvolvidos. O HPV é responsável pelas Neoplasias Intraepiteliais Cervicais (NIC) de alto grau e do câncer cervical, que constituem um problema relevante para a saúde pública global, cursando com elevado ônus psicossocial e econômico aos pacientes, familiares e aos sistemas de saúde. Entretanto, a progressão ao câncer cervical pode ser evitada pela administração de vacinas terapêuticas que utilizam as principais oncoproteínas responsáveis pelo seu desenvolvimento, na tentativa de desencadear uma resposta imunológica mais específica e eficaz contra essa patologia. Atualmente, os tratamentos disponíveis para as neoplasias estão associados a morbidades para a paciente, relacionadas principalmente com o perfil reprodutivo e a recorrência da lesão, demonstrando a necessidade de terapias seguras e efetivas. O objetivo desse estudo foi sintetizar e avaliar criticamente as evidências científicas oriundas de estudos experimentais sobre a segurança, eficácia, e imunogenicidade de vacinas terapêuticas no tratamento de pacientes com NIC de alto grau associadas ao HPV. Realizou-se uma revisão sistemática de ensaios clínicos guiada pelas recomendac?o?es da Cochrane Collaboration. A estratégia de busca foi realizada nas bases de dados eletrônicas MEDLINE, Embase, CENTRAL Cochrane, Web of Science, LILACS e Scopus, sem restrição de data e de idioma. Obteve-se a aprovação/publicação do resgistro do protocolo desta revisão na International Prospective Register of Systematic Reviews-PROSPERO/NHS sob o número CRD42017077428. Os desfechos primários incluíram a segurança, eficácia e imunogenicidade das vacinas terapêuticas testadas em pacientes com NIC 2/3. Para a avaliação metodológica dos estudos, utilizamos a ferramenta de Risco-de-Viés da Cochrane para ensaios clínicos randomizados (ECR) e a ferramenta ROBINS-I (Risk-of-Bias In Non- Randomized Studies) para avaliação dos ensaios clínicos não randomizados. Foram identificados 1.184 estudos, sendo 15 selecionados e analisados. Verificou-se que, a confiabilidade dos resultados pode ser questionada, principalmente, pelo alto risco de vie?s observado em 80% dos estudos avaliados pela ferramenta da Cochrane e em 90,9% dos ensaios avaliados pela ROBINS-I, evidenciado principalmente pelos viéses de seleção e de performance presentes nos estudos. De modo geral, as vacinas terapêuticas mostraram-se heterogêneas quanto à sua formulação, dose, protocolo de intervenção e vias de administração. Na maioria dos estudos (93,3%) as vacinas apresentaramse seguras e bem toleradas, com eficácia clínica observada em 93,3% à partir da regressão das lesões, regressão histopatológica e/ou depuração viral. Adicionalmente, 73,3% dos estudos apresentaram respostas imunológicas favoráveis contra o HPV e 54,5% apresentaram correlação positiva entre imunogenicidade e resposta clínica, indicando resultados promissores que merecem maior investigação. Vacinas terapêuticas, apesar de urgentes, ainda apresentam barreiras a serem superadas antes de estarem disponíveis para a população, necessitando de maiores investimentos em ECR de fase III bem delineados de modo a beneficiar pacientes com NICs de alto grau, evitando assim sua progressão para o câncer cervical, de modo a melhorar os desfechos em saúde com impacto positivo nas taxas de sobrevida e na qualidade de vida dessas pacientes It is estimated that 80% of the sexually active population will be affected by at least one human papillomavirus (HPV) subtype at some moment in their lives, representing a high prevalence and incidence of this sexually transmitted infection (STI), especially in underdeveloped countries. HPV is responsible for high-grade Cervical Intraepithelial Neoplasms (CIN) and cervical cancer, which are a relevant problem for global public health, with a high psychosocial and economic burden in patients, families and health systems. However, progression to cervical cancer can be avoided by the administration of therapeutic vaccines that use the major oncoproteins responsible for its development, in attempts to trigger a more specific and effective immune response against this disorder. Currently, the available treatments for neoplasias are associated with morbidities for the patient, which are mainly related to the reproductive profile and the recurrence of the lesion, demonstrating the need for safe and effective therapies. The purpose of this study was to synthesize and critically evaluate the scientific evidence from experimental studies on the safety, efficacy, and immunogenicity of therapeutic vaccines in the treatment of patients with high-grade CIN associated with HPV. A systematic review of clinical trials guided by Cochrane Collaboration recommendations was carried out. The search strategy was performed in the electronic databases MEDLINE, Embase, Cochrane CENTRAL, Web of Science, LILACS and Scopus, with neither date nor language restriction. Approval for this review protocol was obtained from the International Prospective Register of Systematic Reviews-PROSPERO/NHS under the number CRD42017077428. Primary outcomes such as safety, efficacy, and immunogenicity of therapeutic vaccines tested in patients with CIN 2/3 were included. For the critical appraisal of the studies, we have used the Cochrane Risk-of-Bias tool for randomized clinical trials (RCTs) and the Risk-of-Bias In Non-Randomized Studies tool for assessment of non-randomized clinical trials. A total of 1,184 studies were identified, 15 of which were selected and analyzed. It was verified that the reliability of the results can be questioned mainly by the high risk-of-bias observed in 80% of the studies evaluated by the Cochrane tool, and in 90.9% of the trials assessed by ROBINS-I, as evidenced mainly by the selection bias as well as performance bias present within the studies. Overall, the therapeutic vaccines were heterogeneous with regards to their formulation, dose, intervention protocol and routes of administration. In most of studies (93.3%), the vaccines were safe and well tolerated, with clinical efficacy observed in 93.3% with regards to lesion regression, histopathological regression, and or viral clearance. In addition, 73.3% of the studies showed favorable immunological responses against HPV and 54.5% showed a positive correlation between immunogenicity and clinical response, indicating promising results that might be further investigated. Therapeutic vaccines, although urgent, still present barriers to be overcome before they are available to the population, requiring greater investments in welldesigned phase III RCTs to benefit patients with high-grade CIN, preventing their progression to cervical cancer disease in order to improve health outcomes with a positive impact on patients\' survival rates and quality of life

Apesar de todo o avanço científico nas últimas décadas, a doença de Chagas ainda é um importante problema em saúde pública. A expectativa de vida no Brasil tem aumentado a cada ano e esse crescimento permite o surgimento de uma população idosa em risco de contrair essa doença. Diante disso, os objetivos deste trabalho foram avaliar a resposta imune em ratos Wistar senescentes, infectados pela cepa Y de T. cruzi. O presente estudo mostrou alterações na imunidade inata, quando avaliamos a população de macrófagos peritoneais, bem como a sinalização (expressão de RT1B) e da molécula coestimulatória (CD80) das células APC nos animais senis. A imunidade celular também foi reduzida, uma vez que os animais senis apresentaram uma diminuição significativa na porcentagem de linfócitos T CD4+ e T CD8+, quando comaprados aos jovens. Além disso, foi observado que o envelhecimento reduz a expressão da principal molécula de sinalização, o CD28+, responsável pela ativação de células T. Nos nossos estudos, o envelhecimento foi associado a um aumento no percentual de linfócitos CD4+CD25lowFOXP3+, que representam as células T regulatórias instáveis. Por outro lado, os animais senis não infectados apresentaram uma redução significante no percentual de linfócitos CD4+CD25highFOXP3+, as células T regulatórias estáveis. A senesescência, bem como, o processo infeccioso ocasionaram um aumento nos níveis de corticosterona e no percentual de esplenócitos em apoptose inicial e tardia em animais senis infectados. O envelhecimento também regulou negativamente a transição DN1-DN2, bem como, as subpopulações de timócitos DN3 e DN4 em animais senis, infectados ou não. Observou-se uma redução no percentual de timócitos viáveis nos animais senis, tanto controles quanto infectados. Com relação aos parâmetros do estresse oxidativo, a senilidade elevou os níveis de TBARS, reduziu a atividade da principal enzima antioxidante SOD e na concentração de 8-isoprostano. O substrato da enzima glutationa peroxidase GSH aumentou nesses animais senis, comparados aos jovens. A análise da produção de citocinas evidenciou um aumento nos níveis de TGF-? em animais senis. No 16° dia após a infecção, um aumento nos níveis de IL-12 foram observados apenas nos animais senis infectados. Entretanto, este mesmo grupo apresentou uma redução na produção de IL-2 no 9º dia após a infecção. As concentrações de IL-17 foram maiores nos animais senis infectados e controles, quando comparados aos animais jovens. Tanto a infecção por T. cruzi quanto o envelhecimento ocasionaram um aumento nos níveis de TNF-? nos animais jovens e senis infectados. Com o presente estudo, conseguimos avançar na compreensão dos mecanismos celulares e moleculares envolvidos nas modificações da imunidade ao longo da vida e seus reflexos frente à doença de Chagas. Inúmeras alterações foram observadas, incluindo resposta imune ineficiente, aumento do estresse oxidativo, bem como diminuição da atividade de enzimas antioxidantes e comprometimento no processo de maturação celular intratímica, os quais afetam etapas essenciais no desenvolvimento dos timócitos e exportação de células T para a periferia. Foi observado um desequilíbrio funcional do eixo neuroendócrino, bem como a diminuição da viabilidade celular. Além disso, as mudanças no perfil da produção de citocinas promoveram implicações diretas na competência imunológica desses animais. Despite all the scientific advances in the last decades, Chagas disease is still an important public health problem. Every year, the life expectancy in Brazil has increased and this growth tops with the emergence of an elderly population at risk of contracting this disease. Therefore, the objectives of this study were to evaluate the immune response in senescent Wistar rats, infected with T. cruzi Y strain. Changes in innate immunity was observed in the population of peritoneal macrophages, as well as signaling (RT1B expression) and the costimulatory molecule (CD80) of APC cells in senile animals. Cellular immunity was also reduced, since senile animals showed a significant decrease in the percentage of CD4+ T and CD8+T lymphocytes when compared to young counterparts. A reduced expression of the major signaling molecule, CD28+, responsible for T cell activation was also observed as long as animals age. Aging was associated with an increase in the percentage of CD25+ CD25lowFOXP3+ lymphocytes, which represent T regulator cells. On the other hand, the uninfected senile animals presented a significant reduction in the percentage of CD4+ CD25highFOXP3+ lymphocytes. Senescence as well as the infectious process triggered an increase in corticosterone levels and in the percentage of splenocytes in early and late apoptosis in senile infected animals. Aging also negatively regulated the DN1-DN2 transition, as well as the subpopulations of thymocytes DN3 and DN4 in senile animals, infected or not. A reduction in the percentage of viable thymocytes was observed in control and infected senile animals. Regarding to oxidative stress, aging increased the levels of TBARS, reduced the activity of the main antioxidant enzyme SOD and the concentration of 8-isoprostane. Oppositely, aged animals displayed increased levels of the enzyme glutathione peroxidase GSH as compared to the young ones. The analysis of cytokine production evidenced an increase in TGF-? levels in senile animals. On the 16th day after infection, enhanced concentrations of IL-12 was observed only in senile infected animals. However, this same group showed a reduction in IL-2 production on the 9th day after infection. Senile and control animals displayed higher IL-17 concentrations when compared to young animals. Both T. cruzi infection and aging caused an increase in TNF-? levels in young and senile infected animals. The present study allowed us to better understand the cellular and molecular mechanisms involved in the immune response throughout life and its actions on Chagas\' disease. For that, several alterations were observed such as an inefficient immune response, increased oxidative stress, decreased antioxidant enzyme activity and impairment in the intratymic cell maturation process, impairing thymocyte development and the export of T cells to the periphery, as well decreased cell viability. A functional imbalance of the neuroendocrine axis was also observed, as well as changes in the profile of cytokine production directly impairing the immune competence in these animals.

O câncer de mama é um dos tumores malignos mais comuns e que afeta um grande número de mulheres da população mundial. O processo inflamatório gerado juntamente com o crescimento descontrolado das células tumorais promove um estresse metabólico no microambiente tumoral levando ao acúmulo de adenosina extracelular decorrente da hipóxia tecidual. A adenosina gerada e seus efeitos mediados via receptor A2A (A2AR) interfere em vários subtipos celulares. Neste trabalho, avaliamos o papel do receptor A2A na indução de uma resposta de células Th17 durante o carcinoma mamário experimental, visto que estas desempenham um importante papel no crescimento e na progressão do tumor de mama invasivo. Para isso, utilizamos o modelo de carcinoma mamário murino que desenvolve (4T-1) e o que não desenvolve metástase (67NR). Nossos resultados mostraram que o tumor mamário 4T-1 apresenta alta expressão do receptor A2A comparado com o tumor 67NR. A deficiência do receptor A2a preveniu o crescimento do tumor mamário 4T- 1 e de colônias de células tumorais em sítios secundários da doença, concomitante com a diminuição da resposta de perfil Th17 e do recrutamento de neutrófilos para o sitio primário. Ainda, que as células tumorais 4T-1 apresentaram uma alta expressão dos receptores de adenosina e das ectonucleotidases (CD73 e CD39), sugerindo que a via de sinalização de adenosina exerce um efeito direto nessas células. A administração de adenosina ou AMP (trifosfato de adenosina) em culturas de células tumorais 4T-1 induziu xvi um aumento da expressão de IL-6, CCL20 e CXCL1, mediadores importantes para o recrutamento de linfócitos T produtores de IL-17 e de neutrófilos. Além disso, durante o crescimento do tumor 4T-1, observamos uma alta expressão da enzima ciclooxigenase 2 (Cox-2) comparado com o tumor 67NR. No entanto, os animais deficientes geneticamente do receptor A2A apresentaram uma redução significativa da expressão de Cox-2 no microambiente tumoral comparados com os animais BALB/c. A inibição da enzima Cox-2 com indometacina ou celecoxicib (inibidor seletivo) em animais com tumor 4T-1 preveniu o crescimento do tumor primário e, consequentemente, resultou na redução da expressão de moléculas relacionadas como o perfil de resposta de células Th17 tais como IL-6, CCL20, IL-17A e Ror?t. Esses resultados indicam que o bloqueio da via de sinalização do receptor A2A interfere na indução de resposta de células Th17, abrindo novas perspectivas para o desenvolvimento de terapias alternativas para o controle de tumores invasivos Breast cancer is one of the most common and aggressive malignant tumors, affecting a large number of women in the worldwide population. The inflammatory process generated along with the uncontrolled growth of tumor cells promotes metabolic stress in the tumor microenvironment leading to the accumulation of extracellular adenosine due to the generation of tissue hypoxia. The generated adenosine and its effects mediated by A2A (A2AR) interfere in several cell subtypes. In this study, we evaluated the role of A2A receptor in the induction of a Th17 cell response in experimental breast carcinoma, given that these cells play an important role in invasive breast tumor growth and progression. For this, we employed the murine mammary metastatic (4T-1) and nonmetastatic carcinoma (67NR) model. Our results showed that there is high expression of adenosine A2A receptor in breast tumor 4T-1 compared to 67NR. The A2AR deficiency prevented the growth of metastatic breast tumor 4T-1 and tumor cell colonies in secondary disease sites, together with a decrease in Th17 response profile and in the neutrophils recruitment to the primary site. The tumor cells 4T-1 presented high expression of adenosine receptors and ectonucleotidases (CD73 and CD39), suggesting that the adenosine signaling pathway has a direct effect on these cells. Adenosine or AMP (adenosine triphosphate) administration in tumoral 4T-1 cell cultures induced an increase in IL-6, CCL20 and CXCL1 expression, important mediators in the recruitiment of IL-17 producing T lymphocytes and neutrophils. Besides that, during 4T-1 tumor growth we observed high expression of cyclooxygenase 2 (Cox-2) compared to 67RN tumor. However A2A deficient mice showed a significative reduction in Cox-2 expression in tumoral microenvironment compared to BALB/c mice. Cox-2 inhibition with indometacine or celecoxib (selective inhibitor) in mice with 4T-1 tumor prevented primary tumor growth and, consequently, resulted in reduction of the expression of molecules related to the Th17 profile, as IL-6, CCL20, IL-17A and Ror?t. These results indicate that the blockade of the A2A signaling pathway interfere on the induction of Th17 cells response, opening new perspectives for the development of alternative therapies to the control of invasive tumors

O sistema imune do hospedeiro acometido pela doença de Chagas tem sido alvo de importantes pesquisas que podem favorecer avanços no entendimento dos mecanismos patogênicos relacionados à doença. No presente estudo, investigamos possíveis ações benéficas provenientes de efeitos anti-inflamatórios e antioxidantes da associação terapêutica (melatonina/meloxicam) sobre a cardiomiopatia chagásica crônica. Foram analisadas citocinas e outros mediadores envolvidos no processo inflamatório característico da infecção. Além disso, níveis plasmáticos de Creatina Fosfoquinase MB (CK-MB), número de focos inflamatórios no tecido cardíaco e análise do peso relativo do coração foram avaliados como fatores preditivos da redução dos danos no miocárdio após os diferentes tratamentos. Como modelo experimental, foram utilizados ratos wistar infectados pela cepa Y de T. cruzi e tratados ou não com melatonina, meloxicam, ou associação de ambos. As células inflamatórias e o parasitismo tecidual cardíaco foram avaliados por meio de técnica histológica. A técnica ELISA foi utilizada para detecção e quantificação de citocinas pró-inflamatórias e modulatórias no miocárdio e nos soros dos animais experimentais. Também foram investigados os níveis de nitrito produzidos por macrófagos e por células cardíacas dos grupos experimentais pela reação de Griess. Os parâmetros foram investigados durante a fase: crônica (90, 120 e 180) dias após a infecção. O estudo poderá contribuir para um melhor entendimento das respostas imunológicas anti-T. cruzi, bem como da patogenia da disfunção cardíaca chagásica crônica. The host immune system affected by Chagas disease has been the subject of important research that can promote advances in the understanding the pathogenic mechanisms related to the disease. In the present study, it was investigated the possible beneficial actions of the anti-inflammatory and antioxidant combination therapy (melatonin / meloxicam) on chronic Chagas cardiomyopathy. Some cytokines and other mediators involved in the inflammatory process characteristic of infection were analyzed. In addition, plasma levels of creatine phosphokinase MB (CK-MB), number of inflammatory foci in the cardiac tissue and analysis of the relative weight of the heart were evaluated as predictors of damage reduction in the myocardium after different treatments. As experimental model, we used Wistar rats infected with the Y strain of T. cruzi. Some groups were untreated and others treated with melatonin, meloxicam, or combination of both. Inflammatory cells and heart tissue parasitism were evaluated by histological technique. Some pro-inflammatory and modulatory cytokines were quantified by ELISA technique in the myocardium and serum from experimental animals. The production of nitrite by macrophages and cardiac cells of the experimental groups was also investigated through Griess reaction. The parameters were observed during the chronic stage: (90, 120, and 180) days after infection. The study may contribute to a better understanding of the anti-T. cruzi immunological responses, as well as the pathogenesis of chronic Chagas heart dysfunction.

O câncer de mama é o segundo tipo de câncer mais frequente no mundo e o mais comum entre as mulheres. Agentes estressores afetam significativamente a gênese da doença. O estresse crônico suprime facetas da resposta imune como a atividade das células natural killer (NK) e a proliferação de células T, as quais exercem um papel importante na resposta imune contra o câncer, pois lisam as células tumorais e atuam no combate e monitoramento do crescimento desordenado das mesmas. É concebível que o manejo do estresse possa reduzir a progressão do câncer e melhorar a qualidade de vida de pacientes com a doença. Várias modalidades de redução de estresse tem se mostrado promissoras no tratamento oncológico, como as terapias complementares. O presente estudo teve como objetivo identificar e analisar as evidências disponíveis na literatura científica sobre a utilização de terapias complementares como estratégia de redução do estresse e sua influência no sistema imune de mulheres com câncer de mama. Realizou-se uma revisão integrativa da literatura, utilizando como referencial teórico a Prática Baseada em Evidências. Foram utilizadas cinco bases de dados: LILACS, EMBASE, CINAHL, PubMed e Biblioteca Cochrane. Os critérios de inclusão foram artigos disponíveis na íntegra, em inglês, espanhol ou português e publicados nos últimos 20 anos (1981-2011). Para a localização dos artigos foram cruzados os descritores controlados e não controlados selecionados com base nos vocabulários MeSH e DeCS. Foram encontrados 2612 artigos nas bases de dados. Após a leitura do título e do resumo e a exclusão dos artigos duplicados, foram pré-selecionados 29 estudos. Foi realizada a leitura na íntegra desses artigos, sendo que a amostra final foi dessa revisão foi constituída de 12 artigos. Em relação ao período de publicação, 2 artigos (16%) foram publicados nos anos de 1997 e 1998 e 10 artigos (84%) foram publicados a partir do ano de 2001. Quanto à base de dados em que os estudos foram encontrados, 5 estudos (65%) foram encontrados no PUBMED, 6 (35%) na EMBASE. Outro estudo (8,3%) encontrava-se na base de dados CINAHL e estava repetido nas bases EMBASE e PUBMED. Yoga, toque terapêutico, meditação, reflexologia, massagem e sessões de relaxamento são as principais terapias complementares utilizadas por mulheres com câncer de mama. As principais razões para a adesão a esse novo tipo de tratamento são o alívio de sintomas e diminuição da toxicidade do tratamento convencional, prevenção de recorrência da doença e melhora significativa do sistema imunológico. Melhora significativa da depressão, fadiga, ansiedade, qualidade de vida, diminuição dos níveis de cortisol, aumento da citotoxicidade de células NK e da proliferação de linfócitos T foram detectados após a utilização das terapias complementares, sendo a massagem terapêutica a terapia mais utilizada pelas pacientes. Tais resultados evidenciam a utilização da terapia complementar como uma potente estratégia balizadora da promoção de saúde e tratamento do câncer de mama. Breast cancer is the second most common type of cancer worldwide and the most common cancer among women. Stressors significantly affect the genesis of the disease. Chronic stress suppresses facets of the immune response such as the activity of natural killer cells (NK) and T-cell proliferation. These cells play an important role in the immune response against cancer because they lyse tumor cells and monitor and combat their disordered growth. It is conceivable that the management of stress may reduce the progression of cancer and improve the quality of life of patients with the disease. Several methods of stress reduction have shown promise in treating cancer, including complementary and alternative medicine. This study aimed to identify and analyze the evidence in the literature on the use of complementary and alternative medicine as a strategy for reducing stress and its influence on the immune system of women with breast cancer. We performed an integrative literature review, using the evidence-based practice as the theoretical framework. We used five databases: LILACS, EMBASE, CINAHL, PubMed and the Cochrane Library. Inclusion criteria were full-text articles available in English, Spanish or Portuguese and published in the last 20 years (1981-2011). Controlled and uncontrolled descriptors selected from Mesh and DeCS were crossed for location of the articles, and 2612 articles were found in the databases. After reading the title and abstract, duplicate articles were excluded and 29 studies were selected. After reading the entire articles, the final sample of this review consisted of 12 articles. Concerning the period of publication, 2 articles (16%) were published in 1997 and 1998 and 10 articles (84%) were published from 2001. Regarding the databases where the studies were found, five studies (65%) were found in PUBMED, 6 (35%) in EMBASE. Another study (8.3%) was found in CINAHL and was repeated in EMBASE and PUBMED databases. Yoga, therapeutic touch, meditation, reflexology, massage and relaxation sessions are the main complementary and alternative medicine used by women with breast cancer. The main reasons for choosing this new type of treatment are the relief of symptoms and decrease of toxicity of the conventional treatment, prevention of disease recurrence and significant improvement of the immune system. Significant improvement in depression, fatigue, anxiety, quality of life, decreased levels of cortisol, increased cytotoxicity of NK cells and T lymphocyte proliferation were detected after the use of complementary and alternative medicine, and therapeutic massage therapy was used by most patients. These results highlight the use of complementary and alternative medicine as a potent strategy for health promotion and treatment of breast cancer.

A identificação de moléculas com potenciais ações terapêuticas e a compreensão dos mecanismos envolvidos na modulação da resposta imune frente à administração de substâncias farmacologicamente ativas em modelos experimentais infectados por Trypanosoma cruzi tem contribuído de maneira importante nas investigações de novas terapias para a doença de Chagas. Estudos prévios demonstraram as ações do zinco e da melatonina sobre a imunidade do hospedeiro, durante a fase aguda da infecção chagásica experimental, potencializando a resposta imune gerada contra o parasita. O presente estudo teve como objetivo avaliar o possível efeito imunomodulador da administração de zinco e melatonina em ratos Wistar machos durante a fase crônica da infecção por T. cruzi, utilizando os seguintes parâmetros: dosagens de citocinas IL-2, IL-4, IL-10, IFN-, e TNF-, produção de óxido nítrico, proliferação de esplenócitos, citometria de fluxo para análise fenotípica das subpopulações de células T CD3+CD4+ e CD3+CD8+, células dendríticas, células NK (CD161+) e NKT (CD3+CD161+), macrófagos, moléculas co-estimuladoras CD28, CD80 e CD86, RT1B, análise dos marcadores CD11a e CD25 e detecção do processo de apoptose celular. O efeito imuno-modulador do zinco e da melatonina durante a fase crônica da infecção chagásica pode ser comprovado através redução no percentual de macrófagos, células dendríticas, linfócitos TCD4+ e TCD8+, apoptose celular, das concentrações de óxido nítrico, IFN- e MCP-1. Adicionalmente, a terapia com zinco e melatonina durante a infecção experimental resultou em modificações importantes na resposta imunológica, como evidenciado pelo aumento no percentual de células T reguladoras, da proliferação celular e dos níveis das citocinas IL-2, IL-4 e IL-10. O desenvolvimento de novas estratégias terapêuticas, que possam modular a resposta inflamatória, contribuindo para prevenção dos danos teciduais observados na fase crônica da doença, são alvos importantes no tratamento da doença de Chagas. The identification of molecules with potential therapeutic actions and the comprehension of the mechanisms involved in modulating the immune response by the administration of pharmacologically active substances in experimental models infected with Trypanosoma cruzi have contributed significantly in the investigation of new therapies for Chagas\' disease. Previous studies have demonstrated the actions of zinc and melatonin on host\'s immunity during the acute phase of experimental Chagas\' disease, enhancing the immune response generated against the parasite. The present study aimed to evaluate the possible immunomodulatory effect of zinc and melatonin administration in male Wistar rats during the chronic phase of T. cruzi infection, using the following parameters: measurement of IL-2, IL-4, IL-10, IFN- and TNF-, nitric oxide, proliferation of splenocytes, flow cytometry for phenotypic analysis of CD3+CD4+ and CD3+CD8+ T cells subpopulations, dendritic cells, NK cells (CD161+) and NK T (CD3+CD161+), macrophages, co-stimulatory molecules CD28, CD80 and CD86, RT1B, analysis of markers CD11a and CD25 and detection of the apoptosis process. The immunomodulator effect of zinc and melatonin during the chronic phase of Chagas\' disease was evaluated by a reduction of the percentage of macrophages, dendritic cells, CD4+ and CD8+ lymphocytes, cellular apoptosis, the concentrations of nitric oxide, IFN- and MCP-1. In addition, the treatment with zinc and melatonin during the experimental infection resulted in significant changes in the immune response, as evidenced by the increased percentage of regulatory T cells, cell proliferation and cytokine levels of IL- 2, IL-4 and IL-10. The development of new therapeutic strategies that can modulate the inflammatory response, contributing to prevention of tissue damage observed in the chronic phase, are important targets in the treatment of Chagas disease.

O desenvolvimento de novas estratégias terapêuticas ao melanoma é de particular importância devido à sua baixa resposta aos tratamentos tradicionais. No presente trabalho, utilizamos modelo de melanoma murino experimental para estudarmos os efeitos da fosfoetanolamina (PEA) sintética sobre o desenvolvimento deste tumor. Nossos resultados demonstram que o fosfomonoéster apresentou efeito inibidor da proliferação de células da linhagem B16F10 in vitro, induzindo apoptose após estimulação por 24 a 72h. In vivo, o tratamento (via oral) de animais portadores de melanoma com diferentes doses de PEA (10, 20 e 40mg/Kg), durante 10 ou 20 dias consecutivos, resultou em volumes tumorais pelo menos 70% menores que o de animais controle e diferenças macroscópicas consideráveis. PEA induziu, de maneira dose-dependente, aumento da apoptose e diminuição da proliferação de células tumorais. O tratamento resultou em alterações hematológicas como aumento do número de plaquetas, eritrócitos e leucócitos. Dentre os leucócitos, observou-se uma maior proporção de linfócitos e monócitos após 10 e 20 dias de tratamento, respectivamente. Em adição, PEA induziu uma maior produção da citocina pró-inflamatória IL-6 e das citocinas anti-inflamatórias IL-10 e TGF- e menores níveis da citocina pró-inflamatória IFN-. Os níveis de IL-1, IL-12p70 e IL-17 não foram alterados com o tratamento. Nossos resultados demonstram um papel inibidor da PEA sobre a progressão do melanoma, contribuindo para um melhor entendimento de sua atividade anti-tumoral. The low responsiveness of melanoma to traditional treatments together with its increasing incidence makes the development of new therapeutic strategies against this type of cancer extremely important. In this study, we used a murine melanoma model to evaluate the effects of synthetic phosphoethanolamine (PEA) on the development of this tumor. In vitro, PEA had an inhibitory effect on the proliferation of B16F10 cells, inducing apoptosis after 24 to 72h stimulation. In vivo, oral treatment of melanoma-bearing animals with different doses of PEA (10, 20 e 40mg/Kg) during 10 or 20 consecutive days resulted in reduced tumor volumes (at least 70% compared to the control) and in expressive macroscopic differences. PEA also induced a dose-dependent increase of apoptosis and decrease in tumor cell proliferation. The treatment also resulted in hematological changes, such as increased numbers of platelets, erythrocytes and leukocytes. Among leukocytes, we observed a higher proportion of lymphocytes and monocytes after 10 and 20 days of treatment, respectively. In addition, PEA induced higher levels of the pro-inflammatory cytokine IL-6 and of the anti-inflammatory cytokines IL-10 and TGF-, and it also induced a lower production of the pro-inflammatory cytokine IFN-. No differences were observed in the levels of IL-1, TNF-, IL-12p70 and IL-17 upon treatment. Our results demonstrate an inhibitory role of PEA in the development of melanoma, contributing to a better understanding of its antitumoral activity.
