This study was performed to find halophilic bacteria that have the ability to increase salinity resistance in wheat seedlings, so that any type of wheat cultivar can be grown in saline areas by inoculation with these bacteria without the need for genetic manipulation.A number of highly saline farm soils from several areas around Tabriz and Urmia Lake were screened to isolate bacterial isolates. A total of six highly halophilic isolates were isolated for the study. The effect of bacteria on salinity tolerance in wheat was determined as a factorial of two factors (salinity and bacteria) based on a randomized complete block design with three replications. The effect of six bacterial isolates at three high concentrations of salt (NaCl) (0.5, 1 and 1.5%) on wheat seed growth under standard laboratory conditions for four weeks was evaluated. Two bacterial isolates that had the best performance - in most growth indices such as fresh and dry size and weight of roots and stems - were selected and their effect was assayed under greenhouse conditions. Finally, the best bacteria in terms of function were selected and identified by molecular methodsFrom the six highly halophilic bacterial isolates, two bacteria were selected for greenhouse studies, one of which, caused a 50% increase in most growth factors, was selected for identification. Introduction: Halophiles and halotolerant microorganisms are some of the extremophiles that are able to grow in medium containing sodium chloride and have adapted to life in salinity environments. Halophiles bacteria in saline soils by maintaining the food chain, decomposition of organic matter and improvement of soil structure and fertility improve soil conditions. Materials and methods: In order to isolate the halotoletant bacteria, from the halophyte rhizosphere, four desert areas in Golestan province were sampled. To check the Extremophile of isolates, their resistance was tested for resistant to salinity, drought, temperature and PH. Also, plant growth promoting traits were measured. Results: Fromforty-five strains which were isolated, three strains (G3, G6 and G14) have demonstrated the ability of resistance to 35% salt. Isolates G6 and G3 phosphate solubiliziation power of 301 and 201 ppm, respectively. Isolated G6 micrograms produced auxin 20/7 Mg/ ml. G14 and G6 grow at 50 °C, pH = 10 and osmotic potential -0 /7MPa. While G3 strain grows at 50 °C, pH = 7/ 5 and osmotic potential -0/49. The three strains of the bacterial genera Bacillus and Pseudomonas, respectively. Discussion and conclusion: In this study, isolates due to the growth in concentrations of salt and saturated salt tolerance of extreme environmental conditions and are likely halotolerant or halophile bacteria and its potential for use in various fields of biotechnology including biotech, industrial enzyme production and biological fertilizers for saline soil improvement.

Introduction: Excessive use of tellurite nowadays, has suffered the environment from the toxic effects of the oxyanion. Hence, biological treatment of polluted areas is considered as an environmentally friendly and inexpensive method. Although the toxic effects of tellurite for most microorganisms have been reported, but several species of the bacteria including the halophilic bacteria used in this project can overcome the toxicity of the oxyanion by its reduction to the elemental form. The aim of this study was to identify the mechanism (s) involved in tellurite detoxification. Materials and methods: In order to enhance and maintain enzymatic activity during purification, the test conditions and enzyme production by the strain were optimized. The optimization was done by One Factor at A Time (OFAT) method. Several factors, including: time, various percentages of inoculum, range of pH, concentration of tellurite and various salts effects were optimized. For assurance tellurite removal was examined during the experiment. The enzyme was purified by various methods, including ammonium sulphate precipitation and hydrophobic interaction column chromatography in which the Concentration of 1.4 M saturated ammonium sulphate was applied. The purity of the enzyme was assessed by SDS-PAGE during each phase. Results: The optimum conditions obtained showed that at 30 hours, 3% inoculum, pH 7.5, without tellurite and with 5% NaCl the highest enzyme activity and tellurite removal are observed. Purification of the enzyme greatly reduced the concentration of unrelated proteins and caused a concentrated band which could be one subunit of the enzyme targeted. The partially purified enzyme’s fraction was shown to have nitrate and selenite reductase activity other than tellurite reductase activity. Discussion and conclusion: This study is an approach to the identification of the halophilic microorganisms Physiology and enzymes involved in restoring tellurite. The production of the enzyme responsible for this phenomenon has been optimized and partially purified.

Gomishan wetland is a natural ecosystem located in 35 km north west of Gorgan, in the west vicinity of Khajeh Nafas city and Gomishan. Twice sampling from 3 different geographic positions in dry and rainy seasons, led to the isolation of 224 isolates.
