This paper analyzes the example of the publishers Oldenbourg and the publication of the 'Illustrierte Technische Wörterbücher' (ITW), a multilingual dictionary which competed with a similar project that the Verein Deutscher Ingenieure (VDI) had already initiated. Its willingness to challenge the vast and well-funded VDI with this project--and to tackle the immense difficulties associated with the dictionary itself, which pioneered the standardization of technical terminology across six languages--indicates that for Oldenbourg, this project was of paramount importance to its reputation. The decision to take the challenge and publish the ITW was the starting point of Alfred Schlomann's career as well as the foundation of Oldenbourgs reputation as a premier publishing house and pioneered the standardization of technical terminology. The example shows that the publishers' decisions had been of paramount importance not only for scientists' careers and as gatekeeper of the scientific community but can also influence the development of science.

Gegenstand dieser Arbeit waren Untersuchungen von Einflüssen von Ambruticin VS-3, Fludioxonil und Jerangolid A auf Candida albicans. Aus Untersuchungen an filamentösen Pilzen war bekannt, dass es sich bei dem Target von Ambruticin VS-3 und Fludioxonil um eine Histidinkinase Typ III handelt, die in den Signalweg der osmotischen Stressantwort involviert ist. Jerangolid A kam zum Einsatz, da es eine hohe strukturelle Ähnlichkeit zu Ambruticin VS-3 besitzt, so dass das gleiche Target vermutet wurde. In dieser Arbeit sollte überprüft werden, ob in dem humanpathogenen Hefepilz C. albicans das Target der Substanzen ebenfalls die Histidinkinase Typ III Nik1/Cos1 ist. Unter osmotischen Stressbedingungen wird der HOG-Signalweg aktiviert, der unter anderem die Glycerinbiosynthese induziert. Die Behandlung von C. albicans mit den Testsubstanzen führte, genauso wie osmotische Stressbedingungen, zur Erhöhung der Glycerinkonzentration. Darüber hinaus wurde eine Phosphorylierung der MAPK Hog1 bei den Wildstämmen nach Substanzexposition nachgewiesen. Daraus lässt sich schließen, dass durch die Testsubstanzen in C. albicans der HOG-Signalweg aktiviert wird. Bei allen Untersuchungen erwies sich die COS1-Deletionsmutante als resistent gegenüber den Substanzen, so dass angenommen werden kann, dass diese Histidinkinase Typ III auch in C. albicans das Target der Testsubstanzen ist. Um den Einfluss von Ambruticin VS-3 auf C. albicans genauer zu verstehen, wurden Genexpressionsanalysen durchgeführt. Gene, kodierend für Proteine aus dem HOG-Signalweg und aus der Zellwandbiosynthese, wurden verstärkt exprimiert, wohingegen Gene aus dem Hyphenwachstum herunterreguliert wurden. Der Einsatz von Ambruticin VS-3 in einem in-vitro-Infektionsmodell bewirkte eine Steigerung der Phagozytoseaktivität von mit C. albicans-infizierten Makrophagen. Das lässt vermuten, dass Ambruticin VS-3 die Zellwandbiosynthese in C. albicans sekundär beeinflusst., In this thesis, the influences of ambruticin VS-3, fludioxonil and jerangolid A on the pathogenic yeast Candida albicans were investigated. The target of ambruticin VS-3 and fludioxonil is known to be a type III histidine kinase, which is involved in osmotic stress signalling. This knowledge based on analyses performed with filamentous fungi. However, the function of the histidine kinase is still not well understood. The human pathogen C. albicans was chosen as model system, which possess Nik1/Cos1 as type III histidine kinase and the role of Nik1/Cos1 was investigated as a target. Under osmotic stress conditions the so-called HOG-pathway is activated, which leads to an induction of the glycerol biosynthesis. The treatment with the substances led to an increase of external glycerol of the wild type strains. Further analyses of the phosphorylation status of Hog1 confirmed activation by treatment with ambruticin VS-3 and fludioxonil. Analysis of gene expression showed an upregulation of genes related to the HOG-pathway as well as to cell wall biosynthesis, whereas hyphae-associated genes were downregulated. Cell wall biosynthesis and hyphal growth are indicators of virulence. Due to the evidence of ambruticin influences on the pathogenicity of the yeast, the compounds were tested in in vitro infection models. The internalization of the yeasts by murine macrophages was enhanced when the yeast was pre-incubated with the substance over night. This suggests that ambruticin VS-3 induces changes in the cell wall assembly of the yeast which are recognized by the macrophages. In all investigations, the COS1 deletion mutant was resistant to treatments of ambruticin VS-3, jerangolid A and fludioxonil which implies the histidine kinase Nik1/Cos1 as the target of the selected fungicides.

A brief historical overview of physical chemistry at the University of Geneva as well as a description of the present research activities at the department of physical chemistry are presented.

Im Ergebnis der in den Jahren 2006 und 2007 an der TU Freiberg durchgeführten Gärversuche stand die Forderung nach weiteren Untersuchungen der in der Praxis vorherrschenden Mischsubstrate auf ihre grundsätzliche Vergärbarkeit, Gasproduktion und Gaszusammensetzung. So wurden Ganzpflanzensilage (GPS) Triticale, Kleegras-, Luzernegrassilage, Maissilage, Zuckerrübe, Gehaltsrübe, kaltgepresster Rapskuchen oder fusarienbelasteter Triticaleschrot als Cofermente zu Rindergülle oder Hühnerkot eingesetzt. GPS Triticale und Zuckerrüben lieferten als Monosubstrate keine stabile Vergärung und damit keine auswertbaren Ergebnisse. Spitzenwerte in der Methangasproduktion erzielten Rindergülle in Kombination mit GPS Triticale oder Zuckerrüben. Ebenfalls hohe Methangasausbeuten erzielte die Kombination von Maissilage und Hühnertrockenkot mit Rindergülle. Bei der Cofermentation von mykotoxinbelastetem Getreide ergab sich kein negativer Einfluss auf die Prozessstabilität. Die Zugabe von Enzymen als Betriebshilfsstoff bewirkte eine geringfügige Steigerung der Methangasproduktion. Für die Prozessoptimierung und damit optimale Auslastung der Biogasanlage sollten sprunghafte Veränderungen der Inputzusammensetzung vermieden werden.

Im Rahmen des vom Sächsischen Landesamt für Umwelt, Landwirtschaft und Geologie (vormals: Landesanstalt für Landwirtschaft) geförderten Forschungsprojektes „Ermittlung von Gaserträgen mykotoxinbelasteter Getreidearten für die Co- und Monovergärung“ wurden am Standort Freiberg in der vorhandenen Laborbiogasanlage Fermentationsversuche mit verschiedenen landwirtschaftlichen Substraten durchgeführt, um dadurch wissenschaftlich fundierte Aussagen zum Cofermentationsprozess zu gewinnen. Bei den Versuchen sollte geprüft werden, ob Mykotoxine im Getreide die Gaserträge reduzieren bzw. beeinflussen und ob die Mykotoxinbelastung während der Vergärung abgebaut wird. Das giftige Stoffwechselprodukt von Fusarienpilzen im Getreide ist Mykotoxin. Eine Probe mit fusarienbefallener Triticale wurde vom Landesamt für Umwelt, Landwirtschaft und Geologie (vormals Landesanstalt für Landwirtschaft) zur Verfügung gestellt. Im Rahmen dieses Projektes wurden Versuche mit Triticale, mit fusarienbelasteter Triticale und Ganzpflanzensilage (GPS)-Triticale durchgeführt. Bei Versuchen zur Cofermentation diente Rindergülle als Basissubstrat. Neben der kontinuierlichen Vergärung werden die Substrate zusätzlich in einem diskontinuierlichen Verfahren untersucht.

Recent applications of one-electron equations for embedded electron density introduced originally for multi-level modeling of solvated molecules (T.A. Wesolowski, A. Warshel, J. Phys. Chem. 1993, 97, 8050) are reviewed. The considered applications concern properties directly related to the electronic structure of molecules (or an atom) in condensed phase such as: i) localized electronic excitations in a chromophore involved in a hydrogen-bonded intermolecular complex; ii) UV/Vis spectra of acetone in water; and iii) energy levels of f-orbitals for lanthanide cations in a crystalline environment. For each case studied, the embedding potential is represented graphically and its qualitative features are discussed.

Over the past 12 years, new immunosuppressive agents, better knowledge of anesthesiology and postoperative reanimation as well as refinement in surgical technics modified the indications for and results of orthotopic liver transplantation (OLT). At the beginning of the OLT era, liver tumors that could only be removed by total hepatectomy were one of the most frequent indications. Nowadays, this indication is mostly abandoned in view of a high rate of recurrence and poor long-term results. In contrast, the prognosis of fulminant hepatic failure has been dramatically improved by OLT, once efficient organization systems allowed adequate organ supply and emergency transplantation within a few hours. Three main groups of diseases (cholestatic diseases, inborn errors of metabolism and parenchymal diseases) can be treated by OLT with excellent results (actuarial survivals of 80 to 90% have been reported at one and 2 years). Later graft dysfunction is rare except for virus B recurrence. Primary biliary cirrhosis and the group of inborn errors of metabolism are regarded as the optimal indications of OLT in adults and children respectively. Precocious evaluation of patients, before advanced stages of the disease associated with multiple complications, should prevent them from dying on a waiting list and decrease operative as well as early postoperative risks. Not only does OLT provide mere survival (among 5 patients with lethal hepatic disease, 4 are alive at 2 years from OLT), it also provides a regained quality of life with a virtually normal (for the price of a daily medication intake) family, professional and sportive life. Such achievements prompt us today to propose early transplantation to these patients.
