Nature is based on self-assembling systems that generate functional structures. Inspired by nature, programmable artificial architectures have been developed, often making use of synthetic peptides and nucleic acids. In contrast, only limited examples of programmable carbohydrate assemblies have been reported. This can be partially ascribed to the limited knowledge of carbohydrates’ structure. Structural complexity, heterogeneity of natural carbohydrate samples, and the lack of suitable analytic techniques have prevented the molecular level description of carbohydrate materials. The introduction of synthetic model systems, able to generate chemically defined assemblies, could help the understanding of carbohydrate materials. Here, simple and well-defined oligosaccharides were employed to create model systems to study supramolecular carbohydrate-based assemblies and produce useful data for the formation of tailor-made materials. This approach also shined light on the interactions involved in the formation of natural systems, such as bacterial biofilms, where carbohydrate and peptides interact to form valuable nanocomposites. In chapter 2, I investigated the supramolecular structure formation of synthetic oligosaccharides. Systematic variation in their chain length, substitution pattern, and glycosidic linkages, generated distinctive morphologies, including spherical particles or fiber-like structures. The compounds showed unique intrinsic optical properties (e.g. red edge excitation shift), highly dependent on their aggregation status. Potential applications of glycomaterials in bioimaging and optical devices are envisioned. Among these six compounds, disaccharide 13 (13-D in chapter 3), growing into needle-like structures, offered the perfect model system to explore details of oligosaccharide assemblies and optimize analytical techniques to study carbohydrate materials. The stability of the assembly enabled the implementation of microcrystal electron diffraction (MicroED) for oligosaccharide samples. This technique allowed for the reconstruction of the crystal unit cell and permitted correlating the local molecular organization with the supramolecular assembly. Synthetic analogous of compound 13 with specific single-site modifications were designed to identify key stabilizing interactions. The combination of organic chemistry and electron diffractions methods will be implemented to reveal molecular details of natural polysaccharide assemblies. In chapter 4, a new model system was introduced to study bacterial biofilms, nanocomposites of cellulose and proteins (e.g. curli fibers). Specific E. Coli strains produce phosphoethanolamine (pEtN) cellulose as part of their protective biofilms, providing increased adhesion. I employed synthetic peptides and oligosaccharides to generate artificial biofilm and study the role of pEtN cellulose in biofilm formation. Different amounts and patterns of pEtN substitution in the oligosaccharide modulated the length and aggregation tendency of the peptide fibers. The mechanical properties of the protein-carbohydrate network were affected by the chemical nature of the carbohydrate component, with high adhesion measured for highly substituted pEtN cellulose analogues. Synthetic oligosaccharides able to interrupt fibrillary assembly were identified and could serve as promising drug candidates for the treatment of neurological diseases or as antibacterial agents. Overall, the synthetic oligosaccharide models presented in this thesis will establish the foundation of our understanding of carbohydrate interactions in nature and will promote several applications of carbohydrate materials in nanobiotechnology.

Ebola virus and Marburg virus make up the family Filoviridae. Due to their singlestranded RNA genome in negative orientation, they belong to the order Mononegavirales. Filoviruses cause a severe hemorragic fever in humans and non-human primates with fatality rates up to 90%. Because of the severe illness, and the fact that there is no specific treatment or vaccination, they are classified as BSL-4 pathogen. The Nucleocapsid is built by the non-segmented RNA-genome, the nucleoprotein NP, the viral polymerase L, the polymerase-cofactor VP35 and VP30. Subject of this study is a coiled coil domain within NP and its influence on various functions of NP. First the role of the coiled coil domain in the interaction of NP with VP30 and VP35 was investigated by immunofluorescence analysis. NP-mutants without coiled coil 3D-structure or without coiled coil domain were still able to interact with VP35, but not with VP30. This could be due to two binding domains on NP for VP35. Binding of VP30 to NP is either mediated directly by the coiled coil or requires a 3D confirmation determined by the NP coiled coil. Furthermore, this study is focussing on the influence of the coiled coil domain on NPNP interaction and self assembly of NP. By immunofluorescence analysis it could be proofed that the formation of NP inclusion bodies (inclusions) depends on a functional coiled coil. Knock-out mutants could still be recruited to inclusions build by NPwt, suggesting that the loss of the coiled coil domain only causes a partly loss of the ability for self-interaction. With an complementation assay it could be shown that the coiled coil domain is sufficient to mediate NP-NP interactions. NP forms helices that are supposed to be the framework of the nucleocapsids. Analysis of NP-helices by electron microscopy suggested that the ability to form helices also depends on an intact coiled coil domain or the presence of NPwt. NP mutants alone were not able to assemble to helices. Oligomerization mediated by the coiled coil domain or an interaction with RNA influenced by this domain could be reasons for these findings. The last part of the study is dedicated to the influence of the NP coiled coil domain on viral transcription. In an iVLP assay a dominant negative effect of NP with deleted or mutated coiled coil domain could be observed. This in probably due to a defect of NP with deleted or mutated coiled coil in encapsidation the virus sepecific minigenome that serves as template for the viral polymerase complex which is also indicated by the altered NP helices formation., Das Ebolavirus (EBOV) bildet mit dem Marburgvirus die Familie der Filoviren, die aufgrund ihres einzelsträngigen, negativ orientierten RNA-Genoms der Ordnung der Mononegavirales zugeteilt werden. Filoviren verursachen beim Menschen und bei nicht-menschlichen Primaten ein schweres hämorrhagisches Fieber mit einer Mortalitätsrate bis zu 90%. Deshalb und weil es bisher keine spezifische Therapie und keine Impfung gibt, werden Filoviren als Pathogene der Sicherheitsstufe 4 eingestuft. Der Nukleokapsidkomplex des EBOV besteht aus dem nicht-segmentierten RNAGenom sowie dem Nukleokapsidprotein NP, der viralen Polymerase L, dem Polymerase-Ko-Faktor VP35 sowie VP30 besteht. Gegenstand dieser Arbeit ist eine Coiled-Coil-Domäne innerhalb des NP, deren Einfluss auf verschiedene Funktionen des NP untersucht werden sollte. Hierfür wurde die Rolle der Coiled-Coil bei der Interaktion mit VP30 und VP35 analysiert. NPMutanten, die entweder keine Coiled-Coil-3D-Struktur oder gar keine Coiled-Coil mehr aufwiesen, konnten hier zwar noch mit VP35, aber nicht mehr mit VP30 interagieren. Die Gründe hierfür sind vermutlich, dass VP35 mehrere Bindungsstellen besitzt und VP30 entweder im Bereich der Coiled-Coilbindet oder die durch die Coiled-Coil vermittelte Konformation des NP benötigt. Weiterhin befasst sich diese Arbeit mit der Rolle der Coiled-Coil-Domäne bei der NPNP- Interaktion und dem self assembly. Immunfluoreszenzanalysen ergaben, dass die Bildung von NP-Einschlusskörpern, so genannter inclusions, von der Coiled-Coil- Domäne abhängt. Coiled-Coil-defiziente Mutanten können aber noch durch NPwt in inclusions rekrutiert werden, die Fähigkeit zur Selbstinteraktion ist also nur eingeschränkt. Mittels eines Complementation Assays konnte gezeigt werden, dass die Coiled-Coil-Domäne allein ausreichend für eine NP-NP Bindung sein kann. Immunelektronenmikroskopischen Analysen konnten zudem zeigen, dass die Bildung von NP-Helices, welche das Gerüst der Nukleokapside darstellen, ebenfalls nur bei intakter Coiled-Coil-Domäne oder in Gegenwart von NPwt möglich ist. Eine Rolle könnte hierbei entweder eine durch die Coiled-Coil-Domäne vermittelte Oligomerisierung des NP oder RNA-Interaktion spielen, welche zur Bildung von Helices wichtig ist. Im letzten Teil der Arbeit wurde der Einfluss Coiled-Coil-Domäne auf die virale Transkription untersucht. Hierbei zeigte sich im iVLP-Assay, dass eine nicht intakte Coiled-Coil einen dominant negativen Effekt auf die Transkription hat. Dies ist vermutlich dadurch bedingt, dass ohne intakte Coiled-Coil keine NP-Helices gebildet werden können und so auch keine regulären Nukleokapside, welche für die Polymerase als Matrize zur Transkription und Replikation dienen.

Das gitterfehlangepaßte Kristallwachstum führt unter bestimmten Bedingungen zu einem 3-D Wachstumsmodus, der oft Stranski-Krastanow-Wachstum genannt wird. Resultierende Strukturgrößen liegen in der Größenordnung 10 nm und die Halbleiterstrukturen besitzen daher Quanteneigenschaften. Sie stehen im Fokus grundlagenwissenschaftlichen Interesses, da künstliche Atome und Dimensionalitätseffekte an ihnen untersucht werden können. Auch von der Anwendungsseite wächst das Interesse, da niederdimensionale Strukturen hoher Kristallqualität und mit hoher gestalterischer Freiheit geschaffen werden können. In dieser Arbeit wurden Mischhalbleiter-Heterostrukturen der Dimensionalität d= 0,1 und 2 mittels Gasphasen-MBE hergestellt. Ziel war eine Korrelation der strukturellen mit den elektronischen und optischen Eigenschaften. Selbstformierende Quantendrähte und Quantenpunkte in leitfähigen Kanälen wurden in ihrem Einfluß auf den lateralen Transport untersucht. Weiterhin wird dargestellt, wie zusätzliche, durch Quantenpunkte induzierte Oberflächenzustände eine deutliche Verschiebung der Energie des Oberflächen-Ferminiveau-Pinning einer (100)-GaAs-Oberfläche verursachen. Der senkrechte Elektronentransport durch Quantenpunkte dient der Untersuchung von Dot-induzierten, tiefen elektronischen Zuständen und der Erklärung eines eindimensionalen Modells elektronischer Kopplung zwischen denselben. Zusätzlich führen uns die Ergebnisse optischer Messungen zu einem besseren Verständnis des Vorgangs der Dotformierung und der elektronischen Kopplung zwischen zufällig verteilten Quantenpunkten. The lattice-mismatched epitaxial growth is known to induce a three-dimensional growth mode often referred to as Stranski-Krastanov growth. The resulting structures have typical sizes of 10 nm and possess quantum properties, which are of fundamental physical interest, since artificial atoms and dimensionality effects can be studied. There is a growing interest from an applicational point of view also, since low dimensional structures of a high crystal quality and of a high degree of designerabillity can be created. In this work such structures of a dimensionality d=0,1 and 2 based on compound semiconductors have been designed and prepared by molecular beam epitaxy to perform comparative studies with respect to their electronic, structural and optical properties. Self assembled quantum wires and dots in conductive channels have been examined according to their influence on lateral electrical transport. It is demonstrated how additional surface states from quantum dots cause a distinct shift in the Surface Fermi-level of a GaAs (100) surface. Vertical transport through dots is used to support a model of one-dimensional coupling between deep states induced by the dots. Additionally, optical investigations let us attain a better understanding of the process of dot formation and the electronic coupling between the randomly distributed dots.
