Ongoing climate change and the related ocean warming as well as ocean acidification represent a major challenge for marine organisms. Marine fish are very efficient osmotic regulators, however, the details of the underlying compensation mechanisms remain to be explored, such as the interaction with elevated temperature. In the present study, effects on marine fish from ocean acidification and warming were investigated in long-term incubation experiments on Atlantic cod (Gadus morhua). Isolated perfused gill experiments were conducted to examine in vivo costs of the branchial energy budget and acid-base relevant ion transport under hypercapnia at optimum temperature (10 degree celsius) and in the warmth (18 degree celsius). In addition, potential effects on whole animal level as well as maximum enzyme capacities of central metabolic pathways in liver and muscle tissue were investigated. In conclusion, Gadus morhua possesses a wide range of compensational mechanisms on different systemic levels to cope with the challenge of ocean acidification, whereas high temperature has been confirmed as limiting factor.

Die Konformationszustände von Proteinen sind eng mit deren Protonierungszuständen verknüpft. Im Rahmen der statistischen Mechanik wurde eine neue Methode zur Berechnung der korrelierten Besetzungen von Protonierungs- und Konformationszuständen formuliert. Insbesondere wurden dabei entropische Beiträge zu Dissoziationsreaktionen berücksichtigt. Mögliche Reorientierungen von dipolaren oder geladenen Seitenketten wurden explizit modelliert. Dadurch wird auch die Stabilisierung von unterschiedlichen Protonierungszuständen durch lokale Relaxationen der Proteinstruktur beschrieben. Die entwickelte Methode wurde erfolgreich zur Untersuchung der Fluktuationen im Grundzustand sowie der Relaxationen nach Lichtanregung im Grün Fluoreszierenden Protein angewendet. Die Kenntnis der Verteilung von Protonierungszuständen ist essentiell für das Verständnis der Photophysik dieses Proteins The conformational states of proteins are closely connected to their protonation states. A method to calculate the correlated population of protonation and conformational substates was developed, based on the concepts of statistical mechanics. The entropic contributions to dissoziation reactions were especially considered. Possible reorientations of dipolar or charged sidechains were modelled explicitly. Therefore, the stabilization of protonation states, resulting from the local relaxation of the protein sructure, is described. The method developed was succesfully applicated to the investigation of fluctuations in the groundstate and light induced relaxation of the Green Fluorescent Protein. Knowledge on the distribution of protonation states is important to be able to understand the photophysics of this protein.

Untersucht wurde ob ein neuer Pulskontur Algorithmus des Gerätes Picco (Medical Systems, München) der, die individuelle Compliance der Aorta mit in die Berechnungen des HZV mit einbezieht dem bisherigen Algorithmus, des gleichen Gerätes überlegen ist. Dazu wurden auf der herzchirurgischen Intensivstation H3A im Universitätsklinikum Großhadern 47 Patienten, die sich herzchirurgischen Eingriffen unterzogen hatten, gemessen. Als Referenz Methode zur Ermittlung des HZV wurde ein Pulmonaliskatheter verwendet, den die Patienten aus anderer Indikation bekommen hatten, von dem behandelnden Arzt der nicht an der Studie beteiligt war. Nachdem das Gerät kalibriert worden war wurden stündlich drei sukzessive Einzelmessungen durchgeführt. Hierbei wurden die Pulskurven im Meßzeitraum aufgezeichnet und im nachhinein in ein Picco das mit dem neuen Algorithmus ausgestattet war eingespielt. So konnten die gleichen hämodynamischen Bedingungen simuliert werden. Es wurden 287 Einzelmessungen durchgeführt und ausgewertet. Als Statistische Methoden wurden die Regressionsanalyse und der Bland Altmann Blot [Bland, 1986 #3]durchgeführt. Bei PCCO alt gegen COpa war R² = 0,634 (r=0,796), bei PCCO neu gegen COpa R² = 0,7681 (r=0,8764). Beim Bland Altmann Blot: PCCO alt vs. COpa Bias = 0,01 l/min, 2SD = 2,25l/min; PCCO neu vs. COpa Bias = 0,25 l/min, 2SD = 1,3 l/min. Mit der Vorliegenden Arbeit konnte gezeigt werden das der neue Algorithmus dem alten Überlegen ist und seine Verwendung empfohlen werden kann. Außerdem konnte gezeigt werden das eine dem Pulmonaliskatheter in Genauigkeit ebenbürtige Methode existiert, die ihm allerdings aufgrund der geringeren Invasivität überlegen ist. Es werde weitere prospektive Studien im verschiedenen Bereichen empfohlen, insbesondere bei Patienten mit hohen Schwankungen der Parameter HZV, SVR und MAP. Objective of this survey was to compare a new algorithm for pulse contour measurement, which additionally considers the individual compliance of the aorta for calculating the cardiac output (CO), with the previously used algorithm for the same pulsion continuous cardiac output (PiCCO) device (Medical Systems, Munich, Germany). Measurements were carried out on the surgical intensive care unit H3A of the Munich University Hospital Großhadern in 47 patients that had undergone cardiac surgery. Reference measurements of CO were obtained by standard thermodilution technique using the pulmonary artery catheter all patients had received for a different indication by their doctor in attendance, who was not involved in the study. After calibrating the PiCCO device, three successive single measurements were carried out every hour for six consecutive hours. Pulse waves were recorded and later transferred to another PiCCO device, which was equipped with the new algorithm. This procedure enabled us to simulate identical hemodynamic conditions on both devices. A total of 287 single measurements was acquired and analyzed. Statistical methodes: regression analysis and Bland Altmann Blot [Bland, 1986 #3] For PCCO old vs. COpa was R² = 0,634 (r=0,796), for PCCO new vs. COpa R² = 0,7681 (r=0,8764). We conclude that the new algorithm for PiCCO is superior to the previous one and it´s application can be recommended. Furthermore we were able to demonstrate that PiCCO is at least equivalent to standard thermodilution technique with regard to accuracy, with the advantage of being less invasive. We suggest further prospective studies in order to confirm these findings for various groups of patients, especially those with high fluctuations of CO, SVR and MAP.
