EINLEITUNG. Ischämisch-neovaskuläre Retinopathien (u.a. Frühgeborenenretinopathie oder diabetische Retinopathie) sind die häufigste Erblindungsursache in Industrienationen und stellen somit eine Herausforderung für die Augenheilkunde dar. Der Einsatz von anti-VEGF-A-Antikörpern hat die Therapie revolutioniert, jedoch profitieren nicht alle Patienten davon. Interessanterweise reduziert eine Makrophagendepletion im Modell der Sauerstoff-induzierten Retinopathie (einem Tiermodell der Frühgeborenenretinopathie) pathologische retinale Neovaskularisation. Daher wurde bisher das von Makrophagen synthetisierte VEGF-A als pathophysiologisch relevante Quelle pro-angiogener Stimuli vermutet. Ziel dieser Arbeit war, mit Hilfe zellspezifischer Mausmutanten und in vitro Methoden die Rolle myeloider Zellen im Mausmodell der Sauerstoff-induzierten Retinopathie zu untersuchen, um Konzepte für selektivere therapeutische Eingriffsmöglichkeiten aufzuzeigen. METHODEN. Mäuse, die selektiv für VEGF-A in Zellen der myeloiden Zellreihe defizent sind, konnten durch Kreuzung von VEGFfl/fl- mit LysMCre-Mäusen auf einem genetischen C57BL/6J-Hintergrund erzeugt werden. Sowohl unbehandelte, als auch durch Clodronat-Liposomen makrophagendepletierte VEGF-A-defiziente LysMCre Mäuse wurden im Vergleich zu Geschwisterkontrolltieren im Modell der Sauerstoff-induzierten Retinopathie untersucht. Die Analyse der Netzhäute der Tiere erfolgte an den postnatalen Tagen 14 und 17 mittels quantitativer Real-Time PCR, immunhistochemischer Verfahren und Durchflusszytometrie. Parallel wurde in vitro die Menge des VEGF-A Transkripts in MIO-M1 Zellen, einer humanen Müller-Zelllinie, in alleiniger Kultur und in Ko-Kultur mit der murinen Mikrogliazelllinie BV-2 überprüft. ERGEBNISSE. Übereinstimmend mit der Literatur zeigte sich an systemisch makrophagendepletierten Mäusen im Modell der Sauerstoff-induzierten Retinopathie eine Reduktion der avaskulären und neovaskulären Fläche, die jedoch keine statistische Signifikanz erreichte. Parallel fielen deutliche immunhistochemische VEGF-A Signale in Müllerzellen auf. Eine durchflusszytochemische Analyse von Netzhäuten neugeborener Mäuse bestätigte Makrophagen als die bei weitem größte Gruppe von Immunzellen. Die FACS-basierte Subspezifizierung von Makrophagen und Mikroglia zeigte, dass der Anteil residentieller Mikroglia nach Sauerstoffbehandlung signifikant anstieg. Jedoch hatte die zellspezifische VEGF-A-Defizienz in myeloiden Zellen keinen Einfluss auf die relative mRNA Expression von VEGF-A in der Gesamtretina. Weder die Netzhautmorphologie, noch die Größe der avaskulären Fläche als Ausdruck einer anti-angiogenen Wirkung war in den gendefizienten Mäusen verändert. Immunhistochemisch stellten sich die Müllerzellen nach Sauerstoffexposition morphologisch aktiviert dar. Eine Zellkulturstudie zeigte, dass die relative mRNA Expression von VEGF-A in MIO-M1 Müllerzellen signifikant höher war, wenn sie in Gegenwart von BV-2 Mikrogliazellen kultiviert wurden. SCHLUSSFOLGERUNGEN. Unsere Ergebnisse aus dem Modell der Sauerstoff-indu-zierten Retinopathie legen nahe, dass nicht das aus myeloiden Zellen sezernierte VEGF-A pathologische retinale Neovaskularisation stimuliert. Aufgrund unserer in vitro erhobenen Befunde vermuten wir vielmehr einen indirekten Mechanismus. Wir postulieren, dass Makrophagen über die Freisetzung von Mediatoren die VEGF-A Sekretion aus Müllerzellen induzieren., INTRODUCTION. Ischemic neovascular retinopathies (e.g. retinopathy of prematurity or diabetic retinopathy) still challenge the ophthalmic community as they represent the leading cause for blindness in industrialized countries. While the application of anti-VEGF-A-directed antibodies has revolutionized the treatment regime, some patients do not benefit due to a lack of response. Interestingly, depletion of macrophages reduces pathological neovascularization in the model of oxygen-induced retinopathy (an animal model for retinopathy of prematurity). Since, VEGF-A expression by macrophages has been considered to be the pathophysiologically relevant source of VEGF-A. The aim of this study was to analyze the relevance of myeloid cells in oxygen-induced retinopathy by using cell-specific mouse mutants and in vitro methods to reveal new therapeutical strategies. METHODS. Mice that selectively lack VEGF-A-expression in myeloid cells were generated by crossbreeding VEGFfl/fl and LysMCre mice on a C57BL/6J background. Knockout mice with or without macrophage-depletion through systemic administration of clodronate-liposomes were analyzed in the model of oxygen-induced retinopathy. Their retinas were analyzed on postnatal days 14 and 17 by quantitative Real-Time PCR, immunohistochemistry, and flow cytometry. Furthermore, relative expression of VEGF-A was measured in an in vitro model in MIO-M1 cells, a human Müller cell line, cultured solitary and in co-culture with BV-2 cells, a murine microglia cell line. RESULTS. In accordance with the literature, we found that macrophage-depleted mice in the mouse model of oxygen-induced retinopathy showed a reduced avascular and neovascular area which did not reach statistical significance in our studies. However, striking were the concomitantly prominent VEGF-A signals in immunohistochemical stainings of Müller cells. Flow cytometric analyses in retinas of newborn mice confirmed that macrophages represent by far the most dominant group of immune cells. FACS-based differentiation of macrophages and microglia showed that the relative number of residential microglia significantly increased after oxygen exposure. However, the myeloid-specific knockout of VEGF-A did not affect relative VEGF-A mRNA expression, retinal morphology or the size of the avascular area. Müller cells were morphologically activated in immunohistochemical stainings following oxygen-treatment. Relative VEGF-A mRNA expression in MIO-M1 cells was significantly higher in co-culture with BV-2 cells than alone. CONCLUSIONS. Our data obtained from the model of oxygen-induced retinopathy suggest that myeloid cells do not stimulate retinal neovascularization through direct VEGF-A secretion. Instead, our in vitro data rather indicate that myeloid cells indirectly acitvate retinal VEGF-A via Müller cells. Thus we hypothesize that macrophages stimulate Müller cells to secrete VEGF-A through the release of soluble mediators.

Im Rahmen der vorliegenden Dissertation wurden die Rolle von myeloiden Zellen und die Rolle des Zytokins Thrombopoetin in der bakteriellen Meningitis untersucht. Wesentliche Befunde wurden in einem Mausmodell der bakteriellen Meningitis erhoben. Mit Hilfe dieses Modells können Mechanismen der Inflammation auf zellulärer und Zytokinebene sowie der neuronale Schaden untersucht werden. In Abhängigkeit von der Fragestellung wurden zur Auslösung der Meningitis lebende Pneumokokken, das synthetische bakterielle Lipopeptid Pam3CysSK4 (ein spezifischer Agonist am Toll-Like-Rezeptor 2) oder aktivierte Spenderleukozyten verwendet. In Untersuchungen an Wildtyp-Mäusen und Mäusen mit homozygoter Ablation des Thrombopoietin-Rezeptors (c-Mpl-/-) zeigte sich 24 h nach der Meningitis-Induktion mit Streptokokkus pneumoniae in den c-Mpl-/--Mäusen eine geringere Dichte apoptotischer Zellkerne in Körnerzellschicht des Gyrus dentatus als bei Wildtyp-Mäusen. Die zusätzliche intrathekale Applikation von rekombinanten Thrombopoietin (TPO) führte in infizierten Wildtyp-Mäusen zu einer Zunahme apoptotischer Neurone im Gyrus dentatus. Diese Befunde deuten auf eine proapoptotische Wirkung von TPO während der bakteriellen Meningitis hin. Ergänzende Zellkulturuntersuchungen an primärer ZNS-Mikroglia und einer Makrophagen-Zelllinie deuteten auf eine wichtige Rolle der bakteriellen Toxine Pneumolysin (Pln) und Wasserstoffperoxid (H2O2) bei der Induktion von TPO in diesen Zellen hin. RAG-1-defiziente Mäuse, denen reife T- und B-Lymphozyten fehlen, zeigten im Vergleich zu Wildtyp-Mäusen 24 h nach der Meningitis-Induktion mit Pam3CysSK4, schwerere klinische Krankheitszeichen und eine verstärkte neutrophile Liquorpleozytose, jedoch einen geringer ausgeprägten neuronalen Gewebeschaden. Eine differentielle Regulierung pro- und antiinflammatorischer Zytokine im Hirngewebe und in den Leukozyten im Subarachnoidalraum deutete auf Kompartiment-spezifische Effekte der Lymphozyten während akuter Meningitis hin, darunter eine Abschwächung der meningealen Inflammation und die Förderung neurotoxischer Eigenschaften aktivierter Mikroglia. Des Weiteren wurde eine Methodik entwickelt, mit der die direkte Aktivierung immunkompetenter Zellen im Subarachnoidalraum durch bakterielle Liganden auf spezifische Zellpopulationen limitiert werden kann. Das Verfahren beruhte auf der Aufreinigung der jeweiligen Zellpopulation aus Wildtyp-Spendermäusen mittels geeigneter Antikörpermarkierung und Isolation nach dem MACS-Prinzip. Nach Stimulation mit dem spezifischen TLR2-Agonisten Pam3CysSK4 wurden die Zellen in den Subarachnoidalraum von TLR2-defizienten Mäusen eingebracht. Direkte proinflammatorische Wirkungen des Liganden im Empfänger waren auf diese Weise ausgeschlossen. Nach Methodenetablierung wurden erste Pilotversuche mit aufgereinigten Splenozytenfraktionen durchgeführt. Im Vergleich zu unstimulierten Zellen lösten mit Pam3CysSK4 stimulierte Ly6G-positiv selektierte Neutrophile keine wirtsseitige Inflammationsreaktion aus, während ein robuster Einstrom von Leukozyten des Empfängertiers durch die negativ selektierte Population oder die wiedervereinigten Fraktionen ausgelöst wurde. Diese präliminären Befunde legen nahe, dass monozytäre Zellen und nicht neutrophile Granulozyten im Subarachoidalraum eine Wächterfunktion gegenüber invadierenden grampositiven Bakterien ausüben., The present paper reports on investigations of the role of myeloid cells and of the cytokine thrombopoietin in bacterial meningitis. Central results were gathered in a mouse model of bacterial meningitis which allows to investigate mechanisms of inflammation at the cellular and cytokine level as well as neuronal damage. Depending on the question, meningitis was induced using live pneumococci, the synthetic bacterial lipopeptide Pam3CysSK4 (a specific toll- like receptor 2 agonist), or stimulated splenocyte fractions. In experiments on wild-type mice and mice with homozygous ablation of the thrombopoietin receptor (c-Mpl-/-), a lower density of apoptotic neurons in the granular cell layer of the dentate gyrus 24 hours after meningitis induction with Streptococcus pneumoniae was found in the c-Mpl-/- mice when compared to wild- type mice. Intrathecal application of recombinant thrombopoietin (TPO) led to an increase in appoptotic neurons in the dentate gyrus in infected wild-type mice. These findings suggested a proapoptotic effect of TPO during bacterial meningitis. Further studies on primary CNS microglia and a macrophage cell line revealed an important role of the bacterial toxins pneumolysin (Pln) and hydrogen peroxide (H2O2) in inducing TPO expression by these cells. RAG-1-deficient mice lacking mature T and B lymphocytes showed more severe clinical impairment and increased neutrophil concentrations in the cerebrospinal fluid (CSF) compared to wild-type mice 24 h after meningitis induction with Pam3CysSK4. However, neuronal tissue damage was less pronounced. A differential regulation of pro- and anti-inflammatory cytokines in brain tissue and in the leukocytes in the subarachnoid space indicated a compartment-specific effect during acute meningitis. Specifically, an attenuation of meningeal inflammation and the promotion of neurotoxic properties of activated microglia were observed. Furthermore, a methodology was developed to limit the direct activation of immunocompetent cells by bacterial ligands to specific leukocyte populations in the subarachnoid space. The method is based on the purification of the respective cell population from wild-type donors, using magnetic cell sorting (MACS) after appropriate immunolabeling of the target cells. After stimulation with the specific TLR2 agonist Pam3CysSK4, the cells were injected into the subarachnoid space of TLR2-deficient mice, thereby excluding direct proinflammatory effects of the ligand in the recipient. After establishing the method, pilot experiments were conducted using purified splenocyte fractions. Compared to unstimulated cells, Pam3CysSK4-stimulated Ly6G positive-selected neutrophils did not induce an inflammation reaction. Conversely, a robust inflow of host leukocytes was triggered by the negatively selected population or the combined fractions. These preliminary findings suggest that monocytic cells rather than neutrophil granulocytes exert a sentinel function against invading gram-positive bacteria in the subarachoid space.

Oncogene-induced senescence causes hepatocytes to secrete cytokines, which induce their immune-mediated clearance to prevent tumor initiation, a process termed "senescence surveillance." However, senescent hepatocytes give rise to hepatocellular carcinomas (HCCs), if the senescence program is bypassed or if senescent cells are not cleared. Here, we show context-specific roles for CCR2(+) myeloid cells in liver cancer. Senescence surveillance requires the recruitment and maturation of CCR2(+) myeloid cells, and CCR2 ablation caused outgrowth of HCC. In contrast, HCC cells block the maturation of recruited myeloid precursors, which, through NK cell inhibition, promote growth of murine HCC and worsen the prognosis and survival of human HCC patients. Thus, while senescent hepatocyte-secreted chemokines suppress liver cancer initiation, they may accelerate the growth of fully established HCC.

Melanoma is known for its rapid progression, metastasis to distant organs and therapeutic resistance. Despite high melanoma immunogenicity, the results of immunotherapeutic clinical studies are mostly unsatisfactory. One explanation is the development of strong immunosuppression mediated by highly immunosuppressive regulatory leukocytes, in particular, myeloid-derived suppressor cells (MDSCs). These cells were found to be enriched and activated in the melanoma microenvironment, inducing a profound impairment of anti-tumor immune responses and leading to the tumor progression. Therefore, understanding the mechanisms of MDSC generation, migration to the tumor site and activation as well as their targeting is important for the development of novel strategies for effective melanoma immunotherapy. We suggest that such therapeutic approaches should involve the inhibition of MDSC-mediated immunosuppressive melanoma microenvironment combined with other immunologic treatments.

The influence of age on the myelogram and on the different indices (ratios) was studied on bone marrow aspirates of 131 healthy dogs which were divided into different age groups (A [4-6 months] to G [8 years]). Myelograms were obtained by performing a differential count on 1000 cells. Age dependent differences were particularly seen in the 4 to 6 month old dogs. These dogs had significantly higher percentages of immature erythroid cells (p0.01) than dogs of the other age groups and, consequently, the lowest proportion of myeloid cells. This fact was also reflected by a lower quotient of number of myeloid cells/number of erythroid cells (M:E-ratio) and increased erythroid maturation index (I:ME-ratio). In addition, the proportions of monocytes and lymphocytes were increased and the number of plasma cells was decreased in juvenile dogs. The results in young dogs which were different from the values of adult animals reflect the increased requirements of red cell mass during the time of growth and ontogenesis of the immune system. These findings have to be considered in the interpretation of bone marrow films of young dogs.
