Abstract

The deoxyribonucleic acid (DNA) is subject of a series of modifications with time, which are connected with the development of cancer and the process of aging. However, an efficient and nearly perfect repair system works against them, which eliminates most but not all of these changes. A series of tests has been developed for assessment of genotoxic potential of compounds and mixed pollution of environment. Three fast assay systems (umu test, DIT test, and Visko test) are discussed, whose results are obtained more quickly than by using conventional methods (e.g., Ames test or sister chromatid exchange).

Abstract

The metabolite patterns of 4,4-diphenyl-N-isopropyl-cyclohexylamine-hydrochloride (pramiverine, Sistalgin) in urine, feces, bile and serum of Wistar rats, beagle dogs, rhesus monkey and man were analyzed with radio thin-layer chromatographic techniques. The structures of six pramiverine metabolites were elucidated. Pramiverine is eliminated unchanged in only minute amounts via the renal, the biliary and the fecal route. Consequently an almost quantitative absorption and metabolism takes place. Metabolite patterns in serum and urine differ considerably indicating that some of the metabolites are excreted preferentially by the kidney while others are reabsorbed to various degrees. The identified metabolites are products of dealkylation, deamination and hydroxylation reactions.

Abstract

(2-Amino-5, 6-dimethylbenzimidazolyl)-cobamide (III) is transformed to (2-hydroxy-5, 6-dimethylbenzimidazolyl) cobamide (IV) by nitrous acid. Exchange of the NH2-group by hydrogen with nitrous acid/hypophosphorous acid yields vitamin B12 (I). This reaction completes a cycle vitamin B12 (I)----[carboxy(2-cyanoamino-4,5-dimethylphenyl)amino]cobamide+ ++ (II)----(2-amino-5,6-dimethylbenzimidazolyl)cobamide (III)----vitamin B12 (I), which allows chemical 14C-labelling of vitamin B12. In this procedure cyanogen bromide, which is necessary for the first step, was labelled with [14C] cyanide. By the following reactions a vitamin B12 was formed in which C-2 of the 5, 6-dimethylbenzimidazole moiety is labelled.

Amphetamine is deaminated oxidatively by highly purified rat liver mitochondria. This monoamineoxidase (MAO) is an enzyme of the mitochondrial outer membrane and its activity is about two times higher than that of the microsomal MAO. Anaerobic incubations with mitochondria reduce phenylacetone, an important metabolite of amphetamine, to phenylisopropanol. The ketoreductase is localized in the mitochondrial inner membrane or its matrix.
