Malaria is responsible for 445 000 deaths in 2016. The emergence and the spread of resistant P. falciparum to artemisinin-based combinations is a major health problem in South-East Asia. Research must continue to find compounds with a novel mechanism of action. The apicoplast is an interesting target. It is a Plasmodium organelle derived from a secondary endosymbiosis with a red alga. Red algae could be a special source of new antiplasmodial compounds targeting the isoprenoid biosynthesis pathway in the apicoplast. This thesis focuses on bioactive secondary metabolites identification via a new analytical strategy. A computer program called Plasmodesma was created and achieves an automatic differential analysis of 1H-1H and 1H-13C 2D NMR spectra. Bioactive compounds are isolated and identified by hyphenated HPLC-SPE-NMR. The presence of different classes of compounds including sterols could explain the antiplasmodial activity in studied the red algae species., L’apparition de P. falciparum résistants aux CTAs est un problème majeur en Asie du Sud-Est. Il est nécessaire de rechercher des molécules présentant des mécanismes d’actions nouveaux. Une cible d’intérêt est l’apicoplaste, un organite de Plasmodium issu d’une endosymbiose avec une algue rouge. Les algues rouges pourraient présenter une source privilégiée de nouvelles molécules antipaludiques ciblant notamment les voies de biosynthèse apicoplastique. Cette thèse porte sur l’identification de ces métabolites secondaires bioactifs via le développement d’une nouvelle stratégie analytique. Un programme informatique nommé Plasmodesma a été créé et permet l’analyse différentielle de manière automatique de spectres RMN 2D 1H-1H et 1H-13C. Carrageenans are linear sulphated polysaccharides occuring as cell wall constituents in red algae and are used texturing agents in the food industry. The backbone of the polymer is composes of alternating α(1→3) et β (1→4)-Linked D-galactopyranose units. The structure of the repeating units differs in the degree and position of sulphate esterification. 3,6-anhydro-α-D-galactopyranoses units can also replace the α(1→3)-D-galactopyranose units can also replace the α(1→3)-D-galactopyranose units. Strucural studies of carrageenans have been conducted using carrageenases which are glycoside hydrolases producted by marine bacteria. They degrade specifically carrageenans by cleaving their β(1→4) bounds. The oligo-carrageenans obtained have been analysed by liquid chromatography and characterised by mass spectrometry and NMR. The structure and the proportions of hydrolysis products are representative of the polymer structure. These studies were performed with k- and i-carrageenases on carrageenans containing their biosynthetic precursors, the κ-/μ- an ι-/ν-carrageenans, as well as other hybrid carrageenans. Characterisation of hybrid oligosaccharides within the hydrolytic products has confirmed the heterogeneity of these strucures. Different organisation models (block, random) of the repetitive units within these heterogeneous chains have been proposed.; Les carraghénanes sont des polysaccharides linéaires sulfatés qui composent la paroi cellulaire des algues rouges et qui sont utilisés comme agents texturants dans l’industrie alimentaire. La structure des motifs disaccharidiques de répétition des carraghénanes varie par la position et le nombre des groupements sulfates. L'algue rouge, Gracilaria verrucosa, a été cultivée, durant le printemps 2009, en mode suspendu à différents emplacements et profondeurs représentant les différences biogéochimiques de la lagune de Bizerte au Nord de la Tunisie. La concentration et la distribution des nutriments (ammonium, phosphate) dans l'eau, ainsi que la salinité, la température, l’oxygène et la transparence de l’eau ont été déterminées. Les méthodes analytiques internationales standardisées ont été appliquées. Les effets du mode de culture, de l’emplacement et de la profondeur sur les rendements en poids, protéines, R-Phycoérythrine et sucres totaux de l’algue ont été étudiés. La croissance de Gracilaria a été similaire dans les différents sites mais plus lente dans les profondeurs supérieures à 1 m en raison d’une moindre intensité lumineuse. L’espèce étudiée contient des teneurs élevées en protéines (20-30%) et en R-phycoérythrine (50-85mg/g matière fraîche) et une faible teneur de carbohydrates (30%-35%). Les teneurs en protéines et R-phycoérythrine corroborent le potentiel d’utilisation de cette algue, pour l’extraction de molécules à haute valeur ajoutée et le développement d’activités innovantes. تم استزراع الطحلب الأحمر غراسيلاريا بطريقة التّدلي وذلك بعدّة مواقع وأعماق يمثلون الاختلافات البيوجيوكيمائية لبحيرة بنزرت خلال ربيع سنة 2009. تم تحديد التركيز وتوزيع المغذيات (الآزوت و الفسفور) من جهة وتحديد درجة الملوحة والحرارة ونسبة الأكسيجين الذائب بالماء وصفاء الماء من جهة أخرى، وذلك باستعمال الطّرق التحليلية الموحّدة دوليّا. خلال هذه الدراسة تم تحديد نمو الطحلب وكمية البروتينات والسكريات وصبغ .R-phycoérythrineيمكن أن نستخلص من خلال هذه الدّراسة أن نموّ طحلب الغراسيلاريا مماثل في كل المواقع. كما أن هذا النموّ في الأعماق التي لا تتجاوز المتر عن سطح الماء أقل من النموّ في الأعماق التي تتجاوز المتر وذلك لمحدودية كميّة الضوء. وكما بيّنت هذه الدّراسة أنهذا الطحلب يحتوي على نسب عالية من البروتينات (20- 30%) وصبغ R-phycoérythrine (50 – 80 مغ/غ مادّة رطبة) ونسبة منخفضة من السكريات (30 – 35 %). إنّ نسبة البروتينيات والصبغ المتحصّل عليها تجعل من طحلب الغراسيلاريا مادة خام لاستخلاص صبغ R-phycoérythrine ذات القيمة المضافة العالية. The red alga, Gracilaria verrucosa, was grown using suspended modes at different depths and sites representing the biogeochemical differences of the Bizerte lagoon in the North of Tunisia during a spring 2009. The concentration and distribution of nutrients (ammonium, phosphate) in water, as well as the salinity, temperature and dissolved oxygen were determined. During this period some parameters as growth, concentrations of proteins, carbohydrates and pigments yield were investigated in order to evaluate the influence of the culture mode, site and depth on Gracilaria production. Internationally standardized analytical methods were applied in order to reach these objectives. Similar growth was observed in the lagoon. Gracilaria grew slower at a depths superior to 1.0 m. The major limiting factor of algae growth was found to be the light intensity. The results indicated that Gracilaria verrucosa showed a high range of protein (20%-30%), R-phycoerythrin contents (50-85mg/g weigh matter ) and a low content in carbohydrate (30%-35%).The proteins and R-phycoerythrin contents corroborate the potential use of the studied algae, which constitute useful raw material for the development of R-phycoerythrin extraction activity. In his book, Life on a young planet, A.H. Knoll states that the first documented fossils of green algae date back 750 Ma. However, according to B. Teyssèdre's book, La vie invisible, they are much older. Using a method which combines paleontology and molecular phylogeny, this paper is an inquiry into the Precambrian fossils of some "acritarchs" and of a primitive clade of green algae, the Pyramimonadales. A paraphyletic group of unicellular green algae, named "Prasinophyceae", is represented at Thule (Greenland) ca. 1200 Ma by several morphotypes of the monophyletic Pyramimonadales, including Tasmanites and Pterospermella that are akin to algae still living today. These two, and others, probably had forerunners going back 1450 / 1550 Ma. Some acritarchs that may represent Pyramimonadales producing "phycomas" which split open for dehiscence were confusingly included in the polyphyletic pseudo-taxon "Leiosphaeridia" and are possibly already present at Chuanlinggou, China, ca. 1730 Ma. Many acritarchs that Timofeev obtained by acid maceration of Russian samples dated between 1800 and 2000 Ma were probably unicellular Chlorophyta which synthesized algaenans or other biopolymers resistant to acetolysis. Living Prasinophyceae are undoubtedly green algae (Viridiplantae). Thus, if Prasinophyceae fossils go back certainly to 1200 Ma, probably to 1500 Ma and possibly to 1730 Ma, then the ancestor of green algae (Chlorophyta and Streptophyta) probably separated from the ancestor of red algae (Rhodophyta) as early as 2000 Ma. more...
