Within supramolecular chemistry, host-guest chemistry aims the development of hollow molecules able to encapsulate specifically smaller substrates. Applications are ranging from detection or trapping of molecules to catalysis in confined space. Cryptophanes are a family of cage-shaped molecules containing a hydrophobic cavity able to encapsulate several interesting substrates. Thus, they are studied for the formation of MRI-bioprobes, as they can encapsulate xenon as well as for the trapping of toxic heavy metal cations, which are complexed with huge affinity. The modulation of cryptophanes encapsulation properties is generally performed by changing their cavity size, but the introduction of heteroelements in their structure remains almost unstudied, as it requires some synthetic adaptations. The work carried out in this thesis focuses on the design of new nitrogen-containing cryptophanes called tri-azacryptophanes. The synthesis of two generations of tri-azacryptophanes containing aniline groups is reported, along with their properties of xenon recognition in organic medium. They encapsulate xenon with drastically different properties than their fully oxygenated analogs. Moreover, these properties vary under pH changes, thanks to the protonation of the aniline groups. Finally, the synthesis of an amphoteric tri-azacryptophane is reported. This compound shows first encouraging results for the encapsulation of xenon in water, which is an interesting step toward applications as an MRI-bioprobe, and the presence of nitrogen atoms seems also to increase the affinity for heavy metal cations in basic water., Au sein de la chimie supramoléculaire, la chimie hôte-invité vise le développement de molécules possédant une cavité et permettant l’encapsulation spécifique de certains substrats. Les champs d’applications concernent la détection ou le piégeage de molécules, voire la catalyse en milieu confiné. Les cryptophanes sont une famille de molécules-cages, contenant une cavité hydrophobe, qui peuvent encapsuler plusieurs substrats d’intérêt. Ils sont ainsi étudiés tant pour la formation de biosondes IRM, grâce à leurs propriétés d’encapsulation du xénon, que pour le piégeage de métaux lourds toxiques, qu’ils complexent avec une forte affinité. La modulation des propriétés d’encapsulation des cryptophanes se fait essentiellement en modifiant la taille de leur cavité, mais l’introduction d’hétéroéléments dans leur structure, qui nécessite quelques ajustements synthétiques, reste peu étudiée. Les travaux réalisés pour cette thèse traitent de la conception de nouveaux dérivés azotés nommés tri-azacryptophanes. La synthèse de deux générations de tri-azacryptophanes, possédant des groupements anilines est détaillée, ainsi que leurs propriétés de reconnaissance du xénon en milieu organique. Celles-ci sont drastiquement différentes de celles de leurs analogues oxygénés, et, de plus, varient en fonction du pH, lorsque les fonctions anilines se protonnent. Enfin, la synthèse d’un tri-azacryptophane amphotère est rapportée. Ce composé montre des premiers résultats encourageants pour l’encapsulation du xénon dans l’eau, ce qui constitue une étape importante vers une application comme biosonde, tandis que la présence d’atomes d’azote donneurs favorise nettement le piégeage de cations de métaux lourds par rapport aux analogues oxygénés.
