Seleno-Yeasts (SY) used as feed additives are known to contain different Selenium (Se) species. Seleno-Yeasts has been shown, on previous analytical methods, to contain selenomethionine (SeMet), selenocysteine (SeCys), selenate (SeIV) and selenite (SeVI), and various other organic and inorganic Se forms identified but rarely quantified. A new advanced method has allowed elemental Se (Se0), an inorganic Se species, to be quantified, thereby obtaining better insight into the proportion of inorganic Se in SY products. The study aimed to quantify the Se0 in SY products and assess the proportion of inorganic Se in SY. The Se speciation of 13 fresh commercials SY from different suppliers and batches, was assayed for the total Se, inorganic Se species (SeIV, SeVI and Se0), and organic Se species (SeMet and SeCys). Results on total Se were in line with the expected Se concentrations for all evaluated samples. The proportion of Se present as Se0 ranged from 3.6% to 51.8%. The quantity of Se0 in the SY products, added to SeIV and SeVI, indicated an average proportion of inorganic Se of 14.2% for the 13 analyzed SY products. The proportion of Se as SeMet ranged from 19.0% to 71.8%, (average of 55.8%), and a large variability in the SeMet content was observed. The SeCys content was also variable, with an average of 3.8%, relative to the total Se. In conclusion, advances in the analytical characterization have revealed that SY products can have a significantly high proportion of inorganic Se, which could affect the bioavailability of Se from SY supplements and explain their variable and lower bio-efficacy than pure SeMet supplements, such as hydroxy-selenomethionine. Although iron oxide and hydroxide nanoparticles can be efficient candidates for the removal of selenium oxyanions due to their high adsorption capacity, the role of each iron species has not been fully elucidated. Furthermore, iron species are often found to be less effective for Se(VI) than Se(IV). The challenge and novelty of this study was to develop a carbon material impregnated with different iron phases, including oxides (magnetite/hematite) and hydroxides (goethite/lepidocrocite) capable of removing both Se(IV) and Se(VI). Since the phase and morphology of the iron nanoparticles play a significant role in Se adsorption, the study evaluated both characteristics by modifying the impregnation method, which is based on an oxidative hydrolysis with FeSO4 7H2O and CH3COONa, and the type of carbonaceous support (activated carbon or sucrose-based carbon foam). Impregnated activated carbons provide better removal efficiencies (70–80%) than carbon foams (<40%), due to their high surface areas and point zero charges. These results show that the adsorption of Se(VI) is more favorable on magnetic oxides (78%) and hydroxides (71%) than in hematite (<40%). In addition, the activated carbon decorated with magnetite showed a high adsorption capacity for both selenium species, even in alkaline conditions, when the sorbent surface is negatively charged. A mechanism based on the adsorption of inner-sphere complexes was suggested for Se(IV) immobilization, whereas Se(VI) removal occurred through the formation of outer-sphere complexes and redox processes. Previous studies indicated that the antioxidant and water balance capacity played important roles in improving the vase life of cut flowers. As a trace element, selenium (Se) has been proven to play an important role in extending the vase life of lily cut flowers. However, there is still no report on the effects of Se on the vase life of other cut flowers. In this study, we explored the role of inorganic selenium, named sodium selenite (Na2SeO3), in improving the vase life of Eustoma grandiflorum cut flowers. Compared with the control (distilled water) and the other Na2SeO3 concentrations, 4.0 mg/L Na2SeO3 significantly increased the superoxide dismutase, catalase, ascorbate peroxidase glutathione reductase, glutathione peroxidase and glutathione S-transferase activities, and decreased the malondialdehyde content and the electrolyte leakage. Meanwhile, compared with the control and the other of Na2SeO3 concentrations, 4.0 mg/L Na2SeO3 significantly increased the soluble sugar, proline and soluble protein contents, relative water content, average fresh weight change rate and average water balance value. The above results imply that Na2SeO3 showed an important role in strengthening the antioxidant capacity and maintaining the water balance. Besides, 4.0 mg/L Na2SeO3 significantly increased the flower diameter and the vase life. The above findings suggested that Na2SeO3 extended the vase life and the ornamental value of E. grandiflorum cut flowers by enhancing the antioxidant capacity and water balance, which provides new knowledge for the application of Na2SeO3 in improving the fresh keeping of E. grandiflorum cut flowers.

Selenium is an essential microelement involved in various biological processes. Selenium deficiency increases the risk of human immunodeficiency virus infection, cancer, cardiovascular disease, and inflammatory bowel disease. Selenium possesses anti-oxidant, anti-cancer, immunomodulatory, hypoglycemic, and intestinal microbiota-regulating properties. The non-linear dose-response relationship between selenium status and health effects is U-shaped; individuals with low baseline selenium levels may benefit from supplementation, whereas those with acceptable or high selenium levels may face possible health hazards. Selenium supplementation is beneficial in various populations and conditions; however, given its small safety window, the safety of selenium supplementation is still a subject of debate. This review summarizes the current understanding of the health-promoting effects of selenium on the human body, the dietary reference intake, and evidence of the association between selenium deficiency and disease.

This study was aimed to investigate the effect of in-ovo injection with vitamin E and selenium during the incubation and post-hatching period. 360 fertilized eggs of Ross 308 broiler breeders were incubated, then distributed on 10th day into three groups (120 eggs/group). 1st group: uninjected eggs, 2nd group: eggs were injected with 0.1 ml deionized water/egg in the chorioallantoic cavity, and 3rd group: eggs were injected with 0.1 ml/egg of Introvit-E-Selen in chorioallantoic cavity. After hatching, 270 chicks were randomly distributed into six groups, and reared until 42 days aged as follows: birds in 1st, 3rd and 5th groups drinks tap water only (free from any addition), while birds of other groups, were reared on drinking water supplemented with 12.5 mg vitamin E and 500 µg sodium selenite/liter water. Results showed a significant increase in hemoglobin, lymphocytes% and serum globulin, and a significant decrease in packed cell volume%, heterophils%, heterophils/lymphocytes ratio, glucose, cholesterol, triglycerides and albumin concentration as compared with control. Flavonoids from Ginkgo biloba have antioxidant and free radical scavenging activity. In the present study, we used different concentrations of OS (organic selenium), NS (nano-selenium), and IS (inorganic selenium) to treat the 2-year-old Ginkgo seedlings and studied the effects of different concentrations of selenium treatment on the physiological indexes and flavonoid content of Ginkgo at different growth stages. Our results showed that the content of chlorophyll, soluble sugar, and total flavonoid glycosides in the 1.6-mM NS treatment group increased significantly in July and September (by 43.87%, 59.92%, and 43.99% and 13.86%, 18.60%, and 35.04%), compared to the control respectively, all P < 0.05. To further analyze the changes of flavonoid content and gene expression levels in Ginkgo leaves, transcriptome sequencing analysis was performed on the treated seedlings. Sequencing analysis results showed that NS could promote PAL, COMT, CAD, and F6H gene expression levels in the flavonoid synthesis pathway; OS up-regulates the CCR and HCT genes. IS promotes the expression of C3'H, CCoAOMT, CYP, DFR, and HCT genes. Furthermore, the increase of key genes and enzyme levels promotes the increase of total flavonoid content. These data suggest that the appropriate concentration of OS and NS can promote the expression level of flavonoid synthesis genes and further promote the increase of total flavonoid content in Ginkgo leaves, which is beneficial to the medicinal value of Ginkgo extract. Currently, planting selenium-rich crops using inorganic selenium such as selenate and selenite is used to address human selenium deficiency problems. In this paper, besides the above two traditional inorganic selenium speciation, we chose a new organic selenium speciation of potassium selenocyanoacetate to investigate the different effects of selenium speciation on selenium absorption, selenium transformation and cadmium antagonism via foliar application. Planting experiments showed that the selenium content of garlic bulbs treated with organic selenium was 1.8–3.9 times higher than that of inorganic selenium. Additionally, the absorption and transformation efficiency of organic selenium in garlic was also the highest, reaching over 95 %. Importantly, it was noteworthy that the cadmium content in bulbs treated with organic selenium was significantly lower than the Chinese food safety standard (0.2 mg/kg). Hence, this study provides an efficient organic selenium speciation which is beneficial to meet human selenium requirements and ensure safe utilization of cadmium-contaminated soils. This study aims to investigate the effects of different selenium (Se) sources on the growth performance of Chinese broilers and provide a scientific rationale for adding Se additives to broiler feed. Relevant studies that meet standard inclusion criteria were identified and extracted from China National Knowledge Infrastructure and Wanfang and Chinese Scientific Journal (VIP) databases. A total of 9 studies with 539 subjects were included. A meta-analysis was performed with STATA15.0 to estimate the combined standardized mean difference (SMD) with a 95% confidence interval (95% CI). Heterogeneity test of articles was examined by Q-test, and the results showed that P values of feed conversion ratio, average daily gain (ADG), and average daily intake were all less than 0.05, suggesting a strong heterogeneity among the selected literature. Therefore, the random effect model is selected to calculate the SMD of the three indexes. The combined SMDs (95% CI) of feed:gain, ADG, and average daily feed intake (ADFI) were −0.39 (−1.03, 0.25), 0.26 (−0.29, 0.81), and −1.45 (−3.09, 0.20), respectively, and the P values were all less than 0.05. This study shows that the absolute differences in the growth performance (feed:gain, ADG, and ADFI) of broilers fed with either organic or inorganic Se supplements at the same dose were quite small. The P values of Egger’s test were 0.770, 0.089, and 0.426, respectively, for the above indexes, showing no significant publication bias. Sensitivity analysis ensured the stability and reliability of the results. In summary, the effects of organic and inorganic Se in feed on the growth performance of broilers are statistically equal.

Abstract The chemical properties including toxicity of selenium depend on the concentration and chemical structure present in the natural environment. Inorganic selenium is more toxic than organic selenium; thus, the accurate determination of inorganic selenium in various sample has been needed. HG-AAS, HPLC-ICP-MS, and GC-MS are the current methods for the determination of inorganic selenium. However, these methods have the disadvantage of expensive equipment, complicated sample preparation, and long analysis time. LIBS coupled with ion exchange membrane was used for the determination of inorganic selenium. The detection limit of inorganic selenium by the analytical method was 11.21 mg/kg, and the recoveries of 50 and 100 mg/kg Se(VI) were 98.5 % and 101.80 %, respectively. The speciation method is cost-effective, simple, and not labor intensive for the quantitation of inorganic selenium.

The total selenium content of selenium-enriched agricultural products was determined by inductively coupled plasma mass spectrometry after microwave digestion. The total selenium content in the tested samples ranged from 27.9 μg/kg to 13.6 mg/kg. High performance liquid chromatography–inductively coupled plasma mass spectrometry was employed to determine selenite and selenate in selenium-enriched rice, tea, and garlic. The separation of inorganic and organic selenium was realized within 15 min by using 40 mmol/L diammonium phosphate (pH 5.0) and 60 mmol/L diammonium phosphate (pH 6.0) as the mobile phase with gradient elution. Selenite and selenate were isolated by ultrasonic assisted extraction at 78 °C for 30 min with 0.1 mol/L potassium hydroxide as the extraction solution. The detection limits were 2.5 and 5 μg/kg for selenite and selenate, respectively. The relative standard deviation was less than 5.5%. Recoveries of inorganic selenium were between 88.2% and 113.0%. The proposed method was successfully applied for the determination of inorganic selenium as well as total selenium in selenium-enriched rice, tea, and garlic. The results showed that inorganic selenium in natural selenium-enriched agricultural products is primarily present in selenite composing no more than 20% of total selenium. [ABSTRACT FROM AUTHOR]

After identifying the nanosorbent with FT-IR, CHNS, XRD, SEM, TEM, VSM, and BET surface analysis, parameters affecting the retention and desorption were optimized. Under optimal conditions, the detection limit of 8 ng/L was achieved for selenium (IV), and the method's precisions were less than 12.5 % intra-day RSD% and < 14.4 % for inter-day RSD%. The highest adsorption capacity was obtained as 264 mg/g. Finally, the desired adsorbent was used to extract and determine selenium species in water samples and total selenium in food samples. Cadmium (Cd) is an element injurious for human health and is possibly toxic to organisms at minor concentrations. While some of other trace metallic elements have antagonistic features to it. One of them is the interaction between selenium (Se) and Cd in plant different organs. Literature review disclosed that the intake of Se to some extent can reduce the accumulation of Cd in plants, while the research on of trace metallic elements (Cd) and Se-enriched food (rice) in the living body has rarely been reported. This study intended to explore whether there was a mitigating effect of Se-enriched rice on mice poisoned with Cd. A mouse model of low-dose and high-dose Cd poisoning was established (supplemented with cadmium chloride(CdCl2·2½H20)), followed by feeding two groups (1) Se-enriched rice and (2) setting an equal amount of inorganic Se group. After that, the impact of Se-enriched rice on the antioxidant activity was evaluated. The Se-enriched diet enhanced the total antioxidant capacity (T-AOC), superoxide dismutase (SOD), and enzyme activities of GSH peroxidase (GSH-Px) in mice livers and kidney whereas significantly decreased the malondialdehyde (MDA) contents. Moreover, the degree of physiological damage in mice with low cadmium poisoning was significantly alleviated, and the expression of antioxidant genes (Nrf-2, GPX1, TrxR2, TNF-2) was increased. In conclusion, the Se-enriched diet has a positive effect on the biological effects in mice, and it can be used as a daily diet to resist damage to the body's low Cd state and support enzymatic antioxidant systems by eliminating oxidative injury. The aim of the present study was to assess the effects of selenium feed supplements on the functional properties of eggs. The hens in experimental groups were fed diets supplemented with 0.2 mg/kg selenium from sodium selenite (Group 1), selenium-enriched yeast (Group 2), synthetic L-selenomethionine (Group 3), or hydroxy analog of selenomethionine (Group 4). The Control Group (Group C) was fed with basal feed without supplementation. The highest values of albumen gel firmness were shown in Group C eggs; differences with all experimental groups were significant (p < 0.001 to p = 0.009). It was ascertained that albumen gel firmness correlated with albumen pH (rs = 0.490; p < 0.001), which was highest in eggs from non-supplemented hens. Group 1 eggs and Group C eggs showed lower albumen foaming capacity (p < 0.001) compared to eggs from other groups. Both albumen foaming capacity and albumen foam stability were higher in Group 2 eggs than in Group C eggs (p < 0.001). The highest yolk foaming capacity was found in Group 2 eggs (p < 0.001). Sponge cakes baked with Group C eggs had a smaller volume than those baked with eggs from Group 2 (p = 0.005), Group 3 (p = 0.004) and Group 4 (p = 0.024). The results of the study confirmed that selenium added to the laying hen feed significantly affected the monitored functional properties of both albumen and yolk. The most distinctive effect of selenium was shown in eggs from the group supplemented with selenium-enriched yeast, for which the results of albumen foam capacity and stability and yolk foaming capacity were the best. Accurately quantifying selenium (Se) speciation and transformation in Se-enriched crops is highly significant for human health. The investigation of Se species in Se-enriched crops involves assessing the enrichment of both organic and inorganic Se species, considering their plant families and edible parts. The staple crops of rice, corn, and wheat showed no or less inorganic Se with the increase of total Se; however, potatoes expressed a proportion of selenate [Se(VI)]. In addition, the organic Se proportions in Se-enriched crops of Cruciferous, Brassicaceae , and Umbelliferae plant families were relatively lower than the proportion of inorganic Se. Concurrently, the edible parts of the Se-enriched gramineous or cereal crops enriched with organic Se and crops with fruit, stem, leaf, and root as edible parts contain the maximum percentage of organic Se with a certain proportion of inorganic Se. This study contributes to a sparse body of literature by meticulously discerning appropriate Se-enriched crop selection through a comprehensive evaluation of Se speciation and its organic and inorganic accumulation potential.

This study aimed to evaluate effects of supplementing the diets of laying hens with different sources of Se and different levels of flaxseed on egg Se concentration, yolk antioxidant properties and immune response. In a completely randomized design, 384 Hy-Line W-36 hens (aged 50 weeks) were assigned to a 3 × 4 factorial arrangement, comprising four replicates of eight hens. For 10 weeks, the birds were fed one of three basal diets containing 0, 50, and 100 g/kg of flaxseed and regular Se content from mineral premix, supplemented with no additional Se (SN) and 1 mg/kg of Se from sodium selenite (SS), Se-enriched yeast (SY), and hydroxy selenomethionine (SOH). The greatest Se concentration in the yolk was observed in diets containing organic Se sources. The SOH diet produced the greatest albumen Se concentration, followed by SY, and then SS. Using any Se supplementation improved glutathione peroxidase activity and total antioxidant capacity, compared with SN. However, superoxide dismutase activity and malondialdehyde were not affected by Se supplementation. Cell-mediated immune response was improved by Se supplementation, especially SS. Antibody response against sheep red blood cell (SRBC) was not affected by Se source, except that IgM was greater in the hens that received SS and SY. Neither flaxseed supplementation, nor its interaction with Se source was significant for any trait. Thus, supplementation of diets with 1 mg/kg Se, especially from organic sources, enriched the egg and improved some yolk antioxidant properties and immune system function. Selenium (Se) is a non-metallic trace element essential for normal cellular function, which has been linked with reduced risk of cancer, cardiovascular disease, cognitive decline and thyroid disease in humans. Se deficiency in livestock is associated with white muscle disease, retained placenta, ill-thrift and mastitis. Where Se status or bioavailability from the soil for plants is poor, livestock rely on supplemental Se in their diets predominantly as either sodium selenite (inorganic form) or selenised-yeast (organic form). As lactic acid bacteria (LAB) have been shown to incorporate Se as either organic or elemental (Nano-Se) there may be potential to use silage inoculant bacteria to improve the Se status of feed to provide the Se requirements of livestock. We screened twenty-seven LAB in MRS broth in the presence of sodium selenite for growth and uptake of Se as organic (selenocysteine and selenomethionine), inorganic (selenite and selenate) or/and Nano-Se, with the aim to identify potential candidates for a mini-silo study. Sodium selenite addition into the growth medium of LAB reduced growth rates but also resulted in the conversion of the inorganic sodium selenite into predominately Nano-Se and small quantities of organic-Se. Based on a rank analysis of growth and ability to take up (total Se content) and convert inorganic Se (Nano and organic Se content), three LAB were selected for further investigation as silage inoculants: L. brevis DSMZ (A), L. plantarum LF1 (B), and L. plantarum SSL MC15 (C). Each LAB was used as an inoculant within a grass mini-silo trial, either cultured in the presence of sodium selenite before inoculation or sodium selenite added to the inoculum at inoculation versus controls with no Se. The addition of sodium selenite either into the growth media of LAB or applied at inoculation of grass silage did not interfere with the ability of the LAB to act as a silage inoculant with no difference in silage fermentation characteristic between LAB with no Se added. The addition of sodium selenite either to the LAB growth medium or at inoculation resulted in the conversion of sodium selenite into Nano-Se and organic-Se (Nano-Se, ca. 103 higher than organic), as previously shown in the screening trial. There was no difference between the three LAB for incorporation of Se or in silage quality, indicating the potential to develop silage inoculants to increase the bioavailable form of Se (elemental and organic) to livestock through conversion of inorganic forms during ensiling. BACKGROUND: Selenium is an indispensable trace element for humans and its deficiency can lead to serious health complications. Nearly 70% of the area of China faces selenium deficiency. To deal with this problem, selenium‐enriched rice has been increasingly incorporated into everyday diets. However, there is a lack of in‐depth studies of the absorption, translocation, and transformation of selenium in the different parts of the rice plant when sprayed with sodium selenite. RESULTS: Foliar sodium selenite applied at critical growth stages can significantly improve the total and organic selenium content of plants. Application of 10 mg L−1 sodium selenite led to the most organic selenium (0.03 mg kg−1) in polished rice. Correlation studies of sodium selenite applied to leaves and other plant parts showed that total selenium accumulated most in glume, followed by rice bran, then polished rice, and finally embryo. The behavior of organic selenium was different. Organic selenium accumulated most in polished rice, then embryo, then rice bran, and finally glume. Moreover, 75–85% of the Se found in polished rice and embryo was organic in nature. The present study amid to evaluate the effect of feeding Silver Montazah parents reared in open system house during winter season on different selenium sources and levels on the fertility of cocks, performance and mortality % of hatched chicks. The examined two selenium sources were nano selenium (Nano-Se) and sodium selenite (SS), and the three supplemental levels of each source were 0.10, 0.25 and 0.40 ppm. 180 females and 90 males of Silver Montazah chickens divided into 6 treatments of three replicates of each. Males and females located in separate battery cages in open system house. Semen characterization was carried out, then hens were artificially inseminated by semen from cocks with the same dietary treatment. A total number of 90 eggs/treatment were incubated and fertility and hatchability were calculated. Hatched chicks fed on control diet containing 0.10 mg SS /kg diet up to 2 wks of age then chicks of each treatment received the same Se treatments of their parents. Growth performance of chicks was recorded up to 8 wks of age. The obtained Results showed that: - Using Nano Se improved semen ejaculate volume, sperm motility, growth performance of hatched chicks during first 2 wks of age and their dressing % and PCV% at 8 wks of age, also it improve significant the activities of antioxidant system and liver functions. , using levels 0.25 mg Se/kg enhance the overall semen quality, fertility and hatchability chick performance and their physiological statues. - The overall results of all experiment treatments showed superior effect of using Nano-Se at level 0.25 mg Se /kg diet to enhance the overall reproductive performance of cocks and growth and physiological status of their hatched chicks up to 8 wks. In addition, the examination of blood cells and plasma biochemical measurements showed positive effect on antioxidant system and overall immune status. Based on the obtained results we could recommend using 0.25 mg Nano Se/kg diet to enhance the reproductive performance of Silver Montazah cocks and decrease mortality rate of growing chicks during winter season. Ninety-six male goslings were allocated and assigned to treatment using a completely randomized design. Dietary treatments included a basal diet consisting of corn, wheat, and soybean meal with either no additional selenium (CON), 0.3 mg/kg of inorganic selenium (I-Se; sodium selenite), or 0.3 mg/kg of organic selenium (O-Se; selenium-enriched yeast). After a 56-day feeding period, geese were slaughtered on a common ending day and two geese per pen (n = 24) were used for the analyses conducted in this study. Meat (equal portions of the breast and thigh meat) and liver were collected and evaluated for proximate composition, fatty acid profile, pH, phenolic content, thiobarbituric acid reactive substances (TBARS), and total volatile basic nitrogen (TVB-N) over a 9-day storage period at 4 °C. The meat and liver samples from geese supplemented I-Se or O-Se had greater (p < 0.01) lipid content compared with geese not supplemented with additional selenium. At the conclusion of the 9-day storage period, meat and liver samples from geese supplemented I-Se or O-Se had lower (p < 0.05) pH values, greater (p < 0.05) phenolic content, lower (p < 0.05) TBARS values, and lower (p < 0.05) TVB-N compared with geese not supplemented with additional selenium (CON).

The aim of the study was to compare the effect of selenium (Se) supplementation in organic (lactate-protein complex) or inorganic (sodium selenite) forms on Se concentrations in the blood and organs of goat kids. The experiment involved nineteen male goat kids divided into three groups: C, Se-I, and Se-O. Control group C (n = 5) was without Se supplementation, group Se-I (n = 7) received sodium selenite supplement (0.30 mg Se per animal/day), and group Se-O (n = 7) received lactate-protein selenium complex (0.28 mg Se per animal/day). The supplementation started on the day of weaning and continued for 13 weeks till the day of slaughter. Blood samples for determination of Se concentration and glutathione peroxidase activity were collected on the day of weaning and during weeks 4, 8, and 13 thereafter. Samples of liver tissue, spleen, kidneys, lungs, heart, tongue, diaphragm, shoulder, back, and thigh muscles were taken immediately after slaughter. Significantly higher concentration of Se in group Se-O in comparison with group C was found in thigh muscles (110.4 vs 71.0 µg/kg, P ≤ 0.01), shoulder (105.0 vs 67.2 µg/kg, P ≤ 0.01), back (102.9 vs 61.7 µg/kg, P ≤ 0.01), and heart (180.8 vs 116.7 µg/kg, P ≤ 0.01). Significantly higher concentration of Se in group Se-I in comparison with group C was found in shoulder (83.2 vs 67.2 µg/kg, P ≤ 0.01) and diaphragm (93.6 vs 72.8 µg/kg, P ≤ 0.01). The comparison of the groups Se-I and Se-O showed significantly higher Se concentrations in thigh muscles, heart, back, shoulder, and lungs in group Se-O. Mean Se concentration in tissues of experimental groups was 125.8% in Se-O group and 110.7% in Se-I group in comparison with group C. Our results are suggesting that supplementation of Se in the form of lactate-protein complex is more efficient in comparison with sodium selenite.

The present study aimed to investigate the influence of maternal selenium supplementation on the skeletal muscle development of the offspring. A total of 720 Ross 308 broiler breeders at 24-week-old were allocated into 3 treatments with 6 replicates of 40 hens each and fed with 0 mg/kg-(group Se/C), 0.5 mg/kg organic-(group Se/O), and 0.5 mg/kg inorganic-(group Se/I) selenium, respectively for 8 weeks. The male offspring from each nutritional treatment were divided and housed into 8 cages of 12 birds each and fed with a commercial diet supplemented with selenium from Na2SeO3 at 0.15 mg/kg. Results showed that Se/O group had the highest selenium deposition (P < 0.05) in the egg yolk and albumen. Furthermore, maternal selenium supplementation promoted breast muscle yield; increased serum insulin and IGF-I concentration; upregulated AKT, mammalian target of rapamycin (mTOR), P70S6K, Myf5, MyoD, MyoG, and SelW mRNA levels; and improved the phosphorylation of AKT at Serine 473 residue, mTOR at Serine 2448 residue, and FOXO at Serine 256 residue in skeletal muscles of the offspring. In contrast, the hens’ diet supplemented with selenium could result in reduction of uric acid level in serum and downregulation of Atrogin-1 and MuRF1 mRNA levels in the skeletal muscle of the offspring. Additionally, no significant effect on the skeletal muscle development post-hatch was observed between organic and inorganic selenium supplementation. In conclusion, maternal organic selenium supplementation improved selenium deposition in egg; however, no significant effect has been detected on the breast muscle development of the offspring of broiler breeder compared with inorganic selenium supplementation. Effect of various doses and forms of selenium (organically bound vs. inorganic) on selected parameters of ruminal fluid and blood in sheep was evaluated. The trial was performed with 15 sheep divided into two groups. Animals from group A (n = 8) received a feeding mixture with selenomethionine, while sheep from group B (n = 7) were fed a mixture with sodium selenite. During the first 14 days, animals from both groups were fed a mixture with optimum dose of selenium (1 mg Se/kg dry matter). For another 28 days, all experimental animals received a Se-deficient mixture (0.1 mg Se/kg dry matter), whereas in the last 21 days of the experiment, animals were fed a high-Se diet (5.0 mg Se/kg dry matter). Throughout the trial, 4 samples of blood and ruminal fluid were taken from each animal. The samples of ruminal fluid were analyzed to determine the concentration of Se and identify other parameters of ruminal fermentations. Selenium levels were also determined in ruminal biomass. In blood, Se concentration, glutathione peroxidase (GSH-Px) activity, and other selected biochemical parameters were measured. The results of the present study demonstrate that the actual intake of both organic and inorganic Se is reflected in Se concentration in ruminal fluid and ruminal biomass and, similarly, in Se content and GSH-Px activity in blood. The form of supplemented selenium did not have a significant effect on ruminal fermentation parameters in dependence on different doses of Se contained in feeding rations (except for the negative effect of a sudden start of feeding high levels of organically bound Se on infusoria count, which was accompanied by the increase of GMT, LDH, and AST enzymes activity in ruminal fluid). The results also suggest possible negative interaction between the intake of organically bound selenium and the concentration of copper in blood of sheep.

The goal of the experiment was to compare the effect of four different forms of selenium (Se) - sodium selenite (SS), lactate-protein selenium complex (SL), selenium enriched yeast (SY), and selenium-proteinate (SP) supplemented to pregnant goats on Se concentration and glutathione peroxidase (GSH-Px) activity in the blood of goats on the day of delivery and also on Se concentration and GSH-Px activity in the blood of newborn kids. The experiment involved 33 pregnant goats of White Short-haired breed. The supplementation started 6 weeks before the parturition. The goats were divided into 5 groups: control group C, not supplemented, and 4 trial groups (SL, SP, SS, SY), which received Se in the above stated forms by the means of supplemented pellets (300 g per animal per day) at a rate 900 μg Se/kg of dry matter. The average Se concentrations in the blood of the goats were 79.6 μg/l in group C, 152.6 μg/l in group SL, 167.1 μg/l in group SP, 144.9 μg/l in group SS, and 152.9 μg/l in group SY. Selenium concentrations in all 4 trial groups were significantly higher (P < 0.01) than in control group, however no significant difference was found between individual trial groups. Likewise, the activity of GSH-Px in goat blood increased significantly in all supplemented groups compared to the controls; however we did not discover any significant differences in activity of GSH-Px between the individual selenium-supplemented groups. The Se concentrations in the blood of kids were significantly (P < 0.01) higher in the selenium-supplemented groups (SL - 94.9 μg/l, SP - 87.5 μg/l, SS - 87.6 μg/l, SY - 92.5 μg/l) than in the control group (C - 49.4 μg/l), but we did not discover any differences between the individual experimental groups. The activity of GSH-Px in the blood of the kids tended towards higher values in the supplemented groups than in the control group, but the values were significantly higher (P < 0.05) only in groups SY and SL. We have found significant correlation between GSH-Px activity and Se concentration in the blood of goats (r = 0.86) and newborn kids (r = 0.95). Likewise, there was significant correlation between Se concentration in the blood of goats and their kids (r = 0.74). We discovered that the kids are reaching physiologically only about 60% of Se status in whole blood in comparison with their mothers. Our results are suggesting that all the above forms of Se were similarly utilised and transferred into the foetus in the goats.

This paper evaluates the impact of long-term supplementation of different forms of zinc (Zn) and selenium (Se) on the content of these substances in the blood and hair of goats. Two analogous supplementation experiments were performed. 37 goats divided into four groups were used in the first trial with the Zn supplementation. Group A (n = 10) was a control group (with no Zn administered). A further three groups (B, C, D) were supplemented with Zn in various forms. Group B (n = 9) with zinc oxide, Group C (n = 9) with zinc lactate and Group D (n = 9) with zinc chelate. The second trial with Se supplementation was carried out on 20 goats divided into four groups. Group E (n = 5) was a control group. The other three groups were administered Se. Group F (n = 5) was supplied with a selenium lactate-protein complex, Group G (n = 5) with sodium selenite and Group H (n = 5) with selenium yeast. Three months later blood and hair samples were taken from all animals and Zn and Se concentrations were determined in whole blood, plasma, and hair. Glutathione peroxidase (GSH-Px) activity was determined in the Se supplementation trial group. At the end of the trial the Zn concentrations in plasma and whole blood were without major differences between the groups. The plasma concentration of Zn did not increase from the initial value at the start of the trial. In hair the average concentration of Zn was 95.2-100.0 mg/kgin all groups. No conclusive relation was confirmed between the values of Zn in hair and its concentration in blood. The Se concentration in whole blood (µg/l) at the end of trial in supplemented groups (F - 188.8 ± 24.6; G - 197.2 ± 10.9; H - 190.1 ± 26.3) was significantly higher (P < 0.01) than in the control group (E - 103.1 ± 23.5). Similarly, the activity of GSH-Px (µkat/l) was significantly higher in all supplemented groups (F - 872.3 ± 94.8; G - 659.5 ± 176.4; H - 839.8 ± 150.8) than in the control group (E - 379.1 ± 63.5). Se content in hair (µg/kg) was higher also in all trial groups (F - 242.3 ± 41.5; G - 200.5 ± 46.9; H - 270.0 ± 106.8) than in the control group (E - 174.7 ± 38.0). However, it was significantly (P < 0.05) higher only in Group F. A conclusive correlation was identified between the Se concentration in whole blood and its content in hair (r = 0.54; P < 0.05; n = 20). Based on the results it can be concluded that none of the supplemented forms of Zn increased its concentration in blood, plasma and hair. On the other hand, the administration of Se led to an increase in the Se concentration in blood, increased the activity of GSH-Px in whole blood and the Se content in hair. Based on the proven correlation and regression relation between the Se concentration in blood and its content in hair, hair can be considered as a suitable material for the diagnosis of long-term Se status in goats. Goats with sufficient Se status are those that have more than 160 µg/kg of Se in hair dry weight.

Selenium is a trace element essential for human health that has received considerable attention due to its nutritional value. Selenium’s bioactivity and toxicity are closely related to its chemical form, and several studies have suggested that the organic form of selenium (i.e., selenomethionine) is more bioavailable and less toxic than its inorganic form (i.e., sodium selenite). Probiotics, especially Bifidobacteriium and Lactobacillus spp., have received increasing attention in recent years, due to their intestinal microbial balancing effects and nutraceutical benefits. Recently, the bioconversion (a.k.a biotransformation) of various bioactive molecules (e.g., minerals, primary and secondary metabolites) using probiotics has been investigated to improve substrate biofunctional properties. However, there have been few reports of inorganic selenium conversion into its organic form using Bifidobacterium and Lactobacillus spp. Here we report that the biosynthesis of organic selenium was accomplished using the whole cell bioconversion of sodium selenite under controlled Bifidobacterium bifidum BGN4 culture conditions. The total amount of organic and inorganic selenium was quantified using an inductively coupled plasma-atomic emission spectrometer (ICP-AES). The selenium species were separated via anion-exchange chromatography and analyzed with inductively coupled plasma-mass spectrometry (ICP-MS). Our findings indicated that the maximum level of organic selenium was 207.5 µg/g in selenium-enriched B. bifidum BGN4. Selenomethionine was the main organic selenium in selenium-enriched B. bifidum BGN4 (169.6 µg/g). Considering that B. bifidum BGN4 is a commercial probiotic strain used in the functional food industry with clinically proven beneficial effects, selenium-enriched B. bifidum BGN4 has the potential to provide dual healthy functions as a daily supplement of selenium and regulator of intestinal bacteria. This is the first report on the production of organic selenium using B. bifidum spp.

The aim of this study was to evaluate the effect of a long-term peroral selenium supplementation in the form of sodium selenite and selenium lactate-protein complex by comparing selenium concentrations and glutathione peroxidase activity in blood of goats and their kids as well as comparing selenium concentrations in goat colostrums. For the study, a total of 27 clinically healthy pregnant white shorthair goats were used. They were divided to three groups, i.e., the control group (C) without any selenium supplementation, sodium selenite group (E1) and selenium lactate-protein complex group (E2). For four months, experimental goats received 0.43 mg of selenium per animal per day in diet; goats from the control group were given 0.15 mg of selenium per animal per day. At the beginning of the experiment, goats of all groups showed an average selenium concentration of 96 μg/l in whole blood. On the parturition day, samples of first colostrum from goats and heparinized blood from goats and kids were taken. In the control group (C), average blood selenium concentrations of 111.4 ± 33.5 μg/l were observed on the parturition day. In both experimental groups, selenium concentrations were significantly higher (P < 0.05). Average selenium concentration in the sodium selenite group (E1) was 177.2 ± 34.8 μg/l and in the group supplemented with selenium lactate-protein complex (E2) 159.0 ± 28.5 μg/l. Average glutathione peroxidase (GSH-Px) activity in blood of control goats (C) was 581.9 ± 99.2 μkat/l, in group E1 1 154.6 ± 156.2 μkat/l and in group E2 1 011.6 ± 153.6 μkat/l. GSH-Px activity in experimental groups was significantly higher (P < 0.05) as compared with the control group. Average selenium concentrations in colostrum was in the control group 40.1 ± 12.8 μg/l, in E1 99.0 ± 29.9 μg/l and in group E2 79.0 ± 17.7 μg/l. Colostral selenium concentrations in experimental groups were significantly higher (P < 0.05) as compared with the control group. No significant difference in the monitored parameters was found between experimental groups. In kids of control mothers (kC), average selenium concentrations in blood on the parturition day were 62.4 ± 22.9 μg/l; kids of mothers supplemented with sodium selenite (kE1) showed average selenium levels of 100.0 ± 31.2 μg/l, and the average selenium concentration in kids of mothers receiving lactate-protein complex was 83.4 ± 20.1 μg/l (kE2). Average GSH-Px activity in control kids (kC) was 402.1 ± 153.9 μkat/l. Kids from kE1 showed average activity of GSH-Px 806.1 ± 254.9 μkat/l and kids from group kE2 529.9 ± 119.8 μkat/l. Statistically significant difference (P < 0.05) was found only between kC and kE1 which showed significantly higher selenium concentration and GSH-Px activity. The results of this study confirm that both forms of selenium administered in experimental groups (i.e., sodium selenite and selenium lactate-protein complex) had similar biological effect in goats. However, results obtained in kids indicate a better effect of supplementation with sodium selenite.

Selenium is an essential microelement involved in various biological processes. Selenium deficiency increases the risk of human immunodeficiency virus infection, cancer, cardiovascular disease, and inflammatory bowel disease. Selenium possesses anti-oxidant, anti-cancer, immunomodulatory, hypoglycemic, and intestinal microbiota-regulating properties. The non-linear dose-response relationship between selenium status and health effects is U-shaped; individuals with low baseline selenium levels may benefit from supplementation, whereas those with acceptable or high selenium levels may face possible health hazards. Selenium supplementation is beneficial in various populations and conditions; however, given its small safety window, the safety of selenium supplementation is still a subject of debate. The aim of this study was to determine the effect of higher amount of inorganic selenium (Na2SeO3) on productivity parameters and meat quality of pigs for fattening. Sixty 62-d-old pigs (Landrace x Yorkshire (mother) and Pietrain x Duroc (father)) for fattening which were individually weighed and were randomly assigned to 2 dietary treatments with 2 replicate stalls of 15 pigs each. The pigs were fed for 13 weeks ad libitum with a standard wheat-barleysoybean meal compound diet supplemented with 0.3 mg/kg Na2SeO3 and 75 mg/kg vit. E (Control group) and with 0.5 mg/kg Na2SeO3 and 75 mg/kg vit. E (Experimental group). Meat traits in live pigs were measured by ultrasonic equipment Piglog 105. The samples for the analysis of selenium were taken from the M. longissimus dorsi between 12 and last rib and from the liver. Chemical composition of breast meat was being determined by standard methods. The results of conducted trial showed, that the addition of 0.5 mg/g of sodium selenite in the diets of fattening pigs didn't affect productivity parameters, chemical meat composition, accumulation of selenium in the meat tissue, but significant improved the muscularity of pigs. This study aimed to assess the interaction between vitamin B 6 and selenium (Se) for the flow of Se towards the Se-dependent glutathione peroxidase (GPX) system in response to oxidative stress naturally induced by oestrus in a pubertal pig model. At first oestrus, forty-five gilts were randomly assigned to the experimental diets ( n = 9/group): basal diet (CONT); CONT + 0.3 mg/kg of Na-selenite (MSeB 6 0); MSeB 6 0 + 10 mg/kg of HCl-B 6 (MSeB 6 10); CONT + 0.3 mg/kg of Se-enriched yeast (OSeB 6 0); and OSeB 6 0 + 10 mg/kg of HCl-B 6 (OSeB 6 10). Blood samples were collected at each oestrus (long-term profiles), and daily from day −4 to +3 (slaughter) of the fourth oestrus (peri-oestrus profiles) after which liver, kidneys, and ovaries were collected. For long-term profiles, CONT had lower blood Se than Se-supplemented gilts ( p < 0.01) and OSe was higher than MSe ( p < 0.01). Lower erythrocyte pyridoxal-5-phosphate was found in B 6 0 than B 6 10 ( p < 0.01). No treatment effect was observed on GPX activity. For peri-oestrus profiles, treatment effects were similar to long-term profiles. Treatment effects on liver Se were similar to those for long-term blood Se profiles and OSe had higher renal Se concentrations than MSe gilts ( p < 0.01). Gene expressions of GPX1 , GPX3 , GPX4 , and selenocysteine lyase in liver and kidney were greatest in OSeB 6 10 gilts ( p < 0.05). These results suggest that dietary B 6 modulate the metabolic pathway of OSe towards the GPX system during the peri-oestrus period in pubertal pigs. [ABSTRACT FROM AUTHOR]

Selenium (Se) has emerged as an essential micronutrient but its role in cancer prevention, immunomodulation, thyroid functioning and viral inhibition continues to remain inconclusive. The aim of this review is to dwell upon various factors responsible for mixed results arising out of Se supplementation trials. Some researchers have used inorganic form of Se, sodium selenite, which is less bioavailable and may be toxic if used for prolonged period, while others have opted for organic form, selenomethionine, which may incorporate non-specifically into proteins. In recent times, other organic forms, methylselenocysteine and selenocysteine, being less toxic, more bioavailable and endowed with greater therapeutic value against cancer have attracted the attention of research community. Moreover, appropriate dose (upto 200μg Se per day) may hold the key to bring perceptible beneficial health effects. Further, subjects used in trials should have low plasma Se levels for if they are already having its adequate level then supplementation is likely to be less effective. We conclude that the health effects of Se may largely depend upon its chemical form, dosage and Se-status of population being supplemented. Abstract: A useful preconcentration and determination method was proposed for trace selenium in food samples. The procedure is based on complex formation of Pyronine B with Se(IV) ions in the presence of sodium dodecyl sulphate (SDS) and Ponpe 7.5. The variables affecting complex formation, extraction and phase separation were studied and optimised. Under the experimental conditions used, the calibration graph was linear in the range of 20–1700μgL−1 for Se(IV) ions. The limit of detection was 3.81μgL−1 of Se(IV) and the relative standard deviation for 5 replicate determinations at 250μgL−1concentration level was 2.45%. Recovery values were obtained between 97.8% and 102.8% for spiked samples. The method was successfully applied to the determination of total selenium in some food samples and alcoholic and nonalcoholic beverages. Its validity was checked by the analysis of four certified reference materials. The results obtained by the proposed method were quantitatively in good agreement with the certified values. Selenium is an essential micronutrient in sheep, and deficiency can limit lamb growth and survival. To evaluate how different chemical forms of Se administered to mature ewes at comparative dosages affect ewe and progeny performance, 240 ewes were divided into 8 treatment groups (n = 30 each) and drenched weekly with no Se; at the maximum FDA-allowed concentration with inorganic Na-selenite or organic Se-yeast (4.9 mg Se/wk); with inorganic Na-selenate (8.95 mg Se/wk); or with inorganic Na-selenite and organic Se-yeast at supranutritional concentrations (14.7 and 24.5 mg Se/wk, respectively). The treatment period started approximately 2 wk before breeding and lasted for 62.5 wk. Ewes of the no-Se and Se-yeast groups continued treatments for another 21 to 24 wk through a second lambing season. Chemical form or dosage of Se did not affect ewe reproductive performance based on proportion of ewes lambing in each treatment group, or number of lambs born, nursed, or weaned per ewe (all P > 0.10). Ewes receiving the highest supplementation rate of Se-yeast at 24.5 mg Se/wk had higher BCS (scale 1 to 5) at the end of yr 1 (2.95 vs. 2.66; P = 0.05) than ewes receiving Se-yeast at 4.9 mg Se/wk. Performance was better in lambs from ewes receiving Se-yeast at 24.5 mg Se/wk than in lambs from ewes receiving Se-yeast at 4.9 mg Se/wk or no Se. In yr 1, lambs from ewes receiving Se-yeast at 24.5 vs. 4.9 mg Se/wk were heavier at 120 d of age (37.0 vs. 34.2 kg; P = 0.05). In yr 2, lambs from ewes receiving Se-yeast at 24.5 mg Se/wk were or tended to be heavier at 60 d of age than lambs from ewes receiving no Se (21.2 vs. 19.0 kg; P = 0.04) or lambs from ewes receiving Se-yeast at 4.9 mg Se/wk (19.2 kg; P = 0.09). This effect was more pronounced in ewes raising multiple lambs. We conclude that supranutritional supplementation of ewes with Se-yeast at 24.5 mg Se/wk improves lamb growth and ewe health without negatively affecting reproductive performance. We describe a method for the determination of inorganic selenium in water samples via gas-phase chemiluminescence (GPCL). Se(IV) was first derivatized with 4-nitro- o-phenylenediamine to form 5-nitropiazselenol. The latter was decomposed by persulfate through photocatalytic oxidation to give Se(VI), which was reduced to Se(IV). Selenium hydride was generated from Se(IV) through reduction with sodium borohydride and then preconcentrated using cryotrapping. The cryotrapped hydride was evaporated and carried to a reaction chamber by a stream of helium, where it produced GPCL as a result of ozonation. The method exhibits a wide linear calibration range (from 0.5 μg L to 1.0 mg L) with a detection limit of 0.12 μg L (for n = 11), and a relative standard deviation of 3.90 % (at n = 11) at 5.0 μg L level of selenium. Abstract: Herein, ultrasound-assisted emulsification microextraction (USAEME) and dispersive liquid–liquid microextraction (DLLME) methods based on applying low-density organic solvents have been critically compared for the speciation of inorganic selenium, Se(IV) (selenite) and Se(VI) (selenate) in environmental water samples by gas chromatography-flame ionization detection (GC-FID). At pH 2 and T =75°C for 7min, only Se(IV) was able to form the piazselenol complex with 4-nitro-o-phenylenediamine. Piazselenol was extracted using an extraction solvent and was injected into a GC-FID instrument for the determination of Se(IV). Conveniently, Se(VI) remained in the aqueous phase. Total inorganic selenium was determined after the reduction of Se(VI) to Se(IV) and prior to the above procedures. The Se(VI) concentration was calculated as the difference between the measured total inorganic selenium and Se(IV) content. The effect of various experimental parameters on the efficiencies of the two methods and their optimum values were studied with the aid of response surface methodology and experimental design. Under the optimal conditions, the limit of detections (LODs) for Se(IV) obtained by USAEME-GC-FID and DLLME-GC-FID were 0.05 and 0.11ngmL−1, respectively. The relative standard deviations (RSDs, n =6) for the measurement 10ngmL−1 of Se(IV) were 5.32% and 4.57% with the enrichment factors of 2491 and 1129 for USAEME-GC-FID and DLLME-GC-FID, respectively. new method was established for the speciation of Se by hydride generation-atomic fluorescence spectrometry. Effective separation of organic and inorganic Se in Se-enriched eggs was achieved by precipitating albumen with trichloroacetic acid. The detection limit for Se was 0.07 μg L. The proposed method was successfully applied for the determination of organic and inorganic Se as well as total Se in Se-enriched eggs with the recovery of 90.1-112% and the relative standard deviation ( n = 5) of 0.6-3.4%. The study was conducted to determine the efficacy of organic Se from Se-enriched alga Chlorella spp. in placental transfer to piglets. In group A (n = 8) the sows were fed during the gestation a diet supplemented with inorganic Se (sodium selenite, 0.3 mg/kg). In group B (n = 8) the diet of the sows was supplemented with organic Se from Se-enriched alga (0.3 mg/kg). The Se concentrations in the whole blood (P < 0.05) and in colostrums (P < 0.05) were higher in the group of sows provided with the organic Se form. No differences in GSH-Px activities were found between the two groups. The concentrations of Se in piglet tissues were also higher when the organic form was provided. A 4-week experiment was carried out on 360 laying hens of the Hy Line Brown hybrid. Laying hens were divided into three groups (C, E1 and E2) with 120 hens in each group and kept in 24 cages. Hens were fed layer diets containing 18% of crude protein and 11.60 MJ ME. Hens in the control group C were fed diets that contained 0.2 mg/kg of inorganic selenium (sodium selenite). Experimental groups E1 and E2 were given diets with increased concentrations of selenium as follows: E1 = 0.4 mg/kg of selenium (sodium selenite), E2 = 0.4 mg/kg of organic selenium (Sel- Plex). Selenium concentration in diets affected significantly the content of selenium in albumen (P < 0.001) and yolk (P < 0.05). The highest concentration of selenium was determined in albumen and yolk of eggs produced in group E2 (345 ng/g and 783 ng/g, respectively), then in eggs of group E1 (230 ng/g and 757 ng/g, respectively), and group C had the lowest concentration of selenium in albumen and yolk (181 ng/g and 573 ng/g, respectively). After 28 days of storage at 4 °C, the eggs containing organic selenium had more freshness (VN: C = 32.9, E1 = 2.60, E2 = 2.11). Speciation of selenium (Se) is typically carried out using a sophisticated technique such as ICP-MS after preconcentration using an adsorbent; however, the separation and preconcentration of inorganic Se has not been realized in the solutions containing high concentrations of SO 4 2–. A dithiocarbamate-modified cellulose (DMC) was used in this study for the selective extraction and preconcentration of inorganic Se in wastewater, with a portable liquid electrode plasma-optical emission spectrometry (LEP-OES) being employed for quantification. DMC was found to selectively and quantitatively adsorb selenite (SeIV) over a wide range of pH (1.0–8.0); however, less than 3.0% of selenate (SeVI) was adsorbed in a pH range of 3.0–11. Quantitative extraction of SeIV was achieved even in the presence of 3.5 mol L–1 SO 4 2–. The maximum sample volume from which 10 mg of DMC could quantitatively extract SeIV was found to be 500 mL. KOH (0.60 mL, 1.5 mol L–1) was found to quantitatively desorb SeIV retained on the adsorbent and yielded an enrichment factor of 833. The recovery of Se species from synthetic flue-gas desulfurization wastewater containing SeIV and SeVI at concentrations of 5.0 µmol L–1 was 96.2 ± 1.8% and 105.8 ± 1.8%, respectively. Abstract: The experiment was conducted to compare the effect of different selenium sources on the expression of glutathione peroxidase 1 (GPx1) and iodothyronine deiodinase 1 (Dio1) mRNA in mice by quantitative real-time PCR. A total of 60 male Kunming mice at average body weight of 20g were allotted to three groups in a randomized complete block design, namely two treatments and one control. Mice in Group 1 were fed a basal diet as control, while mice in Groups 2 and 3 were fed the basal diet supplemented with 0.1mg/kg selenium as sodium selenite or selenized yeast, respectively. Whole feeding experiment lasted for 30d. At the end of the feeding trial, liver mRNA levels of GPx1 and Dio1 were determined by quantitative real-time PCR, as well as growth performance, body composition, blood and GPx activity were determined. The results showed that no significant differences in overall growth performance and body composition, including body weight, body length, heart weight, kidney weight and liver weight, were found between the experimental groups (P>0.05). Blood GPx activity increased in all of the selenium supplemented groups compared with control group (P<0.01). However, blood GPx activity in selenized yeast group was higher than that in sodium selenite group (P<0.05). Liver mRNA levels of GPx1 and Dio1 also increased in the two selenium supplemented groups compared with the control group (P<0.05), while there was no significant difference between the sodium selenite and selenized yeast groups (P>0.05). In conclusion, selenium increased the mRNA expression of GPx1 and Dio1 genes in murine liver, and there was no significant difference between the organic or inorganic form of selenium used. The aim of the trial was to determine the efficacy of organic Se from Se-enriched yeast in placental transfer to piglets in the conditions of a Czech pig farm. In group I (n = 8) the sows were fed during gestation and lactation a diet supplemented with inorganic Se (sodium selenite, 0.3 mg/kg of Se for the gestation and 0.38 mg/kg of Se for the lactation diet). In group II (n = 8) the diet of the sows was supplemented with organic Se from Se-enriched yeast (0.3 mg/kg of Se for the gestation and 0.38 mg/kg of Se for the lactation diet). Se concentrations in the whole blood, colostrum and milk were higher (P < 0.01) in the group of sows fed with the organic Se form. No differences in GSH-Px activities in the whole blood were found between the two groups of sows. The concentrations of Se in piglet tissues (heart P < 0.05, liver P > 0.01, muscle P > 0.01) were also higher when the organic form was provided. It is concluded that the use of the organic Se from Se-enriched yeast in sows resulted in greater transfer of Se to their progeny, however it did not have a positive effect on the antioxidant system of the organism. Abstract: A flow injection system with anion exchange resin minicolumns was coupled with dynamic reaction cell (DRC™) ICP-MS for the determination and speciation of selenite and selenate at sub μgL−1 levels. The charged selenate and uncharged selenite were separated on the first resin column in which only selenate was retained. The unretained selenite was then deprotonated with alkaline solution, and the resulting anionic selenite species was collected on the second column serially connected downstream. By setting a sample loop, total selenium can be determined together with selenite and selenate. The selenium species was eluted by nitric acid and carried to DRC™ ICP-MS for their detection. Using ammonia as reaction gas, the detection of 78Se was improved. The enrichment factor was 20 for 10mL of sample. The standard deviations (n =5) of peak heights were 4.9%, 4.1%, and 7.0% for a 5.0×10−2 μgL−1 selenite and selenate, and total Se, respectively. The calibration graphs were linear from 2.0×10−2 to 1.0μgL−1 selenite and selenate. And, the linearity for total selenium was good in the range of 10.0×10−2 to 1.0μgL−1.
