The restoration and treatment of underground voids have always posed significant challenges for constructing environmentally sustainable mines. To investigate the effectiveness of a combined approach involving waste rock filling and grouting roof filling as treatment methods to ensure safety and stability in mining voids, this study employed a comprehensive dynamic analysis approach. It specifically focused on an individual underground metal mine cavity by integrating numerical simulation analysis techniques with onsite displacement monitoring methods. By simulating stress-strain conditions during different stages of cavity formation and subsequent treatments while considering onsite displacement monitoring data, this study extensively analyzed how these treatments impact rock stress levels, strain conditions within rocks themselves, and the stability of the surface riverbed. The results show that: (1) ''waste rock filling + cemented roof filling'' can effectively reduce the stress of surrounding rock, significantly improve the structural stability of goaf, reduce surface displacement and deformation, and ensure the stability of surface riverbed; (2) In the process of mining and filling, the stress and strain sensitive points are mainly distributed in the roof, bottom and intercolumn of the stope; (3) When the goaf is backfilled from top to bottom, the stress-strain changes at each stage are uneven: the maximum principal stress changes slowly in the early stage, but greatly in the later stage; The change of displacement showed a trend of slowing down in the early stage and stabilizing in the later stage: (4) ''waste rock filling + cemented filling'' roof joint can realize the transformation of solid waste into treasure in mines, effectively promote the construction of green mines, and is an effective means of goaf treatment. It provides ideas and means for other similar complex problem analysis and prevention.

Adeno-associated viruses (AAVs) are popular gene therapy delivery vectors, but their application can be limited by anti-vector immunity. Both preexisting neutralizing antibodies (NAbs) and post-administration NAbs can limit transgene expression and reduce the clinical utility of AAVs. The development of novel AAVs will advance our understanding of AAV immunity and may also have practical applications. In this study, we identified five novel AAV capsids from rhesus macaques. RhAAV4282 exhibited 91.4% capsid sequence similarity with AAV7 and showed similar tissue tropism with slightly diminished overall signal. Despite this sequence homology, RhAAV4282 and AAV7 showed limited cross-neutralization. We determined a cryo-EM structure of the RhAAV4282 capsid at 2.57 Å resolution and identified a small segment within the hypervariable region IV, involving seven amino acids that formed a shortened external loop in RhAAV4282 compared with AAV7. We generated RhAAV4282 and AAV7 mutants that involved swaps of this region and showed that this region partially determined neutralization phenotype. We termed this region the hypervariable region IV neutralizing epitope (HRNE). Our data suggests that modification of the HRNE can lead to AAVs with altered neutralization profiles.

Purpose Timely and accurate monitoring of soil salinity content (SSC) is essential for precise irrigation management of large-scale farmland. Uncrewed aerial vehicle (UAV) low-altitude remote sensing with high spatial and temporal resolution provides a scientific and effective technical means for SSC monitoring. Many existing soil salinity inversion models have only been tested by a single variable selection method or machine learning algorithm, and the influence of variable selection method combined with machine learning algorithm on the accuracy of soil salinity inversion remain further studied. Methods Firstly, based on UAV multispectral remote sensing data, by extracting the spectral reflectance of each sampling point to construct 30 spectral indexes, and using the pearson correlation coefficient (PCC), gray relational analysis (GRA), variable projection importance (VIP), and support vector machine-recursive feature elimination (SVM-RFE) to screen spectral index and realize the selection of sensitive variables. Subsequently, screened and unscreened variables as model input independent variables, constructed 20 soil salinity inversion models based on the support vector machine regression (SVM), back propagation neural network (BPNN), extreme learning machine (ELM), and random forest (RF) machine learning algorithms, the aim is to explore the feasibility of different variable selection methods combined with machine learning algorithms in SSC inversion of crop-covered farmland. To evaluate the performance of the soil salinity inversion model, the determination coefficient (R2), root mean square error (RMSE) and performance deviation ratio (RPD) were used to evaluate the model performance, and determined the best variable selection method and soil salinity inversion model by taking alfalfa covered farmland in arid oasis irrigation areas of China as the research object. Results The variable selection combined with machine learning algorithm can significantly improve the accuracy of remote sensing inversion of soil salinity. The performance of the models has been improved markedly using the four variable selection methods, and the applicability varied among the four methods, the GRA variable selection method is suitable for SVM, BPNN, and ELM modeling, while the PCC method is suitable for RF modeling. The GRA-SVM is the best soil salinity inversion model in alfalfa cover farmland, with Rv2 of 0.8888, RMSEv of 0.1780, and RPD of 1.8115 based on the model verification dataset, and the spatial distribution map of soil salinity can truly reflect the degree of soil salinization in the study area. Conclusion Based on our findings, the variable selection combined with machine learning algorithm is an effective method to improve the accuracy of soil salinity remote sensing inversion, which provides a new approach for timely and accurate acquisition of crops covered farmland soil salinity information.

Abstract Quorum sensing (QS) is a crucial regulatory mechanism controlling bacterial signalling and holds promise for novel therapies against antimicrobial resistance. In Gram-positive bacteria, such as Streptococcus pneumoniae, ComA is a conserved efflux pump responsible for the maturation and secretion of peptide signals, including the competence-stimulating peptide (CSP), yet its structure and function remain unclear. Here, we functionally characterize ComA as an ABC transporter with high ATP affinity and determined its cryo-EM structures in the presence or absence of CSP or nucleotides. Our findings reveal a network of strong electrostatic interactions unique to ComA at the intracellular gate, a putative binding pocket for two CSP molecules, and negatively charged residues facilitating CSP translocation. Mutations of these residues affect ComA’s peptidase activity in-vitro and prevent CSP export in-vivo. We demonstrate that ATP-Mg2+ triggers the outward-facing conformation of ComA for CSP release, rather than ATP alone. Our study provides molecular insights into the QS signal peptide secretion, highlighting potential targets for QS-targeting drugs.

Based on the self-developed high permeability water pressure loading system, the physical and mechanical properties and pore structure evolution under various high permeability water pressures were studied. This study aims to reveal the microscopic damage mechanism of concrete when subjected to high permeability water pressures. The results indicate that the internal water content increases with the rising of water pressure under various high permeability conditions. Furthermore, the higher the water pressure, the greater the reduction in strength, particularly evident in low-strength-grade concrete. High permeability water pressure causes to the expansion of gelatinized pores with small sizes into transition pores. Moreover, there is a concurrent alteration in both pore size and pore number under high permeability water pressure.

Rapid and accurate inversion of soil salinity is a key scientific problem that needs to be solved urgently. Due to the accuracy of UAV multispectral remote sensing inversion of salinity based on back propagation neural network (BPNN) is low, in this study, used the UAV multispectral image and field measurements of 60 soil surface salinity as data sources, 16 salinity indexs were constructed using the extracted spectral reflectance, and performed a gray relation analysis to screen salinity index features after applying a film removal to construct the BPNN salinity inversion model. Particle swarm optimization (PSO), thinking evolutionary algorithm (MEA), and genetic algorithm (GA) were applied to optimize the BPNN inverse model, respectively, and the optimization capabilities of the four algorithms were compared and evaluated to optimize the best optimization algorithm. The results showed that the GRA variable screening can effectively remove the redundant information of spectral parameters and reduce the complexity of the salinity inversion model; the PSO, MEA, and GA can effectively improve the robusticity of BPNN inversion model, and GA algorithm has the best optimization effect in terms of inverse model optimization effect, followed by MEA and PSO algorithms; the accuracy of the PSO-BPNN, MEA-BPNN, and GA-BPNN inversion models are better than that of the BPNN model, and GA-BPNN is the best salinity inversion model, which achieves R2 of 0.6659, RMSE of 0.0751, and RPD of 2.0211. This approach can effectively solve salinity monitoring accuracy issues of UAV multispectral inversion.

Abstract Diabetes Mellitus can cause dental pulp cells apoptosis by oxidative stress, and affect the integrity and function of dental pulp tissue. Mitochondria are the main attack targets of oxidative stress and have a critical role in apoptosis. However, whether mitochondria are involved in dental pulp damage caused by diabetes mellitus remains unclear. This study aimed to investigate the role of mitochondria in the apoptosis of odontoblast-like cell line (mDPC6T) induced by glucose oxidative stress, and to explore its possible mechanism. We established an oxidative stress model in vitro using glucose oxidase/glucose to simulate the pathological state under diabetic conditions. We found that the opening of mitochondrial permeability transition pore (mPTP) contributed to the apoptosis of mDPC6T treated with glucose oxidase, as evidenced by enhanced mitochondrial reactive oxygen species (mtROS) and intracellular Ca2+ disorder, significantly reduced mitochondrial membrane potential (MMP) and ATP production. Antioxidant N-acetylcysteine (NAC) or Cyclosporine A (mPTP inhibitor) blocked the mPTP opening, which significantly attenuated mitochondrial dysfunction and apoptosis induced by glucose oxidative stress. In addition, we found that glucose oxidative stress stimulated mPTP opening may through inhibition of Akt-GSK3β pathway. This study provides a new insight into the mitochondrial mechanism underlying diabetes-associated odontoblast-like cell apoptosis, laying a foundation for the prevention and treatment of diabetes-associated pulp injury.

The spatiotemporal variation of the model parameters of the soil-water characteristic curve (SWCC) reflect the soil water holding capacity and soil pore distribution state. It is an integral part of interdisciplinary disciplines such as soil hydrodynamics and ecohydrology. The authors selected the optimal SWCC model for the Lanzhou collapsible loess, used classical statistics and geostatistics methods to study the spatiotemporal variability of the SWCC model parameters, and using the comprehensive comparison of the mean relative differences (MRD), standard deviations (SDRD) and an index of temporal stability (ITS) determined the representativeness measuring point. The results showed that the SWCC parameter α was medium variability in the 0–30 cm soil layer, n and θs were of low variability, and the spatial distribution of the parameters of different soil layers was consistent. Migration direction prediction of θs was very similar in each layer, α, n and θs were all strongly significantly correlated positively. Moreover, the determination coefficient of representative measuring point 16 had the highest prediction accuracy for the measured values of SWCC. The results of this paper can be used as a simple method to predict SWCC and provide theoretical guidance for soil water management and soil collapse erosion monitoring in the collapsible loess area. Gravel mulching is an ancient mulching system with a history of more than 300 years in China. To explore the changes of soil-water content (SWC) and heat transport in watermelon gravel-mulched fields under drip irrigation, we simulated three irrigation quotas (W1, 180 m3/hm2; W2, 270 m3/hm2; and W3, 360 m3/hm2) and three irrigation frequencies (F1, three times; F2, six times; and F3, nine times) based on HYDRUS-2D. The results indicated that peak SWC increased with irrigation quota. The range of fluctuation of SWC decreased as irrigation frequency increased. The temperature of the 0–40 cm soil layer varied with air temperature, but the range of fluctuation decreased with depth. Irrigation affected the distribution of soil water, increased soil heat capacity, and reduced the impact of air temperature on soil temperature, thus delaying the impact of air temperature on soil temperature. High-frequency drip irrigation could therefore effectively improve SWC, reduce water stress during the period of watermelon growth, and effectively delay the effect of air temperature on soil temperature, providing a theoretical basis for developing reasonable irrigation strategies and regulating soil water and heat in gravel-mulched fields. The Harvard Cryo-Electron Microscopy Center for Structural Biology, which was formed as a consortium between Harvard Medical School, Boston Children’s Hospital, Dana-Farber Cancer Institute, and Massachusetts General Hospital, serves both academic and commercial users in the greater Harvard community. The facility strives to optimize research productivity while training users to become expert electron microscopists. These two tasks may be at odds and require careful balance to keep research projects moving forward while still allowing trainees to develop independence and expertise. This article presents the model developed at Harvard Medical School for running a research-oriented cryo-EM facility. Being a research-oriented facility begins with training in cryo-sample preparation on a trainee’s own sample, ideally producing grids that can be screened and optimized on the Talos Arctica via multiple established pipelines. The first option, staff assisted screening, requires no user experience and a staff member provides instant feedback about the suitability of the sample for cryo-EM investigation and discusses potential strategies for sample optimization. Another option, rapid access, allows users short sessions to screen samples and introductory training for basic microscope operation. Once a sample reaches the stage where data collection is warranted, new users are trained on setting up data collection for themselves on either the Talos Arctica or Titan Krios microscope until independence is established. By providing incremental training and screening pipelines, the bottleneck of sample preparation can be overcome in parallel with developing skills as an electron microscopist. This approach allows for the development of expertise without hindering breakthroughs in key research areas.

A concentric twin tube (CTT) can be built by placing a carbon nanotube (CNT) in another identical CNT. Different from diamondene nanotubes, a stable CTT has no inter-shell covalent bond. As a prestressed double-walled nanotube, CTT has a lower structural stability at a finite temperature. According to the molecular dynamics and first-principle calculations, (a) CTTs have three types of relaxed configurations. In a type III CTT, the inner tube buckles to produce a V-shaped cross-section, and the outer tube may be convex or concave. (b) The minimal radii of relaxed zigzag and armchair CTTs with concave outer tubes were found. (c) After relaxation, the circumferences and areas of the two tubes in a type III CTT are different from those of the corresponding ideal CNT. The area change rate (A-CR) and circumference change rate (C-CR) of the outer tube are the first-order Gaussian function of the radius of the ideal CNT (which forms the CTT), and tends to be 73.3% of A-CR or 95.3% of C-CR, respectively. For the inner tube of a CTT, the A-CR is between 29.3% and 37.0%, and the C-CR is close to 95.8%. (d) The temperature slightly influences the findings given above.

We report the near atomic resolution (3.3 Å) of the human polycystic kidney disease 2-like 1 (polycystin 2-l1) ion channel. Encoded by PKD2L1, polycystin 2-l1 is a calcium and monovalent cation-permeant ion channel in primary cilia and plasma membranes. The related primary cilium-specific polycystin-2 protein, encoded by PKD2, shares a high degree of sequence similarity, yet has distinct permeability characteristics. Here we show that these differences are reflected in the architecture of polycystin 2-l1.

Drying could change the microstructure of cement-based materials and inevitably affect their mechanical properties. The isothermal drying process of concrete at three scales and its effect on compressive behavior and microstructure were investigated. The deformations of cement paste, mortar, and concrete in the drying process all exhibit the characteristics of expansion first and then shrinkage. The porosity and average pore diameter increase after drying, which is mainly attributed to the increase of pores less than 100 nm diameter for paste and to the pores within 100~1000 nm for mortar. Drying makes paste denser, while the bonding between paste and aggregate is weakened. Microstructural studies indicate that the increase in compressive strength of concrete caused by isothermal drying is the competition result between the strengthening effect and the weakening effect, and is related to the paste content.

Rift Valley fever virus (RVFV), a Phlebovirus with a genome consisting of three single-stranded RNA segments, is spread by infected mosquitoes and causes large viral outbreaks in Africa. RVFV encodes a nucleoprotein (N) that encapsidates the viral RNA. The N protein is the major component of the ribonucleoprotein complex and is also required for genomic RNA replication and transcription by the viral polymerase. Here we present the 1.6 Å crystal structure of the RVFV N protein in hexameric form. The ring-shaped hexamers form a functional RNA binding site, as assessed by mutagenesis experiments. Electron microscopy (EM) demonstrates that N in complex with RNA also forms rings in solution, and a single-particle EM reconstruction of a hexameric N-RNA complex is consistent with the crystallographic N hexamers. The ring-like organization of the hexamers in the crystal is stabilized by circular interactions of the N terminus of RVFV N, which forms an extended arm that binds to a hydrophobic pocket in the core domain of an adjacent subunit. The conformation of the N-terminal arm differs from that seen in a previous crystal structure of RVFV, in which it was bound to the hydrophobic pocket in its own core domain. The switch from an intra- to an inter-molecular interaction mode of the N-terminal arm may be a general principle that underlies multimerization and RNA encapsidation by N proteins from Bunyaviridae. Furthermore, slight structural adjustments of the N-terminal arm would allow RVFV N to form smaller or larger ring-shaped oligomers and potentially even a multimer with a super-helical subunit arrangement. Thus, the interaction mode between subunits seen in the crystal structure would allow the formation of filamentous ribonucleocapsids in vivo. Both the RNA binding cleft and the multimerization site of the N protein are promising targets for the development of antiviral drugs.

Green hydrogen has shown great potential to power microgrids as a primary source, whereas the resilient operation methodology under extreme events remains an open area. To fill this gap, this letter establishes an operational optimization strategy towards resilient hydrogen-powered microgrids. The frequency and voltage regulation characteristics of primary hydrogen sources under droop control and their electrical-chemical conversion process with nonlinear stack efficiency are accurately modeled by piecewise linear constraints. A resilience-oriented multi-time-slot stochastic optimization model is then formulated for an economic and robust operation under changing uncertainties. Test results show that the new formulation can leverage the primary hydrogen sources to achieve a resilience and safety-assured operation plan, supplying maximum critical loads while significantly reducing the frequency and voltage variations.

Rotavirus live-attenuated vaccines, both mono- and pentavalent, generate broadly heterotypic protection. B-cells isolated from adults encode neutralizing antibodies, some with affinity for VP5*, that afford broad protection in mice. We have mapped the epitope of one such antibody by determining the high-resolution cryo-EM structure of its antigen-binding fragment (Fab) bound to the virion of a candidate vaccine strain, CDC-9. The Fab contacts both the distal end of a VP5* β-barrel domain and the two VP8* lectin-like domains at the tip of a projecting spike. Its interactions with VP8* do not impinge on the likely receptor-binding site, suggesting that the mechanism of neutralization is at a step subsequent to initial attachment. We also examined structures of CDC-9 virions from two different stages of serial passaging. Nearly all the VP4 (cleaved to VP8*/VP5*) spikes on particles from the earlier passage (wild-type isolate) had transitioned from the "upright" conformation present on fully infectious virions to the "reversed" conformation that is probably the end state of membrane insertion, unable to mediate penetration, consistent with the very low in vitro infectivity of the wild-type isolate. About half the VP4 spikes were upright on particles from the later passage, which had recovered substantial in vitro infectivity but had acquired an attenuated phenotype in neonatal rats. A mutation in VP4 that occurred during passaging appears to stabilize the interface at the apex of the spike and could account for the greater stability of the upright spikes on the late-passage, attenuated isolate. The thermal conditions of soil are important in practical agricultural production. The characteristics of heat flux, moisture content, thermal conductivity, and other soil parameters vary with temperature. This study uses VADOSE/W to create a model of heat transmission between soil and atmosphere, simulating daily changes in soil temperature using sand mulching. By using the published data to verify the model, the results show that the fitting effect is good and the reliability of the model is verified. We also used this model to determine the temporal and spatial distributions of soil temperature, temperature differences, and a temperature gradient for sand mulching and bare soil during the growing season of spring wheat. These results indicated that the sand mulching preserved heat at night. The difference in temperature at each depth and the temperature gradient became negative with the gradual increase in solar altitude. The difference in soil temperature at each depth and the temperature gradient tended to become positive by 22:00. These results indicated that the sand mulch effectively maintained the soil temperature in the morning and night during the growing season and impeded the transfer of heat at mid-day. This study provides a new method for determining the transfer of heat in sand-mulched soil, which can guide the effective regulation of soil temperature. [ABSTRACT FROM AUTHOR]

Soil-water characteristic curve (SWCC) and soil hydraulic conductivity are important soil hydrodynamic parameters, which are of great significance in production practice. In 128 soil samples collected from 0-20, 20-40 cm layers at sampling scales of 32×32 m, the authors determined the effect of different periods of time on SWCC and soil hydraulic conductivity in fields mulched gravel. The water holding capacity of soil changes dramatically throughout time, with NEW having the maximum water holding capacity. As the planting time increases, the soil water holding capacity decreases. Both van Genuchten (VG) and Gardner models can fit SWCC in different time periods well, but the fitting accuracy of the VG model is higher. Geostatistics and Kriging interpolation are used to study the spatial variability of the VG model parameters of the SWCCs. The parameters θs, α, and n show a strong spatial correlation as a whole and are slightly affected by random factors. The changing trend of soil unsaturated hydraulic conductivity (K(θ)) in gravel-mulched fields is faster than the CHECK, and the NEW has the fastest change trend. The data suggest that fields mulched with gravel retain more water, with the NEW having the most noticeable water retention effect. The study's findings can be used to investigate the regional variations in soil hydrodynamic characteristics in gravel mulched fields in arid locations. [ABSTRACT FROM AUTHOR]

6-BA, a small molecule compound of cytokinins, has been proven to delay leaf senescence in different species, including Chinese flowering cabbage; however, its specific mechanism remains relatively unknown. In this study, the application of external 6-BA delayed leaf senescence in Chinese flowering cabbage, showing that 6-BA effectively prevented the decrease in the maximum quantum yield (Fv/Fm) and overall chlorophyll content and suppressed the expression of the senescence-associated gene BrSAG12 over a 7-day period of storage. Moreover, treatment with 6-BA decreased the respiratory rate, NAD(H) content, the activities of hexose phosphate isomerase (PHI), succinate dehydrogenase (SDH), cytochrome c oxidase (CCO), and ascorbic acid oxidase (AAO) using enzyme-linked immunosorbent assay, and the transcriptional abundance of related genes by real-time quantitative polymerase chain reaction. Furthermore, 6-BA also increased the activity and expression levels of glucose-6-phosphate dehydrogenase (G6PDH) and 6-phosphate gluconate dehydrogenase (6-PGDH). The group treated with 6-BA retained elevated levels of NADP (H), ATP, total ATPase, and nicotinamide adenine dinucleotide kinase (NADK) activity, as well as the expression of respiratory enzymes. Molecular docking indicated that 6-BA hinders the glycolysis pathway (EMP), tricarboxylic acid cycle (TCA), and cytochrome pathway (CCP), and sustains elevated levels of the pentose phosphate pathway (PPP) through interactions with the PHI, SDH, 6-PGDH, G6PDH, CCO, and AAO proteins, consequently delaying postharvest leaf senescence in Chinese flowering cabbage.

Evaporation from the soil is an important component of evapotranspiration, and mulching greatly affects soil evaporation. We conducted numerical simulations to study the effect of the thickness of sand mulch on soil evaporation. We tested nine treatments: mulching with sand thicknesses of 1, 3, 5, 6, 8, 10, 15 and 20 cm plus an unmulched control (CK). Accumulated evaporation was significantly lower, and the resistance to evaporation was significantly higher, for the mulched treatments than CK. The volumetric soil water content (SWC) was significantly higher for the mulched treatments than CK, but SWC varied little for thicknesses >5 cm. Heating was slower and more uniform for the mulched treatments than for CK. With the increase of the thickness of sand, the temperature transmission was slowed down. The change of soil temperature was not obvious at thicknesses >15 cm. A thickness of 5 cm was the most effective for storing water and preserving heat. Our results provide a theoretical basis and technical guidance for the effective use and management of soil water in farmland in arid regions. [ABSTRACT FROM AUTHOR]

Abstract The rich non‐linear dynamics of the coupled oscillators (under second harmonic injection) can be leveraged to solve computationally hard problems in combinatorial optimization such as finding the ground state of the Ising Hamiltonian. While prior work on the stability of the so‐called Oscillator Ising Machines (OIMs) has used the linearization method, in this letter, the authors present a complementary method to analyze stability using the second‐order derivative test of the energy/cost function. Dynamical systems can offer a novel non-Boolean approach to computing. Specifically, the natural minimization of energy in the system is a valuable property for minimizing the objective functions of combinatorial optimization problems, many of which are still challenging to solve using conventional digital solvers. In this work, we design two oscillator-inspired dynamical systems to solve quintessential computationally intractable problems in Boolean satisfiability (SAT). The system dynamics are engineered such that they facilitate solutions to two different flavors of the SAT problem. We formulate the first dynamical system to compute the solution to the 3-SAT problem, while for the second system, we show that its dynamics map to the solution of the Max-not-all-equal (NAE)-3-SAT problem. Our work advances our understanding of how this physics-inspired approach can be used to address challenging problems in computing.

Characterization of the variability of soil moisture, salt content and organic matter content (SOM) is of great significance in agricultural production management and sustainable soil utilization. We present a case study of the variability and modelling with the depth of soil moisture, salt and SOM in a gravelsand mulched jujube orchard, using Geostatistics and Kriging interpolation. Soil moisture, salt and SOM were measured in 256 samples collected from a gravel-sand mulched jujube orchard in the 0-10, 10-20, 20-30 and 30-50 cm. Soil moisture, salt and SOM were more variable in the surface soil, due to several environmental factors, the coefficients of variation (CV) of soil were lower than 23%, indicating weak to moderate variation. The coefficient of variation of moisture and organic matter decreased with the depth and the salinity increased with the depth. There is a significant correlation between each soil layer, which decreases with the increase of the soil layer. The accuracy of the function model with depth as an independent variable and soil properties as a dependent variable is higher than 0.88. Objectives: The use of X-ray for therapeutics always raises the problem of radiation hazards to living beings. In this research, we explored the radioprotective activity of glutathione (GSH) on cognitive ability of X-ray radiated tumor-bearing mice. Methods: Forty C57BL/6 mice were chosen to establish the GL261 glioma model and randomly divided into four groups: Model group, X-ray group, Pre-GSH group and Pos-GSH group. Morris water maze test was used to test cognitive ability. Moreover, histopathological observation of hippocampus was observed by hematoxylin and eosin (HE) staining. The protein expression of choline acetyl transferase (ChAT) was measured by western blot, simultaneously the contents of acetylcholinesterase (Ach), superoxide dismutase (SOD), methane dicarboxylic aldehyde (MDA),TNF-α and IL-6 were detected by the respective kit. Results: There was a significant difference in X-ray group of the escape latency from the Model group (P<0.05). Besides, HE staining revealed that nucleus in hippocampus cells were pyknotic, glial cells were hyperplastic and the nerve cells were swelling in X-ray group. In Xray group the expression of ChAT and Ache were decreased versus Model group. Finally, the cognitive ability in Pre-GSH and Pos-GSH group was enhanced than X-ray group, in which the cognitive ability of Pos-GSH group was higher than the Pre-GSH group. Discussion: X-ray impaired the brain tissues and cognitive ability of tumor-bearing mice. The damages of brain tissues were alleviated by Pre-GSH and Pos-GSH protection and the efficacy of Pos-GSH protection was superior to Pre-GSH protection. The transient receptor potential ion channel subfamily M, member 7 (TRPM7), is a ubiquitously expressed protein that is required for mouse embryonic development. TRPM7 contains both an ion channel and an α-kinase. The channel domain comprises a nonselective cation channel with notable permeability to Mg2+ and Zn2+. Here, we report the closed state structures of the mouse TRPM7 channel domain in three different ionic conditions to overall resolutions of 3.3, 3.7, and 4.1 Ä. The structures reveal key residues for an ion binding site in the selectivity filter, with proposed partially hydrated Mg2+ ions occupying the center of the conduction pore. In high [Mg2+], a prominent external disulfide bond is found in the pore helix, which is essential for ion channel function. Our results provide a structural framework for understanding the TRPM1/ 3/6/7 subfamily and extend the knowledge base upon which to study the diversity and evolution of TRP channels. Transient receptor potential melastatin subfamily member 4 (TRPM4) is a widely distributed, calcium-activated, monovalentselective cation channel. Mutations in human TRPM4 (hTRPM4) result in progressive familial heart block. Here, we report the electron cryomicroscopy structure of hTRPM4 in a closed, Na+- bound, apo state at pH 7.5 to an overall resolution of 3.7 Å. Five partially hydrated sodium ions are proposed to occupy the center of the conduction pore and the entrance to the coiled-coil domain. We identify an upper gate in the selectivity filter and a lower gate at the entrance to the cytoplasmic coiled-coil domain. Intramolecular interactions exist between the TRP domain and the S4- S5 linker, N-terminal domain, and N and C termini. Finally, we identify aromatic interactions via π-π bonds and cation-π bonds, glycosylation at an N-linked extracellular site, a pore-loop disulfide bond, and 24 lipid binding sites. Vegetation deterioration and soil loss are the main causes of more precipitation leakages and surface water shortages in degraded karst areas. In order to improve the utilization of water resources in such regions, water storage engineering has been considered; however, site selection and cost associated with the special karstic geological structure have made this difficult. According to the principle of the Soil Plant Atmosphere Continuum, increasing both vegetation cover and soil thickness would change water cycle process, resulting in a transformation from leaked blue water (liquid form) into green water (gas or saturated water form) for terrestrial plant ecosystems, thereby improving the utilization of water resources. Using the Soil Vegetation Atmosphere Transfer model and the geographical distributed approach, this study simulated the conversion from leaked blue water (leakage) into green water in the environs of Guiyang, a typical degraded karst area. The primary results were as follows: (1) Green water in the area accounted for <50% of precipitation, well below the world average of 65%; (2) Vegetation growth played an important role in converting leakage into green water; however, once it increased to 56%, its contribution to reducing leakage decreased sharply; (3) Increasing soil thickness by 20 cm converted the leakage considerably. The order of leakage reduction under different precipitation scenarios was dry year > normal year > rainy year. Thus, increased soil thickness was shown effective in improving the utilization ratio of water resources and in raising the amount of plant ecological water use; (4) The transformation of blue water into green water, which avoids constructions of hydraulic engineering, could provide an alternative solution for the improvement of the utilization of water resources in degraded karst area. Based on the meteorological data from 46 stations in the Hai River Basin (HRB) from 1961-2010, the annual and seasonal variation of reference evapotranspiration was analyzed. The sensitivity coefficients combined with the detrend method were used to discuss the dominant factor affecting the reference evapotranspiration (ETo). The obtained results indicate that the annual reference evapotranspiration is dominated by the decreasing trends at the confidence level of 95% in the southern and eastern parts of the HRB. The sensitivity order of climatic variables to ETo from strong to weak is: relativity humidity, temperature, shortwave radiation and wind speed, respectively. However, comprehensively considering the sensitivity and its variation strength, the detrend analysis indicates that the decreasing trends of ETo in eastern and southern HRB may be caused mainly by the decreasing wind speed and shortwave radiation. As for the relationship between human activity and the trend of ETo, we found that ETo decreased more significantly on the plains than in the mountains. By contrast, the population density increased more considerably from 2000 to 2010 on the plains than in the mountains. Therefore, in this paper, the correlation of the spatial variation pattern between ETo and population was further analyzed. The spatial correlation coefficient between population and the trend of ETo is -0.132, while the spatial correlation coefficient between the trend of ETo and elevation, temperature, shortwave radiation and wind speed is 0.667, 0.668, 0.749 and 0.416, respectively. This suggests that human activity has a certain influence on the spatial variation of ETo, while natural factors play a decisive role in the spatial variation of reference evapotranspiration in this area. Accurate and robust scale estimation in visual object tracking is a challenging task. To obtain a scale estimation of the target object, most methods rely either on a multi-scale searching scheme or on refining a set of predefined anchor boxes. These methods require heuristically selected parameters, such as scale factors of the multi-scale searching scheme, or sizes and aspect ratios of the predefined candidate anchor boxes. On the contrary, a centerness-aware anchor-free tracker (CAT) is designed in this work. First, the location and scale of the target object are predicted in an anchor-free fashion by decomposing tracking into parallel classification and regression problems. The proposed anchor-free design obviates the need for hyperparameters related to the anchor boxes, making CAT more generic and flexible. Second, the proposed centerness-aware classification branch can identify the foreground from the background while predicting the normalized distance from the location within the foreground to the target center, i.e., the centerness. The proposed centerness-aware classification branch improves the tracking accuracy and robustness significantly by suppressing low-quality state estimates. The experiments show that our centerness-aware anchor-free tracker, with its appealing features, achieves salient performance in a wide variety of tracking scenarios.

This paper considers the problem of stabilizing switched time-delay linear systems through a state-dependent switching strategy. In contrast to the existing works, we adopt a less restrictive assumption of the system, and show that this assumption is sufficient to guarantee asymptotic stability of the considered system under the min-projection switching strategy. Our results also imply that the min-projection switching strategy, originally designed for delay-free switched systems, is robust with respect to small state delays. An optimization problem is formulated to estimate the upper bound of the tolerable time delay. Numerical examples are presented to show that our method is applicable to a larger class of switched systems and leads to a greater delay bound.

To our knowledge, no structural study to date has characterized, in an intact receptor, the coupling of conformational change in extracellular domains through a single-pass transmembrane domain to conformational change in cytoplasmic domains. Here we examine such coupling, and its unexpected complexity, using nearly full-length epidermal growth factor receptor (EGFR) and negative-stain EM. The liganded, dimeric EGFR ectodomain can couple both to putatively active, asymmetrically associated kinase dimers and to putatively inactive, symmetrically associated kinase dimers and monomers. Inhibitors that stabilize the active or inactive conformation of the kinase active site, as well as mutations in the kinase dimer interface and a juxtamembrane phosphorylation site, shift the equilibrium among the three kinase association states. This coupling of one conformation of an activated receptor ectodomain to multiple kinase-domain arrangements reveals previously unanticipated complexity in transmembrane signaling and facilitates regulation of receptor function in the juxtamembrane and cytoplasmic environments. Negative stain electron microscopy (EM) and adhesion assays show that ct,3 Integrin activation requires headpiece opening as well as extension. An extension-inducing Fab to the 2 leg, in combination with representative activating and inhibitory Fabs, were examined for effect on the equilibrium between the open and closed head- piece conformations. The two activating Fabs stabilized the open headpiece conformation. Conversely, two different inhibitory Fabs stabilized the closed headpiece conformation. Adhesion assays revealed that a42 in the extended-open headpiece conformation had high affinity for ligand, whereas both the bent conformation and the extended-closed headpiece conformation represented the low affinity state. We evaluated imaging plates (IPs) and the DITABIS Micron scanner for their use in recording images of negatively stained single-particle specimens and electron diffraction patterns of two-dimensional crystals. We first established the optimal imaging and read-out conditions for images of negatively stained single-particle specimens using the signal-to-noise ratio of the images as the evaluation criterion. We found that images were best recorded on IPs at a magnification of 67 000×, read out with a gain setting of 20 000 and a laser power setting of 30% with subsequent binning over 2 × 2 pixels. Our results show that for images of negatively stained specimens, for which the resolution is limited to ∼20 Å, IPs are a good alternative to EM film. We also compared IPs with a 2K × 2K Gatan charge-coupled device (CCD) camera for their use in recording electron diffraction patterns of sugar-embedded two-dimensional crystals. Diffraction patterns of aquaporin-0 recorded on IPs and with the CCD camera showed reflections beyond 3 Å and had similar RFriedel as well as Rmerge values. IPs can thus be used to collect diffraction patterns, but CCD cameras are more convenient and remain the best option for recording electron diffraction patterns. [ABSTRACT FROM PUBLISHER]

Rotavirus, a nonturreted member of the Reoviridae, is the causative agent of severe infantile diarrhea. The double-stranded RNA genome encodes six structural proteins that make up the triple-layer particle. X-ray crystallography has elucidated the structure of one of these capsid proteins, VP6, and two domains from VP4, the spike protein. Complementing this work, electron cryomicroscopy (cryoEM) has provided relatively low-resolution structures for the triple-layer capsid in several biochemical states. However, a complete, high-resolution structural model of rotavirus remains unresolved. Combining new structural analysis techniques with the subnanometer-resolution cryoEM structure of rotavirus, we now provide a more detailed structural model for the major capsid proteins and their interactions within the triple-layer particle. Through a series of intersubunit interactions, the spike protein (VP4) adopts a dimeric appearance above the capsid surface, while forming a trimeric base anchored inside one of the three types of aqueous channels between VP7 and VP6 capsid layers. While the trimeric base suggests the presence of three VP4 molecules in one spike, only hints of the third molecule are observed above the capsid surface. Beyond their interactions with VP4, the interactions between VP6 and VP7 subunits could also be readily identified. In the innermost T=1 layer composed of VP2, visualization of the secondary structure elements allowed us to identify the polypeptide fold for VP2 and examine the complex network of interactions between this layer and the T=13 VP6 layer. This integrated structural approach has resulted in a relatively high-resolution structural model for the complete, infectious structure of rotavirus, as well as revealing the subtle nuances required for maintaining interactions in such a large macromolecular assembly. The threat of a pandemic outbreak of influenza virus A H5N1 has become a major concern worldwide. The nucleoprotein (NP) of the virus binds the RNA genome and acts as a key adaptor between the virus and the host cell. It, therefore, plays an important structural and functional role and represents an attractive drug target. Here, we report the 3.3-Å crystal structure of H5N1 NP, which is composed of a head domain, a body domain, and a tail loop. Our structure resolves the important linker segments (residues 397-401, 429-437) that connect the tail loop with the remainder of the molecule and a flexible, basic loop (residues 73-91) located in an arginine-rich groove surrounding Arg150. Using surface plasmon resonance, we found the basic loop and arginine-rich groove, but mostly a protruding element containing Arg174 and Arg175, to be important in RNA binding by NP. We also used our crystal structure to build a ring-shaped assembly of nine NP subunits to model the miniribonucleo-protein particle previously visualized by electron microscopy. Our study of H5N1 NP provides insight into the oligomerization interface and the RNA-binding groove, which are attractive drug targets, and it identifies the epitopes that might be used for universal vaccine development.
