Abstract Objective Recent studies indicated that transmembrane protein 40 (TMEM40) is associated with several types of cancers but is not clear in cervical cancer (CC). The study aimed to examine the role of TMEM40 in CC and related mechanisms. Methods The expression of TMEM40 in CC tissues and cell lines was studied with western blot and real-time quantitative RT-PCR. The effect of TMEM40 on proliferation was evaluated by CCK-8, EdU and colony formation assay. The migration, invasion, cell cycle and apoptosis of CC cells were studied with wound healing, transwell assays and flow cytometry. Tumor growth was evaluated in vivo using a xenogenous subcutaneously implant model. Results The results revealed that the TMEM40 elevation in CC tissues and cell lines was closely correlated with tumor size and lymph node metastasis in clinical patients. Upregulation of TMEM40 with OE-TMEM40 vector promoted the invasion, migration and proliferation, inhibited the apoptosis and led to distinct S cell cycle arrest in CC cell lines. Silencing TMEM40 with shRNA inhibited the invasion, migration and proliferation, promoted apoptosis and led to a G0/G1 cell cycle arrest in CC cell lines. Silence of TMEM40 downregulated the expression of c-MYC, Cyclin D1, matrix metalloproteinase-1 (MMP-1) and matrix metalloproteinase-9 (MMP-9), but in contrast, activated p53 and several apoptosis related proteins such as p53, Caspase-3, Caspase-9 and PARP1. In addition, TMEM40 silencing dramatically decreased tumor growth in mice models. Conclusion The present study demonstrates that TMEM40 upregulation can be a potential prognostic biomarker and contribute to CC development.

Abstract The brown planthopper (BPH), Nilaparvata lugens, is an important pest that affects rice (Oryza sativa) production in Asia. The flavone tricin (5,7,4′-trihydroxy-3′,5′-dimethoxy flavone) is a valuable secondary metabolite commonly found in rice plants that can defend rice plants against infestation by BPH. BPH damage can reduce the metabolic level of tricin in rice. Our preliminary transcriptome research results showed that BPH salivary protein 7 (NlSP7), is highly responsive to tricin stimuli. However, the function of NlSP7 in mediating the interaction between the rice plant and the BPH is unknown. In this study, we cloned the NlSP7 gene in N. lugens and found that its mRNA level was greater in the presence of high tricin content than low tricin content, regardless of whether the BPHs were fed a rice plant diet or an artificial diet containing 100 mg/L tricin. Knocking down NlSP7 resulted in BPH individuals spending more time in the non-penetration and pathway phase, and less time feeding on the phloem of rice plants. These changes decreased BPH food intake, feeding behavior, and fitness, as well as the tricin content of the rice plants. These findings demonstrate that the salivary protein 7 of BPH functions as an effector for tricin metabolism in rice.

Resistant crop varieties can usually decrease the population density of insect pests; however, they can also easily cause the occurrence of highly virulent pest populations when repeatedly grown. Whether herbivorous insects feeding intermittently on a susceptible variety affects their subsequent virulence has rarely been investigated. In this paper, we examined the variations in the virulence of the brown planthopper (BPH), Nilaparvata lugens (Stål) (Hemiptera: Delphacidae), by alternately rearing three resistant rice varieties, Mudgo, ASD7, and Rathu Heenati, with a susceptible rice variety (TN1) in indoor experiments. The results showed that, while the susceptible rice variety was used in alternate rearing for several generations, the BPHs exhibited a higher intrinsic rate of increase (rm) and were identified as less virulent to all three resistant varieties. Such virulence reduction by experience with a susceptible variety could delay the progression of resistance-breaking toward resistant varieties. The results suggested that careful alternation with susceptible varieties in fields is a potential method for pest variety-resistance management.

ABSTRACT: There are many factors affecting the instability of the submarine hydrate-bearing slope (SHBS), and the interaction with hydrate is very complicated. In this paper, the mechanical mechanism of the static liquefaction and instability of submarine slope caused by the dissociation of natural gas hydrate (NGH) resulting in the rapid increase of pore pressure of gas hydrate-bearing sediments (GHBS) and the decrease of effective stress are analyzed based on the time series and type of SHBS. Then, taking the typical submarine slope in the northern South China Sea as an example, four important factors affecting the stability of SHBS are selected, such as the degree of hydrate dissociation, the depth of hydrate burial, the thickness of hydrate, and the depth of seawater. According to the principle of orthogonal method, 25 orthogonal test schemes with 4 factors and 5 levels are designed and the safety factors of submarine slope stability of each scheme are calculated by using the strength reduction finite element method. By means of the orthogonal design range analysis and the variance analysis, sensitivity of influential factors on stability of SHBS are obtained. The results show that the degree of hydrate dissociation is the most sensitive, followed by hydrate burial depth, the thickness of hydrate and the depth of seawater. Finally, the concept of gas hydrate critical burial depth is put forward according to the influence law of gas hydrate burial depth, and the numerical simulation for specific submarine slope is carried out, which indicates the existence of critical burial depth.

Fall armyworm is recognized as one of most highly destructive global agricultural pests. In January 2020, it had first invaded Australia, posing a significant risk to its biosecurity, food security, and agricultural productivity. In this study, the migration paths and wind systems for the case of fall armyworm invading Australia were analyzed using a three-dimensional trajectory simulation approach, combined with its flight behavior and NCEP meteorological reanalysis data. The analysis showed that fall armyworm in Torres Strait most likely came from surrounding islands of central Indonesia on two occasions via wind migration. Specifically, fall armyworm moths detected on Saibai and Erub Islands might have arrived from southern Sulawesi Island, Indonesia, between January 15 and 16. The fall armyworm in Bamaga most likely arrived from the islands around Arafura Sea and Sulawesi Island of Indonesia, between January 26 and 27. The high risk period for the invasion of fall armyworm is only likely to have occurred in January–February due to monsoon winds, which were conducive to flight across the Timor Sea towards Australia. This case study is the first to confirm the immigration paths and timing of fall armyworm from Indonesia to Australia via its surrounding islands.

Abstract Background Bladder cancer (BCa) is one of the most common cancers in the urinary system among the world. Previous studies suggested that TMEM40 expression level was significantly associated with clinicopathological parameters including histological grade, clinical stage and pT status of bladder cancer. However, the molecular mechanism of TMEM40 in BCa remains poorly understood. Methods Real-time quantitative RT-PCR (qRT-PCR) and western blot (WB) were used to examine the expression levels of TMEM40 in BCa tissues, paired non-cancer tissues and cell lines. A series of experiments, including CCK-8, wound healing, flow cytometry, transwell and EdU assays were performed to assess the effects of TMEM40 on cell proliferation, cell cycle and apoptosis, migration and invasion. In addition, tumor growth was evaluated in vivo using a xenogenous subcutaneously implant model. All statistical analyses were executed by using the SPSS 20.0 software. All experimental data from three independent experiments were analyzed by Student’s t test and results were expressed as mean ± standard deviation. Results In this study, we identified the role of TMEM40 in the tumorigenesis of bladder cancer and found that it was upregulated in bladder cancer tissues and cell lines, compared with their normal counterparts. The results demonstrated that effective silence of TMEM40 expression suppressed cell proliferation, blocked G1-to-S cell cycle transition, and inhibited cell migration and invasion in human bladder 5637 and EJ cell lines. Consistently, in vivo data showed that TMEM40 silencing could dramatically decreased tumor growth. Further study revealed that TMEM40 knockdown resulted in accumulation of p53 and p21 protein and decrease of c-MYC and cyclin D1 protein. Conclusion These data suggest that TMEM40 represents a potential oncogene, which exert a crucial role in the proliferation and apoptosis via the p53 signaling pathway in BCa, thus probably serve as a novel candidate biomarker and a potential therapeutic target for patients with BCa.

Cyrtorhinus lividipennis Reuter (Hemiptera: Miridae: Orthotylinae), including nymphs and adults, are one of the dominant predators and have a significant role in the biological control of leafhoppers and planthoppers in irrigated rice. In this study, we investigated the antennal morphology, structure and sensilla distribution of C. lividipennis in different instars using scanning electron microscopy. The antennae of both five different nymphal stages and adults were filiform in shape, which consisted of the scape, pedicel and flagellum with two flagellomeres. There were significant differences found in the types of antennal sensilla between nymphs and adults. The multiporous placodea sensilla (MPLA), basiconica sensilla II (BAS II), and sensory pits (SP) only occurred on the antennae of adult C. lividipennis of both sexes. Moreover, there was chaetica sensilla III (CHA III) only observed in males. Sixteen types of antennal sensilla were recorded altogether. They were microtrichia sensilla (MIC), three types of trichoidea sensilla (TRI I-III), three types of chaetica sensilla (CHA I-III), three types of basiconica sensilla (BAS I-III), two types of coeloconica sensilla (COE I and COE II), placodea sensilla (PLA), campaniform sensilla (CAM), MPLA, and SP. In the five different nymphal stages of C. lividipennis, the length of their antennae was significantly increased with the increase of the instar, as well as the number of the TRI II and TRI III. Moreover, sexual dimorphism usually occurred not only in the distribution (CHA III and SP) and the number of antennal sensilla (MIC, BAS II, TRI II, TRI III and MPLA), but also in the length of flagellum (F1 and F2). The possible functions of antennal sensilla are discussed. Those observations could contribute to a better understanding of the development of the olfactory system, and facilitate future studies on the antennal functions in C. lividipennis.

Recently, both global and local classical randomness-assisted projective measurement protocols have been employed to share Bell nonlocality of an entangled state among multiple sequential parties. Unlike Bell nonlocality, Einstein–Podolsky–Rosen (EPR) steering exhibits distinct asymmetric characteristics and serves as the necessary quantum resource for one-sided device-independent quantum information tasks. In this work, we propose a projective measurement protocol and investigate the shareability of EPR steering with steering radius criterion theoretically and experimentally. Our results reveal that arbitrarily many independent parties can share one-way steerability using projective measurements, even when no shared randomness is available. Furthermore, by leveraging only local randomness, asymmetric two-way steerability can also be shared. Our work not only deepens the understanding of the role of projective measurements in sharing quantum correlations but also opens up a new avenue for reutilizing asymmetric quantum correlations.
