Abstract Objective To investigate the diagnostic value of combined 99Tcm-DX lymphoscintigraphy and CT lymphangiography (CTL) in primary chylopericardium. Methods Fifty-five patients diagnosed with primary chylopericardium clinically were retrospectively analyzed. 99Tcm-DX lymphoscintigraphy and CTL were performed in all patients. Primary chylopericardium was classified into three types, according to the 99Tcm-DX lymphoscintigraphy results. Indoor environments can pose a substantial respiratory infection risk, especially in densely populated areas. It is therefore vital to evaluate and predict infection risks for mobile individuals engaging in indoor activities. However, previous studies have primarily focused on assessing viral transmission probabilities between relatively stationary individuals, disregarding the dynamic features of pedestrian trajectories. To address this issue, we propose an approach that uses precise trajectory data and digital twin technology to evaluate and forecast indoor infection risks for mobile individuals. Our approach involves the utilization of high-precision trajectory data to develop a time-varying virus density field map (VDFM), forming the basis for infection risk assessment. Additionally, we present a deep-learning model that employs the transformer method to predict future time-varying VDFMs and associated infection risks. Furthermore, we introduce digital twin to comprehend real-time interactions between the physical and digital realms within the structure. To validate our approach, we conducted a case study at the Wuhan International Exposition Center, serving as the control room. We performed a sensitivity analysis on various preventive measures, including mask-wearing, social distancing, and indoor infection control. The results highlighted the significant impact of these measures on individual infection risks associated with indoor activities. Our research contributes to the development of tailored prevention and control strategies, which play a critical role in mitigating the spread of respiratory infections. Thus, this study holds significant implications for technology-facilitated public health services.

Background & Aims: A high human cytomegalovirus (HCMV) infection rate accompanied by an increased level of bile duct damage is observed in the perinatal period. The possible mechanism was investigated. Methods: A total of 1,120 HCMV-positive and 9,297 HCMV-negative children were recruited, and depending on age, their liver biochemistry profile was compared. Fetal and infant biliary epithelial cells (F-BECs and I-BECs, respectively) were infected with HCMV, and the differences in cells were revealed by proteomic analysis. Protein–protein interactions were examined by coimmunoprecipitation and mass spectrometry analyses. A murine cytomegalovirus (MCMV) infection model was established to assess treatment effects. Results: Perinatal HCMV infection significantly increased the level of bile duct damage. Neonatal BALB/c mice inoculated with MCMV showed obvious inflammation in the portal area with an abnormal bile duct structure. Proteomics analysis showed higher CD14 expression in F-BECs than in I-BECs. CD14 siRNA administration hindered HCMV infection, and CD14-knockout mice showed lower MCMV-induced bile duct damage. HCMV infection upregulated CD55 and poly ADP-ribose polymerase-1 (PARP-1) expression in F-BECs. Coimmunoprecipitation and mass spectrometry analyses revealed formation of the CD14–CD55 complex. siRNA-mediated inhibition of CD55 expression reduced sCD14-promoted HCMV replication in F-BECs. In MCMV-infected mice, anti-mouse CD14 antibody and PARP-1 inhibitor treatment diminished cell death, ameliorated bile duct damage, and reduced mortality. Conclusions: CD14 facilitates perinatal HCMV infection in BECs via CD55, and PARP-1-mediated cell death was detected in perinatal cytomegalovirus-infected BECs. These results provide new insight into the treatment of perinatal HCMV infection with bile duct damage. Impact and implications: Perinatal human cytomegalovirus (HCMV) infection is associated with bile duct damage, but the underlying mechanism is still unknown. We discovered that CD14 expression is increased in biliary epithelial cells during perinatal HCMV infection and facilitates viral entry through CD55. We also detected PARP-1-mediated cell death in perinatal HCMV-infected biliary epithelial cells. We showed that blocking CD14 or inhibiting PARP-1 reduced bile duct damage and mortality in a mouse model of murine cytomegalovirus infection. Our findings provide a new insight into therapeutic strategies for perinatal HCMV infection.

Coal tar pitch (CTP) has become an ideal choice in the preparation of anode precursors for lithium-ion batteries (LIBs) and sodium-ion batteries (SIBs) because of its abundant carbon content, competitive pricing and adjustable structure properties. In this paper, sulfurized pitch-based carbon (SPC-800) was obtained by allowing CTP to react with sulfur at 350 °C and subsequently achieve carbonization at 800 °C. SPC-800 was more disordered and had a larger layer spacing than carbonized CTP (PC-800). Upon utilization as an anode for LIBs, SPC-800 possessed a higher reversible specific capacity (478.1 mAh g−1 at 0.1 A g−1), while utilization in SIBs displayed a capacity of 220.9 mAh g−1 at 20 mA g−1. This work is an important guide to the design of high-performance anodes suitable for use with both LIBs and SIBs.

Abstract Background Chylothorax is a condition that can be challenging to diagnose due to its nonspecific clinical presentation. Several biochemical parameters of chylous pleural effusion have been identified as important indicators for the diagnosis of chylothorax. Lymphoscintigraphy is utilized to assess chylothorax and determine the location of chyle leakage. The present study aimed to evaluate the correlation between the biochemical parameters of chylous pleural effusion and 99mTc-dextran (99mTc-DX) lymphoscintigraphy in diagnosing chylothorax. Material and methods A total of 120 patients were enrolled in the study, 83 of the patients with unilateral chylothorax, and 37 with bilateral chylothorax. All patients underwent both 99mTc-DX lymphoscintigraphy and pleural effusion laboratory analysis. The 99mTc-DX lymphoscintigraphy images were categorized as positive or negative groups based on the presence or absence of abnormal radioactive tracer accumulation in the thorax, respectively. The biochemical parameters of the two groups were subsequently compared. Results Among these patients, 101 (84.17%) had exudative effusions, while 19 (15.83%) had transudative effusions, as determined by the levels of pleural effusion protein, lactate dehydrogenase and cholesterol. Abnormal tracer accumulation in thorax was observed in 82 patients (68.33%). Our findings indicated that lymphoscintigraphy results were not associated with exudative and transudative chylothorax (P = 0.597). The lymphoscintigraphy positive group displayed significantly higher levels of pleural effusion triglyceride and pleural effusion triglyceride/serum triglyceride ratio in all biochemical parameters, compared to the negative group (P = 0.000 and P = 0.005). We identified cutoff values of 2.870 mmol/L for pleural effusion triglycerides and 4.625 for pleural effusion triglyceride/serum triglyceride ratio, respectively, which can facilitate differentiating the positive and negative cases on lymphoscintigraphy. Conclusion Lymphoscintigraphy technique is a dependable diagnostic tool for the qualitative assessment of chylous pleural effusion. Higher pleural effusion triglyceride level and pleural effusion triglyceride/serum triglyceride ratio indicate a positive result in patients with chylothorax on lymphoscintigraphy, with the cutoff values of 2.870 mmol/L and 4.625 aiding in the diagnosis.

Background & Aims: Our previous study indicated that CD177+ neutrophil activation has a vital role in the pathogenesis of biliary atresia (BA), which is partially ameliorated by N-acetylcysteine (NAC) treatment. Here, we evaluated the clinical efficacy of NAC treatment and profiled liver-resident immune cells via single cell RNA-sequencing (scRNA-seq) analysis to provide a comprehensive immune landscape of NAC-derived immune regulation. Methods: A pilot clinical study was conducted to evaluate the potential effects of intravenous NAC treatment on infants with BA, and a 3-month follow-up was carried out to assess treatment efficacy. scRNA-seq analysis of liver CD45+ immune cells in the control (n = 4), BA (n = 6), and BA + NAC (n = 6) groups was performed and the effects on innate cells, including neutrophil and monocyte–macrophage subsets, and lymphoid cells were evaluated. Results: Intravenous NAC treatment demonstrated beneficial efficacy for infants with BA by improving bilirubin metabolism and bile acid flow. Two hepatic neutrophil subsets of innate cells were identified by scRNA-seq analysis. NAC treatment suppressed oxidative phosphorylation and reactive oxygen species production in immature neutrophils, which were transcriptionally and functionally similar to CD177+ neutrophils. We also observed the suppression of hepatic monocyte-mediated inflammation, decreased levels of oxidative phosphorylation, and M1 polarisation in Kupffer-like macrophages by NAC. In lymphoid cells, enhancement of humoral immune responses and attenuation of cellular immune responses were observed after NAC treatment. Moreover, cell–cell interaction analysis showed that innate/adaptive proinflammatory responses were downregulated by NAC. Conclusions: Our clinical and scRNA-seq data demonstrated that intravenous NAC treatment partially reversed liver immune dysfunction, alleviated the proinflammatory responses in BA by targeting innate cells, and exhibited beneficial clinical efficacy. Impact and implications: BA is a serious liver disease that affects newborns and has no effective drug treatment. In this study, scRNA-seq showed that NAC treatment can partially reverse the immune dysfunction of neutrophil extracellular trap-releasing CD177+ neutrophils and Kupffer cells, and lower the inflammatory responses of other innate immune cells in BA. In consequence, intravenous NAC treatment improved the clinical outcomes of patients with BA in term of bilirubin metabolism.

PurposeTo review our single-center surgical outcomes of redo operations after failed Rex shunt procedures.MethodsFrom September 2017 to October 2021, a total of 20 patients (11 males, 9 females; median age: 8.6 years) with Rex shunt occlusions were admitted to our hospital. Two of these patients were previously operated on in our hospital, and the remaining 18 were from other centers. All patients underwent repeat operations after detailed preoperative evaluations.ResultsPreoperative wedged hepatic vein portography (WHVP) was conducted for 18 patients. Thirteen patients exhibited well-developed Rex recessus and intrahepatic portal vein during WHPV examination, consistent with the intraoperative exploration results. Fifteen patients (75%, 15/20) underwent redo-Rex shunt, four underwent Warren shunt and one underwent devascularization surgery. During the redo-Rex shunt operations, the left internal jugular veins (IJV) were used as bypass grafts in 11 patients; the intra-abdominal veins were used in 4 patients. The patients were followed up for 12–59 months (mean, 24.8 months). After redo Rex shunts, the grafts were patent in 14 patients (93.3%, 14/15), but 1 graft had thrombosis (6.7%, 1/15). Three patients suffered from postoperative anastomotic stenosis, and all of the stenosis was relieved with balloon dilatations. After re-Rex shunts, esophageal varices and spleen size were substantially reduced, and the platelet count significantly increased. Postoperative graft thrombosis was found in 1 patient after Warren shunt (1/4, 25%), and there was no graft stenosis. Compared with Warren surgery, patients who underwent re-Rex shunt had a significantly higher rate of platelet increase.ConclusionsRedo-rex shunts can be finished in most patients with failed Rex shunts. Re-Rex shunt is a preferred surgical choice after a failed Rex shunt when a good bypass graft is available, and the surgical success rate can reach more than 90%. A suitable bypass graft is essential for a successful redo Rex shunt. Preoperative WHVP is recommended for the design of a redo surgical plan preoperatively.

Extra-intestinal Pathogenic Escherichia coli (ExPEC) is defined as an extra-intestinal foodborne pathogen, and several dominant sequence types (STs) ExPEC isolates are highly virulent, with zoonotic potential. Bacteria extracellular vesicles (EVs) carry specific subsets of molecular cargo, which affect various biological processes in bacteria and host. The mechanisms of EVs formation in ExPEC remains to be elucidated. Here, the purified EVs of ExPEC strains of different STs were isolated with ultracentrifugation processes. A comparative analysis of the strain proteomes showed that cytoplasmic proteins accounted for a relatively high proportion of the proteins among ExPEC EVs. The proportion of cytoplasm-carrying vesicles in ExPEC EVs was calculated with a simple green fluorescent protein (GFP) expression method. The RecA/LexA-dependent SOS response is a critical mediator of generation of cytoplasm-carrying EVs. The SOS response activates the expression of prophage-associated endolysins, Epel1, Epel2.1, and Epel2.2, which triggered cell lysis, increasing the production of ExPEC cytoplasm-carrying EVs. The repressor LexA controlled directly the expression of these endolysins by binding to the SOS boxes in the endolysin promoter regions. Reducing bacterial viability stimulated the production of ExPEC EVs, especially cytoplasm-carrying EVs. The imbalance in cell division caused by exposure to H2O2, the deletion of ftsK genes, or t6A synthesis defects activated the RecA/LexA-dependent SOS response, inducing the expression of endolysins, and thus increasing the proportion of cytoplasm-carrying EVs in the total ExPEC EVs. Antibiotics, which decreased bacterial viability, also increase the production of ExPEC cytoplasm-carrying EVs through the SOS response. Changes in the proportion of cytoplasm-carrying EVs affected the total DNA content of ExPEC EVs. When macrophages are exposed to a higher proportion of cytoplasm-carrying vesicles, ExPEC EVs were more cytotoxic to macrophages, accompanied with more-severe mitochondrial disruption and a higher level of induced intrinsic apoptosis. In summary, we offered comprehensive insight into the proteome analysis of ExPEC EVs. This study demonstrated the novel formation mechanisms of E. coli cytoplasm-carrying EVs.

Abstract Background Biliary atresia (BA) is a severe cholangiopathy possibly resulting from virus-induced and immune-mediated injury of the biliary system. IFN-γ, secreted from CD4+ Th1 cells and CD8+ cytotoxic T cells, is a major mediator of liver pathology. Programmed death protein-1 (PD-1) signaling suppresses T cell function. However, how PD-1 modify T cell function in BA remains incompletely understood. Methods Frequencies of PD-1 expressing CD4+ and CD8+ T cells were analyzed in the liver and blood from BA and control subjects. Associations of PD-1+CD4+/CD8+T cell abundances with liver function indices were measured. Function of PD-1 was measured by administration of an anti-PD-1 antibody in a Rhesus Rotavirus (RRV)-induced BA model. Survival, histology, direct bilirubin, liver immune cell subsets and cytokine production were analyzed. Results PD-1 was significantly upregulated in CD4+ and CD8+ T cells in patients with BA compared with control subjects. PD-1 expression in T cells was negatively associated with IFN-γ concentration in liver (PD-1+CD4+T cells in liver vs. IFN-γ concentration, r = − 0.25, p = 0.05; PD-1+CD8+T cells in liver vs. IFN-γ concentration, r = − 0.39, p = 0.004). Blockade of PD-1 increased IFN-γ expression in CD4+ T and CD8+ T cells (RRV vs. anti-PD-1 treated RRV mice: 11.59 ± 3.43% vs. 21.26 ± 5.32% IFN-γ+ in hepatic CD4+T cells, p = 0.0003; 9.33 ± 4.03% vs. 22.55 ± 7.47% IFN-γ+ in hepatic CD8+T cells, p = 0.0001), suppressed bilirubin production (RRV vs. anti-PD-1 treated RRV mice: 285.4 ± 47.93 vs. 229.8 ± 45.86 μmol/L total bilirubin, p = 0.01) and exacerbated liver immunopathology. Conclusions PD-1 plays a protective role in infants with BA by suppressing IFN-γ production in T cells. Increasing PD-1 signaling may serve as a therapeutic strategy for BA.

Abstract Background Extrahepatic portal vein obstruction (EHPVO) is the most important cause of hematemesis in children. Intrahepatic left portal vein and superior mesenteric vein anastomosis, also known as meso-Rex bypass (MRB), is becoming the gold standard treatment for EHPVO. We analyzed the value of preoperative computed tomography (CT) in determining whether MRB is feasible in children with EHPVO. Results We retrieved data on 76 children with EHPVO (50 male, 26 female; median age, 5.9 years) who underwent MRB (n = 68) or the Warren procedure (n = 8) from 2013 to 2019 and retrospectively analyzed their clinical and CT characteristics. The Rex recess was categorized into four subtypes (types 1–4) depending on its diameter in CT images. Of all 76 children, 7.9% had a history of umbilical catheterization and 1.3% had leukemia. Sixteen patients (20 lesions) had associated malformations. A total of 72.4% of Rex recesses could be measured by CT, and their mean diameter was 3.5 ± 1.8 mm (range 0.6–10.5 mm). A type 1, 2, 3, and 4 Rex recess was present in 9.2%, 53.9%, 11.8%, and 25.0% of patients, respectively. MRB could be performed in patients with types 1, 2, and 3, but those with type 4 required further evaluation. The sensitivity, specificity, positive predictive value, negative predictive value, and diagnostic accuracy of CT were 100%, 83.8%, 42.1%, 100%, and 85.5%, respectively. Conclusions Among the four types of Rex recesses on CT angiography, types 1–3 allow for the performance of MRB.

Background and AimsCongenital hyperinsulinism of infancy (CHI) is a rare condition that may cause irreversible severe neurological damage in infants. For children in whom medical management fails, partial or near-total pancreatectomy is then required according to the type of lesion. Currently, open surgery of near-total pancreatic head resection is a mature technique for the treatment of focal-form CHI located in the head of the pancreas, but a minimally invasive laparoscopic procedure has not been reported yet. The aim of this study was to verify the feasibility, safety, and efficacy of laparoscopic pancreatic head resection and Roux-en-Y pancreaticojejunostomy for focal-form CHI.MethodsTwo infants with persistent hypoglycemia and increased insulin levels were diagnosed with CHI and underwent laparoscopic near-total pancreatic head resection due to a suboptimal response to medical therapy and the likelihood of focal disease amenable to surgery. Clinical records, operative findings, and postoperative follow-up were collected and analyzed.ResultsThe operative duration was 300–330 min, and the intraoperative blood loss was minimal. The duration of postoperative abdominal drainage was 4–5 days. Neither intra- nor postoperative abdominal complications occurred. Oral feeding was resumed 3–4 days after the operation, and the blood glucose level was gradually stabilized to within the normal range. Normal blood glucose was observed in both patients over a follow-up period of 3–6 months.ConclusionsLaparoscopic pancreatic head resection and Roux-en-Y pancreaticojejunostomy can be considered a safe and effective procedure with minimal morbidity and excellent outcomes for the treatment of focal CHI in the head of the pancreas.

BackgroundIdiopathic pulmonary fibrosis (IPF) is one of interstitial lung diseases (ILDs) with poor prognosis. S100 calcium binding protein A12 (S100A12) has been reported as a prognostic serum biomarker in the IPF, but its correlation with IPF remains unclear in the lung tissue and bronchoalveolar lavage fluids (BALF).MethodsDatasets were collected from the Gene Expression Omnibus (GEO) database. Person correlation coefficient, Kaplan–Meier analysis, Cox regression analysis, functional enrichment analysis and so on were used. And single cell RNA-sequencing (scRNA-seq) analysis was also used to explore the role of S100A12 and related genes in the IPF.ResultsS100A12 was mainly and highly expressed in the monocytes, and its expression was downregulated in the lung of patients with IPF according to scRNA-seq and the transcriptome analysis. However, S100A12 expression was upregulated both in blood and BALF of patients with IPF. In addition, 10 genes were found to interact with S100A12 according to protein–protein interaction (PPI) network, and the first four transcription factors (TF) targeted these genes were found according to hTFtarget database. Two most significant co-expression genes of S100A12 were S100A8 and S100A9. The 3 genes were significantly negatively associated with lung function and positively associated with the St. George’s Respiratory Questionnaire (SGRQ) scores in the lung of patients with IPF. And, high expression of the 3 genes was associated with higher mortality in the BALF, and shorter transplant-free survival (TFS) and progression-free survival (PFS) time in the blood. Prognostic predictive value of S100A12 was more superior to S100A8 and S100A9 in patients with IPF, and the composited variable [S100A12 + GAP index (gender, age, and physiological index)] may be a more effective predictive index.ConclusionThese results imply that S100A12 might be an efficient disease severity and prognostic biomarker in patients with IPF.

Extraintestinal pathogenic Escherichia coli (ExPEC) is a pathogen that causes host extraintestinal diseases. The ST95 E. coli lineage is one of the dominant ExPEC lineages in humans and poultry. In this study, we took advantage of extensive E. coli genomes available through public open-access databases to construct a detailed understanding of the phylogeny and evolution of ST95. We used a high variability of accessory genomes to highlight the diversity and dynamic traits of ST95. Isolates from diverse hosts and geographic sources were randomly located on the phylogenetic tree, which suggested that there is no host specificity for ST95. The time-scaled phylogeny showed that ST95 is an ancient and long-lasting lineage. The virulence genes, resistance genes, and pathogenicity islands (PAIs) were characterized in ST95 pan-genomes to provide novel insights into the pathogenicity and multidrug resistance (MDR) genotypes. We found that a pool of large plasmids drives virulence and MDR. Based on the unique genes in the ST95 pan-genome, we designed a novel multiplex PCR reaction to rapidly detect ST95. Overall, our study addressed a gap in the current understanding of ST95 ExPEC genomes, with significant implications for recognizing the success and spread of ST95.

Objective:To explore the clinical presentation and molecular genetic characteristics of a cohort of congenital hyperinsulinism (CHI) patients from southern China and also to explore the most appropriate therapeutic approaches.Methods:We retrospectively reviewed a cohort of 65 children with CHI. Mutational analysis was performed for KCNJ11 and ABCC8 genes. The GLUD1 gene was sequenced in patients with hyperammonaemia. GCK gene sequencing was performed in those patients with no mutation identified in the ABCC8, KCNJ11 or GLUD1 genes.Results:ABCC8 mutations were identified in 16 (25%) of the cohort, GLUD1 mutations were identified in five children, and no KCNJ11 or GCK mutations were identified. Moreover, some unique features of ABCC8 gene mutations in southern Chinese CHI patients were found in this study. The most common mutation was a deletion/insertion mutation p.Thr1042GlnfsX75 was found in five unrelated patients, which possibly represents a relatively common mutation in southern China. Five novel ABCC8 mutations were detected. The mutations were p.Phe5SerfsX72, p.Gln273ArgfsX85, p.Leu724del, p.Asp1447Gly and IVS 25-1G>T. Five compound heterozygous mutations of ABCC8 gene were identified in this study, and three of these patients were diazoxide-responsive. Forty patients were diazoxide-responsive, 13 patients were diazoxide-unresponsive and 12 patients received dietary treatment only. A pancreatectomy was performed in 10 patients who were unresponsive to medical treatment.Conclusion:To the best of our knowledge, this is the first study of CHI in south China. Mutations in ABCC8 are the most common causes of CHI in this cohort. Diazoxide and dietary treatment were effective in most patients. Multicentre studies are necessary to obtain the long-term follow-up characteristics of such patients at a national level.

Abstract Background Low-type anorectal malformations (ARMs) are considered benign; however, in China, data regarding such conditions are limited. Thus, this study aimed to assess the middle-term bowel functions and quality of life (QOL) among children with low-type ARM. Methods Children > 3 years of age who underwent treatment for low-type ARM (vestibular fistula [VF] and perineal fistula [PF]) during 2013 and healthy children were included. The children were interviewed during their outpatient visits. The primary outcome measures were bowel function, as assessed using the Baylor Continence Scale (BCS), and QOL, as measured using the Pediatric Quality of Life Inventory (PedsQL 4.0). Results A total of 82 patients responded; mean patient age was 6.8 (range, 3–12) years. The increasing household loads make series arc faults more complex, which are difficult to be detected by traditional circuit breakers and lead to the frequent occurrence of residential fire accidents. In this paper, a comprehensive approach of complex load recognition and series arc detection is proposed on the basis of principal component analysis and support vector machine (PCA-SVM) combination model. Several typical loads were selected and analyzed, especially nonlinear and complex loads like power electronics load and multi-state load. Three time-domain parameters, maximum slip difference (MSD), zero current period (ZCP) and maximum Euclidean distance (MED), and nine frequency-domain harmonics information are collected to complex waveforms. To decrease the computation cost and further to enhance the response velocity, all the time-domain and frequency-domain information were blended and dimensionally reduced to three parameters by principal component analysis (PCA). Prior to the series arc detection, load recognition is trained and completed with the artificial intelligence (AI) algorithm. At last, the comprehensive model of load recognition and series arc detection is achieved based on a support vector machine (SVM). The accuracy of load recognition and series arc detection reaches 99.1% and 99.3%, respectively, demonstrating the excellent performances of the intelligent approach to diagnose the series arcing activities in modern household applications.

For solving the problem of polar performance of the inertial navigation system (INS) at mid-low latitudes, the simulation test system constructed by the “attitude and velocity invariant method of trajectory transfer rule based on the transverse coordinate system (T-AVIM)” of the Earth sphere model is used. The test system structure, especially the IMU conversion formula from mid-low latitudes to polar region simulation test, is introduced, and it is proved that the IMU conversion error can be equivalently superimposed on the bias error of the polar simulated IMU. According to the marine estimation formula for the effect of the reference error on the IMU conversion error, the specific influence of the constant error component and the random error component of the reference system on the simulated IMU is analyzed. The calculation method of the simulated IMU error is given with examples and intuitively explained, and the correctness of the theory is verified through simulation experiments.

Series arcing faults have been recognised as a hazardous phenomenon in the power system of more electric aircraft (MEA), especially with the increasing voltage level and power supply. Instead of the electrical method, the optical method can be used as an approach to detect arc fault with the advantages of fast response and immunity to electromagnetic interference. It is the basis of optical arc detection to determine the distribution spectrum range of direct current (DC) arc fault. Aiming at series arc optical detection in the MEA DC system, a DC series arc test platform was established to measure the series arc spectrum. The spectrum was disassembled though empirical mode decomposition to extract characteristic wavelengths of arc light and de-noised by means of wavelet decomposition and reconstruction (wavelet transformation) to reduce equipment noise. To resolve the baseline drift, the baseline of the arc spectrum measured was calculated and corrected based on Savitzky−Golay. The experimental results indicate that there are several series arc characteristic spectra of the MEA DC system, including 309.3 and 324.5 nm in the ultraviolet range. Moreover, the arc spectrum with different metals, copper and aluminium, were contrasted, indicating the arc spectral behaviour is related to the melting/boiling point and chemical properties of the anode material. The measurement of the series arc spectrum makes it more effective to apply optical detection in the MEA discharging faults diagnosis.

A comprehensive study was conducted at a wave energy device test site located off the northeastern coast of Taiwan to assess the influence of oceanic currents on experimental equipment. A bottom-mounted 600 kHz acoustic Doppler current profiler, equipped with integrated temperature and pressure sensors, was deployed at a depth of approximately 31 m. This study, spanning from 6 June 2023 to 11 May 2024, recorded ocean current profiles by assembling data from 50 pings every 10 min, with a resolution of one meter per depth layer. The findings reveal that variations in water levels were predominantly influenced by the M2 tidal constituent, followed by the O1, K1, and S2 tides. Notably, seawater temperature fluctuations at the seabed were modulated by tides, especially the M2 tide. A significant drop in seawater temperature was also observed as the typhoon passed through the south of Taiwan. In terms of sea surface currents, the measured maximum current speed was 71.89 cm s−1, but the average current speed was only 15.47 cm s−1. Tidal currents indicated that the M4 and M2 tides were the most significant, with semimajor axes and inclination angles of 8.48 cm s−1 and 102.60°, and 7.00 cm s−1 and 97.76°, respectively. Seasonally, barotropic tidal currents were the strongest in winter. Additionally, internal tides were identified, with the first baroclinic mode being dominant. The zero-crossing depths varied between 14 and 18 m. During the summer, the M2 baroclinic tidal current displayed characteristics of the second baroclinic mode, with zero-crossing depths at approximately 7 m and 22 m. This node aligns with results from the empirical orthogonal function analysis and correlates with the depths' significant shifts in seawater temperature as measured by a conductivity, temperature, and depth instrument. Despite the velocities of internal tides not being strong, the directional variance between surface and bottom flows presents critical considerations for the deployment and operation of moored wave energy devices. Potential impacts of projected long-term climate change toward the end of the 21st century on rainfall and peak intensity of six super-typhoons in the western North Pacific (WNP) are assessed using a cloud-resolving model (CRM) and the pseudo-global warming (PGW) method, under two representative concentration pathway (RCP) emission scenarios of RCP4.5 and RCP8.5. Linear long-term trends in June–October are calculated from 38 Coupled Model Intercomparison Project phase 5 (CMIP5) models from 1981–2000 to 2081–2100, with warmings of about 3 °C in sea surface temperature, 4 °C in air temperature in the lower troposphere, and increases of 20% in moisture in RCP8.5. The changes in RCP4.5 are about half the amounts. For each typhoon, three experiments are carried out: a control run (CTL) using analysis data as initial and boundary conditions (IC/BCs), and two future runs with the trend added to the IC/BCs, one for RCP4.5 and the other for RCP8.5, respectively. Their results are compared for potential impacts of climate change. In future scenarios, all six typhoons produce more rain rather consistently, by around 10% in RCP4.5 and 20% in RCP8.5 inside 200–250 km from the center, with increased variability toward larger radii. Such increases are tested to be highly significant and can be largely explained by the increased moisture and water vapor convergence in future scenarios. However, using this method, the results on peak intensity are mixed and inconsistent, with the majority of cases becoming somewhat weaker in future runs. It is believed that in the procedure to determine the best initial time for CTL, which yielded the strongest TC, often within a few hPa in minimum central sea-level pressure to the best track data, an advantage was introduced to the CTL unintentionally. Once the long-term trends were added in future runs, the environment of the storm was altered and became not as favorable for subsequent intensification. From 2001 to 2020, three typhoons with similar moving paths and intensities were found to trigger markedly different cooling off northeastern Taiwan. They were typhoons Utor (2001), Nuri (2008), and Hagupit (2008), which led to maximum sea surface temperature (SST) cooling temperatures of 8.8, 2.7, and 1.4 °C, respectively. The drastic cooling discrepancy implies that the existing understanding of the key mechanism leading to the cooling off northeastern Taiwan could be insufficient. For further exploring the key reason(s) contributing to the marked discrepancy, a regional oceanic modeling system (ROMS) was used to reconstruct the background oceanic environment corresponding to three typhoon passages. Results show that the wide radius of maximum winds of typhoon Utor contributes to the strongest SST cooling by enhancing the Kuroshio intrusion (KI) onto the shelf northeast of Taiwan. Heat budget diagnostics explain why including tidal forcing can substantially promote SST cooling. The process was associated mainly with a stronger vertical advection tied to the influence of de-stratification by tidal mixing. Finally, warmer Taiwan Strait currents, driven by wind forcing the typhoons to pass zonally through the north South China Sea, intruded clockwise into the Longdong coast and accelerated the recovery of sea surface cooling around Longdong. Sinapic acid (SA) is renowned for its many pharmacological activities as a polyphenolic compound. The cause of polycystic ovary syndrome (PCOS), a commonly encountered array of metabolic and hormonal abnormalities in females, has yet to be determined. The present experiment was performed to evaluate the antifibrotic properties of SA in rats with letrozole‐induced PCOS‐related ovarian fibrosis. SA treatment successfully mitigated the changes induced by letrozole in body weight (BW) (p <.01) and relative ovary weight (p <.05). Histological observation revealed that SA reduced the number of atretic and cystic follicles (AFs) and (CFs) (p <.01), as well as ovarian fibrosis, in PCOS rats. Additionally, SA treatment impacted the serum levels of sex hormones in PCOS rats. Luteinizing hormone (LH) and testosterone (T) levels were decreased (p <.01, p <.05), and follicle‐stimulating hormone (FSH) levels were increased (p <.05). SA administration also decreased triglyceride (TG) (p <.01) and total cholesterol (TC) levels (p <.05) and increased high‐density lipoprotein cholesterol (HDL‐C) levels (p <.01), thereby alleviating letrozole‐induced metabolic dysfunction in PCOS rats. Furthermore, SA treatment targeted insulin resistance (IR) and increased the messenger RNA (mRNA) levels of antioxidant enzymes in the ovaries of PCOS rats. Finally, SA treatment enhanced the activity of peroxisome proliferator‐activated receptor‐γ (PPAR‐γ), reduced the activation of transforming growth factor‐β1 (TGF‐β1)/Smads, and decreased collagen I, α‐smooth muscle actin (α‐SMA), and connective tissue growth factor (CTGF) levels in the ovaries of PCOS rats. These observations suggest that SA significantly ameliorates metabolic dysfunction and oxidative stress and ultimately reduces ovarian fibrosis in rats with letrozole‐induced PCOS. Sea surface temperature (SST) responses have been perceived as crucial to consequential tropical cyclone (TC) intensity development. In addition to regular cooling responses, a few TCs could cause extreme SST drops (ESSTDs) (e.g., SST drops more than 6 °C) during their passage. Given the extreme temperature differences and the consequentially marked air–sea flux modulations, ESSTDs are intuitively supposed to play a serious role in modifying TC intensities. Nevertheless, the relationship between ESSTDs and consequential storm intensity changes remains unclear. In this study, satellite-observed microwave SST drops and the International Best Track Archive for Climate Stewardship TC data from 2001 to 2021 were used to elucidate the relationship between ESSTDs and the consequential TC intensity changes in the Western North Pacific typhoon season (July–October). Subsequently, the distributed characteristics of ESSTDs were systematically examined based on statistical analyses. Among them, Typhoon Kilo (2015) triggered an unexpected ESSTD behind its passage, according to existing theories. Numerical experiments based on the Regional Ocean Modeling System were carried out to explore the possible mechanisms that resulted in the ESSTD due to Kilo. The results indicate that heavy rainfall leads to additional SST cooling through the enhanced sensible heat flux leaving the surface layer in addition to the cooling from momentum-driven vertical mixing. This process enhanced the sensible heat flux leaving the sea surface since the temperature of the raindrops could be much colder than the SST in the tropical ocean, specifically under heavy rainfall and relatively less momentum entering the upper ocean during Kilo. Ocean acoustic tomography (OAT) deploys most moored stations on the periphery of the tomographic region to sense the solenoidal current field. Moving vehicle tomography (MVT), an advancement of OAT, not only samples the region from various angles for improving the resolution of mapped currents but also acquires information about the irrotational flow due to the sampling points inside the region. To reconstruct a complete two-dimensional current field, the spatial modes derived from the open-boundary modal analysis (OMA) are preferable to the conventional truncated Fourier series since the OMA technique describes the solenoidal and irrotational flows efficiently in which all modes satisfy the coastline and open boundary conditions. Comparisons of the reconstructions are presented using three different representations of currents. The first two representations explain only the solenoidal flow: the truncated Fourier series and the OMA Dirichlet modes. The third representation, accounting for the solenoidal and irrotational flows, uses all the OMA modes. For reconstructing the solenoidal flow, the OMA representation with the Dirichlet modes performs better than the Fourier series. A large difference appears near the bay mouth, where the OMA-Dirichlet reconstruction shows a better fit to the uniform currents. However, considerable uncertainty exists outside the bay mouth where the irrotational currents dominate. This can be improved by the third representation with the inclusion of the Neumann and boundary modes. The reconstruction results using field data were validated against the acoustic Doppler current profiler (ADCP) measurements. Additionally, incorporating constraints from ADCP measurements enhances the accuracy of the reconstruction. Significance Statement: This study contributes toward improving our understanding of accurately measuring oceanic circulation patterns over large areas without relying solely upon stationary sensors or satellite imagery. The study combines multiple sources, such as shipboard ADCP and tomographic techniques, to obtain a complete picture of what is happening beneath surface waters across entire regions under investigation.
