Abstract Walnut is an important economic tree species while confronting with global environmental stress, resulting in decline in quality and yield. Therefore, it is urgent to elucidate the molecular mechanism for the regulation of walnut response to adversity. The protein phosphatase 2C (PP2C) gene family participates in cellular processes in eukaryotes through reversible phosphorylation of proteins and signal transduction regulation. However, the stress response function of PP2C genes was far to be clarified. Therefore, to understand the stress response mechanism of walnut tree, in this study, a total of 41 PP2C genes with complete ORFs were identified from Juglans regia, whose basic bio-information and expression patterns in response to multiple stresses and ABA were confirmed. The results showed that the ORFs of JrPP2Cs were 495 ~ 3231 bp in length, the predicted JrPP2C proteins contained 164 to 1076 amino acids and the molecular weights were 18,581.96 ~ 118,853.34 Da, the pI was 4.55 ~ 9.58. These JrPP2C genes were unevenly distributed on 14 chromosomes, among which Chr11 and Chr13 contained the most genes. Phylogenetic analysis found that these JrPP2C proteins were classed into 9 subfamilies, among which group F covered most JrPP2Cs. The JrPP2Cs in the same subfamily exhibited similarities in the composition of conserved domains, amino acid sequences of motifs and exon/intron organization in DNA sequences. Each JrPP2C includes 4 ~ 10 motifs and each motif contained 15 ~ 37 amino acids. Among the motifs, motif1, motif2, motif3 and motif8 were most abundant. Most of the JrPP2C genes diversely response to osmotic, cadmium, and Colletotrichum gloeosporioide stress as well as ABA treatments, among which JrPP2C28, JrPP2C17, JrPP2C09, JrPP2C36 were more obvious and deserves further attention. All these results indicated that JrPP2C genes play potential vital roles in plant response to multiple stimulus, and are possibly involved in ABA-dependent signaling pathway.

Biological materials, such as bamboo, are naturally optimized composites with exceptional mechanical properties. Inspired by such natural composites, traditional methods involve extracting nanofibers from natural sources and applying them in composite materials, which, however, often results in less ideal mechanical properties. To address this, this study develops a bottom-up nanofiber assembly strategy to create strong fiber-reinforced composite hydrogels inspired by the hierarchical assembly of bamboo. Self-assembled chitosan-sodium alginate nanofibers (CSNFs) are combined with tannic acid (TA) and poly(vinyl alcohol) (PVA) as the interfacial crosslinker and hydrogel matrix, respectively, to emulate the fundamental cellulose-lignin-hemicellulose composition unit of bamboo. Strong interfacial electrostatic interactions and hydrogen bonding form between the functional groups of these components. These molecular interactions can be further reinforced by constructing higher-order structure through stretch-induced orientation. The resulting composite hydrogel achieves good mechanical performance, including a high tensile strength of up to 60.2 MPa and a simultaneous high strength of 48.0 MPa and ultimate strain of 470%. This approach demonstrates a hierarchical bottom-up strategy to construct strong and robust composite hydrogels by effectively leveraging fundamental molecular interactions. By mimicking bamboo's highly integrated structural composition, it offers a promising solution for creating advanced bioinspired materials with excellent mechanical properties. There is interest in mimicking the properties of biological composites in artificial materials, but the hierarchical structures can be challenging to replicate. Granite is a common engineering material that exhibits complex mechanical properties under external loads. This study conducted experimental research and analysis in conjunction with the active monitoring technology of piezoelectric ceramics. A quantitative analysis method for the mechanical properties of rock materials based on piezoelectric health monitoring was established, and for the first time, the piezoelectric monitoring results of rock were mapped and compared with the uniaxial compression performance indicators of rock. In this study, two sets of cyclic impact experiments were conducted on granite samples using a drop hammer. The piezoelectric signals of the granite samples were detected using piezoelectric ceramic active sensing technology. A piezoelectric ceramic damage monitoring method was proposed, and the damage factor of the granite samples was calculated using the wavelet packet energy method. Subsequently, uniaxial compression experiments were performed on the damaged granite samples to obtain mechanical performance data. Finally, a mathematical relationship model was established between the piezoelectric signal and the uniaxial compressive strength of the rocks. It was found that the damage factor of the piezoelectric monitoring signal of the damaged rock were linearly related to the uniaxial compressive strength of the damaged rock. [ABSTRACT FROM AUTHOR]

In order to resolve the issues of low efficiency and poor precision in the traditional finishing process of hardened internal gears, a method is proposed for calculating the profile curves of a drum-shaped grinding tool suitable for mass finishing of hardened internal gears. Additionally, the impact of drum-shaped grinding tool installation errors on the tooth surface deviation of internal gears is analyzed. Firstly, the processing principle for the generation grinding of internal gears by the drum-shaped grinding tool is introduced. Based on differential geometry, meshing theory, and two-degree-of-freedom enveloping method, a mathematical model is developed for the generation grinding of internal gears. Profile curves of the drum-shaped grinding tool are obtained by solving the meshing equation between the drum-shaped grinding tool and the internal gear. Then, the tooth surface equation for the internal gear is derived in the presence of drum-shaped grinding tool installation errors. By discretizing the error tooth surface of the internal gear, the average normal deviation of the tooth surface is calculated. In the end, the distribution of normal deviation on the tooth surface of the internal gear with different drum-shaped grinding tool installation errors is acquired, and the influence of four kinds of installation errors on the tooth surface of the internal gear is analyzed. The sensitive direction is identified for drum-shaped grinding tool installation errors on the tooth surface of the internal gear. Consequently, this research provides a calculation method for the drum-shaped grinding tool fit for high-precision and high-efficiency finishing of mass-produced hardened internal gear and offers a reference for correcting deviation in the tooth surface of internal gear with installation errors of the drum-shaped grinding tool. Allergic asthma (AA) is closely associated with the polarization of T helper (Th)2 and Th17 cells. Interleukin (IL)-18 acts as an inducer of Th2 and Th17 cell responses. However, expressions of IL-18 and IL-18 receptor alpha (IL-18Rα) in blood Th2 and Th17 cells of patients with AA remain unclear. We therefore investigated their expressions in Th2 and Th17 cells using flow cytometric analysis, quantitative real-time PCR (qPCR), and murine AA model. We observed increased proportions of Th2, Th17, IL-18+, IL-18+ Th2, and IL-18+ Th17 cells in blood CD4+ T cells of patients with AA. Additionally, house dust mite seemed to upregulate further IL-18 expression in Th2 and Th17, and upregulate IL-18Rα expression in CD4+ T, Th2, and Th17 cells of AA patients. It was also found that the plasma levels of IL-4, IL-17A, and IL-18 in AA patients were elevated, and they were correlated between each other. In ovalbumin (OVA)-induced asthma mouse (AM), we observed that the percentages of blood CD4+ T, Th2, and Th17 cells were increased. Moreover, OVA-induced AM expressed higher level of IL-18Rα in blood Th2 cells, which was downregulated by IL-18. Increased IL-18Rα expression was also observed in blood Th2 cells of OVA-induced FcεRIα−/− mice. Collectively, our findings suggest the involvement of Th2 cells in AA by expressing excessive IL-18 and IL-18Rα in response to allergen, and that IL-18 and IL-18Rα expressing Th2 cells are likely to be the potential targets for AA therapy. Stochastic cooling of the high-precision spectrometer ring (SRing) at the High Intensity Heavy-Ion Accelerator Facility (HIAF) project in China, which is used mainly for experiments with radioactive fragment beams, is applied to speed up the cooling process of a stored ion beam. In this article, a new coaxial-type notch filter with an amplitude equalizer in the long branch and an optical-type notch filter with phase-stabilized optical fiber are discussed and evaluated for the SRing stochastic cooling system. Both prototypes of coaxial and optical notch filters are fabricated and tested. The minimum notch depth of coaxial and optical notch filters reaches to 26 and 40 dB, respectively. The performance of both coaxial notch filter and optical fiber notch filter is presented in this work. These developments will be used not only for the longitudinal stochastic cooling system but also have potential for the beam feedback system. [ABSTRACT FROM AUTHOR]

Domain-based electronic and electrical (E/E) architectures have been regarded as a possible upgrade to distributed E/E architectures currently used in electric vehicles. In a distributed E/E design, E/E components are directly connected to the automobile bus. Domain-based architectures split E/E components into distinct domains depending on their functions, which clearly benefits software upgrading and wire harness reduction. However, due to its heterogeneous topology with multiple network protocols, domain-based E/E architecture introduces complicated multilink and multinode delays into the control loop. The delays may degrade and even deteriorate the stability of adaptive cruise control (ACC) employing domain-based E/E architecture. To this end, this article proposes a heterogeneous-topology loop delay analysis by introducing a notion of poly-service loop delay. With a graphical pattern, the analytical process is presented in depth. The worst-case loop delay is calculated using an upper-boundary mathematic equation. Then, a hierarchical cyber-physical control method for ACC is designed. The upper level is intended to achieve desired acceleration based on vehicle and intervehicle motion states. And the lower level is intended to mitigate the negative impact of loop delays and provide reliable acceleration tracking. Urban agglomerations are becoming an increasingly important factor in advancing regional development and reshaping a new pattern of regional competition. However, few studies are focused on the impact of expanding urban agglomerations on reducing carbon emissions and its possible mechanism. Based on 285 city-level panel data from 2006 to 2017, this paper uses a staggered Difference-in-Differences (DID) model to explore the reduction effect and its possible mechanism of sustainable regional development policy, characterized by urban agglomeration expansion policy in the Yangtze River Delta, on carbon emissions with policy shocks in 2010 and 2013. The results are as follows: (1) The urban agglomeration expansion policy shows a significant marginal contribution to the reduction of carbon emissions, especially for the later joined (new) cities, and the reduction effect is particularly significant in the first and third years after the expansion, indicating that there are significant short-term and long-term reduction effects of the expansion policy. (2) The heterogeneities of reduction effect among three provinces are significant. Zhejiang Province enjoys the largest proportion carbon emission reduction effect, followed by Anhui and Jiangsu provinces. To be specific, urban agglomeration expansion in Zhejiang Province reduced carbon emissions and carbon emissions intensity in the overall, incumbent cities and new cities, while it only increased the total carbon emissions of the incumbent cities in Jiangsu province. (3) The heterogeneities of reduction effect brought by 2010 and 2013 are also significant. The urban agglomeration expansion policy in 2010 reduced carbon emissions on the whole cities and the incumbent cities with later joined cities excluded, while it had a significant reduction effect on the total, incumbent cities, and the new cities in 2013. (4) There are two possible mechanisms of this reduction effect. One is the strengthening of economic ties and enhanced environmental synergy between governments, called the market integration mechanism, which only has a significant effect on carbon emission reduction in the incumbent cities. Another is through the upgrade of the structure of regional industries, which has a significant effect in both the incumbent and new cities. These findings suggest that when formulating urban agglomerations polices, governments must take into account the carbon emissions effect, and advance the upgrading of industrial structure in the urban agglomeration. A diagonal-cut type beam position monitor (BPM) has been developed for the High Intensity Heavy-Ion Accelerator Facility (HIAF) project at the Institute of Modern Physics. Compared with other types of BPMs, the diagonal-cut type BPM has almost perfect position linearity, i.e., no non-linear correction required, which is advantageous for beams that are transversally large and have a complex charge distribution. The key parameters for the diagonal-cut type BPM have been simulated and optimized in detail and systematically herein. It was found that the crosstalk is improved by ∼10 dB at 160 MHz by insertion of a separate ring between two horizontal or vertical electrodes of the BPM made of stainless steel with vacuum as a dielectric. Furthermore, the longitudinal and transverse numerical simulation to evaluate the beam impedance on the diagonal-cut type BPM has been performed. The results for the crosstalk, position sensitivity, and electrode capacitance to ground obtained from simulations and laboratory measurements agree well. The vacuum of the BPM prototype after baking out at 250 °C for 72 h is better than 1.0 × 10−11 mbar. The simulated and on-line measured BPM output signal magnitude results are consistent with each other. Advanced ceramic products are widely used in aerospace, automotive, electronic, laboratory equipment, and other industries. To achieve the geometric complexity and desirable properties that are difficult to obtain by conventional manufacturing methods, ceramic additive manufacturing (AM) methods have been studied intensively in recent years. However, the adaptive control with feedback is not currently implemented in any commercially available ceramic three‐dimensional printer. Robocasting is one of the most widely utilized constant‐volumetric‐flow AM processes for creating various ceramic materials at a low cost. This study employed robocasting as a model to implement an adaptive control with a feedback loop in the ceramic AM process. In this research, processing load that was proportional to the processing pressure, width of the print, and length of extrusion were selected to be representative of process signal, quality signal, and control parameter, respectively. First, a database of the load and extrusion length was established. An artificial neural network model was created using that established database. The data‐driven, closed‐loop control was integrated into the robocasting process. Finally, the improvement was validated by comparing the quality of the prints produced by both the closed‐loop process with the adaptive control and the open‐loop process without the adaptive control. Aim: To study the effect of pharmacological inhibition of epidermal growth factor receptor (EGFR) on intracranial aneurysm (IA) initiation. Methods: Human IA samples were analyzed for the expression of p‐EGFR and alpha smooth muscle actin (α‐SMA) by immunofluorescence (IF). Rat models of IA were established to evaluate the ability of the EGFR inhibitor, erlotinib, to attenuate the incidence of IA. We analyzed anterior cerebral artery tissues by pathological and proteomic detection for the expression of p‐EGFR and relevant proteins, and vessel casting was used to evaluate the incidence of aneurysms in each group. Rat vascular smooth muscle cells (VSMCs) and endothelial cells were extracted and used to establish an in vitro co‐culture model in a flow chamber with or without erlotinib treatment. We determined p‐EGFR and relevant protein expression in VSMCs by immunoblotting analysis. Results: Epidermal growth factor receptor activation was found in human IA vessel walls and rat anterior cerebral artery walls. Treatment with erlotinib markedly attenuated the incidence of IA by inhibiting vascular remodeling and pro‐inflammatory transformation of VSMC in rat IA vessel walls. Activation of EGFR in rat VSMCs and phenotypic modulation of rat VSMCs were correlated with the strength of shear stress in vitro, and treatment with erlotinib reduced phenotypic modulation of rat VSMCs. In vitro experiments also revealed that EGFR activation could be induced by TNF‐α in human brain VSMCs. Conclusions: These results suggest that EGFR plays a critical role in the initiation of IA and that the EGFR inhibitor erlotinib protects rats from IA initiation by regulating phenotypic modulation of VSMCs. Herein, we present a methodology and framework for exploiting certain interdisciplinary studies that can particularly benefit from integration. In this paper, rigorous derivation of control theory and statistical analysis of simulation results are organically unified for testifying and optimizing the emergence of order in aerial swarming scenarios under free boundary conditions. Each Unmanned Aerial Vehicle (UAV) is regulated by a simplified mathematical model, based on which a distributed flocking protocol is proposed as a feasible solution for aerial swarms. On condition that the initial interaction network is connected, the LaSalle–Krasovskii invariance principle is implemented to verify the effectiveness of the above algorithm. However, most existing results on flocking are far from being engineering applications. A basic challenge is how to present a low-cost energy and time saving solution on account of the limited flight capability of these UAVs and real-time operational requirements. As is well known, energy consumption can be reduced if unnecessary interactions among individuals are eliminated. Therefore, another contribution of this paper is to propose a precise optimization of an existing flocking algorithm for UAVs with respect to interaction requirements. Energy and time measurements, as well as scalability effects, are assessed in terms of statistical significance and strength. A highly sensitive, specific and simple LC–MS/MS method for quantification of capmatinib (INC280) in rat plasma was presented. The LC–MS/MS method was validated in terms of specificity and selectivity, linearity, accuracy and precision, matrix effect, extraction recovery, dilution integrity, carryover and stability as per the US Food and Drug Administration's bioanalytical method validation guideline. The validated assay was applied for quantification of capmatinib from a pharmacokinetic study in rats following oral administration at the doses of 1.0, 3.0 and 9.0 mg/kg. The calibration curve ranges from 1 to 2000 ng/ml with desirable linearity and r2 > 0.99. The intra‐ and inter‐batch accuracies were within 99.24–103.59 and 97.76–102.83% with coefficients of variation 5.08–7.36 and 3.18–4.99%, respectively. No significant interference was observed by endogenous peak at the retention time of capmatinib and IS. The assay was free from any matrix effect and showed precise recovery across the calibration curve range, and samples were stable under all experimental conditions. The validated assay was successfully applied to analyze plasma samples of pharmacokinetic study in rat to determine the concentration of capmatinib. In summary, a novel method for analyzing capmatinib in rat plasma has been successfully validated and is now being utilized for quantification of capmatinib from pre‐clinical studies.
