1. Relaxin 2 carried by magnetically directed liposomes accelerates rat midpalatal suture expansion and subsequent new bone formation

Abstract

Relaxin (RLN) is an insulin-like peptide hormone that enables softening and lengthening of the pubic symphysis and uterine cervix. Here, we analyzed the effects of RLN2 on the expansion of rat midpalatal suture (MPS) using a magnetically directed liposome-based drug delivery system. Thirty-six male rats were divided into three groups: control (MPS was not expanded), lipo (expanded for 1 week with vehicle liposomes encapsulating ferric oxide and Cy5.5), and RLN-lipo (expanded for 1 week with the liposomes coated with RLN2). Rats were sacrificed after 1 week of expansion or after 2 weeks of retention. To accumulate RLN2-liposomes, a magnetic sheet was fixed to the palatal mucosa of the MPS. In vivo imaging showed magnetically controlled accumulation of liposomes in the MPS for 72 h. Immunohistochemistry revealed RLN2 expression in the MPS after expansion and relaxin receptor (RXFP) 2 expression at the osteogenic front (OF) in the RLN-lipo group; all groups expressed RXFP1 in the MPS. MPS expansion and bone formation were significantly accelerated at the OF in RLN-lipo group compared with the other groups. In the RLN-lipo group, significantly accelerated serrate bone deposition and elevated periostin (POSTN), iNOS, and MMP-1 levels were observed in the MPS. Sclerostin (SOST) expression was significantly reduced in newly formed bone in the RLN-lipo group. Our data revealed that RLN2 enhanced suture expansion via MMP-1 and iNOS secretion in the sutural fibroblasts and new bone formation via POSTN expression in osteoblasts at the OF. These properties may be useful for developing a new less-invasive orthopedic treatment aiming at sutural modification of cranio- and maxillofacial deformity patients. 2. Regulation of Calvarial Osteogenesis by Concomitant De-repression of GLI3 and Activation of IHH Targets

Abstract

Loss-of-function mutations in GLI3 and IHH cause craniosynostosis and reduced osteogenesis, respectively. In this study, we show that Ihh ligand, the receptor Ptch1 and Gli transcription factors are differentially expressed in embryonic mouse calvaria osteogenic condensations. We show that in both Ihh−/− and Gli3Xt−J/Xt−J embryonic mice, the normal gene expression architecture is lost and this results in disorganized calvarial bone development. RUNX2 is a master regulatory transcription factor controlling osteogenesis. In the absence of Gli3, RUNX2 isoform II and IHH are upregulated, and RUNX2 isoform I downregulated. This is consistent with the expanded and aberrant osteogenesis observed in Gli3Xt−J/Xt−J mice, and consistent with Runx2-I expression by relatively immature osteoprogenitors. Ihh−/− mice exhibited small calvarial bones and HH target genes, Ptch1 and Gli1, were absent. This indicates that IHH is the functional HH ligand, and that it is not compensated by another HH ligand. To decipher the roles and potential interaction of Gli3 and Ihh, we generated Ihh−/−;Gli3Xt−J/Xt−J compound mutant mice. Even in the absence of Ihh, Gli3 deletion was sufficient to induce aberrant precocious ossification across the developing suture, indicating that the craniosynostosis phenotype of Gli3Xt−J/Xt−J mice is not dependent on IHH ligand. Also, we found that Ihh was not required for Runx2 expression as the expression of RUNX2 target genes was unaffected by deletion of Ihh. To test whether RUNX2 has a role upstream of IHH, we performed RUNX2 siRNA knock down experiments in WT calvarial osteoblasts and explants and found that Ihh expression is suppressed. Our results show that IHH is the functional HH ligand in the embryonic mouse calvaria osteogenic condensations, where it regulates the progression of osteoblastic differentiation. As GLI3 represses the expression of Runx2-II and Ihh, and also elevates the Runx2-I expression, and as IHH may be regulated by RUNX2 these results raise the possibility of a regulatory feedback circuit to control calvarial osteogenesis and suture patency. Taken together, RUNX2-controlled osteoblastic cell fate is regulated by IHH through concomitant inhibition of GLI3-repressor formation and activation of downstream targets.

Abstract

The origin of Galactic Cosmic rays (GCRs), whose spectrum extends to PeV energies, is one of the longest-standing problems in astroparticle physics. One of the main sources of GCRs are regarded to be Supernova remnants (SNRs). While SNRs are known to accelerate protons, so far there is no evidence that SNRs can accelerate CRs to PeV energies. Providing that ~10% of the parent Cosmic ray energy is converted to gamma rays, the gamma-ray spectrum extending up to ~100 TeV would be a signature of a so-called Galactic PeVatron, an object responsible for the production of protons up to the knee of the Cosmic ray spectrum. The current multi-wavelength data indicate that HESS J1809-193 is one of the most promising Galactic PeVatron candidates. So far, no firm identification on the source nature has been established as there are several possible counterparts at lower energies; one of them being SNR G11.0−0.0. We report here the results of an observational campaign performed by the MAGIC telescopes on HESS J1809-193 since 2019 in the very-high-energy gamma-ray domain (E>100 GeV). The data were obtained with the Very Large Zenith Angle (VLZA) technique, which increased the collection area significantly to about one square kilometer. Abstract

X-ray and gamma-ray binaries are systems consisting of a compact object and normally a non-degenerate companion star. Most of these sources have been shown to emit radiation in a broad frequency range, from radio up to X-rays and sometimes gamma rays. We report on recent results in very high-energy gamma rays above 100 GeV obtained by the MAGIC Collaboration for the Galactic X-ray binaries MAXI J1820+070 and 1A 0535+262, and the gamma-ray binary HESS J0632+057. Multiwavelength data at lower energies are also provided for a better contextualisation of the sources.

Abstract

QSO B0218+357 is currently the only gravitationally lensed source from which very-high-energy (VHE, &100GeV) gamma-ray emission has been detected. We report the multiwavelength monitoring observations of this source performed between 2016 and 2020 in radio interferometry, optical, X-ray and gamma-ray bands. Abstract

Blazars, together with other active galactic nuclei, are the most luminous persistent sources in our universe; and therefore a prime candidate for very-high-energy (>0.2 TeV, VHE) gamma-ray observations. For the two MAGIC telescopes, the Mrk501 galaxy is among the brightest observed blazars due to its proximity. We report a multi-wavelength and multi-messenger study of Mrk501 with data from 2017 to 2020, when Mrk501 showed a VHE flux typically below 10% that of the Crab Nebula. During this time, we performed three long observations with NuSTAR, which characterized the hard X-ray emission during three different low-activity flux levels. This Mrk501 dataset provided the unprecedented opportunity to study multi-wavelength variability and correlations with sensitive instruments during historically low X-ray and VHE gamma-ray emission (below 5% of the Crab Nebula flux in the VHE range), which could be considered as the baseline emission of Mrk501. We complemented the broadband spectral energy distributions (SED) of the identified historically low X-ray and VHE gamma-ray flux with data published by IceCube, in order to evaluate the potential existence of a hadronic component that is stable (or slowly variable), and less visible than the leptonic component that may dominate the emission during typical and flaring activity. Abstract

The high-peaked BL Lac object 1ES 0647+250 is one of the few distant blazars detected at very-high-energy (VHE, E>100 GeV) γ rays during non-flaring activity. Its redshift is still uncertain, with a recently proposed lower limit of z>0.29. This blazar was first detected by the MAGIC telescopes between 2009 and 2011 during its low state, displaying a flux of around 2% of the Crab Nebula above 100 GeV, but it has shown several periods of high activity, where the VHE γ-ray flux increased by more than one order of magnitude. The VHE spectra of four observed periods are characterized and the redshift of this source is estimated using the HE and VHE spectral shapes. The multi-wavelength data set collected from 2009 to 2020 introduced in this work will serve as the seed for further studies, including detailed studies of the broad-band spectral energy distribution for different activity levels and multi-band variability and correlation studies., PoS: Proceedings of Science, 395, ISSN:1824-8039, 37th International Cosmic Ray Conference (ICRC 2021)

Abstract

We present the first search for dark matter (DM) spectral lines in the Galactic centre (GC) region with the MAGIC telescopes. The MAGIC telescopes, located on the Canary island of La Palma (Spain), are sensitive to gamma rays in the energy range from 50 GeV to 50 TeV. MAGIC has performed indirect DM searches in various astrophysical targets, such as dwarf spheroidal galaxies and clusters of galaxies. Observations at high zenith angles significantly increase the telescopes' collection area and sensitivity for gamma rays in the TeV regime. We present the results obtained with more than 200 hours of high-zenith angle observations of the GC region with MAGIC, which allow us to probe promising heavy SUSY models, and to obtain competitive limits to the DM annihilation cross-section at high DM particle mass, compared to existing constraints. 9. Therapeutic effect of nanogel-based delivery of soluble FGFR2 with S252W mutation on craniosynostosis.

Abstract

Apert syndrome is an autosomal dominantly inherited disorder caused by missense mutations in fibroblast growth factor receptor 2 (FGFR2). Surgical procedures are frequently required to reduce morphological and functional defects in patients with Apert syndrome; therefore, the development of noninvasive procedures to treat Apert syndrome is critical. Here we aimed to clarify the etiological mechanisms of craniosynostosis in mouse models of Apert syndrome and verify the effects of purified soluble FGFR2 harboring the S252W mutation (sFGFR2IIIcS252W) on calvarial sutures in Apert syndrome mice in vitro. We observed increased expression of Fgf10, Esrp1, and Fgfr2IIIb, which are indispensable for epidermal development, in coronal sutures in Apert syndrome mice. Purified sFGFR2IIIcS252W exhibited binding affinity for fibroblast growth factor (Fgf) 2 but also formed heterodimers with FGFR2IIIc, FGFR2IIIcS252W, and FGFR2IIIbS252W. Administration of sFGFR2IIIcS252W also inhibited Fgf2-dependent proliferation, phosphorylation of intracellular signaling molecules, and mineralization of FGFR2S252W-overexpressing MC3T3-E1 osteoblasts. sFGFR2IIIcS252W complexed with nanogels maintained the patency of coronal sutures, whereas synostosis was observed where the nanogel without sFGFR2S252W was applied. Thus, based on our current data, we suggest that increased Fgf10 and Fgfr2IIIb expression may induce the onset of craniosynostosis in patients with Apert syndrome and that the appropriate delivery of purified sFGFR2IIIcS252W could be effective for treating this disorder.

Abstract

In February 2019, a flaring state of the extreme blazar candidate TXS 1515-273 was registered by the Fermi-LAT, which triggered observations with the MAGIC telescopes and the X-ray satellites Swift, XMM-Newton and NuStar. The observations led to the discovery of the source at very-high-energy (VHE, 100 GeV < E < $ 100 TeV) gamma-ray energies and the detection of short time scales of variability (1 h) in several X-ray bands. The analysis of the observed variability helped us to constrain the physical parameters of the emission region. Thanks to the high-quality X-ray data, the synchrotron peak location was determined. The source was classified as a high synchrotron peaked source during the flaring activity. We constructed the broadband spectral energy distribution from radio to TeV. We interpreted it assuming leptonic emission and taking into account the constraints from the X-ray variability. We tested two scenarios: a simple one-zone model and a two-component model. Both models were found to describe the data well from X-ray to VHE gamma ray, but the two-zone model allows for a more accurate modelling of the emission at radio and optical energies.

Abstract

Silicon photomultipliers (SiPMs) have become the baseline choice for cameras of the small-sized telescopes (SSTs) of the Cherenkov Telescope Array (CTA). On the other hand, SiPMs are relatively new to the field and covering large surfaces and operating at high data rates still are challenges to outperform photomultipliers (PMTs). The higher sensitivity in the near infra-red and longer signals compared to PMTs result in higher night sky background rate for SiPMs. However, the robustness of the SiPMs represents a unique opportunity to ensure long-term operation with low maintenance and better duty cycle than PMTs. The proposed camera for large size telescopes will feature 0.05 degree pixels, low power and fast front-end electronics and a fully digital readout. In this work, we present the status of dedicated simulations and data analysis for the performance estimation. The design features and the different strategies identified, so far, to tackle the demanding requirements and the improved performance are described.

Abstract

The measurement of an astrophysical flux of high-energy neutrinos by IceCube is an important step towards finding the long-sought sources of cosmic rays. Nevertheless, the long exposure neutrino sky map shows no significant indication of point sources so far. The real-time followup of neutrino events turned out to be the most successful approach in neutrino point-source searches. It brought, among others, the most compelling evidence for a neutrino point source: the flaring gamma-ray blazar TXS 0506+056 in coincidence with a single high-energy neutrino from IceCube (IceCube-170922A). The fast multiwavelength(MWL) follow-up of this alert was key for establishing this coincidence and constraining the subsequent theoretical modeling for this event. In the long run, accurate and contemporaneous MWL spectral measurements are essential ingredients in investigating the physical processes leading to particle acceleration and emission of radiation. A deeper understanding of those processes allows us to put constraints on the potential neutrino emission. Here we present the light curves and simultaneous spectral energy distributions from November 2017 till February 2021 of MAGIC and MWL monitoring of TXS 0506+056. The more than the two year-long rise and high state of the radio light curve of TXS 0506+056, which started near the time of the IceCube neutrino detection, seems to have ended, as indicated by a steep decrease in the first half of 2021. Abstract

The Cherenkov Telescope Array (CTA) is the next-generation ground-based very-high-energy gamma-ray observatory. The Large-Sized Telescope (LST) of CTA is designed to detect gamma rays between 20 GeV and a few TeV with a 23-meter diameter mirror. We have developed the focal plane camera of the first LST, which has 1855 photomultiplier tubes (PMTs) and the readout system which samples a PMT waveform at GHz with switched capacitor arrays, Domino Ring Sampler ver4 (DRS4). To measure the precise pulse charge and arrival time of Cherenkov signals, we developed a method to calibrate the output voltage of DRS4 and the sampling time interval, as well as an analysis method to correct the spike noise of DRS4. Since the first LST was inaugurated in 2018, we have performed the commissioning tests and calibrated the camera. Abstract

Apert syndrome is an autosomal dominantly inherited disorder caused by missense mutations in fibroblast growth factor receptor 2 (FGFR2). Surgical procedures are frequently required to reduce morphological and functional defects in patients with Apert syndrome; therefore, the development of noninvasive procedures to treat Apert syndrome is critical. Here we aimed to clarify the etiological mechanisms of craniosynostosis in mouse models of Apert syndrome and verify the effects of purified soluble FGFR2 harboring the S252W mutation (sFGFR2IIIcS252W) on calvarial sutures in Apert syndrome mice in vitro. We observed increased expression of Fgf10, Esrp1, and Fgfr2IIIb, which are indispensable for epidermal development, in coronal sutures in Apert syndrome mice. Purified sFGFR2IIIcS252W exhibited binding affinity for fibroblast growth factor (Fgf) 2 but also formed heterodimers with FGFR2IIIc, FGFR2IIIcS252W, and FGFR2IIIbS252W. Administration of sFGFR2IIIcS252W also inhibited Fgf2-dependent proliferation, phosphorylation of intracellular signaling molecules, and mineralization of FGFR2S252W-overexpressing MC3T3-E1 osteoblasts. sFGFR2IIIcS252W complexed with nanogels maintained the patency of coronal sutures, whereas synostosis was observed where the nanogel without sFGFR2S252W was applied. Thus, based on our current data, we suggest that increased Fgf10 and Fgfr2IIIb expression may induce the onset of craniosynostosis in patients with Apert syndrome and that the appropriate delivery of purified sFGFR2IIIcS252W could be effective for treating this disorder.

Abstract

Background and objectives: Relaxin (RLN) is an insulin-like hormone associated with extracellular matrix degradation, osteoclastogenesis, and osteoblast differentiation. This study aimed to assess the effect of RLN during and after lateral expansion of murine calvarial sagittal sutures. Materials and methods: RLN was injected topically using a nano-sized liposome carrier into the sagittal sutures of 8- to 10-week-old wild type mice just before lateral expansion. Suture morphology, bone mineral density (BMD), and bone volume were analysed by micro-computed tomography. Collagen deposition and osteoclast differentiation were observed by Verhoeff-Van Gieson (VVG) and tartrate-resistant acid phosphatase (TRAP) staining, respectively. Results: Less collagen staining and higher tissue-specific relaxin/insulin-like family peptide receptor (Rxfp)-1 and -2 expression were observed in the RLN-treated samples after 48 hours. Increased BMD and volume, and thick well-organised osteoid tissue, with multinucleated TRAPpositive cells, were observed in RLN-treated samples after 1 week. Increased Rxfp-1 expression was observed in the sagittal sutures in the mid-suture fibrous tissue following RLN treatment. Rxfp-2 was only expressed in the calvarial bone under tensile stimulation and RLN treatment further increased its expression. Limitations: RLN-liposomes were not detected at any instance under the current experimental conditions. This is a preliminary study and the sample number limits the power of its results. VVG staining cannot quantify collagen contents but can provide preliminary information on the presence of collagen fibres.
