Over the past few decades, advances in various nanophotonic structures to enhance light–matter interactions have opened numerous opportunities for biosensing applications. Beyond the successful development of label-free nanophotonic biosensors that utilize plasmon resonances in metals and Mie resonances in dielectrics, simpler structures are required to achieve improved sensor performance and multifunctionality, while enabling cost-effective fabrication. Here, we present a simple and effectual approach to colorimetric biosensing utilizing a trilayered Gires–Tournois (GT) resonator, which provides a sensitive slow-light effect in response to low refractive index (RI) substances and thus enables to distinguish low RI bioparticles from the background with spatially distinct color differences. For low RI sensitivity, by impedance matching based on the transmission line model, trilayer configuration enables the derivation of optimal designs to achieve the unity absorption condition in a low RI medium, which is difficult to obtain with the conventional GT configuration. Compared to conventional bilayered GT resonators, the trilayered GT resonator shows significant sensing performance with linear sensitivity in various situations with low RI substances. For extended applications, several proposed designs of trilayered GT resonators are presented in various material combinations by impedance matching using equivalent transmission line models. Further, comparing the color change of different substrates with low RI NPs using finite-difference time-domain (FDTD) simulations, the proposed GT structure shows surpassing colorimetric detection.

Conventional imaging and recognition systems require an extensive amount of data storage, pre-processing, and chip-to-chip communications as well as aberration-proof light focusing with multiple lenses for recognizing an object from massive optical inputs. This is because separate chips (i.e., flat image sensor array, memory device, and CPU) in conjunction with complicated optics should capture, store, and process massive image information independently. In contrast, human vision employs a highly efficient imaging and recognition process. Here, inspired by the human visual recognition system, we present a novel imaging device for efficient image acquisition and data pre-processing by conferring the neuromorphic data processing function on a curved image sensor array. The curved neuromorphic image sensor array is based on a heterostructure of MoS2 and poly(1,3,5-trimethyl-1,3,5-trivinyl cyclotrisiloxane). The curved neuromorphic image sensor array features photon-triggered synaptic plasticity owing to its quasi-linear time-dependent photocurrent generation and prolonged photocurrent decay, originated from charge trapping in the MoS2-organic vertical stack. The curved neuromorphic image sensor array integrated with a plano-convex lens derives a pre-processed image from a set of noisy optical inputs without redundant data storage, processing, and communications as well as without complex optics. The proposed imaging device can substantially improve efficiency of the image acquisition and recognition process, a step forward to the next generation machine vision., Designing efficient bio-inspired visual recognition system remains a challenge. Here the authors present a curved neuromorphic image sensor array based on a heterostructure of MoS2 and pV3D3 integrated with a plano-convex lens for efficient image acquisition and data pre-processing.

Analysing dynamics of a single biomolecule using high-resolution imaging techniques has been had significant attentions to understand complex biological system. Among the many approaches, vertical nanopillar arrays in contact with the inside of cells have been reported as a one of useful imaging applications since an observation volume can be confined down to few-tens nanometre theoretically. However, the nanopillars experimentally are not able to obtain super-resolution imaging because their evanescent waves generate a high optical loss and a low signal-to-noise ratio. Also, conventional nanopillars have a limitation to yield 3D information because they do not concern field localization in z-axis. Here, we developed novel hybrid nanopillar arrays (HNPs) that consist of SiO2 nanopillars terminated with gold nanodisks, allowing extreme light localization. The electromagnetic field profiles of HNPs are obtained through simulations and imaging resolution of cell membrane and biomolecules in living cells are tested using one-photon and 3D multiphoton fluorescence microscopy, respectively. Consequently, HNPs present approximately 25 times enhanced intensity compared to controls and obtained an axial and lateral resolution of 110 and 210 nm of the intensities of fluorophores conjugated with biomolecules transported in living cells. These structures can be a great platform to analyse complex intracellular environment.

Nano-structural optical filters embedded in elastomers having high mechanical tunability provide the geometric degree of freedom for selective light manipulation. The active control of spectral information in typical structural optical filters is highly limited due to the substrate rigidity. Herein, we present mechanochromic transmissive optical filters by employing flexible and stretchable polymer-embedded silicon nanostructures. Si-based nanowire arrays (Si-NWAs) have been introduced to exhibit parametric resonance characteristics by controlling the period and/or diameter. Furthermore, the spectral shift phenomenon by increased diffraction efficiency was observed after the application of a uniaxial tensile force, which depends on the period of Si-NWAs with a large index contrast between the silicon nanowire and elastomer. The strain-sensitive properties of tunable Si-NWAs filters induced by light diffraction were calculated by simulation based on wave optics. The spectral tunability and light filtering features were simply demonstrated by stretching the Si-NWAs’ optical filters. Our proposed structure provides potential opportunities for a wide variety of applications, including dynamic color display, visual strain sensor and anti-counterfeiting.

This study was aimed at evaluating the effects of transforming growth factor-β on the differentiation and mRNA expression of organoids made out of human mesenchymal stem cells. Cell organoids composed of gingiva-derived stem cells were cultured in the presence of transforming growth factor-β1 at concentrations ranging from 0, 1, 10, to 20 ng/ml. Evaluations of the cell morphology of the organoids were performed on days 7, 9, 11, and 14. Quantitative cellular viability was completed on day 14. Alkaline phosphatase activity assays were performed to evaluate the differentiation of stem cells on day 14. Real-time polymerase chain reactions were used to determine the expression levels of TGF-β1, RUNX2, OCN, SOX9, and COL1A1 mRNA on day 14. The stem cells produced well-formed organoids on day 7, and the addition of transforming growth factor-β1 did not result in relevant changes in their shape. The organoids grew in size and became more intact with longer incubation times. On day 14, the diameters were 222.2 ± 9.6 , 186.1 ± 4.8 , 197.2 ± 9.6 , and 211.1 ± 19.2 m for transforming growth factor-β1 at final concentrations of 0, 1, 10, and 20 ng/ml, respectively. Quantitative cell viability results from day 14 exhibited no significant difference between the groups ( P > 0.05 ). There was significantly higher alkaline phosphatase activity with the addition of transforming growth factor-β1 with the highest value for the 1 ng/ml group ( P < 0.05 ). Real-time polymerase chain reaction results demonstrated that the mRNA expression levels of RUNX2, OCN, and SOX were higher in 1 ng/ml but did not reach statistical significance. Treatment with 1 ng/ml of transforming growth factor-β1 significantly increased COL1A1 mRNA expression at day 14. The application of transforming growth factor-β1 increased differentiation, which was confirmed by alkaline phosphatase activity and mRNA expression while maintaining cell viability.

The light field camera provides a robust way to capture both spatial and angular information within a single shot. One of its important applications is in 3D depth sensing, which can extract depth information from the acquired scene. However, conventional light field cameras suffer from shallow depth of field (DoF). Here, a vari-focal light field camera (VF-LFC) with an extended DoF is newly proposed for mid-range 3D depth sensing applications. As a main lens of the system, a vari-focal lens with four different focal lengths is adopted to extend the DoF up to ~15 m. The focal length of the micro-lens array (MLA) is optimized by considering the DoF both in the image plane and in the object plane for each focal length. By dividing measurement regions with each focal length, depth estimation with high reliability is available within the entire DoF. The proposed VF-LFC is evaluated by the disparity data extracted from images with different distances. Moreover, the depth measurement in an outdoor environment demonstrates that our VF-LFC could be applied in various fields such as delivery robots, autonomous vehicles, and remote sensing drones.

Counterfeit medicines are a fundamental security problem. Counterfeiting medication poses a tremendous threat to patient safety, public health, and the economy in developed and less developed countries. Current solutions are often vulnerable due to the limited security levels. We propose that the highest protection against counterfeit medicines would be a combination of a physically unclonable function (PUF) with on-dose authentication. A PUF can provide a digital fingerprint with multiple pairs of input challenges and output responses. On-dose authentication can verify every individual pill without removing the identification tag. Here, we report on-dose PUFs that can be directly attached onto the surface of medicines, be swallowed, and digested. Fluorescent proteins and silk proteins serve as edible photonic biomaterials and the photoluminescent properties provide parametric support of challenge-response pairs. Such edible cryptographic primitives can play an important role in pharmaceutical anti-counterfeiting and other security applications requiring immediate destruction or vanishing features., Counterfeit medicines are a threat to patient health and public safety. Here, the authors use random patterns formed by fluorescent silk microparticles with various excitation and emission pairs as an edible physical unclonable function that can directly be attached onto the surface of medicines.

Tracking multiple objects in a video sequence can be accomplished by identifying the objects appearing in the sequence and distinguishing between them. Therefore, many recent multi-object tracking (MOT) methods have utilized re-identification and distance metric learning to distinguish between objects by computing the similarity/dissimilarity scores. However, it is difficult to generalize such approaches for arbitrary video sequences, because some important information, such as the number of objects (classes) in a video, is not known in advance. Therefore, in this study, we applied a one-shot learning framework to the MOT problem. Our algorithm tracks objects by classifying newly observed objects into existing tracks, irrespective of the number of objects appearing in a video frame. The proposed method, called OneShotDA , exploits the one-shot learning framework based on an attention mechanism. Our neural network learns to classify unseen data samples using labels from a support set. Once the network has been trained, it predicts correct labels for newly received detection results based on the set of existing tracks. To analyze the effectiveness of our method, it was tested on the MOTchallenge benchmark datasets (MOT16 and MOT17 datasets). The results reveal that the performance of the proposed method was comparable with those of current state-of-the-art methods. In particular, it is noteworthy that the proposed method ranked first among the online trackers on the MOT17 benchmark.

Core regulatory transcription factors (CR TFs) orchestrate the placement of super-enhancers (SEs) to activate transcription of cell-identity specifying gene networks, and are critical in promoting cancer. Here, we define the core regulatory circuitry of rhabdomyosarcoma and identify critical CR TF dependencies. These CR TFs build SEs that have the highest levels of histone acetylation, yet paradoxically the same SEs also harbor the greatest amounts of histone deacetylases. We find that hyperacetylation selectively halts CR TF transcription. To investigate the architectural determinants of this phenotype, we used absolute quantification of architecture (AQuA) HiChIP, which revealed erosion of native SE contacts, and aberrant spreading of contacts that involved histone acetylation. Hyperacetylation removes RNA polymerase II (RNA Pol II) from core regulatory genetic elements, and eliminates RNA Pol II but not BRD4 phase condensates. This study identifies an SE-specific requirement for balancing histone modification states to maintain SE architecture and CR TF transcription.

Identity determining transcription factors (TFs), or core regulatory (CR) TFs, are governed by cell-type specific super enhancers (SEs). Drugs to selectively inhibit CR circuitry are of high interest for cancer treatment. In alveolar rhabdomyosarcoma, PAX3-FOXO1 activates SEs to induce the expression of other CR TFs, providing a model system for studying cancer cell addiction to CR transcription. Using chemical genetics, the systematic screening of chemical matter for a biological outcome, here we report on a screen for epigenetic chemical probes able to distinguish between SE-driven transcription and constitutive transcription. We find that chemical probes along the acetylation-axis, and not the methylation-axis, selectively disrupt CR transcription. Additionally, we find that histone deacetylases (HDACs) are essential for CR TF transcription. We further dissect the contribution of HDAC isoforms using selective inhibitors, including the newly developed selective HDAC3 inhibitor LW3. We show HDAC1/2/3 are the co-essential isoforms that when co-inhibited halt CR transcription, making CR TF sites hyper-accessible and disrupting chromatin looping., Core regulatory transcription factors are usually regulated by cell-type specific super enhancers (SEs). Background and objectives: NELL-1 is a competent growth factor and it reported to target cells committed to the osteochondral lineage. The secreted, osteoinductive glycoproteins are reported to rheostatically control skeletal ossification. This study was performed to determine the effects of NELL-1 on spheroid morphology and cell viability and the promotion of osteogenic differentiation of stem cell spheroids. Materials and Methods: Cultures of stem cell spheroids of gingiva-derived stem cells were grown in the presence of NELL-1 at concentrations of 1, 10, 100, and 500 ng/mL. Evaluations of cell morphology were performed using a microscope, and cell viability was assessed using a two-color assay and Cell Counting Kit-8. Evaluation of the activity of alkaline phosphatase and calcium deposition assays involved anthraquinone dye assay to determine the level of osteogenic differentiation of cell spheroids treated with NELL-1. Real-time quantitative polymerase chain reaction (qPCR) was used to evaluate the expressions of RUNX2, BSP, OCN, COL1A1, and β-actin mRNAs. Results: The applied stem cells produced well-formed spheroids, and the addition of NELL-1 at tested concentrations did not show any apparent changes in spheroid shape. There were no significant changes in diameter with addition of NELL-1 at 0, 1, 10, 100, and 500 ng/mL concentrations. The quantitative cell viability results derived on Days 1, 3, and 7 did not show significant disparities among groups (p &gt, 0.05). There was statistically higher alkaline phosphatase activity in the 10 ng/mL group compared with the unloaded control on Day 7 (p &lt, 0.05). A significant increase in anthraquinone dye staining was observed with the addition of NELL-1, and the highest value was noted at 10 ng/mL (p &lt, 0.05). qPCR results demonstrated that the mRNA expression levels of RUNX2 and BSP were significantly increased when NELL-1 was added to the culture. Conclusions: Based on these findings, we conclude that NELL-1 can be applied for increased osteogenic differentiation of stem cell spheroids.

Background and Objectives: Cuminum cyminum L. has long been used in the treatment of various diseases in multiple geographical regions. This study was performed to determine the effects of C. cyminum methanolic extract (CCT) on the cellular viability, alkaline phosphatase activity and mineralization of human mesenchymal stem cells. Materials and Methods: Bone marrow-derived stem cells were cultured in the presence of CCT at concentrations of 0, 0.001, 0.01, 0.1 and 1 &mu, g/mL. Evaluations of cell morphology were performed on days 1, 3, 7 and 14. Cellular viability was evaluated on days 1, 3, 5 and 7. On the 7th and 14th day, alkaline phosphatase activity measurements and Alizarin red S staining were conducted to assess the osteogenic differentiation of stem cells. A real-time polymerase chain reaction was used to determine the expression levels of RUNX2, BSP, OCN, COL2A1 and &beta, catenin mRNAs. Results: Stem cells in the control group showed fibroblast-like morphology and the addition of CCT at 0.001, 0.01, 0.1 and 1 &mu, g/mL did not generate noticeable changes in morphology compared with the untreated control group. The application of CCT did not produce significant changes in cellular viability or alkaline phosphatase activity compared with controls. Alizarin Red S staining was significantly increased with the application of CCT. Treatment with CCT increased the expressions of RUNX2, BSP and OCN. Conclusions: These results indicate that CCT enhanced the osteogenic differentiation of stem cells derived from bone marrow by regulating the expressions of RUNX2, BSP and OCN. Thus, the use of CCT may be applied to achieve beneficial effects on the mineralization of stem cells.

Rhabdomyosarcoma (RMS) is an aggressive pediatric malignancy of the muscle, that includes Fusion Positive (FP)-RMS harboring PAX3/7-FOXO1 and Fusion Negative (FN)-RMS commonly with RAS pathway mutations. RMS express myogenic master transcription factors MYOD and MYOG yet are unable to terminally differentiate. Here, we report that SNAI2 is highly expressed in FN-RMS, is oncogenic, blocks myogenic differentiation, and promotes growth. MYOD activates SNAI2 transcription via super enhancers with striped 3D contact architecture. Genome wide chromatin binding analysis demonstrates that SNAI2 preferentially binds enhancer elements and competes with MYOD at a subset of myogenic enhancers required for terminal differentiation. SNAI2 also suppresses expression of a muscle differentiation program modulated by MYOG, MEF2, and CDKN1A. Further, RAS/MEK-signaling modulates SNAI2 levels and binding to chromatin, suggesting that the differentiation blockade by oncogenic RAS is mediated in part by SNAI2. Thus, an interplay between SNAI2, MYOD, and RAS prevents myogenic differentiation and promotes tumorigenesis., Rhabdomyosarcomas are tumours blocked in myogenic differentiation, which despite the expression of master muscle regulatory factors, including MYOD, are unable to differentiate. Here, the authors show that SNAI2 is upregulated by MYOD through super enhancers, binds to MYOD target enhancers, and arrests differentiation.

Vision Meets Drone: Multiple Object Tracking (VisDrone-MOT2021) challenge - the forth annual activity organized by the VisDrone team - focuses on benchmarking UAV MOT algorithms in realistic challenging environments. It is held in conjunction with ICCV 2021. VisDrone-MOT2021 contains 96 video sequences in total, including 56 sequences (~24K frames) for training, 7 sequences (~3K frames) for validation and 33 sequences (~13K frames) for testing. Bounding-box annotations for novel object categories are provided every frame and temporally consistent instance IDs are also given. Additionally, occlusion ratio and truncation ratio are provided as extra useful annotations. The results of eight state-of-the-art MOT algorithms are reported and discussed. We hope that our VisDrone-MOT2021 challenge will facilitate future research and applications in the field of UAV vision. The website of our challenge can be found at http://www.aiskyeye.com/.

A thermal emitter with selective and broadband emission on different sides can mitigate greenhouse effect in hot enclosures., Passive radiative cooling functions by reflecting the solar spectrum and emitting infrared waves in broadband or selectively. However, cooling enclosed spaces that trap heat by greenhouse effect remains a challenge. We present a Janus emitter (JET) consisting of an Ag–polydimethylsiloxane layer on micropatterned quartz substrate. The induced spoof surface plasmon polariton helps overcome inherent emissivity loss of the polymer and creates near-ideal selective and broadband emission on the separate sides. This design results in not only remarkable surface cooling when the JET is attached with either side facing outwards but also space cooling when used as an enclosure wall. Thus, the JET can passively mitigate the greenhouse effect in enclosures while offering surface cooling performance comparable to conventional radiative coolers.

Acclimatable colors in response to environmental stimuli, which are naturally endowed with some living things, can provide an opportunity for humans to recognize hazardous substances without taking empirical risks. Despite efforts to create artificial responsive colors, realistic applications in everyday life require an immediate/distinct colorimetric realization with wide chromatic selectivity. A dynamically responsive virus (M‐13 phage)‐based changeable coloring strategy is presented with a highly lossy resonant promoter (HLRP). An ultrathin M‐13 phage layer for rapid response to external stimuli displays colorimetric behavior, even in its subtle swelling with strong resonances on HLRP, which is modeled using the complex effective refractive index. Optimal designs of HLRP for several material combinations allow selective chromatic responsivity from the corresponding wide color palette without modification of the dynamic responsive layer. As a practical demonstration, the spatially designed colorimetric indicator, which is insensitive/sensitive to external stimuli, provides an intuitive perception of environmental changes with hidden/revealed patterns. Furthermore, the proposed colorimetric sensor is tested by exposure to various volatile organic chemicals and endocrine disrupting chemicals for versatile detectability, and is fabricated in a wafer‐scale sample for large‐area scalability., A large‐area virus coated ultrathin colorimetric sensor with a highly lossy resonant promoter (HLRP) is developed. A dynamically responsive virus‐based changeable coloring strategy with HLRP is an efficacious approach to fuse the advantages of strong resonant photonic structures and dynamically responsive materials. An ultrathin virus layer for rapid response to external stimuli displays colorimetric behavior on HLRP.

Summary Core regulatory transcription factors (CR TFs) establish enhancers with logical ordering during embryogenesis and development. Here we report that in fusion-positive rhabdomyosarcoma, a cancer of the muscle lineage, the chief oncogene PAX3-FOXO1 is driven by a translocated FOXO1 super enhancer (SE) restricted to a late stage of myogenesis. Using chromatin conformation capture techniques, we demonstrate that the extensive FOXO1 cis-regulatory domain interacts with PAX3. Furthermore, RNA sequencing and chromatin immunoprecipitation sequencing data in tumors bearing rare PAX translocations implicate enhancer miswiring across all fusion-positive tumors. HiChIP of H3K27ac showed connectivity between the FOXO1 SE, additional intra-domain enhancers, and the PAX3 promoter. We show that PAX3-FOXO1 transcription is diminished when this network of enhancers is ablated by CRISPR. In this paper, we propose an efficient online multi-object tracking method based on the Gaussian mixture probability hypothesis density (GMPHD) filter and occlusion group management scheme where a hierarchical data association is utilized for the GMPHD filter to reduce the false negatives caused by missed detection. The hierarchical data association consisting of two modules, detection-to-track and track-to-track associations, can recover the lost tracks and their switched IDs. In addition, the proposed grouping management scheme handles occlusion problems with two main parts. The first part, “track merging” can merge the false positive tracks caused by false positive detections from occlusions. The occlusion of the false positive tracks is usually measured with some metric. In this research, we define the occlusion measure between visual objects, as sum-of-intersection-over-each-area (SIOA) instead of the commonly used intersection-over-union (IOU). The second part, “occlusion group energy minimization (OGEM)” prevents the occluded true positive tracks from false “track merging”. Each group of the occluded objects is expressed with an energy function and an optimal hypothesis will be obtained by minimizing the energy. We evaluate the proposed tracker in benchmarks such as MOT15 and MOT17 which are public datasets for multi-person tracking. An ablation study in training dataset reveals not only that “track merging” and “OGEM” complement each other, but also that the proposed tracking method shows more robust performance and less sensitiveness than baseline methods. Also, the tracking performance with SIOA is better than that with IOU for various sizes of false positives. Experimental results show that the proposed tracker efficiently handles occlusion situations and achieves competitive performance compared to the state-of-the-art methods. In fact, our method shows the best multi-object tracking accuracy among the online and real-time executable methods.

The miniaturization of 3D depth camera systems to reduce cost and power consumption is essential for their application in electrical devices that are trending toward smaller sizes (such as smartphones and unmanned aerial systems) and in other applications that cannot be realized via conventional approaches. Currently, equipment exists for a wide range of depth-sensing devices, including stereo vision, structured light, and time-of-flight. This paper reports on a miniaturized 3D depth camera based on a light field camera (LFC) configured with a single aperture and a micro-lens array (MLA). The single aperture and each micro-lens of the MLA serve as multi-camera systems for 3D surface imaging. To overcome the optical alignment challenge in the miniaturized LFC system, the MLA was designed to focus by attaching it to an image sensor. Theoretical analysis of the optical parameters was performed using optical simulation based on Monte Carlo ray tracing to find the valid optical parameters for miniaturized 3D camera systems. Moreover, we demonstrated multi-viewpoint image acquisition via a miniaturized 3D camera module integrated into a smartphone.

In this paper, we introduce a disk-shaped Ge-on-Si photodetector for refractive-index difference sensing at an operating wavelength of 1550 nm. For the implementation of a small-scale sensor, a Ge layer was formed on top of a Si layer to increase the absorption coefficient at the expense of the light-detection area. Additionally, the sensor had a ring waveguide structure along the edge of the disk formed by a recess into the inner part of the disk. This increased the interaction between the dominant optical mode traveling along the edge waveguide and the refractive index of the cladding material to be sensed, and conclusively increased detection sensitivity. The simulation results show that the proposed sensor exhibited a detection sensitivity of &gt, 50 nm/RIU (Refractive Index Unit), a quality factor of approximately 3000, and a minimum detectable refractive index change of 0.95 &times, 10&minus, 2 RIU with a small disk radius of 3 &mu, m. This corresponds to 1.67 times the sensitivity without a recess (&gt, 30 nm/RIU).

Anti-reflection coatings (ARCs) from the cornea nipple array of the moth-eye remarkably suppress the Fresnel reflection at the interface in broadband wavelength ranges. ARCs on flat glass have been studied to enhance the optical transmittance. However, little research on the implementation of ARCs on curved optical lenses, which are the core element in imaging devices, has been reported. Here, we report double-sided, bio-inspired ARCs on bi-convex lenses with high uniformity. We theoretically optimize the nanostructure geometry, such as the height, period, and morphology, since an anti-reflection property results from the gradually changed effective refractive index by the geometry of nanostructures. In an experiment, the transmittance of an ARCs lens increases up to 10% for a broadband spectrum without distortion in spot size and focal length. Moreover, we demonstrate ~30% improved transmittance of an imaging system composed of three bi-convex lenses, in series with double-sided ARCs (DARCs).

Our previous study of DNA methylation in the pediatric soft tissue tumor rhabdomyosarcoma (RMS) demonstrated that fusion-positive (FP) and fusion-negative (FN) RMS tumors exhibit distinct DNA methylation patterns. To further examine the significance of DNA methylation differences in RMS, we investigated genome-wide DNA methylation profiles in discovery and validation cohorts. Unsupervised analysis of DNA methylation data identified novel distinct subsets associated with the specific fusion subtype in FP RMS and with RAS mutation status in FN RMS. Furthermore, the methylation pattern in normal muscle is most similar to the FN subset with wild-type RAS mutation status. Several biologically relevant genes were identified with methylation and expression differences between the two fusion subtypes of FP RMS or between the RAS wild-type and mutant subsets of FN RMS. Genomic localization studies showed that promoter and intergenic regions were hypomethylated and the 3’ untranslated regions were hypermethylated in FP compared to FN tumors. There was also a significant difference in the distribution of PAX3-FOXO1 binding sites between genes with and without differential methylation. Moreover, genes with PAX3-FOXO1 binding sites and promoter hypomethylation exhibited the highest frequency of overexpression in FP tumors. Finally, a comparison of RMS model systems revealed that patient-derived xenografts most closely recapitulate the DNA methylation patterns found in human RMS tumors compared with cell lines and cell line-derived xenografts. In conclusion, these findings highlight the interaction of epigenetic changes with mutational alterations and transcriptional organization in RMS tumors, and contribute to improved molecular categorization of these tumors.

Recently developed approaches in deterministic assembly allow for controlled, geometric transformation of two-dimensional structures into complex, engineered three-dimensional layouts. Attractive features include applicability to wide ranging layout designs and dimensions along with the capacity to integrate planar thin film materials and device layouts. The work reported here establishes further capabilities for directly embedding high-performance electronic devices into the resultant 3D constructs based on silicon nanomembranes (Si NMs) as the active materials in custom devices or microscale components released from commercial wafer sources. Systematic experimental studies and theoretical analysis illustrate the key ideas through varied 3D architectures, from interconnected bridges and coils to extended chiral structures, each of which embed n-channel Si NM MOSFETs (nMOS), Si NM diodes, and p-channel silicon MOSFETs (pMOS). Examples in stretchable/deformable systems highlight additional features of these platforms. These strategies are immediately applicable to other wide-ranging classes of materials and device technologies that can be rendered in two-dimensional layouts, from systems for energy storage, to photovoltaics, optoelectronics, and others.

Background Dietary supplementation of zinc oxide (ZnO) to 2000 to 4000 mg/kg is known to be effective for the prevention and treatment of post-weaning diarrhea in the pig. Such a ‘pharmacological’ supplementation, however, can potentially result in environmental pollution of the heavy metal, because dietary ZnO is mostly excreted unabsorbed. Two experiments (Exp.) were performed in the present study to determine the effects of a lipid-coated ZnO supplement Shield Zn (SZ) compared with those of ZnO. Methods In Exp. 1, a total of 240 21-day-old weanling pigs were fed a diet supplemented with 100 mg Zn/kg as ZnO (ZnO-100), ZnO-2500, SZ-100, or SZ-200 in 24 pens for 14 days on a farm with its post-weaning pigs exhibiting a low incidence of diarrhea. Exp. 2 was performed using 192 24-day-old piglets as in Exp. 1 on a different farm, which exhibited a high incidence of diarrhea. Curved image sensors, which are a key component in bio-inspired imaging systems, have been widely studied because they can improve an imaging system in various aspects such as low optical aberrations, small-form, and simple optics configuration. Many methods and materials to realize a curvilinear imager have been proposed to address the drawbacks of conventional imaging/optical systems. However, there have been few theoretical studies in terms of electronics on the use of a lateral photodetector as a flexible image sensor. In this paper, we demonstrate the applicability of a Si-based lateral phototransistor as the pixel of a high-efficiency curved photodetector by conducting various electrical simulations with technology computer aided design (TCAD). The single phototransistor is analyzed with different device parameters: the thickness of the active cell, doping concentration, and structure geometry. This work presents a method to improve the external quantum efficiency (EQE), linear dynamic range (LDR), and mechanical stability of the phototransistor. We also evaluated the dark current in a matrix form of phototransistors to estimate the feasibility of the device as a flexible image sensor. Moreover, we fabricated and demonstrated an array of phototransistors based on our study. The theoretical study and design guidelines of a lateral phototransistor create new opportunities in flexible image sensors.

The aim of the current study is to examine the improving effect of Sasa borealis stem (SBS) extract extracts on high-fat diet (HFD)-induced hepatic steatosis in rats. To determine the hepatoprotective effect of SBS, we fed rats a normal regular diet (ND), HFD, and HFD supplemented with 150 mg/kg body weight (BW) SBS extracts for five weeks. We found that the body weight and liver weight of rats in the HFD + SBS group were significantly lower than those in the HFD group. Significantly lower serum total cholesterol (TC) and triglyceride (TG) concentrations were observed in the SBS-supplemented group compared with the HFD group. We also found that the HFD supplemented with SBS group showed dramatically reduced hepatic lipid accumulation compared to the HFD alone group, and administration of SBS resulted in dramatic suppression of TG, TC in the HFD-induced fatty liver. In liver gene expression within the SBS treated group, PPAR alpha was significantly increased and SREBP-1c was significantly suppressed. SBS induced a significant decrease in the hepatic mRNA levels of PPAR gamma, FAS, ACC1, and DGAT2. In conclusion, SBS improved cholesterol metabolism, decreased lipogenesis, and increased lipid oxidation in HFD-induced hepatic steatosis in rats, implying a potential application in treatment of non-alcoholic fatty liver disease.

The recent emergence of materials for electronic systems that are capable of programmable self-destruction and/or bio/eco-resorption creates the potential for important classes of devices that cannot be easily addressed using conventional technologies, ranging from temporary biomedical implants to enviromentally benign environmental monitors to hardware secure data systems. Although most previous demonstrations rely on wet chemistry to initiate transient processes of degradation/decomposition, options in “dry transient electronic systems” could expand the range of possible uses. The work presented here introduces materials and composite systems in which sublimation under ambient conditions leads to mechanical fragmentation and disintegration of active devices upon disappearance of a supporting substrate, encapsulation layer, interlayer dielectric and/or gate dielectric. Examples span arrays of transistors based on silicon nanomembranes with specialized device designs to solar cells adapted from commercial components.

Carbon nanotubes (CNTs) are a primary nanomaterial that have outstanding physical and mechanical characteristics, and CNTs can be combined with cement-based materials to alter their heating characteristics. In this study, the types of CNTs used were multiwalled carbon nanotubes (MWCNTs) and single-walled carbon nanotubes (SWCNTs). Experiments were performed to determine the altered heating characteristics of the CNT cement mortars. The parameters of the experiment were CNT type, CNT content, curing age, and applied voltage. The results for the different CNT cement mortars indicate that mixing SWCNTs with water to produce CNT cement mortars was more effective for modifying the heating characteristics compared to mixing MWCNTs with water. In addition, field emission scanning electron microscope (FE-SEM) images supported the results found in the heating experiments.

We present the effect of geometrical parameters, particularly shape, on optical absorption enhancement for thin film solar cells based on crystalline silicon (c-Si) and gallium arsenide (GaAs) using a rigorous coupled wave analysis (RCWA) method. It is discovered that the "sweet spot" that maximizes efficiency of solar cells exists for the design of nanophotonic surfaces. For the case of ultrathin, rod array is practical due to the effective optical resonances resulted from the optimum geometry whereas parabola array is viable for relatively thicker cells owing to the effective graded index profile. A specific value of thickness, which is the median value of other two devices tailored by rod and paraboloid, is optimized by truncated shape structure. It is therefore worth scanning the optimum shape of nanostructures in a given thickness in order to achieve high performance. (C) 2016 Optical Society of America

We calculated diffraction efficiencies of grating structures inspired byMorphobutterfly wings by using a rigorous coupled-wave analysis method. The geometrical effects, such as grating width, period, thickness, and material index, were investigated in order to obtain better optical performance. Closely packed grating structures with an optimized membrane thickness show vivid reflected colors and provide high sensitivity to surrounding media variations, which is applicable to vapor sensing or healthcare indicators.Morphostructures with high index materials such as zinc sulfide or gallium phosphide generate white color caused by broadband reflection that can be used as reflected light sources for display applications.

We present bifunctional nanostructured (NS) coverglasses with enhanced transmittance and self-cleaning function for improving the efficiency of a photovoltaic (PV) module with an InGaP/GaAs/Ge triple-junction solar cell. Prior to the fabrication of NS coverglasses, theoretical investigations were carried out using a rigorous coupled-wave analysis method to determine the desirable glass nanostructures that can effectively enhance the light absorption of the PV module. The transmission properties of the fabricated NS coverglasses with different glass nanostructures were systematically analyzed by considering the absorption spectrum of three subcells of the solar cell in order to find the most effective NS coverglass for enhancing the photocurrent of the PV module and thereby improving the conversion efficiency. The PV module with the most effective NS coverglass showed 4.2% enhanced short-circuit current density and 4.3% enhanced efficiency compared with the PV module with a bare coverglass measured under one sun of the air mass 1.5 global, showing the necessity of integrating a finely designed NS coverglass to effectively improve the efficiency of various PV systems with multijunction solar cells.

Changes in meat quality traits are strongly associated with alterations in postmortem metabolism which depend on genetic variations, especially nonsynonymous single nucleotide variations (nsSNVs) having critical effects on protein structure and function. To selectively identify metabolism-related nsSNVs, next-generation transcriptome sequencing (RNA-Seq) was carried out using RNAs from porcine liver, which contains a diverse range of metabolic enzymes. The multiplex SNV genotyping analysis showed that various metabolism-related genes had different nsSNV alleles. Moreover, many nsSNVs were significantly associated with multiple meat quality traits. Particularly, ch7:g.22112616A>G SNV was identified to create a single amino acid change (Thr/Ala) at the 145th residue of H1.3-like protein, very close to the putative 147th threonine phosphorylation site, suggesting that the nsSNV may affect multiple meat quality traits by affecting the epigenetic regulation of postmortem metabolism-related gene expression. Besides, one nonsynonymous variation, probably generated by gene duplication, led to a stop signal in porcine testicular carbonyl reductase (PTCR), resulting in a C-terminal (E281-A288) deletion. Molecular docking and energy minimization calculations indicated that the binding affinity of wild-type PTCR to 5α-DHT, a C(21)-steroid, was superior to that of C-terminal-deleted PTCR or human carbonyl reductase, which was very consistent with experimental data, reported previously. Furthermore, P284 was identified as an important residue mediating the specific interaction between PTCR and 5α-DHT, and phylogenetic analysis showed that P284 is an evolutionarily conserved residue among animal carbonyl reductases, which suggests that the C-terminal tails of these reductases may have evolved under evolutionary pressure to increase the substrate specificity for C(21)-steroids and facilitate metabolic adaptation. Altogether, our RNA-Seq revealed that selective nsSNVs were associated with meat quality traits that could be useful for successful marker-assisted selection in pigs and also represents a useful resource to enhance understanding of protein folding, substrate specificity, and the evolution of enzymes such as carbonyl reductase.
