ObjectiveThe purpose of the study is to evaluate the effects of anticardiolipin (aCL) and/or anti-β2-glycoprotein-I (aβ2GPI) antibodies, namely antiphospholipid antibodies (aPL), on in vitro fertilization (IVF) outcomes.Materials and methodsThe study group comprised infertile women with aPL undergoing IVF-ET cycles. Controls were infertile women with tubal etiology without aPL. The impact of aPL on reproductive outcomes, such as oocyte quality, embryo quality, and implantation capacity, was compared between the study group and controls. Additionally, peripheral blood T cell subsets, such as T helper (Th)1, Th2, Th17, and T regulatory (Treg) cells and cytokines, were analyzed by the flow cytometry. Differences between the study group and controls were analyzed.ResultsA total of 132 infertile women, including 44 women with aPL, and 88 controls were sequentially recruited for this study. Women with aPL had lower numbers of total and perfect/available embryos and lower rates of MII oocytes, blastocyst formation, perfect and available embryos, implantation, clinical pregnancy, and take-home baby. Additionally, imbalanced Th1/Th2 and Th17/Treg ratios, significantly higher levels of serum IL-2, TNF-α, IFN-γ, and IL-17A, and a significantly lower serum IL-4 were noticed in women with aPL compared to controls.ConclusionWomen with aPL such as aCL and/or aβ2GPI antibodies were associated with adverse IVF outcomes. Early screening for aPL and appropriate consultation for couples undergoing IVF should be considered. In addition, underlying immunopathology and inflammatory immune mechanisms associated with aPL should be further explored.

Abstract Maintaining homeostasis of the decidual immune microenvironment at the maternal–fetal interface is essential for placentation and reproductive success. Although distinct decidual immune cell subpopulations have been identified under normal conditions, systematic understanding of the spectrum and heterogeneity of leukocytes under recurrent miscarriage in human deciduas remains unclear. To address this, we profiled the respective transcriptomes of 18,646 primary human decidual immune cells isolated from patients with recurrent pregnancy loss (RPL) and healthy controls at single-cell resolution. We discovered dramatic differential distributions of immune cell subsets in RPL patients compared with the normal decidual immune microenvironment. Furthermore, we found a subset of decidual natural killer (NK) cells that support embryo growth were diminished in proportion due to abnormal NK cell development in RPL patients. We also elucidated the altered cellular interactions between the decidual immune cell subsets in the microenvironment and those of the immune cells with stromal cells and extravillous trophoblast under disease state. These results provided deeper insights into the RPL decidual immune microenvironment disorder that are potentially applicable to improve the diagnosis and therapeutics of this disease.

Inflammasomes, intracellular, multimeric protein complexes, are assembled when damage signals stimulate nucleotide-binding oligomerization domain receptors (NLRs). Several inflammasomes have been reported, including the NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3), NLRP1, NLRP7, ice protease-activating factor (IPAF), absent in melanoma 2 (AIM2) and NLR family CARD domain-containing protein 4 (NLRC4). Among these inflammasomes, the NLRP3 inflammasome is the most well-studied in terms of structure and function. Unlike other inflammasomes that can only be activated by a finite number of pathogenic microorganisms, the NLRP3 inflammasome can be activated by the imbalance of the internal environment and a large number of metabolites. The biochemical function of NLRP3 inflammasome is to activate cysteine-requiring aspartate proteinase-1 (caspase-1), which converts pro-IL-1β and pro-IL-18 into their active forms, namely, IL-1β and IL-18, which are then released into the extracellular space. The well-established, classic role of NLRP3 inflammasome has been implicated in many disorders. In this review, we discuss the current understanding of NLRP3 inflammasome and its critical role in gynecological disorders and obstetrical complications.

Nanoscale zero-valent iron (NZVI) has a high adsorption capacity for heavy metals, but easily forms aggregates. Herein, preprocessed undulating venus shell (UVS) is used as support material to prevent NZVI from reuniting. The SEM and TEM results show that UVS had a porous layered structure and NZVI particles were evenly distributed on the UVS surface. A large number of adsorption sites on the surface of UVS-NZVI are confirmed by IR and XRD. UVS-NZVI is used for adsorption of Pb2+ and Cd2+ at pH = 6.00 in aqueous solution, and the experimental adsorption capacities are 29.91 and 38.99 mg g−1 at optimal pH, respectively. Thermodynamic studies indicate that the adsorption of ions by UVS-NZVI is more in line with the Langmuir model when Pb2+ or Cd2+ existed alone. For the mixed solution of Pb2+ and Cd2+, only the adsorption of Pb2+ by UVS-NZVI conforms to the Langmuir model. In addition, the maximum adsorption capacities of UVS-NZVI for Pb2+ and Cd2+ are 93.01 and 46.07 mg g−1, respectively. Kinetic studies demonstrate that the determination coefficients (R2) of the pseudo first-order kinetic model for UVS-NZVI adsorption of Cd2+ and Pb2+ are higher than those of the pseudo second-order kinetic model and Elovich kinetic model. Highly efficient performance for metal removal makes UVS-NZVI show potential application to heavy metal ion adsorption.

Abstract Interleukin-28A (IL-28A) modulates CD11c+ dendritic cell (DC) function and promotes type 1T helper (Th1) differentiation, thus suppressing allergic airway diseases. However, the function of the IL-28A isoform IL-28B in these diseases remains largely unknown. In this study, we revealed a novel role of IL-28B in inducing type 1 immunity and protecting against ovalbumin (OVA)-induced allergic asthma in mice. IL-28B overexpression in wild-type mice promoted natural killer (NK) cell polarization in the lung, leading to the increased number of interferon (IFN)-γ-producing NK1 cells as well as Th1 differentiation. Importantly, IL-28B overexpression had no protective effect on OVA-induced asthma in IFN-γ-knockout (IFN-γ−/−) mice. These results demonstrate that IL-28B ameliorates experimental allergic asthma via enhancing NK cell polarization, which might be useful for prevention and treatment of allergic asthma.

A new method using cloud point extraction-flame atomic absorption spectrometry was established by using ammonium pyrrolidine dithiocarbamate as a chelating agent and the cloud point phenomenon of a complexed anion-non-ionic surfactant mixture (Sodium dodecyl sulfate-Triton X-114 mixture). It was used for determining the morphology of chromium in the water and sediments of Haidian Creek. The experimental conditions inorganic electrolytes, pH of a solution, dosage of reagent, the temperature of a water bath, and heating time were optimized according to their effect on the determination of absorbance. The interference of common coexisting ions is discussed. The results show that under the optimized conditions, the standard curve for Cr(III) and Cr(VI) were both linear in the range of 0–9 × 10−3 μg/L. The detection limits for Cr(III) and Cr(VI) were 2.3 and 2.7 μg/L, the correlation coefficients of the standard curve were 0.9904 and 0.9911, and the relative standard deviations were 3.7% and 4.5% (C = 0.04 μg/mL, n = 11), respectively. Compared with the common coexisting ions, Fe3+ produced greater interference that cannot be neglected. A hybrid masking agent was prepared by mixing diethyltriaminepentaacetic acid and 30% triethanolamine solution at 1:25 ratio and then used in experiments. It could mask the interference of Fe3+ in the samples. The proposed method was successfully applied in the determination of chromium in water and sediment samples collected from Haidian Creek. It had recovery rates of 86%–98% and 90%–104% for Cr(VI) and Cr(III) respectively, and it gave satisfactory results. [ABSTRACT FROM AUTHOR]

IL-28B is a member of the newly discovered type III IFN family and exhibits unique antiviral properties compared with other family members. NK cells play a critical role in defending against viruses; however, little is known about the role of IL-28B in NK cell function. In a mouse model of influenza Avirus (mouse adapted influenza A/PR/8/34 strain) infection, long-term overexpression of IL-28B induced by hepatocyte-specific gene delivery exerted a strong antiviral effect in the presence of NK cells. In IL-28B-overexpressing wild-type mice, the percentages and absolute numbers of NK cells in the spleen, liver, and lung were markedly increased, with higher proliferation and accelerated NK cell maturation based on phenotypes staining with CD11b and CD27 or CD11b and KLRG1. Furthermore, the effect of IL-28B on NK cells was macrophage dependent, as confirmed in an in vitro coculture assay and in in vivo macrophage- or alveolar macrophage-depletion experiments. Transwell studies demonstrated that CFSE-labeled NK cell proliferation was driven, in a dose-dependent manner, by unknown soluble factor(s) secreted by IL-28B-stimulated alveolar macrophages, without requiring direct cell-cell contact. Depression is one of the most common mental disorders in modern society. The disease not only seriously harms people’s quality of life, but also brings a huge burden to families and society because of the high suicide rate and disability rate caused by it. Recent studies have shown that the intestinal flora plays an important role the occurrence and progression of depression and dysfunctional gut-brain communication may be an essential pathological mechanism of depression. The “microbiota-gut-brain” (MGB) axis allows the intestinal flora to participate in the two-way information exchange between the intestine and the brain. Most previous studies mainly focus on exploring the individual characteristics of the intestinal flora in patients with depression and the relationship between the intestinal flora and the occurrence of depression, revealing that maintaining a healthy intestinal flora may be a new idea for the prevention and treatment of depression in the future. This article summarizes recent progress in this research field. We hope that this review can provide insights into the action mechanism of anti-depressive agents from the perspective of the MGB axis, and also provide a reference for the application of probiotics in new depression treatments.

AlthoughNKcells are classified as innate immune cells, recent studies have demonstrated the transformation of NKcells into long-lived memory cells that contribute to secondary immune responses in certain mouse models. However, whether NK cells mount an Agspecific memory response to acute influenza virus infection has not yet been examined. Here, we show that, consistent with previous studies, lung NK cells play an important role in controlling viral proliferation after primary influenza virus infection. However, although lung NK cells display a memory phenotype at the late stage of infection, these cells do not protect mice against secondary influenza virus infection. Interestingly, liver NK cells from influenza virus-infected mice possess a memory phenotype and protect mice against secondary influenza virus infection. Memory-like liver NK cells display a CD49a+DX5- phenotype, and the adoptive transfer of purified liver CD49a+DX5- NK cells into naive mice followed by viral infection results in protective immunity and decreased viral titer. Moreover, we demonstrate that primary inactivated influenza virus induces memory NK cells residing in the liver of Rag1-/- mice. Collectively, these data suggest that liver CD49a+DX5- NK cells remember encountered Ag from influenza virus after primary infection and are more protective upon subsequent infection.
