Type I gamma-ray bursts (GRBs) are believed to originate from compact binary mergers usually with a duration of main emission less than 2 s. However, recent observations of GRB 211211A and GRB 230307A indicate that some merger-origin GRBs could last much longer. Since they show strikingly similar properties (indicating a common mechanism), which are different from the classic “long”-short burst (e.g., GRB 060614), we find they form an interesting subclass of type I GRBs, and we suggest to name them as type IL GRB. We find that the prompt emission of type IL GRB is composed of three episodes: (1) a precursor followed by a short quiescent (or weak emission) period, (2) a long-duration main emission, and (3) an extended emission. With this burst pattern, a good candidate, GRB 170228A, was found in the Fermi/Gamma-ray Burst Monitor archive data. Temporal and spectral analyses indeed show that GRB 170228A falls in the same group with GRB 211211A and GRB 230307A in many diagnostic figures. Thus, this burst pattern could be a good reference for rapidly identifying type IL GRBs and very helpful for conducting low-latency follow-up observation. We estimated the occurrence rate and discussed the physical origins and implications for the three emission episodes of type IL GRBs. Our analysis suggests the premerger precursor model, especially the magnetar super flare model, is more favored for type IL GRBs. More observations in multiwavelength and multimessenger are required to deepen our understanding of this subclass of GRB.

The PR interval on the electrocardiogram reflects atrial and atrioventricular nodal conduction time. The PR interval is heritable, provides important information about arrhythmia risk, and has been suggested to differ among human races. Genome-wide association (GWA) studies have identified common genetic determinants of the PR interval in individuals of European and Asian ancestry, but there is a general paucity of GWA studies in individuals of African ancestry. We performed GWA studies in African American individuals from four cohorts (n = 6,247) to identify genetic variants associated with PR interval duration. Abstract Objective The aim of this study is to develop a nomogram model for predicting the occurrence of intramyocardial hemorrhage (IMH) in patients with Acute Myocardial Infarction (AMI) following Percutaneous Coronary Intervention (PCI). The model is constructed utilizing clinical data and the SYNTAX Score (SS), and its predictive value is thoroughly evaluated. Methods A retrospective study was conducted, including 216 patients with AMI who underwent Cardiac Magnetic Resonance (CMR) within a week post-PCI. Clinical data were collected for all patients, and their SS were calculated based on coronary angiography results. Based on the presence or absence of IMH as indicated by CMR, patients were categorized into two groups: the IMH group (109 patients) and the non-IMH group (107 patients). The patients were randomly divided in a 7:3 ratio into a training set (151 patients) and a validation set (65 patients). A nomogram model was constructed using univariate and multivariate logistic regression analyses. The predictive capability of the model was assessed using Receiver Operating Characteristic (ROC) curve analysis, comparing the predictive value based on the area under the ROC curve (AUC). The analysis of gamma-ray burst (GRB) spectra often relies on empirical models lacking a distinct physical explanation. Previous attempts to couple physical models with observed data focus on individual burst studies, fitting models to segmented spectra with independent physical parameters. However, these approaches typically neglect to explain the time evolution of observed spectra. In this study, we propose a novel approach by incorporating the synchrotron radiation model to provide a self-consistent explanation for a selection of single-pulse GRBs. Our study comprehensively tests the synchrotron model under a unified physical condition, such as a single injection event of electrons. By tracing the evolution of cooling electrons in a decaying magnetic field, our model predicts time-dependent observed spectra that align well with the data. Using a single set of physical parameters, our model successfully fits all time-resolved spectra within each burst. Our model suggests that the rising phase of the GRB light curve results from the increasing number of radiating electrons, while the declining phase is attributed to the curvature effect, electron cooling, and the decaying magnetic field. Our model provides a straightforward interpretation of the peak energy’s evolution, linked to the decline of the magnetic field and electron cooling due to the expansion of the GRB emission region. Our findings strongly support the notion that spectral and temporal evolution in GRB pulses originates from the expansion of the GRB emission region, with an initial radius of approximately 10 ^15 cm, and synchrotron radiation as the underlying emission mechanism.

The radiation mechanism (thermal photosphere or magnetic synchrotron) and the progenitor of gamma-ray bursts (GRBs) are under hot debate. Recently discovered, the prompt long-duration (∼10 s, normally from the collapse of massive stars) property of GRB 211211A strongly conflicts with its association with a kilonova (normally from the merger of two compact objects, NS–NS, NS–BH, or NS–WD, duration ≲2 s). In this paper, we find that the probability photosphere model with a structured jet can satisfactorily explain this peculiar long duration, through the duration stretching effect (∼3 times) on the intrinsic longer (∼3 s) duration of an NS–BH merger, the observed empirical 2SBPL spectrum (with soft low-energy index α of ∼−1), and its evolution. In addition, much evidence of the NS–BH merger origin is found, especially the good fit of the afterglow-subtracted optical−near-IR light curves by the significant thermal cocoon emission and the sole thermal “red” kilonova component. Finally, a convincing new explanation for the X-ray afterglow plateau is revealed.

The present study validated the general extended technology acceptance model for e-learning (GETAMEL) with the survey data from the English as a foreign language (EFL) online class during the novel coronavirus lockdown period. A total of 678 undergraduates participated in the survey. Structural equation modeling was employed to analyze the data. The results showed that the influence of perceived usefulness of students on their intentional behavior to use the online learning system was not mediated by their attitude, indicating a very limited role of attitude toward technology in the model. Enjoyment and self-efficacy had no significant effects on the internal constructs, raising theoretical concerns on the applicability of this general model into specific contexts. In addition, we found that experience might be a moderator rather than an antecedent of the internal constructs in the model.

Gut microbiota can influence host gene expression and physiology through metabolites. Besides, the presence or absence of gut microbiome can reprogram host transcriptome and epitranscriptome as represented by N6‐methyladenosine (m6A), the most abundant mammalian mRNA modification. However, which and how gut microbiota‐derived metabolites reprogram host transcriptome and m6A epitranscriptome remain poorly understood. Here, investigation is conducted into how gut microbiota‐derived metabolites impact host transcriptome and m6A epitranscriptome using multiple mouse models and multi‐omics approaches. Various antibiotics‐induced dysbiotic mice are established, followed by fecal microbiota transplantation (FMT) into germ‐free mice, and the results show that bile acid metabolism is significantly altered along with the abundance change in bile acid‐producing microbiota. Unbalanced gut microbiota and bile acids drastically change the host transcriptome and the m6A epitranscriptome in multiple tissues. Mechanistically, the expression of m6A writer proteins is regulated in animals treated with antibiotics and in cultured cells treated with bile acids, indicating a direct link between bile acid metabolism and m6A biology. Collectively, these results demonstrate that antibiotic‐induced gut dysbiosis regulates the landscape of host transcriptome and m6A epitranscriptome via bile acid metabolism pathway. In this study, we investigated an absorber based on a center-aligned tandem nanopillar array for ultra-broadband solar energy harvesting theoretically. A high-efficiency, omnidirectional absorber was obtained by introducing the center-aligned tandem nanopillar array embedded in an Al2O3 dielectric layer. The multi-coupling modes at different wavelengths were interpreted. The strong absorption can be adjusted by changing the radii and heights of nanopillars. According to the simulation results, the average absorptance of the absorber exceeded 94% in the wavelength range from 300 nm to 2000 nm. In addition, the high-efficiency absorption was insensitive to the incident angle and polarization state. The research not only proposed an absorber which possesses a huge potential value for application areas, such as thermal photovoltaic systems, infrared detection, and isotropic absorption sensors, but also pointed out a new way to design an absorber with high efficiency in an ultrabroad wavelength range.

Abstract Background The effect of statin treatment on circulating coenzyme Q10 (CoQ10) has been studied in numerous randomized controlled trails (RCTs). However, whether statin treatment decreases circulating CoQ10 is still controversial. Methods PubMed, EMBASE, and the Cochrane Library were searched to identify RCTs to investigate the effect of statin treatment on circulating CoQ10. We calculated the pooled standard mean difference (SMD) using a fixed effect model or random effect model to assess the effect of statin treatment on circulating CoQ10. The methodological quality of the studies was determined according to the Cochrane Handbook. Publication bias was evaluated by a funnel plot, the Egger regression test, and the Begg–Mazumdar correlation test. Results Twelve RCTs with a total of 1776 participants were evaluated. Compared with placebo, statin treatment resulted in a reduction of circulating CoQ10 (SMD, − 2.12; 95% CI, − 3.40 to − 0.84; p = 0.001), which was not associated with the duration of statin treatment (Exp, 1.00; 95% CI, 0.97 to 1.03; p = 0.994). Subgroup analysis demonstrated that both lipophilic statins (SMD, − 1.91; 95% CI, − 3.62 to 0.2; p = 0.017) and hydrophilic statins (SMD, − 2.36; 95% CI, − 4.30 to − 0.42; p = 0.028) decreased circulating CoQ10, and no obvious difference was observed between the two groups (SMD, − 0.20; 95% CI, − 0.208 to 0.618; p = 0.320). Based on computer-aided drug design (CADD), the active groups of the known active small molecule compounds that can bind to EGFR target protein were analyzed through the molecular docking method. Then, 12 novel asiatic acid derivatives were synthesized by introducing active groups at ring A and C-28 positions of asiatic acid. The structures of these novel compounds were determined by NMR and MS. Furthermore, the anti-tumor activities of these derivatives on human lung cancer cells (A549) and human breast cancer cells (MCF-7) were evaluated by MTT assay. In conclusion, compounds I4 and II3 have stronger anti-cancer activity than parent compounds, the activities were stronger than gefitinib and comparable to afatinib, which may be potential candidate compounds for tumor therapy. Based on the simulated docking of Epidermal growth factor receptor inhibitors with known active small molecule compounds, computer-aided drug design technology was used to analyze key amino acid fragments and determine the active groups binding with key sites. Then, twelve novel analogues of oleanolic acid (OA) were synthesized by introducing active groups at the C-3 and C-28 positions of OA. The structures of these novel analogues were confirmed by NMR and MS. Furthermore, the antitumor activities of these novel analogues were evaluated by MTT assay. As a result, compounds I3 and II3 showed stronger cytotoxicity on tumor cells than positive controls. In conclusion, our study synthesized twelve novel analogues of OA and determined compounds I3 and II3 had better antitumor effect, which may be potential candidate compounds for tumor therapy. Objectives: This study addresses the bioavailability challenges associated with oral nicotinamide mononucleotide (NMN) administration by introducing an innovative NMN formulation incorporated with hydroxyapatite (NMN–HAP). Methods: The NMN–HAP was developed using a wet chemical precipitation and physical adsorption method. To assess its superiority over conventional free NMN, we examined NMN, nicotinamide adenine dinucleotide (NAD+), and nicotinamide riboside (NR) levels in mouse plasma and tissues following oral administration of NMN–HAP. Key findings: NMN–HAP nanoparticles demonstrated a rod-shaped morphology, with an average size of ~50 nm, along with encapsulation efficiency and drug loading capacity exceeding 40%. In vitro, drug release results indicated that NMN–HAP exhibited significantly lower release compared with free NMN. In vivo studies showed that NMN–HAP extended circulation time, improved bioavailability compared with free NMN, and elevated plasma levels of NMN, NAD+, and NR. Moreover, NMN–HAP administration displayed tissue-specific distribution with a substantial accumulation of NMN, NAD+, and NR in the brain and liver. Conclusion: NMN–HAP represents an ideal formulation for enhancing NMN bioavailability, enabling tissue-specific delivery, and ultimately elevating in vivo NAD+ levels. Considering HAP's biocompatible nature and versatile characteristics, we anticipate that this system has significant potential for various future applications. Perforin-1 (PRF), a cytotoxic lymphocyte pore-forming protein, plays an important role in the action of cytotoxic T cells and natural killer cells in that it causes the lysis of abnormal body cells and the elimination of virus-infected cells and tumors. Upon degranulation, PRF inserts itself into the target cell's plasma membrane, forming a pore. The subsequent translocation of pro-apoptotic granzymes (including granzyme B, A, M et al.) into the cytoplasm provides the proteases with access to numerous protein substrates that promote apoptosis after cleavage. These proteases are believed to be the main executioners of target cell apoptosis. Although the PRF and granzyme components are both critical to this process and in some way involved in inducing cell death in target cells, the inhibition of tumor growth could still be efficient in granzyme-deficient mice. It is unclear whether PRF alone can suppress tumors. In this study, we discovered that forced ectopic expression of PRF alone, in the absence of granzymes, could mediate cell death in cancer cells. Notably, transient expression of both full-length and truncated active-form PRF in human Hep G2, SK-BR-3, and HeLa cells was found to induce apparent cell growth inhibition and cell death, as evidenced by chromosome condensation and DNA fragmentation, increased caspase-3 activity, and the release of apoptosis inducing factor (AIF) and cytochrome c from the mitochondria. This PRF-induced cell death could be abrogated by pan-caspase inhibitor (Z-VAD) and mitochondria protector (TAT-BH4). The implication of these results is that ectopically expressed PRF has apoptosis-inducing abilities, and PRF alone is sufficient to induce apoptotic cell death in cells with ectopic expression. Taking this into consideration, our results suggest the possibility of using PRF as a pro-apoptotic gene for tumor therapeutics.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), a novel coronavirus with high nucleotide identity to SARS-CoV and to SARS-related coronaviruses that have been detected in horseshoe bats, has spread across the world and had a global effect on healthcare systems and economies1,2. A suitable small animal model is needed to support the development of vaccines and therapies. Here we report the pathogenesis and transmissibility of SARS-CoV-2 in golden (Syrian) hamsters (Mesocricetus auratus). Immunohistochemistry assay demonstrated the presence of viral antigens in nasal mucosa, bronchial epithelial cells and areas of lung consolidation on days 2 and 5 after inoculation with SARS-CoV-2, followed by rapid viral clearance and pneumocyte hyperplasia at 7 days after inoculation. We also found viral antigens in epithelial cells of the duodenum, and detected viral RNA in faeces. Notably, SARS-CoV-2 was transmitted efficiently from inoculated hamsters to naive hamsters by direct contact and via aerosols. Transmission via fomites in soiled cages was not as efficient. Although viral RNA was continuously detected in the nasal washes of inoculated hamsters for 14 days, the communicable period was short and correlated with the detection of infectious virus but not viral RNA. Inoculated and naturally infected hamsters showed apparent weight loss on days 6–7 post-inoculation or post-contact; all hamsters returned to their original weight within 14 days and developed neutralizing antibodies. Our results suggest that features associated with SARS-CoV-2 infection in golden hamsters resemble those found in humans with mild SARS-CoV-2 infections. The pathogenicity and transmissibility of SARS-CoV-2 in golden (Syrian) hamsters resemble features of COVID-19 in human patients, suggesting that these hamsters could be used to model this disease.
