This paper proposes an improved DeepLabV3+ lightweight algorithm for the identification of installation gaps in spacer replacement robots. By using lightweight MobileNetV3 to extract semantic features of spacer installation gaps, parameters and computational complexity are reduced; Perform dimensionality reduction and dimensionality increase operations on the ASPP module to reduce the number of model parameters; Introduce ECA module to restore the clarity of target boundaries; Use a loss function combining Focal Loss and Dice Loss to enhance segmentation performance. The experimental results show that the improved DeepLabV3+ algorithm improves MIoU, MPA, and prediction speed, while balancing segmentation accuracy and speed, and can effectively segment the installation gap of the spacer.

It is a developing trend for transmission line operation robots to replace manual operations in line operation and maintenance. However, the existing on-line methods of robots are complex and inefficient, which seriously affects the progress of the entire operation and maintenance. In order to solve this problem, this paper studies the automatic on-line and off-line control method of live working robot for 500kV transmission lines. Firstly, the on-line and off-line structure of the robot and its operating mechanism are analyzed, and on this basis, the relevant mathematical model is established. Through the analysis of the model, cascade fuzzy PID is used to control the robot’s attitude leveling. Finally, through MATLAB software simulation, the cascade fuzzy PID control effect is the best.

In this paper, four kinds of coatings Ni + 30% WC, Ni + 15% WC, Ni, and Co are prepared on EH40 steel by laser cladding technology. Electrochemical experiment tests at a simulating low-temperature (–40°C) environment in 0.5 mol·L−1 hydrochloric acid (HCl) solution and 3.5 wt% NaCl solution were conducted, and the polarization curve and electrochemical impedance spectroscopy (EIS) impedance spectrum were obtained. The results show that the Ni + 15% WC coating has the smallest corrosion current and 0.5 mol·L−1 HCl solution and 3.5 wt% NaCl solution have the maximum polarization resistance and impedance arc. Coatings will be treated in a 0.5 mol·L−1 HCl solution and 3.5 wt% NaCl solution at –40°C immersion for 30 days; finally, X-ray photoelectron spectroscopy (XPS) and scanning electron microscope (SEM) were used for test passivation films of Ni + 15% WC samples and sample surface morphology after corrosion. XPS results show that Fe, Mo, and Ni elements can form metal oxides in the 0.5 mol·L−1 HCl solution, and Cr, Fe, Mo, Ni, and W elements can form metal oxides in the 3.5 wt% NaCl solution. SEM results display that relatively severe corrosion appeared on the substrate near tungsten carbide in a 0.5 mol·L−1 HCl solution, and coating will be corroded form inside of the coating owing to the corrosive solution will penetrate the substrate. In 3.5 wt% NaCl solution, severe corrosion of the substrate has not been observed; however, the binding part has been obviously corroded.

To investigate the corrosion behavior of WC–Co composite coating, plasma transferred arc (PTA) welding was applied to prepare WC–Co coating on hull steel EH40 for achieving good metallurgical bonding. The phases of coatings were mainly composed of WC particles, solid solution γ-Co, Fe3Ni2, SiO2 and Mo2C. After electrochemical test, it can be concluded that Co-based coating with 45% WC content has better corrosion resistance in low temperature marine environment for obtaining denser oxide film. Through X-ray photo electron spectroscopy (XPS) analysis, the main corrosion products of immersion were Co(OH)2 and Co3O4. These corrosion products aggregate to form a stable corrosion product film, which plays some protective role for the coating. Hard particle WC is also partially oxidized to WO3. The pitting hole of 60% WC is the most serious. Pitting corrosion is easy to occur at the interface defects.

Ecological source areas play a pivotal role in nature preservation and soil conservation. These areas are critical in optimizing land-use patterns and fostering the establishment facilitating the establishment of protected ecological source areas. In this study, two types of state layers of ecological source area change were extracted based on land cover data and integrated with an intelligent algorithm to analyze the role of ecological factors contributing to land change. Recommendations are proposed to enhance the optimization of the ecological spatial pattern within the Yellow River Basin. The results indicate key findings: (1) The ecological source areas in the Daxia River Basin experienced Fluctuations between 1990 and 2020 due to the interconversion of grassland and unused land. The change was relatively smooth during 1990–2000, with the ecological source areas increasing slightly by 16.016 km2. From 2000 to 2010, the proportion of unutilized land transferred to grassland was 6.3%, which is significantly higher than the annual transfer average of 1.6%. This is mainly due to the ecological protection measures in the cities of Linxia and Hezuo. Between 2010 and 2020, economic construction resulted in the loss of grassland, and the percentage of grassland turned out was 7.3%, which was significantly higher than the annual average of 1.8%. (2) In the Daxia River Basin, rainfall, sunlight intensity, temperature, and elevation had a greater impact on land change during the period from 1990 to 2020, with contributions of −0.4, 0.09, 0.08, and −0.26, respectively. The transcription factor (TF)‐mediated regulatory network controlling lincomycin production in Streptomyces lincolnensis is yet to be fully elucidated despite several types of associated TFs having been reported. SLCG_2919, a tetracycline repressor (TetR)‐type regulator, was the first TF to be characterized outside the lincomycin biosynthetic cluster to directly suppress the lincomycin biosynthesis in S. lincolnensis. In this study, improved genomic systematic evolution of ligands by exponential enrichment (gSELEX), an in vitro technique, was adopted to capture additional SLCG_2919‐targeted sequences harboring the promoter regions of SLCG_6675, SLCG_4123‐4124, SLCG_6579, and SLCG_0139‐0140. The four DNA fragments were confirmed by electrophoretic mobility shift assays (EMSAs). Reverse‐transcription quantitative polymerase chain reaction (RT‐qPCR) showed that the corresponding target genes SLCG_6675 (anthranilate synthase), SLCG_0139 (LysR family transcriptional regulator), SLCG_0140 (beta‐lactamase), SLCG_6579 (cytochrome P450), SLCG_4123 (bifunctional DNA primase/polymerase), and SLCG_4124 (magnesium or magnesium‐dependent protein phosphatase) in ΔSLCGL_2919 were differentially increased by 3.3‐, 4.2‐, 3.2‐, 2.5‐, 4.6‐, and 2.2‐fold relative to those in the parental strain S. lincolnensis LCGL. Furthermore, the individual inactivation of these target genes in LCGL reduced the lincomycin yield to varying degrees. This investigation expands on the known DNA targets of SLCG_2919 to control lincomycin production and lays the foundation for improving industrial lincomycin yields via genetic engineering of this regulatory network. Lincomycin industrially produced by Streptomyces lincolnensis can be adopted to treat infections caused by Gram-positive bacteria. SLCG_Lrp, a transcriptional regulator of the Lrp family, was first identified to positively regulate lincomycin biosynthesis. However, the regulatory role of SLCG_Lrp is yet to be elucidated. This study utilized RNA-seq for comparing the transcriptome profile of original-strain LCGL and the ΔSLCGL_Lrp mutant. A total of 244 genes comprising 116 downregulated and 128 upregulated genes were differentially expressed between LCGL and ΔSLCGL_Lrp. An in-depth analysis revealed that SLCG_Lrp promotes nitrate assimilation but inhibits fatty acid metabolism, as well as directly regulates five regulators participating in the modulation of multiple cellular processes. With individual inactivation of those regulatory genes in S. lincolnensis LCGL, we confirmed the FadR transcriptional regulator SLCG_2185 was obviously correlated with lincomycin production and found it to transcriptionally stimulate the lincomycin biosynthetic cluster. Furthermore, SLCG_2185 overexpression in the high-yield S. lincolnensis LA219X promoted lincomycin production by 17.8%, and SLCG_2185 being co-overexpressed with SLCG_Lrp in LA219X increased lincomycin production by 28.1% compared to LA219X. Therefore, this investigation not only provides a direction for further investigations regarding the regulation mechanism of SLCG_Lrp, but also provides a basis for guiding the further improvement of lincomycin levels. Reliable prediction of the macromechanical properties of composite solid propellants in the microscale can accelerate the development of propellants with high mechanical properties. According to the characteristics of the composition ratio of a four-component hydroxyl-terminated polybutadiene (HTPB) propellant with the component ammonium perchlorate (AP), hydroxyl-terminated polybutadiene, aluminum powder (AL), and cyclotrimethylenetrinitramine (or RDX for short), an improved random delivery algorithm was developed to separately model filler particles with the different sizes. A step-by-step equivalent representative volume element (RVE) model was generated to reflect the microstructures of the propellant. The isotropy and uniformity of the RVE model were also tested using a two-point probability function. The Park-Paulino-Roesler (PPR) cohesive model was introduced to simulate the particle debonding (or dewetting) in solid propellant. The stress-strain curves of the propellant were obtained by the macroscopic test with the extension rate 200 mm/min at different temperatures. Based on these experimental data, the 8 characteristic parameters suitable for the microinterface of the propellant were obtained by using an inversion optimization method. A microscale finite element prediction model of the propellant considering dewetting damage was constructed to study the evolution process of the microdamage of the propellant. Tumor-associated macrophages (TAMs) account for more than 50% of the cells in the tumor immune microenvironment of patients with breast cancer. A high TAM density is associated with a poor clinical prognosis. Targeting TAMs is a promising therapeutic strategy because they promote tumor growth, development, and metastasis. In this study, we found that dimethyl formamide (DMF) significantly inhibited the tumor invasion-promoting ability of TAMs in the co-culture system and further showed that DMF functioned by reducing reactive oxygen species (ROS) production in TAMs. The orthotopic 4T1 cell inoculation model and the spontaneous mouse mammary tumor virus-polyoma middle tumor-antigen tumor model were used to evaluate the antitumor effect of DMF. The results showed that DMF significantly inhibited tumor metastasis and increased T-cell infiltration into the tumor microenvironment. Mechanistically, NRF2 activation was necessary for DMF to exert its function, and DMF can play a role in breast cancer as an anticancer drug targeting TAMs. [ABSTRACT FROM AUTHOR]

Diabetes as a chronic epidemic disease with obvious symptom of hyperglycemia is seriously affecting human health globally due to the diverse diabetic complications. Diabetic cardiovascular autonomic neuropathy (DCAN) is a common complication of both type 1 and type 2 diabetes and incurs high morbidity and mortality. However, the underlying mechanism for DCAN is unclear. It is well known that purinergic signaling is involved in the regulation of cardiovascular function. In this study, we examined whether the P2Y12 receptor could mediate DCAN-induced sympathetic reflexes. Our results revealed that the abnormal changes of blood pressure, heart rate, heart rate variability, and sympathetic nerve discharge were improved in diabetic rats treated with P2Y12 short hairpin RNA (shRNA). Meanwhile, the expression of P2Y12 receptor, interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and connexin 43 (Cx43) in stellate ganglia (SG) was decreased in P2Y12 shRNA-treated diabetic rats. In addition, knocking down the P2Y12 receptor also inhibited the activation of p38 MARK in the SG of diabetic rats. Taken together, these findings demonstrated that P2Y12 receptor in the SG may participate in developing diabetic autonomic neuropathy, suggesting that the P2Y12 receptor could be a potential therapeutic target for the treatment of DCAN. Abstract: Chronic lead exposure causes peripheral sympathetic nerve stimulation, including increased blood pressure and heart rate. Purinergic receptors are involved in the sympathoexcitatory response induced by myocardial ischemia injury. However, whether P2X4 receptor participates in sympathoexcitatory response induced by chronic lead exposure and the possible mechanisms are still unknown. The aim of this study was to explore the change of the sympathoexcitatory response induced by chronic lead exposure via the P2X4 receptor in the stellate ganglion (SG). Rats were given lead acetate through drinking water freely at doses of 0 g/L (control group), 0.5 g/L (low lead group), and 2 g/L (high lead group) for 1 year. Our results demonstrated that lead exposure caused autonomic nervous dysfunction, including blood pressure and heart rate increased and heart rate variability (HRV) decreased. Western blotting results indicated that after lead exposure, the protein expression levels in the SG of P2X4 receptor, IL‐1β and Cx43 were up‐regulated, the phosphorylation of p38 mitogen‐activated protein kinase (MAPK) was activated. Real‐time PCR results showed that the mRNA expression of P2X4 receptor in the SG was higher in lead exposure group than that in the control group. Double‐labeled immunofluorescence results showed that P2X4 receptor was co‐expressed with glutamine synthetase (GS), the marker of satellite glial cells (SGCs). These changes were positively correlated with the dose of lead exposure. Streptomyces lincolnensis is generally utilized for the production of lincomycin A (Lin-A), a clinically useful antibiotic to treat Gram-positive bacterial infections. Three methylation steps, catalyzed by three different S-adenosylmethionine (SAM)-dependent methyltransferases, are required in the biosynthesis of Lin-A, and thus highlight the significance of methyl group supply in lincomycin production. In this study, we demonstrate that externally supplemented SAM cannot be taken in by cells and therefore does not enhance Lin-A production. Furthermore, bioinformatics and in vitro enzymatic assays revealed there exist two SAM synthetase homologs, MetK1 (SLCG_1651) and MetK2 (SLCG_3830) in S. lincolnensis that could convert l-methionine into SAM in the presence of ATP. Even though we attempted to inactivate metK1 and metK2, only metK2 was deleted in S. lincolnensis LCGL, named as ΔmetK2. Following a reduction of the intracellular SAM concentration, ΔmetK2 mutant exhibited a significant decrease of Lin-A in comparison to its parental strain. Individual overexpression of metK1 or metK2 in S. lincolnensis LCGL either elevated the amount of intracellular SAM, concomitant with 15% and 22% increase in Lin-A production, respectively. qRT-PCR assays showed that overexpression of either metK1 or metK2 increased the transcription of lincomycin biosynthetic genes lmbA and lmbR, and regulatory gene lmbU, indicating SAM may also function as a transcriptional activator. When metK1 and metK2 were co-expressed, Lin-A production was increased by 27% in LCGL, while by 17% in a high-yield strain LA219X. [ABSTRACT FROM AUTHOR]

Background/Aims: Chronic diabetic hyperglycemia can damage various of organ systems and cause serious complications. Although diabetic cardiac autonomic neuropathy (DCAN) is the primary cause of death in diabetic patients, its pathogenesis remains to be fully elucidated. Baicalin is a flavonoid extracted from Scutellaria baicalensis root and has antibacterial, diuretic, anti-inflammatory, anti- metamorphotic, and antispasmodic effects. Our study explored the effects of baicalin on enhancing sympathoexcitatory response induced by DCAN via the P2Y12 receptor. Methods: A type 2 diabetes mellitus rat model was induced by a combination of diet and streptozotocin. Serum epinephrine was measured by enzyme-linked immunosorbent assay. Blood pressure and heart rate were measured using the indirect tail-cuff method. Heart rate variability was analyzed using the frequency-domain of electrocardiogram recordings. The expression levels of P2Y12, interleukin-1beta (IL-1β), tumor necrosis factor alpha (TNF-α), and connexin 43 (Cx43) were determined by quantitative real-time reverse transcription-polymerase chain reaction and western blotting. The interaction between baicalin and P2Y12 determined using by molecular docking. Results: Baicalin alleviated elevated blood pressure and heart rate, improved heart rate variability, and decreased the elevated expression levels of P2Y12, IL-1β, TNF-α, and Cx43 in the stellate ganglia of diabetic rats. Baicalin also reduced the elevated concentration of serum epinephrine and the phosphorylation of p38 mitogen-activated protein kinase in diabetic rats. Conclusion: Baicalin decreases sympathetic activity by inhibiting the P2Y12 receptor in stellate ganglia satellite glial cells to maintain the balance between sympathetic and parasympathetic nerves and relieves DCAN in the rat. Background/Aims: The induction of endothelial injury by hyperglycemia in diabetes has been widely accepted. Naringin is a bio-flavonoid. Some studies showed that naringin alleviates diabetic complications, but the exact mechanisms by which naringin improves diabetic anomalies are not yet fully understood. The aim of this research was to study the protective effect of naringin on high glucose-induced injury of human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were cultured with or without high glucose in the absence or presence of naringin for 5 days. The expression of CX3CL1 was determined by quantitative real-time RT-PCR (qPCR) and western blot. The cellular bioenergetic analysis oxygen consumption rate (OCR) was measured with a Seahorse Bioscience XF analyzer. Results: The production of reactive oxygen species (ROS), the expression of CX3CL1 and the level of AKT phosphorylation were increased in HUVECs cultured with high glucose compared with controls. However, naringin rescued these increases in ROS production, CX3CL1 expression and AKT phosphorylation. Nitric oxide (NO) production and OCR were lower in the high glucose group, and naringin restored the changes induced by high glucose. Molecular docking results suggested that Naringin might interact with the CX3CL1 protein. Conclusion: Naringin protects HUVECs from high-glucose-induced damage through its antioxidant properties by downregulating CX3CL1 and by improving mitochondrial function. [ABSTRACT FROM AUTHOR]

Hepatic glucokinase (GK) expression and activity are decreased in type 2 diabetes mellitus (T2DM), and glycogen synthase kinase-3 (GSK-3) inhibits the synthesis of GK. In hepatocytes, the activation of the protein kinase B (PKB/AKT) signaling pathway enhances GK expression and inhibits the phosphorylation of GSK-3β. The dysfunction of certain long noncoding RNAs (lncRNAs) has been associated with a variety of diseases.Aims: This study explored the effects of the lncRNA NONRATT021972 small interfering RNA (siRNA) on the dysfunction of hepatic GK through AKT signaling in T2DM rats.Methods: Livers from type 2 diabetic rats and hepatocytes cultured in high glucose and high fatty acid media were studied. The changes in expression of AKT, GK and GSK 3β were detected by western blotting or RT-PCR. The application of bioinformatics technology (CatRAPID) was used to identify the targets of NONRATT021972 RNA.Results: We found that lncRNA NONRATT021972 levels in the liver were increased in type 2 diabetic rats, and the increase was associated with an increase in the blood glucose levels. The NONRATT021972 siRNA enhanced phospho-AKT (p-AKT) levels, GK expression and hepatic glycogen synthesis. This siRNA also reduced phospho-glycogen synthase kinase-3β (p-GSK-3β) levels and hyperglycemia in T2DM rats, as well as in hepatocytes cultured in high glucose media with fatty acids.
