Soil beneficial microorganism deficiency in the degraded grasslands have emerged as the major factors negatively impacting soil quality and vegetation productivity. EM (effective microorganisms) has been regarded as a good ameliorant in improving microbial communities and restoring degraded soil of agricultural systems. However, knowledge was inadequate regarding the effects of adding EM on the degraded alpine grassland. Four levels of EM addition (0, 150, 200, 250 mL m–2) were conducted to investigate the effects of EM addition on soil properties and microorganisms of degraded alpine grassland. The addition of EM increased aboveground biomass, soil organic carbon, total nitrogen, available phosphorus, and microbial biomass, but decreased soil electric conductivity. Meanwhile, the relative biomasses of gram-negative bacteria decreased, while the ectomycorrhizal fungi and arbuscular mycorrhizal fungi increased after EM addition. The relationship between microbial communities and environmental factors has been changed. The restore effect of EM increased with the increase of addition time. These results indicated that EM addition could be a good practice to restore the health of the degraded alpine grassland ecosystem.

Subshrub encroachment is a frequently occurring phenomenon in arid and semi-arid area, altering the plant community structure and function. However, the effects of the encroachment process on soil bacterial communities are poorly understood on the Loess Plateau, China. In this study, we assessed the changes in soil properties and vegetation characteristics, and their association with alterations in soil bacterial communities in grasslands subjected to different subshrub (Artemisia sacrorum Ledeb.) encroachment intensities (no subshrub encroachment, light subshrub encroachment, moderate subshrub encroachment, and heavy subshrub encroachment). Our results indicated that heavy subshrub encroachment significantly increased plant aboveground biomass and bacterial diversity, but adversely affected plant diversity. Heavy and moderate subshrub encroachment significantly increased soil organic carbon, total nitrogen and water content. Subshrub encroachment at all intensities increased community weighted mean trait values of leaf carbon content, leaf nitrogen content, leaf phosphorus content, specific leaf area, but decreased leaf dry matter content and community functional dispersion. In addition, subshrub encroachment altered the composition of plant and bacterial communities. The partial least squares structural equation model indicated that subshrub encroachment indirectly influenced bacterial communities by affecting vegetation characteristics and soil properties. The determinants of bacterial diversity were vegetation structures (community weighted mean trait values of specific leaf area and plant community composition) and soil properties (organic carbon content, alkaline hydrolysis nitrogen, total phosphorus, available phosphorus, and soil water content). The composition of bacterial community was primarily regulated by plant aboveground biomass and soil properties. Overall, the results of this study improved our understanding of the impact of subshrub encroachment on soil bacteria community on Loess Plateau, and the processes underlying the alterations in the soil bacterial community by subshrub encroachment.

Abstract Background NAC proteins contribute to diverse plant developmental processes as well as tolerances to biotic and abiotic stresses. The pear genome had been decoded and provided the basis for the genome-wide analysis to find the evolution, duplication, gene structures and predicted functions of PpNAC transcription factors. Results A total of 185 PpNAC genes were found in pear, of which 148 were mapped on chromosomes while 37 were on unanchored scaffolds. Phylogeny split the NAC genes into 6 clades (Group1- Group6) with their sub clades (~ subgroup A to subgroup H) and each group displayed common motifs with no/minor change. The numbers of exons in each group varied from 1 to 12 with an average of 3 while 44 pairs from all groups showed their duplication events. qPCR and RNA-Seq data analyses in different pear cultivars/species revealed some predicted functions of PpNAC genes i.e. PpNACs 37, 61, 70 (2A), 53, 151(2D), 10, 92, 130 and 154 (3D) were potentially involved in bud endodormancy, PpNACs 61, 70 (2A), 172, 176 and 23 (4E) were associated with fruit pigmentations in blue light, PpNACs 127 (1E), 46 (1G) and 56 (5A) might be related to early, middle and late fruit developments respectively. Besides, all genes from subgroups 2D and 3D were found to be related with abiotic stress (cold, salt and drought) tolerances by targeting the stress responsive genes in pear. Conclusions The present genome-wide analysis provided valuable information for understanding the classification, motif and gene structure, evolution and predicted functions of NAC gene family in pear as well as in higher plants. NAC TFs play diverse and multifunctional roles in biotic and abiotic stresses, growth and development and fruit ripening and pigmentation through multiple pathways in pear.

This study aims to understand the influence of nitrogen accumulation, fungal endophyte, yield, nitrogen use efficiency, and grain nutritional quality parameters on the yield of quinoa in some areas of China. The endophytic microbial community in plants plays a crucial role in plant growth, development, and health, especially in quinoa plants under different nitrogen fertilizer levels. The results from the present study indicated that appropriate nitrogen application significantly enhanced the nitrogen accumulation and yield of quinoa grains during maturity, increasing by 34.54–42.18% and 14.59–30.71%, respectively. Concurrently, protein content, amylose, total starch, ash, and fat content also increased, with respective growth rates of 1.15–18.18%, 30.74–42.53%, 6.40–12.40%, 1.94–21.94%, and 5.32–22.22%. Our constructed interaction network of bacterial and fungal communities revealed that bacteria outnumbered fungi significantly, and most of them exhibited synergistic interactions. The moderate increase in N150 was beneficial for increasing quinoa yield, achieving nitrogen use efficiency (NUE) of over 20%. The N210 was increased, and both the yield and NUE significantly decreased. This study provides novel insights into the impact of nitrogen fertilizer on quinoa growth and microbial communities, which are crucial for achieving agricultural sustainable development.

The role of amino acid transporter (AAT) genes in facilitating the transmembrane movement of amino acids between cells and various cellular components has been characterized in several plant species. Quinoa (Chenopodium quinoa Willd.), a renowned nutritious crop known for its amino acid composition, has not yet had its AAT genes characterized. Therefore, the identification and characterization of AAT genes in quinoa will help bridge this knowledge gap and offer valuable insights into the genetic mechanisms underlying amino acid transport and metabolism. This study focuses on gene expression, gene structure, duplication events, and a comparison of functions studied to establish the role of AAT genes. A total of 160 non-redundant AAT genes were identified in quinoa and classified into 12 subfamilies, with 8 subfamilies belonging to the amino acid/auxin permease (AAAP) family and 4 to the amino acid-polyamine-organocation (APC) superfamily family. The chromosomal localization, gene structures, and conserved motifs of these genes were systematically analyzed. Expression profiling revealed diverse expression patterns across various tissues and in response to drought and salt stresses. Segmental and tandem duplications were found to contribute to the gene duplication and expansion of the CqAAT gene family. Additionally, CqCAT6 and CqAAP1 were predicted to regulate the long-distance transportation and distribution of amino acids, making them potential candidate genes for further research. Overall, this information could serve as a foundation for the identification and utilization of CqAATs in Quinoa, enhancing our understanding of amino acid transport mechanisms in this important crop.

BackgroundNatural orifice specimen extraction surgery (NOSES) has been widely applied to the treatment of colorectal cancer. This study aim to investigate the short-term and survival outcomes of transrectal specimen extraction after laparoscopic right hemicolectomy.MethodsFrom January 2016 to December 2021, a total of 166 consecutive patients with right colon cancer who underwent laparoscopic right hemicolectomy in Cancer Hospital of Chinese Academy of Medical Sciences and Beijing Hospital were identified. Baseline data, perioperative parameters, anal function, inflammatory indicators and survival outcomes were collected and compared.ResultsTotally, 24 patients who underwent transrectal NOSE were matched with 24 patients who received conventional laparoscopic surgery (LAP). Patients in NOSES group had a significantly lower incidence of incision infection (0 vs 20.8%, P=0.048), faster recovery of gastrointestinal function (2.1 vs 3,1 days, P=0.032) compared with those in LAP group. Abstract Background RNA-cleaving deoxyribozymes (DNAzymes) are catalytic deoxyribonucleic acid molecules that have become a promising new class of gene suppressors by binding and cleaving target mRNA. This study investigated whether DNAzymes targeting Bcl-xL enhanced the effectiveness of radiotherapy and chemotherapy in colorectal cancer (CRC) cells. Methods Two types of CRC cells, SW480 and SW837, were transfected with five DNAzymes. Cell viability, Bcl-xL expression and apoptosis were examined. SW480 xenograft model was used to examine the combined effects of Bcl-xL DNAzymes and 5-FU (or X-rays) on tumor growth. Results Three Bcl-xL DNAzymes, DT882, DT883, and DT884 were identified to be effective in suppressing Bcl-xL expression and causing cell apoptosis. Furthermore, DT882 combined with 5-FU or radiotherapy addictively promoted cell apoptosis and significantly inhibited the growth of SW480 xenografts in vivo. Conclusions These results suggest that Bcl-xL DNAzymes can enhance the radiosensitivity and chemosensitivity in CRC cells via inducing apoptosis.

Background: The RAD51 recombinase is involved in homologous recombination and DNA repair. However, the association of RAD51 with the prognosis of adenocarcinoma at the gastroesophageal junction (ACGEJ) is not clear. We aimed to investigate the association of RAD51 with ACGEJ prognosis. Methods: The difference in the expression level of RAD51 between ACGEJ tumors and control tissues in the microarray datasets (GSE159721, GSE74553, and GSE96669) were compared. The online Kaplan-Meier plotter survival analysis and meta-analysis were used to analyze the association of RAD51 with overall survival in pan-cancers. MiRNAs targeting RAD51 were identified and their expression profiles in ACGEJ tumors were analyzed. Functional enrichment analysis was performed for miRNAs of RAD51. Results: RAD51 was upregulated in ACGEJ tumors compared with control tissues (P < 0.05). High RAD51 level was correlated with a poor prognosis in stomach adenocarcinoma and esophageal cancer. The meta-analysis showed that high RAD51 level was correlated with a poor prognosis in TCGA pan-cancers (P = 0.03). Six regulatory miRNAs of RAD51, including hsa-miR-182, hsa-miR-221, and hsa-miR-34a, were downregulated in ACGEJ tumor tissues and were associated with pathways including “fatty acid biosynthesis” and “viral carcinogenesis”. Conclusions: RAD51 is a potent prognostic biomarker in ACGEJ. MiRNAs including hsa-miR-182, hsa-miR-221, and hsa-miR-34a might play crucial roles in ACGEJ by regulating the RAD51 gene.

Abstract This paper presents a scientific examination of wall paintings at two nonmonastic sites in Gyantse, Tibet: Gazhi Lhakang, which is a family temple built in the mid-eighteenth century by local aristocrats, and the Lotso Residence, which was occupied by Nepalese merchants in the early twentieth century. Samples were analyzed with optical microscopy, scanning electron microscopy with energy-dispersive X-ray spectroscopy, Raman spectroscopy, and X-ray diffraction. Two phases of painting—an early phase and a late phase—were identified in Gazhi Lhakang, including three distinct types of mural stratigraphy. The earlier phase features an unusual technique known as “paperhanging”, wherein the pigments were applied on a layer of Tibetan paper glued to the wall. The later phase at Gazhi Lhakang and the painting of the Lotso Residence feature a relatively simple wall treatment with fewer coating layers and no ground layer. A typical mixture of clay and sand was used for the coating layers, while the structure slightly varied from what has been described in the literature. The techniques of powder embossing, gilding, and gold outlining were adopted in both buildings. The metallic material found at Gazhi Lhakang is a gold-silver alloy, while copper was used as imitation gold at the Lotso Residence. Mineral pigments, such as azurite, malachite, orpiment, cinnabar, and iron oxide, were used for both phases of Gazhi Lhakang. Modern synthetic pigments, such as chrome yellow, emerald green, and synthetic ultramarine, were used for the Lotso Residence, indicating that it was painted after the mid-nineteenth century.

Through the analysis of the recovery inrush current generated by the external fault removal of the converter transformer, it is pointed out that the zero-sequence current caused by the recovery inrush may result in the saturation of the neutral current transformer (CT), whose measurement distortion contributes to the mis-operation of zero-sequence differential current protection. In this paper, a new scheme of zero-sequence differential current protection based on waveform correlation is proposed. By analyzing the characteristics of zero-sequence current under internal fault, external fault and external fault removal, the waveform correlation of the zero-sequence current measured at the terminal of the transformer and the zero-sequence current measured at the neutral point of the transformer is used for identification. The polarity of the CT is selected to guarantee the zero-sequence currents at the terminal and neutral point of the transformer exhibit a "ride through" characteristic under external fault, then the waveform similarity is high, and the correlation coefficient is positive. On the other hand, when internal fault occurs, zero-sequence current waveforms on both sides differ from each other largely, and the correlation coefficient is negative. Through a large number of simulations verified by PSCAD/EMTDC, this criterion can accurately identify internal and external faults, exempt from effects of the recovery inrush. Moreover, it presents certain ability for CT anti-saturation.
