2. An upstream platform for the production of high grade heterologous proteins in the yeast Pichia pastoris

Abstract

Pichia pastoris is a methylotrophic yeast and well established expression system for the production of therapeutic proteins and industrial enzymes. It is characterised by its tightly regulated promoters, natural ability to secrete recombinant proteins and potential to grow to very high cell densities in order to combat low product titres. These high cell densities, in combination with the requirement of methanol as an inducing agent, lead to problems, however, with cell viability being negatively influenced, resulting in the release of host cell proteins, which include proteases that both reduce product quality and complicate purification. This thesis introduces a simple fermentation strategy that reduces the amount of methanol required during fermentation by utilising the non-repressing substrate sorbitol as a co-feed with methanol during induction. This allowed cell growth profiles to remain similar to those seen with current protocols, with final cell densities of 120-140 g/L dry cell weight (DCW) being attained. Cell viability and product yields were unaffected by the new strategy, but protease release was reduced due to a shift in host cell protein impurity profiles. The scalability of fermentations was also greatly increased due to a 60% reduction in heat generation during induction. Finally, a methodology for the determination of cellular robustness was successfully developed using adaptive focussed acoustics: this was used to demonstrate that methanol induction did not negatively impact cellular robustness, and that reduced growth rates were a key requirement of enhanced cellular robustness. These results have all been demonstrated with two strains of P. pastoris, an in-house GS115 strain expressing secreted embryonic alkaline phosphatase (SEAP) and an industrially utilised strain, CLD804 expressing aprotinin.

24. Statelessness and applications for leave to remain: a best practice guide

Abstract

The UK Immigration Rules were amended in April 2013 to include a new category of leave: ‘Part 14 stateless persons’. Statelessness arises when a person is not a national of a state. The guide focuses upon the definition applied by the UK Immigration Rules: ‘a person who is not considered as a national by any State under the operation of its law’. This definition is taken from the 1954 Convention relating to the Status of Stateless Persons (the 1954 Convention). An important feature of the Immigration Rules relating to stateless persons is the scope they provide to secure recognition by the Home Office of statelessness, a legal condition which itself has important consequences in international law. The guide sets out the view of the authors on best practice in advising and representing clients considering or making an application for statelessness leave in the UK. The UK law in this area is in its infancy and is likely to develop particularly quickly. Some key questions of law and practice have yet to be addressed by the courts; the guide provides a snapshot of the state of play as at September 2016. It is aimed at those who may represent clients considering whether and when to make an application for leave to remain as a stateless person. It may also be of use to organisations working with potentially stateless persons who may refer them to legal advisers. It is a joint publication of the Immigration Law Practitioners' Association and the University of Liverpool. The authors are in-house solicitors in the Law Department's Law Clinic.

Abstract

The provisions of the Legal Aid, Sentencing and Punishment of Offenders Act 2012 (LASPO 2012) will remove almost all immigration cases from the scope of legal aid with effect from 1 April 2013. Part One of this paper describes the expected changes LASPO 2012 will make and then explores their anticipated impact. It explains that large numbers of migrants and their family members are likely to be without legal advice and representation after April 2013, including in cases where the state-enforced removal or deportation from the UK of a parent is contemplated, with the consequence of separating parent from child. Part Two explores the possibility that an ‘exceptional case determination’ might provide a route back into legal aid funding, and finds that this is likely to be restricted, in immigration cases, to those making applications relying on Article 8 ECHR and who can demonstrate a particular and individual requirement for legal aid. Part Three explores the rationale for these changes, and concerns about access to justice for migrants and their family members in cases involving acute interference with rights to family and private life. It places these concerns in context, specifically the fundamental and restrictive amendments to the Immigration Rules relating to family migration introduced from July 2012. These amendments are enormously complex and their full legal implications have yet to be tested in the higher courts. Part Four questions whether the changes will in fact achieve their stated aim of cost savings or whether the costs will simply be transferred to other parts of the State (especially to the Tribunal system, in dealing with litigants in person). The paper additionally questions, in Part Five, whether the regulators are equipped to regulate the quality of the fee-charging immigration advice services to which at least some individuals will turn. The paper concludes that, at the very least, it is particularly harsh that the Government has removed the ‘currency’ of legal aid at this time, so that those with limited financial resources have neither access to legal aid advice about the meaning of those Rules nor legal aid representation to test their proper interpretation. Abstract

Clonidine is an adrenergic agonist with high affinity for alpha2 adrenoceptors that also has affinity for imidazoline receptors. Clonidine has previously been shown to reduce immobility in the forced swim test (FST) in mice. In the present study, this effect was blocked by idazoxan (0.06 mg/kg s.c.) and by yohimbine (1.0 mg/kg s.c.) suggesting that clonidine's effects in this test are mediated via its action at alpha2 sites. Imidazoline I2 site ligands have been shown to inhibit monoamine oxidase and thus may also have antidepressant activity. Three compounds with selective affinity for I2 receptors (BU224, BU239, BDF 8082) were also tested in the FST. These compounds showed no activity either alone or in combination with a subthreshold dose of imipramine in the FST. These results suggest that I2 receptor ligands do not show antidepressant-like activity in the FST in mice. Furthermore the activity of the mixed alpha2/I1 agonist clonidine is most likely to be due to its action at alpha2 sites. [ABSTRACT FROM AUTHOR]

42. THE RELEVANCE OF CHONDROITIN AND KERATAN SULPHATE MARKERS IN NORMAL AND ARTHRITIC SYNOVIAL FLUID.

Abstract

This study investigated the synovial fluid concentrations of glycosaminoglycan (GAG), keratan sulphate (KS) epitope 5D4 and chondroitin sulphate (CS) sulphation patterns in healthy volunteers and patients with osteoarthritis (OA) and rheumatoid arthritis (RA). Synovial fluids were collected from knee joints of healthy volunteers (n = 24), and patients with OA (n = 28) and RA (n = 29). Concentrations of GAG and the keratan sulphate epitope SEW were measured in 15 of the healthy volunteers, and all of the OA and RA synovial fluids. Total GAG was measured using a dye-binding method and 5D4 by an ELISA. The unsaturated CS disaccharides ΔC4 and ΔC6 were measured by capillary electrophoresis in all synovial fluids. The concentrations of GAG, 5D4 and ΔC6 in the normal synovial fluid were higher but that of ΔC4 lower than those of the disease groups. The ΔC6:ΔC4 ratios correlated with age (r = − 0.437, P < 0.001) and the mean value was lower in females than males (2.92 compared with 5.22, P < 0.001). After allowing for age and sex, the ΔC6:ΔC4 ratio in the control group was significantly elevated (P < 0.001) compared to both OA and RA. The ratio was also related to proteoglycan markers (r = 0.383 for 5D4 and r = 0.357 for GAG). The finding that 5D4 and ΔC6:ΔC4 ratios are higher in synovial fluid from healthy volunteers compared to OA and RA suggests that they may be markers of the susceptibility of articular cartilage to early damage in arthritis.
