25 páginas, 6 figuras, 2 tablas, Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele., This work was funded by a grant (EADB) from the EU Joint Programme – Neurodegenerative Disease Research. Log jams alter gradients in hydraulic head, increase the area available for hyporheic exchange by creating backwater areas, and lead to the formation of multiple channel branches and bars that drive additional exchange. Here, we numerically simulated stream‐groundwater interactions for two constructed flume systems—one without jams and one with a series of three jams—to quantify the effects of interacting jam structures and channel branches on hyporheic exchange at three stream flow rates. In simulations without jams, average hyporheic exchange rates ranged from 2.1 × 10−4 to 2.9 × 10−4 m/s for various stream discharge scenarios, but with jams, exchange rates increased to a range of 1.3 × 10−3–3.5 × 10−3 m/s. Largely due to these increased hyporheic exchange rates, jams increased stream‐groundwater connectivity or decreased the turnover length that stream water travels before interacting with the hyporheic zone, by an order of magnitude, and drove long flow paths that connected multiple jams and channel threads. Decreased turnover lengths corresponded with greater reaction significance per km, a measure of the potential for the hyporheic zone to influence stream water chemistry. For low‐flow conditions, log jams increased reaction significance per km five‐fold, from 0.07 to 0.35. Jams with larger volumes led to longer hyporheic residence times and path lengths that exhibited multiple scales of exchange. Additionally, the longest flow paths connecting multiple jams occurred in the reach with multiple channel branches. These findings suggest that large gains in hydrologic connectivity can be achieved by promoting in‐stream wood accumulation and the natural formation of both jams and branching channels. Plain Language Summary: Log jams widen streams, create pools, and cause the stream to branch and follow new paths. These processes affect surface water‐groundwater interaction or the exchange between the stream and the water moving within the sediments and streambanks. In this work, we used three‐dimensional computer models of an experimental laboratory flume system to understand the effects of multiple log jams and channel branches on this surface water‐groundwater interaction. We found that when log jams are present in streams, the rate of water moving between the surface and subsurface is greater than it would be otherwise and increases under flood conditions. Log jams also increase chemical contact with the streambed, which can lead to natural improvements in stream water quality. We also found that where log jams are bigger and located near branching channels, stream water that enters the bed follows diverse pathways that span a wide range of travel distances and times before returning to the stream. Essentially, in‐stream wood and channel branching increases water flow in streambed sediments and increases the potential for chemical processing to occur. Objective: To determine the levels of antithyroid antibodies and thyroid hormones in the sera of patients with oral lichen planus (OLP), and to quantify the expression of thyroid proteins in OLP lesions. Subjects and Methods: Venous blood samples were drawn from 110 patients with OLP who had no history of thyroid disease or levothyroxine supplementation (OLP+/LT4−). A random population sample of 657 healthy subjects was used as the control group. Two additional groups were used as comparators. Immunohistochemical and qPCR analyses were performed on tissue specimens collected from the patients with OLP and thyroid disease and healthy subjects. Results: No association was found between the presence of antithyroid antibodies and OLP. More patients in the OLP+/LT4− group showed high levels of thyroid‐stimulating hormone and low levels of free thyroxine than were seen in the control group. Thyroid‐stimulating hormone receptor was more highly expressed in the OLP lesions of patients with thyroid disease than in the healthy oral mucosa. Conclusions: A significant number of patients with OLP who are not previously diagnosed with thyroid disease have thyroid parameters that are compatible with hypothyroidism. The expression of thyroid‐stimulating hormone receptor in OLP lesions suggests that mechanisms related to autoimmune thyroid disease are involved in the aetiology of OLP. [ABSTRACT FROM AUTHOR]

Pharmacogenomics is yet to fulfill its promise of manifestly altering clinical medicine. As one example, a predictive test for tardive dyskinesia (an adverse drug reaction consequent to antipsychotic exposure) could greatly improve the clinical treatment of schizophrenia but human studies are equivocal. A complementary approach is the mouse-then-human design in which a valid mouse model is used to identify susceptibility loci which are subsequently tested in human samples. We used inbred mouse strains from the Mouse Phenome Project to estimate the heritability of haloperidol-induced activity and orofacial phenotypes. 159 mice from 27 inbred strains were chronically treated with haloperidol (3 mg/kg/day via subdermal slow-release pellets) and monitored for the development of vacuous chewing movements (VCMs, the mouse analog of TD) and other movement phenotypes derived from open field activity and the inclined screen test. The test battery was assessed at 0, 30, 60, 90, and 120 days in relation to haloperidol exposure. As expected, haloperidol caused marked changes in VCMs, activity in the open field, and EPS. Unexpectedly, factor analysis demonstrated that these measures were imprecise assessments of a latent construct rather than discrete constructs. The heritability of a composite phenotype was ~0.9 after incorporation of the longitudinal nature of the design. Murine VCMs are a face valid animal model of antipsychotic-induced TD and heritability estimates from this study support the feasibility of mapping of susceptibility loci for VCMs.

Nicotine dependence has been shown to represent a heritable condition, and several research groups have performed linkage analysis to identify genomic regions influencing this disorder though only a limited number of the findings have been replicated.In the present study, a genome-wide linkage scan for nicotine dependence was conducted in a community sample of 950 probands and 1204 relatives recruited through the University of California, San Francisco (UCSF) Family Alcoholism Study. A modified version of the Semi-Structured Assessment for the Genetics of Alcoholism (SSAGA) with additional questions that probe nicotine use was used to derive DSM-IV nicotine dependence diagnoses.A locus on chromosome 2q31.1 at 184 centiMorgans nearest to marker D2S2188 yielded a logarithm (base 10) of odds (LOD) score of 3.54 (point-wise empirical p=0.000012). Additional peaks of interest were identified on chromosomes 2q13, 4p15.33-31, 11q25 and 12p11.23-21. Follow-up analyses were conducted examining the contributions of individual nicotine dependence symptoms to the chromosome 2q31.1 linkage peak as well as examining the relationship of this chromosomal region to alcohol dependence.The present report suggests that chromosome 2q31.1 confers risk to the development of nicotine dependence and that this region influences a broad range of nicotine dependence symptoms rather than a specific facet of the disorder. Further, the results show that this region is not linked to alcohol dependence in this population, and thus may influence nicotine dependence specifically.

The beta decays of N-12 and B-12 have been studied at KVI and JYFL to resolve the composition of the broad and interfering 0(+) and 2(+) strengths in the triple-alpha continuum. For the first time a complete treatment of 3 alpha decay is presented including all major breakup channels. A multilevel, many-channel R-matrix formalism has been developed for the complete description of the breakup in combination with the recently published separate analysis of angular correlations. We find that, in addition to the Hoyle state at 7.65 MeV, more than one 0(+) and 2(+) state is needed to reproduce the spectra. Broad 0(3)(+) and 2(2)(+) states are found between 10.5 and 12 MeV in this work. The presence of beta strength up to the N-12 Q-value window suggests the presence of additional 0(+) and 2(+) components in the C-12 structure at energies above 12.7 MeV.

Absolute branching ratios to unbound states in C-12 populated in the beta decays of N-12 and B-12 are reported. Clean sources of N-12 and B-12 were obtained using the isotope separation on-line (ISOL) method. The relative branching ratios to the different populated states were extracted using single-alpha as well as complete kinematics triple-alpha spectra. These two largely independent methods give consistent results. Absolute normalization is achieved via the precisely known absolute branching ratio to the bound 4.44 MeV state in C-12. The extracted branching ratios to the unbound states are a factor of three more precise than previous measurements. Branching ratios in the decay of Na-20 are also extracted and used to check the results.

The triple-alpha-particle breakup of states in the triple-alpha continuum of C-12 has been investigated by way of coincident detection of all three alpha particles of the breakup. The states have been fed in the beta decay of N-12 and B-12, and the alpha particles measured using a setup that covers all of the triple-alpha phase space. Contributions from the breakup through the Be-8(0(+)) ground state as well as other channels-interpreted as breakup through excited energies in Be-8-have been identified. Spins and parities of C-12 triple-alpha continuum states are deduced from the measured phase-space distributions for breakup through Be-8 above the ground state by comparison to a fully symmetrized sequential R-matrix description of the breakup. At around 10 MeV in C-12, the breakup is found to be dominated by 0(+) strength breaking up through the ghost of the Be-8(0(+)) ground state with L = 0 angular momentum between the first emitted alpha particle and the intermediate Be-8 nucleus. For C-12 energies above the 12.7 MeV 1(+) state, however, L = 2 breakup of a C-12 2(+) state through the Be-8(2(+)) excited state dominates. Furthermore, the possibility of a 2(+) excited state in the 9-12 MeV region of C-12 is investigated.

Two complementary experimental techniques have been used to extract precise branching ratios to unbound states in C-12 from N-12 and B-12 beta-decays. In the first the three alpha-particles emitted after decay are measured in coincidence in separate detectors, while in the second method N-12 and B-12 are implanted in a detector and the summed energy of the three a-particles is measured directly. For the narrow states at 7.654 MeV (0(+)) and 12.71 MeV (1(+)) the resulting branching ratios are both smaller than previous measurements by a factor of similar or equal to 2. The experimental results are compared to no-core shell model calculations with realistic interactions from chiral perturbation theory, and inclusion of three-nucleon forces is found to give improved agreement. The beta-delayed charged particle emission from 11Li has been studied with emphasis on the three-body n+alpha+6He and five-body 2alpha+3n channels from the 10.59 and 18.15 MeV states in 11Be. Monte Carlo simulations using an R-matrix formalism lead to the conclusion that the AHe resonance states play a significant role in the break-up of these states. The results exclude an earlier assumption of a phase-space description of the break-up process of the 18.15 MeV state. Evidence for extra sequential decay paths is found for both states.
