Abstract Background Recent studies have identified the presence of cancer-associated fibroblasts (CAFs) within glioblastoma (GBM), yet their biological roles and underlying mechanisms remain poorly understood. This study aimed to construct a CAF-related prognostic model to guide patient prognosis and treatment strategies. Method We employed various bioinformatics methods, including enrichment analysis, Weighted Gene Co-expression Network Analysis (WGCNA), Lasso regression analysis, and machine learning techniques such as XGBoost and Random Forest, to develop a novel risk index termed CAFscore. Patients were stratified into high and low CAFscore groups for subsequent survival analysis. The area under the curve (AUC) and concordance index (C-index) for CAFscore were calculated and compared against other clinical characteristics and existing prognostic models. Drug sensitivity assessments were conducted using the Oncopredict package. Functional validation of key genes was performed through scratch and invasion assays in GBM cells. Results Our analyses revealed four core CAF-related genes, leading to the establishment of CAFscore. Notably, patients in the high CAFscore group exhibited significantly reduced survival and exhibited enrichment in epithelial-mesenchymal transition (EMT) and inflammation response pathways. Furthermore, CAFscore showed a significant negative correlation with the sensitivity to irinotecan and its analogs, while demonstrating a positive correlation with sensitivity to 505,124 (a TGFβRI inhibitor). LRP10 emerged as a central gene within the CAFscore, displaying markedly elevated expression in GBM and a strong association with CAF infiltration. Silencing LRP10 significantly inhibited the invasive capabilities of GBM cells. Conclusion This study presented the first CAF related prognostic model (CAFscore) in GBM, and demonstrated that the model could effectively guide patient prognosis and potentially inform personalized treatment strategies. The core gene of CAFscore, LRP10, was significantly overexpressed in GBM and might play a pivotal role in regulating CAF infiltration as well as tumor invasion and metastasis, highlighting LRP10 as a promising therapeutic target for GBM management.

Abstract The coupled green energy and chemical production by photocatalysis represents a promising sustainable pathway, which poses great challenges for the multifunction integration of catalytic systems. Here we show a promising green photocatalyst design using Cu-ZnIn2S4 nanosheets and carbon dots as building units, which enables the integration of reaction, mass transfer, and separation functions in the nano-space, mimicking a nanoreactor. This function integration results in great activity promotion for benzyl alcohol oxidation coupled H2 production, with H2/benzaldehyde production rates of 45.95/46.47 mmol g−1 h−1, 36.87 and 36.73 times to pure ZnIn2S4, respectively, owning to the enhanced charge accumulation and mass transfer according to in-situ spectroscopies and computational simulations of the built-in electrical field. Near-unity selectivity of benzaldehyde is achieved via the effective separation enabled by the Cu(II)-mediated conformation flipping of the intermediates and subsequent π-π conjugation. This work demonstrates an inspiring proof-of-concept nanoreactor design of photocatalysts for coupled sustainable systems.

To study the influence of an elbow inlet on the rotating stall characteristics of a waterjet propulsion pump (WJPP), a three-dimensional internal flow field in a WJPP under a straight-pipe inlet and elbow inlet is numerically simulated. By comparing the hydraulic performance of WJPP under the two inlet conditions, the internal relationship between the inlet mode and the flow pattern in the pump is clarified. Based on unsteady pressure fluctuation characteristics and wavelet analysis, the influence of the inlet mode on the rotating stall is revealed, and the stall transient propagation characteristics under critical stall conditions are analyzed. The disturbance effects of the inlet channel geometry disappear under low flow rate conditions, the main disturbance is induced by the high-speed countercurrent, and the flow pattern under the elbow inlet is better than that under the straight-pipe inlet. Under the straight-pipe inlet, the single-stall nucleus in the WJPP temporarily experiences a low-frequency and high-amplitude disturbance, which subsequently transforms into a mode of multi-stall nuclei with high-frequency circumferential disturbance. Under the elbow inlet, the rotating stall always maintains a mode of high-amplitude and low-frequency disturbance, which represents the transient characteristics of a single stall core propagating in the circumferential direction inside the channel. The results of this study have a reference value for structural design optimization in a WJPP.

To investigate the erosion mechanism and model of ultra-high pressure (200 MPa) water jet on grade–A marine steel with different curvatures. The ultra-high pressure water jet was induced by plunger pump. The dynamic strain was collected by dynamic strain gauge during impact to explore the mechanical effect. The microstructure was analyzed using three-dimensional topography and a scanning electron microscope. The results show that the average microstrains of samples with the curvature of 1/4 m−1, 3/4 m−1, and 5/4 m−1 impacted by ultra-high pressure water jet are 707, 952 and 1053, respectively, and the strain amplitudes are 300∼700, 300–600 and 300–560 respectively. The force exerted on the surface of the curved samples manifested as cyclic, pulsating, and alternating stress. With the curvature increase, the roughness decreases by 19.22%, 48.64%, and 21.31%, respectively. Meanwhile, surface damage, material removal, pits, holes, and cracks decrease. Finally, the erosion mechanism is clarified, and the erosion model of the ultra-high pressure water jet on grade–A marine steel with different curvatures is established.}

Photoelectrochemical NO3− reduction (PEC NITRR) not only provides a promising solution for promoting the global nitrogen cycle, but also converts NO3− to the important chemicals (NH3). However, it is still a great challenge to prepare catalysts with excellent NO3− adsorption/activation capacity to achieve high NITRR. Herein, we designed a novel Fe2+Cu2+Fe3+LDH/BiVO4 (FCF-LDH/BVO) catalyst with synergistic effect of chemical adsorption and physical enrichment. Fe2+ in FCF-LDH/BVO provides the rich Lewis acid sites for the adsorption of NO3−, and the appropriate layer spacing of FCF-LDH further promotes the physical enrichment of NO3− in its interior, thus realizing the effective contact between NO3− and active sites (Fe2+). FCF-LDH/BVO showed excellent NH3 production performance (FENH3 = 66.1%, rNH3 = 13.8 μg h−1 cm−2) and selectivity (FENO2- = 2.5%, rNO2- = 4.9 μg h−1 cm−2) in 0.5 mol L−1 Na2SO4 electrolyte. In addition, FCF-LDH/BVO maintains the desirable PEC stability for six cycle experiments, showing great potential for practical application. The 14NO3− and 15NO3− isotope test provides strong evidence for further verification of the origin of N in the generated NH3. This LDH catalyst has a great potential in PEC removal of NO3− from groundwater.

Abstract Owing to the specific electronic-redistribution and spatial proximity, diatomic catalysts (DACs) have been identified as principal interest for efficient photoconversion of CO2 into C2H4. However, the predominant bottom-up strategy for DACs synthesis has critically constrained the development of highly ordered DACs due to the random distribution of heteronuclear atoms, which hinders the optimization of catalytic performance and the exploration of actual reaction mechanism. Here, an up-bottom ion-cutting architecture is proposed to fabricate the well-defined DACs, and the superior spatial proximity of CuAu diatomics (DAs) decorated TiO2 (CuAu-DAs-TiO2) is successfully constructed due to the compact heteroatomic spacing (2-3 Å). Owing to the profoundly low C-C coupling energy barrier of CuAu-DAs-TiO2, a considerable C2H4 production with superior sustainability is achieved. Our discovery inspires a novel up-bottom strategy for the fabrication of well-defined DACs to motivate optimization of catalytic performance and distinct deduction of heteroatom synergistically catalytic mechanism.

Sewage pumps are widely utilized in various applications, including municipal wastewater treatment plants, industrial processes, and residential sewage systems. These pumps are specifically designed to handle the transportation of mixtures consisting of solid and liquid components, commonly observed as two-phase flow. In this study, semi-open sewage pumps with varying clearance sizes have been examined through adjustment of the geometrical model. Multi-factor analysis of variance is employed to investigate the combined influence of different two-phase flow conditions and tip clearance sizes on the performance of sewage pumps. The results show that the performance of the pump decreases significantly as the clearance size increases. Also, the sensitivity of pump efficiency to variations in tip clearance size is contingent upon the flow rate. Under part-load conditions, both particle concentration and clearance size exhibit a notable effect on efficiency. The two-phase flow will greatly enhance the intensity of the unsteady flow near the inlet and outlet of the semi-open impeller, which is the main reason for the performance degradation of the sewage pump. The findings of this research provide valuable insights for enhancing the hydraulic performance and operational stability of semi-open sewage pumps operating under two-phase flow conditions.

Highlights Recent advances in electrochemical energy storage based on nano- and micro-structured (NMS) scaffolds are summarized and discussed. The fundamentals, superiorities, and design principle of NMS scaffolds are outlined. Given the present progress, the ongoing challenges and promising perspectives are highlighted.

The presence of sparsity in both input features and weights within convolutional neural networks offers a valuable opportunity to significantly reduce the number of computations required during inference. Moreover, the practice of compressing input data serves to diminish storage requirements and lower data transfer costs, ultimately enhancing overall power efficiency. However, the compression of randomly sparse inputs introduces challenges in the input matching process, often resulting in substantial hardware overhead and increased power consumption. These challenges arise due to the irregular nature of sparse inputs and the variability in convolutional strides. In response to these challenges, this research introduces an innovative data compression method, named Stride-Aware Sparsity Compression (StarSPA), designed to effectively locate valid input values and expedite the multiplication process. To fully capitalize on this proposed compression method, a weight-stationary approach is employed for efficient convolution. Comprehensive simulations demonstrate that the proposed accelerator achieves speedup factors of $1.17\times $ , $1.05\times $ , $1.09\times $ , $1.23\times $ , and $1.12\times $ when compared to the recent accelerator named SparTen for AlexNet, VGG16, GoogLeNet, ResNet34, and EfficientNetV2, respectively. Furthermore, FPGA implementation of the core reveals a noteworthy $2.55\times $ reduction in hardware size and a $5\times $ enhancement in energy efficiency when compared to SparTen.

BackgroundEnhanced recovery after surgery (ERAS) has been widely used in adult surgery. However, few studies have reported the efficacy of ERAS in paediatric patients with Meckel's diverticulum (MD), the aim of the study was to prospectively evaluate the safety and efficacy of ERAS in treating MD.MethodsA prospective randomised controlled study of children with MD admitted to our hospital from Jan 1, 2021 to Dec 31, 2023 were conducted, we developed and implemented an ERAS program for this patients. All cases were strictly selected according to the inclusion and exclusion criteria. Among these patients, they were randomly assigned to the ERAS group or the traditional (TRAD) group with random number table row randomization. The main observational indicators were operation time, intraoperative hemorrhage, FLACC pain scale results on 2 h, 6 h, 12 h, 24 h after surgery, length of postoperative stay (LOPS), time to first defecation, time to first eating after surgery, time to discontinuation of intravenous infusion, total treatment cost, incidence of postoperative complications, 30-day readmission rate and parental satisfaction rate.ResultsA total of 50 patients underwent Meckel's diverticulectomy during this period, 7 patients were excluded, 23 patients were assigned to the ERAS group and 20 patients were assigned to the TRAD group. There were no significant differences in demographic data and operation time, intraoperative hemorrhage. The FLACC pain scale results on 2 h, 6 h, 12 h, 24 h after surgery were significantly lower in the ERAS group. The LOPS was 6.17 ± 0.89 days in the ERAS group and 8.30 ± 1.26 days in the TRAD group, resulting in a significantly shorter LOPS in ERAS group. ERAS could also reduce the first postoperative defecation time, the time to first eating after surgery and the time to discontinuation of intravenous infusion. The treatment cost was decreased in the ERAS group. The rate of complications and 30-day readmission were not significantly different between the two groups.ConclusionsIn this single-center study, the ERAS protocol for patients with MD requiring surgery was safe and effective.

As a carbon capture and utilization (CCU) technology, gas diffusion electrode (GDE) based electrochemical CO2 reduction reaction (eCO2RR) can convert CO2 to valuable products, such as formate and CO. However, the electrode parameters and operational conditions need to be studied and optimised to enhance the performance and reduce the net cost of the eCO2RR process before its industrial application. In this work, a machine learning algorithm, i.e., extended adaptive hybrid functions (E-AHF) is combined with a multi-physics model for the data-driven three-objective optimisation and techno-economic analysis of the GDE-based eCO2RR process. The effects of eight design variables on the product yield (PY), CO2 conversion (CR) and specific electrical energy consumption (SEEC) of the process are analysed. The results show that the R2 of the E-AHF model for the prediction of PY, CR and SEEC are all higher than 0.96, indicating the high accuracy of the developed machine learning algorithm for the prediction of the eCO2RR process. The process performance experiences a notable improvement after optimisation and is affected by a combination of eight variables, amongst which the electrolyte concentration having the most significant impact on PY and CR. The optimal trade-off single-pass PY, CR and SEEC are 3.25×10−9 kg s−1, 0.663% and 9.95 kWh kg−1 based on flow channels with 1 cm in length, respectively. The SEEC is reduced by nearly half and PY and CR are improved more than two times after optimisation. The production cost of the GDE-based eCO2RR process was approximately $378 t−1product (CO and formate), much lower than that of traditional CO2 utilisation factories ($835 t−1product). The electricity cost accounted for more than 80% of the total cost, amounting to $318 t−1, indicating that cheaper and cleaner electricity sources would further reduce the production cost of the process, which is the key to the economics of this technology.

Researchers have started to recognize the necessity for a well-defined ML governance framework based on the principle of decentralization and comprehensively defining its scope of research and practice due to the growth of machine learning (ML) research and applications in the real world and the success of blockchain-based technology. In this paper, we study decentralized ML governance, which includes ML value chain management, decentralized identity for the ML community, decentralized ownership and rights management of ML assets, community-based decision-making for the ML process, decentralized ML finance, and risk management.

It is important to reduce carbon emissions caused by the energy consumption of pumps. This study used a centrifugal pump with a specific speed of 89.6 as the research object to improve pump efficiency. The adaptive single-objective method was adopted as the automatic optimization tool with computational fluid dynamics, which includes optimal space-filling experimental design, Kriging response surface, and mixed-integer sequential quadratic programming. Eight geometric parameters from the meridian section and the plan of the impeller and diffuser were chosen as the design variables. The maximum efficiency under the design flow conditions was set as the optimization target. The Spearman correlation coefficient analysis results show that the sensitivity of each variable to efficiency and head. Compared with the original scheme, the optimal scheme showed a 2.75% increase in efficiency and a 1.17 m increase in head under the design flowrate. The internal flow field after optimization was also improved. An external characteristic experiment of the original and optimized pumps was performed to validate the numerical results. This automatic optimization method presents great potential to improve the hydraulic performance of centrifugal pumps at a lower cost.

Fluidized bed is the main reactor for the heating and gasification of coal, biomass, and other solid fuels. The diameter, density, and shape of the particles significantly affects the gas–solid​ two-phase flow process. Based on the numerical method combining computational fluid dynamics and discrete element method (CFD–DEM), this paper mainly performs numerical simulation on the gas–solid two-phase flow process of coarse particles under different inlet mass flow rates with diameters of 2.5 mm, 3.0 mm, and 3.5 mm, respectively. The high-speed photographic experiments were carried out to validate the numerical results successfully. The effects of particle diameter on bubble morphology have been compared, such as bed height and initial bed mass change rate. The maximum value of the bed height reached gradually decreases as the particle diameter increases. Two typical C-shaped bubbles were found above the large bubbles during the fluidization processes, which further improve the uniformity of mixing. Slugging appears as the width of the bubbles in the fluidized bed is close to the width of the fluidized bed, which has great effects on the mass and heat transfer. This study could serve as reference for further optimal design of fluidized bed, especially for biomass or chemical looping combustion application with coarse particles.

Abstract In the information explosion society, information security is highly demanded in the practical application, which raised a surge of interest in designing secure and reliable information transmission channels based on the inherent properties of emerging devices. Here, an innovative strategy to achieve the data encryption and reading during the data confidential transmission based on VO2 device is proposed. Owing to the specific insulator‐to‐metal transition property of VO2, the phase transitions between the insulator and metallic states are modulated by the combination of electric field, temperature, and light radiation. These external stimulus‐induced phase diagram is directly associated with the defined VO2 device, which are applicable for control the “0” or “1” electrical logic state for the information encryption. A prototype device is fabricated on an epitaxial VO2 film, which displayed a unique data encryption function with excellent stability. The current study not only demonstrated a multiphysical field‐modulated VO2 device for information encryption, but also supplied some clues for functional devices applications in other correlated oxide materials.

In this study, the performance of submersible mixers in sewage treatment was improved by optimizing the installation position parameters of the mixer. The aim was to enhance the average flow velocity and mixing efficiency in the pool. The study employed ISIGHT software, integrated with Creo Parametric 6.0 software and ANSYS Workbench 2020 software, to analyze the factors affecting mean flow velocity and completed a multi-objective optimized design using Non-dominated Sorting Genetic Algorithm (NSGA-II). The study used the ISIGHT software to analyze the factors affecting mean flow velocity in the pool. The installation position parameters of the submersible mixer were selected as design variables. The study employed Creo Parametric 6.0 software to create a three-dimensional model of the pool and the submersible mixer. ANSYS Workbench 2020 software was used to simulate fluid flow in the pool. The Non-dominated Sorting Genetic Algorithm (NSGA-II) was used for multi-objective optimization. The results of the study indicated an increase of approximately 0.021 m/s in average flow velocity and an improvement of approximately 0.47% in mixing efficiency compared to pre-optimization values. The effective axial propulsion distance and effective radial diffusion radius were significantly increased by 6.71% and 8.33%, respectively, after optimization. The fluid distribution in the pool became more uniform, and the low-speed zone was greatly reduced, resulting in an enhanced flow state of the fluid in the pool and a strengthened mixing effect. The study provides insights into the control of the submersible mixer’s installation position to improve the average flow velocity inside the pool. Automatic optimization of submersible mixer installation locations using the ISIGHT software can effectively improve mixing efficiency, overall plant operating efficiency, and economic benefits in sewage treatment plants. The multi-objective optimization platform based on the ISIGHT platform for wastewater treatment mixer installation location can be successfully applied in engineering practice.

The lightning's nature is that different-polarity leaders extend in air. Only negative leaders' development was previously associated to floating plasma. We found that the floating plasma could also lead the positive leader stepwise development.

ObjectiveExploring the relationship between (weight-adjusted waist index) WWI and arterial stiffness (AS) in the total and different BMI populations among patients with hypertension.MethodsThis study enrolled 5232 hypertensive subjects, a subset of the China H-type Hypertension Registry Study. WWI was calculated as WC (cm) divided by the square root of weight (kg). Brachial-ankle pulse wave velocity (baPWV) was measured to determine AS.ResultsThe mean WWI was 10.97 (0.78)cm/√kg. In multiple logistic analyses showed that there were significant dose-dependent association between WWI with baPWV in a dose-dependent manner in total population (β 57.98, 95% CI 44.06-71.90), and in different BMI group: group 1 (BMI

The ball valve is an important device in the pipeline transportation system of nuclear power plants. Its operational stability and safety directly affect the normal working of nuclear power plants. In this study, the transient numerical simulation of the opening and closing process of a ball valve was conducted on the basis of the flow interruption capability experiment of the ball valve by using the moving mesh method and inlet and outlet variable boundary conditions. The flow rate and pressure difference with time of the opening and closing process of the ball valve were studied. The internal flow characteristics of the ball valve under different relative openings were analyzed in conjunction with the typical back-step flow structure. Results show that the transient numerical results agree well with the experimental results. The internal flow characteristics of the ball valve are similar at the same opening during opening and closing process. At small opening, the spool and outlet channels easily form a back-step flow structure. The disappearance and generation of backflow vortices during opening and closing occur at 85% opening and 75% opening, respectively. With the decrease in opening degree, the difference in vortex core area in the flow channel of the ball valve spool in the opening and closing process gradually appears. The research results provide some reference value for the design and optimization of ball valves.

This study presents a comprehensive investigation of the internal noise characteristics of a mixed-flow pump by combining computational fluid dynamics (CFD) and computational acoustics. The turbulent flow field of the pump is simulated using the unsteady SST k-ω turbulence model in CFD. The contributions of the volute, guide vanes, and impeller to the internal noise are analyzed and compared using the Lighthill theory, FW-H formula, and LMS Virtual Lab software for acoustic simulation. The research findings indicate that the energy of pressure fluctuations in the mixed-flow pump is predominantly concentrated at the blade passing frequency and its low-frequency harmonics. This suggests that the internal noise is mainly in the low-frequency range, with higher energy at the blade passing frequency and its harmonics. Under the 0.6Qdes flow condition, the flow inside the pump becomes more complex, resulting in higher sound pressure levels and sound power levels compared to higher flow conditions. However, for flow conditions ranging from 0.8Qdes to 1.2Qdes, the sound pressure levels gradually increase with increasing flow rate, with the sound pressure level at 1.0Qdes being nearly identical to that at 1.2Qdes. The analysis of sound power level spectra at different flow rates reveals that the distribution characteristics of internal vortex structures directly impact the hydrodynamic noise inside the mixed-flow pump. These research findings provide a significant theoretical basis for noise control in mixed-flow pumps.

Abstract Rheumatoid arthritis (RA) is an essential cause of labor loss and disability for people worldwide. Acanthopanax senticosus polysaccharide (ASPS) is one of the most important active components from A. senticosus, which exhibits various pharmacological activities such as antioxidation and immunomodulation. However, no studies have reported the application of ASPS in treating RA. This study aims to investigate the therapeutic effect of ASPS on RA and reveal its underlying mechanism. The potential therapeutic effect of ASPS against RA is initially verified in this study using the collagen‐induced arthritis model. Moreover, the protective benefits of ASPS are transmitted through the fecal microbiota and blocked by simultaneous antibiotic cocktail treatment, indicating that gut microbiota may be correlated with ASPS. The 16S rRNA sequencing using feces samples and untargeted UPLC‐MS metabolomics using serum samples further reveal that ASPS reprograms the arthritic progression triggered dysbiosis, enhances the expression of γ‐glutamylcysteine (GGC) synthetase, and enriches the serum concentration of GGC. Furthermore, metabolites GGC is found to be able to effectively interrupt NLRP3 inflammasome activation via inhibiting ASC nucleation and therefore attenuate inflammatory arthritis. Taken together, this work highlights ASPS's therapeutic potential against RA, which mainly exhibits its effects via modulating gut microbiota and regulating GGC production.

With the increasingly severe situation of water pollution control, optimal design of the mixing flow field of submersible mixers and improving the mixing uniformity of activated sludge have become key research issues. At present, the research on the submersible mixer is mostly focused on water as the medium, and the flow field characteristics of solid-liquid two-phase flow, which is closer to the actual scene, still need more systematic research. This paper presented numerical simulations of the solid‒liquid two‒phase flow problem at various installation heights based on the coupled CFD‒DEM method in the Euler‒Lagrange framework. The velocity distribution, dead zone distribution, particles’ velocity development, particles’ mixing degree, and particles’ aggregation of the flow field were compared and analyzed for different installation heights. The results show that the flow field has two flow patterns: single‒ and double‒circulation, due to different installation heights, in which the velocity and turbulent kinetic energy of the flow field of the double‒circulation flow pattern are more uniform. The installation height affects the moment particles enter the impeller and the core jet zone, thus affecting the degree of particle mixing and the mixing time. The adjustment of the installation height also has an impact on particle aggregation. These findings indicate that the installation height significantly affects the flow field characteristics and the particle motion distribution. The coupled CFD‒DEM method can analyze the macroscopic phenomenon of the solid‒liquid two‒phase flow field of the submersible mixer from the scale of microscopic particles, which provides a theoretical approach for the optimal design of the mixing flow field. It can provide better guidance for engineering practice.

The unsteady flow characteristics of pump as hydraulic turbine play a vital role in its safe and stable operation, while the ultra-low specific-speed centrifugal pump may face more stability problems due to the limitations of its flow conditions under the turbine working condition. Therefore, in this study, the unsteady characteristics of an ultra-low specific-speed centrifugal pump under turbine conditions are studied using a numerical simulation method, and the numerical simulation is verified using an experimental method. Based on the hydraulic losses of each flow passage component, the energy characteristics of pump as turbine (PAT) are established, and the distribution pattern of total pressure fluctuation in the turbine is studied. The results show that the rotor–stator interaction between the impeller and the tongue makes the hydraulic performance and the internal flow field change periodically. The pressure fluctuation intensities at the tongue, blade inlet edge, and balance hole are large, and the total pressure fluctuation in the three areas is intense in space and time. The internal flow characteristics at typical blade positions show that the secondary flow phenomena such as separation flow and wake flow near the tongue make the pressure gradient larger, which is an important influence mode of the rotor–stator interaction. This study provides a reference and guidance for the unsteady study of low specific-speed PAT.

The fatigue-creep properties of Ni–Fe based superalloy with varying strain amplitude, temperature and holding duration are studied in ambient air. The results demonstrated that the number of cycles decreases greatly with increasing strain amplitude from 0.5% to 1.0%, and the ǁRD texture in the microstructure is conformed along the deformation direction. Both high temperature and holding duration are key factors in the reduction of fatigue cycles. It is determined that the dislocations bypass the second phase particles during deformation process, and the critical shear stress transition is observed. Different from the temperature factor, higher holding time leads to the growth of the second phase, and which finally exceeds the critical size of dislocation cutting, leading to the inducement for dislocations bypassing the second phase.

Background and aimsThe relationship between the new obesity index weight-adjusted-waist index (WWI) and hyperuricemia is unclear. We aimed to explore the association of the WWI and hyperuricemia among the hypertensive population.MethodsA total of 14,078 hypertension participants with complete data were included in our study. WWI was calculated by waist circumference divided by the square root of weight. Specifically, men with 420 μmol/L and women with 360 μmol/L were considered to have hyperuricemia.ResultsThe prevalence of hyperuricemia was 61.1% in men and 51.4% in women. On the whole, multivariate logistic regression analyses found that there was a linear positive correlation of WWI with hyperuricemia in both men (OR: 1.37; 95%CI: 1.25, 1.49) and women (OR: 1.35; 95%CI: 1.26, 1.45). Subgroup analysis found that the relationship between WWI and hyperuricemia was stable in stratified subgroups (all P-interactions >.05).ConclusionWWI showed a positive association with hyperuricemia among hypertension patients.

Abstract Background Inflammatory osteolysis after total joint replacement (TJR) may cause implant failure, periprosthetic fractures, and be a severe threat to global public health. Our previous studies demonstrated that melatonin had a therapeutic effect on wear-particles induced osteolysis. Gut microbiota is closely related to bone homeostasis, and has been proven to be affected by melatonin. However, whether melatonin could play its anti-osteolysis effects through reprogramming gut microbiota remains elusive. Results Here, we demonstrated that melatonin could alleviate Ti-particles induced osteolysis, while this therapeutic effect was blocked by antibiotic cocktail treatment. Interestingly, transplantation of fecal microbiota from mice treated with melatonin reappeared the same beneficial effect. Analysis of the 16S rRNA revealed that melatonin could reverse dysbacteriosis triggered by osteolysis, and elevate the relative abundance of some short chain fatty acid (SCFA) producing bacteria. Moreover, butyrate was enriched by exogenous melatonin administration, while acetate and propionate did not show an evident difference. This was consistent with the results of the metagenomic approach (PICRUSt2) analysis, which revealed a general increase in the synthetic enzymes of butyrate. More importantly, direct supplementation of butyrate could also recapitulate the anti-osteolysis effect of melatonin. Further analysis identified that butyrate alleviated osteolysis via activating its receptor GPR109A, and thus to suppress the activation of NLRP3 inflammasome triggered by Ti-particles. Conclusions Taken together, our results suggested that the benefits of melatonin mainly depend on the ability of modulating gut microbiota and regulating butyrate production. Graphic Abstract

Single blade centrifugal pump is suitable for transporting sewage water with large obstacles in diameter, which is required to achieve high performance. The blade inlet angle is a parameter that affects the performance of the single blade centrifugal pump. However, the selection of blade inlet angle is frequently based on experiences and demonstrates certain arbitrariness. Therefore, six impellers with different blade inlet angles are designed using ANSYS Bladegen and NX software. These impellers are categorized into Groups 1 and 2. The blade inlet angle of the impellers in Group1/Group2 increases/decreases from the front shroud to the rear one, respectively. Subsequently, the numerical simulation is carried out to investigate the effect of the blade inlet angle on the performance of the single blade pump. The blade inlet angle has more obvious influence on the head, efficiency, and shaft power for the impellers in Group 1 than those in Group 2. The lowest pressure is negative for one of the impellers in Group 1. Moreover, an optimum blade inlet angle improves the pump performance. In addition, the alternating load is small on the blades of the impellers in Group 2. The present results are helpful in designing the single blade centrifugal pump and improving its performance.

S U M M A R Y: Background: In clinical routine, preoperative radiographic assessment of lower extremity geometry relies on conventional X-rays. However, the plane goniometric measuring has several limitations in accurately locating anatomical landmarks. The purpose of this study is to propose a fast and accurate 3D-reconstruction-method based on biplanar X-rays with clinical measurements assessment in standing position. Methods: 50 candidates for HTO or DFO with deformity of the lower extremities were included in this study. Biplanar X-rays were performed using the EOS imaging system in conventional double-stance full weight-bearing position (DS) and shifted-foot standing position (SF). The results of hip-knee-ankle angle (HKAA), lateral distal femoral angle (LDFA), and medial proximal tibial angle (MPTA) were evaluated by either 2D manual goniometer (MG) based on X-ray in DS standing position or 3D-reconstruction goniometer based on X-rays in SF standing position. Results: For the reproducibility study, MG and EOS goniometer were both reliable in repeated measures of HKAA, LDFA, and MPTA, with average concordance correlation coefficients (CCCs) all above 0.910. The agreements between MG and EOS measurements were high for HKAA and LDFA with CCCs all above 0.90, while the agreement was low for MPTA with CCC below 0.75. Further linear regression model analysis also revealed a significant correlation between MG and EOS measurements for HKAA (all R2 ≥ 0.93) and LDFA (all R2 ≥ 0.90), but not for MPTA (all R2 ≤ 0.522). Conclusion: In comparison with the traditional 2D manual goniometer, EOS 3D reconstruction based goniometric measuring could provide equivalent results of HKAA and LDFA, and potentially a more accurate result of MPTA. These findings suggest that EOS 3D reconstruction based goniometric measuring is suitable for preoperative evaluation and planning for HTO/DFO. However, future improvements of the 3D reconstruction method are needed for better detection of the femoral condyles and tibial plates without the requirement of shifted-foot standing position. The translational potential of this article: EOS 3D reconstruction based goniometric measuring could provide equivalent or even more accurate results of HKAA, LDFA, and MPTA, in comparison with the traditional 2D manual goniometer, making it suitable for preoperative evaluation and planning for HTO/DFO.

Abstract Background Heat shot protein 90 (HSP90) AA1 functions as an onco-protein to regulate the assembly, manipulation, folding and degradation of its client proteins, including c-MYC. However, little is known about the mechanism of HSP90AA1 regulation. Methods Transcriptome RNA-sequencing data of hepatocellular carcinoma (HCC) samples were used to detect the mRNA expression of FBXL6. Immunoprecipitation/Mass Spectrum (IP/MS) method was used to identify the interacting proteins of FBXL6. The co-immunoprecipitation assay was used to determine the interaction between FBXL6 and HSP90AA1. The in vivo ubiquitination assay was performed to determine the regulation of HSP90AA1 by FBXL6. Luciferase reporter and chromatin immunoprecipitation (ChIP) assays were used to determine the transcriptional regulation of FBXL6 by c-MYC. Immunohistochemical (IHC) staining was performed to study the correlation of FBXL6 and HSP90AA1 protein expression in 87 HCC samples. Cell counting and colony formation assays were implemented to detect the biological effects of FBXL6 on the growth of HCC cells in vitro. The effect of FBXL6 on HCC tumor growth in vivo was studied in a tumor xenograft model in mice. Results Here, we identified the orphan F-box protein FBXL6, a substrate recognition subunit of an SCF (Skp1-Cul1-F-box protein) complex, as the ubiquitin ligase for HSP90AA1. FBXL6 promoted K63-dependent ubiquitination of HSP90AA1 to stabilize it. Through analysis of the TCGA dataset, we found that FBXL6 was significantly increased in HCC tissues and positively correlated with c-MYC pathway. FBXL6 accumulation in HCC causes the stabilization and activation of c-MYC by preventing HSP90AA1 degradation. The activated c-MYC directly binds to the promoter region of FBXL6 to induce its mRNA expression. Conclusion Collectively, our data revealed an unknown FBXL6-HSP90AA1-c-MYC axis which might contribute to the oncogenesis of HCC, and we propose that inhibition of FBXL6 might represent an effective therapeutic strategy for HCC treatment. Shot peening technologies can improve the performance of materials and extend the service life of parts. Cavitation water jet peening (CWJP) is a new shot peening technology that uses the shear effect of a high-speed jet in a submerged environment to produce a cluster of bubbles, thereby achieving a significant increase in impact performance. In this paper, CWJP was used to strengthen the surface of the 7075 aluminum alloy (Al7075), and the effects of CWJP with different scanning speeds on its microstructure and mechanical properties were investigated. The morphology evolution revealed that the plastic deformation of the sample surface became more serious and the surface roughness increased with the decrease in scanning speed. The distribution of residual stress and microhardness along the depth direction of the Al7075 was measured before and after the CWJP impact, and it was found that CWJP causes work hardening on the surface of the Al7075 and can convert the subsurface tensile stress of the Al7075 into compressive stress. The mechanism of grain refinement of the Al7075 during plastic deformation was systematically revealed. Therefore, CWJP can effectively improve the surface properties of the 7075 aluminum alloy.

Abstract Background Long non-coding RNA LINC00346 has been recently suggested as a prognostic marker in pancreatic cancer. However, its biological function in pancreatic cancer has not yet been determined. In this study, we attempted to ascertain the role of LINC00346 in regulating the aggressiveness of pancreatic cancer. Methods The effects of overexpression and knockdown of LINC00346 on the proliferation, cell cycle progression, apoptosis, and gemcitabine resistance were investigated. Bioinformatic analysis, luciferase reporter assay, and RNA immunoprecipitation assay were performed to search for potential microRNAs (miRs) that can interact with LINC00346. Results Overexpression of LINC00346 significantly enhanced the proliferation, colony formation, and tumorigenesis of pancreatic cancer cells. Conversely, knockdown of LINC00346 suppressed pancreatic cancer cell proliferation and caused a cell-cycle arrest at the G2/M-phase. Depletion of LINC00346 also enhanced gemcitabine sensitivity in pancreatic cancer cells both in vitro and in vivo. Mechanistic investigation revealed that LINC00346 acted as a sponge for miR-188-3p and blocked the repression of BRD4 by miR-188-3p in pancreatic cancer cells. Clinical evidence indicated a negative correlation between LINC00346 and miR-188-3p in pancreatic cancer specimens. Rescue experiments showed that LINC00346 attenuated the growth-suppressing and chemosensitizing effects of miR-188-3p on pancreatic cancer cells. In addition, silencing of BRD4 significantly inhibited LINC00346-induced pancreatic cancer cell proliferation and colony formation. Conclusions LINC00346 shows the ability to promote pancreatic cancer growth and gemcitabine resistance, which is in part mediated by antagonization of miR-188-3p and induction of BRD4. Targeting LINC00346 may improve gemcitabine-based therapeutic efficacy.

Submersible mixers are widely used in the sewage treatment process in various fields, such as agriculture and industry. They are mainly responsible for pushing flow and mixing activated sludge particles in a pool. Based on the CFD-DEM coupling method under the Euler–Lagrange framework, the solid–liquid two-phase flow of a submersible mixer was simulated in this paper, and the motion characteristics and distribution laws of particles in the pool were studied in axial, horizontal, and lateral directions, respectively. An evaluation method of distribution uniformity was proposed to analyze the velocity distribution of the flow field, the velocity distribution of particles, and the mixing uniformity of particles. The results show that the movement process of activated sludge particles in the pool can be roughly divided into three stages: the horizontal development stage, absorption–injection stage, and reflux-mixing stage, in which the reflux-mixing stage is the main stage for the uniform distribution of particles in the whole flow field. Particle accumulation occurs mainly in the dead zones of the flow field. Distribution of particles in the axial direction has the most homogeneous extent. Vortices can be generated near pool walls, causing accumulation of particles. This method can be a good guide for engineering practice.

To determine the influences of the main structural parameters on open-design vortex pump performance, optimize the vortex pump performance, and reduce the running vibration and improve stability, orthogonal testing method was introduced in this paper. The selected main factors included impeller outer diameter (D2), impeller outlet width (b2), outlet setting angle of impeller (β2), and inlet setting angle of impeller (β1), and the nine types of impellers were coded according to orthogonal table. After obtaining the preliminarily optimum value range for each factor through range analysis, comprehensive analysis was employed based on the orthogonal test to investigate the main factors and identify the primary and secondary influencing factors affecting the performance of the vortex pump. An optimization scheme was obtained for further design. The results show that the numerical calculation results of the optimization scheme pump are in good agreement with the test results, and it shows the feasibility of the numerical calculation method. The testing results showed that efficiency and head of the optimal model were 4.2% and 9 m higher than those of the prototype model, respectively. Improved efficiency and head met the design requirements. The orthogonal testing method proved the feasibility of performance optimization of the vortex pump. The backflow occurs at the pump entrance and rotates in the same direction with impeller. It moves along the pipe wall from the lateral cavity to the inlet and encourters with the approaching flow.

To find a better method to suppress cavitation, a microchannel design connecting the internal low-pressure area with the outside is proposed for the first time in this paper; the method was adopted to replenish fluid in the interior of the low-pressure area to inhibit cavitation. Through numerical simulation, it is found that the size and position of microchannel have a certain influence on the cavitation inhibition. The results show that the generation and development of cavitation, under the same working conditions, can be effectively restrained by adopting appropriate microchannel (x = 0.05 c, d = 6 cm). Compared with the original hydrofoil, the scale of cavitation is reduced by nearly 50%, and its turbulent kinetic energy remains unchanged. Therefore, it is considered that microchannel technology, as a new means of cavitation suppression, is of great significance to other types of fluid machinery.

A water-jet erosion test was carried out on grade-A ship steel to study the interaction and erosion mechanism of the water jet on the steel surface. When the water jet impacted, a STSS-1 stress-detection module was used to collect the dynamic strain signal on the rear of the ship’s plate, and a scanning electron microscope, transmission electron microscope, X-ray diffractometer, and other equipment were used to analyze the microstructure and phase of the grade-A ship steel before impact. The surface morphology of the material after impact was studied and analyzed. The impact stress of the water jet on the grade-A steel was an alternating stress, and the jet pressure decayed in the radial direction. The material surface was fatigued under the action of the jet alternating stress. After the water-jet erosion, the central area of the grade-A steel was dominated by an elongated cementite hard phase, and the peripheral area had a pearlite structure. A model for the jet erosion and peeling of grade-A ship steel was established to clarify the mechanism of erosion by the water jet.

The submerged high-pressure water jet has the characteristics of high velocity, strong turbulence, and severe cavitation. In order to reveal the formation mechanism of shear cavitation in the submerged high-pressure water jet and to grasp the turbulent structure and velocity distribution characteristics in the jet, the prediction ability of different turbulence models is studied first. The models represent the RANS model and RANS-LES hybrid model which are used to simulate the same cavitation jet, and the results are compared with the experimental results. The most reasonable model is then used to investigate the submerged high-pressure cavitation jet with different cavitation numbers. It is found that the calculation accuracy for small-scale vortexes has a great influence on the prediction accuracy of cavitation in the submerged jet. Both the DDES model and the SBES model can effectively capture the vortexes in the shear layer, and the SBES model can obtain more turbulence details. The result of the simulation under different cavitation numbers using the SBES model agrees well with the experimental result. Under the condition with low cavitation number, an intensive shear layer is formed at the exit of the nozzle, and small-scale vortexes are distributed along the shear layer. Mass transfer rate is relatively high in the region with a stronger vortex, which confirms that the low pressure in the vortex center is the main reason for the generation of cavitation in the shear layer. With the decrease of the cavitation number, the cavitation intensity increases obviously, while the nondimensional velocity along the radial direction changes little, which follows an exponential function.

In order to analyze the effect of impeller with different slot widths on the performance of the low-specific-speed centrifugal pumps, based on the impeller of a single-stage pump with the specific speed of 21, two gap drainage schemes with slot widths of 1.5 mm and 6.0 mm, slot diameter of 180 mm, and lap length of 5 mm were designed. Both experimental and numerical simulation methods were applied to compare the steady performance, which includes the head, efficiency, and the internal flow field distribution, and the unsteady pressure pulsation performance between new designed pumps and the original pump. The results show that gap drainage would cause a certain degree of head reduction, but a smaller slot width could achieve higher efficiency. Meanwhile, a reasonable open seam scheme can reduce the development of pressure pulsation, which provides experience and reference for the stable operation of low-specific-speed centrifugal pumps.

Background:. Accurate preoperative planning for total hip arthroplasty (THA) relies on conventional anteroposterior radiographs. The difficulty of determining the magnification factor of radiographs is a major limitation. Despite the use of markers for calibration, identifying the plane of the hip joint is a major challenge. The aim of this study was to evaluate the accuracy of a novel method for image calibration and preoperative planning in THA involving the use of a biplanar radiographic (EOS imaging) system and a self-designed coin device. Methods:. Biplanar radiographs (with the self-designed coin device) and a conventional anteroposterior radiograph (with a coin) were made for 26 patients after primary THA. The agreement between the actual and calculated diameters for each method was assessed using the concordance correlation coefficient (CCC) and Bland-Altman plots. In addition, 15 patients undergoing primary THA were prospectively enrolled to evaluate the EOS imaging-based method (EOS method), with biplanar radiographs made with use of the coin device. The accuracy of the preoperative predicted size of the implants was evaluated. Results:. Both the EOS and conventional anteroposterior radiograph-based methods were reliable in repeated measurements of the diameter of the artificial femoral head in the reproducibility study, with the average CCCs for both methods >0.990. The agreement between the actual and EOS-based calculated diameters of the artificial femoral head was excellent, with a CCC of >0.990, while the agreement was poor between the actual and anteroposterior radiograph-based calculated diameters, with a CCC of

The process of nanoemulsion was improved by optimizing the concentration of chitosan-N-acetylene-L-cysteine and sodium tripolyphosphate and the volume ratio between the two components. The prepared formulations were characterized in terms of particle size. Support vector regression (SVR) was built up and used to identify the parameters which could influence the particle size of the nanoemulsion. Data were divided into calibration and validation sets and then were modeled by SVR. It was found that the developed model was of high quality. This model was then used to explore the effect of composition and processing factors on the particle size of the nanoemulsion preparation. This study demonstrated that the SVR model may be used to optimize critical parameters to control preparation of this system and to obtain a dominant factor to control the final particle size.

In order to study the shaft system vibration of mixed-flow pump under rotor–stator interaction, the unsteady pressure fluctuation characteristics are measured and the rotor axis orbit obtained based on the Bentley 408 data acquisition system. The relationship between pressure fluctuation and vibration characteristics of shaft system at the blade passing frequency is analyzed. The results show that the pressure fluctuation amplitude is the largest and the rotor–stator interaction effect is the most obvious in the middle of the impeller. Along the direction of the main stream, the velocity energy is converted into pressure energy, the rotor–stator interaction effect is gradually weakened, and the main frequency of the pressure pulsation gradually turns from the 4 X frequency to the 1 X frequency of the impeller rotation frequency. The hydraulic stirring vibration and other factors lead to jagged sharp corners on the original axis orbit. The axis orbit of 1 X frequency is an ellipse with little difference between long and short axis while the 2 X frequency is the opposite, from which the existence of arcuate rotary whirl and misalignment phenomenon of the rotor can be judged. Combined with time–frequency characteristics of pressure pulsation, it can be found that the hydraulic imbalance has a great influence on the vibration of the shafting, while the rotor–stator interaction at the blade passing frequency takes the second place, which is the main factor of inducing the 4 X frequency vibration of the axis orbit. This study targets is that providing practical guidance for improving operation stability and preventing the vibration failure of the mixed-flow pump.

High-pressure submerged cavitation jet is widely used in the fields of material peening, petroleum drilling, and ocean engineering. The impact performance of the jet with intensive cavitation is related to the factors such as working condition and the nozzle geometry. To reveal the relationship between the nozzle divergent angle and the jet pressure on the unsteady characteristics of the jet, high-speed photography with frame rate of 20000 fps is used to record the image of the cavitation clouds. Grayscale analysis algorithm developed in MATLAB is used to study the effects of injecting condition on the special structure, unsteady characteristics, and shedding frequency of the cavitation bubbles. The impact load characteristics of the cavitation jet with different cavitation numbers and stand-off distances are recorded using a high-response pressure transducer. It is found that the cavitation number is the main factor affecting the cavitation morphology of the submerged jet. The lower the cavitation number is, the more intense the cavitation occurs. The outlet divergent angle of the convergent-divergent nozzle also has a significant influence on the development of the cavitation clouds. In the three nozzles with the outlet divergent angles of 40°, 80°, and 120°, the highest bubble concentration is formed usinga nozzle with a divergent angle of 40°, but the high-concentration cavitating bubbles are only distributed in a very small range of the nozzle outlet. The cavities generated by using the nozzle with a divergent angle of 80° can achieve good results in terms of concentration and distribution range, while the nozzle with divergent angle of 120° has lower cavitation performance due to the lack of the constraint at the outlet which intensifies the shear stress of the jet. According to the result of frame difference method (FDM) analysis, the jet cavitation is mainly formed in the vortex structure generated by the shearing layer at the nozzle exit, and the most severe region in the collapse stage is the rear end of the downstream segment after the bubble cloud sheds off. The impact load of the cavitation jet is mainly affected by the stand-off distance of the nozzle from the impinged target, while the nozzle outlet geometry also has an effect on the impact performance. Optimizing the stand-off distance and the outlet geometry of the nozzles is found to be a good way to improve the performance of the cavitation jet.

In order to investigate the effect of various impeller tip clearances on the pressure pulsation in a mixed-flow pump, the energy performance test and pressure pulsation experiment of a mixed-flow pump with different tip clearances are studied simultaneously. The pressure pulsation signals at impeller inlet, middle, and oulet are processed and analyzed by the wavelet transform. The results show that the change of tip clearance can affect the head and efficiency of mixed-flow pump. Under designed flow rate, when the tip clearance increases from 0.2 mm to 1.1 mm, the head and efficiency decrease by 18.1% and 11.6%, respectively. Due to the strong influence of blade passing frequency (BPF) at impeller middle, the period of pressure pulsation curve is about 1/4 of impeller rotation period. At impeller inlet, the low-frequency pulsation with energy concentration is the main disturbance frequency, and, with the increase of tip clearance, not only does the high-value region of wavelet spectrum expand to low-frequency direction but also it is easy to form second-order peaks in the time-averaged wavelet curve. At impeller outlet, affected by BPF and rotor-stator interaction (RSI), the high-frequency disturbance in RSI area decreases first and then increases. The wavelet coherence demonstrates the stable low-frequency disturbances in comparison to others and it will affect the flow field at impeller middle.

Based on current conditions and historical records, profitability in production farming depends on making a right and timely operational decision. Precision farming is a systematic program designed to maximize the productivity of agriculture by carefully tailoring the soil and crop management to meet the specific requirements in each field while preserving environmental quality. This review paper highlights the development of an automated irrigation system with portable wireless sensor networks and decision support methods to remotely measure the environmental parameters in an agriculture field. Radio satellite, mobile phones, sensors, internet-based communication, and microcontroller capture the ecological parameters such as soil moisture, temperature, humidity, and light intensity. The knowledge gained from the sensors is transferred directly to the cloud server by using IoT technology. Users from anywhere in the world can display them through an internet-enabled device. Development of sensor-based application in modern agriculture makes it cost-effective and potentially productive and increases the efficiency through precision agriculture farming. Different limitations have been reported in the previously reviewed publications like the shortage of power in the field that can be solved by using a solar panel that recharges the battery at the same time using electricity. Bluetooth application in the agriculture sector is mainly improved by design system optimization. Problems related to transmission and radio range frequency can be solved by using a power class upgraded antenna.

Studies on the tip leakage vortex (TLV) are extensive, while studies on the secondary tip leakage vortex (S-TLV) are rare. To advance the understanding of the formation mechanism of the S-TLV, turbulent cavitating flows were numerically investigated using the shear stress transport (SST) turbulence model and the Zwart–Gerber–Belamri cavitation model. The morphology and physical quantity distribution of the S-TLV under two cavitation conditions were compared, and its formation mechanism was analyzed. The results reveal that in the lower cavitation number case, there is a low-velocity zone of circumferential flow near the tip in the back half of the blade. The shear vortices formed by the leakage jet gradually accumulate and concentrate in the low-velocity area, which is one of the main sources of the S-TLV. Meanwhile, the radial jet pushes the vortices on the suction surface to the tip, which mixes with the S-TLV. The flow path formed by the radial jet and the leakage jet is in accordance with the rotation direction of the S-TLV, which promotes the S-TLV’s further development. Under the conditions of a small cavitation number and low flow rate, the circumferential velocity and radial velocity of the fluid near the gap have altered significantly, which is conducive to the formation of the S-TLV.

Natural ventilators can maintain the ventilation of buildings and tunnels, and can exhaust fire smoke without requiring energy. In this study, we optimized a natural ventilator by adding axial fan blades (equivalent to adding a fan system) to investigate the effect of wind speed on the ventilation and smoke exhaust performance of an optimized natural ventilator. The experimental results showed that the best configuration of the ventilator was five fan blades at an angle of 25° with set-forward curved fan blades. With this configuration, the ventilation volume of the optimized natural ventilator was increased by 11.1%, and the energy consumption was reduced by 2.952 J. The third experiment showed that, in the case of a fire, the optimized ventilator can reduce the temperature of the ventilator faster than the original ventilator, indicating better smoke exhaust performance. The reason for this effect is that, when the optimized natural ventilator rotates, the rotation of the blades creates a flow field with a more evenly distributed wind speed. The experiments proved that natural ventilators can be optimized by adding a fan system. The results of this study can be applied to effectively improve the ventilation performance of natural ventilators to quickly exhaust smoke in building and tunnel fires, and provide a reference for related research on natural ventilators.

Abstract Background The glutamine synthetase (GS), an astrocyte-specific enzyme, is involved in lipopolysaccharide (LPS)-induced inflammation which activates the mitogen-activated protein kinase (MAPK) signaling. Endocannabinoid 2-arachidonyl glycerol (2-AG) has been described to serve as an endogenous mediator of analgesia and neuroprotection. However, whether 2-AG can directly influence astrocytic GS and MAPK expressions remains unknown. Methods In the present study, the effects of 2-AG on astrocytic GS expression, p38 and ERK1/2 expression, cell viability, and apoptosis following LPS exposure were investigated. Results The results revealed that LPS exposure increased GS expression with p38 activation in the early phase and decreased GS expression with activation of ERK1/2, decrease of cell viability, and increase of apoptosis in the late phase. Inhibition of p38 reversed GS increase in the early phase while inhibition of ERK1/2 reversed GS decrease in the late phase induced by LPS exposure. 2-AG protected astrocytes from increase of apoptosis and decrease of cell viability induced by the late phase of LPS exposure. In the early phase of LPS exposure, 2-AG could suppress the increase of GS expression and activation of p38 signaling. In the late phase of LPS exposure, 2-AG could reverse the decrease of GS expression and activation of ERK1/2 induced by LPS. Conclusion These findings suggest that 2-AG could maintain the GS expression in astrocytes to a relatively stable level through modulating MAPK signaling and protect astrocytes from LPS exposure.

Public blockchains provide a decentralized method for storing transaction data and have many applications in different sectors. In order for users to track transactions, a simple method is to let them keep a local copy of the entire public ledger. Since the size of the ledger keeps growing, this method becomes increasingly less practical, especially for lightweight users such as IoT devices and smartphones. In order to cope with the problem, several solutions have been proposed to reduce the storage burden. However, existing solutions either achieve a limited storage reduction (e.g., simple payment verification), or rely on some strong security assumption (e.g., the use of trusted server). In this paper, we propose a new approach to solving the problem. Specifically, we propose an efficient verification protocol for public blockchains, or EPBC for short. EPBC is particularly suitable for lightweight users, who only need to store a small amount of data that is independent of the size of the blockchain. We analyze EPBC’s performance and security, and discuss its integration with existing public ledger systems. Experimental results confirm that EPBC is practical for lightweight users.
