Abstract

Serum amyloid A (SAA) is a family of proteins, the plasma levels of which may increase >1000-fold in acute inflammatory states. We investigated the role of SAA in sepsis using mice deficient in all three acute-phase SAA isoforms (SAA-TKO). SAA deficiency significantly increased mortality rates in the three experimental sepsis mouse models: cecal ligation and puncture (CLP), cecal slurry (CS) injection, and lipopolysaccharide (LPS) treatments. SAA-TKO mice had exacerbated lung pathology compared to wild-type (WT) mice after CLP. A bulk RNA sequencing performed on lung tissues excised 24 h after CLP indicated significant enrichment in the expression of genes associated with chemokine production, chemokine and cytokine-mediated signaling, neutrophil chemotaxis, and neutrophil migration in SAA-TKO compared to WT mice. Consistently, myeloperoxidase activity and neutrophil counts were significantly increased in the lungs of septic SAA-TKO mice compared to WT mice. The in vitro treatment of HL-60, neutrophil-like cells, with SAA or SAA bound to a high-density lipoprotein (SAA-HDL), significantly decreased cellular transmigration through laminin-coated membranes compared to untreated cells. Thus, SAA potentially prevents neutrophil transmigration into injured lungs, thus reducing exacerbated tissue injury and mortality. 13. Play Therapy for Bereaved Children: Adapting Strategies to Community, School, and Home Settings

Abstract

Play therapy is a highly adaptable treatment method that can be modified according to children's ages, circumstances, and settings in which counseling occurs. Play therapy may be used in schools, community settings, and homes to help children following the death of a significant other. After reviewing basic developmental factors that affect children's ability to comprehend the meaning of death, the article discusses the special circumstances of grief in different situations, including community-based counseling after Hurricane Katrina, school-based group play therapy following a teacher's death, and conjoint parent-child play therapy after a father's death in a terrorist attack. Increasing the acceptability and effectiveness of play therapy, professionals must consider and incorporate family and community traditions and beliefs. Additionally, professionals are encouraged to support teachers and parents in utilizing play-based activities to facilitate children's expressions of grief.

Abstract

Children have not yet developed the coping mechanisms of adults, and it is difficult for them to verbally communicate their needs. Practitioners increasingly recognize the usefulness of nonverbal communication methods to help these children. This revised and updated casebook and text focuses on treatment of children who have experienced crises resulting from psychological, physical, or environmental events, such the violence of war or terrorist acts. This edition notes that managed care has emerged a controlling factor in the treatment planning for children. Counselors have learned to utilize short-term, intermittent, and group therapy to help children recover from their exposure to crisis. Although traditional one-on-one interventions continue to be appropriate for children who have experienced severe circumstances such as suicide in the family or multiple traumas, many children can benefit from briefer methods. The book presents case studies in a two-column format that gives readers content of therapy sessions, along with clinician's accompanying thoughts and rationale for intervention. Chapters include literature reviews, case summaries, assessment and treatment plans, and questions for discussion. It demonstrates a variety of play therapy methods such as art, story telling, group art activities, and games, and provides examples of the power of play as a means of symbolic communication. Challenges included establishing trust and increasing parent and school personnel awareness of demands on university research faculty. 22. Predictors of Child Caregiver Commitment.

Abstract

This study examined the relationship between child caregiver commitment and the caregiver's personal characteristics and work experience, the characteristics of the job, and the quality of the child care center. Questionnaires that included several measurement instruments were sent to 381 full-time child caregivers in 94 centers. Caregiver commitment was measured by means of the Organizational Commitment Questionnaire; job satisfaction was measured by the Job Descriptive Index; and job characteristics were measured by the Job Diagnostic Survey. Center quality was assessed by an analysis of four factors: staff-child ratio; amount of caregiver training; group size; and staff turnover. Results indicated that the most powerful predictor of caregiver commitment was the caregiver's job satisfaction. A relationship to commitment was also found for the caregiver's perception of job characteristics, the caregiver's age, and the caregiver's education. A relationship to commitment was not found for center quality, caregiver wages, and caregiver tenure. A 44-item bibliography is included. (BC)

Abstract

Metabolic changes are assessed after initiation of protease inhibitor (PI)-sparing regimes in antiretroviral-naive patients. The results have shown mild increases in glucose and decreases in insulin sensitivity in all regimens, whereas lipids are modestly higher in the 3-drug efavirenz (EFV)-containing arms and the metabolic dysfunctions of concern after the PI-sparing therapies are abnormal total cholesterol (TC) and lower high-density lipoprotein cholesterol (HDL-C) levels.

Abstract

Several studies in the past have reported positive correlations between circulating Serum amyloid A (SAA) levels and obesity. However, based on limited number of studies involving appropriate mouse models, the role of SAA in the development of obesity and obesity-related metabolic consequences has not been established. Accordingly, herein, we have examined the role of SAA in the development of obesity and its associated metabolic complications in vivo using mice deficient for all three inducible forms of SAA: SAA1.1, SAA2.1 and SAA3 (TKO). Male and female mice were rendered obese by feeding a high fat, high sucrose diet with added cholesterol (HFHSC) and control mice were fed rodent chow diet. Here, we show that the deletion of SAA does not affect diet-induced obesity, hepatic lipid metabolism or adipose tissue inflammation. However, there was a modest effect on glucose metabolism. The results of this study confirm previous findings that SAA levels are elevated in adipose tissues as well as in the circulation in diet-induced obese mice. However, the three acute phase SAAs do not play a causative role in the development of obesity or obesity-associated adipose tissue inflammation and dyslipidemia. [ABSTRACT FROM AUTHOR]
