The ability to resolve the dynamics of matter on its native temporal and spatial scales constitutes a key challenge and convergent theme across chemistry, biology, and materials science. The last couple of decades have witnessed ultrafast electron diffraction (UED) emerge as one of the forefront techniques with the sensitivity to resolve atomic motions. Increasingly sophisticated UED instruments are being developed that are aimed at increasing the beam brightness in order to observe structural signatures, but so far they have been limited to low average current beams. Here, we present the technical design and capabilities of the HiRES (High Repetition-rate Electron Scattering) instrument, which blends relativistic electrons and high repetition rates to achieve orders of magnitude improvement in average beam current compared to the existing state-of-the-art instruments. The setup utilizes a novel electron source to deliver femtosecond duration electron pulses at up to MHz repetition rates for UED experiments. Instrument response function of sub-500 fs is demonstrated with < 100 fs time resolution targeted in future. We provide example cases of diffraction measurements on solid-state and gas-phase samples, including both micro- and nanodiffraction (featuring 100 nm beam size) modes, which showcase the potential of the instrument for novel UED experiments.

Emission inventories are essential for modelling studies and pollution control, but traditional emission inventories are usually updated after a few years based on the statistics of “bottom-up” approach from the energy consumption in provinces, cities, and counties. The latest emission inventories of multi-resolution emission inventory in China (MEIC) was compiled from the statistics for the year 2016 (MEIC_2016). However, the real emissions have varied yearly, due to national pollution control policies and accidental special events, such as the coronavirus disease (COVID-19) pandemic. In this study, a four-dimensional variational assimilation (4DVAR) system based on the “top-down” approach was developed to optimise sulfur dioxide (SO2) emissions by assimilating the data of SO2 concentrations from surface observational stations. The 4DVAR system was then applied to obtain the SO2 emissions during the early period of COVID-19 pandemic (from 17 January to 7 February 2020), and the same period in 2019 over China. The results showed that the average MEIC_2016, 2019, and 2020 emissions were 42.2×106, 40.1×106, and 36.4×106 kg d−1. The emissions in 2020 decreased by 9.2 % in relation to the COVID-19 lockdown compared with those in 2019. For central China, where the lockdown measures were quite strict, the mean 2020 emission decreased by 21.0 % compared with 2019 emissions. Three forecast experiments were conducted using the emissions of MEIC_2016, 2019, and 2020 to demonstrate the effects of optimised emissions. The root mean square error (RMSE) in the experiments using 2019 and 2020 emissions decreased by 28.1 % and 50.7 %, and the correlation coefficient increased by 89.5 % and 205.9 % compared with the experiment using MEIC_2016. For central China, the average RMSE in the experiments with 2019 and 2020 emissions decreased by 48.8 % and 77.0 %, and the average correlation coefficient increased by 44.3 % and 238.7 %, compared with the experiment using MEIC_2016 emissions. The results demonstrated that the 4DVAR system effectively optimised emissions to describe the actual changes in SO2 emissions related to the COVID lockdown, and it can thus be used to improve the accuracy of forecasts.

This paper presents a three-dimensional variational (3DVAR) data assimilation (DA) system for aerosol optical properties, including aerosol optical thickness (AOT) retrievals and lidar-based aerosol profiles, developed for the Model for Simulating Aerosol Interactions and Chemistry (MOSAIC) within the Weather Research and Forecasting model coupled to Chemistry (WRF-Chem) model. For computational efficiency, 32 model variables in the MOSAIC_4bin scheme are lumped into 20 aerosol state variables that are representative of mass concentrations in the DA system. To directly assimilate aerosol optical properties, an observation operator based on the Mie scattering theory was employed, which was obtained by simplifying the optical module in WRF-Chem. The tangent linear (TL) and adjoint (AD) operators were then established and passed the TL/AD sensitivity test. The Himawari-8 derived AOT data were assimilated to validate the system and investigate the effects of assimilation on both AOT and PM2.5 simulations. Two comparative experiments were performed with a cycle of 24 h from 23 to 29 November 2018, during which a heavy air pollution event occurred in northern China. The DA performances of the model simulation were evaluated against independent aerosol observations, including the Aerosol Robotic Network (AERONET) AOT and surface PM2.5 measurements. The results show that Himawari-8 AOT assimilation can significantly improve model AOT analyses and forecasts. Generally, the control experiments without assimilation seriously underestimated AOTs compared with observed values and were therefore unable to describe real aerosol pollution. The analysis fields closer to observations improved AOT simulations, indicating that the system successfully assimilated AOT observations into the model. In terms of statistical metrics, assimilating Himawari-8 AOTs only limitedly improved PM2.5 analyses in the inner simulation domain (D02); however, the positive effect can last for over 24 h. Assimilation effectively enlarged the underestimated PM2.5 concentrations to be closer to the real distribution in northern China, which is of great value for studying heavy air pollution events.

MicroRNAs are short non-coding RNAs that regulate gene expression. Several microRNAs, useful for coronary artery disease assessment, have previously been identified. MicroRNA-33 is located within SREBP introns and controls cholesterol homeostasis. In order to find the possibility of microRNA-33 as a potential biomarker in high cholesterol disease, we developed a mouse model for coronary heart disease by feeding mice with a high-fat diet. The expression differences of microRNA-33, SREBP and ABCA1 genes in the liver, muscle, and lipid tissues were compared between a high-cholesterol group and control group in mice. The results showed that ABCA1 was up-regulated by high cholesterol conditions in liver, muscle and lipid tissues. SREBP1C was up-regulated by high cholesterol conditions in the liver and lipid tissues and down-regulated by high cholesterol conditions in the muscle tissue. MicroRNA-33 and SREBP2 were down-regulated by high cholesterol conditions in the liver and muscle tissues and up-regulated by high cholesterol conditions in the lipid tissue. Our study suggests that antisense therapeutic targeting of microRNA-33 may be a potential biomarker for cardiovascular disease.

Introduction Apart from the psychiatric symptoms, cognitive deficits are also the core symptoms of schizophrenia. Brain network control theory provided information on the role of a specific brain region in the cognitive control process, helping understand the neural mechanism of cognitive impairment in schizophrenia. Objectives To characterize the control properties of functional brain network in first-episode untreated patients with schizophrenia and the relationships between controllability and psychiatric symptoms, as well as exploring the predictive value of controllability in differentiating patients from healthy controls (HCs). Methods Average and modal controllability of brain networks were calculated and compared between 133 first-episode untreated patients with schizophrenia and 135 HCs. The associations between controllability and clinical symptoms were evaluated using sparse canonical correlation analysis. Support vector machine (SVM) and SVM-recursive feature elimination combined with the controllability were performed to establish the individual prediction model. Results Compared to HCs, the patients with schizophrenia showed increased average controllability and decreased modal controllability in dorsal anterior cingulate cortex (dACC). Brain controllability predominantly in somatomotor, default mode, and visual networks was associated with the positive symptomatology of schizophrenia. The established model could identify patients with an accuracy of 0.68. Furthermore, the most discriminative features were located in dACC, medial prefrontal lobe, precuneus and superior temporal gyrus. Conclusions Altered controllability in dACC may play a critical role in the neuropathological mechanisms of cognitive deficit in schizophrenia, which could drive the brain function to different states to cope with varied cognitive tasks. As symptom-related biomarkers, controllability could be also beneficial to individual prediction in schizophrenia. Disclosure No significant relationships.

Introduction There is a growing consensus on brain networks that it is not immutable but rather a dynamic complex system for adapting environment. The neuroimaging research studying how brain regions work collaboratively with dynamic methods had demonstrated its effectiveness in revealing the neural mechanisms of schizophrenia. Objectives To investigate altered dynamic brain functional topology in first-episode untreated schizophrenia patients (SZs) and establish classification models to find objective brain imaging biomarkers. Methods Resting-state-functional magnetic resonance data for SZs and matched healthy controls were obtained(Table1). Power-264-template was used to extract nodes and sliding-window approach was carried out to establish functional connectivity matrices. Functional topology was assessed by eigenvector centrality(EC) and node efficiency and its time-fluctuating was evaluated with coefficient of variation(CV). Group differences of dynamic topology and correlation analysis between Positive and Negative Syndrome Scale(PANSS) scores and topology indices showing group differences, which also were used in establishing classification models, was examed. Results The CV of node efficiency in angular and paracingulate gyrus was larger in SZs. There are 13 nodes assigned into several brain networks displaying altered CV of EC between groups(Figure1.A). Fluctuation of EC of the node in DMN, which was lower in SZs, showed negative correlation with PANSS total scores(Figure1.B). Dynamic functional topology of above nodes was used to train classification models and demonstrated 80% and 71% accuracy for support vector classification(SVC) and random forest(RF), respectively(Figure2). Conclusions Dynamic functional topology illustrated a capability in identifying SZs. Aberrated dynamics of DMN relevant to severity of patient’s symptoms could reveal the reason why it contributed to classification. Disclosure No significant relationships.

The authors developed a three-dimensional variational (3-DVAR) aerosol extinction coefficient (AEC) and aerosol mass concentration (AMC) data assimilation (DA) system for aerosol variables in the Weather Research and Forecasting–Chemistry (WRF–Chem) model with the WRF–Chem using the Model for Simulating Aerosol Interactions and Chemistry (MOSAIC) scheme. They establish an AEC observation operator and its corresponding adjoint based on the Interagency Monitoring of Protected Visual Environments (IMPROVE) equation and investigate the use of lidar AEC and surface AMC DA to forecast mass concentration (MC) profiles of PM2.5 (particulate matter with an aerodynamic diameter of less than 2.5 µm) across China. Two sets of data were assimilated: AEC profiles captured by five conventional Mie scattering lidars (positioned in Beijing, Shijiazhuang, Taiyuan, Xuzhou, and Wuhu) and PM2.5 and PM10 MC data obtained from over 1500 ground environmental monitoring stations across China. Three DA experiments (i.e., a PM2.5 (PM10) DA experiment, a lidar AEC DA experiment, and a simultaneous PM2.5 (PM10) and lidar AEC DA experiment) with a 12 h assimilation period and a 24 h forecast period were conducted. The PM2.5 (PM10) DA reduced the root mean square error (RMSE) of the surface PM2.5 MC in the initial field of the model by 38.6 µg m−3 (64.8 %). When lidar AEC data were assimilated, this reduction was 10.5 µg m−3 (17.6 %), and a 38.4 µg m−3 (64.4 %) reduction occurred when the two data sets were assimilated simultaneously, although only five lidars were available within the simulation region (approximately 2.33 million km2 in size). The RMSEs of the forecasted surface PM2.5 MC 24 h after the DA period in the three DA experiments were reduced by 6.1 µg m−3 (11.8 %), 1.5 µg m−3 (2.9 %), and 6.5 µg m−3 (12.6 %), respectively, indicating that the assimilation and hence the optimization of the initial field have a positive effect on the PM2.5 MC forecast performance over a period of 24 h after the DA period.

Hexagonal boron nitride (h-BN) is considered as one of the most promising materials for next-generation quantum technologies. In this paper, we report bulk-like multilayer h-BN epitaxially grown using a carbon-free precursor on c-plane sapphire with a strong emphasis on material characterization and analysis. In particular, structural, morphological, and vibrational properties, and chemical bonding of such van der Waals materials are presented. Between as-grown h-BN and c-plane sapphire, a compressive residual strain induced by both lattice mismatch and thermal expansion mismatch is examined by both theoretical and experimental studies. Atomic force microscopy revealed and scanning electron microscopy supported the presence of wrinkles across the entire surface of the film, likely due to biaxial compressive strain further verified by Raman spectroscopy. Stacking orders in h-BN with a clearly layered structure were confirmed by high resolution transmission electron microscopy, showing that the films have crystallographic homogeneity. Chemical analysis of the as-grown films was done by x-ray photoelectron spectroscopy, which confirmed the formation of stoichiometric h-BN films with excellent uniformity. This wafer-scale chemical vapor deposition-grown layered h-BN with 2D morphology facilitates applications in the fields of quantum- and deep ultraviolet-photonics.

A three-dimensional variational data assimilation system is implemented within the Weather Research and Forecasting/Chemistry model, and the control variables consist of eight species of the Model for Simulation Aerosol Interactions and Chemistry scheme. In the experiments, the three-dimensional profiles of aircraft speciated observations and surface concentration observations acquired during the California Research at the Nexus of Air Quality and Climate Change field campaign are assimilated. The data assimilation experiments are performed at 02:00 local time 2 June 2010, assimilating surface observations at 02:00 and aircraft observations from 01:30 to 02:30 local time. The results show that the assimilation of both aircraft and surface observations improves the subsequent forecasts. The improved forecast skill resulting from the assimilation of the aircraft profiles persists a time longer than the assimilation of the surface observations, which suggests the necessity of vertical profile observations for extending aerosol forecasting time.

A three-dimensional variational data assimilation scheme is designed for satellite AOD based on the IMPROVE (Interagency Monitoring of Protected Visual Environments) equation. The observation operator that simulates AOD from the control variables is established by the IMPROVE equation. All of the 16 control variables in the assimilation scheme are the mass concentrations of aerosol species from the Model for Simulation Aerosol Interactions and Chemistry scheme, so as to take advantage of this scheme in providing comprehensive analyses of species concentrations and size distributions as well as be calculating efficiently. The assimilation scheme can save computational resources as the IMPROVE equation is a quadratic equation. A single-point observation experiment shows that the information from the single-point AOD is effectively spread horizontally and vertically.

SIRT1 is the gene that codes for Sirtuin 1, an NAD (nicotinamide adenine dinucleotide)-dependent class III histone deacetylase. This gene plays a key role in adipose tissue and muscle development in animals. Chinese Luxi cattle (n = 169) were selected to identify SIRT1 SNPs (single nucleotide polymorphisms) and investigate the relationship of these SNPs with carcass traits. Five SNPs (g.-382G > A, g.-274C > G, g.17324T > C, g.17379A > G, and g.17491G > A) were identified by direct sequencing. SNPs g.-382G > A and g.-274C > G were located within the promoter region of this gene. SNP g.-382G > A was significantly associated with dressing percentage, meat percentage, and striploin and ribeye weights, and the g.-274C > G polymorphism had a strong effect on carcass, tenderloin, and high rib weights in Luxi cattle. These findings will provide possible clues for the biological roles of SIRT1 underlying beef cattle carcass traits.

The objective of this study was to evaluate the effect of diets containing different amounts of wheat, as a partial or whole substitute for corn, on digestibility, digestive enzyme activities, serum metabolite contents and ruminal fermentation in beef cattle. Four Limousin×LuXi crossbred cattle with a body weight (400±10 kg), fitted with permanent ruminal, proximal duodenal and terminal ileal cannulas, were used in a 4×4 Latin square design with four treatments: Control (100% corn), 33% wheat (33% substitution for corn), 67% wheat (67% substitution for corn), and 100% wheat (100% substitution for corn) on a dry matter basis. Balance constraints are important for background error covariance (BEC) in data assimilation to spread information between different variables and produce balance analysis fields. Using statistical regression, we develop a balance constraint for the BEC of aerosol variables and apply it to a three-dimensional variational data assimilation system in the WRF/Chem model; 1-month forecasts from the WRF/Chem model are employed for BEC statistics. The cross-correlations between the different species are generally high. The largest correlation occurs between elemental carbon and organic carbon with as large as 0.9. After using the balance constraints, the correlations between the unbalanced variables reduce to less than 0.2. A set of data assimilation and forecasting experiments is performed. In these experiments, surface PM2.5 concentrations and speciated concentrations along aircraft flight tracks are assimilated. The analysis increments with the balance constraints show spatial distributions more complex than those without the balance constraints, which is a consequence of the spreading of observation information across variables due to the balance constraints. The forecast skills with the balance constraints show substantial and durable improvements from the 2nd hour to the 16th hour compared with the forecast skills without the balance constraints. The results suggest that the developed balance constraints are important for the aerosol assimilation and forecasting.

The effects of three different feeding systems on beef cattle production performance, rumen fermentation, and rumen digesta particle structure were investigated by using 18 Limousin (steers) with a similar body weight (575±10 kg) in a 80-d experiment. The animals were equally and randomly divided into three treatment groups, namely, total mixed ration group (cattle fed TMR), SI1 group (cattle fed concentrate firstly then roughage), and SI2 group (cattle fed roughage firstly then concentrate). Abstract This study aimed to explore the diuretic activity of linalyl acetate (LA). LA is an essential oil, it is an integral phyto-constituent of various plants. In this study, acute and chronic diuretic activities were explored by measuring the levels of different electrolytes and pH in the urine of experimental rats. Rats were divided into five groups. The control group was given 10 mg/kg normal saline, the treated group was given 10 mg/kg furosemide, and the remaining 3 groups received different doses of LA including 25, 50, and 75 mg/kg through intraperitoneal route, to determine its diuretic potential. Urine volume for acute diuretic activity was measured for 6 hours however for chronic diuretic activity was measured for 6 days. For a comparative study of LA with a control group and treated group with reference drug, diuretic index was used. Moreover, the underlying mechanism of the diuretic activity was also explored by comparing atropine, L-NAME, and indomethacin. The results of each group with 6 rats in each group were obtained by ± standard error of the mean of every group. Analysis of Variance (ANOVA) was used for statistical analysis. Results revealed that the LA 75 mg/kg dose showed comparable results as of furosemide. Moreover, this study revealed the involvement of muscarinic receptors to produce diuresis in comparison with atropine with very little involvement of prostanoids and no effect on NO pathway induced by indomethacin and L-NAME respectively. It is concluded that LA possess anti-diuretic potential. Muscarinic receptors might be involved in producing diuretic effects.

INTRODUCTION: Physical activity is an essential element in our daily life that leads to long-term health benefits. Physical activity refers to movement of the body that requires energy. Body mass index (BMI) indicates a ratio of body weight to squared height, which is a useful health indicator. On the contrary, body composition describes the body by measuring percentages of fat and muscle in human bodies. MATERIALS AND METHODS: This cross-sectional study aimed to determine the relationship between physical activities, BMI and body composition among pre-university students from one of the universities in Selangor, Malaysia. Stratified random sampling was employed to recruit 70 pre-university students into this study. RESULTS: From the study, 50% of the respondents are categorized as minimally active. In addition, there is significant difference between the physical activity levels of male and female respondents. The relationship between physical activity and BMI indicates a very weak negative correlation. Similarly, the correlation between physical activity and fat mass is a weak negative relationship. Meanwhile, there is a weak positive correlation between physical activity and muscle mass. CONCLUSION: Therefore, it can be concluded that when physical activity increases, BMI and body fat mass will decrease, while muscle mass will increase. Moreover, it was shown that there was a significant relationship between physical activity and body composition. [ABSTRACT FROM AUTHOR]

We study the impact of the geomagnetic storm of 7–9 September 2017 on the low- to mid-latitude ionosphere. The prominent feature of this solar event is the sequential occurrence of two SYM-H minima with values of −146 and −115 nT on 8 September at 01:08 and 13:56 UT, respectively. The study is based on the analysis of data from the Global Positioning System (GPS) stations and magnetic observatories located at different longitudinal sectors corresponding to the Pacific, Asia, Africa and the Americas during the period 4–14 September 2017. The GPS data are used to derive the global, regional and vertical total electron content (vTEC) in the four selected regions. It is observed that the storm-time response of the vTEC over the Asian and Pacific sectors is earlier than over the African and American sectors. Magnetic observatory data are used to illustrate the variation in the magnetic field particularly, in its horizontal component. The global thermospheric neutral density ratio; i.e., O∕N2 maps obtained from the Global UltraViolet Spectrographic Imager (GUVI) on board the Thermosphere Ionosphere Mesosphere Energetics and Dynamics (TIMED) satellite are used to characterize the storm-time response of the thermosphere. These maps exhibit a significant storm-time depletion of the O∕N2 density ratio in the northern middle and lower latitudes over the western Pacific and American sectors as compared to the eastern Pacific, Asian and African sectors. However, the positive storm effects in the O∕N2 ratio can be observed in the low latitudes and equatorial regions. It can be deduced that the storm-time thermospheric and ionospheric responses are correlated. Overall, the positive ionospheric storm effects appear over the dayside sectors which are associated with the ionospheric electric fields and the traveling atmospheric disturbances. It is inferred that a variety of space weather phenomena such as the coronal mass ejection, the high-speed solar wind stream and the solar radio flux are the cause of multiple day enhancements of the vTEC in the low- to mid-latitude ionosphere during the period 4–14 September 2017.

Support layer of thin film composite (TFC) membrane plays an important role in forward osmosis (FO) performance. A new type of support layer or nanocomposite substrate was developed by incorporating titanium dioxide (TiO2)/graphene oxide (GO) into polysulfone (PSF) matrix. Prior to performance evaluation, the developed substrates were characterized with respect to surface chemistry, roughness and cross-sectional morphology. The results showed that both surface hydrophilicity and roughness of PSF-based substrates were increased upon incorporation of nanomaterials. Substrates with long finger-like voids extended from the top to the bottom could be developed upon incorporation of TiO2 (SubstrateTiO2) or TiO2/GO mixture (SubstrateTiO2/GO). The improved surface hydrophilicity and favorable structure formed are the main factors leading to higher water flux of nanocomposite substrate. Moreover, the water flux of FO using TFC membranes could be enhanced using this nanocomposite substrate. Comparing to the control TFC membrane, the TFC membranes made of SubstrateTiO2 and SubstrateTiO2/GO exhibited greater water flux with minimum increase in reverse draw solute flux. Based on the results obtained, it can be concluded that the incorporation of TiO2 and/or GO nanoparticles into PSF substrate could potentially improve the TFC membrane performance during FO applications. Introduction: Non invasive positive pressure ventilation (NIPPV) is the first line treatment for hypercapnic acute respiratory failure (ARF) secondary to COPD exacerbation in selected patients. Limited data exist supporting the use of fiberoptic bronchoscopy (FOB) during this clinical setting. The aim of this study is to assess the role of FOB during NIPPV in patients with decompensated COPD acute exacerbation. Methods: This study is a randomized prospective case control pilot study carried out on 50 patients - admitted to critical care units at Alexandria University Hospital, Egypt - suffering from hypercapnic ARF secondary to COPD exacerbation with Kelly Matthay Score from 2 to 4. All patients received NIPPV. Patients were divided randomly into 2 equal groups: group I (cases) (25 patients) was subjected to additional intervention of early FOB during the first 6–12 h from admission while group II (control) (25 patients) received the conventional treatment and NIPPV only. Outcome parameters measured were changes in ABG data, duration of NIPPV, rate of its success, ICU stay and mortality as well as the safety of FOB and possible complications. Results: No significant difference was detected between the 2 groups regarding the baseline characteristics. No serious complications happened from FOB, and Oxygen desaturation happened in 4/25 patients (16%), Tachycardia in 2/25 patients (8%). In group I, 23 patients (92%) were successfully weaned from NIPPV versus 16 patients (64%) in group II (p = 0.037). Total duration of NIPPV was 28.52 h in group I versus 56.25 h in group II (p = 0.001). Length of ICU stay was 4.84 days in group I versus 8.68 days in group II (p = 0.001). Only 1 patient died in group I versus 3 patients in group II (p = 0.609). Conclusion: The early application of FOB during NIPPV in patients with ARF due to COPD exacerbation was shown to be safe. Significant improvement in the outcome of patients who underwent FOB was noticed in terms of improved ABG data, shorter duration of NIPPV, higher percentage of success and shorter ICU stay while no significant difference was detected in mortality.

Underground goaf in metallurgical mining area leads to surface subsidence and has destructive effect on blast furnace. This paper studies the influence of underground goaf on blast furnace.The overlying strata and surface subsidence due to the formed gob area after the underground mining led to a different degree of destruction to blast furnace. Hence, blast furnace was modeled to theoretically analyze the additional stress applied to blast furnace body and foundation when withstanding the surface deformation. On this basis, the anti-deformation structure was designed to analyze the influences of Class I - IN surface deformation on the setting of Blast furnace body unit.

Ocean acidification (OA) is usually thought to change the speciation of trace metals and increase the concentration of free metal ions, hence elevating metal bioavailability. In this study, embryos of the oyster Crassostrea angulata and abalone Haliotis discus hannai were cultured under 4 pCO2 conditions (400, 800, 1500 and 2000 µatm) with Cu and Zn added. Fertilization rate was measured 2 h post-fertilization (hpf), while larval deformation and larval shell length were measured 24 hpf. Our results show that OA can alleviate Cu and Zn inhibition of C. angulata fertilization by 86.1 and 26.4% respectively, and Zn inhibition of H. discus hannai fertilization by 43.7%. However, OA enhanced the inhibitory effect of Cu on fertilization of H. discus hannai by 34.7%. OA enhanced the toxic effect of Cu on larval normality of C. angulata by 22.0% and the effect of Cu and Zn on larval normality of H. discus hannai by 71.4 and 37.2%, respectively. OA also enhanced the inhibitory effects of Cu and Zn on larval calcification in H. discus hannai by 8.8 and 8.6%, respectively. However, OA did not change the effect of Cu on the calcification of C. angulata larvae. OA decreased Zn inhibition of oyster larval calcification from 3.1 to 1.5%. Based on our results, the toxic effects of metal on early development of molluscs are not always increased by rising pCO2 and differ across developmental stages, egg structure and species. This complexity suggests that caution should be taken when carrying out multiple environmental stressor tests on molluscan embryos.

The distribution of the invasive aquatic plant Eichhornia crassipes is considered to be limited by winter survival. Therefore, winter warming, as well as characteristics of overwintering organs, are expected to affect its distribution and survival. An experiment was conducted to analyze the effects of winter warming and stem base treatments (size or burial) on winter survival, regrowth and thus performance of floating or rooted plants of E. crassipes using a simulated warming system in a greenhouse. Winter warming significantly increased the percentage survival in both forms of the plant and facilitated its regrowth and clonal propagation. Stem base size played an important role in determining survival, regrowth and vegetative propagation. Moreover, water cover and sediment burial of stem bases facilitated overwintering. After winter survival, a larger fraction of the biomass of E. crassipes was allocated to shoots. These results suggest that, with climate warming, the invasive ability of E. crassipes will be enhanced, with distribution spreading to the north into central and north China, favouring plants with large stem bases.

Autophagosome cancer vaccines can promote cross-presentation of multiple tumour antigens and induce cross-reactive T cell responses. However, so far, there is no effective method for obtaining a highly immunogenic autophagosomal cancer vaccine because autophagosomes, once formed, quickly fuse with lysosomes and cannot easily escape from cells. Here we report a functional Ti 2 NX nanodot that caps the autophagosome membrane lipid phosphatidylinositol-4-phosphate, blocking the fusion of autophagosomes with lysosomes and producing stable nanodot-coated autophagosomes in tumours. The formed nanodot-coated autophagosomes can escape from cancer cells to lymph nodes, where they activate tumour-specific T cells. We show that our approach reduces tumour burden and provide long-term immune surveillance protection for cured mice. The simultaneous occurrence of high temperatures and hypoxia events caused mass die-offs of aquatic animals. It is crucial to investigate the relationship between hypoxia tolerance and thermal tolerance of aquatic animals to predict the biological and ecological outcomes under global climate change scenarios. In this study, the hypoxia tolerance and thermal tolerance of Pacific abalone, Haliotis discus hannai, were measured by methods based on adhesion capacity (hypoxia adhesion duration and heat adhesion duration) and heart rate fluctuations (breakpoint of dissolved oxygen and Arrhenius breakpoint temperature). Weak correlations were found between hypoxia tolerance and thermal tolerance (Spearman correlation, r = -0.09, P = 0.2069; Pearson correlation, r = -0.04, P = 0.3313). Furthermore, a total of 21 significant SNPs and 19 candidate genes (such as cubn, lrp6, gria2, rft2, and casp8) were identified to be associated with hypoxia tolerance of Pacific abalone by conducting whole genome resequencing and genome-wide association study (GWAS). But there was no overlap between candidate genes associated with hypoxia tolerance and candidate genes associated with thermal tolerance, validating the weak correlation between hypoxia tolerance and thermal tolerance. This study highlights that individuals with greater hypoxia tolerance do not necessarily have greater thermal tolerance. Purpose: Mitigating tobacco-related disparities in the Appalachian region and rural areas is crucial. This study seeks to gauge cigarette smoking prevalence in Virginia counties, uncover rurality and Appalachian-linked disparities, and explore local drivers of these gaps., Method: A 2011-2019 Virginia BRFSS data were used to estimate county-level cigarette smoking rates in adults aged 18 or older. Counties were categorized as urban/rural and Appalachian/non-Appalachian, with a focus on rural-Appalachian. Disparities in cigarette smoking rates and associated factors were analyzed via the Blinder-Oaxaca decomposition method. The study assessed 4 dimensions of the Centers for Disease Control and Prevention's social vulnerability index (SVI): socioeconomic, minority status, household composition, and housing. Additionally, county-specific factors such as tobacco agriculture, physician availability, coal mining, and tobacco retailer density were examined., Findings: Rural areas exhibited a 6.18% higher cigarette smoking prevalence compared to urban areas (P<.001). SVI dimensions accounted for 53.2% of the disparity, county features explained 16.4%, and 30.4% remained unexplained. Appalachian areas had a 6.79% higher cigarette smoking prevalence than non-Appalachian areas (P<.001). SVI dimensions explained 51.4% of the disparity, county features accounted for 21.8%, leaving 26.8% unexplained. Rural-Appalachian areas showed a 7.8% higher cigarette smoking prevalence (P<.001). SVI dimensions contributed to 51.7% of the disparity, county features explained 9.6%, and 38.7% remained unexplained., Conclusions: Substantial disparities in cigarette smoking prevalence exist in underserved areas of Virginia, including rural, Appalachian, and rural-Appalachian regions. While SVI dimensions, physician availability, tobacco agriculture, and coal mining contribute, yet notable gaps remain unexplained. Aralia elata (Miq.) Seem, is an important cash crop in northeastern China. The tender shoots are rich in amino acids, vitamins, and trace elements, and the saponins of leaves and roots have antioxidant and immune-boosting properties. In August of 2023, root rot disease was observed in 4-year-old A. elata plants in Dongzhou County (41.79 °N, 124.11 °E), Liaoning Province, China, with an overall incidence of approximately 40%. While previous studies have described root rot in A. elata caused by Fusarium oxysporum (Zheng et al. 2024), this new root rot showed different symptoms. The leaves of the diseased plants were water-soaked and wilted but not chlorotic. The internal tissue of the roots was disintegrated and soft, while the xylem appeared brown and empty with only the epidermis remaining. Diseased root tissues were sterilized with 75% ethanol for 30 s, treated with 2% sodium hypochlorite for 2 min, and rinsed twice in sterile distilled water. Twelve isolates with similar morphological characteristics were obtained (isolation frequency of 80%), the isolates were plated on PDA medium and incubated at 25℃. Single-zoospore cultures were obtained and transferred to soybean agar medium (SA). After growth on SA for 7 days, the formation of rounded colonies of white aerial mycelia was observed. The sporangia were ovoid or ellipsoid, papillate, terminally borne, with an average length and width of 30.24 (18.35 to 45.39) × 24.16 (14.55 to 37.64) μm. The isolates were homothallic, produced plerotic oogonia with smooth walls with an average diameter of 27.97 (21.06 to 35.90 μm), and yielded one paragynous or anohiginous antheridium, sized 12.21 (10.11 to 14.72) μm × 9.75 (8.39 to 11.31) μm. Based on these morphological characteristics, the isolates were identified as part of the Phytophthora genus (Montealegre et al. 2016). The internal transcribed spacer (ITS) region and cytochrome oxidase subunit Ⅱ (COXⅡ) gene were amplified using the respective primers ITS1/ITS4 and FM66/FM58 (Wiseman et al. 2018). The ITS and COXⅡ sequences were compared with sequences in GenBank, showing 99.83% to 100% sequence identity with Phytophthora cactorum (accession nos. OR347848 and OR351295, respectively). The sequences have been deposited in GenBank with the accession numbers PP479774 (ITS) and PP489221 (COXⅡ). Construction of a maximum-likelihood phylogenetic tree of the identified sequences using MEGA-X indicated that the isolate could be identified as P. cactorum. To confirm pathogenicity, shallow wounds were made in the taproots of 10 one-year-old A. elata and were inoculated with four 9-mm diameter agar plugs from one-week-old oomycete cultures on SA agar per root, and the pathogenicity test was repeated twice. Another 10 wounded plants inoculated with agar plugs without oomycete served as the controls. The plants were planted in flowerpots filled with sterilized forest soil and cultured in a greenhouse at 25℃ with 14 h of light per day. After 20 days, the inoculated plants showed similar symptoms to those initially observed in the field. No symptoms of root rot were observed in the controls. The P. cactorum was re-isolated from the inoculated plants, and identified based on the morphology and ITS and COXⅡ sequence, fulfilling Koch's postulates. P. cactorum has been reported to cause root rot in A. elata in Korea and Japan (Lee et al. 2005; Uchida et al. 1984). To our knowledge, this is the first report of P. cactorum causing root rot in A. elata in China. P. cactorum is a serious threat to A. elata cultivation, and disease management strategies need to be developed.

Osteosarcoma (OS) is the most prevalent secondary sarcoma associated with retinoblastoma (RB). However, the molecular mechanisms driving the interactions between these two diseases remain incompletely understood. This study aims to explore the transcriptomic commonalities and molecular pathways shared by RB and OS, and to identify biomarkers that predict OS prognosis effectively. RNA sequences and patient information for OS and RB were obtained from the University of California Santa Cruz (UCSC) Xena and Gene Expression Omnibus databases. When RB and OS were first identified, a common gene expression profile was discovered. Weighted Gene Co-expression Network Analysis (WGCNA) revealed co-expression networks associated with OS after immunotyping patients. To evaluate the genes shared by RB and OS, univariate and multivariate Cox regression analysis were then carried out. Three machine learning methods were used to pick key genes, and risk models were created and verified. Next, medications that target independent prognostic genes were found using the Cellminer database. The comparison of differential gene expression between OS and RB revealed 1216 genes, primarily linked to the activation and proliferation of immune cells. WGCNA identified 12 modules related to OS immunotyping, with the grey module showing a strong correlation with the immune-inflamed phenotype. This module intersected with differential genes from RB, producing 65 RB-associated OS Immune-inflamed Genes (ROIGs). Analysis identified 6 hub genes for model construction through univariate Cox regression and three machine learning techniques. A risk model based on these hub genes was established, demonstrating significant prognostic value for OS. Genes shared between OS and RB contribute to the progression of both cancers through multiple pathways. The ROIGs risk score model independently predicts the overall survival of OS patients. Purpose: To investigate the utility of combining clinical and contrasted-enhanced tomography (CECT) parameters for the preoperative evaluation of perineural invasion (PNI) in gallbladder carcinoma (GBC)., Methods: A total of 134 patients with GBC (male/female, 52/82; age, 64.4 ± 9.7 years) were divided into PNI-positive (n = 63) and PNI-negative groups (n = 71). Clinical characteristics (demographic information, liver function indicators and tumor markers) and CECT parameters (tumor type, tumor size, gallbladder stone, invasion of gallbladder neck/cystic duct, clinical T stage and N stage) were collected and compared between two groups. Binary logistic regression analysis, receiver operating characteristic curves analyses and Delong test were used in further statistical analyses in clinical T3-4 stage (cT3-4) GBC patients. Overall survival (OS) rates after surgery were compared between PNI-negative group and PNI-positive group of cT3-4 GBC patients., Results: The majority of GBC patients with PNI were classified as cT3-4 (61/63, 96.8%), while only 3.2% (2/63) of PNI-positive cases were identified at cT1-2. Among cT3-4 GBC, OS was significantly lower in the PNI-positive group than the PNI-negative group after surgery (HR,1.661; 95% CI, 1.044-2.643; P = 0.032). Gender and gallbladder neck/cystic duct invasion were independent predictive factors for cT3-4 GBC patients with PNI. A combination of gender and gallbladder neck/cystic duct invasion showed the best diagnostic performance than that of individual parameters (all P < 0.05)., Conclusions: Preoperative T staging using CECT enables the initial assessment of PNI status in GBC patients. Background: Detecting and segmenting intracranial aneurysms (IAs) from angiographic images is a laborious task., Objective: To evaluates a novel deep-learning algorithm, named vessel attention (VA)-Unet, for the efficient detection and segmentation of IAs., Methods: This retrospective study was conducted using head CT angiography (CTA) examinations depicting IAs from two hospitals in China between 2010 and 2021. Training included cases with subarachnoid hemorrhage (SAH) and arterial stenosis, common accompanying vascular abnormalities. Testing was performed in cohorts with reference-standard digital subtraction angiography (cohort 1), with SAH (cohort 2), acquired outside the time interval of training data (cohort 3), and an external dataset (cohort 4). The algorithm's performance was evaluated using sensitivity, recall, false positives per case (FPs/case), and Dice coefficient, with manual segmentation as the reference standard., Results: The study included 3190 CTA scans with 4124 IAs. Sensitivity, recall, and FPs/case for detection of IAs were, respectively, 98.58%, 96.17%, and 2.08 in cohort 1; 95.00%, 88.8%, and 3.62 in cohort 2; 96.00%, 93.77%, and 2.60 in cohort 3; and, 96.17%, 94.05%, and 3.60 in external cohort 4. The segmentation accuracy, as measured by the Dice coefficient, was 0.78, 0.71, 0.71, and 0.66 for cohorts 1-4, respectively. VA-Unet detection recall and FPs/case and segmentation accuracy were affected by several clinical factors, including aneurysm size, bifurcation aneurysms, and the presence of arterial stenosis and SAH., Conclusions: VA-Unet accurately detected and segmented IAs in head CTA comparably to expert interpretation. Background: Interleukin-1β (IL-1β) plays a crucial role in cartilage degeneration by inducing inflammatory cascades in chondrocytes, impairing their normal biological functions. Long non-coding RNA NKILA (lncRNA NKILA) has been implicated in osteoarthritis (OA), but its specific molecular mechanisms remain unclear. This study aims to elucidate the function and molecular regulatory mechanism of lncRNA NKILA in articular chondrocytes under IL-1β stimulation., Methods: Primary human articular chondrocytes were cultured to investigate the effects of IL-1β on chondrocyte proliferation, apoptosis, and extracellular matrix metabolism. We employed Reverse Transcription Quantitative Polymerase Chain Reaction (RT-qPCR), western blot, flow cytometry, immunofluorescence, and nuclear mass separation assays to explore the interaction between lncRNA NKILA and the NFκB signaling pathway. Additionally, animal experiments were conducted to evaluate the therapeutic potential of modulating lncRNA NKILA expression in vivo., Results: IL-1β treatment led to decreased chondrocyte proliferation and increased apoptosis. Our study demonstrated that IL-1β downregulates lncRNA NKILA, which weakens its inhibitory effect on the NFκB (Nuclear Factor Kappa B) signaling pathway. This downregulation results in increased NFκB activity and exacerbates chondrocyte degeneration. Notably, the upregulation of lncRNA NKILA significantly alleviated OA symptoms, indicating that NKILA could be a promising therapeutic target., Conclusion: IL-1β reduces lncRNA NKILA expression, weakening its inhibition of NFκB signaling and promoting articular chondrocyte degeneration. Inflammation and extracellular matrix (ECM) degradation are two major factors involved in the pathogenesis of osteoarthritis (OA). Wedelolactone, a natural compound classified as a coumestan, is isolated from the medicinal plants Eclipta alba and Wedelia calendulacea. In this study, we assessed the protective effects of Wedelolactone on chondrocytes in OA. Our findings show that pretreatment with Wedelolactone effectively inhibited the IL-1β-induced upregulation of COX‑2, iNOS, TNF-α, and IL6 in chondrocytes, contributing to inflammation suppression. Moreover, pretreatment with Wedelolactone followed by IL-1β treatment significantly increased the expression of Collagen II and SOX9, while decreasing the expression of Adamts5, MMP1, MMP3, and MMP13, thereby promoting ECM protection. Through Network pharmacology Analysis, we identified 14 key targets that link Wedelolactone and OA. GO and KEGG pathway analysis suggested that Wedelolactone primarily impacted OA by targeting inflammatory responses, particularly the NF-κB signaling pathway. Further studies demonstrated Wedelolactone prevented IL-1β-induced activation of NF-κB signaling pathway by inhibiting the translocation of p65 and the preventing the degradation of IκBα in human chondrocytes. Molecular docking studies also indicated that Wedelolactone can directly bind to the NF-κB complex, thereby inhibited the nuclear localization of p65. In vivo experiments demonstrated that Wedelolactone can alleviate cartilage damage in DMM mice model. In summary, Wedelolactone appears to mitigate inflammation and cartilage degeneration by suppressing the NF-κB signaling pathway, thereby alleviating OA progression. Effective repair of large bone defects through bone tissue engineering (BTE) remains an unmet clinical challenge. Successful BTE requires optimal and synergistic interactions among biocompatible scaffolds, osteogenic factors, and osteoprogenitors to form a highly vascularized microenvironment for bone regeneration and osseointegration. We sought to develop a highly effective BTE system by using 3D printed citrate-based mPOC/hydroxyapatite (HA) composites laden with BMP9-stimulated human urine stem cells (USCs). Specifically, we synthesized and characterized methacrylate poly(1,8 octamethylene citrate) (mPOC), mixed it with 0%, 40% or 60% HA (i.e., mPOC-0HA, mPOC-40HA, or mPOC-60HA), and fabricated composite scaffold via micro-continuous liquid interface production (μCLIP). The 3D-printed mPOC-HA composite scaffolds were compatible with human USCs that exhibited high osteogenic activity in vitro upon BMP9 stimulation. Subcutaneous implantation of mPOC-HA scaffolds laden with BMP9-stimulated USCs revealed effective bone formation in all three types of mPOC-HA composite scaffolds. Histologic evaluation revealed that the mPOC-60HA composite scaffold yielded the most mature bone, resembling native bone tissue with extensive scaffold-osteointegration. Collectively, these findings demonstrate that the citrate-based mPOC-60HA composite, human urine stem cells, and the potent osteogenic factor BMP9 constitute a desirable triad for effective bone tissue engineering.

A machine learning (ML) strategy is suggested to optimize dual-layer oxide thin film transistor (TFTs) performance. In this study, Bayesian optimization (BO), an algorithm recognized for its efficiency in optimizing material design, is applied to guide the design of a channel layer composed of IZO and IGZO. The sputtering fabrication process, which has attracted attention as an oxide semiconductor channel layer deposition method, is fine-tuned using ML to enhance multiple electrical characteristics of transistors: field-effect mobility, threshold voltage, and subthreshold swing. Using BO, the sputtering conditions─plasma power, pressure, and gas ratio, which intricately influence device performance─were modified using 19 data sets of 84 scenarios. It reveals that the modulated process conditions improve field-effect mobility up to 46.7 cm 2 V -1 s -1 , achieving more than double the performance of conventional IGZO TFTs. Furthermore, it was observed that threshold voltage is optimized to zero voltage, and the subthreshold swing is considerably improved, contributing to reduced power consumption. This study demonstrates that leveraging ML to optimize TFTs design not only accelerates the design process but also improves device performance dramatically. Overall, this ML strategy manages complex correlations among process parameters, properties, and performance and sets a precedent for the expeditious optimization of semiconductor devices.

Exploiting solid powder fluorescence holds significant potential in diverse domains including medicine and forensics. Conventional fingerprint detection methods often fall short due to low contrast, sensitivity, and high toxicity. To addressing these challenges, we present a novel method for latent fingerprint detection using fluorescent carbon dots (CDs) encapsulated into conventional or mesoporous SiO 2 colloidal spheres (CD@SiO 2 or CDs@m-SiO 2 ) through a surface functionalization-assisted cooperative assembly process. The synthesized monodisperse CDs@SiO 2 and CDs@m-SiO 2 spheres, with tuning particle size, adjustable porosity and pore size, and highly dispersed CDs, exhibit improved fingerprint visibility and contrast on various substrates such as glass, stainless steel, and plastic. CDs located in SiO 2 with excellent affinity effectively avoids their solid-state self-quenching phenomenon, which, coupling with mesoporous SiO 2 shell, maximumly retains their fluorescence properties. Autophagy is a lysosome-dependent cellular degradation pathway that responds to a variety of environmental and cellular stresses, which is defective in aging and age-related diseases, therefore, targeting autophagy with small-molecule activators has potential therapeutic benefits. In this study, we successfully completed the first total synthesis of Ivesinol, an identified antibacterial natural product, and efficiently constructed a library of its analogs. To measure the effect of Ivesinol analogs on autophagic activity, we performed cell imaging-based screening approach, and observed that several Ivesinol analogs exhibited potent autophagy-regulating activity. Specifically, the derivative B2 significantly activated autophagy activity in concentration- and time-dependent manners, and even outperformed the commonly used activator Torin1 in activating autophagy in MCF-7 cells at 0.5 μM. Bioinformatics analysis showed that B2 treatment significantly impacted ubiquitin mediated proteolysis and AMPK signaling pathway, with functionally related gene sets displaying strong correlations. Based on these findings, we proposed that B2 activates autophagy by mechanisms involved in downregulation of key HSP70 family members, activation of the UPR, and ultimately leading to autophagy. Background: Patterns of arterial remodeling may be associated with outcomes in patients with severe middle cerebral artery (MCA) stenosis after endovascular treatment (EVT). This study aims to investigate the potential correlation between arterial remodeling patterns in patients with severe MCA stenosis, and plaque characteristics and procedure-related perforator stroke (PS)., Methods: Consecutive patients with MCA atherosclerotic disease who underwent EVT at the First Affiliated Hospital of Zhengzhou University from January 2018 to June 2023 were retrospectively enrolled in this study. The clinical and surgical data of the patients were evaluated. Remodeling index values >1.05, 0.95-1.05, and <0.95 indicated positive remodeling (PR), intermediate remodeling (IR), and negative remodeling (NR), respectively. Based on the remodeling index values calculated by high-resolution magnetic resonance imaging (HR-MRI), the arterial remodeling patterns were classified as NR or non-NR; non-NR was defined as both PR and IR. The primary endpoint was procedure-related PS. Plaque characteristics included the plaque area, plaque burden, contrast-enhancement ratio, and plaque distribution. Plaque features and the incidence of procedure-related PS were compared between the NR and non-NR groups. Differences in the categorical variables between the patients with NR and non-NR were assessed using the Chi-square test (χ 2 ) or Fisher's exact test. Differences in the continuous variables between the two groups were assessed using the Student t -test or Mann-Whitney U -test., Results: A total of 65 patients (mean age: 57.6 years) were enrolled in the study. Of the patients, 45 (69.2%) showed NR and 20 (30.8%) showed non-NR. The primary endpoint of procedure-related PS occurred in 3 patients (3/65, 4.6%). On HR-MRI, the entire cohort had a mean plaque area of 10.0 mm 2 , a mean plaque burden of 92.0 %, and a mean enhancement ratio of 1.3. Compared to the non-NR group, the NR group had a smaller plaque area (9.2 vs. 11.6 mm 2 , P=0.001). The probability of the primary end point was 15.0% in the non-NR group and 0.0% in the NR group (P=0.026)., Conclusions: PS after MCA elective stenting may be related to non-NR based on HR-MRI. Background: Immune checkpoint inhibitor (ICI) has been widely used in the treatment of advanced cancers, but predicting their efficacy remains challenging. Traditional biomarkers are numerous but exhibit heterogeneity within populations. For comprehensively utilizing the ICI-related biomarkers, we aim to conduct multidimensional feature selection and deep learning model construction., Methods: We used statistical and machine learning methods to map features of different levels to next-generation sequencing gene expression. We integrated genes from different sources into the feature input of a deep learning model, by means of self-attention mechanism., Results: We performed feature selection at the single-cell sequencing level, PD-L1 (CD274) analysis level, tumor mutational burden (TMB)/mismatch repair (MMR) level, and somatic copy number alteration (SCNA) level, obtaining 96 feature genes. Based on the pan-cancer dataset, we trained a multi-task deep learning model. We tested the model in the bladder urothelial carcinoma testing set 1 (AUC = 0.62, n = 298), bladder urothelial carcinoma testing set 2 (AUC = 0.66, n = 89), non-small cell lung cancer testing set (AUC = 0.85, n = 27), and skin cutaneous melanoma testing set (AUC = 0.71, n = 27)., Conclusion: Our study demonstrates the potential of the deep learning model for integrating multidimensional features in predicting the outcome of ICI. The Sm 3+ doped SrO-Nb 2 O 5 -Al 2 O 3 -Ga 2 O 3 -SiO 2 glasses in this work were prepared using the conventional melt quenching method. The effects of Al 2 O 3 /Ga 2 O 3 ratio on the structure and orange light emission properties were studied by XRD, Raman spectroscopy, spectrophotometer and J-O theory, respectively. With the increase of Al 2 O 3 content, the absorption coefficient of the glass sample gradually increases, which might be attributed to an increase in non-bridged oxygen bonds caused by a change in the glass network structure. Under 403 nm excitation, the emission spectra show clear peaks at 602 nm and 649 nm, representing the 4 G 5/2  →  6 H 7/2 , and 4 G 5/2  →  6 H 9/2 transitions, respectively. When the Al 2 O 3 /Ga 2 O 3 ratio is 0.25, the sample luminescence intensity is the highest, and the emission cross section of A2 glass sample is 4.34 × 10 -22  cm 2 . The CIE color coordinates, color purity, and color temperature values of all samples were determined, and they were all located in the orange-red light region. Persistent pulmonary hypertension of the newborn (PPHN) is a syndrome of acute respiratory failure characterized by systemic hypoxemia and elevated pulmonary arterial pressure, which leads to pathological changes in pulmonary vascular remodeling and endothelial cell function. Long non-coding RNA (lncRNA) H19 has been shown to be involved in the regulation of arterial endothelial cell function, but its regulatory role in PPHN is not fully understood. In the present study, mouse pulmonary artery endothelial cells (MPAECs) were cultured in a hypoxic conditions. Subsequently, the regulatory function of lncRNA H19 on MPAECs was explored by constructing adenoviruses knocking down and overexpressing lncRNA H19. The results revealed that the hypoxic conditions could induce the proliferation and migration of MPAECs, as well as the high expression of lncRNA H19 in MPAECs. Knockdown of lncRNA H19 expression in MPAECs reversed hypoxic environment-induced functional changes in endothelial cells, whereas overexpression of lncRNA H19 further enhanced the proliferation and migration of MPAECs. In addition, lncRNA H19 upregulated the hypoxia-inducible factor-1α (HIF-1α)/vascular endothelial growth factor (VEGF) pathway through sponge of miNA-20a-5p, which in turn promoted changes in endothelial cell function. LncRNA H19 may interfere with vascular remodeling in hypoxia-induced pulmonary hypertension by upregulating the expression of HIF-1α and VEGF in vascular endothelial cells., Competing Interests: Declarations. Background: Although optical coherence tomography (OCT) guidance with the 'MLDMAX' criteria is very useful for de-novo lesions during percutaneous coronary intervention (PCI), there are limited studies on its benefits in patients with in-stent restenosis (ISR). This study aimed to compare the clinical outcomes of patients with ISR who underwent repeat PCI (re-PCI) with OCT or angiographic guidance., Methods: This retrospective study enrolled 2142 patients with ISR who underwent re-PCI at Nanjing First Hospital from January 2016 to January 2023. The primary endpoint was the incidence of target vessel failure (TVF) post-re-PCI within 3 years. The secondary endpoints included each component of TVF., Results: After propensity score matching, 161 matched pairs were enrolled. OCT guidance of re-PCI was associated with a significantly lower risk of TVF compared with angiographic guidance alone [hazard ratio (HR), 0.51; 95% confidence interval (CI), 0.31-0.83; P = 0.007] in patients with ISR. However, only 68.9% of patients with ISR met the final criteria of 'MLDMAX' post-re-PCI, which was associated with a lower risk of TVF compared with patients without meeting the OCT criteria (HR, 0.24; 95% CI, 0.11-0.54; P < 0.001), and satisfied the angiographic criteria (HR, 0.40; 95% CI, 0.19-0.85; P = 0.017)., Conclusion: Compared with angiographic guidance, OCT guidance significantly reduced TVF risk following re-PCI for ISR lesions, especially for patients who met the final criteria of 'MLDMAX'., (Copyright © 2024 Wolters Kluwer Health, Inc. The structure and dynamics of ferroelectric domain walls are essential for polarization switching in ferroelectrics, which remains relatively unexplored in two-dimensional ferroelectric α-In 2 Se 3 . Interlayer interactions engineering via selecting the stacking order in two-dimensional materials allows modulation of ferroelectric properties. Here, we report stacking-dependent ferroelectric domain walls in 2H and 3R stacked α-In 2 Se 3 , elucidating the resistance switching mechanism in ferroelectric semiconductor-metal junction devices. In 3R α-In 2 Se 3 , the in-plane movement of out-of-plane ferroelectric domain walls yield a large hysteresis window. Conversely, 2H α-In 2 Se 3 devices favor in-plane domain walls and out-of-plane domain wall motion, producing a small hysteresis window. High electric fields induce a ferro-paraelectric phase transition of In 2 Se 3 , where 3R In 2 Se 3 reaches the transition through intralayer atomic gliding, while 2H In 2 Se 3 undergoes a complex process comprising intralayer bond dissociation and interlayer bond reconstruction.
