We constructed a new ecotoxicological tool to test for the presence of teratogens in rivers by exploiting the regenerative capabilities of Hydra vulgaris because using living organisms is considered good practice in environmental monitoring for detecting detrimental pressures. Outputs on the Hydra assay were integrated with two biotic indices based on benthic invertebrates and macrophytes to evaluate whether the assay acts as a tool for detecting generalised freshwater conditions or as a biological test model revealing only the effects of dissolved teratogens. Several hypostomae of H. vulgaris were reared in waters collected in diverse southern central Latium rivers, where physiochemical descriptors were surveyed for environmental characterisation. The regeneration rate and amount of aberrations were evaluated to obtain a teratogenic risk index (TRI) score and then compared with the control. TRI calculations showed 4 sites (18.18%) in Class I (no risk), 15 sites (68.18%) in Class II (low risk), 2 sites (9.09%) in Class III (moderate risk) and 1 site (4.55%) in Class IV (high risk). No Class V sites (very high risk) were found. No significant correlations emerged among physicochemical parameters (except for PO43–), environmental descriptors, Simpson diversity indices and biotic indices and the TRI. These results promote an integrated approach to improving links between biological and ecological responses to anthropogenic impacts. We explored a multilevel approach to providing a monitoring tool to detect teratogens dissolved in waters by extending the potential of tracking human-mediated detrimental environmental changes using overlapping information from short-term responses in an embryo toxicological assay based on freshwater polyps and two biotic indices based on plants and invertebrates. Vegetation management of archaeological sites is a recurrent problem when considering the conservation of historic structures and biodiversity. In the UNESCO site of the Etruscan necropolis "La Banditaccia" (Cerveteri, Italy), several plants grow on the ancient tumuli, which are subjected to different management practices. In order to assess the influence of interventions on plant colonisation, a comparative analysis of several tumuli was performed, through structural, chorological, phytosociological and statistical evaluations. This investigation identified and defined an ecological gradient among the tumuli. The cover values of Therophytes and Hemicryptophytes decreased by half with regular cutting, while those of Geophytes and Phanerophytes increased. Mesophilic field plants (MFP), both ephemeral and perennial, represented the most abundant group. Mediterranean bushes and forests (MBF) showed a positive trend from the entrance to the last tumuli of the necropolis. This trend was clearly defined by PCA, and it seems community changes were caused by the absence of mowing activities and to a lower tourist frequentation. SIMPER analysis highlighted which species on each tumulus mainly influence their dissimilarity. This knowledge, when combined with the evaluation of the potential risk of the different plant communities growing on the tumuli, can help to identify the best management practices. Since effects of alien invasive free-floating plants can be relevant in aquatic ecosystems, we investigated the non-native Lemna minuta impact on four aquatic animal groups: Hydra vulgaris (Coelenterates), Asellus aquaticus (Arthropods), Gambusia affinis (Fish), Bufo bufo tadpoles (Amphibians). An indoor experiment was conducted keeping animals in water held in tanks with L. minuta mats of 0.5 (WI1), 1.5 cm thick (WI2) and without mats (WOU). Water parameters (DO, DO%, pH) and animal responses (survival rate, vitality) were measured every 48 h (0–288 h). Treatments with mats showed significant impacts on animals which were more severe with increasing mat thickness. Strong decreasing of oxygen and pH associated with mat occurrence had a large impact on animals. In WI2 all individuals died within 144 h (H. vulgaris, B. bufo within 96h), while in WI1 there was a higher survival rate and vitality (excluding B. bufo died within 96 h) and in WOU no deaths. Running water habitats are among the most altered aquatic systems by human activities driving an increase in the organic components and the associated bacterial load as well. To contribute in improving the monitoring activities in running waters, here we tested the validity of the new Micro Biological Survey (MBS) method to specifically assess the bacterial load in running waters focusing on Total Viable Counts (at 22°C and 37°C) and Escherichia coli (at 44°C) in order to propose a new prognostic tool for watercourses. MBS method is an alternative colorimetric method for counting bacterial load in water and food samples that is easy to use and leads to a reliable and simple interpretation of results, being also faster and less expensive than traditional methods. Then, we compared MBS with the traditionally used reference method for the bacterial load, and with the most used biotic index for Italian watercourses based on the benthic invertebrates: the Extended Biotic Index (EBI). The last comparison was performed to validate the use of MBS in biomonitoring activities since the benthic invertebrate multi-species assemblage (and then EBI) alter own structure mainly depending on the organic component variation. During the first part of the study, the assessment of both linearity (regressions among bacterial concentrations) and accuracy (significant correlation between a measured value and a value used as reference) confirmed the validity of the MBS method. Second, the linear regressions between the three investigated microbial parameters vs. both physical-chemical descriptors and EBI, revealed the usefulness of MBS as a valid tool for routine microbiological analyses involved in rapid and easy field monitoring activities. Noninvasive methods in shell shape variation may help to understand evolution, ecology, stress and role of molluscan in aquatic ecosystems. Imaging analysis is a suitable diagnostic tool in morphological studies to (1) evaluate the health status of investigated animals, and (2) monitor sea coastal habitats. We introduce the feasibility of the cone-beam computed tomography as an optimal technique for 3D surface scanning to obtain virtual valve surfaces of Mytilus galloprovincialis, and analyze them exploiting the geometric morphometric facilities. Statistical output revealed morphological difference between mussels coming from different extensive rearing systems highlighting how the entire valve surface contributed to discriminate between groups when we compared 2-and 3D analyses. Many factors drive the morphological differences observed in the valve shape variation between the two sites, such as geographical genetic differentiation, natural environmental effects and culture conditions. The simplicity of the proposed methodology avoids damage and handling of individuals, makes this approach useful for morphological data collection, and helps to detect detrimental agents for sea ecosystems by using molluscans. The ability of mammals to feel noxious stimuli lies in a heterogeneous group of primary somatosensory neurons termed nociceptors, which express specific membrane receptors, such as the Transient Receptor Potential (TRP) family. Here, we show that one of the most important nociceptive-like pathways is conserved in the freshwater coelenterate Hydra vulgaris, the most primitive organism possessing a nervous system. In particular, we found that H. vulgaris expresses TRPM3, a nociceptor calcium channel involved in the detection of noxious heat in mammals. Furthermore, we detected that both heat shock and TRPM3 specific agonist (i.e., pregnenolone sulfate) induce the modulation of the heat shock protein 70 (HSP70) and the nitric oxide synthase (NOS), two genes activated by TRP-mediated heat painful stimuli in mammals. As expected, these effects are inhibited by a TRPM3 antagonist (i.e., mefenamic acid). Interestingly, the TRPM3 agonist and heat shock also induce the expression of nuclear transcription erythroid 2-related factor (Nrf2) and superoxide dismutase (SOD), known markers of oxidative stress; noteworthy gene expression was also inhibited by the TRPM3 antagonist. ABSTRACT River impoundments and waste water discharge are a serious threat to the integrity and biodiversity of river ecosystems, especially in central Italy. Benthic macroinvertebrates were sampled in autumn and summer along the Aniene River to assess the cumulative biological effect of the numerous dams and sewage treatment plants that affect its middle and lower course. We hypothesized that (i) increasing habitat impairment would promote the formation of nestedness in species assemblage, where species poor locations support only a sub-set of organisms from richer sites; (ii) specific life-history traits would confer sensitivity to habitat degradation. Patterns of macroinvertebrate richness and diversity along the river tracked the distribution of dams and sewage treatment plants. Partial Mantel test showed that dissimilarity in assemblages increased with the number of dams and treatment plants between reaches after controlling for longitudinal distance. Assemblages were significantly nested, and nestedness appeared related to both water quality gradients (phosphorous, turbidity) that reflected anthropogenic inputs, and to natural gradient in altitude. Reaches with nested assemblages (supporting a sub-set of the species pool) were characterized by greater representations of taxa with shorter life cycles, while, in contrast, species rich sites supported taxa with longer life cycles and lower dispersal ability. These results suggest that the cumulative effect of dams and sewage treatment plants promoted the formation of nested subsets in species distribution. Moreover, it appeared that certain functional traits that conferred sensitivity also dictated the progressive non-random loss of taxa in face of multiple anthropogenic stressors. These findings have conservation implications in the regions, but need to be considered preliminary since anthropogenic and natural factors co-varied systematically along the study river precluding the identification of single factor effects. The main aim of this study was to improve the knowledge about the concordance among macrophytes and macroinvertebrates to provide complementary information and facilitate the procedures for quality assessment of river ecosystems. Macrophytes and macroinvertebrates were collected in 11 sampling sites along a central Apennine calcareous river in October 2008 and June 2009. The concordance between the two biomonitoring groups was tested according to several environmental parameters. The comparison of data matrix similarities by Mantel test showed differences in the assemblage of macrophytes and macroinvertebrates along the river since correlation values were 0.04, p > 0.05 in October 2008 and 0.39, p > 0.05 in June 2009. The study revealed lack of concordance between the two groups, emphasizing that the information provided by macrophytes and macroinvertebrates does not overlap in terms of response to environmental parameters.
