Abstract Background The number of known inherited metabolic diseases (IMDs) has been expanding, and the rate of diagnosis is improving with the development of innovative approaches including next generation sequencing (NGS). However, a substantial proportion of IMDs remain undetected by traditional diagnostic approaches. We aim to highlight the spectrum of IMDs diagnosed by the Undiagnosed Diseases Network (UDN) and to learn from the UDN diagnostic processes that were able to detect IMDs. Methods We conducted a retrospective analysis of 757 UDN participants diagnosed from 2015 until 2023 using the cohort database, which were divided into a cohort with IMDs (n = 194; 27%) and a cohort whose phenotypes were not explained by an IMD (n = 563; 73%), based on the International Classification of Inherited Metabolic Disorders (ICIMD). Then, we divided the causes of the metabolic 194 diagnoses into seven groups that included all the ICIMD categories. We inspected which clinical and laboratory approaches contributed to a final UDN diagnosis. We also present a UDN case example from each group to highlight the diagnostic yields that resulted from combining newer diagnostic approaches in the UDN and illustrate potential pitfalls of current NGS methods. Results These 194 cases of IMDs included examples from 21/25 (84%) of the ICIMD categories. Of the UDN subjects 164/194 (85%) were diagnosed with IMDs through NGS. Conclusion The spectrum of IMDs detected in the UDN cohort is large and growing and appropriate use of newer multiple diagnostic approaches should further increase diagnosis of IMDs that are presently missed by the traditional laboratory screening methods.

9. Exploring the Experiences of Working Mothers Navigating the Kaleidoscope Career Post COVID-19 Pandemic: A Case Study

- Abstract
The attrition of working mothers in technology workplaces is a pressing issue that demands attention from IT, HR, and D&I leaders who strive to forge inclusive environments within predominantly male industries. Through this study, I illuminated the multifaceted challenges these working mothers faced within the technology sector, exploring their experiences and pinpointing reasons for their departure. I offer evidence-backed recommendations for organizations to retain and better support these invaluable employees struggling to stay employed as IT professionals. Through this holistic single case study, I grasped the nuances of working mothers' experiences during the COVID-19 pandemic, offering a comprehensive, rich perspective on female attrition and providing detailed insights into these four working mothers' experiences after leaving their careers. Using a holistic single case study design, I facilitated a deep dive into the specific experiences of working mothers in IT during the global pandemic crisis. NVivo version 14 further streamlined the organization, coding, and analysis process for data analysis. I efficiently unpacked a large volume of qualitative data to unveil common themes and patterns encapsulating these participants' experiences. I distilled the findings from the preliminary questionnaires and interviews into three primary themes: balancing work and family, career challenge sacrifice, and lack of career authenticity, aligning with the Kaleidoscope career model parameters of balance, challenge, and authenticity. This study revealed that over 50% of women under 35 left IT during the pandemic (Lynkova, 2023). These findings underscore the urgent need for creating inclusive workplaces that nurture innovation and drive business success, as Microsoft did during the pandemic (Boyd, 2023). Microsoft is an excellent example of an employer that supports diverse employee needs. During the pandemic, Microsoft leaders prioritized digitizing remote and hybrid workplaces to ensure employees could work efficiently from their location (Boyd, 2023). IT leaders should follow Microsoft's lead and prioritize building more inclusive workplaces that foster innovation and drive business success instead of forcing in-person work mandates that exclude marginalized employees. 10. Conflicting Understandings of Multicultural Society, Global World, and English: Multimodal Content Analysis of 5 Korean Elementary EFL Textbooks

- Abstract
In response to becoming a more multicultural society, the South Korean Ministry of Education has implemented a multicultural education policy since 2006 by promoting English as key for young learners' communicative competence in a global world. Despite numerous previous studies on government-led curriculum and textbooks for English as a foreign language (EFL) across many countries, little attention has been given to the cultural and linguistic aspects of primary-level EFL textbooks. Within the framework of intercultural communicative competence (ICC), this study investigated multicultural understandings, social values, and beliefs embedded in the reading passages of five Korean Grade 6 EFL textbooks. By employing multimodal content analysis, it also scrutinized what messages and meanings are constructed with semiotic devices and the extent of the textbooks to foster EFL students' ICC. Findings revealed that the textbooks (re)produce national pride and otherness and promote the idea of English for a White global world. The lack of interactive communication events in diverse settings was also found problematic in developing students' new identities as global citizens. The study argues for critical approaches to multicultural education and intercultural development in EFL education by reconceptualizing English as a mediator for developing intercultural contact with diverse others.

11. Thinking Queer with Vietnamese EFL Textbooks

- Abstract
Queer is an inclusive term that embraces Lesbian, Gay, Bisexual, Transgender, and Queer (LGBTQ+), among other sexual identities. Thinking queer examines how fixed meanings or knowledge, or a particular image or identity, can be challenged, unfixed, unlearned, inquired, and placed into new learnings and understandings. In this paper, we think queer together with terms, images, and dialogs in Vietnamese English as a Foreign Language (EFL) textbooks. We adapted the Critical Visual Discourse Analysis (CVDA) and methods of critique to guide our thinking with the two questions: 1) How are gender roles represented in Vietnamese EFL textbooks? and 2) Are there queer terms, images, dialogs represented in Vietnamese EFL textbooks? If so, how are they depicted? The findings revealed biases in gender roles and invisibility of queer representations. We conclude by offering pedagogical implications for future studies.

With multivalent ion batteries being considered viable post‐lithium energy storage technologies, the last few years have seen increasing interest in intercalation type cathode materials for Ca‐ion batteries (CIBs). Prussian blue analogues (PBAs) display many positive attributes such as open structural framework, large interstitial sites, and versatile transition metal redox activity. The PBA cathodes reported for CIBs often contain K‐ion or Na‐ion due to the syntheses involving the use of potassium or sodium hexacyanoferrate. It has been long overlooked whether the presence of K‐ion or Na‐ion in pristine PBAs could affect the Ca‐ion storage in the PBAs when assessing their CIB performance. Through a combined experimental and computational investigation, this work reports that when K+/Na+ is present in the pristine PBAs, K/Na intercalation is preferred by the PBA structure over Ca intercalation even in the Ca cells. Ca‐ion intercalation can be increased when the K+/Na+ content in the pristine PBAs is reduced, shifting from a K or Na‐dominated intercalation process to a Ca/K or Ca/Na co‐intercalation process. This work consolidates that PBAs are an interesting and versatile intercalation host for several ions, but investigating PBAs as intercalation cathodes requires careful consideration of all ions present in the battery system.

Autosomal dominant congenital disorder of glycosylation (CDG) type Iw (OMIM# 619714) is caused by a heterozygous mutation in the STT3A gene. Most CDGs have an autosomal recessive (AR) mode of inheritance, but several cases with an autosomal dominant (AD) form of an AR CDG have been recently identified. This report describes a 17-year-old male who was referred to the Undiagnosed Diseases Network (UDN) with a history of macrocephaly, failure to thrive, short stature, epilepsy, autism, attention-deficit/hyperactivity disorder, mild developmental delay, intermittent hypotonia, dysmorphic features, and mildly enlarged aortic root. Trio exome sequencing was negative. His biochemical workup included normal plasma amino acids, ammonia, acylcarnitine profile and urine organic and amino acids. His UDN genome sequencing (GS) identified a previously unreported de novo STT3A variant (c.1631A > G: p.Asn544Ser). This variant removes a glycosylation site and was predicted to be destabilizing by structural biology modeling. The patient was formally diagnosed by the UDN Metabolomics Core as having an abnormal transferrin profile indicative of CDG type Iw through metabolomic profiling. We report here an affected male with phenotypic, molecular, and metabolic findings consistent with CDG type Iw due to a heterozygous STT3A variant. This case highlights the importance of further testing of individuals with the phenotypic and metabolic findings of an AR disorder who are heterozygous for a single disease-causing allele and can be shown to have a new AD form of the disorder that represents clinical heterogeneity.

Purpose Loneliness is a negative emotional state which is common in later life. The accumulative effects of loneliness have a significant impact on the physical and mental health of older adults. We aim to qualitatively explore the experiences of loneliness in later life and identify relevant behaviours and indicators which will inform novel methods of loneliness detection and intervention. Methods We conducted 60 semi-structured interviews with people aged 65 and over between September 2022 and August 2023. Data were analysed using a reflective thematic approach with early theme development on NVIVO software. Results Three themes were identified from the experiences of loneliness in older adults. 1) Unique responses to loneliness, including crying, increased eating or drinking and sleep difficulties, 2) Age-related losses, such as networks, roles, and abilities to engage in activities reducing over time and 3) Individual differences in overcoming loneliness, where strategies such as keeping busy and adopting a positive mindset were impacted by motivation and mood of older adults. Conclusion Distinct signs and relevant factors to loneliness in later life have been identified which can be detected by future sensing technologies. Findings of this in-depth qualitative study highlight that loneliness is a subjective experience requiring a holistic and person-centred approach to detection and intervention.

19. Influence of physical activity measurement on the association between Life’s Essential 8 and incident cardiovascular disease in older women

Objective: The American Heart Association’s Life’s Essential 8 (LE8) metric includes self-reported physical activity as one of the metrics for assessing cardiovascular health. Self-reported physical activity is prone to misclassification, whereas accelerometer measures are less biased. We examined associations of LE8 and incident cardiovascular disease (CVD) using self-reported and accelerometer-measured physical activity. Methods: Participants in the Women’s Health Initiative (WHI) Objective Physical Activity and Cardiovascular Health Study (n = 4,243; mean age = 79 ± 7 years) with no CVD history completed the WHI physical activity questionnaire and the Community Healthy Activities Model Program for Seniors (CHAMPS) questionnaire prior to wearing a hip-worn accelerometer for up to seven days in 2012–2014. LE8 components (physical activity, diet, sleep, body mass index, smoking, blood pressure, non-HDL cholesterol, and glucose) were scored according to guidelines. Scores were created using five physical activity measures: WHI questionnaire (LE8WHI), CHAMPS (LE8CHAMPS), accelerometer-measured physical activity (LE8A), and sample quantiles of accelerometer-measured physical activity (LE8AQ) and daily steps (LE8STEPS). Hazard ratios (HR) for physician-adjudicated CVD were estimated using Cox regression. Results: 707 incident CVD events occurred over an average 7.5 years. Multivariable HRs (95 % CI) comparing women in the highest vs. lowest quartiles of LE8 scores were: LE8WHI = 0.53 (0.43–0.67), LE8CHAMPS = 0.47 (0.38–0.60), LE8A = 0.44 (0.36–0.56), LE8AQ = 0.44 (0.35–0.55), and LE8STEPS = 0.45 (0.35–0.57). Conclusions: The LE8-incident CVD association varies by physical activity measurement, however all methods showed reduced risk. Device-measures of physical activity may be more accurate in the LE8, but when impractical to implement, also support use of self-reported measures.

22. Twenty-first century marine climate projections for the NW European shelf seas based on a perturbed parameter ensemble

The northwest European shelf (NWS) seas are environmentally and economically important, and an understanding of how their climate may change helps with their management. However, as the NWS seas are poorly represented in global climate models, a common approach is to dynamically downscale with an appropriate shelf sea model. We develop a set of physical marine climate projections for the NWS. We dynamically downscale 12 members of the HadGEM3-GC3.05 perturbed parameter ensemble (approximately 70 km horizontal resolution over Europe), developed for UKCP18, using the shelf sea model NEMO CO9 (7 km horizontal resolution). These are run under the RCP8.5 high-greenhouse-gas-emission scenario as continuous simulations over the period 1990–2098. We evaluate the simulations against observations in terms of tides, sea surface temperature (SST), surface and near-bed temperature and salinity, and sea surface height. These simulations represent the state of the art for NWS marine projections. We project an SST rise of 3.11 °C (± 2σ = 0.98 °C) and a sea surface salinity (SSS) freshening of −1.01 (± 2σ = 0.93; on the (unitless) practical salinity scale) for 2079–2098 relative to 2000–2019, averaged over the NWS (approximately bounded by the 200 m isobar and excluding the Norwegian Trench, the Skagerrak and Kattegat), a substantial seasonal stratification increase (23 d over the NWS seas), and a general weakening of the NWS residual circulation. While the patterns of NWS changes are similar to our previous projections, there is a greater warming and freshening that could reflect the change from the A1B emissions scenario to the RCP8.5 concentrations pathway or the higher climate sensitivity exhibited by HadGEM3-GC3.05. Off the shelf, south of Iceland, there is limited warming, consistent with a reduction in the Atlantic Meridional Overturning Circulation and associated northward heat transport. These projections have been publicly released, along with a consistent 200-year present-day control simulation, to provide an evidence base for climate change assessments and to facilitate climate impact studies. For example, we illustrate how the two products can be used to estimate climate trends, unforced variability and the time of emergence (ToE) of the climate signals. We calculate the average NWS SST ToE to be 2034 (with an 8-year range) and 2046 (with a 33-year range) for SSS. We also discuss how these projections can be used to describe NWS conditions under 2 and 4 °C global mean warming (compared with 1850–1900), as a policy-relevant exemplar use case.

Abstract The Northwest European shelf experienced unprecedented surface temperature anomalies in June 2023 (anomalies up to 5 °C locally, north of Ireland). Here, we show the shelf average underwent its longest recorded category II marine heatwave (16 days). With state-of-the-art observation and modelling capabilities, we show the marine heatwave developed quickly due to strong atmospheric forcing (high level of sunshine, weak winds, tropical air) and weak wave activity under anticyclonic weather regimes. Once formed, this shallow marine heatwave fed back on the weather: over the sea it reduced cloud cover and over land it contributed to breaking June mean temperature records and to enhanced convective rainfall through stronger, warmer and moister sea breezes. This marine heatwave was intensified by the last 20-year warming trend in sea surface temperatures. Such sea surface temperatures are projected to become commonplace by the middle of the century under a high greenhouse gas emission scenario.

Summary: Background: Dual inhibition of cytotoxic T-lymphocyte associated protein 4 (CTLA-4) and programmed death ligand 1 (PD-L1) has been shown to be an effective treatment strategy in many cancers. We sought to determine the objective response rate of combination durvalumab (D) plus tremelimumab (TM) in parallel cohorts of patients with carefully selected rare cancer types in which these agents had not previously been evaluated in phase II trials and for which there was clinical or biological rationale for dual immune checkpoint inhibitor therapy to be active. Methods: We designed a multi-centre, non-blinded, open-label phase II basket trial with each of the following 8 rare cancers considered a separate phase II trial: salivary carcinoma, carcinoma of unknown primary (CUP) with tumour infiltrating lymphocytes and/or expressing PD-L1, mucosal melanoma, acral melanoma, osteosarcoma, undifferentiated pleomorphic sarcoma, clear cell carcinoma of the ovary (CCCO) or squamous cell carcinoma of the anal canal (SCCA). The primary objective was to evaluate the response rate of the combination of D and TM, and the secondary objectives were to evaluate the tolerability and safety of D and TM combination. Eligible patients had advanced, metastatic or recurrent, or unresectable cancer with no known life-prolonging treatment option, age ≥16 years, ECOG performance status 0 or 1. Patients received D (1500 mg IV) + TM (75 mg IV) on Day 1 q4 weeks for 4 cycles followed by D q4 weeks until disease progression. This trial is registered with ClinicalTrials.gov, NCT02879162. Findings: From December 14th, 2016, to August 14, 2019, 140 patients enrolled into seven cohorts. The rare melanoma cohorts were closed due to lack of accrual. Of the 140 patients enrolled, 138 were eligible, 138 were evaluable for toxicity and 128 (91%) were evaluable for response. Durable responses were noted in all cohorts except for osteosarcoma. The overall response rate for eligible patients was 16% (95% CI: 10–23%). The response rates in each cancer cohort were undifferentiated pleomorphic sarcoma 15% (n = 3/20; 95% CI 3–38%), salivary carcinoma 20% (n = 4/20; 95% CI: 6–44%), CUP 17% (n = 3/18; 95% CI 4–41%), SCCA 10% (n = 2/20; 95% CI 12–32%) and CCCO 21% (n = 8/39; 95% CI 9–37%). Grade 3/4 adverse events were rare, where 4 patients experienced grade 4 related events and39 patients experienced grade 3 events. Interpretation: Durvalumab + tremelimumab treatment resulted in meaningful responses in salivary carcinoma and CCCO and deserves further exploration in front-line studies. Background Ventricular repolarization time (ECG QT and JT intervals) is associated with malignant arrhythmia. Genome‐wide association studies have identified 230 independent loci for QT and JT; however, 50% of their heritability remains unexplained. Previous work supports a causal effect of lower serum calcium concentrations on longer ventricular repolarization time. We hypothesized calcium interactions with QT and JT variant associations could explain a proportion of the missing heritability. Methods and Results We performed genome‐wide calcium interaction analyses for QT and JT intervals. Participants were stratified by their calcium level relative to the study distribution (top or bottom 20%). We performed a 2‐stage analysis (genome‐wide discovery [N=62 532] and replication [N=59 861] of lead variants) and a single‐stage genome‐wide meta‐analysis (N=122 393, [European ancestry N=117 581, African ancestry N=4812]). We also calculated 2‐degrees of freedom joint main and interaction and 1‐degree of freedom interaction P values. In 2‐stage and single‐stage analyses, 50 and 98 independent loci, respectively, were associated with either QT or JT intervals (2‐degrees of freedom joint main and interaction P value

Background The long-term outcomes of COVID-19 hospitalisation in individuals with pre-existing airway diseases are unknown. Methods Adult participants hospitalised for confirmed or clinically suspected COVID-19 and discharged between 5 March 2020 and 31 March 2021 were recruited to the Post-hospitalisation COVID-19 (PHOSP-COVID) study. Participants attended research visits at 5 months and 1 year post discharge. Clinical characteristics, perceived recovery, burden of symptoms and health-related quality of life (HRQoL) of individuals with pre-existing airway disease (i.e., asthma, COPD or bronchiectasis) were compared to the non-airways group. Results A total of 615 out of 2697 (22.8%) participants had a history of pre-existing airway diseases (72.0% diagnosed with asthma, 22.9% COPD and 5.1% bronchiectasis). Detecting and clearing legacy landmines, Improvised Explosive Devices (IED), and Unexploded Ordnances (UXO) using a force made up of humans or animals is extremely risky, labour- and time-intensive. It is crucial to quickly map millions of buried landmines/IDE/UXO, and remove them at a reasonable cost to minimise potential risks and make this labour-intensive task easier. Using unmanned vehicles and robots outfitted with various remote sensing modalities appears to be the ideal way to carry out this task in a non-invasive manner while employing a geophysical investigative method. In this study, a small-scale customised drone – the so-called Maggy – was developed to simplify and automate the procedures of cleaning explosive devices. It was instrumented with innovative intelligent automated techniques and magnetometer sensor technologies. Maggy’s performance was assessed in field tests conducted in Latvia and the United Kingdom. The outcomes, obtained in the open-air minefields and the benchmark assessments, verify the viability of the technologies, methods, and approaches integrated into Maggy for the efficient and economical detection of legacy landmines and IDE/UXO. This research provides the related research community with fundamental design and implementation parameters (e.g. flight speed, flight altitude) in building and using magnetometer-integrated Unmanned Aerial Systems (UAS). The improved versions of the developed easy-to-use compact technology are aimed to be deployed by humanitarian demining teams to expedite their clearing operations safely and efficiently.

38. An exploration of interpersonal touch in relation to mental health difficulties

"Nothing eases suffering like human touch" - Bobby Fischer. The sense of touch is one of the first senses humans develop (Ward Platt, 2006). It connects us to our physical environment and allows us to explore and learn from our surroundings, whilst being receptive to changes in temperature, texture, pressure and speed (Jones, 2016; Lederman, 1982; O'Shaughnessy, 1989). Tactile input is detected by receptors on the skin, transmitting that information via the central nervous system for final reception by the brain (Abraira & Ginty, 2013). In addition to our physical environment, touch also connects us to our social environment, defined by the interpersonal and social connections we make. In the context of our social environment, touch is a form of non-verbal communication with a role to facilitate communication of emotions, social bonding, attachments and intimacy (Hertenstein et al., 2006). A specific type of touch, linked to social bonding and social connection, is called affective touch. Affective touch is a slow, gentle, stroke-like motion that activates specific Ctactile (CT) afferents on hairy skin (Nordin, 1990). Activation of the CT afferents is associated with greater perceived pleasantness of touch (Löken et al., 2009). Experiences and attitudes to touch can vary and are influenced by past experiences, such as how much physical affection was given in childhood, and current experiences, such as touch from friends and family (Trotter et al., 2018). The perception of touch, regarding how pleasant it is perceived, also varies and will be influenced by experiences of touch. For example, individuals with low exposure to touch, rate touch as less pleasant (Sailer & Ackerley, 2019) and individuals with anxious attachment styles, rate touch as less pleasant (Krahé et al., 2018). Other factors can influence the perception of touch, such as mental health difficulties. For example, individuals with anorexia or autism, perceive touch as less pleasant (Croy et al., 2016; Crucianelli et al., 2016). Despite differences in perception of touch, the therapeutic benefits of it have been widely reported. Social touch has been shown to buffer stress and produce calming effects (Morrison, 2016). Affective touch specifically has also been shown to produce calming effects through decreased heart rate, in infants and adults (Fairhurst et al., 2014; Pawling et al., 2017; Triscoli et al., 2017). Interpersonal touch, in the form of massage therapy, has been shown to yield therapeutic benefits such as a reduction in pain, distress, aggression and stress (Diego et al., 2002; Hernandez-Reif et al., 1998; Hernandez-Reif et al., 2001). Within a mental health setting, touch has been shown to be reassuring, comforting and increase social connection between nurses and clients (Gleeson & Higgins, 2009). Ethics of touch and the use of touch in mental health settings need to be considered, for example how this impacts the power dynamics within a therapeutic relationship, specifically considering whether the use of touch is clinically appropriate, how touch may be interpreted by a client and the potential for sexual exploitation of clients (Zur, 2007). Depression and anxiety are common mental health difficulties that have been shown to benefit from touch. Field (2002) concluded that individuals with greater intimacy with their parents had lower depression scores and individuals with less intimacy with their parents had higher depression scores. Massage therapy has already been shown to reduce depression through decreases in cortisol and serotonin levels, and increased dopamine levels (Field et al., 2005) and reduce anxiety through decreases in salivary cortisol (Field et al., 1996). The evidence of positive effects of touch in common mental health difficulties could be used to inform treatment and intervention. This thesis explores interpersonal touch in relation to mental health difficulties. Chapter I is a systematic review of the literature around touch-based interventions for anxiety and depression. The review aims to provide an updated synthesis of the impact of massage therapy on anxiety and/or depression, and the direction of this relationship. Chapter II provides an empirical study exploring the association between depressive symptoms and the perception of affective touch. This study was conducted online and used videos depicting social touch to evaluate the impact of depressive symptoms on perceived pleasantness of social touch.

39. Low Macular Pigment Optical Density Is Associated with Manifest Primary Open-Angle Glaucoma in Older Women

Background: Lower density of carotenoids lutein and zeaxanthin (L/Z) in the macula (i.e., macular pigment) has been linked to greater risk for age-related eye disease. Objectives: We evaluated whether macular pigment optical density (MPOD) was associated with manifest primary open-angle glaucoma (POAG) among older women in the Carotenoids in Age-Related Eye Disease Study 2 (CAREDS2). Methods: MPOD was measured with customized heterochromatic flicker photometry in women who attended CAREDS2 (2016–2019) and CAREDS1 (2001–2004) study visits. Manifest POAG at CAREDS2 was assessed using visual fields, disc photos, optical coherence tomography, and medical records. Age-adjusted linear and logistic regression models were used to investigate the cross-sectional association between POAG and MPOD at CAREDS2, and MPOD measured 15 years earlier at CAREDS1. Results: Among 426 CAREDS2 participants (mean age: 80 y; range: 69–98 y), 26 eyes with manifest POAG from 26 participants were identified. Glaucomatous eyes had 25% lower MPOD compared to nonglaucomatous eyes [mean (SE): 0.40 (0.05) compared with 0.53 (0.01)] optical density units (ODU), respectively (P = 0.01). Compared with MPOD quartile 1, odds for POAG were lower for women in quartiles 2–4 (P-trend = 0.01). After excluding eyes with age-related macular degeneration, associations were similar but not statistically significant (P-trend = 0.16). Results were similar for MPOD measured at CAREDS1. Conclusions: Our results add to growing evidence that low MPOD may be a novel glaucoma risk factor and support further studies to assess the utility of dietary interventions for glaucoma prevention.

Demand for natural gas continues to climb in the United States, having reached a record monthly high of 104.9 billion cubic feet per day (Bcf/d) in November 2023. Hydraulic fracturing, a technique used to extract natural gas and oil from deep underground reservoirs, involves injecting large volumes of fluid, proppant, and chemical additives into shale units. This is followed by a “shut-in” period, during which the fracture fluid remains pressurized in the well for several weeks. The microbial processes that occur within the reservoir during this shut-in period are not well understood; yet, these reactions may significantly impact the structural integrity and overall recovery of oil and gas from the well. To shed light on this critical phase, we conducted an analysis of both pre-shut-in material alongside production fluid collected throughout the initial production phase at the Hydraulic Fracturing Test Site 2 (HFTS 2) located in the prolific Wolfcamp formation within the Permian Delaware Basin of west Texas, USA. Specifically, we aimed to assess the microbial ecology and functional potential of the microbial community during this crucial time frame. Prior analysis of 16S rRNA sequencing data through the first 35 days of production revealed a strong selection for a Clostridia species corresponding to a significant decrease in microbial diversity. Here, we performed a metagenomic analysis of produced water sampled on Day 33 of production. This analysis yielded three high-quality metagenome-assembled genomes (MAGs), one of which was a Clostridia draft genome closely related to the recently classified Petromonas tenebris. This draft genome likely represents the dominant Clostridia species observed in our 16S rRNA profile. Annotation of the MAGs revealed the presence of genes involved in critical metabolic processes, including thiosulfate reduction, mixed acid fermentation, and biofilm formation. These findings suggest that this microbial community has the potential to contribute to well souring, biocorrosion, and biofouling within the reservoir. Our research provides unique insights into the early stages of production in one of the most prolific unconventional plays in the United States, with important implications for well management and energy recovery.

Background: Patterns of physical activity and sedentary behavior among postmenopausal women are not well characterized. Objectives: To describe the patterns of accelerometer-assessed physical activity and sedentary behavior among postmenopausal women. Design: Cross-sectional study. Methods: Women 63–97 years (n = 6126) wore an ActiGraph GT3X + accelerometer on their hip for 1 week. Latent class analysis was used to classify women by patterns of percent of wake time in physical activity and sedentary behavior over the week. Results: On average, participants spent two-thirds of their day in sedentary behavior (62.3%), 21.1% in light low, 11.0% in light high, and 5.6% in moderate-to-vigorous physical activity. Five classes emerged for each single-component model for sedentary behavior and light low, light high, and moderate-to-vigorous physical activity. Six classes emerged for the multi-component model that simultaneously considered the four behaviors together. Conclusion: Unique profiles were identified in both single- and multi-component models that can provide new insights into habitual patterns of physical activity and sedentary behavior among postmenopausal women. Implications: The multi-component approach can contribute to refining public health guidelines that integrate recommendations for both enhancing age-appropriate physical activity levels and reducing time spent in sedentary behavior.

Background Pre-existing cardiovascular disease (CVD) or cardiovascular risk factors have been associated with an increased risk of complications following hospitalisation with COVID-19, but their impact on the rate of recovery following discharge is not known.Objectives To determine whether the rate of patient-perceived recovery following hospitalisation with COVID-19 was affected by the presence of CVD or cardiovascular risk factors.Methods In a multicentre prospective cohort study, patients were recruited following discharge from the hospital with COVID-19 undertaking two comprehensive assessments at 5 months and 12 months. Patients were stratified by the presence of either CVD or cardiovascular risk factors prior to hospitalisation with COVID-19 and compared with controls with neither. Full recovery was determined by the response to a patient-perceived evaluation of full recovery from COVID-19 in the context of physical, physiological and cognitive determinants of health.Results From a total population of 2545 patients (38.8% women), 472 (18.5%) and 1355 (53.2%) had CVD or cardiovascular risk factors, respectively. Compared with controls (n=718), patients with CVD and cardiovascular risk factors were older and more likely to have had severe COVID-19. Full recovery was significantly lower at 12 months in patients with CVD (adjusted OR (aOR) 0.62, 95% CI 0.43 to 0.89) and cardiovascular risk factors (aOR 0.66, 95% CI 0.50 to 0.86).Conclusion Patients with CVD or cardiovascular risk factors had a delayed recovery at 12 months following hospitalisation with COVID-19. Targeted interventions to reduce the impact of COVID-19 in patients with cardiovascular disease remain an unmet need.Trail registration number ISRCTN10980107.

Abstract Heart rate variability (HRV) is a cardiac autonomic marker with predictive value in cardiac patients. Ultra-short HRV (usHRV) can be measured at scale using standard and wearable ECGs, but its association with cardiovascular events in the general population is undetermined. We aimed to validate usHRV measured using ≤ 15-s ECGs (using RMSSD, SDSD and PHF indices) and investigate its association with atrial fibrillation, major adverse cardiac events, stroke and mortality in individuals without cardiovascular disease. In the National Survey for Health and Development (n = 1337 participants), agreement between 15-s and 6-min HRV, assessed with correlation analysis and Bland–Altman plots, was very good for RMSSD and SDSD and good for PHF. In the UK Biobank (n = 51,628 participants, 64% male, median age 58), after a median follow-up of 11.5 (11.4–11.7) years, incidence of outcomes ranged between 1.7% and 4.3%.
