Abstract

This study focuses on how catalyst concentration, activator concentration, and initial polymerization temperature impact in‐situ anionic ring‐opening polymerization (AROP) of ω‐lauryllactam. Catalyst and activator roles are fulfilled by NaH and toluene‐2,4‐diisocyanate (TDI), respectively, with varying catalyst/activator ratios to assess the influence of a bifunctional activator on the polymerization process. The materials produced undergo a thorough analysis, with a specific emphasis on solidification time. The examination extends to scrutinizing how different concentrations of catalyst/activator and polymerization temperatures affect crucial physical and chemical parameters. The study identifies NaH‐6 mol%/TDI‐3 mol% as the optimal formulation for solidification among the three explored temperatures. Notably, at 180 and 200°C, PA12 exhibits enhanced monomer conversion when a catalyst/activator ratio of 1.7 or 2 is applied. These findings underscore the significant impact of the catalyst/activator ratio and their individual concentrations on the polymer's final properties. This influence extends to factors such as crystallinity, polymer chain regularity, and dynamic mechanical properties. Abstract

The ADP-ribosylation factors (ARFs) and ARF-like (ARL) GTPases serve as essential molecular switches governing a wide array of cellular processes. In this study, we used proximity-dependent biotin identification (BioID) to comprehensively map the interactome of 28 out of 29 ARF and ARL proteins in two cellular models. Through this approach, we identified ~3000 high-confidence proximal interactors, enabling us to assign subcellular localizations to the family members. Notably, we uncovered previously undefined localizations for ARL4D and ARL10. Clustering analyses further exposed the distinctiveness of the interactors identified with these two GTPases. We also reveal that the expression of the understudied member ARL14 is confined to the stomach and intestines. We identified phospholipase D1 (PLD1) and the ESCPE-1 complex, more precisely, SNX1, as proximity interactors. Functional assays demonstrated that ARL14 can activate PLD1 in cellulo and is involved in cargo trafficking via the ESCPE-1 complex. Overall, the BioID data generated in this study provide a valuable resource for dissecting the complexities of ARF and ARL spatial organization and signaling. [ABSTRACT FROM AUTHOR]

Abstract

Necrotizing enterocolitis (NEC) is a life-threatening condition for premature infants in neonatal intensive care units. Finding indicators that can predict NEC development before symptoms appear would provide more time to apply targeted interventions. In this study, stools from 132 very-low-birth-weight (VLBW) infants were collected daily in the context of a multi-center prospective study aimed at investigating the potential of fecal biomarkers for NEC prediction using proteomics technology. Eight of the VLBW infants received a stage-3 NEC diagnosis. Stools collected from the NEC infants up to 10 days before their diagnosis were available for seven of them. Their samples were matched with those from seven pairs of non-NEC controls. The samples were processed for liquid chromatography-tandem mass spectrometry analysis using SWATH/DIA acquisition and cross-compatible proteomic software to perform label-free quantification. ROC curve and principal component analyses were used to explore discriminating information and to evaluate candidate protein markers. A series of 36 proteins showed the most efficient capacity with a signature that predicted all seven NEC infants at least a week in advance. Overall, our study demonstrates that multiplexed proteomic signature detection constitutes a promising approach for the early detection of NEC development in premature infants. Abstract

Airborne mineral aerosols emitted in high-latitude regions can impact radiative forcing, biogeochemical cycling of metals, and local air quality. The impact of dust emissions in these regions may change rapidly, as warming temperatures can increase mineral dust production and source regions. As there exists little research on mineral dust emissions in high-latitude regions, we have performed the first study of the physico-chemical properties of mineral dust emitted from a sub-Arctic proglacial dust source, using a method tailored to the remote conditions of the Canadian North. Soil and aerosol samples (PM10 and deposited mineral dust) were collected in May 2018 near the Ä'äy Chù (Slims River), a site exhibiting strong dust emissions. WHO air quality thresholds were exceeded at several receptor sites near the dust source, indicating a negative impact on local air quality. Notably, temporally averaged particle size distributions of PM10 were very fine as compared to those measured at more well-characterized, low-latitude dust sources. In addition, mineralogy and elemental composition of ambient PM10 were characterized; PM10 elemental composition was enriched in trace elements as compared to dust deposition, bulk soil samples, and the fine soil fractions (d < 53 µm). Finally, through a comparison of the elemental composition of PM10, dust deposition, and both fine and bulk soil fractions, as well as of meteorological factors measured during our campaign, we propose that the primary mechanisms for dust emissions from the Ä'äy Chù Valley are the rupture of clay coatings on particles and/or the release of resident fine particulate matter. Abstract

Summary AXL is activated by its ligand GAS6 and is expressed in triple-negative breast cancer cells. In the current study, we report AXL expression in HER2-positive (HER2 + ) breast cancers where it correlates with poor patient survival. Using murine models of HER2 + breast cancer, Axl, but not its ligand Gas6, was found to be essential for metastasis. We determined that AXL is required for intravasation, extravasation, and growth at the metastatic site. We found that AXL is expressed in HER2 + cancers displaying epithelial-to-mesenchymal transition (EMT) signatures where it contributes to sustain EMT. Interfering with AXL in a patient-derived xenograft (PDX) impaired transforming growth factor β (TGF-β)-induced cell invasion. Last, pharmacological inhibition of AXL specifically decreased the metastatic burden of mice developing HER2 + breast cancer. Abstract

Cancer cells exploit the epithelial-to-mesenchymal transition (EMT) program to become metastatic. Cytoskeletal regulators are required in mesenchymal cells where they promote EMT and EMT-induced migration. In a search for regulators of metastasis, we conducted shRNA screens targeting the microtubule plus-end tracking proteins (+TIPs). We show that the +TIP ACF7 is essential both for the maintenance of the EMT program and to promote migration. We find that the E3 ubiquitin ligase HectD1 promotes ACF7-proteasome-mediated degradation. Depletion of HectD1 stabilized ACF7, and this enhanced EMT and migration. Decreased HectD1 expression increased metastases in mouse models and conferred increased resistance to the cytotoxic drug cisplatin. A retrospective analysis of biopsies from breast cancer patients also reveals a correlation between higher ACF7 or lower HectD1 expression with poor clinical outcomes. Together, these results suggest that the control of ACF7 levels by HectD1 modulates EMT and the efficiency of metastasis. [ABSTRACT FROM AUTHOR]

Abstract

The Poincaré Seminar is held twice a year at the Institute Henri Poincaré in Paris. The goal of this seminar is to provide up-to-date information about general topics of great interest in physics. Both the theoretical and experimental results are covered, with some historical background. Particular care is devoted to the pedagogical nature of the presentation. This volume is devoted to Einstein's 1905 papers and their legacy. After a presentation of Einstein's epistemological approach to physics, and the genesis of special relativity, a centenary perspective is offered. The geometry of relativistic spacetime is explained in detail. Single photon experiments are presented, as a spectacular realization of Einstein's light quanta hypothesis. A previously unpublished lecture by Einstein, which presents an illuminating point of view on statistical physics in 1910, at the dawn of quantum mechanics, is reproduced. The volume ends with an essay on the historical, physical and mathematical aspects of Brownian motion. Abstract

Reactions of [Rh( κ2- O, O-acac)(PMe3)2] (acac=acetylacetonato) and α,ω-bis(arylbutadiynyl)alkanes afford two isomeric types of MC4 metallacycles with very different photophysical properties. As a result of a [2+2] reductive coupling at Rh, 2,5-bis(arylethynyl)rhodacyclopentadienes ( A) are formed, which display intense fluorescence ( Φ=0.07-0.54, τ=0.2-2.5 ns) despite the presence of the heavy metal atom. Rhodium biphenyl complexes ( B), which show exceptionally long-lived (hundreds of μs) phosphorescence ( Φ=0.01-0.33) at room temperature in solution, have been isolated as a second isomer originating from an unusual [4+2] cycloaddition reaction and a subsequent β-H-shift. We attribute the different photophysical properties of isomers A and B to a higher excited state density and a less stabilized T1 state in the biphenyl complexes B, allowing for more efficient intersystem crossing S1→Tn and T1→S0. Control of the isomer distribution is achieved by modification of the bis- (diyne) linker length, providing a fundamentally new route to access photoactive metal biphenyl compounds. Abstract

Background: Necrotizing enterocolitis (NEC) is the most frequent life-threatening gastrointestinal disease experienced by premature infants in neonatal intensive care units. The challenge for neonatologists is to detect early clinical manifestations of NEC. One strategy would be to identify specific markers that could be used as early diagnostic tools to identify preterm infants most at risk of developing NEC or in the event of a diagnostic dilemma of suspected disease. As a first step in this direction, we sought to determine the specific gene expression profile of NEC. Methods: Deep sequencing (RNA-Seq) was used to establish the gene expression profiles in ileal samples obtained from preterm infants diagnosed with NEC and non-NEC conditions. Data were analyzed with Ingenuity Pathway Analysis and ToppCluster softwares. Results: Data analysis indicated that the most significant functional pathways over-represented in NEC neonates were associated with immune functions, such as altered T and B cell signaling, B cell development, and the role of pattern recognition receptors for bacteria and viruses. Among the genes that were strongly modulated in neonates with NEC, we observed a significant degree of similarity when compared with those reported in Crohn's disease, a chronic inflammatory bowel disease. Conclusions: Gene expression profile analysis revealed a predominantly altered immune response in the intestine of NEC neonates. Moreover, comparative analysis between NEC and Crohn's disease gene expression repertoires revealed a surprisingly high degree of similarity between these two conditions suggesting a new avenue for identifying NEC biomarkers.
