Novel asymmetrical Fe(III) Schiff base coordination complexes, with the N4O2 coordination environment, exhibiting abrupt spin crossover (SCO) behaviour with a wide hysteresis loop of 27 K, have been synthesised from an ethylenediamine based asymmetrical Schiff base ligand viz. N-(2-aminoethyl)salicylaldimine (saen) and characterised via single crystal XRD measurements, variable temperature magnetic studies and Mössbauer measurements. The single crystal XRD measurements prove that the compound is a mixture of two fractions, one with methanol and the other without methanol, viz, [Fe(saen)2]NO3·CH3OH at 299 K (1–299) and at 173 K (1–173) and [Fe(saen)2]NO3 at 173 K (2–173), confirming that the Fe(III) centre has a distorted octahedral coordination geometry having two deprotonated asymmetrical tridentate saen ligands, counterbalanced by a nitrate anion and methanol solvent molecule in 1 while 2 has a non-solvate form. An extended hydrogen bonding network between the amino hydrogen atoms of the saen ligand, nitrate counteranion, and methanol solvate molecule is observed in fraction 1, while an extended zig-zag hydrogen bonding between the nitrate anion and amino hydrogens of saen is observed in fraction 2. Furthermore, the complex has been characterised by variable temperature magnetic susceptibility measurements and 57Fe Mössbauer spectroscopy, confirming the SCO nature. [ABSTRACT FROM AUTHOR]

L-Carnitine (LC) is known to play key roles in lipid metabolism and antioxidative activity, implicating enhanced cryotolerance of bovine blastocysts. However, sustainability of LC supplementation during culture period on preimplantation development beyond the blastocyst stage has not been investigated so far. Therefore, all embryos were cultured under fatty acid-free conditions, one group with LC (LC embryos) and the control group without LC (control) supplementation. Transfer to recipients was conducted on day 6. Elongation-stage embryos were recovered on day 14; metrics of embryo recollection, developmental rates as regards early elongation-stage as well as mean embryo length did not differ between the groups. Gene expression analyses via NGS revealed 341 genes to be differentially regulated between elongation-stage embryos derived from LC supplementation compared to controls. These played mainly a role in molecular functions and biological processes like oxidoreductase activity, ATP-dependent activity, cellular stress, and respiration. Pathways like oxidative phosphorylation and thermogenesis, extracellular matrix receptor signaling, PI3K-Akt, and focal adhesion were affected by differentially regulated genes. Moreover, all DEGs located on the mitochondria were significantly downregulated in LC embryos, being in line with lower mitochondrial copy number and mtDNA integrity compared to the control group. Finally, we uncovered alterations of the bioenergetic profile on day 7 as a consequence of LC supplementation for the first time, revealing significantly higher oxygen consumption rates, ATP linked respiration and spare capacity for LC embryos. In summary, we uncovered direct effects of LC supplementation during the culture period on the bioenergetic profile along with sustainable effects on mtDNA copy numbers and transcriptome profile of bovine day 14 embryos. A new iron(III) complex (Et3NH)2[Fe(L)2](ClO4)·MeOH (1) where H2L = 2-{(E)-[2-hydroxyphenyl)imino]methyl}phenol has been synthesised and characterised by single crystal XRD, elemental analysis and DC magnetic susceptibility measurements. The dianionic ligands L2− coordinate in a tridentate fashion with the Fe(III) through their deprotonated phenolic oxygens and azomethine nitrogen atoms, resulting in a trans-FeO4N2 chromophore. Variable-temperature magnetic measurements were performed between 300 and 5 K under an applied field of 0.1 T and show that 1 is in the high spin state (S = 5/2) over the whole measured temperature range. This is confirmed by Mössbauer spectroscopy at 77 and 300 K. [ABSTRACT FROM AUTHOR]

A skewed male-to-female ratio in cattle is believed to be due to the biased embryo losses during pregnancy. The changes in biochemical secretion such as miRNAs by the embryo due to altered maternal environment could cause a sex biased selective implantation resulting in a skewed male to female ratio at birth. Nevertheless, it is still not clear whether the male and female embryos could modify their miRNA expression patterns differently in response to altered physiological developmental conditions. Therefore, this study was focused on identifying sex specific miRNA expression patterns induced in the embryo during the elongation period in response to the maternal environment. For this, in vitro produced day female and male embryos were transferred to Holsteins Frisian cows and heifers. The elongated female and male embryos were then recovered at day 13 of the gestation period. Total RNA including the miRNAs was isolated from each group of elongated embryo samples were subjected to the next generation miRNA sequencing. Sequence alignment, identification and quantification of miRNAs were done using the miRDeep2 software package and differential miRNA expression analyses were performed using the edgeR bioconductor package. The recovery rate of viable elongating embryos at day 13 of the gestation period was 26.6%. In cows, 2.8 more viable elongating male embryos were recovered than female embryos, while in heifers the sex ratio of the recovered elongating embryos was close to one (1.05). The miRNA analysis showed that 254 miRNAs were detected in both male and female elongated embryos developed either in cows or heifers, of which 14 miRNAs including bta-miR-10b, bta-miR-148a, bta-miR-26a, and bta-miR-30d were highly expressed. Moreover, the expression level of 32 miRNAs including bta-let-7c, bta-let-7b, bta-let-7g, bta-let-7d and bta-let-7e was significantly different between the male and female embryos developed in cows, but the expression level of only 4 miRNAs (bta-miR-10, bta-mR-100, bta-miR-155 and bta-miR-6119-5p) was different between the male and female embryos that were developed in heifers. Furthermore, 19 miRNAs including those involved in cellular energy homeostasis pathways were differentially expressed between the male embryos developed in cows and heifers, but no significantly differentially expressed miRNAs were detected between the female embryos of cows and heifers. Thus, this study revealed that the sex ratio skewed towards males in embryos developed in cows was accompanied by increased embryonic sexual dimorphic miRNA expression divergence in embryos developed in cows compared to those developed in heifers. - Abstract
Post calving metabolic stress reduces the fertility of high producing dairy cows possibly by altering the expression of genes in the maternal environment via epigenetic modifications. Therefore, this study was conducted to identify endometrial DNA methylation marks that can be associated with pregnancy outcomes in postpartum cows at the time of breeding. For this, twelve days post-calving, cows were either offered a control diet or supplemented daily with rumen-protected methionine. Cows showing heat 50-64 days postpartum were artificially inseminated. Endometrial cytobrush samples were collected 4-8 h after artificial insemination and classified based on the pregnancy out comes as those derived from cows that resulted in pregnancy or resulted in no pregnancy. The DNAs isolated from endometrial samples were then subject to reduced representative bisulfite sequencing for DNA methylation analysis. Results showed that in the control diet group, 1,958 differentially methylated CpG sites (DMCGs) were identified between cows that resulted in pregnancy and those that resulted in no pregnancy of which 890 DMCGs were located on chr 27: 6217254-6225600 bp. A total of 537 DMCGs were overlapped with 313 annotated genes that were involved in various pathways including signal transduction, signalling by GPCR, aldosterone synthesis and secretion. Likewise, in methionine supplemented group, 3,430 CpG sites were differentially methylated between the two cow groups of which 18.7% were located on Chr27: 6217254-6225600 bp. A total of 1,781 DMCGS were overlapped with 890 genes which involved in developmental and signalling related pathways including WNT-signalling, focal adhesion and ECM receptor interaction. Interestingly, 149 genes involved in signal transduction, axon guidance and nonintegrin membrane-ECM interactions were differentially methylated between the two cow groups irrespective of their feeding regime, while 453 genes involved in axon guidance, notch signalling and collagen formation were differentially methylated between cows that received rumen protected methionine and control diet irrespective of their fertility status. Overall, this study indicated that postpartum cows that could potentially become pregnant could be distinguishable based on their endometrial DNA methylation patterns at the time of breeding. Abstract Background African animal trypanosomosis, transmitted cyclically by tsetse flies or mechanically by other biting flies, causes serious inflictions to livestock health. This study investigates the extent of non-tsetse transmitted animal trypanosomosis (NTTAT) by Trypanosoma (T.) evansi and T. vivax in domestic animals in the tsetse-free regions of Northern Ethiopia, Afar and Tigray. Methods A cross sectional study was conducted on 754 dromedary camels, 493 cattle, 264 goats, 181 sheep, 84 donkeys, 25 horses and 10 mules. The microhaematocrit centrifugation technique was used as parasitological test. Plasma was collected for serodiagnosis with CATT/T.evansi and RoTat 1.2 immune trypanolysis (ITL) while buffy coat specimens were collected for molecular diagnosis with T. evansi type A specific RoTat 1.2 PCR, T. evansi type B specific EVAB PCR and T. vivax specific TvPRAC PCR. Results The parasitological prevalence was 4.7% in Tigray and 2.7% in Afar and significantly higher (z = 2.53, p = 0.011) in cattle (7.3%) than in the other hosts. Cotton varieties that are high yielding and resistant to pests are required to improve production and productivity and to capitalize on the crop's enormous potential and its critical role in Ethiopia's expanding textile industry. Lack of improved cotton technology has forced farmers to recycle local varieties for ages which have become very susceptible to pests which are the major causes of very low productivity and quality of cotton in the country. Among major pests, bollworms (Helicoverpa armigera and Pectinophora gossypiella) account for 36–60% of yield losses. In the absence of genetically resistant or tolerant varieties, genetically engineered bollworm-resistant Bacillus thuringiensis (Bt) cotton has offered a great opportunity to reduce crop losses from bollworms. The objective of the study was to evaluate the efficacy of bollworm resistance and adaptability of Bt cotton varieties across cotton growing environments in Ethiopia and provide recommendations. Two Bt cotton hybrids (JKCH 1947 and JKCH 1050), one Bt OPV (Sudan), and three OPV conventional varieties (Weyito 07, Stam-59A, and Deltapine-90) were evaluated at seven different agro-ecologies using a randomized complete block design (RCBD) with three replications. Results showed significant differences among genotypes for yield and other traits. Hybrids JKCH 1947 and JKCH 1050 were the top high yielders under high and mild bollworm infestations, with mean seed cotton yield of 3.10 t·ha
−1 each and lint yield of 1.20 and 1.19 t·ha−1 , respectively, whereas the standard check Deltapine-90 (popular variety) recorded a mean seed cotton and lint yield of 2.3 t·ha−1 and 0.8 t·ha−1 , respectively. Combined analysis showed that genotypes, environment, and the genotypes × environment interactions had a highly significant effect (P < 0.05) on fiber quality. Weyito 07 and the two hybrids (JKCH 1947 and JKCH 1050) had upper half mean fiber lengths in the range of 27.78 to 32.11 mm. For fiber strength, genotypes Weyito 07, JKCH 1050, Stam-59A, and JKCH 1947 had 33.50 g/tex, 28.59 g/tex, 28.00 g/tex, and 27.75 g/tex, respectively. The fiber quality values of the hybrids were within acceptable limits, with staple lengths ranging from 27.78 to 28.44 mm and fiber strengths ranging from 27.75 to 28.59 g/tex. Results show potential adaptation of the hybrids under different cotton growing environments and their superior yield performance due also to added protection of yield losses from damage by bollworms. The contrast is bigger under high insect pressure conditions due to the genetically engineered Bt trait compared to the conventional varieties. The effective field resistance against bollworms in most locations shows that wider use of these hybrids can enhance cotton productivity and quality in Ethiopia. [ABSTRACT FROM AUTHOR]- Published
