36. SMAC-armed oncolytic virotherapy enhances the anticancer activity of PD1 blockade by modulating PANoptosis.

Abstract

Background: Oncolytic viruses (OVs) are increasingly recognized as promising tools for cancer therapy, as they selectively infect and destroy tumor cells while leaving healthy cells unharmed. Despite considerable progress, the limited therapeutic efficacy of OV-based virotherapy continues to be a significant challenge in cancer treatment. Methods: The SMAC/DIABLO gene was inserted into the genome of vesicular stomatitis virus (VSV) to generate VSV-S. Head and neck squamous cell carcinoma (HNSCC) cell lines and orthotopic mouse models were employed for research. Morphological changes were observed using both light microscopy and transmission electron microscopy. Molecular alterations were analyzed through Western blotting and ELISA kits. The tumor secretome was characterized using a combination of biotinylation and LC-MS analysis. Immune cell changes were evaluated by flow cytometry and immunohistochemistry. Results: Compared to its parental virus, VSV-S not only increases apoptosis by overexpressing SMAC during VSV infection but also triggers elevated levels of PANoptosis (pyroptosis, apoptosis, and necroptosis) in HNSCC cells via activation of caspase-1/gasdermin D (GSDMD) signaling. As a result, VSV-S-induced PANoptosis promotes CD8+ T cell tumor infiltration and enhances their cytotoxic capacity, eventually potentiating T cell-mediated antitumor immunity. Moreover, VSV-S reduces PDL1 levels in HNSCC cells and, in combination with PD1 blockade, produces a more potent antitumor effect than either therapy alone. Conclusions: Our findings demonstrate that the combination of VSV-S and PD1 blockade offers a synergistic therapeutic strategy for HNSCC, supporting the advancement of VSV-based virotherapy as a promising strategy to improve outcomes for HNSCC patients. Abstract

Head and neck cancer (HNC) represents a heterogeneous group of malignancies with increasing global incidence and notable mortality. Early detection is essential for improving survival rates and minimizing recurrence; however, existing diagnostic methods are often invasive and complex. There is a need for noninvasive and more effective approaches for early detection and real-time monitoring of HNC. Saliva contains various biomolecules that may serve as indicators of HNC. As a result, saliva-based biomarkers have emerged as a transformative approach in the diagnosis and treatment of HNC due to their ease of collection, non-invasiveness, and potential to provide details about biomolecular changes associated with cancer progression. This narrative review synthesizes the current literature on the potential of saliva as a noninvasive diagnostic tool for HNC. It highlights various biomarkers found in saliva, including cell-free DNA, RNA, proteins, and metabolites, and explores emerging technologies in saliva detection that could transform the future of HNC management. Continued research efforts and larger-scale validation studies are essential to fully realize the potential of saliva-based biopsy and help pinpoint notable biomarkers to improve patient outcomes and reduce mortality associated with HNC worldwide. Abstract

Background: The purpose of this study was to provide further insights into whether age and/or sex are associated with prognosis in oral tongue squamous cell carcinoma. Methods: This was a retrospective cohort study utilizing hospital registry data from 2006 to 2016 obtained from the National Cancer Database. Identified patients were divided into various cohorts based on age, sex, and staging. A descriptive analysis was performed using chi‐square tests and overall survival rates were estimated using Kaplan–Meier method. Results: A total of 17 642 patients were included in the study. The 5‐year overall survival rates were 82.0% (95% CI: 79.8%–84.0%) in younger patients versus 67.5% (95% CI: 66.7%–68.3%, p‐value <0.0001) older patients. The median overall survival for females was 143.4 months (95% CI: 133.2–NA) versus 129.8 (95% CI: 125.4–138.7, p‐value <0.0001) in males. Conclusions: Our analysis suggests that younger age and female sex are both predictors of improved survival in oral tongue squamous cell carcinoma. [ABSTRACT FROM AUTHOR]

Abstract

Background: This study assesses the prevalence of sexually transmitted infections (STIs) in first time visitors to the STIs clinic in Hangzhou, China, considering different genders, ages and symptoms. And also explores howthe COVID‐19 pandemic has affected on STIs. Methods: From 2019 to 2023, 27,283 first time visitors were tested for nine distinct STIs, including Human Papillomavirus (HPV), Human Immunodeficiency Virus (HIV), syphilis, Herpes Simplex Virus type 2 (HSV‐2), Ureaplasma urealyticum (UU), Chlamydia trachomatis (CT), Neisseria gonorrhoeae (NG), Mycoplasma genitalium (MG), and vaginal Candida. Results: Symptomatic male and female visitors showed overall STI‐positive rates of 39.27% and 59.20%, respectively(p <.001). The top three pathogens in both genders were HPV (47.56% and 56.71%), UU (29.21% and 56.47%), and HSV‐2 (22.41% and 52.94%). Among asymptomatic visitors, the total STI‐positive rate was 36.63% in males and 52.03% in females. Age‐stratified analysis revealed higher STI rates in visitors ≤ 20 or >50 years, regardless of gender and symptoms. During the COVID‐19 pandemic, symptomatic visitors showed lower positive rates for HPV, HIV, syphilis, and HSV‐2, while Candida, UU, CT, NG, and multiple infections increased. Among asymptomatic visitors, HPV had the lowest positive rate, while NG and multiple infections increased during the pandemic. Conclusion: STI prevalence is notably high, particularly in those aged ≤ 20 and >50 years. It emphasizes the need for enhanced health education, condom use, and vaccination.
