The anti-Parkinson monoamine oxidase (MAO)-B inhibitor rasagiline (Azilect) was shown to possess neuroprotective activities, involving the induction of brain-derived- and glial cell line-derived neurotrophic factors (BDNF, GDNF). Employing conventional neurochemical techniques, transcriptomics and proteomic screening tools combined with a biology-based clustering method, we show that rasagiline, given chronically post-MPTP (N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine), exerts neurorescue/neurotrophic activity in mice midbrain dopamine neurons. Rasagiline induced the activation of cell signaling mediators associated with neurotrophic factors responsive-tyrosine kinase receptor (Trk) pathway including ShcC, SOS, AF6, Rin1 and Ras and the increase in the Trk-downstream effector phosphatidylinositol 3 kinase (PI3K) protein. Confirmatory Western and immunohistochemical analyses indicated activation of the substrate of PI3K, Akt and phosphorylative inactivation of glycogen synthase kinase-3β and Raf1. Thus, the activation of Ras-PI3K-Akt survival pathway may contribute to rasagiline-mediated neurorescue effect. It is interesting to determine whether a similar effect is seen in parkinsonian patients after long-term treatment with rasagiline.

TVP1022, the S-enantiomer of rasagiline (Azilect®) (N-propargyl-1R-aminoindan), exerts cyto/cardio-protective effects in a variety of experimental cardiac and neuronal models. Previous studies have demonstrated that the protective activity of TVP1022 and other propargyl derivatives involve the activation of p42/44 mitogen-activated protein kinase (MAPK) signaling pathway. In the current study, we further investigated the molecular mechanism of action and signaling pathways of TVP1022 which may account for the cyto/cardio-protective efficacy of the drug. Using specific receptor binding and enzyme assays, we demonstrated that the imidazoline 1 and 2 binding sites (I(1) & I(2)) are potential targets for TVP1022 (IC(50) =9.5E-08 M and IC(50) =1.4E-07 M, respectively). Western blotting analysis showed that TVP1022 (1-20 µM) dose-dependently increased the immunoreactivity of phosphorylated p42 and p44 MAPK in rat pheochromocytoma PC12 cells and in neonatal rat ventricular myocytes (NRVM). This effect of TVP1022 was significantly attenuated by efaroxan, a selective I(1) imidazoline receptor antagonist. In addition, the cytoprotective effect of TVP1022 demonstrated in NRVM against serum deprivation-induced toxicity was markedly inhibited by efaroxan, thus suggesting the importance of I(1)imidazoline receptor in mediating the cardioprotective activity of the drug. Our findings suggest that the I(1)imidazoline receptor represents a novel site of action for the cyto/cardio-protective efficacy of TVP1022.

In in vitro fertilization (IVF) treatments, early identification of embryos with high implantation potential is required for shortening time to pregnancy while avoiding clinical complications to the newborn and the mother caused by multiple pregnancies. Current classification tools are based on morphological and morphokinetic parameters that are manually annotated using time‐lapse video files. However, manual annotation introduces interobserver and intraobserver variability and provides a discrete representation of preimplantation development while ignoring dynamic features that are associated with embryo quality. A fully automated and standardized classifiers are developed by training deep neural networks directly on the raw video files of >6200 blastulation‐labeled and >5500 implantation‐labeled embryos. Prediction of embryo implantation is more accurate than the current state‐of‐the‐art morphokientic classifier. Embryo classification improves with video length where the most predictive images show only partial association with morphological features. Deep learning substitute to human evaluation of embryo developmental competence thus contributes to implementing single embryo transfer methodology.

AbstractAccumulating evidence suggests that many cytotoxic signals occurring in the neurodegenerative brain can initiate neuronal death processes, including oxidative stress, inflammation, and accumulation of iron at the sites of the neuronal deterioration. Neuroprotection by iron chelators has been widely recognized with respect to their ability to prevent hydroxyl radical formation in the Fenton reaction by sequestering redox-active iron. An additional neuroprotective mechanism of iron chelators is associated with their ability to upregulate or stabilize the transcriptional activator, hypoxia-inducible factor-1α (HIF-1α). HIF-1α stability within the cells is under the control of a class of iron-dependent and oxygen-sensor enzymes, HIF prolyl-4-hydroxylases (PHDs) that target HIF-1α for degradation. Thus, an emerging novel target for neuroprotection is associated with the HIF system to promote stabilization of HIF-1α and increase transcription of HIF-1–related survival genes, which have been reported to be regulated in patient's brains afflicted with diverse neurodegenerative diseases. In accordance, a new potential therapeutic strategy for neurodegenerative diseases is explored, by which iron chelators would inhibit PHDs, target the HIF-1–signaling pathway and ultimately activate HIF-1–dependent neuroprotective genes. Test anxiety interferes with the ability to communicate knowledge during tests. Preparatory test anxiety may lead to excessive learning that reduces concentration and leads to mistakes and ineffective learning. This preliminary study developed the Preparatory Test Anxiety Questionnaire (PTAQ), tested its reliability and construct validity, and assessed whether students with preparatory test anxiety exhibit excessive learning. Additionally, it explored the relations between the subscales and the total scores of the PTAQ and the TAI. Sample included 364 undergraduate college psychology students. Results indicated that the PTAQ is comprised of the ‘cognition-emotion’ and ‘behavior’ subscales and has an adequate internal consistency and construct validity. The subscales and total score of the PTAQ were positively correlated with those of the TAI. Regression analysis showed that the Emotion-Cognition scale of the PTAQ significantly added to the explained variance, while the Behavior scale did not. Results are discussed with regard to preparatory test anxiety.
