In this study, different mass concentrations (0%, 1%, 2%, 3%, and 4%, based on starch mass) of tamarind seed polysaccharides (TSP) were added to endow potato starch (PS) bread with gluten-like properties in order to improve the processing performance and texture quality of gluten-free bread. The results indicated that adding TSP increased the peak viscosity of PS, improve its viscoelasticity, and endow it with gluten-like network structural characteristics. With increasing TSP concentration, the baking loss rate of gluten-free bread significantly decreased from 26.01% to 20.36%, and the specific volume increased from 1.27 mL/g to a maximum value of 3.19 mL/g. The addition of TSP significantly improved the texture characteristics of bread, as evidenced by a reduction in its hardness, cohesiveness, chewiness and resilience, resulting in a softer texture. Adding TSP delayed the staling of bread, thereby extending the shelf life. Principal component analysis (PCA) showed strong correlations between bread quality and baking performance and gelatinization characteristics of PS. This study has expanded the application of TSP in food field and provides new ideas and a theoretical basis for the development of gluten-free starch products.

To address the issues of significant object discrepancies, low similarity, and image noise interference between source and target domains in object detection, we propose a supervised learning approach combined with knowledge distillation. Initially, student and teacher models are jointly trained through supervised and distillation-based approaches, iteratively refining the inter-model weights to mitigate the issue of model overfitting. Secondly, a combined convolutional module is integrated into the feature extraction network of the student model, to minimize redundant computational effort; an explicit visual center module is embedded within the feature pyramid network, to bolster feature representation; and a spatial grouping enhancement module is incorporated into the region proposal network, to mitigate the adverse effects of noise on the outcomes. Ultimately, the model undergoes a comprehensive optimization process that leverages the loss functions originating from both the supervised and knowledge distillation phases. The experimental results demonstrate that this strategy significantly boosts classification and identification accuracy on cross-domain datasets; when compared to the TFA (Task-agnostic Fine-tuning and Adapter), CD-FSOD (Cross-Domain Few-Shot Object Detection) and DeFRCN (Decoupled Faster R-CNN for Few-Shot Object Detection), with sample orders of magnitude 1 and 5, increased the detection accuracy by 1.67% and 1.87%, respectively. Video anomaly detection (VAD) plays a crucial role in fields such as security, production, and transportation. To address the issue of overgeneralization in anomaly behavior prediction by deep neural networks, we propose a network called AMFCFBMem-Net (appearance and motion feature cross-fusion block memory network), which combines appearance and motion feature cross-fusion blocks. Firstly, dual encoders for appearance and motion are employed to separately extract these features, which are then integrated into the skip connection layer to mitigate the model's tendency to predict abnormal behavior, ultimately enhancing the prediction accuracy for abnormal samples. Secondly, a motion foreground extraction module is integrated into the network to generate a foreground mask map based on speed differences, thereby widening the prediction error margin between normal and abnormal behaviors. To capture the latent features of various models for normal samples, a memory module is introduced at the bottleneck of the encoder and decoder structures. This further enhances the model's anomaly detection capabilities and diminishes its predictive generalization towards abnormal samples. The experimental results on the UCSD Pedestrian dataset 2 (UCSD Ped2) and CUHK Avenue anomaly detection dataset (CUHK Avenue) demonstrate that, compared to current cutting-edge video anomaly detection algorithms, our proposed method achieves frame-level AUCs of 97.5% and 88.8%, respectively, effectively enhancing anomaly detection capabilities. Next generation fuel-efficient jet engines are running hotter presenting a structural challenge for the exhaust systems and structures adjacent to the engines. A conventional and affordable titanium alloy with superior oxidation resistance provides significant weight reductions and associated cost savings by eliminating the need for high density material systems such as nickel-base superalloys for service temperatures in between current titanium and nickel, enabling major technology advancement in high temperature aerospace applications. This paper presents an overview of Arconic’s engineered material ARCONIC-THORTM to address the needs of future aerospace systems.

The aeroengine and airframe applications of titanium alloys are often limited by their insufficient oxidation resistance to the aggressive environment at higher temperatures. A high temperature oxidation resistant titanium alloy (Arconic-THORTM) has been developed. This alloy is an alpha-beta alloy with superior oxidation resistance and improved creep resistance. The oxidation weight gain of Arconic-THORTM is much lower than Ti-6242 and Beta 21s in the temperature range up to 750˚C. The room and elevated temperature properties of Arconic-THORTM are comparable to those for Ti-6242. Arconic-THORTM also shows superior, post-thermal-exposure tensile strength, ductility, and fatigue properties, and is capable of being cold formed, hot formed, and superplastic formed, welded and heat treated to different product geometries. The microstructures and mechanical properties for sheet manufactured from the production ingots are presented and discussed in this study.

Utilization of gamma titanium aluminide alloys in aerospace and automotive/industrial applications has placed significant demand on melting sources for products to be used in cast, wrought, and direct-machining applications. There is also an increased demand for input stock used in gas atomization of powders. Current technologies used in ingot manufacturing include plasma arc melting, vacuum arc melting, and induction skull melting + centrifugal casting. Subsequent processing may include forging, re-melting + casting, or machining directly into components. Over the past six years, Arconic Engineered Structures has developed a robust melting method using plasma cold-hearth melting technology, including the design and implementation of a new 3-torch system to produce Ti-48-2-2 cast bars. General discussions concerning plasma cold-hearth melting, manufacturing challenges, and metallurgical attributes associated with cast Ti-48-2-2 bars will be reviewed. Emphasis will be on understanding the impact of hot isostatic pressing on internal voids, residual stress cracking and resulting mechanical properties.

Background and Objectives: Notch is a fascinating signaling pathway. It is extensively involved in tumor growth, cancer stem cells, metastasis, and treatment resistance and plays important roles in metabolic regulation, tumor microenvironment, and tumor immunity. However, the role of Notch in ovarian cancer (OC) has yet to be fully understood. Therefore, this study systematically described the expression, mutation, and copy number variation of genes in the Notch signaling pathway in OC and evaluated the relationship between gene mutation and Overall Survival (OS) prognosis. Materials and Methods: Notch risk score (NTRS) was established by univariate Cox regression analysis combined with Lasso regression analysis, and the efficacy of NTRS in predicting prognosis and immunotherapy response in patients with OC was verified. We further assessed the correlations of NTRS with clinical features, immune infiltration level, immune checkpoint expression, and immune characteristics. Additionally, differential expression and functions of the fourteen signature genes were confirmed via vitro assays. Results: The results showed that Notch genes (NTGs) were markedly differentiated between tumor and normal tissues, which may help to explain the high heterogeneity in the biological characteristics and therapeutic outcomes of human OC. A Notch risk (NTR) prognostic model based on 11 key NTGs was successfully constructed. Tumors with high Notch risk scores (NTRS) were independently associated with shorter overall survival and poorer immunotherapy outcomes. We further assessed the correlations of NTRS with immune characteristics. The results showed that NTGs play a key role in regulating the tumor immune microenvironment. Additionally, we validated the baseline and induced expressions of 14 prognosis-related NTGs in our own OC samples. In vitro assays confirmed that the knockdown of NCOR2 and APH1B and overexpression of HEY2 and SKP2 could inhibit the proliferation, invasion, and migration of OC cells. Conclusions: These findings emphasize that Notch multilayer changes are associated with the prognosis of patients with OC and the characteristics of immune cell infiltration. Our predictive signature may predict the prognosis and immunotherapy response of OC patients in an independent manner. Ovarian cancer is one of the most common gynecological malignancies in women, with a poor prognosis and high mortality. With the expansion of single-cell RNA sequencing technologies, the inner biological mechanism involved in tumor recurrence should be explored at the single-cell level, and novel prognostic signatures derived from recurrence events were urgently identified. In this study, we identified recurrence-related genes for ovarian cancer by integrating two Gene Expression Omnibus datasets, including an ovarian cancer single-cell RNA sequencing dataset (GSE146026) and a bulk expression dataset (GSE44104). Based on these recurrence genes, we further utilized the merged expression dataset containing a total of 524 ovarian cancer samples to identify prognostic signatures and constructed a 13-gene risk model, named RMGS (recurrence marker gene signature). Based on the RMGS score, the samples were stratified into high-risk and low-risk groups, and these two groups displayed significant survival difference in two independent validation cohorts including The Cancer Genome Atlas (TCGA). Also, the RMGS score remained significantly independent in multivariate analysis after adjusting for clinical factors, including the tumor grade and stage. Furthermore, there existed close associations between the RMGS score and immune characterizations, including checkpoint inhibition, EMT signature, and T-cell infiltration. Finally, the associations between RMGS scores and molecular subtypes revealed that samples with mesenchymal subtypes displayed higher RMGS scores. In the meanwhile, the genomics characterization from these two risk groups was also identified. In conclusion, the recurrence-related RMGS model we identified could provide a new understanding of ovarian cancer prognosis at the single-cell level and offer a reference for therapy decisions for patient treatment. Background and Aim. Increasing evidence suggests that the peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α)/fibronectin type III domain-containing 5 (FNDC5)/brain-derived neurotrophic factor (BDNF) pathway might be critical for neuroprotection. Our present study is aimed at investigating the antidepressant-like effects of curcumin (CUR) in a chronic unpredictable mild stress- (CUMS-) induced depression rat model and explore whether the PGC-1α/FNDC5/BDNF pathway is the major driving force behind the therapeutic effects of CUR. Methods. All rats were randomly divided into four groups, namely, control, CUMS, CUMS + CUR , and CUMS + CUR + SR 18292 (PGC-1α inhibitor). Behavioral tests were conducted to assess the antidepressant-like effects of CUR. The expressions of PGC-1α, estrogen-related receptor alpha (ERRα), FNDC5, and BDNF were determined to investigate the regulatory effects of CUR on the PGC-1α/FNDC5/BDNF pathway. The PGC-1α inhibitor SR18292 was used to explore the role of PGC-1α in the induction of BDNF by CUR. Results. Daily gavage of 100 mg/kg CUR successfully attenuated the abnormal behaviors induced by CUMS and effectively prevented CUMS-induced reduction of PGC-1α, ERRα, FNDC5, and BDNF expressions. CUR also enhanced PGC-1α and ERRα translocation from cytoplasm to nucleus. Furthermore, we found that CUR supplementation effectively promoted neurocyte proliferation and suppressed neuronal apoptosis induced by CUMS. Of note, the PGC-1α inhibitor SR18292 remarkably reversed the beneficial effects of CUR on depressed rats, indicating an important role of PGC-1α in the antidepressant-like effects of CUR. Conclusion. Collectively, our data evaluating the neuroprotective action of CUR in the CUMS rats highlights the involvement of the PGC-1α/FNDC5/BDNF pathway in the antidepressant-like effects of CUR. Cultivating new crop cultivars with multiple abiotic stress tolerances is important for crop production. The abscisic acid‐stress‐ripening (ASR) protein has been shown to confer abiotic stress tolerance in plants. However, the mechanisms of ASR function under stress condition remain largely unclear. In this study, we characterized all ASR family members in common wheat and constitutively overexpressed TaASR1‐D in a commercial hexaploid wheat cultivar Zhengmai 9023. The transgenic wheat plants exhibited increased tolerance to multiple abiotic stresses and increased grain yields under salt stress condition. Overexpression of TaASR1‐D conferred enhanced antioxidant capacity and ABA sensitivity in transgenic wheat plants. Further, RNA in situ hybridization results showed that TaASR1‐D had higher expression levels in the vascular tissues of leaves and the parenchyma cells around the vascular tissues of roots and stems. Yeast one‐hybrid and electrophoretic mobility shift assays revealed that TaASR1‐D could directly bind the specific cis‐elements in the promoters of TaNCED1 and TaGPx1‐D. In conclusion, our findings suggest that TaASR1‐D can be used to breed new wheat cultivars with increased multiple abiotic stress tolerances, and TaASR1‐D enhances abiotic stress tolerances by reinforcing antioxidant capacity and ABA signalling. Durum wheat has limited culinary utilizations partly due to its extremely hard kernel texture. Previously, we developed transgenic durum wheat lines with expression of the wildtype Puroindoline a (Pina) and characterized PINA's effects on kernel hardness, total flour yield and dough mixing properties in durum wheat. The medium-hard kernel texture is potentially useful for exploring culinary applications of durum wheat. In the present study, we examined the milling parameters and flour attributes of the transgenic lines, including particle size distribution, damaged starch and water binding capacity. PINA expression results in increased break and reduction flour yield but decreased shorts. PINA expression also leads to finer flour particles and decreased starch damage. Interestingly, PINA transgenic lines showed increased peak viscosity and breakdown viscosity but leave other flour pasting parameters generally unaltered. PINA transgenic lines were associated with increased small monomeric proteins, appearing to affect gluten aggregation. Our data together with several previous results highlight distinct effects of PINs on pasting properties depending on species and variety. The medium-hard kernel texture together with improved pasting parameters may be valuable for producing a broader range of end-products from durum wheat. [ABSTRACT FROM AUTHOR]

Mineral elements in soil solutions are thought to be the precursor of the formation of reactive minerals, which play an important role in global carbon (C) cycling. However, information regarding the regulation of mineral elements release in soil is scarce. Here, we examined the long-term (i.e., 23 yrs) effects of fertilisation practices on Fe minerals in a red soil in Southern China. The results from chemical analysis and Fourier-transform infrared spectroscopy showed that long-term swine manure (M) treatment released greater amounts of minerals into soil solutions than chemical fertilisers (NPK) treatment, and Fe played a dominant role in the preservation of dissolved organic C. Furthermore, Fe K-edge X-ray absorption near-edge fine structure spectroscopy demonstrated that reactive Fe minerals were mainly composed of less crystalline ferrihydrite in the M-treated soil and more crystalline goethite in the NPK-treated soil. In conclusion, this study reported spectroscopic evidence of the improvement of reactive Femineral content in the M-treated soil colloids when compared to NPK-treated soil colloids. Introduction: On July 1, 2011, the Chinese government launched a national Action Plan for antibiotic stewardship targeting antibiotic misuse in public hospitals. The aim of this study was to evaluate the impacts of the Action Plan in terms of frequency and intensity of antibiotic utilization and patients costs in public general hospitals. Methods: Administrative pharmacy data from July 2010 to June 2014 were sampled from 65 public general hospitals and divided into three segments: (1) July 2010 to June 2011 as the preparation period; (2) July 2011 to June 2012 as the intervention period; and (3) July 2012 to June 2014 as the assessment period. The outcome measures included (1) antibiotic prescribing rates; (2) intensity of antibiotic consumption; (3) patients costs; and (4) duration of peri-operative antibiotic treatment in clean surgeries of thyroidectomy, breast, hernia, and orthopedic procedures. Longitudinal and cross-sectional analyses were conducted. Results: Longitudinal analyses showed significant trend changes in the frequency and intensity of antibiotic consumption, the patients’ costs on antibiotics, and the duration of antibiotic treatment received by surgical patients undergoing the 4 clean procedures during the intervention period. Cross-sectional analyses showed that the antibiotic prescribing rates were reduced to 35.3% and 12.9% in inpatient and outpatient settings, that the intensity of antibiotic consumption was reduced to 35.9 DDD/100 bed-days, that patients’ costs on antibiotics were reduced significantly, and that the duration of peri-operative antibiotic treatment received by surgical patients undergoing the 4 types of clean procedures decreased to less than 24 hour during the assessment period. Conclusion: The Action Plan, as a combination of managerial and professional strategies, was effective in reducing the frequency and intensity of antibiotic consumption, patients’ costs on antibiotics, and the duration of peri-operative antibiotic treatment in the 4 clean surgeries. Abstract: We report the application of Vitamin D3 (VD3) in nanoparticles of oleoyl alginate ester (OAE)(OAE-VD3). The internalization of fluorescent OAE-VD3 by Caco-2 cells was visualized by confocal laser scanning microscopy. In vivo pharmacokinetic studies showed that incorporation into OAE nanoparticles resulted in increased absorption of VD3. Its application in the treatment of rickets was assayed using a model of nutritionally induced vitamin D-deficiency rickets. The results showed that the encapsulated VD3 had better efficacy than that of the free drug in vivo. Our studies provide evidence that OAE nanoparticles are valuable as nutraceutical delivery vehicles to enhance the absorption of VD(3). [ABSTRACT FROM AUTHOR]

Kernel hardness is a key trait of wheat seeds, largely controlled by two tightly linked genes Puroindoline a and b (Pina and Pinb). Genes homologous to Pinb, namely Pinb2, have been studied. Whether these genes contribute to kernel hardness and other important seed traits remains inconclusive. Using the high-quality bread wheat reference genome, we show that PINB2 are encoded by three homoeologous loci Pinb2 not syntenic to the Hardness locus, with Pinb2-7A locus containing three tandem copies. PINB2 proteins have several features conserved for the Pin/Pinb2 phylogenetic cluster but lack a structural basis of significant impact on kernel hardness. Pinb2 are seed-specifically expressed with varied expression levels between the homoeologous copies and among wheat varieties. Using the high-quality genome information, we developed new Pinb2 allele specific markers and demonstrated their usefulness by 1) identifying new Pinb2 alleles in Triticeae species; and 2) performing an association analysis of Pinb2 with kernel hardness. The association result suggests that Pinb2 genes may have no substantial contribution to kernel hardness. Our results provide new insights into Pinb2 evolution and expression and the new allele-specific markers are useful to further explore Pinb2's contribution to seed traits in wheat.
