6. The co-occurrence of anemia and cardiometabolic disease risk demonstrates sex-specific sociodemographic patterning in an urbanizing rural region of southern India

Abstract

BACKGROUND/OBJECTIVES: To determine the extent and sociodemographic determinants of anemia, overweight, metabolic syndrome (MetS) and the co-occurrence of anemia with cardiometabolic disease risk factors among a cohort of Indian adults. SUBJECT/METHODS: Cross-sectional survey of adult men (n = 3322) and nonpregnant women (n = 2895) aged 18 years and older from the third wave of the Andhra Pradesh Children and Parents Study that assessed anemia, overweight based on body mass index, and prevalence of MetS based on abdominal obesity, hypertension and blood lipid and fasting glucose measures. We examined associations of education, wealth and urbanicity with these outcomes and their co-occurrence. RESULTS: The prevalence of anemia and overweight was 40% and 29% among women, respectively, and 10% and 25% among men (P < 0.001), respectively, whereas the prevalence of MetS was the same across sexes (15%;P = 0.55). The prevalence of concurrent anemia and overweight (9%), and anemia and MetS (4.5%) was highest among women. Household wealth was positively associated with overweight and MetS across sexes (P < 0.05). Independent of household wealth, higher education was positively correlated with MetS among men (odds ratio (95% confidence interval): MetS: 1.4 (0.99, 2.0)) and negatively correlated with MetS among women (MetS: 0.54 (0.29, 0.99)). Similar sex-specific associations were observed for the co-occurrence of anemia with overweight and MetS. CONCLUSIONS: Women in this region of India may be particularly vulnerable to co-occurring anemia and cardiometabolic risk, and associated adverse health outcomes as the nutrition transition advances in India. Abstract

Back pain is a common and debilitating disorder with largely unknown underlying biology. Here we report a genome-wide association study of back pain using diagnoses assigned in clinical practice; dorsalgia (119,100 cases, 909,847 controls) and intervertebral disc disorder (IDD) (58,854 cases, 922,958 controls). We identify 41 variants at 33 loci. The most significant association (ORIDD = 0.92, P = 1.6 × 10−39; ORdorsalgia = 0.92, P = 7.2 × 10−15) is with a 3’UTR variant (rs1871452-T) in CHST3, encoding a sulfotransferase enzyme expressed in intervertebral discs. The largest effects on IDD are conferred by rare (MAF = 0.07 − 0.32%) loss-of-function (LoF) variants in SLC13A1, encoding a sodium-sulfate co-transporter (LoF burden OR = 1.44, P = 3.1 × 10−11); variants that also associate with reduced serum sulfate. Genes implicated by this study are involved in cartilage and bone biology, as well as neurological and inflammatory processes. © 2022, The Author(s).

Abstract

BACKGROUND/OBJECTIVES: Iron is fundamental to many basic biological functions, and animal studies suggest that iron deficiency early in life can have a lasting impact on the developing brain. SUBJECTS/METHODS: We used a population-based cohort of mothers and their children to assess the effect of iron status among pregnant women on the cognitive ability of their offspring. But to avoid the inherent confounding that occurs within observational epidemiology studies we examined the association of maternal genotype at single-nucleotide polymorphisms in the genes HFE (rs1799945) and (rs1800562), TF (rs3811647) and TMPRSS6 (rs1800562), which are related to iron, haemoglobin or transferrin levels, on their child's cognitive test scores at age 8. RESULTS: We found strong associations between HFE and TMPRSS6 genotypes and mother's haemoglobin levels early in pregnancy (P-values are all ≤ 4.1 x [10.sup.- 5]) and a genetic score comprised of alleles at these loci was even more strongly associated with haemoglobin levels (P = 3.0 x [10.sup.-18]), suggesting that this was a good instrument to use to look at the effect of prenatal iron levels on offspring cognition. However, mother's genotype at the above loci was not associated with offspring IQ at age 8. CONCLUSIONS: We therefore concluded that there is no evidence of an effect of exposure to low levels of iron (within the normal range) in pregnancy on offspring cognition at age 8. However, pregnant women in the UK with low haemoglobin levels are prescribed iron supplements and so we were unable to look at the effect of iron deficiency in our study. 15. The long-term effect of dietary advice in men with coronary disease: follow-up of the Diet and Reinfarction trial (DART)

Abstract

Objective: To assess the long-term effect of dietary advice on diet and mortality after a randomised trial of men with a recent history of myocardial infarction. Design: Questionnaire survey and mortality follow-up after a trial of dietary advice. Setting: Twenty-one hospitals in south Wales and south-west England. Subjects: Former participants in the Diet and Reinfarction Trial. Main outcome measures: Current fish intake and cereal fibre intake. All-cause mortality, stroke mortality and coronary mortality. Results: By February 2000, after 21 147 person years of follow-up, 1083 (53%) of the men had died. Completed questionnaires were obtained from 879 (85%) of the 1030 men alive at the beginning of 1999. Relative increases in fish and fibre intake were still present at l0 y but were much smaller. The early reduction in all-cause mortality observed in those given fish advice (unadjusted hazard 0.70 (95% CI 0.54, 0.92)) was followed by an increased risk over the next 3 y (unadjusted hazard 1.31 (95% CI 1.01, 1.70). Fat and fibre advice had no clear effect on coronary or all-cause mortality. The risk of stroke death was increased in the fat advice group-the overall unadjusted hazard was 2.03 (95% CI 1.14, 3.63). Conclusions: In this follow-up of a trial of intensive dietary advice following myocardial infarction we did not observe any substantial long-term survival benefit. Abstract

Published in final edited form as: Nat Genet. 2019 March ; 51(3): 431–444. doi:10.1038/s41588-019-0344-8., Autism spectrum disorder (ASD) is a highly heritable and heterogeneous group of neurodevelopmental phenotypes diagnosed in more than 1% of children. Common genetic variants contribute substantially to ASD susceptibility, but to date no individual variants have been robustly associated with ASD. With a marked sample size increase from a unique Danish population resource, we report a genome-wide association meta-analysis of 18,381 ASD cases and 27,969 controls that identifies five genome-wide significant loci. Leveraging GWAS results from three phenotypes with significantly overlapping genetic architectures (schizophrenia, major depression, and educational attainment), seven additional loci shared with other traits are identified at equally strict significance levels. Dissecting the polygenic architecture, we find both quantitative and qualitative polygenic heterogeneity across ASD subtypes. These results highlight biological insights, particularly relating to neuronal function and corticogenesis and establish that GWAS performed at scale will be much more productive in the near term in ASD., The iPSYCH project is funded by the Lundbeck Foundation (grant numbers R102-A9118 and R155-2014-1724) and the universities and university hospitals of Aarhus and Copenhagen. Abstract

The beam spin asymmetry of the exclusive pseudoscalar channel e→p→e′p′η was measured for the first time in the deep-inelastic regime (W>2 GeV/c2 and Q2>1 GeV2/c2) using a longitudinally polarized 5.78 GeV electron beam at Jefferson Lab with the CEBAF Large Acceptance Spectrometer. The data were accumulated in 144 four-dimensional bins of Q2, xB, −t and ϕ over a wide kinematic range, where ϕ is the azimuthal angle between the lepton and hadron scattering planes, The measured azimuthal dependence with large amplitudes of the sin⁡ϕ moments is a clear indication of a substantial contribution to the polarized cross-section from transversely polarized virtual photons. In the framework of generalized parton distributions (GPDs) this contribution is expressed via longitudinal-transverse interference between chiral-even and chiral-odd GPDs. The experimental results are compared to the existing theoretical models demonstrating the sensitivity to the product of chiral-odd and chiral-even GPDs and provide new constraints to the existing GPD parameterizations. Abstract

Osteoarthritis is the most common musculoskeletal disease and the leading cause of disability globally. Here, we performed a genome-wide association study for osteoarthritis (77,052 cases and 378,169 controls), analyzing four phenotypes: knee osteoarthritis, hip osteoarthritis, knee and/or hip osteoarthritis, and any osteoarthritis. We discovered 64 signals, 52 of them novel, more than doubling the number of established disease loci. Six signals fine-mapped to a single variant. We identified putative effector genes by integrating expression quantitative trait loci (eQTL) colocalization, fine-mapping, and human rare-disease, animal-model, and osteoarthritis tissue expression data. We found enrichment for genes underlying monogenic forms of bone development diseases, and for the collagen formation and extracellular matrix organization biological pathways. Ten of the likely effector genes, including TGFB1 (transforming growth factor beta 1), FGF18 (fibroblast growth factor 18), CTSK (cathepsin K), and IL11 (interleukin 11), have therapeutics approved or in clinical trials, with mechanisms of action supportive of evaluation for efficacy in osteoarthritis.

Abstract

International audience; The beam spin asymmetry of the exclusive pseudoscalar channel e→p→e′p′η was measured for the first time in the deep-inelastic regime ( W>2 GeV/c2 and Q2>1 GeV2/c2 ) using a longitudinally polarized 5.78 GeV electron beam at Jefferson Lab with the CEBAF Large Acceptance Spectrometer. The data were accumulated in 144 four-dimensional bins of Q2 , xB , − t and ϕ over a wide kinematic range, where ϕ is the azimuthal angle between the lepton and hadron scattering planes, The measured azimuthal dependence with large amplitudes of the sin⁡ϕ moments is a clear indication of a substantial contribution to the polarized cross-section from transversely polarized virtual photons. In the framework of generalized parton distributions (GPDs) this contribution is expressed via longitudinal-transverse interference between chiral-even and chiral-odd GPDs. The experimental results are compared to the existing theoretical models demonstrating the sensitivity to the product of chiral-odd and chiral-even GPDs and provide new constraints to the existing GPD parameterizations.

We present precision measurements of the target and beam-target spin asymmetries from neutral pion electroproduction in deep-inelastic scattering (DIS) using the CEBAF Large Acceptance Spectrometer (CLAS) at Jefferson Lab. Abstract

We report the first measurements of the $E$ beam-target helicity asymmetry for the $\vec{\gamma} \vec{n} \to K^{0}\Lambda$, and $K^{0}\Sigma^{0}$ channels in the energy range of 1.70$\leq W\leq$2.34 GeV. The CLAS system at Jefferson Lab used a circularly polarized photon beam and a target consisting of longitudinally polarized solid molecular hydrogen deuteride (HD) with low background contamination for the measurements. Comparisons with predictions from the KaonMAID, SAID, and Bonn-Gatchina models are presented. These results will help separate the isospin $I=0$ and $I=1$ photo-coupling transition amplitudes in pseudoscalar meson photoproduction.

Abstract

The photoproduction of ω mesons off the proton has been studied in the reaction γp → p ω using the CEBAF\ud Large Acceptance Spectrometer (CLAS) and the frozen-spin target in Hall B at the Thomas Jefferson National\ud Accelerator Facility. For the first time, the target asymmetry T has been measured in photoproduction from the\ud decay ω → π+π−π0, using a transversely polarized target with energies ranging from just above the reaction\ud threshold up to 2.8 GeV. Significant nonzero values are observed for these asymmetries, reaching about 30–40%\ud in the third-resonance region. New measurements for the photon-beam asymmetry Σ are also presented, which\ud agree well with previous CLAS results and extend the world database up to 2.1 GeV. These data and additional\ud ω photoproduction observables from CLAS were included in a partial-wave analysis within the Bonn-Gatchina\ud framework. Significant contributions from s-channel resonance production were found in addition to t-channel\ud exchange processes.

Abstract

The cross section for coherent $\omega$-meson photoproduction off the deuteron has been measured for the first time as a function of the momentum transfer $t = (P_{\gamma}-P_{\omega})^2$ and photon energy $E_{\gamma}$ using the CLAS detector at the Thomas Jefferson National Accelerator Facility. The cross sections are measured in the energy range $1.4 < E_{\gamma} < 3.4$ GeV. A model based on $\omega-N$ rescattering is consistent with the data at low and intermediate momentum transfer, $|t|$. For $2.8 < E_{\gamma} < 3.4$ GeV, the total cross-section of $\omega-N$ scattering, based on fits within the framework of the Vector Meson Dominance model, is in the range of 30-40 mb.

Abstract

New results on the single-differential and fully-integrated cross sections for the process $\gamma_{v} p \rightarrow p' \pi^{+} \pi^{-}$ are presented. The experimental data were collected with the CLAS detector at Jefferson Laboratory. Measurements were carried out in the kinematic region of the reaction invariant mass $W$ from 1.3 to 1.825 GeV and the photon virtuality $Q^2$ from 0.4 to 1.0 GeV$^2$. The cross sections were obtained in narrow $Q^{2}$ bins (0.05 GeV$^{2}$) with the smallest statistical uncertainties achieved in double-pion electroproduction experiments to date. The results were found to be in agreement with previously available data where they overlap. A preliminary interpretation of the extracted cross sections, which was based on a phenomenological meson-baryon reaction model, revealed substantial relative contributions from nucleon resonances. The data offer promising prospects to improve knowledge on the $Q^{2}$-evolution of the electrocouplings of most resonances with masses up to $\sim$1.8 GeV., Comment: 22 pages, 22 figures, will be sent to Phys. Abstract

International audience; The photoproduction of ω mesons off the proton has been studied in the reaction γp→pω using the CEBAF Large Acceptance Spectrometer (CLAS) and the frozen-spin target in Hall B at the Thomas Jefferson National Accelerator Facility. For the first time, the target asymmetry T has been measured in photoproduction from the decay ω→π+π−π0, using a transversely polarized target with energies ranging from just above the reaction threshold up to 2.8 GeV. Significant nonzero values are observed for these asymmetries, reaching about 30–40% in the third-resonance region. New measurements for the photon-beam asymmetry Σ are also presented, which agree well with previous CLAS results and extend the world database up to 2.1 GeV. These data and additional ω photoproduction observables from CLAS were included in a partial-wave analysis within the Bonn-Gatchina framework. Significant contributions from s-channel resonance production were found in addition to t-channel exchange processes.

Abstract

We report the first measurements of the $E$ beam-target helicity asymmetry for the $\vec{\gamma} \vec{n} \to K^{0}\Lambda$, and $K^{0}\Sigma^{0}$ channels in the energy range 1.70$\leq W\leq$2.34 GeV. The CLAS system at Jefferson Lab uses a circularly polarized photon beam and a target consisting of longitudinally polarized solid molecular hydrogen deuteride with low background contamination for the measurements. The multivariate analysis method boosted decision trees was used to isolate the reactions of interest. Comparisons with predictions from the KaonMAID, SAID, and Bonn-Gatchina models are presented. These results will help separate the isospin $I=0$ and $I=1$ photo-coupling transition amplitudes in pseudoscalar meson photoproduction., Comment: 14 pages, 9 figures; Revised to match published version
