The paper discusses the chemical-physical-petrographic features and decay processes of San Nicola Church (11th−14th century) building materials, one of the most representative Medieval Churches in Sardinia, stylistically attributable to the Romanesque architecture. The monument was built up into two stages and shows a characteristic size uniformity of the ashlars. The masonry is mainly made up of rhyodacitic pyroclastites belonging to the Sardinian Eocene-Miocene magmatic phase (38–15 My). These volcanic rocks were widely used in Medieval architecture for the excellent workability, but its minero-petrographic features greatly favor the decay, due to the action of chemical-physical alteration processes. Although the alteration degree and macroscopic forms of decay vary from zone to zone in the monument, according to the different compositional features of the volcanic stones, weathering and exposure condition, the pyroclastic rocks are generally affected by greater alteration than other igneous lithotypes, due to their petrographic-volcanological characteristics (e.g., low-medium welding grade, medium-high porosity). The results concerning mineralogical and petrographic features, respectively, carried out by the application of X-Ray Powder Diffraction (XRPD), Optical Microscope (OM), and Scanning Electron Microscope (SEM) techniques, both on fresh building rocks and associated alterations, are here presented and discussed. This study focuses on the analysis of Islamic ceramics from the Alcaçova of Santarém, Portugal. To establish the ceramic raw material provenance and ceramic technology, a set of raw materials was sampled and compared with the archaeological ceramics. Raw materials were dry sieved to increase the clay content, to reconstruct the ceramist homogenization process, and to evaluate chemical and mineralogical variation. Heritage sites built with natural stone are at risk from fires, which can alter stone properties and compromise its structural integrity. Over 60 studies in the past three decades have examined fire impact on natural stone, providing insights for their prevention and restoration. The primary objectives are to develop effective strategies to mitigate fire risks, protect heritage structures, and ensure the preservation of our cultural legacy. Two noteworthy Portuguese limestones used as heritage building materials: Lioz (LL) and grey Ançã stone (GAS), were studied regarding the effect of high-temperature exposure for simulating fire at 200 °C, 400 °C and 600 °C, followed by cooling in water to reproduce fire extinguish in natural stone buildings. The findings provided insights into how the different temperatures impact the stone morphological, physical and mechanical properties. Color measurements (CIE L*a*b*) showed a color difference from 3 to 32 %; SEM-EDS confirmed microstructure modifications after fire exposure with cracks formation and intragranular porosity development. Among the diverse physical and mechanical properties of the stones, uniaxial compressive strength decreased from 1 to 33 %, Leeb D hardness decreased up to 12.2 %, lowering in open porosity was detected in the range 70-289 % and ultrasound speed propagation were significantly affected after thermal cycle at 600 °C with a negative variation reaching 49 %. Results from TGA show a loss of mass due to retained water (∼40 °C) and loss of hydration water at ∼ 120 °C in both limestones. The total mass loss (∼42-∼44 %) is associated with the loss of H 2 O, CO 2 . In conclusion, stones with higher toughness and compression strength exhibited reduced damage at high temperatures due to their enhanced resistance to fracturing under stress. As limestone's mechanical strength decreases under high temperatures, it's advisable to increase its thickness to ensure sufficient support for loads and intended conditions of use. Alpha7 nicotinic acetylcholine receptors (α7 nAChR) are widely distributed throughout the central nervous system and are found at particularly high levels in the hippocampus and cortex. Several lines of evidence indicate that pharmacological enhancement of α7 nAChRs function could be a potential therapeutic route to alleviate disease-related cognitive deficits. A recent pharmacological approach adopted to increase α7 nAChR activity has been to identify selective positive allosteric modulators (PAMs). α7 nAChR PAMs have been divided into two classes: type I PAMs increase agonist potency with only subtle effects on kinetics, whereas type II agents produce additional dramatic effects on desensitization and deactivation kinetics. Here we report novel observations concerning the pharmacology of the canonical type II PAM, PNU120596. Using patch clamp analysis of acetylcholine (ACh)-mediated currents through recombinant rat α7 nAChR we show that positive allosteric modulation measured in two different ways is greatly attenuated when the temperature is raised to near physiological levels. Furthermore, PNU120596 largely removes the strong inward rectification usually exhibited by α7 nAChR-mediated responses.

The alpha7 nicotinic acetylcholine receptor (nAChR) has been implicated in Alzheimer's disease and schizophrenia, leading to efforts targeted toward discovering agonists and positive allosteric modulators (PAMs) of this receptor. In a Ca2+ flux fluorometric imaging plate reader assay, SB-206553 (3,5-dihydro-5-methyl -N-3-pyridinylbenzo [1, 2-b:4,5 -b']-di pyrrole-1(2H)-carboxamide), a compound known as a 5-hydroxytryptamine(2B/2C) receptor antagonist, produced an 8-fold potentiation of the evoked calcium signal in the presence of an EC(20) concentration of nicotine and a corresponding EC(50) of 1.5 muM for potentiation of EC(20) nicotine responses in GH4C1 cells expressing the alpha7 receptor. SB-206553 was devoid of direct alpha7 receptor agonist activity and selective against other nicotinic receptors. Confirmation of the PAM activity of SB-206553 on the alpha7 nAChR was obtained in patch-clamp electrophysiological experiments in GH4C1 cells, where it failed to evoke any detectable currents when applied alone, yet dramatically potentiated the currents evoked by an EC(20) (17 microM) and EC(100) (124 microM) of acetylcholine (ACh). Native nicotinic receptors in CA1 stratum radiatum interneurons of rat hippocampal slices could also be activated by ACh (200 microM), an effect that was entirely blocked by the alpha7-selective antagonist methyllycaconitine (MLA). These ACh currents were potentiated by SB-206553, which increased the area of the current response significantly, resulting in a 40-fold enhancement at 100 microM. In behavioral experiments in rats, SB-206553 reversed an MK-801 (dizocilpine maleate)-induced deficit in the prepulse inhibition of acoustic startle response, an effect attenuated in the presence of MLA. This latter observation provides further evidence in support of the potential therapeutic utility of alpha7 nAChR PAMs in schizophrenia.
