Obesity, a global public health challenge, is closely associated with gut microbiota alterations. Probiotics have the potential to prevent obesity, but their effectiveness can vary depending on the specific strain. This study focused on evaluating the anti-obesity and probiotic properties of four Bifidobacterium species isolated from healthy Chinese individuals in vitro. The findings indicated that Bifidobacterium lactis YGMCC2013 showed notable cholesterol assimilation ability, bile salt hydrolase activity, gastrointestinal tolerance, cellular adhesion, and production of short-chain fatty acids. The anti-obesity effect of YGMCC2013 was then assessed in mice fed a high-fat diet. The results demonstrated that YGMCC2013 reduced body and liver weight, adipose tissue hypertrophy and inflammation, and influenced specific microbial genera associated with adipose tissue characteristics. Notably, the study observed associations between certain microbial genera and different types of adipose tissue, indicating that YGMCC2013 may impact the balance and function of adipose tissue through its effects on the gut microbiota. Furthermore, YGMCC2013 enhanced the thermogenic capacity of brown and beige adipocytes, and improved the structure and function of the gut microbiota. These improvements were reflected by an increase in short-chain fatty acids produced by beneficial bacteria in the intestine, and the activation of metabolic pathways related to energy and carbohydrate metabolism. Moreover, YGMCC2013 modulated the expression of il-27 and tgr5 signaling pathways in adipose tissue, which might be involved in regulating adipose tissue inflammation and energy expenditure, respectively. These findings collectively demonstrate that YGMCC2013 has potential as a probiotic candidate for preventing obesity and improving metabolic health by influencing the gut-adipose tissue axis.

Crown ethers are famous for the highly selectively grab Sr(II) from concentrated nitric acid solution due to the size match, but they suffer from the high leakage into the liquid phase caused by the presence of a large number of hydrophilic groups. To reduce their leakage, two novel porous silica-based adsorbents, (DtBuCH18C6 + Dodec)/SiAaC-g-ABSA and (DtBuCH18C6 + Dodec)/SiAaC-g-3-ABSA, were prepared by vacuum impregnation with organic contents of about 55.9 wt.% and 56.1 wt.%, respectively. The two adsorbents have good reusability and structural stability, and the total organic carbon leakage rates in 2 M HNO3 solution are lower than 0.56 wt.% and 0.29 wt.%, respectively. Batch adsorption experiments revealed that the two adsorbents possessed good adsorption selectivity towards Sr, with SFSr/M over 40, except that of SFSr/Ba in 2 M HNO3 solution. The adsorption equilibrium of Sr in 2 M HNO3 solution was reached within 1 h, with saturated adsorption capacities of 36.9 mg/g and 37.5 mg/g, respectively. Furthermore, the XPS results indicate that the adsorption mechanism is the coordination of the crown ether ring with Sr. This work not only develops two novel adsorbents for the separation of Sr in nitric acid environments; it also provides a method for effectively reducing the water solubility of crown ethers.

Hypercholesterolemia, a risk factor for cardiovascular disease (CVD), often requires therapeutic agents with varying degrees of side effects. This has created a need for safe and natural alternatives such as medications or functional foods that can improve lipid metabolism and reduce cholesterol levels. In recent years, Next-generation probiotics (NGPs) have recently emerged as a potential solution, offering distinct mechanisms compared to traditional probiotics. Among the NGPs, Bacteroides, a dominant bacterial genus in the human gut, has gained significant attention due to its prevalence, ability to break down plant polysaccharides, and production of short-chain fatty acids (SCFAs). Recent evidence has demonstrated that Bacteroides effectively reduces cholesterol levels, prevents obesity, and lowers the risk of CVD. However, research on Bacteroides is currently limited to a few species, leaving rooms for exploration of the beneficial functions of different species in this genus. In this study, we isolated 66 Bacteroides strains, including 9 distinct species, from healthy adults’ fecal samples. By comparing their ability to assimilate cholesterol, we found that the transformation ability was not specific to any particular species. Notably, Bacteroides dorei YGMCC0564 revealed superior cholesterol-lowering capabilities and bile salt hydrolase (BSH) activity in vitro, surpassing that of Lactobacillus GG (LGG). YGMCC0564 exhibited favorable probiotic characteristics, including high survival rate in vitro simulation of gastrointestinal digestion, excellent adhesion ability, susceptibility to antibiotics, absence of hemolysis or virulence genes, and substantial production of SCFAs. The strain also demonstrated remarkable bile salt deconjugation activities and upregulation of the BT_416 gene associated with cholesterol, providing insights into a possible molecular mechanism underlying its cholesterol-reducing activity. These findings establish YGMCC0564 as a promising NPG candidate for improving cardiovascular health.

Abstract Concrete-filled steel tube (CFST) columns are widely used in civil engineering because of their excellent bearing capacity; however, the reinforcement of CFST columns lacks effective measures. To strengthen CFST columns quickly and effectively, two methods, namely, winding FRP (fiber reinforced polymer) or steel strips, were explored in this work. Two unconfined CFST columns, eight FRP-strengthened CFST columns and four welded steel strip-strengthened CFST columns were manufactured and tested. The failure modes and axial load–strain curves of all specimens under compression load were concluded and compared. The effects of the primary parameters, such as FRP layers (1, 2, 3 and 4 layers) and steel strip thickness (3.0 and 6.0 mm), on the bearing capacity and deformation capacity were also investigated. The ultimate load of CFST columns increased from 28.72 to 64.16% after being confined by FRP with one to four layers. The ultimate load of the welded steel strip-strengthened CFST column with 3.0 mm steel strips and 6.0 mm steel strips increased by 28.46% and 49.82%, respectively, compared with the unconfined CFST column. Thus, the increase in FRP layers and steel strip thickness can markedly improve the compressive behavior of the FRP/welded steel strip-strengthened CFST columns. The cost performance of the two different reinforcement methods also showed that the cost of the welded steel strip-strengthened CFST column is nearly 40% of that of the FRP-strengthened CFST column when the same strengthening effect was obtained, which indicated that the welded steel strip-strengthened CFST column is more cost-efficient than CFST columns confined by FRP. Finally, six existing models for the ultimate load of FRP-strengthened CFST columns were presented and evaluated. From the evaluation results, the Zhang et al.’s model, Lu et al.’s model and Hu et al.’s model for FRP-strengthened CFST columns were shown to provide the best applicability and accuracy. Based on the Mander et al.’s model, a model for the ultimate load of welded steel strip-strengthened CFST columns was proposed and evaluated. The proposed model can accurately predict the ultimate load of welded steel strip-strengthened CFST columns.

IntroductionChronic viral hepatitis (CH) is a stage prior to cirrhosis and primary cancer. Standard protocols for CH assessment during the long follow-up period are of great importance for precise treatment and living quality improvement. In this study, we aimed to analyze multiple serum indexes in chronic hepatitis B (CHB)-infected patients and to discuss their combined values in clinical applications.MethodsTotal 503 lines of laboratory data from 2012 to 2021 were extracted from103 CHB patients who were followed-up in our hospital. They were divided into the remission group and the progression group according to their complete clinical information and laboratory data. A series of models of serum indexes were analyzed to illustrate the fluctuation trend of @ach index in a time-dependent manner.ResultsThe models revealed that abundant serum alpha-fetoprotein (AFP) in the remission group was characteristically associated with hepatocyte destruction markers aspartate aminotransferase (AST) and alanine aminotransferase and favored a much longer progression-free period (P 0.0001). A model-derived equation consisting of serum AFP and AST values showed a good performance (83% reliability) to distinguish the two groups.DiscussionThis study clearly demonstrates the intrinsic quantitative relationship between serum AFP and liver aminotransferases involving antivirus treatment response. The model-based equation compensates for serum hepatitis B virus DNA detection during outpatient follow-up and it may serve as a useful laboratory tool for CHB progression assessment.

Developing a new drug delivery system is one of the useful approaches to overcome the limited use of berberine (BBR) to enhance its absorption and bioavailability. We prepared a novel berberine–glycyrrhizic acid (BBR–GL) complex formulation to increase the plasma concentration and bioavailability of BBR by improving BBR solubility and lowering the absorption barrier. The complex formulation with BBR and GL in the ratio 1:1 was developed through the self-assembly process and evaluated in vitro. Compared with BBR and BBR/GL physical mixture, the BBR–GL complex showed different characteristics by SEM, DSC, FT-IR, and PXRD measurement. In pharmacokinetic evaluation, the BBR–GL complex significantly increased the plasma concentration of BBR and the major metabolite berberrubine (BBB), with the AUC of BBR elevated to 4.43-folds, while the complex was safe as BBR. Furthermore, doxorubicin (DOX) was used to induce cardiotoxicity. Hematological study, histopathological examinations, electrocardiography (ECG), cardiac secretion measurement, and biochemical index analysis proved that the model of doxorubicin-induced cardiotoxicity (DIC) was conducted successfully. With the AUC of BBR increasing in the BBR–GL complex and the absorbed complex itself, the BBR–GL complex enhanced prevention effect to DIC and exhibited a significant prevention effect to attenuate heart damage. Our findings demonstrated that a novel BBR-loaded BBR–GL complex formulation could increase BBR plasma concentration. Improvement of BBR bioavailability by the BBR–GL complex could coordinate with GL to attenuate DIC. Concerning the safety of the drug delivery system at present, the BBR–GL complex could be a potential therapeutic formulation for the prevention of cardiac damage in the clinical application of doxorubicin.

Context Xinmai 'an tablet has been used to improve myocardial blood supply. Recently, some compounds from its formula have shown that they can treat pulmonary arterial hypertension (PAH). Objective This study investigates the effects of Xinmai 'an extract (XMA) on PAH and further tests the co-therapeutic enhancement with sildenafil (SIL). Materials and methods Pulmonary artery smooth muscle cells were subjected to stimulation with SIL (12.5 μM) and XMA (250 μg/mL) for 48 h. Sprague–Dawley rats were randomly grouped into eight groups (n = 8 per group): (I) control group received saline; (II) MCT group received MCT (60 mg/kg); (III) SIL-Low group received MCT + SIL at 10 mg/kg/day; (IV) SIL-high group received MCT + SIL at 30 mg/kg/day; (V) XMA-High group received MCT + XMA at 251.6 mg/kg/day; (VI) SIL (Low)+XMA (Low) group received SIL (10 mg/kg) + XMA at 62.9 mg/kg/day; (VII) SIL (Low)+XMA (Medium) group received SIL (10 mg/kg) + XMA at 125.8 mg/kg/day; (VIII) SIL (Low)+XMA (High) group received SIL (10 mg/kg) + XMA at 251.6 mg/kg/day. Both XMA and SIL were given by gavage and were maintained daily for 2 weeks. Results XMA could improve SIL’s efficacy in the treatment of PAH by decreasing cell viability more effectively at non-cytotoxic concentrations (250 μg/mL) and reducing Right Ventricular Systolic Pressure (RVSP) in PAH rat. Potential mechanisms might at least in part be through activating the MAPK signalling pathway. Discussion and conclusions The combination of XMA and SIL can improve the efficacy of pulmonary hypertension and reduce the dosage of SIL.

Background Previously, we developed a monoclonal antibody (mAb) NJ001 that binds to the antigen SP70 in human non‐small cell lung cancer (NSCLC) cells and showed it could inhibit lung adenocarcinoma (AD) growth. Here, we investigated the effect and mechanisms of NJ001 in lung AD metastasis. Methods Human lung AD cells (SPC‐A1 and A549) were treated with different concentrations of mAb NJ001, and the effects of NJ001 on cell migration and invasive activity were investigated using wound‐healing and Matrigel assays, respectively. The molecular mechanism of this inhibition was explored by microarrays, qRT‐PCR, western blot, luciferase assays and electrophoretic mobility shift assays (EMSA). Results MAb NJ001 markedly suppressed lung AD cell migration; and the invasiveness of SPC‐A1 and A549 cells treated with mAb NJ001 was diminished by 65%. Tissue inhibitor of matrix metalloproteinase‐3 (TIMP‐3) was highly expressed in SPC‐A1 cells treated with mAb NJ001, whereas knockdown of TIMP‐3 by shRNA significantly increased SPC‐A1 and A549 invasiveness. MAb NJ001 affects lung AD by inhibiting TIMP‐3 through direct transcriptional regulation of FOXP1 binding sites in the TIMP‐3 promoter region, as shown in luciferase assays and EMSA. Conclusions MAb NJ001 inhibits invasiveness and metastasis in lung AD through the FOXP1 binding sites in the TIMP‐3 promoter region. It may have clinical applications in preventing and treating metastatic lung AD.

Abstract Background Chronic nonhealing wounds represent one of the most common complications of diabetes and require advanced treatment strategies. Increasing evidence supports the important role of mesenchymal stem cells in diabetic wound healing; however, the underlying mechanism remains unclear. Here, we explored the effects of umbilical cord-matrix stem cells (UCMSCs) on diabetic wound healing and the underlying mechanism. Methods UCMSCs or conditioned medium (UCMSC-CM) were injected into the cutaneous wounds of streptozotocin-induced diabetic mice. The effects of this treatment on macrophages and diabetic vascular endothelial cells were investigated in vivo and in vitro. Results Our results reveal that UCMSCs or UCMSC-CM accelerated wound healing by enhancing angiogenesis. The number of host macrophages recruited to the wound tissue by local infusion of UCMSCs was greater than that recruited by fibroblast transplantation or control. The frequency of M2 macrophages was increased by UCMSC transplantation or UCMSC-CM injection, which promoted the expression of cytokines derived from M2 macrophages. Furthermore, when cocultured with UCMSCs or UCMSC-CM, lipopolysaccharide-induced macrophages acquired an anti-inflammatory M2 phenotype characterized by the increased secretion of the cytokines interleukin (IL)-10 and vascular endothelial growth factor and the suppressed production of tumor necrosis factor-α and IL-6. UCMSC-CM-activated macrophages significantly enhanced diabetic vascular endothelial cell functions, including angiogenesis, migration, and chemotaxis. Moreover, the action of UCMSC-CM on macrophages or vascular endothelial cells was abrogated by the administration of neutralizing antibodies against prostaglandin E2 (PGE2) or by the inhibition of PGE2 secretion from UCMSCs. Conclusions Our findings demonstrate that UCMSCs can induce the functional restoration of vascular endothelial cells via the remodeling of macrophage phenotypes, which might contribute to the marked acceleration of wound healing in diabetic mice. Abstract Background MYB-CC transcription factors (TFs) genes have been demonstrated to be involved in the response to inorganic phosphate (Pi) starvation and regulate some Pi-starvation-inducible genes. However, their role in drought stress has not been investigated in bread wheat. In this study, the TaMYBsm3 genes, including TaMYBsm3-A, TaMYBsm3-B, and TaMYBsm3-D, encoding MYB-CC TF proteins in bread wheat, were isolated to investigate the possible molecular mechanisms related to drought-tolerance in plants. Results TaMYBsm3-A, TaMYBsm3-B, and TaMYBsm3-D were mapped on chromosomes 6A, 6B, and 6D in wheat, respectively. TaMYBsm3 genes belonged to MYB-CC TFs, containing a conserved MYB DNA-binding domain and a conserved coiled–coil domain. TaMYBsm3-D was localized in the nucleus, and the N-terminal region was a transcriptional activation domain. TaMYBsm3 genes were ubiquitously expressed in different tissues of wheat, and especially highly expressed in the stamen and pistil. Under drought stress, transgenic plants exhibited milder wilting symptoms, higher germination rates, higher proline content, and lower MDA content comparing with the wild type plants. P5CS1, DREB2A, and RD29A had significantly higher expression in transgenic plants than in wild type plants. Conclusion TaMYBsm3-A, TaMYBsm3-B, and TaMYBsm3-D were associated with enhanced drought tolerance in bread wheat. Overexpression of TaMYBsm3-D increases the drought tolerance of transgenic Arabidopsis through up-regulating P5CS1, DREB2A, and RD29A.

Currently, in a target detection framework, a feature pyramid is widely used to capture differences resulting from the change in size of a detected object. Examples include the one-stage detector, the Deconvolutional Single-Shot Detector (DSSD), the RefineDet and the two-stage detector DetNet. Although these target detection frameworks with characteristic pyramid structure have achieved good results, they are limited by the facts that 1) they only use one feature layer of multi-scale information for prediction, and 2) they do not fully incorporate multi-scale inclusion of feature information. Therefore, based on Single-shot detector (SSD) architecture, we propose a novel one-stage target detector framework-fusion shared network (FSNet) which makes full use of feature information between multiple scale feature maps to more effectively detect objects of different scales. First, we introduce a fusion attention mechanism to fuse features between feature layers of different scales. Secondly, we combine this with multi-head structure fusion multi-layer features extracted as new features by a backbone network. Finally, we feed multi-layer fusion features into shared prediction module (shared PM). In the module, a new class feature pyramid for target detection is formed wherein the feature map used for prediction is composed of multiple levels of feature layers. In order to verify the effectiveness of this new FSNet target detection algorithm, we used the 320X320 input for FSNet, obtaining 80.7% mean accuracy (mAP) and 15 frames per second (Frame Per Second, FPS) in the PASCAL VOC 2007 test.

Prey vary dramatically in quality, and maternal diet is generally assumed to substantially influence offspring survivorship, growth, and reproduction in spiders. Numerous studies that have tested this hypothesis have focused exclusively on parental generation or have considered relatively few fitness components of juvenile offspring. However, maternal diet may have a substantial effect on fitness performance beyond juvenile offspring. Here, we investigated the influence of one-time maternal feeding on multiple offspring fitness components, including the survival rate and growth of juvenile offspring as well as the mating and reproductive success of adult offspring in Hylyphantes graminicola, a sheetweb spider with an extremely short lifespan (∼1 month). We fed field-collected adult female spiders two different diets only once immediately before oviposition: midges (Tendipes sp.) only (MO) or flies (Drosophila melanogaster) only (FO). Juvenile offspring of MO females had significantly higher survival rate, faster growth, and larger male size at maturity than FO offspring. Although maternal diet did not significantly influence mating behavior or fecundity of female offspring overall, those of MO females laid eggs earlier and their eggs also hatched earlier and had a higher hatching rate than those of FO females. Intriguingly, one-time maternal feeding was sufficient to have such an influence on offspring fitness even beyond juvenile offspring in H. graminicola. This one-time maternal effect may be widespread in other spiders and other invertebrates with a short lifespan. Radioactive strontium 90Sr with a half-life of 29 years is one of the most concerned elements in both nuclear accidents and spent fuel reprocessing. Herein, using macroporous silica SiO2 as support, methylacrylic acid as functional monomer and ethylene glycol dimethacrylate as cross-linking agent, a novel resin named SiMaC was prepared via in situ polymerization method for strontium removal. The as-preapred SiMaC was characterized by SEM-EDS, TG-DSC, and BET. Various factors such as pH, m/V, time, and initial concentration of Sr(II) were studied. The maximum adsorption capacity towards Sr(II) was as high as 142.5 mg/g at 298 K. The adsorption equilibrium was almost obtained within 30 min. Moreover, SiMaC was easily and fastly regenerated by 0.1 M HCl and exhibited no obvious decrease in adsorption capacity after five times of successive adsorption-desorption cycles. Most attractively, the uptake rate of Sr(II) reached more than 99% in real river and lake water mediums, as well as reached 76% in real seawater mediums. Human cytomegalovirus (HCMV) is a double-stranded DNA virus that belongs to the β-herpesvirus family and infects 40–90% of the adult population worldwide. HCMV infection is usually asymptomatic in healthy individuals but causes serious problems in immunocompromised people. We restricted this narrative review (PubMed, January 2022) to demonstrate the interaction and molecular mechanisms between the virus and host immune cells with a focus on HCMV-encoded miRNAs. We found a series of HCMV-encoded miRNAs (e.g., miR-UL112 and miR-UL148D) are explicitly involved in the regulation of viral DNA replication, immune evasion, as well as host cell fate. MiRNA-targeted therapies have been explored for the treatment of atherosclerosis, cardiovascular disease, cancer, diabetes, and hepatitis C virus infection. It is feasible to develop an alternative vaccine to restart peripheral immunity or to inhibit HCMV activity, which may contribute to the antiviral intervention for serious HCMV-related diseases.

Abstract SP70 is a novel tumor biomarker in patients with nonsmall cell lung cancer (NSCLC). However, its role as a marker for predicting the response to chemotherapy for patients with advanced NSCLC has not been investigated. A total of 152 patients were enrolled. Serum SP70, carcinoembryonic antigen (CEA), cytokeratin 19 fragment (CYFRA21‐1), and neuron‐specific enolase (NSE) were detected before and after 2 cycles of chemotherapy. The correlation between serum tumor biomarker levels and chemotherapy responses and their association with epidermal growth factor receptor (EGFR) mutation status and progression‐free survival (PFS) were analyzed. Anticardiolipin antibody (ACA) is a kind of autoantibody and is one of the antiphospholipid antibodies (aPLs). Phospholipids with a negative charge on platelets and endothelial cell membranes are ACA target antigens. ACA is common in systemic lupus erythematosus and other autoimmune diseases and is closely associated with thrombosis, thrombocytopenia, and spontaneous abortion. In 1983, Harris established a method for detecting ACA, and research on the antibody has gained worldwide attention and has developed rapidly. For this review, we browsed articles that cover most of the ACA-related studies in the last 25 years and extracted influential ideas and conclusions in this field.

The mechanism for pathogenesis of human papillomavirus (HPV) in the cervix has been investigated intensively. However, detailed differences in the distribution and function of innate immune cells between high-risk HPV types, especially during the chronic inflammation phase, have not been described fully. In this study, histologic pathology results of 245 women with HPV type 16 only (HPV16+) or type 18 only (HPV18+) were analyzed retrospectively from January 2015 to November 2016. More severe lesions of the cervix were observed in HPV16+ women compared with those in HPV18+ women. In total, 212 cervical brush specimens were collected from women suffering from chronic inflammation, HPV16+, or HPV18+ from December 2016 to December 2018. Flow cytometry analysis showed that abundant NK cells along with aberrant Treg cells were found in the HPV16-infected cervix. Quantitative real-time PCR demonstrated that higher expression levels of IFN-γ but muted IL-2 and KLRG-1 expression was detected in the cervix of patients with HPV16+ compared to HPV18+, which were further confirmed using 20 paraffin sections of cervical conization tissue. The ex vivo cytotoxicity experiment showed that the cytotoxicity of NK cells was significantly decreased in the cervix of HPV16+ patients compared with that of HPV18+ patients. Collectively, our results suggested that HPV16 disables the increased NK cells in the early lesion of the cervix, indicating that the local immune system of the cervix is hyporesponsive to HPV16 infection and this may explain its bias for malignant transformation.

Mesenchymal stem cells (MSCs) and hepatocytes are two attractive sources of cell-based therapies for acute liver failure (ALF). The cotransplantation of hepatocytes with MSCs can improve the therapeutic performance for the treatment of ALF. However, the therapeutic potential of conditioned medium (CM) derived from MSCs cocultured with hepatocytes (MSC-H-CM) remains unclear. The purpose of this study was to investigate the effects of MSC-H-CM on damaged hepatocytes in vitro and on D-galactosamine-induced ALF in vivo. D-Galactosamine-treated L02 cells cultured in MSC-H-CM exhibited higher of cell viability and total protein synthesis than L02 cells cultured in MSC-CM, CM derived from hepatocytes (H-CM), MSC-CM + H-CM, or with nonconditioned medium (NCM). Lactate dehydrogenase and aspartate aminotransferase levels were lower in the supernatant of damaged L02 cells cultured in MSC-H-CM than in that of L02 cells cultured in other types of CM. The lowest percentage of apoptotic cells was observed after the MSC-H-CM treatment. When CM was injected into the tail vein of rats with ALF, MSC-H-CM was the most successful at preventing the release of liver injury biomarkers and in promoting the recovery of liver structure. The greatest survival rate 7 days after the first treatment was observed in the MSC-H-CM-treated rats. Our results reveal that the delivery of MSC-H-CM could be a novel strategy for integrating the therapeutic potentials of hepatocytes and MSCs for the treatment of ALF.

This study proposes a multiheaded object detection algorithm referred to as MANet. The main purpose of the study is to integrate feature layers of different scales based on the attention mechanism and to enhance contextual connections. To achieve this, we first replaced the feed-forward base network of the single-shot detector with the ResNet−101 (inspired by the Deconvolutional Single-Shot Detector) and then applied linear interpolation and the attention mechanism. The information of the feature layers at different scales was fused to improve the accuracy of target detection. The primary contributions of this study are the propositions of (a) a fusion attention mechanism, and (b) a multiheaded attention fusion method. Our final MANet detector model effectively unifies the feature information among the feature layers at different scales, thus enabling it to detect objects with different sizes and with higher precision. We used the 512 × 512 input MANet (the backbone is ResNet−101) to obtain a mean accuracy of 82.7% based on the PASCAL visual object class 2007 test. These results demonstrated that our proposed method yielded better accuracy than those provided by the conventional Single-shot detector (SSD) and other advanced detectors.

The role of background matching in camouflage has been extensively studied. However, contour modification has received far less attention, especially in twig-mimicking species. Here, we studied this deceptive strategy by revealing a special masquerade tactic, in which the animals protract and cluster their legs linearly in the same axis with their bodies when resting, using the spider Ariamnes cylindrogaster as a model. We used cardboard papers to construct dummies resembling spiders in appearance and colour. To differentiate the most important factors in the concealment effect, we manipulated body size (long or short abdomen) and resting postures (leg clustered or spread) of the dummies and recorded the responses of predators to different dummy types in the field. The results showed that dummies with clustered legs received significantly less attention from predators, regardless of the body length. Thus, we conclude that A. cylindrogaster relies on the resting posture rather than body size for predator avoidance. This study provides, to the best of our knowledge, empirical evidence for the first time that twig-mimicking species can achieve effective camouflage by contour modification.

Apert syndrome (AS) is a type of autosomal dominant disease characterized by premature fusion of the cranial sutures, severe syndactyly, and other abnormalities in internal organs. Approximately 70% of AS cases are caused by a single mutation, S252W, in fibroblast growth factor receptor 2 (FGFR2). Two groups have generated FGFR2 knock-in mice Fgfr2S252W/+ that exhibit features of AS. During the present study of AS using the Fgfr2S252W/+ mouse model, an age-related phenotype of bone homeostasis was discovered. The long bone mass was lower in 2 month old mutant mice than in age-matched controls but higher in 5 month old mutant mice. This unusual phenotype suggested that bone marrow-derived mesenchymal stem cells (BMSCs), which are vital to maintain bone homeostasis, might be involved. BMSCs were isolated from Fgfr2S252W/+ mice and found that S252W mutation could impair osteogenic differentiation BMSCs but enhance mineralization of more mature osteoblasts. A microarray analysis revealed that Wnt pathway inhibitors SRFP1/2/4 were up-regulated in mutant BMSCs. This work provides evidence to show that the Wnt/β-catenin pathway is inhibited in both mutant BMSCs and osteoblasts, and differentiation defects of these cells can be ameliorated by Wnt3a treatment. The present study suggested that the bone abnormalities caused by deregulation of Wnt pathway may underlie the symptoms of AS.

Chemoreception is a key feature in selection of host plant by phytophagous insects, and odorant-binding proteins (OBPs) are involved in chemical communication of both insects and vertebrates. The legume pod borer, Maruca vitrata Fabricius (Lepidoptera: Crambidae) is one of the key pest species of cowpea and widely distributed throughout tropical and subtropical regions, causing up to 80% of yield loss. In this study, we investigated the electrophysiological responses of female M. vitrata to floral volatiles from V. unguiculata. Seventeen electroantennogram-active compounds were identified from floral volatiles of V. unguiculata by coupled gas chromatography-electroantennography (GC-EAD) and gas chromatography-mass spectrometry (GC-MS). Then, we cloned two novel full-length GOBP genes (MvitGOBP1 and MvitGOBP2) from the antennae of M. vitrata using reverse transcription PCR. Protein sequence analysis indicated that they shared high sequence similarity with other Pyralididae insect GOBPs and had the typical six-cysteine signature. Real-time PCR analysis indicated that MvitGOBP1-2 mRNA was highly expressed in the antennae of female adult with several thousands-fold difference compare to other tissue. Next, the recombinant MvitGOBP1-2 was expressed in Escherichia coli and purified using Ni ion affinity chromatography. Fluorescence binding assays demonstrated that MvitGOBP1-2 had different binding affinities with 17 volatile odorant molecules including butanoic acid butyl ester, limonene, 4-ethylpropiophenone, 1H-indol-4-ol, butanoic acid octyl ester and 2-methyl-3-phenylpropanal. In the field trapping experiment, these six floral volatiles could effectively attract female moths and showed significant difference compared with the blank lure. These results suggested that MvitGOBPs and the seventeen floral volatiles are likely to function in the olfactory behavior response of female moths, which may have played crucial roles in the selection of oviposition sites. The six compounds that we have identified from the volatiles of V. unguiculata may provide useful information for exploring efficiency monitoring and integrated pest management strategies of this legume pod borer in the field.

A S252W mutation of fibroblast growth factor receptor 2 (FGFR2), which is responsible for nearly two-thirds of Apert syndrome (AS) cases, causes retarded development of the skeleton and skull malformation resulting from premature fusion of the craniofacial sutures. We utilized a Fgfr2(+/S252W) mouse (a knock-in mouse model mimicking human AS) to demonstrate decreased bone mass due to reduced trabecular bone volume, reduced bone mineral density, and shortened growth plates in the long bones. In vitro bone mesenchymal stem cells (BMSCs) culture studies revealed that the mutant mice showed reduced BMSC proliferation, a reduction in chondrogenic differentiation, and reduced mineralization. Our results suggest that these phenomena are caused by up-regulation of p38 and Erk1/2 phosphorylation. Treatment of cultured mutant bone rudiments with SB203580 or PD98059 resulted in partial rescue of the bone growth retardation. The p38 signaling pathway especially was found to be responsible for the retarded long bone development. Our data indicate that the S252W mutation in FGFR2 directly affects endochondral ossification, resulting in growth retardation of the long bone. We also show that the p38 and Erk1/2 signaling pathways partially mediate the effects of the S252W mutation of FGFR2 on long bone development.

BACKGROUND: Males usually produce mating plugs to reduce sperm competition. However, females can conceivably also produce mating plugs in order to prevent unwanted, superfluous and energetically costly matings. In spiders-appropriate models for testing plugging biology hypotheses-mating plugs may consist of male genital parts and/or of amorphous covers consisting of glandular or sperm secretions. In the giant wood spider Nephila pilipes, a highly sexually dimorphic and polygamous species, males are known to produce ineffective embolic plugs through genital damage, but nothing is known about the origin and function of additional conspicuous amorphous plugs (AP) covering female genitals. METHODOLOGY: We tested alternative hypotheses of the nature and function of AP in N. pilipes by staging mating trials with varying degrees of polyandry. No APs were ever formed during mating trials, which rules out the possibility of male AP formation. Instead, those females that oviposited produced the AP from a liquid secreted during egg sac formation. Polyandrous females were more likely to lay eggs and to produce the AP, as were those that mated longer and with more total insertions. Our further tests revealed that, in spite of being a side product of egg sac production, AP, when hardened, prevented any subsequent copulation. CONCLUSIONS: We conclude that in the giant wood spider (Nephila pilipes), the amorphous mating plugs are not produced by the males, that repeated copulations (most likely polyandrous) are necessary for egg fertilization and AP formation, and that the AP represents a female adaptation to sexual conflict through prevention of unwanted, excessive copulations. Considering the largely unknown origin of amorphous plugs in spiders, we predict that a similar pattern might be detected in other clades, which would help elucidate the evolutionary interplay of various selection pressures responsible for the origin and maintenance of mating plugs.

Behavioral plasticity has been proposed as a means by which animals alter their phenotypes in response to changing conditions. Animals may display behavioral plasticity as a consequence of environmental variation. The detritus-based, bell-shaped cobweb spider Campanicola campanulata is an ideal model to study behavioral plasticity, because its web architecture is easy to be quantified, and the functions of different parts of the web are clear. Though the plasticity of cobweb architecture has been reported in a few species, retreats as important defensive structures have rarely been considered before because retreats in most cobwebs are relatively small compared with the web size. We studied the web-building behaviors of C. campanulata under different feeding regimes. We set up 3 spider treatments with different feeding conditions: marginally well fed, moderately well fed, and extremely well fed, and observed the differences in the web architecture among them. In addition, we measured the mechanical properties of anchor silk, and also calculated the foraging and defense investment of the spiders. The results showed that marginally well-fed spiders build cobwebs with significantly longer length of anchor silk, lower retreat to the ground, more number and longer gumfooted lines, and larger capture area, while extremely well-fed spiders build cobwebs with significantly bigger retreat volume and higher height of retreat to the ground. In addition, marginally well-fed spiders invest significantly less during cobweb construction. However, there was no significant difference between the breaking force and elongation at break in anchor silk among different treatments. These results demonstrated that marginally well-fed spiders invest more in foraging, and extremely well-fed spiders invest more in defense, and the spider made a balance between foraging and predator avoidance in response to changes in physiological state. As one of the most important invasive and destructive pests in the world, the fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae) reduces crop yield and causes economic loss. It is critical to find an appropriate controlling approach. This study aimed to determine the insecticidal activities of botanical insecticides (30% tea saponin, 0.6% matrine and a mixture thereof) on fall armyworm larvae, as well as the effects of sublethal concentrations of an optimal ratio of the mixture, on the survival rate, developmental duration, pupation rate, emergence rate, and body weight of fall armyworm. In addition, we evaluated the control effect of the insecticides (30% tea saponin, 0.6% matrine, optimal ratio of mixture along with 0.3% azadirachtin and 36% chlorfenapyr) and toxicity to natural enemies in field conditions. The results showed that all botanical insecticides and mixtures were toxic, and the insecticidal bioactivity of 10:1 volume ratio mixture was the highest at 96 hours, with a median lethal concentration (LC50) of 56.561 mg/mL. Meanwhile, the sublethal concentration of a 10:1 volume ratio of the mixture significantly affected the survival rate, prolonged developmental duration and reduced emergence rates and pupal weights, but it did not affect the pupation rates of fall armyworm. Furthermore, it has harmless or slightly harmful effects on natural enemies. Our study provides a novel environmentally friendly and natural enemy-friendly approach to controlling fall armyworms. Topics covered include the damage that males make to the inner walls of the female genital tract, the impact of seminal fluid on the suppression of female mating, and the genital tract injury that greatly suppressed female willingness to mate as well as the development of mating plugs that prevented mating.

Sexual conflict is common in animals, and female sexual cannibalism represents an extreme form of sexual conflict. Males in many species have evolved a variety of strategies to circumvent or decrease the risk of female sexual cannibalism. Opportunistic mating, by which a male mates with a female when she is disturbed or when she is feeding or undertaking moulting, is one of such kinds of strategies, and widely occurs in many animals, especially in spiders. However, whether the occurrence of male opportunistic mating depends on the intensity of female sexual cannibalism remains largely unexplored. We predicted a positive correlation between them. In this study, we tested this prediction by performing a series of mating trials in the laboratory using 3 species of web-building spiders with different intensities of female sexual cannibalism: Nephila pilipes , Nephilengys malabarensis , and Parasteatoda tepidariorum. We found that the occurrence of male opportunistic mating was positively, though not statistically significantly, correlated with the intensity of female sexual cannibalism, thus supporting our hypothesis. All together, we provide evidence that male opportunistic mating may have evolved to respond to the selection pressure posed by female sexual cannibalism. [ABSTRACT FROM AUTHOR]
