Fresh medicine is the use of fresh Chinese herbs after mining. The aim of the present work was to investigate the feasibility of ultra-high-pressure processing in the fresh medicine field. The typical fresh medicine Dendrobium officinale was used as the research material. The effects of pressure level (100 - 500 MPa), processing duration (0.5 - 2.5 min), number of processing cycle (1 - 5), and temperature (20 - 100°C) on the polysaccharide loss (in pressure transmitting medium) and dissolution efficiency (in boiling water) were studied. The results show that all parameters not only caused a dramatic loss of polysaccharides but also significantly increased the dissolution quantity, with the largest loss resulting from the number of repetitive processing cycles, which reached 47.11% after five processing steps. Analysis of the infrared spectroscopy and antioxidant activity results showed that the structure and antioxidant activity of the polysaccharides extracted from the stems processed by the parameters with the maximum value were markedly different, and the antioxidant activity of the polysaccharides extracted from the stems processed five times was the weakest. Ultra-high-pressure processing has good prospects in fresh medicine, while further research on process optimisation is necessary. [ABSTRACT FROM AUTHOR]

OBJECTIVE: The aim of this study was to investigate the influence of long non-coding ribonucleic acid (lncRNA) urothelial carcinoma associated 1 (UCA1) on glucose metabolism in rats with diabetic nephropathy (DN), and to explore its regulatory mechanism. MATERIALS AND METHODS: A total of 30 healthy Sprague-Dawley (SD) rats were selected in this study. All rats were randomly divided into three groups, including the control group, the model group, and the lncRNA UCA1 inhibitor group. The rat model of DN was successfully established via intraperitoneal injection of streptozotocin (STZ). The pathological changes in kidney tissues were detected via hematoxylin-eosin (HE) staining. The levels of blood urea nitrogen (BUN), serum creatinine (Scr), and urinary protein (UP) were detected using the biochemical method. Meanwhile, the content of serum tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) was detected via enzyme-linked immunosorbent assay (ELISA). In addition, the messenger RNA (mRNA) and protein levels of phosphatidylinositol 3-hydroxy kinase (PI3K) and protein kinase B (Akt) in kidney tissues were detected via reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting, respectively. RESULTS: The model group showed severe pathological damage to the kidney, compared with the control group. Meanwhile, the levels of BUN, Scr and UP, and the content of serum TNF-α and IL-6 increased significantly in the model group. The mRNA and the protein levels of PI3K and Akt in kidney tissues of the model group were significantly up-regulated as well. LncRNA UCA1 inhibitor group exhibited relieved pathological damage to the kidney, compared with the model group. The levels of BUN, Scr and UP, and the content of serum TNF-α and IL-6 remarkably decreased in UCA1 inhibitor group. Furthermore, the mRNA and the protein levels of PI3K and Akt in kidney tissues of UCA1 inhibitor groups were significantly down-regulated. CONCLUSIONS: LncRNA UCA1 can relieve the pathological damage to the kidney, improve renal function, and alleviate inflammatory response in DN rats. The quantification of germanium in rice (Oryza sativa L.) was studied by atomic fluorescence spectrometry (AFS). The optimum experimental conditions or the generation of germanium hydride were 360-V photomultiplier tube voltage, 80-mA hollow cathode lamp, 2.5% (w/v) KBH4 in 0.5% KOH solution and 20% H3PO4 mixture with 5% H2SO4. With these experimental conditions, the correlation coefficient (R 2) of calibration curve was up to 0.9992. As a convenient and sensitive calibration curve, it was proposed for the determination of trace amounts of germanium in brown and milled rice samples. The results showed that the content of germanium in brown rice was higher than that in milled rice. The means of the content of germanium were 4.98 and 18.10 ng/g in milled rice and brown rice, respectively. PRACTICAL APPLICATIONS Rice is an important food for human nutrition because it supplies starch, protein and the majority of micronutrients to humans, particularly in Asia. So determination the content of Germanium in rice is important for human health. In this study, we developed the method of AFS which was suitable for determination rice grain. With this method, the content of Ge could be analyzed accurately in rice, so that the good materials would be selected form germplasm resources of rice, which could be used to study physiological and genetic mechanisms and enhance germanium in diets for human. The genetic diversity of 118 tobacco accessions, including flue-cured tobacco, sun-/air-cured tobacco, burley tobacco, oriental tobacco and wild tobacco, was characterised using intersimple sequence repeat (ISSR) and inter-retrotransposon amplification polymorphism (IRAP) markers. ISSR and IRAP banding patterns and genetic distance (GD) values showed the low level of genetic diversity within and among cultivated tobacco types. There was higher GD and average heterozygosity among wild tobacco types than those among cultivated tobacco. Genetic diversity of tobacco germplasm was low, with a high level of genetic identity (>0.77) between the different types. However, neighbour-joining cluster analysis of marker-based GDs showed that the accessions from the same tobacco type, as classified by manufacturing quality traits, were nearly clustered into the same group. These results will help in the formulation of appropriate strategies for variety improvement in tobacco, and ISSR and IRAP markers of the genetic diversity will contribute to further study and improvement of tobacco. Yield and its components were investigated by using a population of 241 recombinant inbred lines (F9 RILs) derived from an elite hybrid rice cross of‘Zhenshan 97’ × ‘Minghui 63’. Quantitative trait loci (QTLs) for causal analysing of yield traits were detected at different yield component (YC) influences by conditional and unconditional QTL mapping methods. The number of QTLs significantly affecting yield was different at component-special influence. Some QTLs controlling yield identified in one component influence were undetectable at the others. More QTLs for yield could be detected at different YC influences. It is possible to reveal that causal gene expression for yield could be different at different YC influences. Mapping QTLs for component effects of yield could help us in understanding the nature of cause-effect traits for the formation of grain yield. [ABSTRACT FROM AUTHOR]

The heterosis controlled by genetic main effects and genotype × environment (GE) interaction effects for protein content and lysine content traits of indica hybrid rice, Oryza sativa L., was studied by using a genetic model for quantitative traits of triploid endosperm. The experiment was conducted over 2 years in a factorial design that included nine cytoplasmic male-sterile lines as females and five restorer lines as males. It was revealed that heterosis of protein content and lysine content were simultaneously controlled by genetic main effects and GE interaction effects. Maternal general heterosis and maternal interaction heterosis were observed. Embryo heterosis or cytoplasm heterosis for lysine content and endosperm heterosis for protein content were more important in general heterosis. Embryo interaction heterosis and cytoplasm interaction heterosis were more important for protein content, but endosperm heterosis was only important for lysine content in GE interaction heterosis. It was shown that some indica hybrid crosses had significant positive heterosis for protein content. Negative heterosis for lysine content was observed in most hybrid crosses. [ABSTRACT FROM AUTHOR]

The objective of this study was to examine the clinical findings, magnetic resonance imaging (MRI), pathological features, and treatment experiments of patients with hypertrophic cranial pachymeningitis (HCP). The clinical findings, MRI, and pathological appearances of 9 patients with HCP were analyzed retrospectively. The thickened dura mater was markedly enhanced after contrast media injection. The lesion near the brain hemisphere presented long regions of T1- and T2-weighted abnormal signal intensities. The abnormal signal intensities of the brain tissue were decreased significantly. Pathological examination demonstrated chronic inflammation changes, with cerebral dura mater fibrous tissue showing obvious hyperplasia, and the periphery of the blood vessel showing a great quantity of infiltrating phlegmonosis cells. HCP mainly presents headache and paralysis of multiple cranial nerves. The distinctive signs on brain MRIs involve strengthening the signal in the cerebral dura.

We propose an all-fiber band-rejection filter with a tunable bandwidth, which is realized by putting a normal long-period fiber grating in series with a rotary long-period fiber grating written in a twisted single-mode fiber by CO(2) laser pulses. Bandwidth tuning is achieved by applying torsion to the composite grating. Our experimental filter shows a bandwidth tuning of approximately 16.3 nm at a rejection level of approximately 15 dB and a polarization-dependent loss lower than approximately 0.9 dB.

The relationships among potassium (K), calcium (Ca), sodium (Na), magnesium (Mg), iron (Fe), zinc (Zn), copper (Cu), and manganese (Mn) contents in milled rice (Oryza stavia L.) of 274 genotypes and the relationships between these mineral element contents and other rice quality traits including 3 cooking quality traits, 17 amino acid contents, and protein content were investigated. The results showed that there were significant correlations among most of mineral element contents. Mg, Fe, and Mn contents were significantly correlated with most of the other mineral element contents, while Cu content had significantly negative associations with the K and Mg contents of rice. The relationships between mineral element contents and cooking quality traits showed that gel consistency (GC) was significantly correlated with K, Cu, and Mn contents of rice. Amylose content (AC) was significantly associated with K, Na, Mg, Cu, and Mn contents. The alkali spreading value (ASV) had closely positive relationships with Ca, Mg, and Mn contents. In addition, 8 mineral element contents had obvious correlations with different amino acid contents. Mg, Ca, and Zn contents were significantly correlated with most of the 17 amino acid contents, but Na content did not correlate with amino acid contents except aspartic acid of rice. Furthermore, significant associations were found between protein content and Na, Mg, Zn, Cu, or Mn content. Six principal components were extracted to explain 84.50% of the total variances and contained the information provided by the original 29 variables according to the principal component analysis.

The dynamic changes of genetic effects, including main effects, and genotype x environment (GE) interaction effects on brown rice thickness (BRT) across environments were investigated by using the developmental genetic models. Seven cytoplasmic male sterile lines of indica rice (Oryza sativa L.) as females and five restoring lines as males were used in a factorial design to produce grains of F(1)s and F(2)s in two environments (years) for developmental genetic analysis. The results indicate that genetic effects, especially GE interaction effects of triploid endosperm genes, cytoplasm genes, and diploid maternal plant genes were important to the performance of BRT at various filling stages of rice. The BRT was genetically controlled by the net genetic effects of genes expressed at the early and late filling stages (1-7 days and 15-21 days after flowering, respectively). The differences in net genetic effects under different environments for endosperm, cytoplasm, and maternal plant genes were found, and the net GE interaction effects were more important to BRT at the early filling and mature stages of rice. Some net genetic effects, especially for net cytoplasm effects spasmodically expressed, were detected among filling stages. Higher additive and cytoplasm main effects, along with their interaction effects, were found, which would be useful for selection for BRT in breeding programs. The synthetically useful approaches to 9-acyloxy 1,5-dichloroanthracene derivatives are reported. The system selectively reduces the carbonyl group flanked by the peri substituents of the anthracenediones to give the corresponding 1,5-dichloro-9(10H)-anthracenone. Simple regioselective acylation of anthracenone is applied with appropriate acyl chlorides in CH2Cl2 with catalytic amount of pyridine to give the novel 9-acyloxy 1,5-dichloroanthracene derivatives. Considerable interest has developed in the mechanism of how anthracenone achieves this desirable selectivity. In an attempt to understand the mechanism of this reaction, solid-state structures of anthracene derivatives have been obtained. In addition, the inhibition of lipid peroxidation in model membranes was determined as was their ability to inhibit the telomere-addition function of the human telomerase enzyme together with their inhibition of the Taq polymerase enzyme. In contrast to (+)-alpha-tocopherol, 3b, 3c, 3d, 3g, and 3i do not enhance lipid peroxidation in model membranes. Implications for 9-acyloxy 1,5-dichloroanthracene analogues as potential anticancer agents are discussed.

Aim: To study the biochemical-pharmacological properties of the recombinant human butyrylcholinesterase (rhBChE) and thereby to size up the potential possibility of using it as a detoxifying agent in succinylcholine intoxication., Methods: CHO-dhfr cells were transfected with plasmids by electroporation. BChE activity was determined colorimetrically by 5, 5'-dithiobis-(2-nitrobenzoic acid) (DTNB) method. Antigenicity was estimated by enzyme-linked immunosorbent assay and Western blot., Results: The maximal expression amounted to 25.83 ng.h-1/10(6) cells. The rhBChE was highly similar to the native human BChE (nhBChE) in terms of its catalytic property, substrate affinity, inhibitor sensitivity, reactivation, stability, and immunoreactivity with anti-nhBChE antibodies. Mice challenged with 1.5 lethal dose of succinylcholine preincubated with rhBChE survived without any symptoms of intoxication., Conclusion: The rhBChE and nhBChE exhibit similar biochemical-pharmacological features. It is of potential value in practical use.

A unique EcoRI site was introduced at the 3' end of cholera toxin B subunit (CTB) gene by site-directed mutagenesis, polynucleotides encoding 120-145aa epitope of HBV PreS2 were chemically synthesized and fused to the 3' end of cholera toxin B subunit gene. The fused gene was over-expressed (about 30 micrograms ml-1) in E. coli, and more than 95% of the fusion protein was secreted into the medium. The fusion protein expressed was purified by affinity chromatography. The chimera protein obtained bound to ganglioside GM1, and had the antigenicity of both cholera toxin B subunit and HBV PreS2 as confirmed by ELISA. After mice were immunized intramuscularly with the fusion protein, anti-CTB antibody and anti-PreS2 antibody were produced. These results indicated that the fusion protein retained not only the biological function of CTB but also the antigenicity and the immunogenicity of cholera toxin B subunit and HBV PreS2 epitope. This work provided a sound basis for further studies on the construction of engineered peptide vaccine.

One case of gouty patient with aseptic loosening of cementless total hip arthroplasty was reported. Mechanical and biological factors may both play some roles in this ollsening hip. From both cup and stem, no direct evidence of loosening was found from pathologic aspect. Further study may need to define relationship between gout and loosening of total hip arthroplasty.

The complete nucleotide sequence has been obtained of the H1 hemagglutinin (HA) gene of a high-yielding (H) mutant of the A/NJ/11/76(H1N1) strain of swine influenza virus in studies of a viral reassortant (X-53a) bearing this gene. This determination has permitted comparison with human influenza H1N1 prototype viruses A/WSN/33 and A/PR/8/34, with which 80% and 94% amino acid homology was found between HA1 and HA2, respectively. Partial sequences have been determined for other viral reassortants containing either H or L (low-yielding phenotype) genes derived from A/NJ/11/76. Sequence of the HA1 region of an L mutant prototype was virtually completed and differed from that of the H mutant by only four amino acid changes. Sequence analysis of four other viruses was restricted to regions of the HA with which monoclonal antibodies capable of distinguishing L and H mutants are presumed to react. Therefore, changes in these sequences are relevant to changes in viral phenotype. Change at residue 155 from Gly to Glu is associated with change from L to H HA phenotype. This site, structurally equivalent to amino acid 158 on the Wiley et al. HA model [Wiley, D. C., Wilson, I. A. & Skehel, J. J. (1981) Nature (London) 289, 373-378] is near the tip of the HA monomer adjacent to the proposed receptor binding site and therefore credibly could influence both viral antigenicity and replication. Because both L and H variants exist in nature and because revertants may be selected in the laboratory as replication variants in the absence of immunoselection, these studies provide evidence for fortuitous antigenic change in association with change in biological function, which is determined by a single base change.

Restriction fragments of HBV-DNA, cleaved by endonuclease HhaI, containing HBcAg gene were trimmed by BAL-31 exonuclease to remove different lengths of the precore sequence. They were inserted into plasmid pUR222 at EcoRI site through synthetic linker ligation. Transformants in E. coli BMH7118 showing different levels of HBcAg gene expression were screened and analyzed for their nucleotide sequences in the junction region both by Maxam and Gilbert's chemical degradation method and by M13 chain termination method. Results of sequence analysis of different transformants revealed a partial palindromic (loop and stem) structure, at -7 to -35 nucleotide with regard to ATG of the HBcAg gene as position +1, which has dramatic effect on the level of expression of the inserted gene using the same promoter, SD sequence and identical N-terminus. The amount of HBcAg synthesized differed from 9% in the high expressing plasmid to less than 0.01% of the total cell proteins in the low expressing transformants. The findings were compared to results obtained by other workers in studies of HBcAg expression in procaryotes and their significance in the expression of eucaryotic genes in procaryotic cells were discussed.

The nucleotide sequences of cloned adw hepatitis B core antigens (HBcAg) which have different levels of expression have been determined by dideoxy chain-terminating method. The results indicate that the different levels of gene expression is primarily due to the different structures of upstream sequences before the initiator of HBcAg genes. The hepatitis B core gene in the low-expressed clone forms a secondary structure before the initiator. This secondary structure has been removed by Bal-31 exonuclease in the high-expressed clone and half of the structure removed in the mid-expressed clone. It is apparent that the transcription/translation of the hepatitis B core gene is somehow blocked because of the secondary structure. Therefore, the amount of HBcAg synthesized in E. coli is dramatically reduced.
