Urbanization is expected to alter animal communities, but few studies have generalized beyond patterns in individual cities. We used community-science data on reptiles and amphibians from 17 cities in North America to determine: 1) how species richness differs between cities and surrounding areas, 2) whether the degree of landcover change in cities vs. surroundings is related to the magnitude of change in species richness, 3) whether urban communities are more similar to each other or to their surrounding areas, and 4) what characteristics of reptile and amphibian species predict how sensitive they are to urbanization. We found that species richness tended to be somewhat lower (10–20%) in cities as compared to surrounding areas. For reptiles, the magnitude of decrease in richness appeared to be primarily related to the degree of change in impervious surface cover, whereas for amphibians these changes tracked differences in canopy cover. There was little evidence of urban homogenization– city communities tended to cluster with their surrounding areas rather than with other cities. For individual species, those that were less represented in observations from cities tended to be more dependent on canopy cover and on specialized breeding habitats such as streams or vernal pools. Our results suggest that cities are capable of supporting a relatively high richness of reptiles and amphibians. However, maintenance of this richness may require attention to habitat protection and restoration, particularly for less common aquatic habitats.

We describe a directed search for continuous gravitational waves in data from the sixth LIGO science run. The target was the nearby globular cluster NGC 6544 at a distance of 2.7 kpc. The search covered a broad band of frequencies along with first and second frequency derivatives for a fixed sky position. The search coherently integrated data from the two LIGO interferometers over a time span of 9.2 days using the matched-filtering F-statistic. We found no gravitational-wave signals and set 95% confidence upper limits as stringent as 6.0 X 10^{-25} on intrinsic strain and 8.5 X 10^{-6} on fiducial ellipticity. These values beat the indirect limits from energy conservation for stars with characteristic spindown ages older than 300 years and are within the range of theoretical predictions for possible neutron-star ellipticities. An important feature of this search was use of a barycentric resampling algorithm which substantially reduced computational cost; this method will be used extensively in searches of Advanced LIGO and Virgo detector data., Comment: 15 pages, 4 figures

This paper presents updated estimates of source parameters for GW150914, a binary black-hole coalescence event detected by the Laser Interferometer Gravitational-wave Observatory (LIGO) in 2015 [Abbott et al. Phys. Rev. Lett. 116, 061102 (2016).]. Abbott et al. [Phys. Rev. Lett. 116, 241102 (2016).] presented parameter estimation of the source using a 13-dimensional, phenomenological precessing-spin model (precessing IMRPhenom) and an 11-dimensional nonprecessing effective-one body (EOB) model calibrated to numerical-relativity simulations, which forces spin alignment (nonprecessing EOBNR). Here, we present new results that include a 15-dimensional precessingspin waveform model (precessing EOBNR) developed within the EOB formalism. We find good agreement with the parameters estimated previously [Abbott et al. Phys. Rev. Lett. 116, 241102 (2016).], and we quote updated component masses of 35 +5 -3 M· and 30 +5 -3 M· (where errors correspond to 90% symmetric credible intervals). We also present slightly tighter constraints on the dimensionless spin magnitudes of the two black holes, with a primary spin estimate < 0.65 and a secondary spin estimate < 0.75 at 90% probability. Abbott et al. [Phys. Rev. Lett. 116, 241102 (2016).] estimated the systematic parameter-extraction errors due to waveform-model uncertainty by combining the posterior probability densities of precessing IMRPhenom and nonprecessing EOBNR. The gravitational-wave signal GW150914 was first identified on Sept 14 2015 by searches for short-duration gravitational-wave transients. These searches identify time-correlated transients in multiple detectors with minimal assumptions aboutthe signal morphology, allowing them to be sensitive to gravitational waves emitted by a wide range of sources including binary black-hole mergers. Over the observational period from September 12th to October 20th 2015, these transient searches were sensitive to binary black-hole mergers similar to GW150914 to an average distance of $\sim 600$ Mpc. In this paper, we describe the analyses that first detected GW150914 as well as the parameter estimation and waveform reconstruction techniques that initially identified GW150914 as the merger of two black holes. We find that the reconstructed waveform is consistent with the signal from a binary black-hole merger with a chirp mass of $\sim 30 \, M_\odot$ and a total mass before merger of $\sim 70 \, M_\odot$ in the detector frame., Comment: 20 pages, 12 figures, https://dcc.ligo.org/LIGO-P1500229/public

Effective gene therapy for the severe hemoglobin (Hb) disorders, sickle-cell anemia and thalassemia, will require an efficient method to transfer, integrate, and express a globin gene in primary erythroid cells. To evaluate recombinant adeno-associated virus (rAAV) for this purpose, we constructed a rAAV vector encoding a human gamma-globin gene (pJM24/vHS432A gamma). Its 4725-nucleotide genome consists of two 180-bp AAV inverted terminal repeats flanking the core elements of hypersensitive sites 2, 3, and 4 from the locus control region of the beta-globin gene cluster, linked to a mutationally marked A gamma-globin gene (A gamma) containing native promoter and RNA processing signals. CD34+ human hematopoietic cells were exposed to rAAV particles at a multiplicity of infection of 500-1000 and cultured in semisolid medium containing several cytokines. A reverse transcriptase polymerase chain reaction assay distinguished mRNA signals derived from transduced and endogenous human gamma-globin genes. Twenty to 40% of human erythroid burst-forming unit-derived colonies expressed the rAAV-transduced A gamma-globin gene at levels 4-71% that of the endogenous gamma-globin genes. The HbF content of pooled control colonies was 26%, whereas HbF was 40% of the total in pooled colonies derived from rAAV transduced progenitors. These data establish that rAAV containing elements from the locus control region linked to a gamma-globin gene are capable of transferring and expressing that gene in primary human hematopoietic cells resulting in a substantial increase in HbF content.

In order to clarify common assumptions on the form of energy and momentum in elasticity, a generalized conservation format is proposed for finite elasticity, in which total energy and momentum are not specified a priori. Velocity, stress, and total energy are assumed to depend constitutively on deformation gradient and momentum in a manner restricted by a dissipation principle and certain mild invariance requirements. Under these assumptions, representations are obtained for energy and momentum, demonstrating that (i) the total energy splits into separate internal and kinetic contributions, and (ii) the momentum is linear in the velocity. It is further shown that, if the stress response is strongly elliptic, the classical specifications for kinetic energy and momentum are sufficient to give elasticity the standard format of a quasilinear hyperbolic system. An aerosol with a high number density of particles with droplet sizes smaller than 3 mu m in diameter is produced by rapidly expanding an emulsion comprised of an aqueous solution and supercritical fluid CO2 at pressures from 1071 to 5000 psi at 35degree to 100degreeC. After dilution with air, fine droplet aerosols of aqueous pharmaceuticals, such as rhDNase, are suitable for pulmonary drug delivery. Dehydration of the aqueous droplets yields fine-particle dry powders, potentially important in pharmaceutical processing and in the generation of fine particles of metal oxides by pyrolysis of metal nitrates or acetates. Thin films of metals or metal oxides can be formed by directing an aerosol containing coating precursors onto heated surfaces. [ABSTRACT FROM AUTHOR]

36. Opiate and cocaine exposed newborns: growth outcomes.

- Abstract
This investigation examined growth parameters at birth in 204 infants born to mothers who used cocaine and/or opiates during pregnancy. Analyses considered both type (cocaine, opiate or both) and pattern of in utero drug exposure. A unique feature of the investigation was the large group of opiate exposed infants. Singleton newborn infants born to cocaine and/or opiate using mothers, were recruited. Using a structured interview and urine toxicology screens, information was obtained on the type and pattern of in utero drug exposure for each infant. Outcome measures included birth weight, length, and head circumference. Birth weight and length were significantly different by type of drug exposure with the opiate only infants the largest (p = .0001) and longest (p = .008). Differences in head circumference size were not statistically significant (p = .58). Mean Zscores were 1 S.D. lower for birth weight and length and 1.5 S.D. lower for head circumference when compared to National Center for Health Statistics (NCHS) growth standards. This study provides support that in utero cocaine exposure may confer more risk for somatic growth retardation at birth than opiate exposure even when controlling for nicotine and alcohol exposure, amount of prenatal care, gender, maternal age, education and marital status. Neuropathological examinations were carried out at necropsy on 274 cases of intrauterine death or neonatal death at or before three days after birth. Fifty six (20.4%) subjects had evidence of prenatal ischaemic brain damage. On review of the maternal case notes to ascertain antenatal clinical associations there was an increased incidence of intrauterine growth retardation, either based on birth weight for gestational age (odds ratio (OR) 2.0; 95% confidence interval (CI) 1.1 to 3.7) or diagnosed antenatally (OR 2.7; 95% CI 1.3 to 5.6). Oligohydramnios was also more common (OR 2.9; 95% CI 1.2 to 7.0). The association of intrauterine growth retardation and white matter damage remained after excluding fetuses with a major congenital anomaly (OR 2.4; 95% CI 1.1 to 5.1). The findings suggest that chronic intrauterine hypoxia may be associated with damage to cerebral white matter among fetuses and infants who die. The relation between ischaemic white matter damage and cerebral palsy among survivors remains speculative. [ABSTRACT FROM PUBLISHER]

A putative retravirus was isolated from a cell culture derived by fusion of mink and dog cell lines with cultured placental cells from a patient with systemic lupus erythematosus. The virus was detected by reverse transcriptase assay of supernatant medium after prolonged subculturing. Study Objective: To determine if aerosolization of purified human plasma alpha 1-antitrypsin is an effective means for increasing lower respiratory anti-neutrophil-elastase defenses in alpha 1-antitrypsin deficiency.Design: Nonrandomized, before-and-after trial with a 7-day treatment period. Companion studies in animals to determine lung epithelial permeability to alpha 1-antitrypsin.Patients: Twelve patients with homozygous Z-type alpha 1-antitrypsin deficiency and mild to moderate emphysema.Interventions: Aerosol administration of human plasma alpha 1-antitrypsin, 100 mg every 12 hours for 7 days. Single, 100-mg aerosol dose to anesthetized sheep with indwelling thoracic lymph duct catheters for direct assessment of lung permeability.Measurements and Main Results: Treatment resulted in increased alpha 1-antitrypsin levels in the lung epithelial lining fluid (0.28 +/- 0.07 microM before therapy to 5.86 +/- 1.03 microM after therapy) and increased anti-neutrophil-elastase capacity (0.78 +/- 0.38 microM before therapy to 4.16 +/- 0.95 microM after therapy). Aerosolized alpha 1-antitrypsin diffused across the respiratory epithelium and entered lung interstitial lymph (in sheep) and reached the systemic circulation (in sheep and humans). No side effects were noted.Conclusion: Short-term aerosol administration of human plasma alpha 1-antitrypsin to patients with alpha 1-antitrypsin deficiency is safe and feasible, resulting in a return to normal of anti-neutrophil-elastase defenses in the lower respiratory tract. Bioprosthetic valve thrombosis (BPVT) is a recognised complication of prosthetic aortic valves and can be found in up to 13% of patients after transcatheter implantation. The mechanism of BPVT is not well known, abnormal flow conditions in the new and native sinuses and lack of functional endothelialisation are suspected causes. BPVT may result in valve dysfunction, possibly related to degeneration, and recurrence of patient symptoms, or remain subclinical. BPVT is best diagnosed at multiphase gated computed tomography (CT) angiography as the presence of reduced leaflet motion (RELM) and hypoattenuating aortic leaflet thickening (HALT). Although CT is used to exclude BPVT in symptomatic patients and those with increased valve gradients, the value of screening and prophylactic anticoagulation is debatable. [ABSTRACT FROM AUTHOR]

A six stage model was applied to a geographically defined population of 210 singleton children born at term who had a diagnosis of cerebral palsy at 5 years of age. Thirty five children were identified as those most likely to have cerebral palsy of intrapartum origin; in 26 of these there was evidence of suboptimal care. It is suggested that this simple model should be tested on populations of children with cerebral palsy and the underlying principles used when considering the likely cause of cerebral palsy in individual children. Abstract Background The role of patient race in medical decision-making is heavily debated. While some evidence suggests that patient race can be used by physicians to predict disease risk and determine drug therapy, other studies document bias and stereotyping by physicians based on patient race. It is critical, then, to explore physicians' attitudes regarding the medical relevance of patient race. Methods We conducted a qualitative study in the United States using ten focus groups of physicians stratified by self-identified race (black or white) and led by race-concordant moderators. Physicians were presented with a medical vignette about a patient (whose race was unknown) with Type 2 diabetes and untreated hypertension, who was also a current smoker. Participants were first asked to discuss what medical information they would need to treat the patient. Then physicians were asked to explicitly discuss the importance of race to the hypothetical patient's treatment. To identify common themes, codes, key words and physician demographics were compiled into a comprehensive table that allowed for examination of similarities and differences by physician race. Common themes were identified using the software package NVivo (QSR International, v7). Results Forty self-identified black and 50 self-identified white physicians participated in the study. All physicians - regardless of their own race - believed that medical history, family history, and weight were important for making treatment decisions for the patient. However, black and white physicians reported differences in their views about the relevance of race. Several black physicians indicated that patient race is a central factor for choosing treatment options such as aggressive therapies, patient medication and understanding disease risk. Moreover, many black physicians considered patient race important to understand the patient's views, such as alternative medicine preferences and cultural beliefs about illness. However, few white physicians explicitly indicated that the patient's race was important over-and-above medical history. Instead, white physicians reported that the patient should be treated aggressively regardless of race. Conclusions This investigation adds to our understanding about how physicians in the United States consider race when treating patients, and sheds light on issues physicians face when deciding the importance of race in medical decision-making.

50. Analysis of preterm deliveries below 35 weeks' gestation in a tertiary referral hospital in the UK. A case-control survey

Abstract Background Preterm birth remains a major public health problem and its incidence worldwide is increasing. Epidemiological risk factors have been investigated in the past, but there is a need for a better understanding of the causes of preterm birth in well defined obstetric populations in tertiary referral centres; it is important to repeat surveillance and identify possible changes in clinical and socioeconomic factors associated with preterm delivery. The aim of this study was to identify current risk factors associated with preterm delivery and highlight areas for further research. Findings We studied women with singleton deliveries at St Michael's Hospital, Bristol during 2002 and 2003. 274 deliveries between 23-35 weeks' gestation (preterm group), were compared to 559 randomly selected control deliveries at term (37-42 weeks) using standard statistical procedures. Both groups were >80% Caucasian. Previous preterm deliveries, high maternal age (> 39 years), socioeconomic problems, smoking during pregnancy, hypertension, psychiatric disorders and uterine abnormalities were significantly associated with preterm deliveries. Both lean and obese mothers were more common in the preterm group. Women with depression/psychiatric disease were significantly more likely to have social problems, to have smoked during pregnancy and to have had previous preterm deliveries; when adjustments for these three factors were made the relationship between psychiatric disease and pregnancy outcome was no longer significant. 53% of preterm deliveries were spontaneous, and were strongly associated with episodes of threatened preterm labour. Medically indicated preterm deliveries were associated with hypertension and fetal growth restriction. Preterm premature rupture of the membranes, vaginal bleeding, anaemia and oligohydramnios were significantly increased in both spontaneous and indicated preterm deliveries compared to term controls. Conclusions More than 50% of preterm births are potentially preventable, but remain associated with risk factors such as increased uterine contractility, preterm premature rupture of the membranes and uterine bleeding whose aetiology is unknown. Despite remarkable advances in perinatal care, preterm birth continues to cause neonatal deaths and long-term morbidity. Significant breakthroughs in the management of preterm birth are likely to come from research into the mechanisms of human parturition and the pathophysiology of preterm labour using multidisciplinary clinical and laboratory approaches.
