Background: Cardiovascular disease (CVD) risk factors are highly prevalent among Hispanic/Latino adults, while the prevalence of MRI infarcts is not well-documented. We, therefore, sought to examine the relationships between CVD risk factors and infarcts with brain structure among Hispanic/Latino individuals. Methods: Participants included 1,886 Hispanic/Latino adults (50-85 years) who underwent magnetic resonance imaging (MRI) as part of the Study of Latinos--Investigation of Neurocognitive Aging-MRI (SOL-INCA-MRI) study. CVD risk was measured approximately 10.5 years before MRI using the Framingham cardiovascular risk score, a measure of 10-year CVD risk (low (<10%), medium (10 - < 20%), and high (≥20%)). MR infarcts were determined as present or absent. Outcomes included total brain, cerebral and lobar cortical gray matter, hippocampal, lateral ventricle, and total white matter hyperintensity (WMH) volumes. Linear regression models tested associations between CVD risk and infarct with MRI outcomes and for modifications by age and sex. Results: Sixty percent of participants were at medium or high CVD risk. Medium and high CVD risk were associated with lower total brain and frontal gray matter and higher WMH volumes compared to those with low CVD risk. High CVD risk was additionally associated with lower total cortical gray matter and parietal volumes and larger lateral ventricle volumes. Men tended to have greater CVDRF-related differences in total brain volumes than women. The association of CVD risk factors on total brain volumes increased with age, equal to an approximate 7-year increase in total brain aging among the high-CVD-risk group compared to the low-risk group. The presence of infarct(s) was associated with lower total brain volumes, which was equal to an approximate 5-year increase in brain aging compared to individuals without infarcts. Infarcts were also associated with smaller total cortical gray matter, frontal and parietal volumes, and larger lateral ventricle and WMH volumes. The paper analyzes stockouts resulting from hurricanes to study whether consumers are persistent in their brand choices. We analyze structural state dependence in brand choice using variation from brand switching during stockouts caused by hurricanes. We derive a simple test for structural state dependence based on the time series of choice persistence for households affected by the stockouts. Using data from the bottled water category, we show that demand increases substantially before hurricanes, causing households to purchase different brands. We find that purchase behavior reverts back to its pre-hurricane trajectory immediately after a hurricane, and we are not able to reject the null hypothesis of no structural state dependence. By contrast, the common approach of estimating structural state dependence based on temporal price variation via a discrete choice model yields a positive effect using data for the same category. We argue that our approach is better suited to identify the causal impact of past choices because it requires fewer assumptions and is based on more plausibly exogenous variation in brand switching resulting from stockouts. We discuss the assumptions underlying the model, the identifica-tion of search cost and preference parameters, the necessary normalizations of utility parameters, counterfactuals that require a search model framework, and different estimation approaches. The goal of this paper is to consolidate knowledge and provide a unified treatment of various aspects of sequential search models that are relevant for empirical work.

The authors analyze the impact of a tax on sweetened beverages using a unique data set of prices, quantities sold, and nutritional information across several thousand taxed and untaxed beverages for a large set of stores in Philadelphia and its surrounding area. The tax is passed through at an average rate of 97%, leading to a 34% price increase. Demand in the taxed area decreases by 46% in response to the tax. Cross-shopping to stores outside of Philadelphia offsets more than half of the reduction in sales in the city and decreases the net reduction in sales of taxed beverages to only 22%. There is no significant substitution to bottled water and modest substitution to untaxed natural juices. The authors show that tax avoidance through cross-shopping severely constrains revenue generation and nutritional improvement, thus making geographic coverage an important policy decision. 24. Age‐related Modularity Changes of the Structural Brain Network: The Framingham Study.

Background: While studies have consistently shown that brain network efficiency decreases with age, data on age‐related reconfiguration of network modules are scarce. We assessed structural brain networks from healthy adults of the Framingham Study to explore age‐related modularity differences. Method: Structural brain networks of 2,903 healthy adults (mean age 55, SD 0.96, range 25‐95, 53% female) were obtained from DTI probabilistic tractography. Connectivity matrices were constructed from 72 cortical and subcortical regions. First, we calculated efficiency to check the validity of our data. Second, we constructed a multilayer network with each layer representing one age decile and calculated modularity for each layer using a robust implementation of the Louvain community‐detection algorithm (Blondel et al., 2008). Third, we compared modular structures between each layer and a reference group (youngest) using the variation of information (VI), an index measure of distance between two modular patterns (e.g. modularity at different ages). Finally, we calculated the participation coefficient, a measure of modular node integration, and the node flexibility, a measure of module allegiance change and assessed their relationships with age. Result: Efficiency decreased with age (b = ‐0.34, SE 0.02, p<0.001), being stable until approximately 50 and declining linearly thereafter (see Figure 1, panel a). Modularity showed a u‐shaped trajectory with most integration at the fifth decile of age (see Figure1, panel b). VI index revealed that modular reconfiguration was greatest between the youngest and the oldest age decile (0.43). Participation coefficient was highest at the fifth decile (see Figure2, panel c) and in frontal nodes (see Figure2, panel a), where it also exhibited high variability. Frontal variability followed a complex pattern (Figure2, panel b). Conversely, central and temporal‐occipital nodes showed less participation and variability (see Figure2, panels a and b), and the lowest flexibility (see Figure3). Conclusion: Brain network modules reconfigure with aging. This reconfiguration is non‐uniform. While midline‐near modules, including the cingulate and parieto‐occipital cortices, show relative stability, frontal modules seemed to reconfigure considerably, as seen by increased average flexibility (change in modular allegiance) and less integration with advancing age, as seen by declining participation coefficients. The paper compares the profitability of personalized pricing policies that are generated from different models of demand and using different data inputs. We study the profitability of personalized pricing policies in a setting with consumer-level panel data. To compare pricing policies, we propose an inverse probability-weighted estimator of profits, discuss how to handle nonrandom price variation, and show how to apply it in a typical consumer-packaged good market with supermarket scanner data. We generate pricing policies from Bayesian hierarchical choice models, regularized regressions, neural networks, and nonparametric classifiers using different sets of data inputs. We find that the performance of machine learning methods is highly varied, ranging from a 30.7% loss to a 14.9% gain relative to a blanket couponing strategy, whereas hierarchical models generate profit gains in the range of 13−16.7%. Across all models, information on consumers' purchase histories leads to large improvements in profits, whereas demographic information has only a small impact. We find that out-of-sample fit statistics are uncorrelated with profit estimates and provide poor guidance toward model selection. Rupture of an intracranial aneurysm leads to subarachnoid hemorrhage, a severe type of stroke. To discover new risk loci and the genetic architecture of intracranial aneurysms, we performed a cross-ancestry, genome-wide association study in 10,754 cases and 306,882 controls of European and East Asian ancestry. We discovered 17 risk loci, 11 of which are new. We reveal a polygenic architecture and explain over half of the disease heritability. We show a high genetic correlation between ruptured and unruptured intracranial aneurysms. We also find a suggestive role for endothelial cells by using gene mapping and heritability enrichment. Drug-target enrichment shows pleiotropy between intracranial aneurysms and antiepileptic and sex hormone drugs, providing insights into intracranial aneurysm pathophysiology. Finally, genetic risks for smoking and high blood pressure, the two main clinical risk factors, play important roles in intracranial aneurysm risk, and drive most of the genetic correlation between intracranial aneurysms and other cerebrovascular traits.

Ndr kinases, such as Neurospora crassa COT1, are important for cell differentiation and polar morphogenesis, yet their input signals as well as their integration into a cellular signaling context are still elusive. Here, we identify the cot-1 suppressor gul-4 as mak-2 and show that mutants of the gul-4/mak-2 mitogen-activated protein (MAP) kinase pathway suppress cot-1 phenotypes along with a concomitant reduction in protein kinase A (PKA) activity. Furthermore, mak-2 pathway defects are partially overcome in a cot-1 background and are associated with increased MAK1 MAPK signaling. A comparative characterization of N. crassa MAPKs revealed that they act as three distinct modules during vegetative growth and asexual development. In addition, common functions of MAK1 and MAK2 signaling during maintenance of cell-wall integrity distinguished the two ERK-type pathways from the p38-type OS2 osmosensing pathway. In contrast to separate functions during vegetative growth, the concerted activity of the three MAPK pathways is essential for cell fusion and for the subsequent formation of multicellular structures that are required for sexual development. Taken together, our data indicate a functional link between COT1 and MAPK signaling in regulating filamentous growth, hyphal fusion, and sexual development.

Enhancing NeuroImaging Genetics through Meta-Analysis Consortium (ENIGMA)—Genetics working group., The cerebral cortex underlies our complex cognitive capabilities, yet little is known about the specific genetic loci that influence human cortical structure. To identify genetic variants that affect cortical structure, we conducted a genome-wide association meta-analysis of brain magnetic resonance imaging data from 51,665 individuals. We analyzed the surface area and average thickness of the whole cortex and 34 regions with known functional specializations. We identified 199 significant loci and found significant enrichment for loci influencing total surface area within regulatory elements that are active during prenatal cortical development, supporting the radial unit hypothesis. Loci that affect regional surface area cluster near genes in Wnt signaling pathways, which influence progenitor expansion and areal identity. Variation in cortical structure is genetically correlated with cognitive function, Parkinson's disease, insomnia, depression, neuroticism, and attention deficit hyperactivity disorder.
