Abstract: We present the results of time-integrated searches for astrophysical neutrino sources in both the northern and southern skies. Data were collected using the partially-completed IceCube detector in the 40-string configuration between 2008 April 5 and 2009 May 20, totaling 375.5 days livetime. An unbinned maximum likelihood ratio method is used to search for astrophysical signals. The data sample contains 36,900 events: 14,121 from the northern sky, mostly muons induced by atmospheric neutrinos and 22,779 from the southern sky, mostly high energy atmospheric muons. The analysis includes searches for individual point sources and targeted searches for specific stacked source classes and spatially extended sources. While this analysis is sensitive to TeV-PeV energy neutrinos in the northern sky, it is primarily sensitive to neutrinos with energy greater than about 1 PeV in the southern sky. No evidence for a signal is found in any of the searches. Limits are set for neutrino fluxes from astrophysical sources over the entire sky and compared to predictions. The sensitivity is at least a factor of two better than previous searches (depending on declination), with 90% confidence level muon neutrino flux upper limits being between E^2 dN/dE ~ 2 - 200 \times 10^-12 TeV cm^-2 s^-1 in the northern sky and between 3 -700 \times 10^-12 TeV cm^-2 s^-1 in the southern sky. The stacked source searches provide the best limits to specific source classes. The full IceCube detector is expected to improve the sensitivity to E^-2 sources by another factor of two in the first year of operation., 44 pages, 21 figures. Submitted to the Astrophysical Journal

13. Specific bioactive collagen peptides (PETAGILE®) as supplement for horses with osteoarthritis: A two‐centred study.

Abstract: Summary: The aim of the study was to evaluate the clinical efficacy of specific bioactive collagen peptides (BCP), here administered orally as PETAGILE®, on horses with mild to moderate, naturally occurring osteoarthritis. Data from a two‐centred pilot study were used for the meta‐analysis. Thirty‐eight privately owned horses of various breeds were available. In one centre, 18 of these patients (6 ± 3 years; 519 ± 100 kg BW) received either 25 g (n = 6) or 50 g (n = 12) BCP/day orally for 12 weeks. In the second centre, 20 horses (18 ± 4 years; 413 ± 94 kg BW) received either a placebo (control; n = 10) or 25 g BCP/day. The attending veterinarians performed an orthopaedic examination including flexion tests and evaluated the degree of lameness, rotation pain, step length and arc of foot flight during trot (8 parameters) at the beginning and after 6 and 12 weeks. The horse owners answered a weekly questionnaire about their perception of lameness, mobility and the horses’ willingness to run. In the 50 g BCP group, in six of eight parameters, a strong effect (Cohen's r > .5) was detected with two parameters (lameness and flexion pain) significantly improved already after 6 weeks. In the 25 g BCP group, a moderate effect (Cohen's r = .3–0.5) was seen in six parameters, with three parameters improved already after 6 weeks. The owners reported a strong effect for mobility and willingness to run (Cohen's r = .69 and .62, respectively) and a moderate effect (Cohen's r = .49 and 0.41) for the development of lameness in the 50 g and 25 g BCP group in comparison with the placebo treatment. This study revealed promising effects of the safe oral‐specific BCP supplementation on symptoms of osteoarthritis in horses already after 3 months. The higher dosage of 50 g BCP/day had superior impact. Further long‐term investigations on specific BCP efficacy in horses with osteoarthritis, preferably in blinded and placebo‐controlled studies, should be performed to confirm these first positive results. Abstract: Context. Transient neutrino sources such as Gamma-Ray Bursts (GRBs) and Supernovae (SNe) are hypothesized to emit bursts of high-energy neutrinos on a time-scale of \lesssim 100 s. While GRB neutrinos would be produced in high relativistic jets, core-collapse SNe might host soft-relativistic jets, which become stalled in the outer layers of the progenitor star leading to an efficient production of high-energy neutrinos. Aims. To increase the sensitivity to these neutrinos and identify their sources, a low-threshold optical follow-up program for neutrino multiplets detected with the IceCube observatory has been implemented. Methods. If a neutrino multiplet, i.e. two or more neutrinos from the same direction within 100 s, is found by IceCube a trigger is sent to the Robotic Optical Transient Search Experiment, ROTSE. The 4 ROTSE telescopes immediately start an observation program of the corresponding region of the sky in order to detect an optical counterpart to the neutrino events. Results. No statistically significant excess in the rate of neutrino multiplets has been observed and furthermore no coincidence with an optical counterpart was found. Conclusion. The search allows, for the first time, to set stringent limits on current models predicting a high-energy neutrino flux from soft relativistic hadronic jets in core-collapse SNe. Abstract: This paper presents searches for flaring sources of neutrinos using the IceCube neutrino telescope. For the first time, a search is performed over the entire parameter space of energy, direction and time looking for neutrino flares of 20 microseconds to a year duration from astrophysical sources among the atmospheric neutrino and muon backgrounds. Searches which integrate over time are less sensitive to flares because they are affected by a larger background of atmospheric neutrinos and muons that can be reduced by the time constraint. Flaring sources considered here, such as active galactic nuclei, soft gamma-ray repeaters and gamma-ray bursts, are promising candidate neutrino emitters. We used mainly data taken between April 5, 2008 and May 20, 2009 by a partially completed configuration of IceCube with 40 strings. For the presented searches an unbinned maximum likelihood method is used with a time-dependent prior to test several different source hypotheses. An "untriggered" search covers any possible time-dependent emission from sources not correlated to any other observation using other astrophysical messengers such as photons. Moreover, a similar time scan is performed for a predefined catalogue of sources that exhibit intense photon flares. Searches triggered by multi-wavelength information on flares from blazars and soft gamma-ray repeaters are performed using the 40 string data and also the data taken by the previous configuration of 22 strings in operation between May 31, 2007 and April 5, 2008. Flares for which extensive and continuous monitoring is available from Fermi-LAT and SWIFT and flares detected by imaging Cherenkov telescopes with shorter time-scale monitoring are considered. Abstract: Self-annihilating or decaying dark matter in the Galactic halo might produce high energy neutrinos detectable with neutrino telescopes. We have conducted a search for such a signal using 276 days of data from the IceCube 22-string configuration detector acquired during 2007 and 2008. The effect of halo model choice in the extracted limit is reduced by performing a search that considers the outer halo region and not the Galactic Center. We constrain any large scale neutrino anisotropy and are able to set a limit on the dark matter self-annihilation cross section of \simeq 10^{-22} cm^3/s for WIMP masses above 1 TeV, assuming a monochromatic neutrino line spectrum., Comment: 12pages, 13figures

Abstract: IceCube has become the first neutrino telescope with a sensitivity below the TeV neutrino flux predicted from gamma-ray bursts if gamma-ray bursts are responsible for the observed cosmic-ray flux above 10^18 eV. Two separate analyses using the half-complete IceCube detector, one a dedicated search for neutrinos from pγ interactions in the prompt phase of the gamma-ray burst fireball and the other a generic search for any neutrino emission from these sources over a wide range of energies and emission times, produced no evidence for neutrino emission, excluding prevailing models at 90% confidence., Physical Review Letters

Abstract: We report the first observation of an anisotropy in the arrival direction of cosmic rays with energies in the multi-TeV region in theSouthern sky using data from the IceCube detector. Between 2007 June and 2008 March, the partially deployed IceCube detector was operated in a configuration with 1320 digital optical sensors distributed over22 strings at depths between 1450 and 2450 m inside the Antarctic ice. IceCube is a neutrino detector, but the data are dominated by a large background of cosmic-ray muons. Therefore, the background data aresuitable for high-statistics studies of cosmic rays in the southern sky. The data include 4.3 billion muons produced by downward-going cosmic-ray interactions in the atmosphere; these events were reconstructed with a median angular resolution of 3° and a median energy of ∼20 TeV. Their arrival direction distribution exhibits an anisotropy in right ascension with a first-harmonic amplitude of (6.4±0.2 stat.±0.8 syst.) × 10-4. © 2010 The American Astronomical Society.

Abstract: We present new results of searches for neutrino point sources in the northern sky, using data recorded in 2007-08 with 22 strings of the IceCube detector (approximately one-fourth of the planned total) and 275.7 days of livetime. The final sample of 5114 neutrino candidate events agrees well with the expected background of atmospheric muon neutrinos and a small component of atmospheric muons. No evidence of a point source is found, with the most significant excess of events in the sky at 2.2 sigma after accounting for all trials. The average upper limit over the northern sky for point sources of muon-neutrinos with E^-2 spectrum is E^2 Phi_nu_mu < 1.4x10^-11 TeV cm^-2 s^-1, in the energy range from 3 TeV to 3 PeV, improving the previous best average upper limit by the AMANDA-II detector by a factor of two., 5 pages, 4 figures. Abstract: Dietary consumption of food supplements has been found to modulate skin functions and can therefore be useful in the treatment of skin aging. However, there is only a limited number of clinical studies supporting these claims. In this double-blind, placebo-controlled study, the effectiveness of the specific bioactive collagen peptide (BCP) VERISOL® on eye wrinkle formation and stimulation of procollagen I, elastin and fibrillin biosynthesis in the skin was assessed. A hundred and fourteen women aged 45-65 years were randomized to receive 2.5 g of BCP or placebo, once daily for 8 weeks, with 57 subjects being allocated to each treatment group. Skin wrinkles were objectively measured in all subjects, before starting the treatment, after 4 and 8 weeks as well as 4 weeks after the last intake (4-week regression phase). A subgroup was established for suction blister biopsies analyzing procollagen I, elastin and fibrillin at the beginning of the treatment and after 8 weeks of intake. The ingestion of the specific BCP used in this study promoted a statistically significant reduction of eye wrinkle volume (p < 0.05) in comparison to the placebo group after 4 and 8 weeks (20%) of intake. Moreover a positive long-lasting effect was observed 4 weeks after the last BCP administration (p < 0.05). Additionally, after 8 weeks of intake a statistically significantly higher content of procollagen type I (65%) and elastin (18%) in the BCP-treated volunteers compared to the placebo-treated patients was detected. For fibrillin, a 6% increase could be determined after BCP treatment compared to the placebo, but this effect failed to reach the level of statistical significance. In conclusion, our findings demonstrate that the oral intake of specific bioactive collagen peptides (Verisol®) reduced skin wrinkles and had positive effects on dermal matrix synthesis. © 2014 S. Abstract: Various dietary supplements are claimed to have cutaneous anti-aging properties; however, there are a limited number of research studies supporting these claims. The objective of this research was to study the effectiveness of collagen hydrolysate (CH) composed of specific collagen peptides on skin biophysical parameters related to cutaneous aging. In this double-blind, placebo-controlled trial, 69 women aged 35-55 years were randomized to receive 2.5 g or 5.0 g of CH or placebo once daily for 8 weeks, with 23 subjects being allocated to each treatment group. Skin elasticity, skin moisture, transepidermal water loss and skin roughness were objectively measured before the first oral product application (t0) and after 4 (t1) and 8 weeks (t2) of regular intake. Skin elasticity (primary interest) was also assessed at follow-up 4 weeks after the last intake of CH (t3, 4-week regression phase). At the end of the study, skin elasticity in both CH dosage groups showed a statistically significant improvement in comparison to placebo. After 4 weeks of follow-up treatment, a statistically significantly higher skin elasticity level was determined in elderly women. With regard to skin moisture and skin evaporation, a positive influence of CH treatment could be observed in a subgroup analysis, but data failed to reach a level of statistical significance. Abstract: We investigate the effects of stochastic density fluctuations on the dust temperatures and the resulting infrared (IR) emission spectra of interstellar clouds as an extension of preceding investigations by Hegmann & Kegel. We consider absorption and scattering by dust grains in spherical clouds which are, on average, homogeneous but have a fluctuating density. The spatial variation of the density is described by means of a Markov process. This clump model introduces two parameters: the correlation length and the Gaussian width of the density fluctuations. As the intensity inherits the randomness of the density n, the ordinary radiative transfer equation has to be replaced by a generalized transfer equation of Fokker–Planck type. In the first part, we investigate the influence of our model parameters on the radiative transport in the ultraviolet (UV) and use the results to calculate the dust temperature in radiative equilibrium. Afterwards, the IR emission of the dust is modelled for the same set of clump parameters. We find that the presence of clumps decreases the effective extinction and therefore leads to substantial differences in UV illumination and dust temperatures, compared with the homogeneous case. Because of the distribution of dust temperatures, the presence of clumps also affects the IR emission and thus possible observations. In the second part, we use a fit with two blackbody spectra to determine the cloud dust mass from our synthetic IR fluxes. It is shown that in a clumpy environment the overall dust mass is generally underestimated. This effect correlates with the degree of cloud fragmentation. [ABSTRACT FROM AUTHOR]

Abstract: This research examines young unmarried women's ways of talking about AIDS, AIDS prevention, and its relationship to mass media AIDS messages in Thailand. Data were derived from a survey conducted in four districts of Kanchanaburi province. Three hundred ninety-seven unmarried women were extracted from the approximately 1,800 original subjects for this analysis. Respondents were asked about the subjects and extent of their conversations about AIDS, the choice of discussion partners, considerations of social appropriateness in talking about the disease, as well as their risk perception. Important findings were that (a) women tend to talk about AIDS primarily with friends and siblings, (b) their reception level of mass media messages is related to the number of topics discussed and frequency of talks by the subjects, and (c) socioeconomic status and age are related to the variety and frequency of talking about AIDS. Abstract: This paper describes the response of the IceCube neutrino telescope located at the geographic south pole to outbursts of MeV neutrinos from the core collapse of nearby massive stars. IceCube was completed in December 2010 forming a lattice of 5160 photomultiplier tubes that monitor a volume of ∼1 km3 in the deep Antarctic ice for particle induced photons. The telescope was designed to detect neutrinos with energies greater than 100 GeV. Owing to subfreezing ice temperatures, the photomultiplier dark noise rates are particularly low. Hence IceCube can also detect large numbers of MeV neutrinos by observing a collective rise in all photomultiplier rates on top of the dark noise. With 2 ms timing resolution, IceCube can detect subtle features in the temporal development of the supernova neutrino burst. For a supernova at the galactic center, its sensitivity matches that of a background-free megaton-scale supernova search experiment. The sensitivity decreases to 20 standard deviations at the galactic edge (30 kpc) and 6 standard deviations at the Large Magellanic Cloud (50 kpc). IceCube is sending triggers from potential supernovae to the Supernova Early Warning System. The sensitivity to neutrino properties such as the neutrino hierarchy is discussed, as well as the possibility to detect the neutronization burst, a short outbreak of νe ’s released by electron capture on protons soon after collapse. Abstract: SN 2008D, a core collapse supernova at a distance of 27 Mpc, was serendipitously discovered by the Swift satellite through an associated X-ray flash. Core collapse supernovae have been observed in association with long gamma-ray bursts and X-ray flashes and a physical connection is widely assumed. This connection could imply that some core collapse supernovae possess mildly relativistic jets in which high-energy neutrinos are produced through proton-proton collisions. The predicted neutrino spectra would be detectable by Cherenkov neutrino detectors like IceCube. A search for a neutrino signal in temporal and spatial correlation with the observed X-ray flash of SN 2008D was conducted using data taken in 2007-2008 with 22 strings of the IceCube detector. Events were selected based on a boosted decision tree classifier trained with simulated signal and experimental background data. The classifier was optimized to the position and a "soft jet" neutrino spectrum assumed for SN 2008D. Using three search windows placed around the X-ray peak, emission time scales from 100 - 10000 s were probed. No events passing the cuts were observed in agreement with the signal expectation of 0.13 events. Current treatments are limited and often attempt to treat OA symptoms rather than improve joint structure and function. Collagen hydrolysates as oral supplements are a promising therapeutic option to achieve this advanced therapeutic aim in both species. The effects of oral supplementation were therefore investigated in canine OA patients., Method: In a systematic, placebo-controlled, double-blind interventional study in 31 dogs with naturally occurring OA, the efficacy of oral supplementation of specific bioactive collagen peptides (BCP) was tested in comparison to the approved combination of the active substances omega-3 fatty acids and vitamin E. The dogs were examined on a horizontal treadmill with 4 integrated piezoelectric force plates at the beginning and end of a twelve-week test period. At both points, the owners completed a specific questionnaire containing the validated Canine Brief Pain Inventory (CBPI) and the dogs were fitted with accelerometers to record total daily activity data., Results: Only the oral supplementation of BCP resulted in a significant improvement of several kinetic parameters measured using a force-plate fitted treadmill, and the quality of life assessed by CBPI, while accelerometry was unaffected by the intervention., Conclusion: The results of this three-month BCP supplementation study using objective measurement parameters in dogs with naturally occurring OA demonstrate an efficacy, suggesting the therapeutic use of BCP in canine OA patients and demonstrating the relevance of this collagen hydrolysate formulation for the treatment of OA in human patients as well., Competing Interests: Jutta Hugenberg is employed by GELITA AG, Eberbach, Germany, which funded the study. Abstract: The intake of specific collagen peptides (SCPs) has been shown to decrease activity-related knee pain in young, physically active adults. This trial investigated the effect of a 12-week SCP supplementation in a wider age range of healthy men and women over 18 years with functional knee and hip pain during daily activities. A total of 182 participants were randomly assigned to receive either 5 g of specific collagen peptides (CP-G) or a placebo (P-G). Pain at rest and during various daily activities were assessed at baseline and after 12 weeks by a physician and participants using a 10-point numeric rating scale (NRS). The intake of 5 g SCP over 12 weeks significantly reduced pain at rest ( p = 0.018) and during walking ( p = 0.032) according to the physician's evaluation. Participants in the CP-G also reported significantly less pain when climbing stairs ( p = 0.040) and when kneeling down ( p < 0.001) compared to the P-G. Additionally, after 12 weeks, restrictions when squatting were significantly lower in the CP-G compared with the P-G ( p = 0.014). The daily intake of 5 g of SCP seems to benefit healthy adults with hip and knee joint discomforts by reducing pain during daily activities.

Abstract: The aim of the study was to evaluate the clinical efficacy of specific bioactive collagen peptides (BCP), here administered orally as PETAGILE ® , on horses with mild to moderate, naturally occurring osteoarthritis. Data from a two-centred pilot study were used for the meta-analysis. Thirty-eight privately owned horses of various breeds were available. In one centre, 18 of these patients (6 ± 3 years; 519 ± 100 kg BW) received either 25 g (n = 6) or 50 g (n = 12) BCP/day orally for 12 weeks. In the second centre, 20 horses (18 ± 4 years; 413 ± 94 kg BW) received either a placebo (control; n = 10) or 25 g BCP/day. The attending veterinarians performed an orthopaedic examination including flexion tests and evaluated the degree of lameness, rotation pain, step length and arc of foot flight during trot (8 parameters) at the beginning and after 6 and 12 weeks. The horse owners answered a weekly questionnaire about their perception of lameness, mobility and the horses' willingness to run. In the 50 g BCP group, in six of eight parameters, a strong effect (Cohen's r > .5) was detected with two parameters (lameness and flexion pain) significantly improved already after 6 weeks. In the 25 g BCP group, a moderate effect (Cohen's r = .3-0.5) was seen in six parameters, with three parameters improved already after 6 weeks. The owners reported a strong effect for mobility and willingness to run (Cohen's r = .69 and .62, respectively) and a moderate effect (Cohen's r = .49 and 0.41) for the development of lameness in the 50 g and 25 g BCP group in comparison with the placebo treatment. This study revealed promising effects of the safe oral-specific BCP supplementation on symptoms of osteoarthritis in horses already after 3 months. The higher dosage of 50 g BCP/day had superior impact. Further long-term investigations on specific BCP efficacy in horses with osteoarthritis, preferably in blinded and placebo-controlled studies, should be performed to confirm these first positive results., (© 2017 Blackwell Verlag GmbH.)

Abstract: Background: Brittle nail syndrome is a common problem among women and refers to nails that exhibit surface roughness, raggedness, and peeling., Aim: The goal of this study was to investigate whether daily oral supplementation with collagen peptides alleviates the symptoms of brittle nails and improves nail growth rate., Methods: In this open-label, single-center trial, 25 participants took 2.5 g of specific bioactive collagen peptides (BCP, VERISOL ® ) once daily for 24 weeks followed by a 4-week off-therapy period. Nail growth rate and the frequency of cracked and/or chipped nails as well as an evaluation of symptoms and global clinical improvement score of brittle nails were assessed by a physician during treatment and 4 weeks after discontinuation., Results: Bioactive collagen peptides treatment promoted an increase of 12% nail growth rate and a decrease of 42% in the frequency of broken nails. Additionally, 64% of participants achieved a global clinical improvement in brittle nails, and 88% of participants experienced an improvement 4 weeks post-treatment. Abstract: In this double-blind, placebo-controlled clinical study, we investigated the efficacy of specific bioactive collagen peptides (BCP) on the cellulite treatment of normal and overweight women. In total, 105 women aged 24-50 years with moderate cellulite were randomized to orally receive a daily dosage of 2.5 g BCP or a placebo over 6 months. The degree of cellulite was evaluated before starting the treatment and after 3 and 6 months of intake. In addition, skin waviness, dermal density, and the length of subcutaneous borderline were assessed. BCP treatment led to a statistically significant decrease in the degree of cellulite and a reduced skin waviness on thighs (P < 0.05) in normal weight women. Moreover, dermal density was significantly improved (P < 0.05) compared to placebo. The subcutaneous borderline showed a significant shortening after BCP intake compared to the beginning of the study, indicating cellulite improvement, but the data failed to reach statistical significance compared to placebo. The efficacy of BCP treatment was also confirmed in overweight women, although the impact was less pronounced in comparison with women of normal body weight. The results of the study demonstrated that a regular ingestion of BCP over a period of 6 months led to a clear improvement of the skin appearance in women suffering from moderate cellulite. Based on the current data, it can be concluded that a long-term therapy with orally administered BCP leads to an improvement of cellulite and has a positive impact on skin health.

Abstract: Various dietary supplements are claimed to have cutaneous anti-aging properties; however, there are a limited number of research studies supporting these claims. The objective of this research was to study the effectiveness of collagen hydrolysate (CH) composed of specific collagen peptides on skin biophysical parameters related to cutaneous aging. In this double-blind, placebo-controlled trial, 69 women aged 35-55 years were randomized to receive 2.5 g or 5.0 g of CH or placebo once daily for 8 weeks, with 23 subjects being allocated to each treatment group. Skin elasticity, skin moisture, transepidermal water loss and skin roughness were objectively measured before the first oral product application (t0) and after 4 (t1) and 8 weeks (t2) of regular intake. Skin elasticity (primary interest) was also assessed at follow-up 4 weeks after the last intake of CH (t3, 4-week regression phase). At the end of the study, skin elasticity in both CH dosage groups showed a statistically significant improvement in comparison to placebo. After 4 weeks of follow-up treatment, a statistically significantly higher skin elasticity level was determined in elderly women. With regard to skin moisture and skin evaporation, a positive influence of CH treatment could be observed in a subgroup analysis, but data failed to reach a level of statistical significance. Abstract: c-Jun N-terminal kinases (JNKs, also called stress activated protein kinases) and the extra-cellular signal responsive kinases (ERKs) exert different functions in mitogenesis, maturation and differentiation of immune and epithelial cells. We investigated specific functions of individual JNK and ERK isoforms in skin permeability barrier repair and in wound healing. JNK1, but not JNK2 or JNK3, deficient mice revealed a delay in the permeability barrier repair after superficial injury to the skin (tape-stripping) as well as a delay in the healing of full skin thickness wounds. Skin barrier injury induced an increase in epidermal JNK1 enzyme activity in mouse skin in vivo, and JNK1 activity correlated with the degree of differentiation in organotypic keratinocyte cultures. Skin injury activated epidermal ERK2 enzyme activity with biphasic maxima after 30 min and 3h, and the activity was independent from the differentiation state in keratinocyte culture. Abstract: IceCube has become the first neutrino telescope with a sensitivity below the TeV neutrino flux predicted from gamma-ray bursts if gamma-ray bursts are responsible for the observed cosmic-ray flux above 10(18) eV. Two separate analyses using the half-complete IceCube detector, one a dedicated search for neutrinos from pγ interactions in the prompt phase of the gamma-ray burst fireball and the other a generic search for any neutrino emission from these sources over a wide range of energies and emission times, produced no evidence for neutrino emission, excluding prevailing models at 90% confidence.

Abstract: Protection of the skin against microbiological infection is provided by the permeability barrier and by antimicrobial proteins. We asked whether the expression of murine β-defensins (mBDs)-1, -3, and -14-orthologs of human β-defensins hBD-1, -2, and -3, respectively--is stimulated by mechanically/physicochemically (tape stripping or acetone treatment) or metabolically (essential fatty acid-deficient (EFAD) diet) induced skin barrier dysfunction. Both methods led to a moderate induction of mBD-1 and mBD-14 and a pronounced induction of mBD-3 mRNA. Protein expression of the mBDs was increased as shown by immunohistology and by western blotting. Artificial barrier repair by occlusion significantly reduced the increased expression of mBD-14 after mechanical injury and of all three mBDs in EFAD mice, supporting an interrelationship between permeability and the antimicrobial barrier. mBD-3 expression was stimulated in vitro by tumor necrosis factor-α (TNF-α), and a neutralizing anti-TNF-α antibody significantly reduced increased mBD-3 expression after barrier injury in mouse skin, indicating that induction of mBD-3 expression is mediated by cytokines. The expression of mBD-14 was stimulated by transforming growth factor-α and not by TNF-α. In summary, we demonstrated upregulation of mBD1, -3, and -14 after mechanically and metabolically induced skin barrier disruption, which may be an attempt to increase defense in the case of permeability barrier dysfunction.

Abstract: Point source searches with the IceCube neutrino telescope have been restricted to one hemisphere, due to the exclusive selection of upward going events as a way of rejecting the atmospheric muon background. We show that the region above the horizon can be included by suppressing the background through energy-sensitive cuts. This improves the sensitivity above PeV energies, previously not accessible for declinations of more than a few degrees below the horizon due to the absorption of neutrinos in Earth. We present results based on data collected with 22 strings of IceCube, extending its field of view and energy reach for point source searches. No significant excess above the atmospheric background is observed in a sky scan and in tests of source candidates. Upper limits are reported, which for the first time cover point sources in the southern sky up to EeV energies.

Abstract: A search for muon neutrinos from neutralino annihilations in the Sun has been performed with the IceCube 22-string neutrino detector using data collected in 104.3 days of live time in 2007. No excess over the expected atmospheric background has been observed. Upper limits have been obtained on the annihilation rate of captured neutralinos in the Sun and converted to limits on the weakly interacting massive particle (WIMP) proton cross sections for WIMP masses in the range 250-5000 GeV. These results are the most stringent limits to date on neutralino annihilation in the Sun.

Abstract: The repair of the permeability barrier to prevent the entry of harmful substances into the body is a goal in wound healing. Semi-occlusive foils, which provide an artificial barrier, are commonly used for the treatment of wounds. We examined the effects of foils on wound contraction, cell migration, and reepithelization. Full-thickness skin wounds in mice were covered with occlusive latex foils or semi-occlusive water vapor-permeable hydrocolloid foils for either the entire, the first half, or the second half of the wound-healing period. We found that application of foils for the entire healing period initially reduced wound healing during the first week of treatment, whereas healing was enhanced during the second week. Foils were found to reduce wound contraction, but enhanced reepithelization during the second week of wound healing because of increased proliferation and migration of keratinocytes. These effects were also noted when the hydrocolloid foils were applied for the second part of the healing period, only. The fully occlusive latex foil led to irritation of the skin, whereas less irritation occurred under semi-occlusive conditions. In summary, we found that artificial barrier repair with semi-occlusive foils in wounds reduced wound contraction and enhanced cell migration and reepithelization without irritation.

Abstract: In aged skin, decreased levels of stratum corneum ceramides have been described. Epidermal ceramides are generated by sphingomyelin hydrolysis or synthesis from sphingosin and fatty acids and are degraded by ceramidase. We recently showed that epidermal acid sphingomyelinase (A-SMase) generates ceramides with structural function in the stratum corneum lipid bilayers, which provide for the permeability barrier function of the skin. Here, we examined the activities of epidermal A-SMase, ceramide synthase, and ceramidase in chronologically aged versus young hairless mouse skin. We found reduced A-SMase and ceramide synthase activities in the epidermis of aged mice. However, studies on enzyme localization revealed unchanged, ongoing high A-SMase activity in the outer epidermis, which correlated with reported normal barrier function found in aged skin under basal conditions. Reduced A-SMase and ceramide synthase activity was noted in the inner epidermis, correlating with reduced capacity for permeability barrier repair in aging. Ceramidase activity was not age dependent. In summary, we found reduced activities of ceramide-generating SMase and ceramide synthase in the inner epidermis of aged skin, explaining its reduced capacity in barrier repair. In contrast, A-SMase activity in the outer epidermis was unchanged, indicating that this enzyme is crucially involved in basal permeability barrier homeostasis.
