Rotaviruses are non-enveloped viruses that each consist of 11 double-stranded RNA molecules. These viruses are able to persist in the environment, and therefore play a fundamental role in the epidemiology of gastroenteritis and severe diarrhoea in children worldwide. While mussels have been primarily used as indicators of chemical pollution, they can also be used to monitor viral contamination. The purpose of this study was to demonstrate that the Mytilus galloprovincialis mussel can also be used to detect microbial contamination, owing to its tendency to naturally concentrate viruses and other pathogens.

Living in extreme environments, marine organisms face constant exposure to a range of stressors, such as high radiation levels, fluctuations in temperature, and oxidative stress. Understanding extremophile fishes is crucial because it gives us valuable insights into the biochemical, physiological, and developmental processes that govern life, by observing how they operate under natural stressors. Among the most fascinating adaptations is the existence of specialised enzymes and compounds that function as potent antioxidants, successfully counteracting reactive oxygen species' deleterious effects. In this review, we analysed the findings from several studies on Antarctic and deep-sea fish species, while highlighting the environmental stressors effects toward the antioxidant system. The antioxidant defences of the considered extremophile fishes have been extensively studied, but there is still much to learn to fully understand this complex system, while the relative research is still ongoing. Adamussium colbecki is a scallop endemic of the Antarctic Ocean, the only modern survivor of the Adamussiini tribe and one of the few bivalves living in polar environments. Compared with other Antarctic animals, very little is known concerning the evolutionary adaptations which allow this species to thrive at sub-zero temperatures. Due to its local abundance and sensitivity to environmental changes, A. colbecki is an interesting model for studying the effects of pollution and climate change in the Antarctic Ocean. Here, we report, for the first time, the application of transcriptomic tools to the study of the effects of a short-to-medium term exposure to a +1.5 °C water temperature increase on three tissues. Although this approach did not highlight any significant change in response to thermal stress, we observed slight alterations in energetic metabolism and nutrient adsorption in the digestive gland, most likely linked with stabling in experimental tanks. The results of our study suggest that A. colbecki may be particularly vulnerable to the effects of climate change due to its complete inability to adapt to temperature increase at the transcriptomic level. Antarctica is the continent with the lowest local human impact; however, it is susceptible to pollution from external sources. Emerging pollutants such as perfluoroalkyl substances pose an increasing threat to this environment and therefore require more in-depth investigations to understand their environmental fate and biological impacts. The present study focuses on expression analysis at the transcriptional level of genes coding for four antioxidant enzymes (sod1, sod2, gpx1, and gpx4) in the liver and kidney of an Antarctic fish species, Trematomus newnesi (Boulenger, 1902). mRNA levels were also assessed in fish exposed to 1.5 μg/L of perfluoro-octanoic acid for 10 days. The kidney showed a higher level of expression than the liver in wildlife specimens. In the liver, the treatment induced an increase in gene expression for all the considered enzymes, whereas in the kidney, it induced a general decrease. The obtained results advance the scientific community's understanding of how the potential future presence of anthropogenic contaminants in the Southern Ocean can affect the antioxidant system of Antarctic fishes. The Antarctic endemic fish genus Trematomus (Trematominae, Notothenioidei) includes 15 species very diverse in morphology, lifestyle and feeding ecology. Co-occurring on the continental shelf, they occupy different habitats and a wide range of ecological niches as the result of adaptive radiation during their evolutionary history. Ecomorphological differentiation is a key feature of adaptive radiations, with a general trend for specialization following divergence. Here, we investigated the trophic adaptive morphology and ecology of six Trematomus species from Terra Nova Bay (Ross Sea) through feeding apparatus metrics and geometric morphometrics. The suction index (SI), the mechanical advantage in jaw closing (MA), the relative surface of the adductor mandibulae muscle and nine morphological traits related to feeding structures were analysed. Head shape clearly differentiates the benthic (T. bernacchii, T. hansoni and T. pennellii) from the pelagic (T. eulepidotus and T. borchgrevinki) species. The position of the eyes and the orientation of the mouth also contribute to specific morphological differences and specialization. Interestingly, T. newnesi stands at an intermediate position and the mouth is clearly oriented upwards compared to the other congeneric species. [ABSTRACT FROM AUTHOR]

The Antarctic continent is one of the most pristine environments on planet Earth, yet one of the most fragile and susceptible to the effects of the ongoing climate change. The overwhelming majority of the components of Antarctic marine trophic chain are stenotherm organisms, highly adapted to the extreme, but extremely stable, freezing temperatures of the Antarctic ocean, which have not changed significantly during the past fifteen million years. Notothenioid fishes are the most abundant representatives of ichthyofauna at these latitudes, being ubiquitously found in coastal areas across the entire continent. While different Antarctic fish species have been previously subjected to studies aimed at defining their range of thermal tolerance, or at studying the response to acute thermal stress, just a handful of authors have investigated the effects of the exposure to a moderate increase of temperature, falling within the expected forecasts for the next few decades in some areas of the Antarctica. Here, the emerald rockcod Trematomus bernacchii was used as a model species to investigate the effects of a 20-day long exposure to a +1.5 ° C increase in the brain, gills and skeletal muscle, using a RNA-sequencing approach. In parallel, the experimental design also allowed for assessing the impact of stabling (including acclimation, the handling of fishes and their confinement in tanks during the experimental phase) on gene expression profiling. The results of this study clearly identified the brain as the most susceptible tissue to heat stress, with evidence of a time-dependent response dominated by an alteration of immune response, protein synthesis and folding, and energy metabolism-related genes. While the gills displayed smaller but still significant alterations, the skeletal muscle was completely unaffected by the experimental conditions. The stabling conditions also had an important impact on gene expression profiles in the brain, suggesting the presence of significant alterations of the fish nervous system, possibly due to the confinement to tanks with limited water volume and of the restricted possibility of movement. Besides providing novel insights in the molecular mechanisms underlying thermal stress in notothenioids, these findings suggest that more attention should be dedicated to an improved design of the experiments carried out on Antarctic organism, due to their extreme susceptibility to the slightest environmental alterations. Metal bioaccumulation and metallothionein (MT) expression were investigated in the gills and liver of the red-blooded Antarctic teleost Trematomus hansoni to evaluate the possibility for this species to face, with adequate physiological responses, an increase of copper and cadmium concentrations in its tissues. Specimens of this Antarctic fish were collected from Terra Nova Bay (Ross Sea) and used for a metal exposure experiment in controlled laboratory conditions. The two treatments led to a significant accumulation of both metals and increased gene transcription only for the MT-1. The biosynthesis of MTs was verified especially in specimens exposed to Cd, but most of these proteins were soon oxidized, probably because they were involved in cell protection against oxidative stress risk by scavenging reactive oxygen species. The obtained data highlighted the phenotypic plasticity of T. hansoni, a species that evolved in an environment characterized by naturally high concentrations of Cu and Cd, and maybe the possibility for the Antarctic fish to face the challenges of a world that is becoming more toxic every day. Metallothioneins (MTs) are low-molecular weight sulfur-rich proteins, widely distributed in nature. They play a homeostatic role in the control and detoxification of metal ions. Previous studies indicated that MTs also have the capacity to scavenge reactive oxygen species. This study aimed to investigate the evolution of the protein in the notothenioid fish, evolved under the selective pressure of relatively high oxygen partial pressures, characteristics of cold Antarctic seawaters, and relatively high concentrations of metal ions, Cd and Cu in particular. The cDNA sequences of MT isoforms were characterized in members of the Nototheniidae, Bathydraconidae, Artedidraconidae, and Channichthyidae families. The phylogenetic relationships of MTs from these families and other teleosts were inferred by using Maximum Likelihood and Bayesian methods. The analysis of coding region and untranslated (UTR) sequences indicated the presence of two MT clades, each containing one of the two MT isoforms, MT-1 and MT-2. Our results indicated, for the first time for these proteins, that the evolution of MT genes has been characterized by strong purifying selection, whereas it did not observe any evidence of positive selection. In addition, phylogenetic analysis of the UTRs suggested that functional changes, in particular related to the MT-1 gene expression, had accompanied the duplication event. [ABSTRACT FROM AUTHOR]

This work aimed to evaluate the effects of zinc (Zn) relating to cadmium (Cd)-induced toxicity and the role played by MTF-1. This transcription factor regulates the expression of genes encoding metallothioneins (MTs), some Zn transporters and the heavy chain of γ-glutamylcysteine synthetase. For this reason, two cell lines of mouse fibroblasts were used: a wild-type strain and a knockout strain to study the effects. Cells were exposed to complete medium containing: (1) 50 μM ZnSO4 (Zn), (2) 1 μM CdCl2 (Cd 1), (3) 2 μM CdCl2 (Cd 2), (4) 50 μM ZnSO4 + 1 μM CdCl2 (ZnCd 1) and (5) 50 μM ZnSO4 + 2 μM CdCl2 (ZnCd 2) for 4, 18 and 24 h. Following exposure, cell viability, the intracellular content of metals, glutathione (GSH) and MT and the gene expression of the two isoforms of MT was evaluated. The results obtained suggest that a lower Cd content in the co-treatments is responsible for the protection offered by Zn due to the probable competition for a common transporter. Furthermore, Zn determines an increase in GSH in co-treatments compared to treatments with Cd alone. Finally, the MTF-1 factor is essential for the expression of MT-1 but not of MT-2 nor probably for the heavy chain of γ-glutamylcysteine synthetase. [ABSTRACT FROM AUTHOR]

In recent decades, the interest in PFAS has grown exponentially around the world, due to the toxic effects induced by these chemical compounds in humans, as well as in other animals and plants. However, current knowledge related to the antistress responses that organisms can express when exposed to these substances is still insufficient and, therefore, requires further investigation. The present study focuses on antioxidant responses in Squalius cephalus and Padogobius bonelli, exposed to significant levels of PFAS in an area of the Veneto Region subjected to a recent relevant pollution case. These two ubiquitous freshwater species were sampled in three rivers characterised by different concentrations of PFAS. Several biomarkers of oxidative stress were evaluated, and the results suggest that PFAS chronic exposure induces some physiological responses in the target species, at both cellular and tissue scales. The risk of oxidative stress seems to be kept under control by the antioxidant system by means of gene activation at the mitochondrial level. Moreover, the histological analysis suggests an interesting protective mechanism against damage to the protein component based on lipid vacuolisation. [ABSTRACT FROM AUTHOR]

Per- and polyfluoroalkyl substances (PFASs) have multiple emission sources, from industrial to domestic, and their high persistence and mobility help them to spread in all the networks of watercourses. Diffuse pollution of these compounds can be potentially mitigated by the application of green infrastructures, which are a pillar of the EU Green Deal. In this context, a phytoremediation pilot plant was realised and supplied by a contaminated well-located in Lonigo (Veneto Region, Italy) where surface and groundwaters were significantly impacted by perfluoroalkyl acids (PFAAs) discharges from a fluorochemical factory. The investigation involved the detection of perfluorobutanoic acid (PFBA), perfluorooctanoic acid (PFOA), perfluorobutanesulfonic acid (PFBS) and perfluorooctanesulfonic acid (PFOS) inside the inlet and outlet waters of the phytoremediation pilot plant as well as in reed grasses grown into its main tank. The obtained results demonstrate that the pilot plant is able to reduce up to 50% of considered PFAAs in terms of mass flow without an evident dependence on physico-chemical characteristics of these contaminants. Moreover, PFAAs were found in the exposed reed grasses at concentrations up to 13 ng g−1 ww. A positive correlation between PFAA concentration in plants and exposure time was also observed. In conclusion, this paper highlights the potential efficiency of phytodepuration in PFAS removal and recommends improving the knowledge about its application in constructed wetlands as a highly sustainable choice in wastewater remediation. Abstract The effect of increasing ocean water temperature on morpho-functional traits of Antarctic marine species is under intense attention. In this work, we evaluated the effects of acute heat stress on the gills of the Antarctic haemoglobinless Chionodraco hamatus and the red blooded Trematomus bernacchii in terms of morphology, heat shock response, antioxidant defense and NOS/NO system. We showed in both species that the exposure to high temperature (4 °C) induced structural alterations, such as epithelial lifting and oedema of secondary lamellae. By immunolocalization we also observed that HSP-90, HSP-70, Xantine Oxidase, Heme Oxigenase and NOS are expressed in both species under control conditions. After heat stress the signals increase in C. hamatus being absent/or reduced in T. bernacchii. Our preliminary results suggest a specie-specific morpho-functional response of the gills of the two Antarctic teleosts to heat stress. The production of reactive oxygen species (ROS) is a normal consequence of the aerobic cell metabolism. Despite their high and potentially detrimental reactivity with various biomolecules, the endogenous production of ROS is a vital part of physiological, immunological, and molecular processes that contribute to fitness. The role of ROS in host-parasite interactions is frequently defined by their contribution to innate immunity as effectors, promoting parasite death during infections. In vertebrates, ROS and antioxidant system enzymes, such as superoxide dismutase (SOD) are also involved in acquired immune memory, where they are responsible for T-cell signalling, activation, proliferation, and viability. Based on recent findings, ROS are now also assumed to play a role in immune priming, i.e., a form of memory in invertebrates. In this study, the potential involvement of Cu,Zn SODs in immunity of the red flour beetle Tribolium castaneum is described for the first time, applying an approach that combines an in silico gene characterisation with an in vivo immune priming experiment using the Gram-positive entomopathogen Bacillus thuringiensis. We identified an unusually high number of three different transcripts for extracellular SOD and found that priming leads to a fine-tuned modulation of SOD expression, highlighting the potential of physiological co-adaptations for immune phenotypes.
