1. The use of recombinant human growth hormone in patients with Mucopolysaccharidoses and growth hormone deficiency: a case series

Abstract

Abstract Background The treatment with recombinant human growth hormone in patients affected by Mucopolysaccharidoses (MPS) is considered whenever a concurrent diagnosis of growth hormone deficiency is demonstrated. The short- and long-term effects of recombinant human growth hormone in this selected cohort is still debated, given the natural progression of disease-related skeletal malformations and the paucity of treated patients reported in literature. The presented case series provides detailed information about the response to recombinant growth hormone in MPS patients diagnosed with growth hormone deficiency. Cases presentation The growth patterns of 4 MPS female patients (current age: 11.7–14.3 years) treated with recombinant human growth hormone due to growth hormone deficiency have been retrospectively analyzed. Two patients, diagnosed with MPS IH, had undergone haematopoietic stem cell transplantation at an early age; the remaining two patients were affected by MPS IV and VI and were treated with enzyme replacement therapy. 4/4 patients presented with a progressive growth deceleration before the diagnosis of growth hormone deficiency was confirmed. This trend was initially reverted by a remarkable increase in height velocity after the start of recombinant growth hormone. We recorded an average increase in height velocity z-score of + 4.23 ± 2.9 and + 4.55 ± 0.96 respectively after 6 and 12 months of treatment. After the first 12–24 months, growth showed a deceleration in all the patients. While in a girl with MPS IH recombinant human growth hormone was discontinued due to a lack in clinical efficacy, 3/4 patients grew at a stable pace, tracking the height centile achieved after the cited initial increase in height velocity. Furthermore, mineral bone density assessed via bone densitometry, showed a remarkable increase in the two patients who were tested before and after starting treatment. Conclusions Recombinant human growth hormone appears to have effectively reverted the growth deceleration experienced by MPS patients diagnosed with growth hormone deficiency, at least during the first 12–24 months of treatment.

Abstract

The identification of multilocus imprinting disturbances (MLID) appears fundamental to uncover molecular pathways underlying imprinting disorders (IDs) and to complete clinical diagnosis of patients. However, MLID genetic associated mechanisms remain largely unknown. To characterize MLID in Beckwith-Wiedemann (BWS) and Silver-Russell (SRS) syndromes, we profiled by MassARRAY the methylation of 12 imprinted differentially methylated regions (iDMRs) in 21 BWS and 7 SRS patients with chromosome 11p15.5 epimutations. MLID was identified in 50% of BWS and 29% of SRS patients as a maternal hypomethylation syndrome. By next-generation sequencing, we searched for putative MLID-causative mutations in genes involved in methylation establishment/maintenance and found two novel missense mutations possibly causative of MLID: one in NLRP2, affecting ADP binding and protein activity, and one in ZFP42, likely leading to loss of DNA binding specificity. Both variants were paternally inherited. In silico protein modelling allowed to define the functional effect of these mutations. We found that MLID is very frequent in BWS/SRS. In addition, since MLID-BWS patients in our cohort show a peculiar pattern of BWS-associated clinical signs, MLID test could be important for a comprehensive clinical assessment. Finally, we highlighted the possible involvement of ZFP42 variants in MLID development and confirmed NLRP2 as causative locus in BWS-MLID.

Abstract

Studies showed that Brazilian Amazon indigenous territories (ITs) are efficient models for preserving forests by reducing deforestation, fires, and related carbon emissions. Considering the importance of ITs for conserving socio-environmental and cultural diversity and the recent climb in the Brazilian Amazon deforestation, we used official remote sensing datasets to analyze deforestation inside and outside indigenous territories within Brazil's Amazon biome during the 2013–2021 period. Deforestation has increased by 129% inside ITs since 2013, followed by an increase in illegal mining areas. In 2019–2021, deforestation was 195% higher and 30% farther from the borders towards the interior of indigenous territories than in previous years (2013–2018). Furthermore, about 59% of carbon dioxide (CO2) emissions within ITs in 2013–2021 (96 million tons) occurred in the last three years of analyzed years, revealing the magnitude of increasing deforestation to climate impacts. Therefore, curbing deforestation in indigenous territories must be a priority for the Brazilian government to secure these peoples' land rights, ensure the forests' protection and regulate the global climate.

Abstract

Objective. This work aims at characterizing LiF:Mg,Ti thermoluminescence detectors (TLDs) for dosimetry of a 250 MeV proton beam delivered at ultra-high dose rates (UHDR). Possible dose rate effects in LiF:Mg,Ti, as well as its usability for dosimetry of narrow proton beams are investigated. Approach. LiF:Mg,Ti (TLD-100TM Microcubes, 1 mm × 1 mm × 1 mm) was packaged in matrices of 5 × 5 detectors. The center of each matrix was irradiated with single-spot low-LET (energy >244 MeV) proton beam in the (1–4500) Gy s−1 average dose rates range. 6. Periodontitis Is Associated with Consumption of Processed and Ultra-Processed Foods:Findings from a Population-Based Study

Abstract

The association between periodontitis and lifestyle factors has been widely investigated. However, an association between periodontitis and dietary patterns has not been explored. Therefore, this study investigated the association between periodontitis and food consumption among a Southern Brazil population. Data from the 1982 Pelotas Birth Cohort were used (n = 537). The exposure, periodontitis, was clinically measured and classified using the AAP/CDC system, then two latent variables were defined: ‘initial’ and ‘moderate/severe’ periodontitis. The consumption of in natura, processed, and ultra-processed foods (NOVA classification) was the outcome and measured in calories using the food frequency questionnaire (FFQ). Confounders were sex, maternal education, smoking status, xerostomia, and halitosis. Data were analyzed by structural equation modeling. ‘Initial’ periodontitis was associated with a higher consumption of in natura food (standardized coefficient (SC) 0.102; p-value = 0.040), versus processed (SC 0.078; p-value = 0.129) and ultra-processed (SC 0.043; p-value = 0.400) foods. ‘Moderate/severe’ periodontitis was associated with higher consumption of ultra-processed foods (SC 0.108; p-value = 0.024), versus processed (SC 0.093; p-value = 0.053) and in natura (SC 0.014; p-value = 0.762) foods. ‘Moderate/severe’ periodontitis appears to be associated with the consumption of processed and ultra-processed foods.

7. Soft tissue vascular tumor-like lesions in adults: imaging and pathological analysis pitfalls per ISSVA classification

Abstract

Abstract Objectives To compare the magnetic resonance imaging (MRI) and Doppler ultrasound (DUS) findings with the pathological findings of soft tissue vascular tumors (STVTs) according to the 2018 ISSVA (International Society for the Study of Vascular Anomalies) classification to differentiate vascular tumors from vascular malformations. Methods This retrospective study included patients with STVTs who underwent contrast-enhanced MRI and pathological analysis at our hospital between 2010 and 2020. The presumptive diagnosis based on the on-site imaging and histological analysis was compared with imaging and histological analysis conducted off-site utilizing the ISSVA criteria. Results This study included 31 patients with 31 vascular tumors located in the head and neck (n = 3), trunk (n = 2), and extremities (n = 26). The off-site pathological analysis confirmed benign vascular tumors in 54.8% of cases (non-involuting congenital hemangioma: 35.5%; epithelioid hemangioma: 13%; pyogenic granuloma: 3%; and spindle cell hemangioma: 3%). Based on the off-site histological analysis, 25.8% were reclassified as having a vascular malformation whereas three had other benign lesions. Only phleboliths were associated with a vascular malformation (p = 0.03). The concordance between off-site MRI and pathological findings was fair (k = 0.3902 (0.0531–0.7274)), whereas that between on-site and off-site pathological analyses was poor (k = −0.0949 (−0.4661 to 0.2763)). Conclusion Benign vascular tumors have non-specific imaging features on imaging with some overlap with atypical vascular malformations. Therefore, histological analysis is recommended. Imaging and pathological analyses should be performed in accordance with the ISSVA classification to minimize inter-observer discrepancies. Critical relevance statement Imaging features of benign vascular tumors on MRI are non-specific, leading to discrepancies with pathological findings and potential overlap with atypical vascular malformations. Imaging and histological analyses should be performed in accordance with ISSVA guidelines to improve patient management. Key Points The imaging features of benign vascular tumors are non-specific. Histological analysis is recommended for soft tissue vascular tumors in adults. Analyses of soft tissue vascular tumors should be performed in accordance with ISSVA guidelines. Graphical Abstract

Reef fishes are a major component of tropical marine biodiversity, but their abundance and distribution are poorly assessed beyond conventional SCUBA diving depths. Based on a baited-video survey of fish assemblages in Southwestern Atlantic's most biodiverse region we show that species composition responded mainly to the two major hard-bottom megahabitats (reefs and rhodolith beds) and to the amount of light reaching the bottom. Both megahabitats encompassed typical reef fish assemblages but, unexpectedly, richness in rhodolith beds and reefs was equivalent. The dissimilar fish biomass and trophic structure in reefs and rhodolith beds indicates that these systems function based on contrasting energy pathways, such as the much lower herbivory recorded in the latter. Rhodolith beds, the dominant benthic megahabitat in the tropical Southwestern Atlantic shelf, play an underrated role as fish habitats, and it is critical that they are considered in conservation planning. Abstract

Objective: Sensory deficits are important risk factors for delirium but have been investigated in single-center studies and single clinical settings. This multicenter study aims to evaluate the association between hearing and visual impairment or bi-sensory impairment (visual and hearing impairment) and delirium. Design: Cross-sectional study nested in the 2017 "Delirium Day" project. Setting and participants: Patients 65 years and older admitted to acute hospital medical wards, emergency departments, rehabilitation wards, nursing homes, and hospices in Italy. Methods: Delirium was assessed with the 4AT (a short tool for delirium assessment) and sensory deficits with a clinical evaluation. We assessed the association between delirium, hearing and visual impairment in multivariable logistic regression models, adjusting for: Model 1, we included predisposing factors for delirium (ie, dementia, weight loss and autonomy in the activities of daily living); Model 2, we added to Model 1 variables, which could be considered precipitating factors for delirium (ie, psychoactive drugs and urinary catheters). Results: A total of 3038 patients were included; delirium prevalence was 25%. Patients with delirium had a higher prevalence of hearing impairment (30.5% vs 18%; P < .001), visual impairment (24.2% vs 15.7%; P < .01) and bi-sensory impairment (16.2% vs 7.5%) compared with those without delirium. In the multivariable logistic regression analysis, the presence of bi-sensory impairment was associated with delirium in Model 1 [odds ratio (OR) 1.5, confidence interval (CI) 1.2-2.1; P = .00] and in Model 2 (OR 1.4; CI 1.1-1.9; P = .02), whereas the presence of visual and hearing impairment alone was not associated with delirium either in Model 1 (OR 0.8; CI 0.6-1.2, P = .36; OR 1.1; CI 0.8-1.4; P = .42) or in Model 2 (OR 0.8, CI 0.6-1.2, P = .27; OR 1.1, CI 0.8-1.4, P = .63). Conclusions and implications: Our findings support the importance of routine screening and specific interventions by a multidisciplinary team to implement optimal management of sensory impairments and hence prevention and the management of the patients with delirium.

No. of bitstreams: 0 Previous issue date: 2021-09-01 Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Marine protected areas (MPAs) represent a useful tool for resource management, as well as to conserve and/or restore biological communities. The level of protection is key factor influencing the marine biodiversity, where a more enforced protection is expected to drive positive outcomes. In 2008, a large MPAs network (∼11,380 km 2) was established in one of off the most populated and industrialized areas in the world (i.e., São Paulo State coast, southeast Brazil). Given many goods and services provided by marine ecosystems, this MPA network represents the most challenging marine conservation initiative in Brazil. Harboring​ areas with different socio-ecological contexts and management regimes, this MPA network provides a unique opportunity to investigate the effects of cumulative impacts. We contrasted the biomass and size structure of reef fish in three subtropical islands under different levels of enforcement. We analyzed the influence of variables as island size, benthic cover, depth, topographic complexity, wave exposure, and protection level on the biomass of reef fish assemblages. Protection level was the main attribute responsible to explain the high biomass of fish target species and small territorial herbivores. In sites sheltered from the waves, the biomass of groupers was ∼1600% higher within enforced area than that from open-access area. Beyond the idea of positive effects of enforcement on reef fish biomass and size, we add evidences that even under multiple stressors, the area-based management is still a strong tool to marine conservation. Abstract

Background The treatment with recombinant human growth hormone in patients affected by Mucopolysaccharidoses (MPS) is considered whenever a concurrent diagnosis of growth hormone deficiency is demonstrated. The short- and long-term effects of recombinant human growth hormone in this selected cohort is still debated, given the natural progression of disease-related skeletal malformations and the paucity of treated patients reported in literature. The presented case series provides detailed information about the response to recombinant growth hormone in MPS patients diagnosed with growth hormone deficiency. Cases presentation The growth patterns of 4 MPS female patients (current age: 11.7–14.3 years) treated with recombinant human growth hormone due to growth hormone deficiency have been retrospectively analyzed. Two patients, diagnosed with MPS IH, had undergone haematopoietic stem cell transplantation at an early age; the remaining two patients were affected by MPS IV and VI and were treated with enzyme replacement therapy. 4/4 patients presented with a progressive growth deceleration before the diagnosis of growth hormone deficiency was confirmed. This trend was initially reverted by a remarkable increase in height velocity after the start of recombinant growth hormone. We recorded an average increase in height velocity z-score of + 4.23 ± 2.9 and + 4.55 ± 0.96 respectively after 6 and 12 months of treatment. After the first 12–24 months, growth showed a deceleration in all the patients. While in a girl with MPS IH recombinant human growth hormone was discontinued due to a lack in clinical efficacy, 3/4 patients grew at a stable pace, tracking the height centile achieved after the cited initial increase in height velocity. Furthermore, mineral bone density assessed via bone densitometry, showed a remarkable increase in the two patients who were tested before and after starting treatment. Conclusions Recombinant human growth hormone appears to have effectively reverted the growth deceleration experienced by MPS patients diagnosed with growth hormone deficiency, at least during the first 12–24 months of treatment.

17. First study on age and growth of the deep-water Goblin Shark, Mitsukurina owstoni (Jordan, 1898)

Abstract

Due to poorly mineralizing structures, ageing deep-water elasmobranchs requires nonconventional techniques. The aim of the present study was to develop a reliable ageing technique using the vertebral centrum to provide information about the age and growth parameters in the Goblin Shark, Mitsukurina owstoni (Jordan, 1898). One vertebral centrum from an individual measuring 315.2 cm in total length was analysed. A minimum age of 27 years was estimated. By incorporating priors based on the growth of deep-water sharks and an additional likelihood on L∞ using data on large male Goblin Sharks, a Bayesian von Bertalanffy growth model was estimated with a male L∞ of 364 cm total length, weigh 215 kg at L∞, grow slowly with a k equal to 0.049, mature at 16.5 years, and live up to 55 years. Our results are essential to provide useful life history information, with the aim of elucidating the cryptic ecology and biology of this deep-water shark.

Abstract

Organic selenium, tellurium and sulfur compounds have been studied due to their pharmacological properties. For instance, the β-aryl-chalcogenium azide compounds have demonstrated antitumoral action in vitro. However, yet no pharmacological actions of this class of compounds were determined in vivo. Caenorhabditis elegans is a nematode that presents innumerable advantages in relation to mammalian models, such as having a small and transparent body, which allows the visualization of its internal anatomy, besides short life and low cost. Based on that, the aim of this work was to investigate the pharmacological and toxicological properties of β-aryl-chalcogenium azide compounds in C. elegans. As well, to evaluate the capacity of organochalcogenium compounds to repair oxidative damage induced by hydrogen peroxide and the possible mechanism of action of these compounds using CF1553 transgenic strain with superoxide dismutase (SOD-3) tagged with GFP. Our results showed that β-aryl-chalcogenium azide have low toxicity in wild-type worms and the pre-treatment protected against the damage induced by hydrogen peroxide at higher tested concentration. Associated with this, we observed that this protection is due in part to the increased expression of the antioxidant enzyme SOD-3. In conclusion, β-aryl-chalcogenium azide compounds caused low toxicity and induced stress-resistance by modulating SOD-3 expression in C. elegans.

Abstract

Introduction: Mucopolysaccharidosis (MPS) IIIB is a lysosomal disorder in which a deficiency in α-N-acetylglucosaminidase impairs the degradation of heparan sulphate, which accumulates in tissues causing multiple organs dysfunction. This disease is associated with significant central nervous system (CNS) abnormalities, but a presentation with a tumour-like lesion has never been reported so far. Clinical presentation: The present report describes the case of a 5-year-old girl suffering from MPS IIIB who developed a cerebellar lesion with evident mass effect. She underwent surgery with a subsequent notable improvement of her clinical picture. Surprisingly, the pathological analysis revealed the lesion to have the typical MPS features. Conclusion: This case would describe a neglected possible presentation of MPS IIIB with a lesion mimicking a neoplasm, which could even be successfully treated with surgery.

No. of bitstreams: 0 Previous issue date: 2019-01-01 Palosuojelurahasto Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) No-take marine reserves (NTRs), i.e. areas with total fishing restrictions, have been established worldwide aiming to promote biodiversity and ecosystem conservation. Brazil has 3.3% of its exclusive economic zone protected by 73 different NTRs, however, most of them currently lack scientific knowledge and understanding of their ecological role, particularly regarding rocky reefs in subtropical regions. In this context, this study aimed to contrast a network of NTRs with comparable fished sites across a coastal biogeographic gradient to investigate the effect of fishing and habitat variability on the abundance and body size of rocky reef fish. We used Baited Remote Underwater stereo-Video (stereo-BRUVs) and Diver Operated stereo-Video (stereo-DOVs) systems to simultaneously sample reef fish and habitat. Model selection and results identified habitat and biogeographic variables, such as distance from shore, as important predictor variables, explaining several aspects of the fish assemblage. The effect of protection was important in determining the abundance and body size of targeted species, in particular for epinephelids and carangids. Conversely, species richness was correlated with habitat complexity but not with protection status. Abstract

Accumulating evidence suggests that pesticides have played a role in the increased rate of honey bee colony loss. One of the most commonly used pesticides in the United States is the neonicotinoid imidacloprid. Although the primary mode of action of imidacloprid is on the insect nervous system, it has also been shown to cause changes in insects' digestive physiology and alter the microbiota of Drosophila melanogaster larvae. The honey bee gut microbiome plays a major role in bee health. Although many studies have shown that imidacloprid affects honey bee behavior, its impact on the microbiome has not been fully elucidated. Here, we investigated the impact of imidacloprid on the gut microbiome composition, survivorship, and susceptibility to pathogens of honey bees. Consistent with other studies, we show that imidacloprid exposure results in an elevated mortality of honey bees in the hive and increases the susceptibility to infection by pathogens. However, we did not find evidence that imidacloprid affects the gut bacterial community of honey bees. Our in vitro experiments demonstrated that honey bee gut bacteria can grow in the presence of imidacloprid, and we found some evidence that imidacloprid can be metabolized in the bee gut environment. However, none of the individual bee gut bacterial species tested could metabolize imidacloprid, suggesting that the observed metabolism of imidacloprid within in vitro bee gut cultures is not caused by the gut bacteria. Overall, our results indicate that imidacloprid causes increased mortality in honey bees, but this mortality does not appear to be linked to the microbiome. IMPORTANCE Growing evidence suggests that the extensive use of pesticides has played a large role in the increased rate of honey bee colony loss. Despite extensive research on the effects of imidacloprid on honey bees, it is still unknown whether it impacts the community structure of the gut microbiome. Here, we investigated the impact of imidacloprid on the gut microbiome composition, survivorship, and susceptibility to pathogens of honey bees. We found that the exposure to imidacloprid resulted in an elevated mortality of honey bees and increased the susceptibility to infection by opportunistic pathogens. However, we did not find evidence that imidacloprid affects the gut microbiome of honey bees. We found some evidence that imidacloprid can be metabolized in the bee gut environment in vitro , but because it is quickly eliminated from the bee, it is unlikely that this metabolism occurs in nature. Thus, imidacloprid causes increased mortality in honey bees, but this does not appear to be linked to the microbiome.

Abstract

We present a systematic study of the orbital inclination effects on black-hole transients fast time-variability properties. We have considered all the black-hole binaries that have been densely monitored by the Rossi XTE satellite. We find that the amplitude of low-frequency quasi periodic oscillations (QPOs) depends on the orbital inclination. Type-C QPOs are stronger for nearly edge-on systems (high inclination), while type-B QPOs are stronger when the accretion disk is closer to face-on (low inclination). Our results also suggest that the noise associated with type-C QPOs is consistent with being stronger for low-inclination sources, while the noise associated to type-B QPOs seems inclination independent. These results are consistent with a geometric origin of the type-C QPOs - for instance arising from relativistic precession of the inner flow within a truncated disk - while the noise would correspond to intrinsic brightness variability from mass accretion rate fluctuations in the accretion flow. The opposite behavior of type-B QPOs - stronger in low inclinations sources - supports the hypothesis that type-B QPOs are related to the jet, the power of which is the most obvious measurable parameter expected to be stronger in nearly face-on sources., 8 pages, 6 figures, accepted by MNRAS

Abstract

Bacteria producing antimicrobial activity were identified among 86 isolates from aquatic environments of Brazilian Amazon Basin. Antimicrobial activity against at least one indicator strain was detected for 59 isolates (68.6%). Inhibitory activity was mostly against Gram-positive bacteria, such as Listeria monocytogenes and Bacillus cereus. The antimicrobial substances produced by 19 strains that showed higher inhibitory activity were partially characterized. These antimicrobial substances showed thermal resistance for up to 100°C and partial resistance to proteolytic treatment. Some antimicrobials were partially resistant to pronase E at 2 mg ml-1 or to treatment with trichloroacetic acid. Detection of antimicrobial activity on polyacrylamide gels revealed that the molecular weight of the compounds was lower than 14 kDa. Several strains showed antibacterial activity, which in some cases appear to be related to antimicrobial peptides. The potential of these microorganisms to produce useful antimicrobial compounds is great and must be better explored.Bactérias produtoras de atividade antimicrobiana foram identificadas entre 86 isolados de ambientes aquáticos da Bacia Amazônica. Destes, 59 isolados (68.6%) apresentaram atividade antimicrobiana contra pelo menos uma bactéria indicadora. A atividade inibitória foi principalmente observada contra bactérias Gram-positivas, como Listeria monocytogenes e Bacillus cereus. As substâncias antimicrobianas produzidas por 19 linhagens que demonstraram maior atividade inibitória foram parcialmente caracterizadas, apresentando resistência térmica até 100°C e resistência parcial ao tratamento proteolítico. Algumas substâncias foram parcialmente inativadas somente quando tratadas com ácido tricloroacético ou com pronase E na concentração de 2 mg ml-1. A detecção da atividade antimicrobiana em géis de poliacrilamida mostrou que os compostos apresentaram peso molecular inferior à 14 kDa. Várias linhagens apresentaram atividade antibacteriana, que em alguns casos estaria relacionada com peptídios antimicrobianos. O potencial destes microrganismos para produzir substâncias antimicrobianas é grande e merece ser mais explorado.

OBJETIVOS: Avaliar a influência da utilização do colírio anestésico, bem como verificar o comportamento da medida da pressão intra-ocular (Po) com a adaptação (aprendizado) do paciente à tonometria de não-contato (TNC). MATERIAIS E MÉTODOS: Estudo clínico randomizado duplo-mascarado em que se avaliaram 24 olhos de 24 voluntários normais, sem experiência prévia com a TNC, divididos em 2 grupos: grupo I (TNC com instilação prévia de colírio anestésico seguido de tonometria de aplanação de Goldmann (TAG)) e grupo II (TNC com instilação prévia de colírio placebo seguido de TAG). As medidas da Po foram realizadas em três visitas com intervalo de um a três dias aproximadamente no mesmo horário. RESULTADOS: Constatou-se a Po por meio da TAG como a única variável preditora estatisticamente significante (p < 0,05) para a TNC. Não houve diferença na variação da Po entre os grupos (p = 0,884), tampouco entre as visitas (p = 0,144). Houve melhor correlação entre a TNC e TAG no grupo II em relação ao grupo I em todas as visitas, porém esta diferença não se mostrou estatisticamente significante (p = 0,4853). CONCLUSÕES: A utilização de anestésico tópico e a adaptação de indivíduos normais ao exame de tonometria de não-contato não demonstraram influência significante sobre a medida da Po obtida por este método.PURPOSE: To evaluate the influence of the previous instillation of anesthetic eye drops, as well as to verify intraocular pressure (IOP) performance with adaptation to noncontact tonometry (NCT). Cancer immunoprevention, a recent development of tumor immunology based on vaccines and other immunological maneuvers to actively reduce the risk of cancer development, will greatly benefit from immunomic approaches for its progress in basic research and for its translation to human applications. The main approaches discussed in this chapter are the use of genetically-modified mouse models, microarray studies and data mining approaches to define antitumor immune responses and to discover new oncoantigens, and agent-based mathematical models to simulate tumor-immune system interactions and to perfect vaccination protocols.

Abstract

The Large Observatory For x-ray Timing (LOFT) was studied within ESA M3 Cosmic Vision framework and participated in the final down-selection for a launch slot in 2022-2024. Thanks to the unprecedented combination of effective area and spectral resolution of its main instrument, LOFT will study the behaviour of matter under extreme conditions, such as the strong gravitational field in the innermost regions of accretion flows close to black holes and neutron stars, and the supra-nuclear densities in the interior of neutron stars. The science payload is based on a Large Area Detector (LAD, 10 m 2 effective area, 2-30 keV, 240 eV spectral resolution, 1 deg collimated field of view) and a WideField Monitor (WFM, 2-50 keV, 4 steradian field of view, 1 arcmin source location accuracy, 300 eV spectral resolution). The WFM is equipped with an on-board system for bright events (e.g. GRB) localization. The trigger time and position of these events are broadcast to the ground within 30 s from discovery. Abstract

Mathematical models are quantitative representation of phenomena built up in the framework of a theory using the language of mathematics. Living organisms are natural complex systems and modelling may play a crucial role since models can also be built with approximate and imperfect knowledge of the phenomenon and model parameters (initial data, entities, relations between entities) can be adjusted to fit modeling results to experimental measurements. These models can then be used to understand general behavior of the phenomenon in different situations; perform model experiments or simulations to understand the role of single constituents and relations; plan new experiments; test theoretical assumptions and suggest theory modifications. Modelling can therefore stimulate scientific creativity and produce better theoretical descriptions of the reality. We describe here our efforts in the creation of models of the immune system and of the competition between immune defenses and tumor cells. We developed an agent-based model of the effects of a vaccine designed to prevent mammary carcinoma incidence in transgenic mice. This model faithfully recapitulates not only the outcome of vaccination experiments, but also the dynamics of immune responses elicited by the vaccine. We then used a genetic algorithm to drive the model and predict optimized vaccination schedules which are currently being tested in vivo. We discuss here also the implications of biologic diversity on model development and the perpspectives to develop natural-scale models of the immune system.

Abstract

We present an evolutionary approach to the search for effective vaccination schedules using mathematical computerized model as a fitness evaluator. Our study is based on our previous model that simulates the Cancer - Immune System competition activated by a tumor vaccine. The model reproduces pre-clinical results obtained for an immunoprevention cancer vaccine (Triplex) for mammary carcinoma on HER-2/neu mice. A complete prevention of mammary carcinoma was obtained in vivo using a Chronic vaccination schedule. Our genetic algorithm found complete immunoprevention with a much lighter vaccination schedule. The number of injections required is roughly one third of those used in Chronic schedule.

Abstract

We present a systematic analysis of the fast time variability properties of the transient black hole binary GRO J1655-40, based on the complete set of RossiXTE observations. We demonstrate that the frequencies of the quasi-periodic oscillations and of the broad band noise components and their variations match accurately the strong field general relativistic frequencies of particle motion in the close vicinity of the innermost stable circular orbit, as predicted by the relativistic precession model. We obtain high precision measurements of the black hole mass (M = (5.31 +/- 0.07) M solar masses, consistent with the value from optical/NIR observations) and spin (a/M = 0.290 +/- 0.003), through the sole use of X-ray timing., 16 pages, 4 figures, 3 tables. Infectious diseases are the world’s leading cause of premature deaths, killing almost 50,000 people every day. In recent years, drug resistance to human pathogenic bacteria has been commonly reported from all over the world (N’guessan et al., 2007). The abusive and indiscriminate use of antimicrobial compounds over many years is the main factor responsible for the appearance of the phenomenon of bacterial resistance to such compounds (Andremont, 2001). With increased incidence of resistance to antibiotics, natural products from plants could be interesting alternatives (Lu et al., 2007; Mbwambo et al., 2007). Some plant extracts and phytochemicals are known to have antimicrobial properties, and can be of great significance in therapeutic treatments. In the last few years, a number of studies have been conducted in different countries to demonstrate such efficacy (BenoitVical et al., 2006; Senatore et al., 2007; Singh et al., 2007). On the other hand, free radicals are known to be the major cause of various chronic and degenerative diseases. Oxidative stress is associated with pathogenic mechanisms of many diseases including atherosclerosis, neurodegenerative diseases, cancer, diabetes and inflammatory diseases, as well as aging processes. It is defined as an imbalance between production of free radicals and reactive metabolites, so-called oxidants, and it also includes their elimination by protective mechanisms, referred to as antioxidative systems. This imbalance leads to damage of important biomolecules and organs with potential impact on the whole organism. Antioxidants can delay, inhibit or prevent the oxidation of oxidizable materials by scavenging free radicals and diminishing oxidative stress (Durackova, 2010; Reuter et al., 2010). Natural antioxidants have been studied extensively for decades in order to find compounds protecting against a number of diseases related to oxidative stress and free radical-induced damage. Abstract

On 2010 October 13, the X-ray astronomical satellite Rossi XTE, during the observation of the newly discovered accretion powered X-ray pulsar IGR J17480--2446, detected a lunar occultation of the source. From knowledge of lunar topography and Earth, Moon, and spacecraft ephemeris at the epoch of the event, we determined the source position with an accuracy of 40 mas (1{\sigma} c.l.), which is interesting, given the very poor imaging capabilities of RXTE (\sim 1\circ). For the first time, using a non-imaging X-ray observatory, the position of an X-ray source with a subarcsecond accuracy is derived, demonstrating the neat capabilities of a technique that can be fruitfully applied to current and future X-ray missions., Comment: 5 pages, 3 figures, accepted for publication on APJL

Abstract

Papain, a phytotherapeutic agent, has been used in the treatment of eschars and as a debriding chemical agent to remove damaged or necrotic tissue of pressure ulcers and gangrene. Its benefits in these treatments are deemed effective, since more than 5000 patients, at the public university hospital at Rio de Janeiro, Brazil, have undergone papain treatment and presented satisfactory results. Despite its extensive use, there is little information about toxic and mutagenic properties of papain. This work evaluated the toxic and mutagenic potential of papain and its potential antioxidant activity against induced-H2O2oxidative stress inEscherichia colistrains. Cytotoxicity assay, Growth inhibition test, WP2-Mutoxitest and Plasmid-DNA treatment, and agarose gel electrophoresis were used to investigate if papain would present any toxic or mutagenic potential as well as if papain would display antioxidant properties. Papain exhibited negative results for all tests. This agent presented an activity protecting cells againstH2O2-induced mutagenesis.

Abstract

This work shows a simple, single-stage, scalable method for the continuous production of high-quality carbon nanotube-polymer transparent conductive films from carbon feedstock. Besides the ease of scalability, a particular advantage of this process is that the concentration of nanotubes in the films, and thus transparency and conductivity, can be adjusted by changing simple process parameters. Therefore, films can be readily prepared for any application desired, ranging from solar cells to flat panel displays. Our best results show a surface resistivity of the order of 300 Ω square-1 for a film with 80% transparency, which is promising at this early stage of process development.

Abstract

Reactive oxygen species (ROS) can induce lesions in different cellular targets, including DNA. Stannous chloride (SnCl2) is a ROS generator, leading to lethality in Escherichia coli (E. coli), with the base excision repair (BER) mechanism playing a role in this process. Many techniques have been developed to detect genotoxicity, as comet assay, in eukaryotic cells, and plasmid DNA agarose gel electrophoresis. In this study, an adaptation of the alkaline gel electrophoresis method was carried out to ascertain the induction of strand breaks by SnCl2 in bacterial DNA, from E. coli BER mutants, and its repair pathway. Results obtained show that SnCl2 was able to induce DNA strand breaks in all strains tested. Moreover, endonuclease IV and exonuclease III play a role in DNA repair. On the whole, data has shown that the alkaline gel electrophoresis assay could be used both for studying DNA strand breaks induction and for associated repair mechanisms.Espécies reativas de oxigênio (ERO) podem induzir lesões em diferentes alvos celulares, incluindo o DNA. O cloreto estanoso (SnCl2) é um gerador de ERO que induz letalidade em E. coli, sendo o reparo por excisão de bases (BER) um mecanismo importante neste processo. Técnicas como o ensaio cometa (em eucariotos) e a eletroforese de DNA plasmidial em gel de agarose têm sido utilizadas para detectar genotoxicidade. No presente estudo, uma adaptação do método de eletroforese em gel alcalino de agarose foi usada para verificar a indução de quebras, pelo SnCl2, no DNA de E. coli, bem como a participação de enzimas do BER na restauração das lesões. Os resultados mostraram que o SnCl2 induziu quebras no DNA de todas as cepas testadas. Além disso, endonuclease IV e exonuclease III estão envolvidas na reparação dos danos. Em resumo, os dados obtidos indicam que a metodologia de eletroforese em gel alcalino de agarose pode ser empregada tanto para o estudo de quebras no DNA, quanto para avaliação dos mecanismos de reparação associados.

Abstract

This study presents results obtained from 83 patients with bilateral vocal cord paralysis in adduction treated between 1982 to 2001, with CO(2) laser microlaryngoscopy. In relation to the different types of surgery followed, three distinct treatment groups were included: group 1 (1982-1984) included 5 patients treated with vaporization of the vocal process of the arytenoid and the homolateral posterior third of the true vocal cord. Group 2 (1983-1990) contained 19 patients who were treated with arytenoidectomy and removal of the homolateral posterior half of the true vocal cord and group 3 (1990-2001), including 59 patients who were treated with arytenoidectomy and removal of the homolateral posterior half or two thirds of both the true and false vocal cord. Functional results were assessed by means of spirometry, spectrography and aerophonic examinations performed at 5, 90, 180, and 240 days postoperatively. The results show that removal of the posterior third of the true vocal cord and false vocal cord, combined with arytenoidectomy, is the surgical treatment of choice to resolve respiratory insufficiency in these patients.

The first derivative amplitude measurements show that Beer's law is obeyed in the range 0-7.5 µg ml-1 for V at 465 nm and 0-4.5 µg ml-1 for Cu at 443 nm. The sensitivity of the method is greater than the zero order method, and foreign ions like UVI, ThIV, CrVI' which interfere in zero order method, do not interfere even in large excess. Vanadium and copper present in a mixture are simultaneously determined without solving the simultaneous equations by measuring the first derivative amplitudes at 412.6 and 430.2 nm for V and Cu, respectively. The method is applied for the determination of Cu in food materials, V in alloy steels, and both V and Cu in chromium-vanadium steel.

Abstract

'Compositional chromosomal mapping', namely the assessment of the GC level of chromosomal bands, led to the identification, in the human chromosomes, of the GC-richest H3+ bands and of the GC-poorest L1+ bands, which were so called on the basis of the isochore family predominantly present in the bands. The isochore organization of the avian genome is very similar to those of most mammals, the only difference being the presence of an additional, GC-richest, H4 isochore family. In contrast, the avian karyotypes are very different from those of mammals, being characterized, in most species, by few macrochromosomes and by a large number of microchromosomes. The 'compositional mapping' of chicken mitotic and meiotic chromosomes by in-situ hybridization of isochore families showed that the chicken GC-richest isochores are localized not only on a large number of microchromosomes but also on almost all telomeric bands of macrochromosomes. On the other hand, the GC-poorest isochores are generally localized on the internal regions of macrochromosomes and are almost absent in microchromosomes. Thus, the distinct localization of the GC-richest and the GC-poorest bands observed on human chromosomes appears to be a general feature of chromosomes from warm-blooded vertebrates.

Abstract

The Authors discuss post-operative failures and complications detected in their case-report of 1857 patients, operated for otosclerotic disease between 1982 and 1993 (399 large fenestra and 1458 small fenestra stapedotomies), with reference to the data from international literature. Their study reveals that the most frequent clinical event caused by complications arising is still hypoacusia of various degree, both conductive (2.7%) and sensorineural (6.6%); more precisely, conductive hearing loss points to revision surgery, while sensorineural lesions, often limited to high frequencies (5.7%), may unfortunately take the form of anacousia (0.9%). Some extra-auditory complications (temporary facial paralysis, ear drum perforation, disgeusia, tinnitus) are also present with relative frequency in stapes surgery (4.2%); among them, however, only impairment or appearance of tinnitus (0.6%) turn out to be a persistent disorder. Analysis of the results underlines that the complications rate is higher in the case of coexisting morphological anomalies (such as stenosis of the external ear canal, disjointing or amputation of the incus, difficulty in approaching the oval window, reobliteration of the oval window, facial nerve prolapse) or when intraoperative problems arise (excessive bleeding, profuse perylymph flow), which make the operation more difficult to perform. The data obtained in this study show that stapedotomy (using a 0.6 mm diameter prosthesis), gives a significantly lower rate of sensorineural complications (4.1%), than platinectomy (15.6%), because of the reduction of labirinthine traumatism. Lastly, the Authors performed 64 revision operations, evaluating intraoperative findings and hearing recovery rate. Causes of failure most frequently detected during revision surgery were: incus necrosis (24.6%) and prosthesis displacement (21.3%) with possible reobliteration of the oval window (23%); however it has not always been possible to detect pathogenesis of a postoperative conductive hearing loss. All things considered, results of revision surgery seem to be less satisfactory (56% improvements, 4.9% severe sensorineural hearing loss) than those obtained after the first operation.

Abstract

Treatment of allergic vasomotor rhinitis is to be regarded with the factors that modify the symptomatology. In fact must be considered the morphological changes (septum deviation, adenoids, turbinates hypertrophy, polyps) infections (bacteria, chlamydiae, micetes) and specific allergens. Identification of allergens and sensitization threshold is to be studied; then specific hyposensitization will be assessed. In the present study, 68 subjects having nasal reactivity have been observed and underwent to different ways of treatment: 18 with permanent stenosis identified by rhinomanometry and not modified with vasoconstrictors were operated; 25 of the 50 patients with normal nasal cavities showed contemporary infections: after a specific antimicrobic or antimycotic treatment, a clear improvement was obtained documented by rhinomanometry before and after nasal stimulations. The 50 patients with normal morphology underwent a local hyposensitization against the allergens. As a matter of fact: a) in all cases a clear improvement was obtained in phase of increasing; b) after one year of maintainance just 13 over 50 (26%) returned to previous conditions. The Authors remark how local immunotherapy by the nasal way give good possibilities in a high percentage of cases in the following conditions: correct clinical evaluation, especially concerning the identification of factors determining vasomotor rhinitis; employment of precise techniques for diagnosis; observation of clinical data and results turning out from instrumental investigations, especially concerning the nasal provocation test evaluated by rhinomanometry.

Abstract

This paper presents the results of our analysis about the influence of Information Technology (IT) malicious traffic on an IP-based automation environment. We utilized a traffic generator, called MACE (Malicious trAffic Composition Environment), to inject malicious traffic in a Modbus/TCP communication system and a sniffer to capture and analyze network traffic. The realized tests show that malicious traffic represents a serious risk to critical information infrastructures. We show that this kind of traffic can increase latency of Modbus/TCP communication and that, in some cases, can put Modbus/TCP devices out of communication. [ABSTRACT FROM AUTHOR]

Abstract

Multiobjective optimization (MO) techniques allow a designer to model a specific problem considering a more realistic behavior, which commonly involves the satisfaction of several targets simultaneously. A fundamental concept, which is adopted in the multicriteria optimization task, is that of Pareto optimality. In this paper we test several well-known procedures to deal with multiobjective optimization problems (MOP) and propose a novel modified procedure that when applied to the existing Normal Boundary Intersection (NBI) method and Normal Constraint (NC) method for more than two objectives overcomes some of their deficiencies. For the three and four objective applications analyzed here, the proposed scheme presents the best performance both in terms of quality and efficiency to obtain a set of proper Pareto points, when compared to the analyzed existing approaches.
