Abstract Objectives To explore the role of nodule-pleural relationship, including nodule with pleural tail sign (PTS), nodule with pleural contact and nodule with pleural unrelated in CT-guided percutaneous transthoracic needle biopsy (PTNB)-induced pneumothorax, and whether employing different puncture routes has an impact on the incidence of pneumothorax in PTNB of nodules with PTS. Methods Between April 1, 2019, to June 30, 2021, 775 consecutive PTNB procedures of pulmonary nodules in the Peking University Cancer Hospital were retrospectively reviewed. The univariate and multivariate regression analysis were used to identify the risk factors for pneumothorax in PTNB. Results The nodule with pleural contact group has a lower incidence of pneumothorax than the nodule with PTS group (p = 0.001) and the nodule with pleural unrelated group (p = 0.002). 8. A Personalized Course Recommendation Model Integrating Multi-Granularity Sessions and Multi-Type Interests

- Abstract
The open online course (MOOC) platform has seen an increase in usage, and there are a growing number of courses accessible for people to select. An effective method is urgently needed to recommend personalized courses for users. Although the existing course recommendation models consider that users' interests change over time, they often model users' learning records as a single time-granularity sequence and ignore the collaboration between different time-granularity sessions when recommending courses. In addition, most course recommendation models tend to use the deep network, which weakens the memory ability of the model. Few methods simultaneously consider long and short-term interests and individual course interests in the latest session, which results in a decline in model performance. To resolve these problems, we design an innovative personalized course recommendation model that Integrating Multi-granularity Sessions and Multi-type Interests (IMSMI), which converts user-course interaction sequences as multi-granularity sessions and uses different types of attention mechanisms to capture multi-type interests. Meanwhile, we introduce the residual connections to further strengthen the memory capability of IMSMI. Experimental results using the XuetangX dataset available to the public demonstrate that IMSMI significantly surpasses other competing models on evaluation metrics. Compared to the next best model, Recall@3 is increased by 20.50%, and MRR@3 is increased by 18.07%.

Abstract The mineralization mechanism responsible for the fossilization of archaeological textiles in close proximity to metal artifacts presents a sophisticated preservation process at both macro and micro levels. This study examines archaeological textiles dating from 2200 BC to AD 1900, sourced from three distinct archaeological sites. The focus is on understanding the microstructural degradation of fibers within a specific burial environment and the preservation achieved through mineralization. These archaeological fibers of archaeological textiles exhibit morphological preservation in the immediate vicinity of copper-based objects. Utilizing tools such as a digital camera, scanning electron microscopy with energy dispersive spectroscopy (SEM–EDS), high-resolution synchrotron-based microtomography (μCT), and enzyme-linked immunosorbent assay (ELISA), we examined fiber morphology, conducted elemental analysis, identified fiber types, and analyzed fiber characteristics. Our findings reveal the presence of smooth-surfaced wools and silks, fibers covered with calculi, and fiber impressions—all subjected to mineralization. These mineralized fibers can be categorized into three distinct stages of mineralization, each exhibiting varying carbon content. We inferred a correlation between mineralization rate and carbon content while also identifying mineralization density distribution on these textiles. Lastly, this study provides insights into the preservation states of textiles across three different mineralization stages, enriching our understanding of the deterioration of organic archaeological material.

Abstract Background The infrazygomatic crest mini-screw has been widely used, but the biomechanical performance of mini-screws at different insertion angles is still uncertain. The aim of this study was to analyse the primary stability of infrazygomatic crest mini-screws at different angles and to explore the effects of the exposure length (EL), screw-cortical bone contact area (SCA), and screw-trabecular bone contact area (STA) on this primary stability. Methods Ninety synthetic bones were assigned to nine groups to insert mini-screws at the cross-combined angles in the occlusogingival and mesiodistal directions. SCA, STA, EL, and lateral pull-out strength (LPS) were measured, and their relationships were analysed. Twelve mini-screws were then inserted at the optimal and poor angulations into the maxillae from six fresh cadaver heads, and the same biomechanical metrics were measured for validation. Cuproptosis is a newly identified form of copper-dependent cell death that differs from other known pathways. This discovery provides a new way to explore copper-based nanomaterial applications in cancer therapy. This study used a layer-by-layer self-assembling method to load Cu2-xS nanoparticle (NP) cores with the siRNA of the PD-L1 immune escape-related gene and wrap a silk fibroin (SF) shell to form a multifunctional copper-based SF nanoplatform, denoted as CuS-PEI-siRNA-SFNs. CuS-PEI-siRNA-SFNs induced cuproptosis and exerted an antitumor effect via multiple mechanisms, including photothermal therapy (PTT), chemodynamic therapy (CDT), and immune activation. The presence of significant dihydrolipoamide S-acetyltransferase (DLAT) oligomers in 4T1 cells treated with CuS-PEI-siRNA-SFNs indicated the triggering of cuproptosis. Furthermore, in vivo experimental results showed that CuS-PEI-siRNA-SFNs efficiently accumulated in the tumor tissues of 4T1 tumor-bearing mice inhibited primary tumor and lung metastasis, and displayed excellent biosafety and antitumor activity. This study demonstrated that the synergistic effect of cuproptosis, PTT, CDT, and immune activation showed promise for treating metastatic breast cancer.

15. Effect of the diet level of whole-plant corn silage on the colonic microflora of Hezuo pigs

Background Whole-plant corn silage (WPCS) is an important roughage source for livestock, and have critical influences on rumen or intestinal microbiota, thus affecting the growth performance and feed efficiency. Our previous studies showed that adding WPCS to the diet of Hezuo pigs could promote the growth and fiber digestibility. While the aim of this study is to understand the effect of dietary WPCS on the colonic microflora in Hezuo pigs, which is essential for improving the roughage exploitation of pigs. Methods Thirty-two Hezuo pigs with similar body weight (7.88 ± 0.81) kg were selected and randomly divided into four groups with eight pigs in each group. Pigs in the control group were fed a basal diet, pigs in the experimental groups (Groups I, II, and III) were fed basal diet supplemented with 5%, 10%, and 15% WPCS, respectively, under 120 d experimental period. Six pigs from each group were picked for collecting colonic contents samples. 16S rRNA sequencing was performed to analyze the colonic microbiota of experimental pigs. Results The results showed that community richness indexes Chao1 and Observed_species in group III of Hezuo pig were significantly lower than that of the other three groups, community diversity indexes Shannon and Simpson were significantly higher in group I and II in comparison to the control group, and significantly lower in group III in comparison to the control group, group I and II. Adding WPCS to the diet of Hezuo pigs has no influence on the colonic dominant phylum, Clostridium sensu stricto 1 and Rikenellaceae RC9 gut group were most prevalent in the colon of Hezuo pig. When compared with the control group, the relative abundance of Streptococcus was significantly decreased in three experimental groups, while p-251-o5, Parabacteroides, Prevotellaceae UCG-003, Prevotellaceae UCG-001, and F082 exhibited significantly higher relative abundances in at least two experimental groups. Fibrobacter, Rikenellaceae RC9 gut group in group I, UCG-010 in group II, Bacteroides in group III exhibited increased relative abundance as compared with the control group. PICRUSt functional annotation indicated that the functions of cellular process and signaling were significantly increased in all WPCS-rationed groups, cancers, nervous system, immune system and environmental adaptation were all differed from groups I and II; three predominant pathways of translation, nucleotide metabolism and signal were only differed from the group II. Conclusions Feeding with 5% and 10% WPCS for Hezuo pigs could improve their colonic microflora diversity, and increase the relative abundance of fiber-digesting bacteria, which may potentially help to improve the fibre digestibility of Hezuo pigs by regulating the microbial function of cellular process and signaling, nucleotide metabolism, translation.

18. Prediction of specific structural damage to the knee joint using qualitative isokinetic analysis

Abstract Background An isokinetic moment curve (IMC) pattern-damaged structure prediction model may be of considerable value in assisting the diagnosis of knee injuries in clinical scenarios. This study aimed to explore the association between irregular IMC patterns and specific structural damages in the knee, including anterior cruciate ligament (ACL) rupture, meniscus (MS) injury, and patellofemoral joint (PFJ) lesions, and to develop an IMC pattern-damaged structure prediction model. Methods A total of 94 subjects were enrolled in this study and underwent isokinetic testing of the knee joint (5 consecutive flexion-extension movements within the range of motion of 90°-10°, 60°/s). Qualitative analysis of the IMCs for all subjects was completed by two blinded examiners. A multinomial logistic regression analysis was used to investigate whether a specific abnormal curve pattern was associated with specific knee structural injuries and to test the predictive effectiveness of IMC patterns for specific structural damage in the knee. Results The results of the multinomial logistic regression revealed a significant association between the irregular IMC patterns of the knee extensors and specific structural damages (“Valley” - ACL, PFJ, and ACL + MS, “Drop” - ACL, and ACL + MS, “Shaking” - ACL, MS, PFJ, and ACL + MS). The accuracy and Macro-averaged F1 score of the predicting model were 56.1% and 0.426, respectively. Conclusion The associations between irregular IMC patterns and specific knee structural injuries were identified. However, the accuracy and Macro-averaged F1 score of the established predictive model indicated its relatively low predictive efficacy. For the development of a more accurate predictive model, it may be essential to incorporate angle-specific and/or speed-specific analyses of qualitative and quantitative data in isokinetic testing. Furthermore, the utilization of artificial intelligence image recognition technology may prove beneficial for analyzing large datasets in the future.

Berberine (BBR) is a natural alkaloid, which has played an important role in the field of medicine since its discovery in the late 19th century. However, the low availability of BBR in vivo prevents its full effect. In recent years, a large number of studies confirmed that BBR has a protective effect on the nervous system through various functions, yet the issue of the inability to systematically understand the protection of BBR on the nervous system remains a gap that needs to be addressed. Many existing literature introductions about berberine in neurodegenerative diseases, but the role of berberine in the nervous system goes far beyond these. Different from these literatures, this review is divided into three parts: preparation method, mechanism, and therapeutic effect. Various dosage forms of BBR and their preparation methods are added, in order to provide a reasonable choice of BBR, and help to solve the problem of low bioavailability in treatment. More importantly, we more comprehensively summarize the mechanism of BBR to protect the nervous system, in addition to the treatment of neurodegenerative diseases (anti-oxidative stress, anti-neuroinflammation, regulation of apoptosis), two extra mechanisms of berberine for the protection of the nervous system were also introduced: bidirectional regulation of autophagy and promote angiogenesis. Also, we have clarified the precise mechanism by which BBR has a therapeutic effect not only on neurodegenerative illnesses but also on multiple sclerosis, gliomas, epilepsy, and other neurological conditions. To sum up, we hope that these can evoke more efforts to comprehensively utilize of BBR nervous system, and to promote the application of BBR in nervous system protection.

Abstract Background Insufficient interfragmentary compression force (IFCF) frequently leads to unstable fixation of osteoporotic lateral tibial plateau fractures (OLTPFs). A combined cancellous lag screw (CCLS) enhances IFCF; however, its effect on OLTPF fixation stability remains unclear. Therefore, we investigated the effect of CCLS on OLTPF stability using locking plate fixation (LPF). Materials and methods Twelve synthetic osteoporotic tibial bones were used to simulate OLTPFs, which were fixed using LPF, LPF-AO cancellous lag screws (LPF-AOCLS), and LPF-CCLS. Subsequently, 10,000 cyclic loadings from 30 to 400 N were performed. The initial axial stiffness (IAS), maximal axial micromotion of the lateral fragment (MAM-LF) measured every 1000 cycles, and failure load after 10,000 cycles were tested. The same three fixations for OLTPF were simulated using finite element analysis (FEA). IFCFs of 0, 225, and 300 N were applied to the LPF, LPF-AOCLS, and LPF-CCLS, respectively, with a 1000-N axial compressive force. The MAM-LF, peak von Mises stress (VMS), peak equivalent elastic strain of the lateral fragment (EES-LF), and nodes of EES-LF > 2% (considered bone destruction) were calculated. Results Biomechanical tests revealed the LPF-AOCLS and LPF-CCLS groups to be superior to the LPF group in terms of the IAS, MAM-LF, and failure load (all p 2% in the LPF were higher than those in the LPF-AOCLS and LPF-CCLS. Conclusion IFCF was shown to enhance the stability of OLTPFs using LPF. Considering overscrewing, CCLS is preferably recommended, although there were no significant differences between CCLS and AOCLS.

23. Life Cycle Environmental Impact Assessment on Two Aquaculture Models in the Yangtze River Basin

With the rapid development of aquaculture in China in recent decades, the country now faces environmental problems, such as high energy consumption and severe environmental pollution. The Yangtze River Basin is the most important area for freshwater aquaculture production in China, accounting for more than half of the total production. The environmental issues induced by aquaculture have received widespread consideration in this area. However, the freshwater farming-induced environmental issues in the Yangtze River Basin were rarely evaluated in prior research. Life cycle assessment (LCA) is a valid tool for assessing environmental impacts and resources and is widely deployed in the industrial sector. It can quantify the impact of a product or service on different environmental indicators at various stages of the production process, thereby aiding in the identification of the best measures to reduce environmental impacts. The LCA is more effective than traditional environmental impact assessment methods; thus, it has been gradually applied in other fields, such as agriculture and aquaculture, in recent years. In aquaculture, LCA has been used to evaluate the environmental impact of various farming models or species in a laboratory setting. However, little attention has been paid to large-scale systems, such as the Yangtze River Basin. The two most important farming models in China are the pond farming model and the integrated rice-fish farming model. Among them, the integrated rice-fish farming model is regarded as environmentally friendly and an important step towards realizing the sustainable development of aquaculture in terms of the efficient utilization of water and land resources. Therefore, in this study, the environmental effects of pond farming and integrated rice-fish farming models were investigated in the Yangtze River Basin using the LCA method. A total of 20 monitoring points were established in the Yangtze River Basin, including 5 for integrated rice-fish farming and 15 for pond culture. In the integrated rice-fish farming model, the life cycle consists of two stages: Feed supply and breeding. In pond culture, the life cycle consists of three stages: Feed supply, power supply, and breeding. We analyzed the values of energy use (EU), global warming potential (GWP), acidification potential (AP), eutrophication potential (EP), and water use (WU) in the pond farming and integrated rice-fish farming models using the LCA method, assessed the contribution rate of each phase for environmental indices, and compared the environmental impact of the two farming models. In order to establish the inputs and outputs of an aquaculture system, experimental measurements and surveys were conducted to collect data. The collected data mainly included feed consumption, electricity consumption, water consumption, total phosphorus and total nitrogen emissions, and greenhouse gas (CH4 and N2O) emissions. The data were analyzed by the LCA software eBalance. In this study, the weight gain of 1 ton of aquaculture products served as the functional unit, and the results were standardized and evaluated in order to compare the environmental impacts of various impact indicators. The results showed that the standardized and weighted evaluation values of WU, EP, GWP, AP, and EU in the integrated rice-fish farming model were 11.650, 0.770, 0.141, 0.096, and 0.003, respectively, and the total environmental impact indicator was 12.660. In the pond farming model, the values of WU, EP, GWP, AP, and EU were 31.453, 1.187, 0.210, 0.174, and 0.007, respectively, and the total environmental impact indicator was 33.031. The environmental impacts of integrated rice-fish farming were lower than those of pond farming. The contribution rate analysis of the three stages of the life cycle to environmental impact showed that the contribution rates of EU, GWP, and AP in the feed supply stage were higher than those in the power supply and breeding stages. EP was primarily associated with the feed supply stage and pollutant discharge stages of the breeding process, and WU was principally concentrated on the breeding stage. Overall, the LCA results showed that the integrated rice-fish farming model had a more environmentally friendly effect than the pond farming model, which indicates that the Yangtze River Basin has considerable development potential. WU was the most influential environmental impact indicator and the main restriction factor in aquaculture; it was mainly associated with the breeding stage. Therefore, moderately increasing stocking density was an effective strategy for reducing water resource consumption and improving water resource utilization rates. In addition to WU, the main environmental impact index of the pond culture model and integrated rice-fish farming models comprised EP, followed by GWP, AP, and EU. The contribution rate analyses of the two models showed that the aquaculture-induced environmental impacts could be mitigated by improving feed production technology, establishing accurate feeding technologies, applying advanced effluent water treatment technologies, and appropriately increasing stocking density in the Yangtze River Basin. In summary, in this study, we compared the environmental impacts between the pond culture and integrated rice-fish farming models in the Yangtze River Basin using the LCA method. In addition, we analyzed the differences between environmental impacts at different production stages. These data served as a reference for the sustainable development and optimization of aquaculture in the Yangtze River Basin.

Purposes A Knowledge Tracing Model based on Multiple Behavior Features Embedded Memory Networks (MFKT) was proposed to fully utilize the learning and forgetting features in interaction records. The MFKT model considers both learning and forgetting behaviors in the learning process. Methods First, two major features, learning and forgetting, are extracted from the interaction records, and then the extracted learning features are embedded into the memory network by scalar crossover, while the forgetting features are embedded by vector combination, which is used to enhance the learning ability of MFKT model for the students’ interaction sequences. In addition, after different students’ answers are completed, the difference in knowledge level growth is considered and a knowledge growth layer to the original memory network is added for calculating the knowledge growth obtained from students’ responses. Conclusion Experiment results on public datasets show that MFKT is more in line with real learning patterns of students and can realize more accurate tracing of students’ knowledge status.

Acanthocephalans of the order Polymorphida mainly parasitic in birds and mammals, are of veterinary, medical and economic importance. However, the evolutionary relationships of its 3 families (Centrorhynchidae, Polymorphidae and Plagiorhynchidae) remain under debate. Additionally, some species of Polymorphida (i.e. Bolbosoma spp. and Corynosoma spp.) are recognized as zoonotic parasites, associated with human acanthocephaliasis, but the mitochondrial genomes for representatives of Bolbosoma and Corynosoma have not been reported so far. In the present study, the complete mitochondrial genomes B. nipponicum and C. villosum (Acanthocephala: Polymorphidae) are reported for the first time, which are 14 296 and 14 241 bp in length, respectively, and both contain 36 genes [including 12 PCGs, 22 tRNA genes and 2 rRNA genes] and 2 non-coding regions (NCR1 and NCR2). The gene arrangement of some tRNAs in the mitogenomes of B. nipponicum and C. villosum differs from that found in all other acanthocephalans, except Polymorphus minutus. Phylogenetic results based on concatenated amino acid (AA) sequences of the 12 protein-coding genes (PCGs) strongly supported that the family Polymorphidae is a sister to the Centrorhynchidae rather than the Plagiorhynchidae, and also confirmed the sister relationship of the genera Bolbosoma and Corynosoma in the Polymorphidae based on the mitogenomic data for the first time. Our present findings further clarified the phylogenetic relationships of the 3 families Plagiorhynchidae, Centrorhynchidae and Polymorphidae, enriched the mitogenome data of the phylum Acanthocephala (especially the order Polymorphida), and provided the resource of genetic data for diagnosing these 2 pathogenic parasites of human acanthocephaliasis.

Hepatic steatosis is prevalent among cultured fish, yet the molecular mechanisms remain incompletely understood. This study aimed to assess changes in hepatic metabolic function in tilapia and to explore the underlying molecular mechanisms through transcriptomic analyses. Tilapia were allocated into two groups: a normal control (Ctr)-fed group and a high-fat diet (HFD)-fed group. Serum biochemical analyses revealed that HFD feeding led to liver damage and lipid accumulation, characterized by elevated levels of glutamic-pyruvic transaminase (GPT), glutamic-oxaloacetic transaminase (GOT), triglycerides (TGs), and total cholesterol (TC). Transcriptome analysis showed that 538 genes were significantly downregulated, and 460 genes were significantly upregulated in the HFD-fed fish. Gene Ontology (GO) enrichment analysis showed that these differentially expressed genes (DEGs) were apparently involved in the lipid metabolic process and monocarboxylic acid metabolic process. Meanwhile, Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis indicated significant alterations in pathways of steroid biosynthesis, porphyrin metabolism, terpenoid backbone biosynthesis, and retinol metabolism after HFD feeding. Additionally, results from Gene Set Enrichment Analysis (GSEA) revealed that gene expression patterns in pathways including oxidative phosphorylation, protein export, protein processing in the endoplasmic reticulum, and ribosome biogenesis were positively enriched in the HFD-fed tilapia. These findings provide novel insights into the mechanisms underlying HFD-induced hepatic dysfunction in fish, contributing to the optimization of feeding strategies in aquaculture.

This review provides a summary of research progress on salt coagulants, acid coagulants, enzyme coagulants, novel coagulants, polysaccharide additives, and various coagulant formulations. The coagulation mechanisms of various coagulants are also discussed. Accordingly, this paper seeks to offer reliable theoretical guidance for the development of novel coagulants and the realization of fully automated tofu production.

Herb compatibility is the soul of traditional Chinese Medicine prescriptions. Coptidis rhizoma (CR) (Coptis chinensis Franch., Coptis deltoidea C.Y.Cheng et Hsiao, or Coptis teeta Wall.; family Ranunculaceae), is a well-known herb. The bitter and cold nature of CR can irritate the spleen and stomach, and certain ingredients in CR may trigger allergic reactions. Herb combinations can help alleviate the side effects caused by CR. Through data analysis and literature research, there are many herbs combined with CR have a high frequency, but only a few are currently used as formulae in clinical practice. The results showed that these six herb pairs are usually widely studied or used as prescriptions in the clinic. This paper describes the six herb pairs from the key traditional uses, changes in bioactive constituents, and compatibility effects, especially with Euodiae fructus (family Rutaceae), Scutellariae radix (family Lamiaceae), Magnoliae Officinalis cortex (family Magnoliaceae), Glycyrrhizae radix et rhizoma (family Fabaceae), Ginseng radix et rhizoma (family Araliaceae), and Aucklandiae radix (family Asteraceae), and found that herbs are more effective when used in combination. Therefore, it is feasible to establish some methods to study herb pairs comprehensively from different perspectives. This paper aims to provide the latest and most comprehensive information on the six herb pairs and summarize the pattern of CR compatibility effects. It aims to attract more attention, and further experimental studies will be conducted to investigate and evaluate the effects of herb pairs containing CR. These data can also provide valuable references for researchers and also provide more possibilities for future applications in clinical practice and new drug development.

As a new generation of transportation, electric vehicles play an important role in carbon-peak targets. The development of electric vehicles needs the support of a charging network, and improper planning of charging stations will result in a waste of resources. In order to expand the charging network of electric vehicles and give full play to the low-carbon and efficient characteristics of electric vehicles, this paper proposed a charging station planning method that considers the characteristics of carbon emission trends. This paper combined the long short-term memory (LSTM) network with the stochastic impacts by regression on population, affluence, and technology (STIRPAT) model to predict the carbon emission trend and quantified the correlation between the construction speed of a charging station and the evolution characteristics of carbon emission by Pearson’s correlation coefficient. A multi-stage charging station planning model was established, which captures the dynamic characteristics of the charging demand of the transportation network and determines the station deployment scheme with economic and low-carbon benefits on the spatiotemporal scale. The Pareto frontier was solved by using the elitist non-dominated sorting genetic algorithm. The model and solution algorithm were verified by the actual road network in a certain area of Shanghai. The results showed that the proposed scheme can meet the charging demand of regional electric vehicles in the future, improve the utilization rate of charging facilities, and reduce the carbon emission of transportation networks.

Kuiper's statistic is a good measure for the difference of ideal distribution and empirical distribution in the goodness-of-fit test. However, it is a challenging problem to solve the critical value and upper tail quantile, or simply Kuiper pair, of Kuiper's statistics due to the difficulties of solving the nonlinear equation and reasonable approximation of infinite series. In this work, the contributions lie in three perspectives: firstly, the second order approximation for the infinite series of the cumulative distribution of the critical value is used to achieve higher precision; secondly, the principles and fixed-point algorithms for solving the Kuiper pair are presented with details; finally, finally, a mistake about the critical value cnα for (α,n)=(0.01,30) in Kuiper's distribution table has been labeled and corrected where n is the sample capacity and α is the upper tail quantile. The algorithms are verified and validated by comparing with the table provided by Kuiper. The methods and algorithms proposed are enlightening and worth of introducing to the college students, computer programmers, engineers, experimental psychologists and so on.

The interaction between cis-2 bis(benzofuro)[60]fullerene derivative and NO dominated double gas molecule was studied at the 6–311G(d,p) basis set level by density functional theory B3LYP-D3 method. The geometric structures of six interaction systems were optimized, and the optimized configurations were analyzed by electronic properties, infrared chromatography and independent gradient model to reveal the essence of intermolecular interaction. The results shows that NONO2, NON2, NOCO, NOCO2 and NOHCN are co-adsorbed on the surface of the compound in physical form, and the binding energies are −77.62 kJ/mol, −24.24 kJ/mol, −27.43 kJ/mol, −39.28 kJ/mol and −36.29 kJ/mol, respectively. The van der Waals interaction between gas molecules makes the three molecular interaction system have synergistic adsorption or competitive adsorption effect. Among them, the synergistic effect of NONO2 co-adsorption system is the biggest, and there is a strong attraction between NO and NO2 molecules. Compared with the interaction system of single gas molecule with cis-2 bis(benzofuro) [60]fullerene derivative, the interaction intensity of double gas molecule with the derivative is significantly improved.

Abstract µ-Conotoxin GIIIB (µ-CTX GIIIB) is a polypeptide containing three disulfide bridges, produced by the sea snail Conus geographus. This study was aimed to explored the cytotoxic effects of µ-CTX GIIIB on mouse skeletal musculoblast (Sol8). Sol8 cells were exposed to ouabain and veratridine to establish the cell injury model, and then treated with µ-CTX GIIIB. CCK-8 was adopted to evaluate the cytotoxicity of µ-CTX GIIIB. Then, proteomics and transcriptome were conducted, and the explore the differentially expressed genes (DEGs) and differentially expressed proteins (DEPs) affected by µ-CTX GIIIB were found. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis was used to investigate the affected signaling pathways. µ-CTX GIIIB increased the cell survival rate of injured Sol8 cells. We found and identified 1,663 DEGs and 444 DEPs influenced by µ-CTX GIIIB. 106 pairs of correlated DEGs and DEPs were selected by combining transcriptome and proteome data. The results of KEGG and GO analysis showed that µ-CTX GIIB affected the cell cycle, apoptosis, DNA damage and repair, lipid metabolism and other biological processes of Sol8 cells. µ-CTX GIIIB could affected cell cycle regulation, DNA damage repair, and activation of tumor factors, with potential carcinogenic effects. Our results provide an important basis for the study of in vitro toxicity, the mechanism of toxicity and injury prevention by µ-CTX GIIIB.

The extensive outbreak of Sargassum horneri in China has not merely imposed a severe threat to the ecological environment and human life in coastal waters but also impeded the development of waterway transportation and the local economy. Consequently, we isolated polysaccharides from S. horneri, designated as SHP, and evaluated the antioxidant activity of SHP both in vitro and in vivo by investigating the effect of SHP on H2O2-induced African green monkey kidney cells (Vero cells) and zebrafish. The results demonstrated that SHP can enhance the activities of superoxide dismutase, catalase, and glutathione peroxidase in zebrafish. It also effectively inhibits micro malondialdehyde and ROS levels in Vero cells and zebrafish to mitigate the oxidative damage caused by H2O2, thereby achieving the protective effect of SHP on Vero cells and zebrafish. In conclusion, SHP holds the potential as a natural antioxidant. SHP can be contemplated for utilization as a natural antioxidant in the biomedical, cosmetic, and food industries, thereby alleviating the environmental stress caused by S. horneri and achieving resource utilization.

42. Alterations in Soil Bacterial Community and Its Assembly Process within Paddy Field Induced by Integrated Rice–Giant River Prawn (Macrobrachium rosenbergii) Farming

Integrated rice–aquatic animal farming has become a vital strategy for enhancing food security. To assess the effects of integrated rice–giant river prawn (Macrobrachium rosenbergii) farming (IRPF) on agricultural ecosystems, we used 16S rRNA gene sequencing to analyze soil bacterial communities in comparison with traditional rice monoculture (RM). Our study revealed that the IRPF did not significantly affect the diversity of the soil bacterial community. However, during the initial culture stage, IRPF markedly increased the relative abundance of the phylum candidate division NC10 and the genus Candidatus Methylomirabilis, enhancing nitrogen-cycling-related functions within the bacterial community. Additionally, IRPF reduced the complexity and stability of these communities in the early to middle culture stages. While stochastic processes usually dominate the assembly of these communities, IRPF restricted bacterial migration and reduced the influence of these stochastic processes. Furthermore, IRPF had a significant impact on environmental factors within paddy soils, strongly correlating with shifts in bacterial communities, particularly through variations in soil nitrite concentration. In conclusion, the influence of IRPF on the bacterial community in paddy soils was primarily observed during the early and middle culture stages, and the impacts of IRPF on the soil bacterial community were primarily driven by environmental changes, especially soil nitrite concentration. These findings provide theoretical insights and a reference for understanding the microbiological impacts of different integrated rice–fish farming systems on agricultural ecosystems.

Cyprinus carpio is a significant freshwater species with substantial nutritional and economic value. Rice–carp co-culture represents one of its principal cultivation methods. However, in the system, the optimal farming density for carp and the impact of high stocking density on their muscle nutritional composition have yet to be explored. Thus, the objective of the current study was to investigate the influences of stocking density on the muscle nutrient profiles and metabolism of C. carpio in rice–fish co-culture systems. Common carp were cultured at three stocking densities, low density (LD), medium density (MD), and high density (HD), over a period of 60 days. Following this, comprehensive analyses incorporating physiological, biochemical, and multi-omics sequencing were conducted on the muscle tissue of C. carpio. The results demonstrated that HD treatment led to a reduction in the antioxidant capacity of C. carpio, while resulting in elevated levels of various fatty acids in muscle tissue, including saturated fatty acids (SFAs), omega-3 polyunsaturated fatty acids (n-3 PUFAs), and omega-6 polyunsaturated fatty acids (n-6 PUFAs). The metabolome analysis showed that HD treatment caused a marked reduction in 43 metabolites and a significant elevation in 30 metabolites, primarily linked to lipid and amino acid metabolism. Additionally, transcriptomic analysis revealed that the abnormalities in lipid metabolism induced by high-stocking-density treatment may be associated with significant alterations in the PPAR signaling pathway and adipokine signaling pathway. Overall, our findings indicate that in rice–fish co-culture systems, high stocking density disrupted the balance of antioxidant status and lipid metabolism in the muscles of C. carpio.

The rice–fish co-culture is an important model of carp farming in China; however, research on the dynamics and assembly of water bacterial communities in this system remains limited. Therefore, this study aimed to explore the dynamics and assembly of bacterial communities, as well as their correlation with environmental factors in paddy water. For these purposes, this study was divided into two groups: a rice–carp co-culture (WRC) group and a rice monoculture (WRM) group, with 20 rice fields in each group. After 60 days of farming, the concentrations of NH4+-N, NO2−-N, TN, and PO43− were significantly higher in the WRC group than those in the WRM group. Alpha diversity analysis showed that the Shannon index in the WRC group was significantly decreased compared with the WRM group. At the phylum level, the relative abundance of Actinobacteria significantly increased, while the relative abundance of Proteobacteria and Bacteroidetes significantly decreased in the paddy water of the WRC group. The neutral community model (NCM) indicated that a random process played a dominant role in the construction of bacterial communities in the two groups, and common carp cultivation increased migration rates, thereby affecting community assembly. The co-occurrence network displayed that common carp cultivation led to looser interactions between bacterial communities. In addition, the contents of nutrients significantly affected the abundance of bacteria in paddy water. In summary, carp cultivation decreased the diversity of bacteria and changed the relative abundance of dominant bacteria, thereby affecting the stability and assembly of bacterial communities.

ABSTRACTImbalanced gut microbiota (GM) and abnormal fecal bile acid (BA) are thought to be the key factors for diarrhea-predominant irritable bowel syndrome (IBS-D), but the underlying mechanism remains unclear. Herein, we explore the influence of the GM–BA–Takeda G-protein-coupled receptor 5 (TGR5) axis on IBS-D. Twenty-five IBS-D patients and fifteen healthy controls were recruited to perform BA-related metabolic and metagenomic analyses. Further, the microbiota-humanized IBS-D rat model was established by fecal microbial transplantation (FMT) to investigate the GM–BA–TGR5 axis effects on the colonic barrier and visceral hypersensitivity (VH) in IBS-D. Finally, we used chenodeoxycholic acid (CDCA), an important BA screened out by metabolome, to evaluate whether it affected diarrhea and VH via the TGR5 pathway. Clinical research showed that GM associated with bile salt hydrolase (BSH) activity such as Bacteroides ovatus was markedly reduced in the GM of IBS-D, accompanied by elevated total and primary BA levels. Moreover, we found that CDCA not only was increased as the most important primary BA in IBS-D patients but also could induce VH through upregulating TGR5 in the colon and ileum of normal rats. TGR5 inhibitor could reverse the phenotype, depression-like behaviors, pathological change, and level of fecal BSH in a microbiota-humanized IBS-D rat model. Our findings proved that human-associated FMT could successfully induce the IBS-D rat model, and the imbalanced GM–BA–TGR5 axis may promote colonic mucosal barrier dysfunction and enhance VH in IBS-D.IMPORTANCEVisceral hypersensitivity and intestinal mucosal barrier damage are important factors that cause abnormal brain–gut interaction in diarrhea-predominant irritable bowel syndrome (IBS-D). Recently, it was found that the imbalance of the gut microbiota–bile acid axis is closely related to them. Therefore, understanding the structure and function of the gut microbiota and bile acids and the underlying mechanisms by which they shape visceral hypersensitivity and mucosal barrier damage in IBS-D is critical. An examination of intestinal feces from IBS-D patients revealed that alterations in gut microbiota and bile acid metabolism underlie IBS-D and symptom onset. We also expanded beyond existing knowledge of well-studied gut microbiota and bile acid and found that Bacteroides ovatus and chenodeoxycholic acid may be potential bacteria and bile acid involved in the pathogenesis of IBS-D. Moreover, our data integration reveals the influence of the microbiota–bile acid–TGR5 axis on barrier function and visceral hypersensitivity.

Background: The functional movement screen (FMS) has been used to identify deficiencies in neuromuscular capabilities and balance among athletes. However, its effectiveness in detecting movement anomalies within the population afflicted by knee osteoarthritis (KOA), particularly through the application of a family-oriented objective assessment technique, remains unexplored. The objective of this study is to investigate the sensitivity of the FMS and daily activities in identifying kinematic abnormalities in KOA people employing a markerless motion capture system.Methods: A total of 45 persons, presenting various Kellgren–Lawrence grades of KOA, along with 15 healthy controls, completed five tasks of the FMS (deep squat, hurdle step, and in-line lunge) and daily activities (walking and sit-to-stand), which were recorded using the markerless motion capture system. The kinematic waveforms and discrete parameters were subjected to comparative analysis.Results: Notably, the FMS exhibited greater sensitivity compared to daily activities, with knee flexion, trunk sagittal, and trunk frontal angles during in-line lunge emerging as the most responsive indicators.Conclusion: The knee flexion, trunk sagittal, and trunk frontal angles during in-line lunge assessed via the markerless motion capture technique hold promise as potential indicators for the objective assessment of KOA.

Colorectal cancer is the third most common type of cancer worldwide and has become one of the major human disease burdens. In clinical practice, the treatment of colorectal cancer has been closely related to the use of irinotecan. Irinotecan combines with many other anticancer drugs and has a broader range of drug combinations. Combination therapy is one of the most important means of improving anti-tumor efficacy and overcoming drug resistance. Reasonable combination therapy can lead to better patient treatment options, and inappropriate combination therapy will increase patient risk. For the colorectal therapeutic field, the significance of combination therapy is to improve the efficacy, reduce the adverse effects, and improve the ease of treatment. Therefore, we explored the clinical advantages of its combination therapy based on mechanism or metabolism and reviewed the rationale basis and its limitations in conducting exploratory clinical trials on irinotecan combination therapy, including the results of clinical trials on the combination potentiation of cytotoxic drugs, targeted agents, and herbal medicine. We hope that these can evoke more efforts to conduct irinotecan in the laboratory for further studies and evaluations, as well as the possibility of more in-depth development in future clinical trials.
